Chromium is a toxic heavy metal, which primarily exists in two inorganic forms, Cr (VI) and Cr (III). Highly soluble hexavalent chromium is carcinogenic due to its oxidizing nature. It is well established that the intestinal bacteria including Lactobacilli have regulatory effect on intestinal homeostasis and a breakdown in the relationship between intestinal cells and bacteria results in the manifestation of gastrointestinal (GI) disorders. In this study Cr (VI) resistance was developed in Lactobacillus strains and the reduction of Cr (VI) was evaluated. All resistant strains showed similarities with their respective normal strains and did not acquire resistance to various antibiotics. A complete bacterial reduction of 32ppm Cr (VI) was observed within 6 to 8 hours. The presence of chromate reducing enzyme have also been established following the partial purification (2 to 5 fold) and characterization of chromate reductase in Lactobacillus strains. The chromate reductase of our strains showed optimum activity at pH 6.0 and 30°C. To our knowledge; these strains are fast in Cr (VI) reduction than any other known bacteria. The results suggest that chromate- resistant Lactobacillus strains would be useful for chromium detoxification from GI-tract as well as for bioremediation of hexavalent chromium from contaminated environment.

The intestinal bacteria are known to play a significant role in intestinal homoeostasis and the mucosal immune system. In vitro interactions of Ampicillin (0.5-2.0 microg mL(-1)), Amphotericin-B (25-200 microg mL(-1)) and Ciprofloxacin (50-500 ng mL(-1)) with Escherichia coli, Pseudomonas sp. (Gram-negative), Lactobacillus sp., Staphylococcus sp. (Gram-positive), total mixed population of gut bacteria and intestinal epithelial cells were studied. In vitro exposure of Ciprofloxacin showed significant dose-dependent inhibition throughout the growth phase in bacteria. Similar patterns of concentration-dependent changes in membrane transport enzymes and structural constituents, dehydrogenase activity associated with respiratory and energy-producing processes and esterase activity test linked to the general heterotrophic activity of the cell were observed in both bacteria and epithelial cells. Distillery effluent or spent wash discharged as waste water contains various toxic chemicals that can contaminate water and soil and may affect the common crops if used for agricultural irrigation. Toxic nature of distillery effluent is due to the presence of high amounts of organic and inorganic chemical loads and its high-acidic pH. Experimental effects of untreated (Raw) distillery effluent, discharged from a distillery unit (based on fermentation of alcohol from sugarcane molasses), and the post-treatment effluent from the outlet of conventional anaerobic treatment plant (Treated effluent) of the distillery unit were studied in mung bean (Vigna radiata, L.R. Wilczek). Mung bean is a commonly used legume crop in India and its neighboring countries. Mung bean seeds were presoaked for 6h and 30 h, respectively, in different concentrations (5-20%, v/v) of each effluent and germination, growth characters, and seedling membrane enzymes and constituents were investigated. Results revealed that the leaching of carbohydrates and proteins (solute efflux) were much higher in case of untreated effluent and were also dependent to the presoaking time. Other germination characters including percentage of germination, speed of germination index, vigor index and length of root and embryonic axis revealed significant concentration-dependent decline in untreated effluent. Evaluation of seedlings membrane transport enzymes and structural constituents (hexose, sialic acid and phospholipids) following 6 h presoaking of seeds revealed concentration-dependent decline, which were much less in treated effluent as compared to the untreated effluent. Treated effluent up to 10% (v/v) concentration reflected low-observed adverse effect levels.

Toxicants including heavy metals reaching the intestine following ingestion through food and water primarily interact with an ecosystem of eukaryotic and prokaryotic cells. Gut bacteria having a dynamic interrelationship with intestinal epithelial cells are known to play important and specific metabolic, trophic, and protective functions. The present study was undertaken to compare the effects of hexavalent chromium on rat intestinal epithelial cells and the resident gut bacteria following in vitro and in vivo exposures. The survival rate and viability pattern of two types of cells were comparable. Under in vitro conditions, the gut bacteria were quick to reduce Cr (VI) in early time periods, while, at 30 h time, both types of cells showed similar capacity for the reduction of Cr (VI). Chromium intoxication (10 ppm of Cr (VI) in drinking water for 10 weeks) caused significant decrease in membrane alkaline phosphatase and Ca(2 +)-Mg(2 +)-ATPase activities of intestinal epithelial cells as well as of three gut bacteria viz. Escherichia coli, Pseudomonas sp, and Lactobacillus sp. Major structural membrane constituents like carbohydrates and phospholipids also showed significant decline in both types of cells. These findings indicate that 10 ppm and higher Cr concentrations may cause toxic insult, resulting in impaired intestinal functional efficacy. It also implies that the gut bacteria can be used at least for preliminary screening of heavy metals gastrointestinal toxicity.

The present study was undertaken to investigate the effects of dengue virus (DV) infection in male mice given drinking water containing 250 ppm Cr (VI) and the normal control male mice given plain water to drink. On the basis of intake of water in 24 h, the average dose of Cr (VI) in each mouse was 14.8 mg/kg. After 3, 6, and 9 weeks of drinking Cr (VI), a set of five mice from each group were inoculated intracerebrally (ic) with a 1000 x LD(50) (100 times the lethal dose that kills 50% mice) dose of DV, and the effects on the spleen were studied at the fourth and eightth day postinoculation. It was observed that Cr (VI) drinking and DV infection led to reduction in the weight of the spleen, but the peak reduction was seen in Cr (VI)-fed mice infected with DV, being 30, 34, and 61% at 3, 6, and 9 weeks respectively. A similar response was seen with respect to the cytotoxic activity of spleen homogenates, phagocytic activity of macrophages, and the mitogenic response of spleen cells to concanavalin A from different groups of animals, being most marked (58 to 60%) at the ninth week of Cr (VI) drinking. This shows a summation of adverse effects of DV infection in mice preexposed to Cr (VI).

The present study was undertaken to investigate the spectrum of bacteria present in the River Gomti water before and after chlorination for drinking purposes. We observed that the strains of Pseudomonas aeruginosa that survived chlorination on three out of seven occasions were resistant to almost all the antibiotics tested. The chlorine-resistant bacteria had mucoid colonies and grew better at 24 degrees C. All attempts to isolate the plasmid responsible for chlorine resistance were unsuccessful. Laboratory experiments using different strains of the P. aeruginosa in distilled water showed that only the resistant strain survived chlorine treatment at a dose of < or =500 microg/L. Similar results were obtained when water collected from seven different sites on the River Gomti was treated with graded doses of chlorine. At the higher dose of chlorine, all the bacteria died in 30 min, whereas with lower doses all the bacteria survived. The present study underscores the importance of measuring water chlorine concentrations to assure they are sufficiently high to remove pathogenic bacteria from drinking water. To our knowledge, this is the first report in the literature of the selection of multidrug-resistant bacteria by suboptimal chlorine treatment of water.

Although widely distributed in eukaryotic cells glycoproteins appear to be rare in prokaryotic organisms. The prevalence of the misconception that bacteria do not glycosylate their proteins has been a subject matter of discussion for a long time. Glycoconjugates that are linked to proteins or peptides, generated by the ribosomal translational mechanism have been reported only in the last two to three decades in a few prokaryotic organisms. Most studied prokaryotic glycoproteins are the S-layer glycoproteins of Archeabacteria. Apart from these, membrane-associated, surface-associated, secreted glycoproteins and exoenzymes glycoproteins are also well documented in both, Archea and Eubacteria. From the recent literature, it is now clear that prokaryotes are capable of glycosylating proteins. In general, prokaryotes are deprived of the cellular organelles required for glycosylation. In prokaryotes many different glycoprotein structures have been observed that display much more variation than that observed in eukaryotes. Besides following similar mechanisms in the process of glycosylation, prokaryotes have also been shown to use mechanisms that are different from those found in eukaryotes. The knowledge pertaining to the functional aspects of prokaryotic glycoproteins is rather scarce. This review summarizes developments and understanding relating to characteristics, synthesis, and functions of prokaryotic glycoproteins. An extensive summary of glycosylation that has been reported to occur in bacteria has also been tabulated. Various possible applications of these diverse biomolecules in biotechnology, vaccine development, pharmaceutics and diagnostics are also touched upon.

A novel glycoprotein (Gp45) has been isolated and purified from Escherichia coli. To our knowledge, Gp45 is the third glycoprotein isolated from E. coli membrane and it is the second in the non-pathogenic strain of the organism. For the isolation of Gp45, cell extract or membrane fraction was treated with sodium deoxycholate for 4 h and precipitated with trichloroacetic acid (TCA). The supernatant fraction of TCA containing the Gp45 was further purified on DEAE-Sephadex A-25. SDS-PAGE showed a single band at 45 kDa position that stained with periodic-Schiff reagent. It contained 60% carbohydrate and 40% protein content. The monosaccharide composition also substantiated the characteristics of the glycoprotein. The E. coli grown in presence of (14)C-glucosamine further confirmed the localization and biosynthesis of this glycoprotein on the membrane during the growth phase. Bacitracin, a general inhibitor of the glycosylation, inhibited its biosynthesis.

Chromium is a naturally occurring heavy metal found commonly in the environment in trivalent, Cr(III), and hexavalent, Cr(VI), forms. Cr(VI) compounds have been declared as a potent occupational carcinogen among workers in chrome plating, stainless steel, and pigment industries. The reduction of Cr(VI) to Cr(III) results in the formation of reactive intermediates that together with oxidative stress oxidative tissue damage and a cascade of cellular events including modulation of apoptosis regulatory gene p53, contribute to the cytotoxicity, genotoxicity and carcinogenicity of Cr(VI)-containing compounds. On the other hand, chromium is an essential nutrient required to promote the action of insulin in body tissues so that the body can use sugars, proteins and fats. Chromium is of significant importance in altering the immune response by immunostimulatory or immunosuppressive processes as shown by its effects on T and B lymphocytes, macrophages, cytokine production and the immune response that may induce hypersensitivity reactions. This review gives an overview of the effects of chromium on the immune system of the body.

The cessation of eating under normal physiological conditions is a voluntary act induced by a conscious sensation known as satiety. In pathological situations there is loss of appetite or anorexia. We have been able to isolate and characterize a proteoglycan from membranes of mono- and dicotyledonous plant sprouts which reduced the food intake significantly when injected into murine model systems without any rebound. Due to its membrane origin, it has been termed ‘satiomem’. The plant proteoglycan (50 kDa) consisted of 70–85% carbohydrates and 15–30% proteins.

Inadvertent intake of inorganic arsenic and chromium through drinking water and food causing their toxic insults is a major health problem. Intestinal bacteria including Lactobacilli play important regulatory roles on intestinal homeostasis, and their loss is known to cause gastrointestinal (GI) disorders. Probiotic Lactobacilli resistance to arsenite and chromium-VI could be an importantfactorfor the perspective attenuation of Gl-disorders caused by these toxic metals/metalloid. In the present study resistance of arsenite (up to 32 ppm), Cr-VI (up to 64 ppm), and arsenite plus Cr-VI (32 ppm each) were developed under in vitro condition following chronological chronic exposures in Lactobacilli strains. Comparative study of biochemical parameters such as membrane transport enzymes and structural constituents; dehydrogenase and esterase activity tests, which are respective indicators for respiratory and energy producing processes, and the general heterotrophic activity of cells, of resistant strains showed similarities with their respective normal parent strains. The resistant strains were also found to be sensitive to antibiotics. Findings indicate that these resistant probiotic Lactobacilli would be useful in the prophylactic interventions of arsenic and chromium GI-toxicity.

Hunger and satiety are complex interplay of several factors in human and animal species. Reduced food intake has also been observed under various pathological conditions. Earlier, we have been able to isolate an endogenous glycoprotein from erythrocyte membranes, which causes anorexia in rats. In the present study, a similar anorexigenic proteoglycan from Mung bean sprout membranes has been isolated and purified. The proteoglycan (50 kDa) consisted of 70–85% carbohydrate with galactose, glucose galactosamine and mannose as the main sugars. Protein part on analysis showed higher glutamic acid and serine content. This proteoglycan reduces food intake when injected in rats deprived of food for 96 hr as well as normally fed rats, mice and rabbits without any rebound. The TCA-soluble proteoglycan from different plant sources have also been compared for their anorexigenic activity. The similarities observed among plant and animal cell membrane proteoglycans with satietins isolated from human blood plasma could be due to membrane origin of satietins. Aspirin, a commonly used therapeutic non-steroidal anti-inflammatory drug (NSAID) is known to cause gastric mucosal damage. Intestinal bacteria having a regulatory effect on intestinal homeostasis play significant role in NSAID-induced intestinal injury. Bacteria and specific cell lines are considered to be suitable for toxicity screening and testing of chemicals. Therefore, to evaluate and compare in vitro toxicity, cultures of rat intestinal epithelial cells (IEC), isolated bacteria and IEC-6 cell line were assessed for viability, morphometric analysis, membrane transport enzymes and structural constituents for membrane damage, dehydrogenase activity test for respiratory and energy producing processes and esterase activity test for intra- and extra-cellular degradation, following the post exposure to aspirin (0—50 µg mL- 1). Similar pattern of dose-dependent changes in these parameters were observed in three types of cells. Similar in situ effects on IEC validated the in vitro findings. These findings indicate that higher aspirin concentrations may alter cellular functions of IEC and gut bacteria. Furthermore, results suggest that gut bacteria and IEC-6 cell line can be used for the initial screening of gastrointestinal cellular toxicity caused by NSAIDs.

The major nonoccupational source of chromium (Cr) for humans is through ingestion with food and water, but its effect on the gut microflora has not been studied. The present study was, therefore, undertaken to investigate the effects of chronic ingestion of potassium dichromate (chromium VI) on the resident gut bacteria of male Wistar rats. A group of rats was kept on drinking water containing 10 ppm chromium VI (Cr [VI]) (called Cr-stressed animals) and the other group was given plain water. After 10 weeks, Lactobacillus, Pseudomonas sp., and Escherichia coli were isolated from the cecum of the rats and various studies were performed. The most significant findings of the present study were the stimulation of growth of facultative gut bacteria from the Cr-stressed rats and the significant increase of growth even in the presence of lower concentrations of Cr. Furthermore, the capacity to reduce Cr (VI) was significantly decreased along with the increased tolerance of the bacteria to Cr (higher minimum inhibitory concentration [MIC] values), which was associated with the development of antibiotic resistance. The effects were most marked with the Pseudomonas sp. and least with the E. coli. The antibiotic resistance developed with the Lactobacillus may be a blessing in disguise, because the bacteria may continue to provide benefits even in patients given antibiotic therapy. The gut bacteria thus provide the first line of defense to the body by converting toxic Cr (VI) to a less toxic Cr (III) and may act as a prebiotic.

The combined effects on the intestinal cells of guinea pigs following feeding them with lathyrus and manganese (Mn) for 90 days were studied in this investigation. Guinea pigs given Mn (4 ppm of their diets) for 90 days showed no change in either intestinal bioconstituents or marker enzymes, with the exception of gamma-glutamyl transpeptidase (GGT) and quinone reductase (QR). Exposure to a diet of 80% lathyrus only resulted in significant (p. 05) inhibition of intestinal alkaline phosphatase (ALP), sucrase, GGT, QR, and glutathione-s-transferase (GST) along with significant (p. 05) depletion of total hexose and phospholipids. Animals given lathyrus and Mn showed a significant (p. 05) decrease in intestinal ALP, Ca +2 Mg +2 -ATPase, sucrase, GGT, GST, and QR along with significant (p. 05) depletion in total hexose and phospholipids and concomitant enhancement in cholesterol when compared to controls. The data clearly indicate that combined treatment with lathyrus and Mn potentiates intestinal toxicity more than does Mn or lathyrus alone.

In order to use facultative gut bacteria as an alternate to animals for the initial gastrointestinal toxicity screening of heavy metals, a comparative study on rat intestinal epithelial cells and resident gut bacteria was undertaken.in vitro growth rate of four gut bacteria, dehydrogenase (DHA) and esterase (EA) activity test, intestinal epithelial and bacterial cell membrane enzymes and in situ effect of arsenite were analysed.Growth profile of mixed resident population of gut bacteria and pure isolates of Escherichia coli, Pseudomonas sp., Lactobacillus sp., and Staphylococcus sp. revealed an arsenite (2-20 ppm) concentration-dependent inhibition. The viability pattern of epithelial cells also showed similar changes. DHA and EA tests revealed significant inhibition (40%-72%) with arsenite exposure of 5 and 10 ppm in isolated gut bacteria and epithelial cells. Decrease in membrane alkaline phosphatase and Ca2+ -Mg2+ -ATPase activities was in the range of 33%-55% in four bacteria at the arsenite exposure of 10 ppm, whereas it was 60%-65% in intestinal epithelial villus cells. in situ incubation of arsenite using intestinal loops also showed more or less similar changes in membrane enzymes of resident gut bacterial population and epithelial cells.The results indicate that facultative gut bacteria can be used as suitable in vitro model for the preliminary screening of arsenical gastrointestinal cytotoxic effects.

Arsenic is a naturally occurring metalloid and the drinking water contamination by inorganic arsenic remains a major public health problem. The trivalent arsenic (arsenite) is more toxic than the pentavalent form (arsenate), and is known to cause gastrointestinal toxicity. Specific immortal cell lines are considered to be suitable for toxicity screening and testing of chemicals as they are easy to handle and possess most of the biochemical pathways present in the corresponding cells present in vivo. The present study was designed to evaluate and compare the in vitro toxicity of arsenite on rat intestinal epithelial cell line (IEC-6) and primary cultures of rat intestinal epithelial cells (IEC). To evaluate in vitro toxicity, cultures of IEC and IEC-6 cells were assessed for viability, morphometric analysis, membrane transport enzymes and structural constituents for membrane damage, dehydrogenase activity test for respiratory and energy producing processes and esterase activity test for intra and extra cellular degradation, following the post exposures to arsenite (0-20 ppm). Significantly similar concentration-dependent changes in these toxicity-screening parameters in IEC and IEC-6 were observed. Highest tested concentration of arsenite (20 ppm) was found to be detrimental in both IEC and IEC-6. Furthermore, to evaluate arsenite toxicity in epithelial cells of rat intestine, intestinal loops were filled with arsenite solutions and incubated for 30 min in situ. In situ studies also showed a significant arsenite concentration-dependent decline in epithelial cell membrane transport enzyme activities and total hexose and sialic acid contents. Concomitant release of membrane enzymes, hexose and sialic acid in the intestinal luminal fluid following higher arsenite exposures further indicated partial membrane damage. Similar morphological changes in IEC and IEC-6 were also evident. These findings also suggest that IEC-6 cell lines are suitable for initial screening of gastrointestinal cellular toxicity caused by arsenite.

The cells of the immune system form a strong line of defence against foreign substances. The present study was undertaken to investigate the capacity of different cells of Wistar rats to reduce potentially carcinogenic hexavalent chromium (Cr-VI) into less toxic trivalent chromium in vitro. 5 x 10(6) cells were incubated with 10 or 25 microg ml(-1) of Cr (VI) in the form of K2Cr2O7 at 37 degrees C in the presence of 5% CO2 in air. At various time periods the remaining amount of Cr (VI) was measured and the percentage of Cr (VI) reduced was calculated. Among the single cell suspensions from the splenic cells a peak reduction of 55% was observed with the total spleen cells, 40% with the B-lymphocyte-enriched subpopulation, 10% with T-lymphocytes and 24% with the macrophages. The reduction by splenic and peritoneal macrophages was similar. Total thymocytes reduced 54% of the Cr (VI). Since the most common route of entry of chromium is through drinking water and food, intestinal cells were also investigated. Among the intestinal cells the maximum reduction of 100% (of 10 microg ml(-1)) was observed with the upper villus cells and 72% with the middle villus cells while reduction was the least (4%) with the crypt cells. The reduction in the intestinal loop in situ was 100%. The time taken by each cell type for the peak reduction to Cr (VI) was markedly different. The findings thus show that the capacity of different cells in the body differs vastly in their capacity and time taken to reduce hexavalent chromium. The most efficient handling of Cr (VI) by the intestine, due to the presence of a variety of cells and bacteria, protects the body from its adverse effects.

The growth rate of Escherichia coli was stimulated when cells were in media containing lead up to a concentration of 300 ppm. Higher concentrations inhibited growth. Metal analysis revealed that in the presence of lead E. coli concentrates 22.8 mg of lead per gram (dry weight) of cells. Analysis of cellular subfractions indicated that membrane fraction concentrated over 95% of the lead taken up by cells, of which a major portion was found to be associated with membrane lipids. Alterations in alkaline phosphatase, Ca2+–Mg2+ -ATPase activities and the carbohydrate and phospholipid contents in membrane fractions were also observed when cells were grown in the presence of lead. A time- and concentration-dependent increase in the release of carbohydrates by the cells was also evident. The results suggest that besides thriving in higher lead surroundings, E. coli possess a marked ability to concentrate substantial amount of inorganic lead.

Vanadium is an important trace metal widely distributed in environment. Interaction of vanadate with skeletal muscle sarcolemma and basement membrane has been focussed. Scatchard analysis indicated the presence of more than one binding site for vanadate. Vanadate inhibits sarcolemmal and intestinal brush border membrane enzymes in a non-competitive manner. Membrane-vanadium interaction may lead to several structural and functional changes. The binding of vanadium to basement membrane may have some protective role.

Micromolar and millimolar Ca(2+)-requiring neutral protease (calpain I and calpain II) along with their endogenous inhibitor calpastatin were isolated and partially purified from the same preparation of rat intestinal epithelial cells. Calpain I and II were partially purified by 1300 and 900-fold with 57 and 53 per cent yield, respectively. The optimum assay conditions revealed pH 7.5, 20 min incubation at 25 degrees C and 0.24% casein substrate for both calpains. The optimum calcium concentration obtained for calpain I and II were 25 microM and 4 mM, respectively. Distribution of rat intestinal epithelial cells calpain I and II along with calpastatin during cell differentiation stages in weanling to senescence age were studied. Calpain I in weanling rats was in an increasing order from villus to crypt regions. Adult rats indicated well expressed consistent calpain I throughout the differentiation stages. Whereas, significant lowering towards crypt region cells were evident in old rats. Calpain II in weanling and adult rats was found to be consistent throughout the differentiation stages. Old animals revealed an increasing trend from villus to crypt region with insignificant activity present in upper villus cells. Concomitantly, different concentrations of calpastatin were observed throughout the differentiation stages in all the age groups. Moreover, the levels of calpains exceeded that of calpastatin in most of the epithelial cell populations during developmental stages. In addition to casein, intestinal epithelial cell membranes were found to be equally good substrates for calpains. Proteolytic susceptibility of weanling, adult and old rat membrane proteins varied significantly all along the ageing process in rats. Simultaneous age-dependent calpastatin response were also evident. Taken together the results obtained provided strong evidence that calpain plays significant role in rat intestinal cell differentiation and ageing process with calpastatin as its specific regulatory protein.

In rats injected intraperitoneally with actinomycin D (2 mg/kg body weight) 12 h earlier, the yield of the 30S ribonucleoprotein particles isolated from liver nuclei by extraction with 0.1 M NaCl at pH 8.0 decreased by 60 per cent. The protein-to-RNA ratio of these particles increased to 32:1 from the ratio 4.4:1 found in the same particles isolated from the nuclei of liver of control rats. The particles isolated from the liver nuclei of rats injected with actinomycin D were depleted of all charge isomers of the two most prominent polypeptides (33,000 and 39,000 daltons) present in the particles of liver of control animals. The most abundant protein in these particles was a 43,000 dalton polypeptide. This polypeptide is the least prominent of the 3 major polypeptides present in the control particles. The same charge isomers of the 43,000 dalton polypeptide were present in the nuclear ribonucleoprotein particles isolated from the liver of control animals and from the liver of animals treated with actinomycin D 12 h earlier. In control animals the nuclear ribonucleoprotein monoparticles isolated from kidney contained 3 major polypeptides of the same molecular weight with the same distribution of their charge isomers as were present in the particles isolated from liver nuclei. The injection of actinomycin D 12 h earlier was without any effect on the protein composition of the 30S nuclear ribonucleoprotein particles of rat kidney.

Samples of dust extracted from the lungs and lymph nodes of certain domestic animals from West Bengal, Orissa, Bihar provinces of India were quantitatively analysed using photon excited energy dispersive X-ray fluorescence technique. Thin specimens were prepared for analysis to minimize matrix enhancement and absorption effects. Amongst the various elements analysed, Hg, Fe, Cu, Zn and Pb were found to be present in appreciable amounts. An important finding was the presence of a very high concentration of Hg in extracted dust samples from West Bengal. The significance of the various results obtained in this investigation is discussed.

Summary Ribonucleoprotein particles sedimenting with an approximate sedimentation coefficient of 30S were prepared from rat liver nuclei. The polypeptides of the particles were separated by two-dimensional gel electrophoresis into three major molecular weight species with five charge isomers each ranging in apparent pI from 6.6 to 8.7. In the ethanol administered rats, the yield of ribonucleoprotein particles decreased by 15% whereas, in case of 18 hr regenerating liver, the decrease in the yield was 65% of the control. The protein-to-RNA ratio of 18 hr regenerating liver increased to 30:1 from the normal liver particle ratio of 4.4:1. The 18 hr regenerating liver particles were depleted of charge isomers of the prominent polypeptides. A normal pattern was retained after 30 hr of regeneration. Ribonucleoprotein particles isolated from the nuclei of alcohol abused livers had four major proteins composed of charge isomers with the apparent pI ranging from 6.4 to 7.5.

The tissue distribution, elimination and covalent binding of [1-14C]-acrylonitrile (VCN) have been investigated in the rat. Rats, given an oral dose of 46.5 mg/kg (0.5 LD50) VCN, excreted 40% of the 14C in urine, 2% in feces, 9% in expired air as 14CO2, 0.5% as H14CN and 4.8% as unchanged VCN in 24 h. Bile flow increased 3 times after the administration of VCN and over a period of 6 h, 27% of the 14C was recovered in bile. The red blood cells retained significant amounts of radioactivity for more than 10 days after treatment, whereas the 14C activity declined sharply in plasma. Initially, the highest levels of radioactivity were found in the stomach and stomach content followed by the intestine. In liver, kidney, brain, spleen, adrenal, lung and heart tissues the radioactivity of the acid soluble fractions declined while covalent binding to macromolecules remained unchanged. In subcellular fractions of liver, kidney, spleen, brain, lung, and heart, 20-40% of the total radioactivity was bound to nuclear, mitochondrial and microsomal fractions whereas in cytosol only 6-14% was bound over a period of 6 h.

Satiety means an internal state that leads to termination of eating. We have isolated an anorexigenic glycoprotein (M.W. 50,000 dalton) from human and rat erythrocyte membrane and from rat liver plasma membrane. The substance isolated from all these membrane sources has almost same onset and offset anorectic effect in rats deprived of food for 96 h as well as in normally fed rats without any rebound. Similar properties of membrane anorectic substance and plasma satietin indicated that it has membrane origin. The results also suggest that the loss of appetite in chronic diseases involving damage or turn-over of cell membranes could be due to release of a glycopeptide from cellular membranes into the circulation.

Preliminary studies related to immunotoxicologic effects of 2,5-hexanedione, the final major metabolite of n -hexane/MnBk, were carried out in rats following single or repeated exposures. Female albino rats were given either single or seven consecutive oral doses of 0.1, 0.2, or 0.5 × LD 50 of 2,5-hexanedione, and a time-related kinetic study was performed using hematology, histology, cellularity, and organ weight/body weight as major parameters. Following single exposure of 2,5-hexanedione to rats, a dose-dependent thymic atrophy was evident at the end of 7 days. The atrophy was reversible when the animals were given 7 days nonexposure rest. In contrast, there was no thymic atrophy when the animals were exposed for 7 consecutive days. Significant decline in the cellular populations of various lymphoid organs was also observed in rats exposed either to single or repeated doses of 2,5-hexanedione. Results obtained in the present study indicate that 2,5-hexanedione, a known potent neurotoxic substance, adversely affects the lymphoid organs of the immune system in rats.

We have used an X-ray fluorescence spectroscopic technique to quantify the trace elements present in Indian asbestos samples. Elements such as Cr, Mn, Fe, Ni, Cu, Zn and Ba were commonly found in all the asbestos varieties. The differences in the trace element content of Indian samples as compared to other global samples could be attributed to the different geochemical processes involved in the formation of ores at different places. The importance of trace metals has been briefly discussed in relation to their likely adverse biological effects.

The excretion, tissue distribution, and binding of [14C]-formaldehyde were studied at different time intervals in male rats following a single intraperitoneal injection of 72 mg CH2O (14.7 μCi)/ kg body weight. Within 30 min, 10% of the total dose was recovered in expired air as14CO2 and by the end of 72 hr, 41% of the administered dose was eliminated through expired air. The total elimination of14CH2O activity in urine and feces in 72 hr was 15%. Erythrocytes retained significant amounts of radioactivity, even at the end of 72 hr. Substantial levels of radioactivity were detected in most tissues one hr after administration, indicating a fast absorption and rapid distribution. Subcellular fractionation of the tissues showed that the highest levels of relative percent binding was in the microsomal fraction, whereas cytosol fractions contained lowest levels of bound radioactivity. DNA, RNA, protein and lipid fractions of liver and spleen tissues showed significantly elevated levels of14C-incorporation as compared to other tissues. Thein vivo incorporation of14C-activity showed an increased association of14CH2O with RNA in all the tissues. The maximum registration of radioactivity in RNA was at 48 hr after administration. Significantly higher amounts of14C-activity were registered in DNA of all tissues. The maximum registration of radiolabel in DNA of most tissues was at 12 hr after the14CH2O administration. The liver DNA showed maximal levels at 3 hr with a second peak at 48 hr.

High-performance liquid chromatography (h.p.l.c.) is used to fractionate the proteins of the 30–40 S nuclear ribonucleoprotein particles. The major core proteins of the particles are eluted from a SynChropak AX-300 anion-exchange column before the more acidic higher-molecular-weight minor particle proteins. Each of the three major core proteins which can be separated from the other particle proteins by preparative polyacrylamide gel electrophoresis are eluted from the SynChropak AX-300 column as one peak. Isoelectric focusing separates each of these three apparently homogeneous peaks into a series of charge isomers ranging in isoelectric pH (pI) from 5.5 to 9.0. The core proteins of the ribonucleoprotein particles have a strong affinity to each other and form aggregates. The elution of each of the charge isomers of the three major proteins in one peak and their elution from an anion exchanger before the elution of the more acidic higher-molecular-weight minor proteins of the nuclear ribonucleoprotein particles is explained by the formation of these aggregates. The separation of the total proteins of the nuclear ribonucleoprotein particles by h.p.l.c. is similar to the separation which can be obtained by preparative electrophoresis but the l.c. technique is simpler, substantially quicker, and adaptable to large-scale preparation.

Summary The effect of intake of ethanol on choline- and ethanolamine kinase activities in liver and brain tissues of mice was studied over a period of 31 days. With a single dose of ethanol there was an initial slight increase in hepatic kinase activity, while brain kinase activity decreased. The activities returned to the normal level after 24 h. Intragastric intubation of 1 ml of 30% aqueous ethanol daily for 1 month to mice led to a decrease in activity of both enzymes in both tissues.

The tissue distribution, elimination and covalent binding of 2,3-14C-and 1-14C-acrylonitrile (VCN) were studies in male Sprague-Dawley rats given an oral dose of 46.5 mg kg-1. Exhalation of unchanged VCN, 14CO2 and H14CN was monitored at selected intervals. Only 5% of the total dose administered was recovered was unchanged VCN. Rate given 2,3-14C-VCN exhaled only 2% of 14C activity was 14CO2 and none was recovered as H14CN, whereas rats given 1-14C-VCN exhaled about 12% of 14C activity as 14CO2 and 0.5% as H14CN. In the initial 24 h, 40% of radioactivity from 1-14C-VCN appeared in urine, while 60% was recovered in the urine of rats given 2,3-14C-VCN. The red blood cells retained significant amounts of radioactivity from both the compounds for more than 10 days after administration, whereas the 14C activity in plasma declined sharply. The highest level of radioactivity from both compounds was recovered in the gastrointestinal tract. In liver, kidney, brain, spleen, adrenal, lung and heart tissues the unbound percent radioactivity decreased, while irreversible percent covalent binding to macromolecules in relation to total increased concomitantly. Subcellular fractionation of the tissues showed that most of the covalently bound radioactivity was distributed in non-cytosolic fractions. As compared to 1-14C-VCN administered animals, the percentage of covalent binding of 2,3-14C-VCN was significantly higher even 72 h after dosing. The relationship between covalent binding and acrylonitrile toxicity is discussed.

Male albino rats were given intraperitoneal injections of choline chloride (0. 1,0.33 or 0.5 × lethal dose 50) for a total period of one month and then killed at the end of 30,90 and 240 days for the study of pathotoxicokinetics of choline. Chronic choline administration in rats caused a decrease in growth rate, a dose dependent modulating effect on the somatic tissue indices of lung and lymph nodes, as well as cellularity of lymph nodes. In another experiment, the effect of choline on mica induced pulmonary lesions was studied. The combined effect of choline and mica caused adenocarcinoma of bronchiolar epithelium and marked lymphadenopathy with abnormal cells in the lymph nodes at the termination of experiment (330 days). The results of the present investigation suggest that excess choline availability not only produces pulmonary pathological lesions by itself but it also further enhances the lung lesions in altered pulmonary conditions

Resume Des particules nucleaires ribonucleoproteiques 30S ont ete isolees de foie et de rein de rat. Les principales proteines de ces particules contiennent de la N G , N G -dimethylarginine. Apres injection intraperitoneale de [ 3 H-methyl]-S-adenosyl- l -methionine, les proteines de ces particules incorporent du radiomethyl tout au long de la periode de 6 heures etudiee. L'incorporation de radiomethyl dans les proteines de ces particules se revele plus faible que son incorporation dans les histones, particulierement dans le rein. Une injection simultanee d'actinomycine D et de [ 3 H-methyl]-S-adenosyl- l -methionine aboutit a la suppression rapide de l'incorporation de radiomethyl dans les proteines des particules dans le foie, et a un degre moindre, egalement dans le rein. Une electrophorese dans une premiere dimension en gradient de pH non a l'equilibre et dans la seconde dimension ont permis en presence de dodecylsulfate de sodium de separer les proteines des particules nucleaires ribonucleoproteiques 30S de foie ou de rein de rat en trois especes principales selon leur poids moleculaire, chacune composee d'une serie d'isomeres de charge a predominance basique. Aucune depletion de ces proteines, ni aucun changement dans la distribution de leurs isomeres de charge n'ont ete observes dans les particules ribonucleoproteiques 30S isolees de foie ou de rein de rat au cours des premieres 6 heures suivant l'injection d'actinomycine D. Il est donc probable que la diminution d'incorporation de radiomethyl dans les proteines des particules ribonucleoproteiniques nucleaires 30S represente une diminution de la production de groupes methyl dans les proteines des particules. Au cours de sa transformation en mRNA, le RNA precurseur est associe aux proteines de la particule. La diminution de production de groupes methyl sur les proteines de la particule pourrait provenir de l'inhibition par l'actinomycine D de la transformation du precurseur du mRNA. Une interference de l'actinomycine D, non seulement avec la synthese de RNA, mais egalement avec la transformation du RNA semble etre indiquee par la decouverte que l'activite specifique du RNA associe aux particules nucleaires ribonucleoproteiques 30S reste quasiment constante entre 2 a 6 heures apres injection de cette drogue.

The immunotoxic potential of 2,5-hexanedione (2,5-Hxdn), the end metabolite of n -hexane/methyl n -butyl ketone, was evaluated in a mouse model involving multiple pathomorphological, hematological, and immunological assays. Young adult male Swiss albino mice were given either single or seven consecutive oral doses of 0.2 × LD 50 of 2,5-Hxdn. None of the treated mice exhibited any sign of hind limb weakness up to 1 week. On the eighth day, half the animals were sacrificed for initial pathomorphological studies of various organs and the other half were subjected to several immune function tests. The results revealed treatment-related reduction in cellularity of spleen, thymus, and mesentric lymph nodes and pathotoxicological changes. Further, immune function tests such as delayed-type hypersensitivity reaction, plaque-forming cell assay, phagocytosis by adherent peritoneal exudate cells, and resistance to endotoxin shock were considerably impaired. These results suggest that 2,5-Hxdn treatment causes profound impairment of immunity in mice even before the onset of peripheral neuropathy.
