The Dolichol kinase (DOLK) gene encodes the polytopic DOLK protein associated with the endoplasmic reticulum (ER) N-glycosylation pathway catalyzing the final step in the biosynthesis of dolichol phosphate. Dolichol phosphate is an oligosaccharide carrier required for N-glycosylation of DOLK protein, with its deficiency leading to a severe hypo glycosylation phenotype in humans which can cause congenital disorders of glycosylation and death in early infancy. The aim of the present study is to identify the phylogenetic relationship between human and ortholog species based on their conserved sequences in DOLK gene. Sequence alignment of DOLK was carried out in this study and the evolutionarily conserved regulatory sequences were identified using bioinformatics. Promoter sequence of human DOLK was compared with orthologous sequences from different organisms. Conserved non-coding sequences (CNS) and motifs in promoter regions were found by analyzing upstream promoter sequences of Homo sapiens DOLK and its orthologous genes in other organisms. Conserved sequences were predicted in the promoter regions in CNS1 and CNS2. Conserved protein sequences were also identified by alignment of the orthologous sequences. Organisms with similar gene sequences are assumed to be closely related and the ER N-glycosylation pathway is conserved in them.

Water is used for various religious purposes, including ablution for billions of Muslims around the world. Water savings in ablution can make a positive contribution towards achieving the water-related sustainable development goals (SDGs). A survey-based study was conducted at St Mary's Mosque in Sydney, Australia, to understand the water usage during ablution and how water can be saved by Muslims in mosques. It has been found that the participants of the survey use 4 to 8 L of water to perform one ablution. About 82% of the participants are careful about saving water in ablution. It has also been found that about 70% of the participants believe that wastage of water during ablution is 'haram' (unlawful). Some 95% of the participants believe that religious motivation can play a significant role in saving water during ablution, and Imams can make a notable positive difference by encouraging their followers to save water during ablution. It has also been found that 64% of the participants are willing to use harvested rainwater for ablution. The findings of this study will help Muslims in Australia and around the globe to save water and contribute to the circular economy, as saving potable water (by reducing water use during ablution and using harvested rainwater at the source of water collection) saves energy and encourages responsible consumption. Event-based models focus on modelling of peak runoff from rainfall data. Conceptual models indicate simplified models that provide reasonably accurate answers despite their crude nature. Rainfall-runoff models are used to transform a rainfall event into a runoff event. This paper focuses on reviewing computational simulation of rainfall-runoff processes over a catchment. Lumped conceptual, event-based rainfall-runoff models have remained the dominant practice for design flood estimation in Australia for many years due to their simplicity, flexibility, and accuracy under certain conditions. Attempts to establish regionalization methods for prediction of design flood hydrographs in ungauged catchments have seen little success. Therefore, as well as reviewing key rainfall-runoff model components for design flood estimation with a special focus on event-based conceptual models, this paper covers the aspects of regionalization to promote their applications to ungauged catchments.
