AIM: To evaluate the long-term response to the fixed combination of dorzolamide/timolol in patients with primary open angle glaucoma (POAG) and the addition of other intraocular pressure (IOP) lowering medications such as prostaglandin analogs and brimonidine.METHODS: A retrospective, non-randomized, and descriptive clinical study was performed with 182 eyes diagnosed with POAG. Patients were divided into three groups:a group with fixed combination of dorzolamide/timolol only, a second group with prostaglandin analogs plus fixed combination of dorzolamide/timolol, and a third group with the addition of brimonidine to the same fixed combination. IOP data were gathered retrospectively and the differences between groups were calculated.RESULTS: IOP was reduced satisfactorily in all three groups; however, a progressive IOP reduction was noted in the group with the fixed combination plus prostaglandin analogs. In this group, a progressive, significant and more homogeneous response of the reduction was noted in comparison with the other groups.CONCLUSION: IOP reduction was efficacious in all three groups. The addition of prostaglandin analogs showed progressive IOP reduction, progressive response and absence of long-term drift. Brimonidine did not show a significant additive effect.

Protein biosynthesis in trypanosomidae. II. The metabolic fate of dl -leucine-1-C 14 in Trypanosoma cruzi . Experimental Parasitology 21 , 154–159. Trypanosoma cruzi is able to metabolize dl -leucine-1-C 14 incorporating its radioactivity into various metabolic fractions. Utilization of leucine is higher in the presence of glucose than in its absence. Both substrates are metabolized individually or together as shown by the respiratory CO 2 production. The path of leucine catabolism seems to be similar to that which has already been described for mammals. Leucine seems also to enter the Krebs cycle and possibly other alternative pathways since glycolic acid is produced with other nonvolatile acids and, thus, indicates the possibility of the glyoxylate shunt. Antibiotics do not affect the metabolism of leucine, with the exception of that which is concerned with its incorporation into proteins.

A 48-fold purification of phosphohexose isomerase from T. spiralis is reported. The enzyme has an optimal pH of 8.0 and is thermostable with an optimal temperature of 55 ° C. It apparently does not depend on protein-SH groups for activity nor does it require any metal for activation, although Co ++ and Mn ++ are inhibitory. The enzyme is inhibited by GNH2-6-P and 6-PG. The ratio of G-6-P to F-6-P at equilibrium was found to be 65:35 at 38 ° C. 2 ΔH o is −8,198 cal/mole with a ΔF o of −385 cal/mole and a change in entropy of −25.1 units/mole.

1. 1. Conditions for the in vivo incorporation of dl -leucine-1-C14 into the proteins of Trypanosoma cruzi are reported in this paper. Best results were obtained with KC1 0·154 M as suspension medium as compared to 0·25 M sucrose. 2. 2. Optimal pH in the absence of glucose was 7·6, while in its presence was 8·0. 3. 3. The addition of glucose increases the incorporation of the isotope into total proteins, while various metabolites of the Krebs cycle have an opposite effect. 4. 4. Cell fractionation studies indicate a rapid labeling of microsomal protein. Chromatography and radioautography show that isoleucine and valine are also labeled in the protein hydrolysate. 5. 5. Our results suggest that an active mechanism, similar to those reported for higher organisms, might be involved in protein biosynthesis in T. cruzi.
