Rubus moluccanus L., commonly known as Molucca bramble or broad-leaf bramble is a member of family Rosaceae which occurs mainly in tropical Asia and Australia. It has applications in indigenous medicines and is used in the treatment of number of ailments like headache, diarrhoea, dysentery, abdominal pain, nocturnal micturition of children and enhancement of female fertility. This plant is a rich and valuable source of bioactive flavonoids, terpenes and other chemical compounds. The presence of these active chemical compounds exerts antibacterial, antifungal, antihelminthic and antioxidant activities. Regardless of its use in various diseases and disorders, the information about this plant is scanty. So the present review assessed and summarised the knowledge on taxonomy, morphology, geographical distribution, nutritional value, ethnobotany, phytochemistry and biological activities of R. moluccanus. Raspberry (Rubus idaeus L.) is susceptible to aphid-borne viruses. We studied the incidence of four of them – black raspberry necrosis virus (BRNV), raspberry leaf mottle virus (RLMV), raspberry vein chlorosis virus (RVCV), and Rubus yellow net virus (RYNV) – in raspberry plants and aphids in and around Norwegian raspberry crops for three years (2019, 2021, and 2022). Most of the samples were from symptomatic plants. Applying RT-PCR, 274 leaf samples and 107 aphid samples were analyzed. All four viruses were found, but BRNV dominated: it was detected in 93% of the 178 leaf samples with virus and was the only virus that occurred more frequently as a single infection than in co-infections with the other viruses. The old cv. Veten had the highest virus incidence (97%) among the sampled plants, followed by uncultivated raspberry in the boundary vegetation (82%). All aphids identified were Amphorophora idaei and Aphis idaei. BRNV and/or RLMV was detected in 27% of the aphid samples. Notably, BRNV was detected in 30% of A. idaei samples, a species not known as a BRNV vector. In subsequent transmission experiments we found that although A. idaei can acquire BRNV within one hour, it did not transmit the virus to healthy raspberry plants. In contrast, Am. idaei , a known BRNV vector, was able to acquire the virus within one minute and transmit it within one hour of inoculation. The stems of Rubus idaeus L., a byproduct of the fruit–food industry, are known sources of bioactive compounds. The main objective of this study was to investigate the composition of polyphenolic compounds in R. idaeus stems. Seven cultivated raspberry varieties, thirteen garden samples, including five well-known raspberry varieties, and thirteen wild raspberry samples from different locations in Estonia were analyzed. The HPLC-MS method detected 62 substances, of which 42 were identified, 12 were tentatively identified, and 8 compounds remained unknown. Protocatechuic acid pentoside was dominant in most varieties and in all garden and wild raspberry samples. Dihydroxybenzoic acid hexoside 1, p-coumaroyl quinic acid 1, quercetin 4'-glucuronide, and p-coumaric acid glycoside were found in significant quantities. Correlations among the contents of individual compounds were established. When studying the dynamics of polyphenolic compound accumulation in, for example, the GR1 sample over a year, it was found that, in raspberry stems, the largest amount of them accumulated in April and slightly less in January and October. Investigating the dependence of the accumulation of polyphenols on the parts of the stem, it was found that the upper parts have the highest phenolic contents. Rubus idaeus is a pivotal cultivated species of raspberry known for its attractive color, distinct flavor, and numerous health benefits. It can be used in pharmaceutical, cosmetics, agriculture and food industries not only as fresh but also as a processed product. Nowadays due to climatic changes, genetic diversity of cultivars has decreased dramatically. However, until now, the status of wild R. idaeus resources in China have not been exploited. In this study, we investigated the resources of wild R. idaeus in China to secure its future potential and sustainability. The MaxEnt model was used to predict R. idaeus suitable habitats and spatial distribution patterns for current and future climate scenarios, based on wild domestic geographic distribution data, current and future climate variables, and topographic variables. The results showed that, mean temperature of the coldest quarter (bio11), precipitation of the coldest quarter (bio19), precipitation of the warmest quarter (bio18), and temperature seasonality (bio4) were crucial factors affecting the distribution of R. idaeus. Presently, the suitable habitats were mainly distributed in the north of China including Xinjiang, Inner Mongolia, Gansu, Ningxia, Shaanxi, Shanxi, Hebei, Beijing, Liaoning, Jilin, Heilongjiang. According to our results, in 2050s, the total suitable habitat area of R. idaeus will increase under SSP1-2.6 and then will be decreased with climate change, while in the 2090s, the total suitable habitat area will continue to decrease. From the present to the 2090s, the centroid distribution of R. idaeus in China will shift towards the east and the species will always be present in Inner Mongolia. Inspired by the methods developed in J. H. Bramble, T. Dupont, and V. Thomée (“Projection methods for Dirichlet’s problem in approximating polygonal domains with boundary-value corrections,” Math. Comput., vol. 26, no. 120, pp. 869–879, 1972), we introduce a new technique that yields a symmetric formulation and has similar performance. The new method is based on a Robin-type problem on an approximate polygonal domain. Optimal error estimates in the energy norm are proved for piecewise quadratics and cubics. The incidence of diabetes mellitus, a pressing global health concern that is rapidly increasing, has prompted the exploration of medicinal plants for potential remedies, particularly those within the Rubus genus. This comprehensive review aims to connect traditional knowledge with scientific insights, shedding light on the antidiabetic properties of various Rubus species. An exhaustive exploration of the literature revealed that Rubus chingii Hu, Rubus idaeus, Rubus ulmifolius, Rubus fruticosus, Rubus amabilis, and some other Rubus species exhibited noteworthy antidiabetic effects, each operating via distinct mechanisms. Furthermore, Rubus species serve as abundant reservoirs of bioactive compounds, including flavonoids, phenolic acids, tannins, alkaloids, glycosides, antioxidants, triterpenoids, and sterols. As indicated by various in vivo and in vitro studies, these compounds notably improve insulin secretion, increase hepatic glycogen synthesis, inhibit key digestive enzymes, enhance the functions of β-cells in the pancreas, and effectively reduce blood glucose levels. This review, which describes the antidiabetic potential of Rubus species in terms of both pharmacological effects and traditional uses, offers valuable insights for future scientific initiatives in the development of innovative antidiabetic therapies. The first record of bramble sharks (Echinorhinus brucus) in the United Arab Emirates is presented. In situ observations of multiple bramble sharks were made at depths between 460 and 720 m from two piloted submersibles and a remotely operated vessel, representing the first known observations of this species in its native deep-water habitat in Arabia and the Indian Ocean. Notably, this research expands on the documented regional distribution of E. brucus for the Gulf of Oman/Arabian Sea and extends this species' regional records to deeper mesophotic zones. These findings underscore the need for further research to understand the ecology and distribution of this cryptic shark species, particularly given its global endangered status and the limited knowledge of its regional population dynamics. Salmonberry (Rubus spectabilis) and thimbleberry (Rubus nutkanus) are important native berry plants for Indigenous communities and restoration sites in the coastal region of British Columbia (BC). Compared to cultivated berry plants, information on the diseases impacting native berry plant species is limited. In late August 2022, powdery mildew symptoms and signs were observed on field plots of salmonberry and thimbleberry plants at the Agassiz Research and Development Centre, BC. The disease symptoms progressed until the end of September. The morphology of conidiophores and conidia when examined under a compound and scanning electron microscope showed they matched with those of Podosphaera aphanis. DNA from diseased foliage was subjected to PCR amplification with primers representing the internal transcribed spacer (ITS) region of rDNA followed by sequencing. The corresponding sequences from both hosts showed >99% similarity to P. aphanis in GenBank and grouped with several accessions of P. aphanis following phylogenetic analysis. In artificially inoculated trials conducted in the greenhouse, powdery mildew symptoms developed within 3−4 weeks post-inoculation and P. aphanis isolates from salmonberry and thimbleberry were cross-pathogenic to each host. To our knowledge, this is the first study characterizing the powdery mildew pathogen infecting salmonberry and thimbleberry and demonstrating cross-pathogenicity to both hosts. Genomic selection (GS) is becoming increasingly widespread and applied due to the promising results obtained, cost savings in generating single nucleotide polymorphism (SNP) markers, and the development of statistical models that allow to improve the analysis robustness and accuracy. The composition and size of the training population have a major influence on GS, which poses challenges for interspecific biparental populations. Another factor is the use of different reference genomes from other species to perform SNP calling, which could make it possible to explore variability in interspecific crosses comprehensively. Late leaf rust is a disease caused by the pathogen Acculeastrum americanum, and there are reports on genetic resistance in Rubus occidentalis, which leads to the need for interspecific hybridizations, aiming to combine the fruit quality of R. idaeus with the resistance of R. occidentalis. The present study was carried out with a population of 94 interspecific raspberry hybrids. We evaluated the effect of different reference genomes on the SNP markers discovery, as well as training population optimization strategies on the accuracy of genomic predictions, namely the CV-α, leaving-one-family-out (LOFO), pairwise families, and stratified k-fold. The average predictive accuracies ranged from − 0.33 to 0.44 and We demonstrated higher prediction accuracy and more precise estimates when we combined stratified sampling to compose the training set (CV-α and k-fold stratified CV) and the panel of Unique markers. These results corroborate that genomic prediction aligned with SNP calling and training population optimization strategies can significantly increase genetic gains in interspecific biparental crosses. Over the last 10 years, global raspberry production has increased by 47.89%, based mainly on the red raspberry species (Rubus idaeus). However, the black raspberry (Rubus occidentalis), although less consumed, is resistant to one of the most important diseases for the crop, the late leaf rust caused by Acculeastrum americanum fungus. In this context, genetic resistance is the most sustainable way to control the disease, mainly because there are no registered fungicides for late leaf rust in Brazil. Therefore, the aim was to understand the genetic architecture that controls resistance to late leaf rust in raspberries. For that, we used an interspecific multiparental population using the species mentioned above as parents, 2 different statistical approaches to associate the phenotypes with markers [GWAS (genome-wide association studies) and copula graphical models], and 2 phenotyping methodologies from the first to the 17th day after inoculation (high-throughput phenotyping with a multispectral camera and traditional phenotyping by disease severity scores). Our findings indicate that a locus of higher effect, at position 13.3 Mb on chromosome 5, possibly controls late leaf rust resistance, as both GWAS and the network suggested the same marker. Of the 12 genes flanking its region, 4 were possible receptors, 3 were likely defense executors, 1 gene was likely part of signaling cascades, and 4 were classified as nondefense related. Although the network and GWAS indicated the same higher effect genomic region, the network identified other different candidate regions, potentially complementing the genetic control comprehension. In this study, the effect of maltodextrin, powdered sugar, and corn starch carrier agents used at different ratios (5% and 10%) in the convective dryer at 65 ºC and hybrid dryer (microwave + convective) at 350 W + 65 ºC to produce blackberry powder was investigated. Drying kinetics, energy analyses, physical, flow properties, and biochemical analyses of blackberry powder production processes were investigated. Drying rates in drying processes varied between 0.0052–0.0477 g moisture/g dry matter minute. Effective moisture diffusion values were determined between 3.36 × 10–8-2.57 × 10–7 m2/s. Specific moisture absorption rate and specific energy consumption values were found to vary between 0.0019–0.0034 kg/kWh and 237.15–530.00 kWh/kg, respectively. Tapped density was determined in the range of 1.666–2.765 g/ml, while bulk density was determined in the range of 1.319–1.937 g/ml. The wettability values of blackberry powders were found to vary between 2.00–27.67 s. Drying processes did not preserve the color values of fresh blackberry puree (p < 0.05). In bioactive findings, total phenol content values were 16.756–25.876 µg GAE/g−1 dw, total monomeric anthocyanin values were 229–1.469 µg cy−3-glu/g−1 dw, total flavonoid values 3.958–5.080 mg KE/kg dw and total antioxidant activity values 406–500 µmol TE/g−1 dw. 1. Recent research has shown that encapsulated raspberry powder (RP) is a natural colourant for foodstuffs. However, no research has been conducted on its use in chicken nuggets. In addition, the effect of RP on products with and without phosphate addition is unknown. This study assessed the effects of RP (control, 0.5%, 1.0%) and phosphate (0.0%, 0.3%) on the pH and colour quality properties of nuggets. 2. In the production of RP, red raspberry (Rubus ideaus L.) juices were encapsulated using maltodextrin in a spray-dryer. Antioxidant activity, total anthocyanin, total phenolics, colour, moisture and pH analyses of the RP were performed. 3. Nuggets were packaged in modified atmosphere packaging (MAP; 40%CO2 + 60%N2) and were stored at 2.0 ± 0.5°C for 120 d. The pH and external and internal surface colour (L*, a*, b*, C* and h) values were measured on d 0, 15, 30, 45, 60, 75, 90, 105 and 120 of storage. 4. The addition of phosphate increased the pH in the samples, while these decreased with the addition of RP (p < 0.05). During storage, the highest pH were seen in the phosphate samples and the lowest in the nuggets with 1.0% RP addition (p < 0.05). 5. With the addition of phosphate, the external surface a* value of nuggets increased (p < 0.05). Depending on the level of RP added to the nuggets, the external surface L* value decreased and a* and b* values increased (p < 0.05). After d 30 of storage, the a* value increased in the samples with RP addition and this increase was higher in the with phosphate nuggets (p < 0.05). 6. The internal surface a* value increased with the addition of RP during nugget production (p < 0.05). The increase in a* value was greater in samples with added phosphate (p < 0.05). During storage, the highest a* values were seen in nuggets treated with phosphate + 0.1% RP (p < 0.05). BACKGROUND: Besides providing nutritional value, functional foods have potential beneficial effects on health. Consumers worldwide seek food products with added value based on physical and mental well-being. OBJECTIVE: This study aimed to analyze the urinary metabolome of volunteers who consumed a drink based on Costa Rican blackberries (Rubus adenotrichos) to identify the presence of urolithins and other possible bioactive compounds related to beneficial effects on health. METHODS: Fifteen healthy non-smoking male volunteers aged between 19 and 43 years old who consumed 250 ml of blackberry-based drink for breakfast for eight consecutive days were selected. Urine samples were obtained before and after drink consumption, which were submitted to analysis by Ultra-Performance Liquid Chromatography coupled to a Mass Detector (UPLC/ESI-Q-TOF-MS). Subsequently, a metabolomic analysis of the results was performed to identify the presence or absence of urolithins and other bioactive metabolites related to blackberry juice consumption. RESULTS: It was possible to detect high intensity presence of urolithin A (n = 6) and high intensity presence of both urolithin A and B (n = 5) in urine samples in several volunteers. However, finding them was impossible in others, or they were at very low intensities (n = 4). Likewise, it was possible to identify Menth-1-en-4,8 diol glucuronide and 1-O-E-Cinnamoyl-(6-arabinosylglucose) as other compounds related to the consumption of blackberries. CONCLUSIONS: The consumption of Costa Rican blackberries allowed us to demonstrate the presence of three metabotypes within the study population: 40% of strong urolithin A (UAP) producers, 33.3% of both urolithin (A and B) producers (UBP), and 26.7% of weak urolithin A producers (0P). Salinity is one of the most important abiotic stress factors that affect plant growth and limit agricultural productivity. In this study, the effects of iron (FeNP), silver (AgNP), and silicon dioxide (SiNP) nanoparticles on the morphological and physiological parameters of in vitro boysenberry plants grown under salinity stress (NaCl) were investigated. According to our study results, higher values were obtained from SiNP application in terms of shoot development parameters; FeNP application was found to be more successful for root development; AgNP application was effective in terms of SPAD, leaf relative water content (LRWC), and relative growth rate (RGR); and FeNP application increased superoxide dismutase (SOD) and catalase (CAT) enzyme activities. Salt stress significantly affected root development, SPAD values, LRWC and RGR, and SOD and CAT enzyme activities. As a result, under salt stress conditions, SiNP, FeNP, and AgNP applications can significantly reduce the negative effects of stress and promote the vegetative development of the plant compared to control conditions. Raspberry (Rubus idaeus L.) fruit are known for their extremely short shelf life. Decay, leakiness, and loss of firmness are the most common limiting factors contributing to their short storage life. However, storage in elevated CO2 and reduced O2 atmospheres can delay senescence in fruit by reducing softening, respiration and ethylene production rates, and pathogen growth. In this study, raspberries were exposed to four different CO2 atmospheres—15 kPa CO2 and 6 kPa O2 (15 kPa); 8 kPa CO2 and 13 kPa O2 (8 kPa); 5 kPa CO2 and 16 kPa O2 (5 kPa); or 0.03 kPa CO2 and 21 kPa O2 (0.03 kPa)—and were evaluated for their postharvest quality periodically during two weeks of storage in 2020 and 2021. Raspberry fruits kept in a 15 kPa CO2 atmosphere followed by 8 kPa CO2 had higher firmness, brighter red color, and the least fungal decay or leakiness. In all atmospheres, the total anthocyanin content increased over time, although the rate of increase was slowed by high CO2. The raspberries' visual attributes deteriorated over time in all atmospheres, but high CO2 atmospheres slowed the rate of deterioration. After five days, the quality of air-stored raspberries was significantly degraded, while the raspberries stored in elevated CO2 maintained good quality for up to ten days. [ABSTRACT FROM AUTHOR]

We studied Rubus wahlbergii and related species in R. sect. Corylifolii, primarily in Sweden, using DNA microsatellites. The results show that the material can be divided into three species: R. wahlbergii s.s. which occurs along the Swedish coast from Bohuslän to Uppland, R. nordicus which occurs on the western coast all the way south to north‐western Skåne, and a previously unrecognized taxon which we here describe as R. wahlbergioides. The latter has scattered localities in Halland, Skåne, Blekinge, Öland and Småland, but is common around Kivik in Skåne and Kalmar in Småland. It is characterised by leaves that are without felt underneath, terminal leaflets with a somewhat more triangular shape and coarser serration than those of R. wahlbergii and also has more glands on the stem and the inflorescence. In the northern part of the distribution, R. nordicus is easily distinguished morphologically from R. wahlbergii, e.g. by a strong tendency to form 7‐foliolate leaves, but in Skåne and Halland, the two species are harder to distinguish. Mycosed mites Acalitus orthomera were observed on blackberry fruits cv. 'Tupy' in Los Reyes de Salgado, Michoacan, Mexico. Acalitus orthomera appeared during the fruiting period of the crop (February-May) of 2023. Sarocladium terricola (J. Mill, Giddens and A. Foster) was isolated from fruits on which the mycosed mites were found, and they are the etiological agents of mycosis. The mycosed mites were first identified morphologically from in vitro cultures and then molecularly by the use of standard PCR using ITS4 and ITS5 primers on the fungal mycelia. This is the first report that Sarocladium terricola is a natural enemy of Acalitus orthomera on blackberry. [ABSTRACT FROM AUTHOR]

Key message: The combination of a flow cytometric seed screen and genotyping of each single seed offers a cost-effective approach to detecting complex reproductive pathways in flowering plants. Reproduction may be seen as one of the driving forces of evolution. Flow cytometric seed screen and genotyping of parents and progeny are commonly employed techniques to discern various modes of reproduction in flowering plants. Nevertheless, both methods possess limitations constraining their individual capacity to investigate reproductive modes thoroughly. We implemented both methods in a novel manner to analyse reproduction pathways using a carefully selected material of parental individuals and their seed progeny. The significant advantage of this approach lies in its ability to apply both methods to a single seed. The introduced methodology provides valuable insights into discerning the levels of apomixis, sexuality, and selfing in complex Rubus taxa. The results may be explained by the occurrence of automixis in Rubus, which warrants further investigation. The approach showcased its effectiveness in a different apomictic system, specifically in Taraxacum. Our study presents a comprehensive methodological approach for determining the mode of reproduction where flow cytometry loses its potential. Red raspberries, Rubus idaeus L. 1753 are famous fruits which possess high value bioactive compounds. In this study, we report the complete chloroplast genome of R. idaeus, it displayed a typical quadripartite structure with 155687 bp in length. The genome encodes 127 genes including 79 protein coding genes, 8 rRNA genes and 40 tRNA genes, the overall GC content is 37.2%. Phylogenetic analysis revealed a close relationship between R. idaeus and R. sachalinensis in Section Malaehobatus. [ABSTRACT FROM AUTHOR]

Raspberry ketone accounts for the characteristic aroma of the raspberry fruit. In order to explore the genes involved in raspberry ketone synthesis, the transcriptome in fruit tissues of two red raspberry varieties "Polka" and "Orange legend", were sequenced and 24213 single genes were obtained. As the red raspberry fruit ripening, genes involved in flavonoid and anthocyanin synthesis were up-regulated, while those associated with lignin synthesis were down-regulated. A gene (RinPKS4) highly related to raspberry ketone synthesis was identified by transcriptome analysis, and RinPKS4 gene was over-expressed in raspberry in order to further understand the function of RinPKS4 gene in raspberry ketone synthesis. The results showed that the gene expression level of RinPKS4 in the leaf tissues of a transgenic lines increased by about 4-fold and the content of raspberry ketone increased by 42.64% compared with the wide type. Agrilus cuprescens (Ménétries, 1832), the rose stem girdler, is a Palearctic Buprestidae (Coleoptera) pest of Rosa and Rubus species. Its introduction and consequent spread across North America may now make it the most widely distributed Agrilus species, globally.No evidence reviewed indicated A. cuprescens was present in North America long before it was first reported on Rosa in New Jersey, USA, in 1913. The species incrementally progressed westward, reaching the Pacific coast in Washington, USA, 101 years later. Future spread is most likely within the northern Canadian prairies and the Pacific Northwest regions. Its presence in the latter region poses a contemporary threat to epicentres of Rubus production in both the United States and Canada. As of 2022, confirmed spread within the region extended nearly to the Canadian border in Washington and the southern Willamette Valley in Oregon.The most comprehensive, foundational investigations of A. cuprescens in North America were conducted in the late 1930s and early 1940s in New York, USA, and in Utah, USA, during the late 1950s and early 1960s following damaging outbreaks on Rubus. Ongoing research in the Pacific Northwest is attempting to refine information on this pest as it progresses towards naturalization within a new North American ecological and agricultural context.This review aggregates and clarifies the geographical progression, biological understandings and management of this species in North America, which has been fraught with a convoluted history of misidentifications, and varying degrees of consideration given to it as a pest of concern. This study aimed to determine the amounts of phenols, antioxidant activity, and sensory compounds in three commercial cultivars of blackberries popular in Lithuania: 'Polar', 'Brzezina', and 'Orkan'. Blackberry pomace was analyzed by the spectrophotometric method for total phenolic content, total flavonoid content and radical-scavenging capacity using the DPPH• and ABTS•+ assays. The phenolic profiles, organic acids, and sugars were analyzed by HPLC. The Heracles II electronic nose, which is based on ultrafast gas chromatography, was used for the quantification of volatile organic compounds. The results show that the total phenolic content of blackberry pomace varied from 2380.60 to 2088.00 mg 100 g−1 and that the total flavonoid content varied from 161.29 to 148.10 mg 100 g−1, depending on the cultivar. A total of 14 polyphenols were also identified, with epigallocatechin and anthocyanin cyanidin-3-O-glucoside being quantified in the highest concentrations (7.28 to 9.72 and 6.19 to 9.79 mg g−1, respectively) and being the predominant phenolic compounds in the blackberry-pomace samples. The odor profiles of blackberry pomace from different cultivars varied. The main volatile organic compounds found in all blackberry pomace were 1-Nonanol and cis-3-Hexen-1-ol, are associated with herbaceous and citrusy aromas. Raspberry (Rubus idaeus L.) is a crop with increasing commercial value due to its nutritional properties and demand in national and international markets. Throughout the growing process, there have been reports of the presence of galls on roots and stems, which reduce plant size and yields. The current study used phylogenetic reconstruction to identify bacteria associated with these symptoms in raspberry plantations in the Mexican states of Jalisco and Michoacán. A total of 69 representative strains selected according to their colony morphotype were identified based on partial sequencing of the 16S ribosomal gene. Forty-nine of them were tested for pathogenicity, including of which three strains induced gall symptoms in raspberry and tomato plants. A set of 28 strains identified within the Agrobacterium and Rhizobium genera were chosen for amplification of the atpD, glnA, gyrB, and rpoB genes. Strain CPO 2.419 was selected for whole genome sequencing, with a total length of 5,679,921 base pairs assembled into 46 contigs. This strain was identified as A. tumefaciens (GenBank JAVIYJ000000000.1) using phylogenetic reconstruction. According to the OrthoANIu and dDDH percentage values, the CPO 2.419 strain belongs to the A. tumefaciens complex, along with the closely related strains of A. tumefaciens LMG 232 (99.4, 89.1), A. tumefaciens CNPSo 675 (98.5, 84.8), and A. tumefaciens ATCC 4720T (98.1, 78), respectively. This study contributes to the understanding of the genomic Agrobacterium diversity in raspberry-producing areas in Mexico and highlights the relevance of genomics for accurate bacterial identification, with important implications for agriculture. Rubus imperialis Chum. Schl. (Rosaceae) have demonstrated some pharmacological activities, including gastroprotective action. However, genotoxic effects of R. imperialis extract was also reported. Since niga‐ichigoside F1 (NIF1) is a major compound of this plant species, and which has proven pharmacological properties, it is essential to investigate whether this compound is responsible for the observed toxicity. Therefore, the objective of this study was to analyze the effects of NIF1 on HepG2/C3A cells for possible cytogenotoxicity, cell cycle and apoptosis influence, and expression of genes linked to the DNA damage, cell cycle, cell death, and xenobiotic metabolism. The results showed no cytogenotoxic effects of NIF1 at concentrations between 0.1 and 20 μg/ml. Flow cytometry also showed no cell cycle or apoptosis disturbance. In the gene expression analysis, none of the seven genes investigated showed altered expression. The data indicate that NIF1 has no cytogenotoxic effects, and no interruption of the cell cycle, or induction of apoptosis, apparently not being responsible for the cytotoxic effects observed in the crude extract of R. imperialis. The adoption of the European Green Deal will limit acaricide use in high value crops like raspberry, to be replaced by biological control and other alternative strategies. More basic knowledge on mites in such crops is then necessary, like species, density, and their role as vectors of plant diseases. This study had four aims, focusing on raspberry leaves at northern altitude: (1) identify mite species; (2) study mite population densities; (3) investigate mite intra-plant distribution; (4) investigate co-occurrence of phytophagous mites, raspberry leaf blotch disorder and raspberry leaf blotch virus (RLBV). Four sites in south-eastern Norway were sampled five times. Floricanes from different parts of the sites were collected, taking one leaf from each of the upper, middle, and bottom zones of the cane. Mites were extracted with a washing technique and processed for species identification and RLBV detection. Mites and leaves were tested for RLBV by reverse transcription polymerase chain reaction (RT-PCR) with virus-specific primers. Phytophagous mites, Phyllocoptes gracilis, Tetranychus urticae, and Neotetranychus rubi, and predatory mites, Anystis baccarum and Typhlodromus (Typhlodromus) pyri were identified. All phytophagous mites in cultivated raspberry preferred the upper zone of floricanes, while in non-cultivated raspberry, they preferred the middle zone. The presence of phytophagous mites did not lead to raspberry leaf blotch disorder during this study. RLBV was detected in 1.3% of the sampled plants, none of them with leaf blotch symptoms, and in 4.3% of P. gracilis samples, and in some spider mite samples, implying that Tetranychids could also be vectors of RLBV. The genus Rubus encompasses over 1000 species, including raspberries and blackberries, known for their rich nutritional and health-promoting properties. This review aims to provide a comprehensive overview of the nutritional values, health benefits, and potential medical and pharmaceutical applications of Rubus species. The fruits, roots, shoots, and leaves of these plants are distinguished by their high content of polyphenols, vitamins, and minerals, which contribute to their potent anti-oxidant, anti-inflammatory, anti-neurodegenerative, and anti-cancer effects. The diverse phytochemical profiles of the Rubus species support their use in the prevention and treatment of chronic diseases such as cardiovascular diseases, diabetes, and certain cancers. Additionally, the Rubus species are valuable as pharmaceutical raw materials due to their bioactive compounds. Despite the focus on a few widely cultivated species, numerous wild and lesser-known Rubus species offer significant untapped potential as functional foods, nutraceuticals, and pharmaceuticals. Future research should explore the detailed mechanisms of their bioactivities, develop effective extraction and formulation techniques, and integrate these findings into public health strategies. Various reports revealed that chemical constituents from many species of Rubus exhibit diverse biological activities. In this study, a novel flavonoid with a 2-(phenanthren-9-yl)-4H-chromen-4-one structure (1), a 5-phenylthiophene-2-carbaldehyde derivative (5) first isolated from a natural source, together with five known compounds including three polyketides (2–4) and two sesquiterpenoids (6–7) were isolated from a traditional Chinese medicine Rubus rosifolius S.Vidal (Rosaceae). The structures of new compounds were elucidated by detailed spectroscopic analysis including NMR and X-ray single-crystal diffraction. The bioassays results indicated that, compound 1 displayed significant cytotoxicity against human colon cancer cell line HCT116 with IC50 value of 8.6 ± 1.9 μM, and compound 5 exhibited moderate cytotoxicity against human breast cancer cell line MDA-MB-435 with IC50 value of 24.1 ± 0.8 μM. Background and Aims Rubus ser. Glandulosi provides a unique model of geographical parthenogenesis on a homoploid (2 n  = 4 x) level. We aim to characterize evolutionary and phylogeographical patterns in this taxon and shed light on the geographical differentiation of apomicts and sexuals. Ultimately, we aim to evaluate the importance of phylogeography in the formation of geographical parthenogenesis. Methods Rubus ser. Glandulosi was sampled across its Eurasian range together with other co-occurring Rubus taxa (587 individuals in total). Double-digest restriction site-associated DNA sequencing (ddRADseq) and modelling of suitable climate were used for evolutionary inferences. Key Results Six ancestral species were identified that contributed to the contemporary gene pool of R. ser. Glandulosi. Sexuals were introgressed from Rubus dolichocarpus and Rubus moschus in West Asia and from Rubus ulmifolius agg. Rubus canescens and Rubus incanescens in Europe, whereas apomicts were characterized by alleles of Rubus subsect. Rubus. Gene flow between sexuals and apomicts was also detected, as was occasional hybridization with other taxa. Conclusions We hypothesize that sexuals survived the last glacial period in several large southern refugia, whereas apomicts were mostly restricted to southern France, whence they quickly recolonized Central and Western Europe. The secondary contact of sexuals and apomicts was probably the principal factor that established geographical parthenogenesis in R. ser. Glandulosi. Sexual populations are not impoverished in genetic diversity along their borderline with apomicts, and maladaptive population genetic processes probably did not shape the geographical patterns. A novel negative-sense single-stranded RNA virus showing genetic similarity to viruses of the genus Rubodvirus has been found in raspberry plants in the Czech Republic and has tentatively been named raspberry rubodvirus 1 (RaRV1). Phylogenetic analysis confirmed its clustering within the group, albeit distantly related to other members. A screening of 679 plant and 168 arthropod samples from the Czech Republic and Norway revealed RaRV1 in 10 raspberry shrubs, one batch of Aphis idaei, and one individual of Orius minutus. Furthermore, a distinct isolate of this virus was found, sharing 95% amino acid identity in both the full nucleoprotein and partial sequence of the RNA-dependent RNA polymerase gene sequences, meeting the species demarcation criteria. This discovery marks the first reported instance of a rubodvirus infecting raspberry plants. Although transmission experiments under experimental conditions were unsuccessful, positive detection of the virus in some insects suggests their potential role as vectors for the virus. In our study, using chromatographic techniques, we isolated three bioactive compounds, which were structurally elucidated as (E)-2-(3-(3,4-dimethoxyphenyl)acrylamido)-N-methylbenzamide (1), 4-Hydroxyquinoline-2-carboxylic acid (2), and (E)-2-Cyano-3-(4-hydroxyphenyl)acrylic acid (3), using spectroscopic methods. The anti-melanogenic, anti-inflammatory, antioxidant, and anti-aging properties were evaluated in vitro by measuring the activity of pharmacological targets including tyrosinase, melanin, NF-κB, hyaluronidase, elastase, collagenase, and Nrf2. Our results show that compound 1 is the most active with IC50 values of 14.19 μM (tyrosinase inhibition), 22.24 μM (melanin inhibition), 9.82–12.72 μM (NF-κB inhibition), 79.71 μM (hyaluronidase inhibition), 80.13 μM (elastase inhibition), 76.59 μM (collagenase inhibition), and 116–385 nM (Nrf2 activation) in the THP-1, HEK001, WS1, and HMCB cells. These findings underscore the promising profiles of the aqueous extract of R. urticifolius at safe cytotoxic concentrations. Additionally, we report, for the first time, the isolation and characterisation of these nitrogenous compounds in the R. urticifolius species. Finally, compound 1, isolated from R. urticifolius, is a promising candidate for the development of more effective and safer compounds for diseases related to skin pigmentation, protection against inflammation, and oxidative stress. BACKGROUND: Red raspberry (Rubus idaeus L.) is an important nectar source for honey production in some specific habitats as well as an important crop, so the definition of the features of this kind of honey is noteworthy. However, due to its rarity on the market, red raspberry honey is poorly characterized. The aim of this work was the phytochemical characterization of honey containing red raspberry from different geographical origins, through melissopalynological analyses concurrently with untargeted metabolomics achieved with different chromatographic techniques coupled to mass spectrometry: solid‐phase micro‐extraction/gas chromatography/mass spectrometry (SPME‐GC–MS) and high‐performance liquid chromatography/Orbitrap mass spectrometry (HPLC‐Orbitrap). RESULTS: Only 4 out of the 12 samples involved in the study contained raspberry pollen as dominant pollen, although these honeys did not group in the hierarchical cluster analysis nor in the classical multidimensional scaling analyses used for data evaluation. The first result was the detection of mislabelling in two samples, which contained raspberry pollen only as minor or important minor pollen. Of the 188 compounds identified by HPLC‐Orbitrap and of the 260 identified by SPME‐GC–MS, 87 and 31 compounds were present in all samples, respectively. The structurally related compounds nicotinaldehyde and nicotinamide, nicotinic acid and nicotinyl alcohol were present in 100% of the samples and correlated with R. idaeus pollen count (r > 0.60, Pearson's correlation analysis). CONCLUSION: This study reveals important aspects about the characterization of red raspberry honey and could give new insights on bee diet and preferences, since niacin compounds resulted interestingly to be related to the presence of red raspberry pollen. © 2024 Society of Chemical Industry. The stability of cell walls and their constituents, extractives, and water during the development and ripening stages of Rubus ulmifolius Schott fruits (blackberry fruit) was explored. The cell wall plays a crucial role in providing structural support and protection to the fruit, maintaining its shape and integrity. It acts as a barrier against mechanical stress, pathogens, and environmental factors. The ripening stages of blackberry fruit were previously classified in four stages according to the size and volatile components in each one. In the present work, we examine the water and sugar content, thermodynamic properties of the cell wall, and colour determination in each of the four ripening stages. The results indicate that the thermal stability of the fruit decreases during ripening, attributed to depolymerisation and changes in the physicochemical properties of the cell wall polysaccharides. Furthermore, the research reveals that the fruit's colour and sugar content undergo changes throughout its development and ripening process. These findings suggest that the stability of the cell walls in ripening fruits is influenced by their constituents and interactions, and that ripening is accompanied by alterations. WHO (World Health Organization) reports from recent years warn about the growing number of antibiotic-resistant bacterial strains. Therefore, there is an urgent need to constantly search for new substances effective in the fight against microorganisms. Plants are a rich source of chemical compounds with antibacterial properties. These compounds, classified as secondary metabolites, may act independently or support the action of currently used antibiotics. Due to the large number of metabolites isolated from the plant kingdom and new plant species being studied, there is a need to develop new strategies/techniques or modifications of currently applied methods that can be used to select plant extracts or chemical compounds isolated from them that enter into positive, synergistic interactions with currently used antibiotics. One such method is the dual-disk synergy test (DDST). It involves the diffusion of active compounds in the agar environment and influencing the growth of microorganisms grown on it. The method was used to assess the interaction of extracts from the fruit and shoots of some cultivated varieties of Rubus idaeus and Rubus occidentalis with selected antibiotics. The research was conducted on strains of bacteria pathogenic to humans, including Staphylococcus aureus, Corynebacterium diphtheriae, Escherichia coli, Pseudomonas aeruginosa, Helicobacter pylori, and Candida albicans, showing synergy, antagonism, or lack of interaction of the tested substances—plant extract and antibiotic. As a result, it was found that the diffusion method is useful in screening tests to assess the impact of antibiotic–herbal substance interactions on Gram-positive and Gram-negative microorganisms. Rubus ellipticus Smith. (Family Rosaceae), often known as the yellow Himalayan raspberry (Yellow Hissar), is one of the most widely used edible fruits in Indian folk medicinal systems. The current review aims to identify the gap between research and existing applications of this fruit to help scientists explore the current trends and opportunities for future development. Fruits of R. ellipticus are the source of several classes of compounds. Fruits of R. ellipticus are also rich in nutrients such as carbohydrates, vitamins, and minerals. It has been shown to have significant medical value in a variety of studies, including as an anti-diabetic, nephroprotective, anti-inflammatory, analgesic, antipyretic, antitumor, wound healing, antifertility, oviposition deterrent, antibacterial, and antioxidant. Fruits of R. ellipticus have been the subject of several in vitro and in vivo investigations, all of which have corroborated their wide range of biological activities and demonstrated their potential for the identification of new therapeutic candidates and the development of innovative herbal food supplements. Additional mechanism-based pharmacological evaluation and clinical research should provide an adequate scientific basis for the traditional usage of R. ellipticus fruits, which is currently not sufficiently supported by the available research on its active components and molecular mechanisms. Objective: The study was designed to test solvent extracts of Rubus suavissimus (RS) for compounds that regulate lipid metabolism and to measure their relative activity as quantitative markers of lipid metabolism-regulating activities based on the spectrum-effect relationship. Method: RS was extracted with 95% ethanol, the different extraction parts were successively extracted with petroleum ether, methylene chloride, ethyl acetate and n-butanol, respectively. The solution of different extraction parts was applied to the induced differentiated 3T3-L1 preadipocytes, and the oil red O staining and triglyceride release from the cells were determined to screen the best metabolism-regulating site. Analysis of different extraction sited by UPLC-Q-TOF-MS/MS technique to separate and identify the components. Based on specific assays, image intensity analysis, grey correlation and partial least squares regressions, the spectrum-effect relationship of the most abundant active components and their ability to regulate lipid metabolism were determined, and the quality markers were screened. Results: Ethyl acetate and n-butanol extraction site was the best metabolism-regulating site. Four components, including ellagic acid, centaurin-3-O-rutinoside, rubusoside and enantio-kauri-16-ene-19-carboxylic acid-13-O-β-D-glucoside were highly associated with metabolism-regulating effects. It could be used as quality markers for regulating lipid metabolism. Conclusion: Investigation of the quality markers of RS extracts based on the spectrum-effect relationship is of great importance for elucidating the pharmacodynamics, screening the core quality markers for therapeutic activity, and ensuring the safety and rational application of traditional medicines. Rubus includes a group of important plants with medicinal and culinary significance, as well as ornamental value. However, due to its status as one of the largest genera in the Rosaceae family and frequent occurrences of apomixis, hybridization, and polyploidy among its species, the morphological identification of this genus is highly challenging. The plastid genome serves as a valuable tool for studying the evolutionary relationships among plants. Therefore, based on the raw data of the whole genomes from six popular Rubus taxa, the complete Cp genomes were assembled, annotated, and subjected to comparative and phylogenetic analyses. In this research, six newly complete Cp genomes were reported, which all had a representative quadripartite formation, with a similar GC content (37.06%–37.26%), and their size ranged from 155,493 bp to 156,882 bp. They all encode 111 unique genes, containing 79 PCGs, 28 tRNA, and 4 rRNA. The analysis of gene structure of different groups showed that the sequence and content of genes were relatively conservative, and there was no gene rearrangement. Most of their PCGs had a high frequency codon usage bias and all genes were in purifying selection states. A nucleotide variable analysis revealed that the IR areas had less variation than the SC areas, and there was the greatest diversity in the SSC area. Eleven hypervariable areas were identified, containing rpl32-trnL, rpl32, rps16-trnQ, trnT-trnL, trnQ-psbK, trnK-rps16, and rps15-ycf1, which could be used as labels for genetic diversity and taxa identification. The phylogenetic trees of 72 Rosaceae plants were constructed based on ML and BI methods. The results strongly support the theory that the Rubus genus was a monophyletic group and sampled species could be arranged into seven subgenera. Overall, this study sheds its new light into the phylogeny of the Rubus genus, providing valuable insights for future studies of the Cp genomes from the expanded taxa of the Rosaceae family. This study examines the impact of ungulate browsing on the understory dynamics of the Cansiglio Forest, the only European beech high forest in the Southern Limestone Alps. Utilizing seven pairs of fenced versus unfenced forest stands established over two decades, we assessed how fencing affects understory structure and tree regeneration. Significant differences were found in cover, height and vascular species composition in both tree regeneration and shrub layers. Fenced areas showed a significantly higher number and greater average height of young trees, supporting the hypothesis that ungulate browsing negatively impacts tree regeneration. Despite previous studies attributing the lack of silver fir (Abies alba Mill.) regeneration primarily to red deer (Cervus elaphus Linnaeus, 1758) browsing, our findings did not show a significant association of this species with fenced conditions. While Rubus abundance was higher inside fenced areas, overall vascular species richness did not differ. Our results highlight the evident effects of ungulate browsing and suggest the need to consider other factors influencing forest regeneration and understory development. Raspberry (Rubus idaeus L.) is susceptible to aphid-borne viruses. We studied the incidence of four of them – black raspberry necrosis virus (BRNV), raspberry leaf mottle virus (RLMV), raspberry vein chlorosis virus (RVCV), and Rubus yellow net virus (RYNV) – in raspberry plants and aphids in and around Norwegian raspberry crops for three years (2019, 2021, and 2022). Most of the samples were from symptomatic plants. Applying RT-PCR, 274 leaf samples and 107 aphid samples were analyzed. All four viruses were found, but BRNV dominated: it was detected in 93% of the 178 leaf samples with virus and was the only virus that occurred more frequently as a single infection than in co-infections with the other viruses. The old cv. Veten had the highest virus incidence (97%) among the sampled plants, followed by uncultivated raspberry in the boundary vegetation (82%). All aphids identified were Amphorophora idaei and Aphis idaei. BRNV and/or RLMV was detected in 27% of the aphid samples. Notably, BRNV was detected in 30% of A. idaei samples, a species not known as a BRNV vector. In subsequent transmission experiments we found that although A. idaei can acquire BRNV within one hour, it did not transmit the virus to healthy raspberry plants. In contrast, Am. idaei, a known BRNV vector, was able to acquire the virus within one minute and transmit it within one hour of inoculation. Our study will improve the identification and management of BRNV.

Abstract There is a growing trend towards enhancing the post-harvest shelf life and maintaining the nutritional quality of horticultural products using eco-friendly methods. Raspberries are valued for their diverse array of phenolic compounds, which are key contributors to their health-promoting properties. However, raspberries are prone to a relatively short post-harvest lifespan. The present study aimed to investigate the effect of exogenous melatonin (MEL; 0, 0.001, 0.01, and 0.1 mM) on decay control and shelf-life extension. The results demonstrated that MEL treatment significantly reduced the fruit decay rate (P ≤ 0.01). Based on the findings, MEL treatment significantly increased titratable acidity (TA), total phenolics content (TPC), total flavonoid content (TFC), and total anthocyanin content (TAC). Furthermore, the MEL-treated samples showed increased levels of rutin and quercetin content, as well as antioxidant activity as measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reduction activity potential (FRAP). Additionally, the samples exhibited higher levels of phenylalanine ammonia-lyase (PAL) and catalase (CAT) enzymes compared to the control samples. Moreover, the levels of pH, total soluble solids (TSS), and IC50 were decreased in the MEL-treated samples (P ≤ 0.01). The highest amount of TA (0.619 g/100 ml juice), rutin (16.722 µg/ml juice) and quercetin (1.467 µg/ml juice), and PAL activity (225.696 nm/g FW/min) was observed at 0.001 mM treatment, while, the highest amount of TAC (227.235 mg Cy-g/100 ml juice) at a concentration of 0.01 mM and CAT (0.696 u/g FW) and TAL activities (9.553 nm/100 g FW) at a concentration of 0.1 mM were obtained. Considering the lack of significant differences in the effects of melatonin concentrations and the low dose of 0.001 mM, this concentration is recommended for further research. The hierarchical cluster analysis (HCA) and principal component analysis (PCA) divided the treatments into three groups based on their characteristics. Based on the Pearson correlation between TPC, TFC, TAC, and TAA, a positive correlation was observed with antioxidant (DPPH and FRAP) and enzyme (PAL and CAT) activities. The results of this study have identified melatonin as an eco-friendly compound that enhances the shelf life of raspberry fruits by improving phenolic compounds, as well as antioxidant and enzyme activities.

The diversity of Rubus in South America is much understudied and a taxonomic framework needs to be established as a basis for future revisionary and phylogenetic work. Our review identified 110 names based on South American specimens which were published since 1767. Each name was then classified according to its botanical description and type material. Additionally, where necessary, we suggest appropriate lecto-, neo-, or epitypes. A comprehensive list of synonyms is provided and representative herbarium specimens for each country are cited to tentatively document geographical range. In total, we accept 46 species of Rubus recorded across South America, propose 19 new synonyms, restore R. organensis, previously a synonym of R. brasiliensis, provide a replacement name for the latter, and include new country records of R. azuayensis, R. laegaardii and R. rusbyi. (1) Background: Endophytic bacteria represent an important component of plant wellness. They have been widely studied for their involvement in plant development and enhancement of stress tolerance. In this work, the endophytic communities of roots, stems, and leaves of blackberry (Rubus ulmifolius Schott) were studied in three different niches: natural, riverside, and human-impacted niches. (2) Results: The microbiome composition revealed that Sphingomonadaceae was the most abundant family in all samples, accounting for 9.4–45.8%. In contrast, other families seem to be linked to a specific tissue or niche. Families Microbacteriaceae and Hymenobacteraceae increased their presence in stem and leaf samples, while Burkholderiaceae abundance was important in riverside samples. Alpha and beta diversity analyses showed that root samples were the most diverse, and they gathered together in the same cluster, apart from the rest of the samples. (3) Conclusions: The analysis of the microbiome of R. ulmifolius plants revealed that the composition was essentially the same in different niches; the differences were primarily influenced by plant tissue factors with a core genome dominated by Sphingomonadaceae. Additionally, it was observed that R. ulmifolius can select its own microbiome, and this remains constant in all tissues evaluated regardless the niche of sampling. [ABSTRACT FROM AUTHOR]
