17. Investigation of Tribological Properties of Inconel 601 under Environmentally Friendly MQL and Nano-Fluid MQL with Pack Boronizing

Friction and high temperatures greatly affect the hardness and processing efficiency of superalloys. Therefore, it is important to provide a coating on their surfaces with a hard layer. In this study, pack boronizing was applied on Inconel 601 to improve its microstructure and tribological properties. In this regard, tribological tests were performed under MQL, nano-MQL1 (MQL + CuO), and nano-MQL2 (MQL + TiO2) environments. The research results showed that the lowest wear depth, friction force, coefficient of friction (CoF), and volume loss values were obtained in pack-boronized Inconel 601 in a nano-MQL2 environment. In the nano-MQL2 environment, the wear depth decreased by 17.81% (from 57.922 µm to 47.605 µm) with package-boronized Inconel 601 compared to as-received Inconel 601 at a 45 N load. Pack-boronized Inconel 601 experienced an average reduction of 30.23%, 41.60%, and 52.32% in friction force when switching from dry to MQL, nano-MQL1, and nano-MQL2 environments, respectively. It was also observed that the coefficient of friction (CoF) and volume loss values decreased with pack boronizing in an MQL/nano-MQL environment. In a nano-MQL2 environment at 15 N load, volume losses for as-received and boron-coated Inconel 601 were determined as 0.288 mm3 and 0.249 mm3, respectively (13.54% decrease). The findings of this study demonstrate that pack boronizing and MQL and nano-MQL techniques enhance the tribological characteristics of Inconel 601 alloys.

BACKGROUND: In a randomized, controlled trial that compared liraglutide, a glucagon-like peptide 1 analogue, with placebo in patients with type 2 diabetes and high cardiovascular risk who were receiving usual care, we found that liraglutide resulted in lower risks of the primary end point (nonfatal myocardial infarction, nonfatal stroke, or death from cardiovascular causes) and death. However, the long-term effects of liraglutide on renal outcomes in patients with type 2 diabetes are unknown. METHODS: We report the prespecified secondary renal outcomes of that randomized, controlled trial in which patients were assigned to receive liraglutide or placebo. The secondary renal outcome was a composite of new-onset persistent macroalbuminuria, persistent doubling of the serum creatinine level, end-stage renal disease, or death due to renal disease. The risk of renal outcomes was determined with the use of time-to-event analyses with an intention-to-treat approach. Changes in the estimated glomerular filtration rate and albuminuria were also analyzed. RESULTS: A total of 9340 patients underwent randomization, and the median follow-up of the patients was 3.84 years. The renal outcome occurred in fewer participants in the liraglutide group than in the placebo group (268 of 4668 patients vs. 337 of 4672; hazard ratio, 0.78; 95% confidence interval [CI], 0.67 to 0.92; P=0.003). This result was driven primarily by the new onset of persistent macroalbuminuria, which occurred in fewer participants in the liraglutide group than in the placebo group (161 vs. 215 patients; hazard ratio, 0.74; 95% CI, 0.60 to 0.91; P=0.004). The rates of renal adverse events were similar in the liraglutide group and the placebo group (15.1 events and 16.5 events per 1000 patient-years), including the rate of acute kidney injury (7.1 and 6.2 events per 1000 patient-years, respectively). CONCLUSIONS: This prespecified secondary analysis shows that, when added to usual care, liraglutide resulted in lower rates of the development and progression of diabetic kidney disease than placebo. Cadmium is a non-essential heavy metal that is highly toxic for most of the plants, even at low concentrations. Cadmium tolerant endophytic bacteria isolated from different sources have been effectively used for mitigating cadmium toxicity and plant growth promotion. In the present study, the potential of an endophytic bacterial strain known as Serratia sp. IU01, previously isolated from Solanum nigrum, was evaluated in terms of alleviating cadmium toxicity and promoting host plant growth. A significant increase in the biomass of two genotypes of Brassica napus L. exposed to different concentrations of cadmium was observed after three weeks inoculation with Serratia sp. IU01 compared to sole cadmium treated plants. High performance liquid chromatography (HPLC) analysis revealed the steady-levels of one of the key antioxidants, i.e., glutathione, were significantly higher in the sole cadmium treated plants compared to control plants. However, inoculation of the cadmium treated plants with the bacterial endophyte resulted in a significant reduction in glutathione contents compared to sole cadmium treated plants, which suggest the antioxidants released by the endophyte in the host environment have assisted the host plant in minimizing the magnitude of the resultant oxidative damage. The precursor of glutathione, i.e., cysteine, showed a similar pattern in the sole cadmium treated and inoculated plants. Our data suggested that inoculation of Brassica napus with the bacterial isolate Serratia sp. IU01, under cadmium stress provide a physiological advantage to the plants in terms of growth promotion and alleviating cadmium toxicity. [ABSTRACT FROM AUTHOR]

This paper presents findings from a community-based participatory study exploring the lived experiences and key livelihood changes post-intervention of a vocational skills training for young people with disabilities in rural Uganda. Twenty-four youth with disabilities (13 female, 11 male) who had previously taken the vocational training were trained to become peer researchers and conducted 72 in-depth interviews with a more recent cohort of youth with disabilities. Findings were gathered into core themes of capacity building, security and interaction. They showed that training in a skill is an important part of the economic empowerment journey for youth with disabilities in rural Uganda. Beyond this, transition from training to work, marketing, proving competence, managing chronic pain whilst working and probable risk are also areas that need guidance and support. Renewed hope for better livelihood prospects was mixed with a degree of uncertainty. Some were unprepared for the complexities around community respect and had also not considered that their financial situation may get worse before it gets better, as part of the risk of self-employment.

Replacement technology and its use in the industrial design process. The concept of substitution technology does not simply refer to a new, better quality technology but rather to technology that dissolves or replaces existing technology, and for this it makes things more difficult for many industrial organizations. From here, the research tackled the topic of substitution technology, trying to employ it and working to link it to the process of designing industrial products through the use of renewable energies as an alternative source of nutrition, and studying the impact of this on their functional and engineering performance.Hiring and working on linking it to the process of designing industrial products through the use of renewable energies as an alternative source of nutrition, and studying the impact of this on their functional and engineering performance. In addition to addressing renewable energy sources of all kinds as a form of substitution technology and studying ways to harness these energies to move the engineering design process to another level to keep pace with the rapid development of these energies and try to reach the maximum engineering benefit based on what we reached after the extensive study of many renewable energy sources.The importance of research is to confirm the importance of incorporating modern energy sources into the design process and during the product development process, and to study the extent of this impact on several factors such as product and similarity in addition to aesthetic factors and others. The research reached the imperative of the process of replacing traditional energy sources with other sustainable sources, pushing the products to the necessity of accompanying that process to give better job performance and to preserve the environment, and thus push the design process to advanced and necessary stages to go hand in hand with the rapid development of energy sources.

Three cultivars of tomato (Solanum lycopersicum L.) i.e. Rio Grande, Moneymaker and Gala were evaluated for their tissue culture responses on different types of media containing different formulations of growth regulators. Among the three cultivar tested on eight different callus induction media (CIM) formulations, Rio Grande was the most responsive cultivar showing 93.3% callus induction responses on CIM1, having 1 mgL
-1 BAP and 0.1 mgL-1 NAA, followed by Moneymaker with 46.67% callus induction responseson CIM7 containing 0.5 mgL-1 BAP, 0.5 mgL-1 NAA, 0.5 mgL-1 I AA and 1 mgL-1 Kinetin fromhypocotyl explants.Among the eight different media compositions, CIM1 resulted in maximum callus induction showing 34.1% and 27.23% average callus induction from hypocotyls and cotyledons' explants, respectively. Successfully proliferated callus were transferred to 7 types of shoot induction media (SIM). Shoot induction was observed on both SIM1 having 1 mgL-1 BAP and 0.1mgL-1 NAA and SIM2 having 2 mgL-1 BAP and 0.2 mgL-1 NAA but SIM1 showed maximum number of shoots primordia from calli. Calli with shoot primordia were transferred to shoot multiplication media i.e. MS media containing 2 mgL-1 IAA, 0.2 mgL-1 BAP, all the calli showed shoot multiplication. Rootinduction media i.e. MS or ½ MS media containing 2 mgL-1 IBA were used to induce rooting in the regenerated plantlets.Rio Grande and Moneymaker produced roots successfully and were successfully acclimatized to greenhouse after well developed rooting system. The possible changes in precipitation of Syrian due to climate change are projected in this study. The symmetrical uncertainty (SU) and multi-criteria decision-analysis (MCDA) methods are used to identify the best general circulation models (GCMs) for precipitation projections. The effectiveness of four bias correction methods, linear scaling (LS), power transformation (PT), general quantile mapping (GEQM), and gamma quantile mapping (GAQM) is assessed in downscaling GCM simulated precipitation. A random forest (RF) model is performed to generate the multi model ensemble (MME) of precipitation projections for four representative concentration pathways (RCPs) 2.6, 4.5, 6.0, and 8.5. The results showed that the best suited GCMs for climate projection of Syria are HadGEM2-AO, CSIRO-Mk3-6-0, NorESM1-M, and CESM1-CAM5. The LS demonstrated the highest capability for precipitation downscaling. Annual changes in precipitation is projected to decrease by −30 to −85.2% for RCPs 4.5, 6.0, and 8.5, while by < 0.0 to −30% for RCP 2.6. The precipitation is projected to decrease in the entire country for RCP 6.0, while increase in some parts for other RCPs during wet season. The dry season of precipitation is simulated to decrease by −12 to −93%, which indicated a drier climate for the country in the future.

The role of efficient fuel mixing and a stable flame holder is crucial in enhancing the performance and capabilities of scramjet engines for high-speed flight. The present research paper has tried to disclose the fuel mixing efficiency of 3-lobe annular nozzle on the mixing mechanism of the fuel jet behind the strut. In addition, using internal air jet flow for increasing the circulation strength and fuel mixing behind the strut is also examined in this study. Numerical simulation of the flow and fuel jet behind the strut is done to reveal the main physics related to the mechanism of fuel mixing inside the combustor with the proposed injection system. The results of our simulation show that using annular 3-lobe fuel jet improve the fuel mixing via production of the multiple vortex pairs within the combustor behind the strut. 34. Optimizing Gaussian process regression (GPR) hyperparameters with three metaheuristic algorithms for viscosity prediction of suspensions containing microencapsulated PCMs.

Suspensions containing microencapsulated phase change materials (MPCMs) play a crucial role in thermal energy storage (TES) systems and have applications in building materials, textiles, and cooling systems. This study focuses on accurately predicting the dynamic viscosity, a critical thermophysical property, of suspensions containing MPCMs and MXene particles using Gaussian process regression (GPR). Twelve hyperparameters (HPs) of GPR are analyzed separately and classified into three groups based on their importance. Three metaheuristic algorithms, namely genetic algorithm (GA), particle swarm optimization (PSO), and marine predators algorithm (MPA), are employed to optimize HPs. Optimizing the four most significant hyperparameters (covariance function, basis function, standardization, and sigma) within the first group using any of the three metaheuristic algorithms resulted in excellent outcomes. All algorithms achieved a reasonable R-value (0.9983), demonstrating their effectiveness in this context. The second group explored the impact of including additional, moderate-significant HPs, such as the fit method, predict method and optimizer. While the resulting models showed some improvement over the first group, the PSO-based model within this group exhibited the most noteworthy enhancement, achieving a higher R-value (0.99834). Finally, the third group was analyzed to examine the potential interactions between all twelve HPs. This comprehensive approach, employing the GA, yielded an optimized GPR model with the highest level of target compliance, reflected by an impressive R-value of 0.999224. It discusses the evaluation of acne severity, evidence-based guidance on acne treatment, and strategies for the management of this chronic disease. Effective treatment of active lesions and prevention of sequela is likely to improve the health of many United Arab Emirates patients with acne. - Abstract
Background: Multiple Sclerosis (MS) is an often debilitating disease affecting the myelin sheath that encompasses neurons. It can be accompanied by a myriad of pathologies and adverse effects such as neurogenic lower urinary tract dysfunction (NLUTD). Current treatment modalities for resolving NLUTD focus mainly on alleviating symptoms while the source of the discomfort emanates from a disruption in brain to bladder neural circuitry. Here, we leverage functional magnetic resonance imaging (fMRI), repetitive transcranial magnetic stimulation (rTMS) protocols and the brains innate neural plasticity to aid in resolving overactive bladder (OAB) symptoms associated with NLUTD., Methods: By employing an advanced neuro-navigation technique along with processed fMRI and diffusion tensor imaging data to help locate specific targets in each participant brain, we are able to deliver tailored neuromodulation protocols and affect either an excitatory (20 min @ 10 Hz, applied to the lateral and medial pre-frontal cortex) or inhibitory (20 min @ 1 Hz, applied to the pelvic supplemental motor area) signal on neural circuitry fundamental to the micturition cycle in humans to restore or reroute autonomic and sensorimotor activity between the brain and bladder. Through a regimen of questionnaires, bladder diaries, stimulation sessions and analysis, we aim to gauge rTMS effectiveness in women with clinically stable MS., Discussion: Some limitations do exist with this study. In targeting the MS population, the stochastic nature of MS in general highlights difficulties in recruiting enough participants with similar symptomology to make meaningful comparisons. As well, for this neuromodulatory approach to achieve some rate of success, there must be enough intact white matter in specific brain regions to receive effective stimulation. While we understand that our results will represent only a subset of the MS community, we are confident that we will accomplish our goal of increasing the quality of life for those burdened with MS and NLUTD., Trial Registration: This trial is registered at ClinicalTrials.gov (NCT06072703), posted on Oct 10, 2023., (© 2024. BACKGROUND : Since the pandemic of SARS-CoV-2 began, our understanding of the pathogenesis and immune responses to this virus has continued to evolve. It has been shown that this infection produces natural detectable immune responses in many cases. However, the duration and durability of immunity and its effect on the severity of the illness are still under investigation. Moreover, the protective effects of antibodies against new SARS-CoV-2 variants still remain unclear., Objectives: To assess the incidence and associated demographic features of SARS-CoV-2 infection in anti-nucleocapsid IgG-positive and anti-nucleocapsid IgG-negative healthcare workers., Material and Methods: This prospective longitudinal cohort study was conducted in Peshawar Medical College group of hospitals of Prime Foundation. Anti-nucleocapsid IgG sero-positive and anti-nucleocapsid IgG sero-negative healthcare workers were followed for a period of 6 months (from 1 Aug 2020 to 31 Jan 2021), and the incidence of SARS-CoV-2 was confirmed by RT-PCR., Results: A total number of 555 cohorts were followed for a period of 6 months; of them 365 (65.7%) were anti-nucleocapsid-negative (group A) and 190 (34.3%) were anti-nucleocapsid-positive (group B) healthcare workers. The mean age of the study cohort was 33.85 ± 9.80 (anti-N (-), 34.2 ± 10.58; anti-N ( +), 33.5 ± 9.50). The median antibody level in anti-nucleocapsid-positive HCWs was 15.95 (IQR: 5.24-53.4). Male gender was the majority in both groups (group A, 246 (67%), group B, 143 (48%)) with statistically significant difference (P < 0.05). Majority of the HCWs were blood group B in both groups (34% each). None of the 190 anti-nucleocapsid-positive HCWs developed subsequent SARS-CoV-2 re-infection, while 17% (n = 65) HCWs developed infection in anti-nucleocapsid-negative group during the 6-month follow-up period., Conclusion: In conclusion, none of the anti-nucleocapsid-positive HCWs developed SARS-CoV-2 re-infection in this study, and the presence of IgG anti-nucleocapsid antibodies substantially reduce the risk of re-infection for a period of 6 months., (© 2022. 41. Crosstalk Between Iron and Sulfur Homeostasis Networks in Arabidopsis .

- Abstract
The widespread deficiency of iron (Fe) and sulfur (S) is becoming a global concern. The underlying mechanisms regulating Fe and S sensing and signaling have not been well understood. We investigated the crosstalk between Fe and S using mutants impaired in Fe homeostasis, sulfate assimilation, and glutathione (GSH) biosynthesis. We showed that chlorosis symptoms induced by Fe deficiency were not directly related to the endogenous GSH levels. We found dynamic crosstalk between Fe and S networks and more interestingly observed that the upregulated expression of IRT1 and FRO2 under S deficiency in Col-0 was missing in the cad2-1 mutant background, which suggests that under S deficiency, the expression of IRT1 and FRO2 was directly or indirectly dependent on GSH. Interestingly, the bottleneck in sulfite reduction led to a constitutively higher IRT1 expression in the sir1-1 mutant. While the high-affinity sulfate transporter ( Sultr1;2 ) was upregulated under Fe deficiency in the roots, the low-affinity sulfate transporters ( Sultr2;1, and Sultr2;2 ) were down-regulated in the shoots of Col-0 seedlings. Moreover, the expression analysis of some of the key players in the Fe-S cluster assembly revealed that the expression of the so-called Fe donor in mitochondria ( AtFH ) and S mobilizer of group II cysteine desulfurase in plastids ( AtNFS2 ) were upregulated under Fe deficiency in Col-0. 42. Identifying higher risk subgroups of health care workers for priority vaccination against COVID-19.

Background: Health care workers (HCWs) are exposed to high risk of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection due to close contact with infected patients in hospital. The objective of this study was to estimate the seroprevalence and to identify the exposure risk of various subgroups among HCWs to prioritize them for early vaccination., Methods: This was a multicentre cross-sectional study conducted between 15 and 29 June 2020. A total of 987 HCWs were recruited randomly from two major tertiary-care hospitals of Peshawar city, Pakistan. The HCWs included doctors, nurses, paramedics and hospital support staff. The US Food and Drug Administration (FDA)-approved kit was used for the detection of SARS-CoV-2 antibodies., Results: Overall, 310 (31.4%) HCWs were seropositive for SARS-CoV-2 antibodies (95% confidence interval, CI: 28.5-34.4). Seroprevalence was higher in males (33.5%) and in age group 51-60 years (40.9%). Seropositivity increased with increasing age from 8.3% in age group ⩽20 to 40.9% in 51-60 years of age group ( p < 0.05). The highest seroprevalence was identified in paramedical staff (42·5%, 95% CI: 36.6-48.6) followed by nursing staff (38·8%, 95% CI: 32.1-45.7). In logistic regression, being a male HCW led to higher risk of seropositivity (odds ratio, OR: 1.50, 95% CI: 1·06-2.13. p < 0.05) compared with female staff members. The odds of seropositivity was higher in nurses (OR: 3·47, 95% CI: 1.99-6.05. p < 0.01), paramedical staff (OR: 3·19, 95% CI: 1.93-5.28. p < 0.01) and hospital support staff (OR: 2·47, 95% CI: 1.29-4.7. p < 0.01) compared with consultants., Conclusion: Overall, our results concluded that nursing and paramedical staff are at higher risk and should be vaccinated on priority., Competing Interests: Conflict of interest statement: The authors declared no potential conflicts of interest with respect to the research, authorship, and/or publication of this article., (© The Author(s), 2022.)

Purpose: Voiding dysfunction (VD) leading to urinary retention is a common neurogenic lower urinary tract symptom in patients with multiple sclerosis (MS). Currently, the only effective management for patients with MS with VD is catheterization. Transcranial Rotating Permanent Magnet Stimulator (TRPMS) is a noninvasive, portable, multifocal neuromodulator that simultaneously modulates multiple cortical regions and the strength of their functional connections. In this pilot trial (ClinicalTrials.gov Identifier: NCT03574610), we investigated the safety and therapeutic effects of TRPMS in modulating brain regions of interest (ROIs) engaged with voiding initiation to improve VD in MS women., Materials and Methods: Ten MS women with VD (having % post-void residual/bladder capacity [%PVR/BC] ≥40% or being in the lower 10th percentile of the Liverpool nomogram) underwent concurrent functional magnetic resonance imaging/urodynamic study (fMRI/UDS) with 3 cycles of bladder filling/emptying, at baseline and post-treatment. Predetermined ROIs and their activations at voiding initiation were identified on patients' baseline fMRI/UDS scans, corresponding to microstimulator placement. Patients received 10 consecutive 40-minute treatment sessions. Brain activation group analysis, noninstrumented uroflow, and validated questionnaires were compared at baseline and post-treatment., Results: No treatment-related adverse effects were reported. Post-treatment, patients showed significantly increased activation in regions known to be involved at voiding initiation in healthy subjects. %PVR/BC significantly decreased. Significant improvement of bladder emptying symptoms were reported by patients via validated questionnaires., Conclusions: Both neuroimaging and clinical data suggested TRPMS effectively and safely modulated brain regions that are involved in the voiding phase of the micturition cycle, leading to clinical improvements in bladder emptying in patients with MS.

44. SARS-CoV-2: big seroprevalence data from Pakistan-is herd immunity at hand?

Purpose: Seroprevalence surveys from different countries have reported SARS CoV-2 antibodies below 20% even in the most adversely affected areas and herd immunity cannot be predicted till more than half of the population gets the disease. The purpose of this survey was to estimate the magnitude of community-based spread of the infection, associated immunity, and the future prospects and proximity to a 'herd community'., Methods: The study was undertaken as a cluster randomized, cross-sectional countrywide survey. This largest community-based seroprevalence data of SARS-CoV-2 were collected between 15th and 31st July, 2020 from seven randomly selected cities belonging to the three most populous provinces of Pakistan. The FDA approved kit of ROCHE was used for detection of SARS-CoV-2 antibodies., Results: Serum samples of 15,390 participants were tested for SARS CoV-2 antibodies with an overall seroprevalence of 42.4%. The seroprevalence ranged from 31.1% to 48.1% in different cities with the highest in Punjab province (44.5%). In univariable analysis, the odds of seropositivity was higher in men compared to women (OR: 1.10, 95% CI: 1.01-1.19, P < 0.05). In multivariable analysis, the risk of being seropositive was lower (OR 0.72, 95% CI: 0.60-0.87, P < 0.01) in younger group (≤ 20 years) than in those aged above 60 years., Conclusion: The study concluded that despite a reasonable seroprevalence, the country is yet to reach the base minimum of estimations for herd immunity. Currently, the only effective management for VD and urinary retention in MS patients is catheterization, prompting us to look for novel therapeutic options beyond the bladder, such as the brain. Transcranial rotating permanent magnet stimulator (TRPMS) is a non-invasive, portable, multifocal neuromodulator that simultaneously modulates multiple cortical regions, enhancing or attenuating strengths of functional connections between these regions. The objective of this pilot clinical trial is to evaluate the feasibility of a TRPMS trial to address lower urinary tract symptoms in MS patients, through investigating the therapeutic effects of TRPMS in modulating brain regions during voiding initiation and mitigating VD in female MS individuals., Methods: Ten adult female MS patients with VD (defined as having %post-void residual/bladder capacity (%PVR/BC) ≥ 40% or Liverpool nomogram percentile < 10%) will be recruited for this study. Concurrent urodynamic and functional MRI evaluation with a bladder filling/emptying task repeated three to four times will be performed at baseline and post-treatment. Predetermined regions of interest and their blood-oxygen-level-dependent (BOLD) activation at voiding initiation will be identified on each patient's baseline anatomical and functional MRI scan, corresponding to the microstimulators placement on their individualized TRPMS treatment cap to either stimulate or inhibit these regions. Patients will receive 10 40-min treatment sessions. Non-instrumented uroflow and validated questionnaires will also be collected at baseline and post-treatment to evaluate clinical improvement., Discussion: Despite the crucial role of the central nervous system in urinary control and its sensitivity to MS, there has been no treatment for urinary dysfunction targeting the brain centers that are involved in proper bladder function. This trial, to our knowledge, will be the first of its kind in humans to consider non-invasive and individualized cortical modulation for treating VD in MS patients. 46. Antimicrobial Prophylaxis in Clean Pediatric Surgical Procedures: A Necessity or Redundancy?

- Abstract
Background The clinical utility of antimicrobial prophylaxis in clean pediatric surgical cases remains enigmatic. The present study aims to evaluate the prevalence of surgical site infections in instances where antibiotic prophylaxis is not employed prior to clean pediatric surgical procedures. Methods A retrospective cross-sectional study that included data of all pediatric clean surgical procedures from January 2018 till January 2020 was conducted. All children undergoing clean surgical procedures who did not receive antibiotics at least two weeks prior to the procedure were included in the study. The exclusion criteria included patients with congenital heart disease, ventriculoperitoneal shunt, nephrotic syndrome, immunodeficiency, and prior administration of antimicrobial prophylaxis. All patients were followed for two to four weeks for any signs of surgical site infections. Results Of the 178 patients included, 119 were male and 59 were female, with the mean age hovering at 8.19 ± 2.87 years. Orchidopexy and herniotomy were the most commonly performed surgical procedures, and were performed in 56 (31.46%) and 54 (30.33%) patients, respectively. Only one case of postoperative surgical site wound infection was reported, accounting for a prevalence rate of 0.56%. Conclusion In clean pediatric surgical procedures, the risk of surgical site infections is exceedingly low. The unnecessary use of antibiotics in children can cause deleterious adverse effects and promote antimicrobial resistance. 47. Sulfur metabolic engineering enhances cadmium stress tolerance and root to shoot iron translocation in Brassica napus L.

- Abstract
Serine acetyltransferase (SAT) (EC 2.3.1.30) is the rate-limiting enzyme of cysteine (Cys) biosynthesis, providing the decisive precursor for the ubiquitous defense thiol glutathione (GSH). Together with O-acetylserine (thiol) lyase (OAS-TL; EC 2.5.1.47) SAT generates Cys in the cytosol, plastids, and mitochondria of vascular plants. The current study aimed to overproduce Cys and GSH for enhanced stress tolerance via overexpression of the feedback-insensitive isoform of serine acetyltransferase from tobacco, i.e., NtSAT4. Constitutive overexpression of NtSAT4 in Brassica napus resulted in the 2.6-fold-4-fold higher SAT activity in different subcellular compartment-specific lines. This higher SAT activity led to a 2.5-fold-3.5-fold higher steady-state level of free Cys and 2.2-fold-5.3-fold elevated level of GSH in leaves compared with nontransformed plants. Among the compartment-specific lines, the mitochondrial targeted NtSAT4 overexpressor line M-182 showed the highest levels of Cys (3.5-fold) and GSH (5.3-fold) compared with wild-type plants. Overexpression of NtSAT4 conferred a physiological advantage in terms of enhanced tolerance against oxidative stress with hydrogen peroxide and the heavy metal cadmium (Cd). The NtSAT4 overexpressor lines showed a significantly higher amount of iron (Fe) translocation from roots to shoots compared with nontransformed plants. 48. Sulfate-Induced Stomata Closure Requires the Canonical ABA Signal Transduction Machinery.

- Abstract
Phytohormone abscisic acid (ABA) is the canonical trigger for stomatal closure upon abiotic stresses like drought. Soil-drying is known to facilitate root-to-shoot transport of sulfate. Remarkably, sulfate and sulfide-a downstream product of sulfate assimilation-have been independently shown to promote stomatal closure. For induction of stomatal closure, sulfate must be incorporated into cysteine, which triggers ABA biosynthesis by transcriptional activation of NCED3. Here, we apply reverse genetics to unravel if the canonical ABA signal transduction machinery is required for sulfate-induced stomata closure, and if cysteine biosynthesis is also mandatory for the induction of stomatal closure by the gasotransmitter sulfide. We provide genetic evidence for the importance of reactive oxygen species (ROS) production by the plasma membrane-localized NADPH oxidases, RBOHD, and RBOHF, during the sulfate-induced stomatal closure. In agreement with the established role of ROS as the second messenger of ABA-signaling, the SnRK2-type kinase OST1 and the protein phosphatase ABI1 are essential for sulfate-induced stomata closure. Finally, we show that sulfide fails to close stomata in a cysteine-biosynthesis depleted mutant. Our data support the hypothesis that the two mobile signals, sulfate and sulfide, induce stomatal closure by stimulating cysteine synthesis to trigger ABA production.

Awareness and knowledge about any disease is the first step to prevent and treat it, so this study evaluates osteoporosis awareness and knowledge in university students. Our results showed a high awareness and good knowledge of osteoporosis. Females were better informed than males. Knowledge increased with increasing education level., Purpose: Osteoporosis is a worldwide health problem, including Saudi Arabia where vitamin D deficiency is common. Prevention of osteoporosis must begin by increasing awareness of the disease from a young age. This study aimed to assess awareness and knowledge of osteoporosis among young adults (18-30 years) attending Saudi universities, exploring the relationship between education and gender and the sources of information in this age group., Methods: A cross-sectional survey was conducted in 337 students (176 females; 161 males) randomly selected from four Saudi universities during January-December 2017. Education level ranged from preparatory year to undergraduate and postgraduate levels. A self-reported questionnaire was designed to assess awareness and knowledge of osteoporosis across several domains, including risk factors for the disease, prevalence, symptoms, prevention, and treatment., Results: Overall, 92% of students had some awareness of osteoporosis through a variety of sources, predominantly via friends. Just over half of all students had a good or high knowledge level overall (53.4 ± 16.6%). Knowledge score correlated with education (r
2 = 0.28) and gender (r2 = 0.27); p < 0.0001. Females were better informed than males (57.7 ± 15.4% vs 48.8 ± 16.8%; p < 0.0001). Knowledge increased with increasing education level (preparatory year (47.8 ± 15.3%), undergraduate (53.5 ± 16.5%), and postgraduate (61.8 ± 15.8%); all p < 0.0001)., Conclusion: Knowledge of osteoporosis was good among university students in Saudi Arabia, higher in females and with increasing years of education. Overall, students were more knowledgeable about risk factors compared to other aspects such as symptoms, prevention, or treatment of osteoporosis.- Published

- Abstract
Plants close stomata when root water availability becomes limiting. Recent studies have demonstrated that soil-drying induces root-to-shoot sulfate transport via the xylem and that sulfate closes stomata. Here we provide evidence for a physiologically relevant signaling pathway that underlies sulfate-induced stomatal closure in Arabidopsis ( Arabidopsis thaliana ). We uncovered that, in the guard cells, sulfate activates NADPH oxidases to produce reactive oxygen species (ROS) and that this ROS induction is essential for sulfate-induced stomata closure. In line with the function of ROS as the second-messenger of abscisic acid (ABA) signaling, sulfate does not induce ROS in the ABA-synthesis mutant, aba3 - 1 , and sulfate-induced ROS were ineffective at closing stomata in the ABA-insensitive mutant abi2 - 1 and a SLOW ANION CHANNEL1 loss-of-function mutant. We provided direct evidence for sulfate-induced accumulation of ABA in the cytosol of guard cells by application of the ABAleon2.1 ABA sensor, the ABA signaling reporter ProRAB18 : GFP , and quantification of endogenous ABA marker genes. In concordance with previous studies, showing that ABA DEFICIENT3 uses Cys as the substrate for activation of the ABSCISIC ALDEHYDE OXIDASE3 (AAO3) enzyme catalyzing the last step of ABA production, we demonstrated that assimilation of sulfate into Cys is necessary for sulfate-induced stomatal closure and that sulfate-feeding or Cys-feeding induces transcription of NINE-CIS-EPOXYCAROTENOID DIOXYGENASE3, limiting the synthesis of the AAO3 substrate. Consequently, Cys synthesis-depleted mutants are sensitive to soil-drying due to enhanced water loss. Our data demonstrate that sulfate is incorporated into Cys and tunes ABA biosynthesis in leaves, promoting stomatal closure, and that this mechanism contributes to the physiological water limitation response., (© 2018 American Society of Plant Biologists.
