A random transposition mutant library of B. gladioli UAPS07070 was analyzed for searching mutants with impaired microbial antagonism. Three derivates showed diminished antimicrobial activity against a sensitive strain. The mutated loci showed high similarity to the quorum sensing genes of the AHL-synthase and its regulator. Another mutant was affected in a gene coding for a LysrR-type transcriptional regulator. The production of toxoflavin, the most well known antimicrobial-molecule and a major virulence factor of plant-pathogenic B. glumae and B. gladioli was explored. The absence of a yellowish pigment related to toxoflavin and the undetectable transcription of toxA in the mutants indicated the participation of the QS system and of the LysR-type transcriptional regulator in the regulation of toxoflavin. Additionally, those genes were found to be related to the swarming phenotype. Lettuce inoculated with the AHL synthase and the lysR mutants showed less severe symptoms. We present evidence of the participation of both, the quorum sensing and for the first time, of a LysR-type transcriptional regulator in antibiosis and swarming phenotype in a strain of B. gladioli

The Bransfield Strait, located between the South Shetland Islands and the north-western end of the Antarctic Peninsula, is a back-arc basin transitional between rifting and spreading. We compiled a geomorphological structural map of the Bransfield Basin combining published data and the interpretation of bathymetric images. Several analogue experiments reproducing the interaction between the Scotia, Antarctic, and Phoenix plates were carried out. The fault configuration observed in the geomorphological structural map was well reproduced by one of these analogue models. The results suggest the establishment of a transpressional regime to the west of the southern segment of the Shackleton Fracture Zone and a transtensional regime to the south-west of the South Scotia Ridge by at least c. 7 Ma. A probable mechanism for the opening of the Bransfield Basin requires two processes: 1) Significant transtensional effects in the Bransfield Basin caused by the configuration and drift vector of the Scotia Plate after the activity of the West Scotia Ridge ceased at c. 7 Ma. 2) Roll-back of the Phoenix Plate under the South Shetland Islands after cessation of spreading activity of the Phoenix Ridge at 3.3 ± 0.2 Ma, causing the north-westward migration of the South Shetland Trench. The presence of endophytic Acetobacter diazotrophicus was tested for pineapple plants (Ananas comosus [L.] Merr.) grown in the field. Diazotrophic bacteria were isolated from the inner tissues of surface sterilized roots, stems, and leaves of pineapple plants. Phenotypic tests permitted the selection of presumptive nitrogen-fixing A. diazotrophicus isolates. Restriction fragment length polymorphisms (RFLPs) of small subunit (SSU) rDNA using total DNA digested with endonuclease SphI and with endonuclease NcoI, hybridizations of RNA with an A. diazotrophicus large subunit (LSU) rRNA specific probe, as well as patterns in denaturing protein electrophoresis (SDS-PAGE) and multilocus enzyme tests allowed the identification of A. diazotrophicus isolates. High frequencies of isolation were obtained from propagative buds that had not been nitrogen-fertilized, and lower frequencies from 3-month-old plants that had been nitrogen-fertilized. No isolates were recovered from 5- to 7-month-old nitrogen-fertilized plants. All the A. diazotrophicus isolates recovered from pineapple plants belonged to the multilocus genotype which shows the most extensive distribution among all host species previously analyzed. Oral health/illness and surrounding socio-demographic and behavioral phenomena are poorly documented in the Mexican elderly. To establish the rationales explaining oral health/illness and tailor a Health Promotion intervention (1995-1997) accordingly, an interview nested in a questionnaire was done at baseline, with the recording of 228 interviews transcribed verbatim. Every transcript was subsequently assessed by content-analysis. Results showed that lack of oral hygiene, inadequate calcium intake and old age were the most common explanations of tooth-loss. Only a few factors were felt to be under control: the same state of relative adaptation that characterized people affected by gum problems. The themes that explained them were lack of oral hygiene, and (non-specific) infections. When interviewees evaluated the impact of good oral health on their lives, many made evaluations pertaining to quality of life, adequate function, and an attractive appearance generally. Introduction: Opsoclonus-myoclonus-ataxia (OMA) syndrome is a rare neurological disorder characterized by involuntary conjugate saccadic eye movements, myoclonus, and ataxia. Few reports exist on patients with HIV and OMA., Case Report: A 41-year-old man diagnosed with HIV-1 infection in 1997 coursed with multiple anti-retroviral schemes as a consequence of poor adherence. In 2008 he presented an HIV-1 viral load of 100,000 copies/mL and a CD4+ T cell count of 10 cells/mm3. In 2013 our patient arrived with an 11-month history of progressive opsoclonus and ataxia. He had undetectable plasma HIV-1 RNA load and CD4+ of 606 cells/mm3. No opportunistic infections were found. Cerebrospinal fluid analysis showed mildly elevated protein concentration and HIV-1 viral load of 534 copies/mL. Cerebrospinal fluid co-receptor tropism test showed selective CCR5 usage. A brain magnetic resonance imaging showed hippocampal atrophy and T2-weighted hyperintensities. Our patient exhibited a dramatic recovery and cerebrospinal fluid HIV clearance after adjustment of anti-retroviral treatment based on genotyping resistance and tropism analyses., Conclusions: In patients with HIV presenting cengral nervous system dysfunction without opportunistic infections, cerebro-spinal fluid and plasma HIV-1 viral load, resistance and tropism tests should be performed to assess a potential viral escape and to design the appropriate anti-retroviral therapy in an individual patient basis.

Objectives: Early initiation of antiretroviral therapy (ART) during acute HIV infection is associated with favourable clinical and epidemiological outcomes. Barriers to prompt treatment initiation limit the benefits of universal access to ART in Mexico. We sought to create an algorithm for the immediate detection and treatment of patients with acute HIV infection., Methods: A nationwide cohort of patients with acute HIV infection was created in 2015. In order to identify cases and treat them promptly at our centre, an interdisciplinary group coordinated through an instant-messaging tool using smart phones was established. When a probable case was detected, a discussion was initiated to confirm the diagnosis and facilitate the administrative processes to initiate ART as soon as possible. We compared time to ART initiation with that in a comparison group of patients with chronic HIV infection enrolled during the same period (May 2015 to February 2017) through routine care, using survival analysis estimators and log-rank tests., Results: We recruited 29 patients with acute HIV infection. The median time to ART initiation was 2 days in these patients, in contrast to 21 days for patients with chronic infection. There were no significant differences in the percentages of patients engaged in care, on treatment or virologically suppressed at 1 year of follow-up., Conclusions: Implementing immediate ART initiation programmes is feasible in Mexico, in spite of the substantial administrative barriers that exist in the country. The degradation of two immobilized dyes by Klebsiella sp. UAP-b5 was studied. In batch experiments, the azo dyestuffs Basic Blue 41 and Reactive Black 5 were immobilized onto corn cobs by adsorption, and the adsorption process was characterized by a pseudo-second-order kinetic equation. Klebsiella sp. UAP-b5 was previously isolated from the corn waste and shown to decolorize these dyes in liquid systems. Here, we demonstrate anaerobic decolorization and reductive biodegradation of these dyes by means of spectrophotometry, HPLC, and IR spectroscopy of the solid waste and desorption solutions. We also demonstrate adsorption of compounds that resemble known degradation products.

This study presents the first genetic characterization of five Trypanosoma rangeli isolates from Minas Gerais, in the southeast of Brazil and their comparison with Colombian populations by minicircle classification, RAPD-PCR and LSSP-PCR analyses. Our results demonstrated a homogenous T. rangeli population circulating among Didelphis albiventris as reservoir host in Brazil while heterogeneous populations were found in different regions of Colombia. KP1(+) minicircles were found in 100% isolates from Brazil and in 36.4% of the Colombian samples, whereas the KP2 and KP3 minicircles were detected in both groups. RAPD-PCR and LSSP-PCR profiles revealed a polymorphism within KP1(+) and KP1(-) T. rangeli populations and allowed the division of T. rangeli in two branches. The Brazilian KP1(+) isolates were more homogenous than the KP1(+) isolates from Colombia. The RAPD-PCR were entirely consistent with the distribution of KP1 minicircles while those obtained by LSSP-PCR were associated in 88.9% and 71.4% with KP1(+) and KP1(-) populations, respectively.

Tissue tropism, the role of reinfection in the development of Chagas' disease, and the selection of subpopulations of Trypanosoma cruzi were evaluated in hamsters inoculated with the VIC strain of T. cruzi. Adult allogeneic male hamsters were inoculated once or reinoculated by the intraperitoneal route up to four times with 2000 blood trypomastigotes. Animals were studied by blood culture, histopathology, immunohistochemistry, and molecular techniques (PCR and low-stringency single specific primer-PCR). Homogeneity of the T. cruzi population observed in different tissues suggests that selective tropism of the VIC strain extends only to various muscle tissues in hamsters and that reinfection is not a factor in the development of the inflammatory processes, although it may aggravate it, possibly due to an increase in tissue parasitism, which might induce autoimmune mechanisms. Beginning the study of chronic pathologic changes in pancreas of hamsters experimentally infected with Trypanosoma cruzi Vic strain, hepatocyte metaplasia was observed in one animal from infected group. This is the first report of oncocytes in Chagas' disease, which could be due to aberrant regenerative response to pancreas inflammatory process.

Background: We have previously shown that the prevalence of hepatitis associated with the hepatitis C virus (HCV) in patients with end stage renal disease in our institution is 10.2%. However, quantification of viral RNA in plasma and its relation with clinical variables has never been studied in our patients. Thus, the aim of the present work was to quantify the HCV viral load in patients with ESRD in dialysis, and to correlate these values with the dialysis modality and the viral genotype., Methods: We performed a transverse, prospective and comparative study in patients with HCV infection in hemodialysis, continuous ambulatory peritoneal dialysis and patients in peritoneal dialysis, but with history of hemodialysis. Viral load was quantified with RT-PCR by using a commercial kit known as Amplicor HCV 2.0. Clinical variables studied were: age, gender, end stage renal disease etiology, modality and time in dialysis, transfusions, serum albumin, aminotransferases, blood urea nitrogen, and serum creatinine., Results: Twenty four patients in dialysis with HCV infection entered into the study. Of these patients, 25% were on peritoneal dialysis, 29% on peritoneal dialysis with history of hemodialysis, and 46% were in hemodialysis. The average viral load (copies x 10(6)/mL) was 1.41 +/- 3.01. Viral load was lower in patients on peritoneal dialysis than in patients treated, or with history of hemodialysis (0.20 +/- 0.12 vs 2.04 +/- 0.88; p < 0.05). We observed no differences in viral load among patients with different viral genotypes., Discussion: The average viral load of our patients in dialysis is lower than the levels usually observed in hepatitis C infected patients without end stage renal disease. The lower viral load in patients treated with peritoneal dialysis, and no history of hemodialysis, probably denotes lower risk of chronic liver disease in these subpopulation.

The last pandemia of the century, that of the Acquired Immunodeficiency Syndrome (AIDS) caused by the Human Immunodeficiency Virus (HIV), continues to advance with gigantic steps at about 18,000 new infections per day worldwide. In the last four years, breakthroughs have been achieved and new medications have been introduced, which have impacted the progression of HIV and its disease, decreasing associated morbidity and mortality. During the course of infection, HIV replicates actively producing as much as 10(10) genetically different virions (quasi-species), which relates to immune escape, higher pathogenicity and drug resistance. Persistent viral replication causes T CD4+ cell destruction and immunodeficiency through several mechanisms. Currently, there are 14 approved anti-retrovirals that when used as triple regimens have been able to decrease opportunistic infections, hospitalization and mortality rates. Unfortunately, these regimens still have many limitations, do not cure and can only suppress the virus effectively in 50% of the treated patients. Besides, when they are inadequately used there is associated resistance development. On the other hand, indications for treatment initiation are changing continuously and heading towards a conservative approach. In the case of salvage regimens, there are only general guidelines that have not been evaluated in clinical studies. Resistance assays have great limitations and their use is very specific. All these factors have made antiretroviral treatment a very complicated issue that should be prescribed and followed by an expert.

The bioavailability of two oral formulations of cyclosporin A (Sandimmun and Neoral) was assessed in 10 children with end-stage renal disease (ESRD), while at a steady state on a dialytic procedure. The study was performed according to a randomized, double blind, cross-over design, allowing a 1-month washout period between studies. Each patient received 2.5 mg/microg of oral cyclosporin A every 12 h, either Sandimmun (SAN) or Neoral (NEO). Serum concentrations of cyclosporin A were determined serially during a 24 h period, after the 5th dose of cylosporin. Serum concentrations against time curves were constructed and bioavailability of both medications, expressed as AUC and Cmax, were compared. A statistically significant increase was observed in the AUC and Cmax of NEO, which were 90% and 130% higher, respectively, than those of SAN. Considering that the internationally accepted criteria for bioequivalence allows a 20% variation in AUC and Cmax, it appears that Neoral and Sandimmun do not bear bioequivalence in children with ESRD. Notwithstanding, there were no significant differences in trough levels between both formulations. We conclude that, if trough levels are the only source of information for dosing design, Neoral could be substituted for Sandimmun on a 1:1 basis. However, a 1:1 drug substitution is not suitable when AUC is used in children with ESRD.

From this present data it has been evaluated a new complement fixation test, comparatively to indirect immunofluorescence to diagnose chronic Chagas' disease, utilizing one watery extract of epimastigotes of Trypanosoma cruzi and three other ethanolic extracts: one from epimastigotes, one from tripomastigotes and a third one of amastigotes obtained from cultures. Utilizing 236 serum samples indirect immunofluorescence test was performed: 109 positives (20 of them with positive parasitologic diagnostic) and 127 negatives (96 of healthy blood donors and 31 with other diseases). The results have showed that is possible a positive reaction in diluted samples up to 1:16. The best limits of reactivity found were the dilutions 1:4 for the ethanolic extract of amastigotes and 1:2 for the others antigens. The correlation index among the new complement fixation test and indirect immunofluorescence test showed that the ethanolic extract from epimastigotes was the best antigen to be utilized to diagnosis purposes. Its co-positivity index with indirect immunofluorescence was 0.92207 and the co-negative index was 0.90000. Concluding, the new complement fixation test showed itself as a fast, sensible, easily applicable semiquantitative microtechnique to the diagnosis of chronic Chagas' disease.

This data evaluated the susceptibility of T. infestans to seven different strains of T. cruzi from chagasic patients of the Triângulo Mineiro, isolated and adapted in Swiss mice. A total of 200 bugs were fed in mice in acute phase injected with the different strains of T. cruzi and at each 30 days, during 5 months, their feces were obtained by abdominal compression or spontaneous dejection. The overall positivity of T. infestans infection with these different strains of T. cruzi was 62.08%. There was a high mortality rate (60%) during the examination period. We conclude that T. infestans has a low susceptibility to these strains. This could explain the low rate of positivity in the xenodiagnosis in chagasic chronic patients of the Triângulo Minerio.

This research characterizes the acute and chronic phases of Chagas' disease in hamster through parasitological and histopathological studies. The acute phase was achieved with 44 young hamsters injected intraperitoneally with 100,000 blood trypomastigotes of Benedito and Y strains of T. cruzi. The chronic phase was induced in 46 hamsters injected intraperitoneally with 35,000 trypomastigotes of Vicentia, Benedito and Y strains. Animals were sacrificed at regular intervals of 24 hours of acute phase and from the 3rd to the 10th month of infection of chronic phase. In the acute phase, parasites were easily recovered from all animals and there was an inflammatory reaction characterized by mononuclear and polymorphous leukocyte infiltration of variable degree in the majority of tissues and organs, specially in the connective loose and fatty tissues, smooth muscle myocardium and skeletal muscle. In the chronic phase the lesions occurred in the same tissues and organs, but the inflammatory response was less severe and characterized by mononuclear infiltration mainly with focal or zonal fibrosis in the myocardium. In 50% of infected animals parasites were found in myocardium and recovered from pericardic, peritoneal and ascitic fluids in some animals. Signs of heart failure, sudden death and enlargement of bowel were observed regularly. We concluded that the hamster is an useful model for Chagas' disease studies.
