Abstract

BackgroundCigarette smoking is a known risk factor for Alzheimer's disease (AD). However, the association between neurodegeneration and other substances has not been fully determined. It is of vital importance to evaluate this relationship in populations at high risk of dementia. Since substance use possibly modifies the progression rate of cognitive decline, we studied this association in a unique and well-phenotyped cohort from the University of Antioquia: carriers of the PSEN1-E280A genetic variant.ObjectiveTo determine the association between substance use and cognitive decline in carriers of the PSEN1-E280A genetic variant.MethodsA retrospective cohort study was conducted with 94 carriers and 69 noncarriers recruited between January 2019 and April 2020. A psychiatrist interviewed the participants using the Consumption of Alcohol, Cigarettes and other Substances questionnaire. The participants were also submitted to cognitive evaluation. The relationship between cognitive decline and substance use was explored through a mixed effects regression model.ResultsThere was an association between cigarettes and better performance on tasks related to perceptual organization, verbal fluency, and memory in carriers. Alcohol had a positive or negative effect on memory according to the type of alcoholic beverage. Results on marijuana use were no conclusive. Coffee was associated with progressive improvements in executive function and verbal fluency.ConclusionCigarette and alcohol were associated with an improvement of some cognitive assessments, possibly by a survival bias. In addition, coffee was related to improvements in executive function and language; therefore, its short-term neuroprotective potential should be studied.

Abstract

Across Latin American and Caribbean countries (LACs), the fight against dementia faces pressing challenges, such as heterogeneity, diversity, political instability, and socioeconomic disparities. These can be addressed more effectively in a collaborative setting that fosters open exchange of knowledge. In this work, the Latin American and Caribbean Consortium on Dementia (LAC-CD) proposes an agenda for integration to deliver a Knowledge to Action Framework (KtAF). First, we summarize evidence-based strategies (epidemiology, genetics, biomarkers, clinical trials, nonpharmacological interventions, networking, and translational research) and align them to current global strategies to translate regional knowledge into transformative actions. Then we characterize key sources of complexity (genetic isolates, admixture in populations, environmental factors, and barriers to effective interventions), map them to the above challenges, and provide the basic mosaics of knowledge toward a KtAF. Finally, we describe strategies supporting the knowledge creation stage that underpins the translational impact of KtAF.

Abstract

Alzheimer's disease (AD) and frontotemporal dementia (FTD) are neurodegenerative disorders that result in a significant burden to both patients and caregivers. By 2050, the number of people with dementia in Latin America will increase 4-fold. A deep understanding of the relevant genetic factors of AD and FTD is fundamental to tackle this reality through prevention. A review of different genetic variants that cause AD or FTD in Latin America was conducted. We searched Medline and PubMed databases using the keywords "Alzheimer's disease," "frontotemporal dementia," "mutation," "America," and "Latin America," besides specific Latin American countries. Forty-five items were chosen and analyzed. PSEN1 mutations are the commonest cause of genetic early-onset Alzheimer's disease (EOAD), followed by PSEN2 and APP mutations. Genetic FTD can be mainly explained by GRN and MAPT mutations, as well as C9orf72 G4C2 repeat expansion. APOE ε4 can modify the prevalence and incidence of late-onset Alzheimer's disease (LOAD), in addition to the cognitive performance in affected carriers.

19. Sensory characterization of the Ethiopian germplasm of Coffea arabica L. conserved in the Colombian Coffee Collection.

Abstract

The sensory quality of coffee (Coffea arabica L.) has become increasingly important and is considered the best way to increase the competitiveness and profitability of the sector. Given the narrow genetic base cultivated, the use of its genetic resources conserved in germplasm banks is particularly relevant, especially for germplasm collected in Ethiopia, the country from which coffee originated and the center of its diversity of profiles. However, the potential benefits of using these resources is unknown. Therefore, the objective of this study was to characterize the sensory quality of the Ethiopian germplasm conserved in the Colombian Coffee Collection (CCC) to determine its variability and potential for use. The sensory qualities of drinks from 378 accessions of CCC were characterized. The results showed that 34% of the accessions evaluated presented sensory descriptors different from those of the traditional varieties. For the Global Impression attribute, the highest scores were associated with citrus, spice and floral notes. Additionally, prospecting rather than the area of origin of the germplasm influenced its sensory diversity, possibly due to the established objectives of the collection. The conserved germplasm exhibited important sensory variability, which is linked to rare descriptors in traditional varieties. Characterization activities of genetic resources, such as those conducted in this study, add value and serve as an initial step toward their application in breeding. The results obtained allow the selection of genotypes with distinct sensory profiles, which can be incorporated into genetic improvement programs aimed at developing future varieties with these attributes. Abstract

Coffee berry disease (CBD) is not present in the Americas and presents a potential risk for growing coffee. Therefore, Colombia, which has been in scientific cooperation with the Centro de Investigação de Ferrugens do Cafeeiro (CIFC) of Portugal for more than 30 years, has been evaluating the genetic resistance of nine populations of C. arabica to 13 isolates of Colletotrichum kahawae JM Waller and PD Bridge, which are diverse in terms of aggressiveness and geographical origin. The phenotypes observed in the interaction between C. arabica and C. kahawae were used to develop a statistically reliable scale (p-value ≥ 0.001) to categorize resistance in C. arabica into five classes, and this scale was used to classify the nine populations of C. arabica evaluated. The results allowed us to corroborate the potential of Timor Hybrid CIFC 1343 (TH CIFC 1343) as a source of genetic resistance to CBD and to identify new genetic sources not yet explored for the development of varieties in Colombia that may eventually mitigate the effects of CBD in the face of increasing rainfall events and minimum temperatures due to climate change, which can favor disease development. Additionally, the results suggest that the existence of races in the C. arabica–C. kahawae complex is probable, and a selection of genotypes was identified as a possible differential series of races in C. kahawae. Abstract

Background & aimsSex and sex hormones can affect responses of patients with nonalcoholic fatty liver disease (NAFLD) to metabolic stress and development of hepatocyte injury and inflammation.MethodsWe collected data from 3 large U.S. studies of patients with NAFLD (between October 2004 and June 2013) to assess the association between histologic severity and sex, menopause status, synthetic hormone use, and menstrual abnormalities in 1112 patients with a histologic diagnosis of NAFLD. We performed logistic or ordinal logistic regression models, adjusting for covariates relevant to an increase of hepatic metabolic stress.ResultsPremenopausal women were at an increased risk of lobular inflammation, hepatocyte ballooning, and Mallory-Denk bodies than men and also at an increased risk of lobular inflammation and Mallory-Denk bodies than postmenopausal women (P < .01). Use of oral contraceptives was associated with an increased risk of lobular inflammation and Mallory-Denk bodies in premenopausal women, whereas hormone replacement therapy was associated with an increased risk of lobular inflammation in postmenopausal women (P < .05).ConclusionsBeing a premenopausal woman or a female user of synthetic hormones is associated with increased histologic severity of hepatocyte injury and inflammation among patients with NAFLD at given levels of hepatic metabolic stress.

Abstract

In this study we identify diverse experiences of Psychology students during online classes, within the framework of covid-19, and in face-to-face classes during the post-pandemic period. Based on a sociocultural psychology approach, we analyzed aspects of these experiences and their impact on the students' learning, emotions and socialization with peers. Five open-ended questionnaires were applied online over two semesters of face-to-face classes and a content analysis was conducted. We observed that students valued the face-to-face classes, felt happy, more focused on academics and content with their social life. However, they considered that online classes are advantageous because they allow them to save money and time in traveling to school. We thus concluded that in both modalities the students learned to recognize and highlight the advantages and inconveniences of each one., En el presente trabajo identificamos diversas experiencias de estudiantes de la carrera de Psicología durante las clases en línea, en el marco de la covid-19, y en las presenciales durante la pospandemia. Desde un enfoque de psicología sociocultural, analizamos aspectos de dichas experiencias sobre su aprendizaje, sus emociones y su socialización con pares. Se aplicaron en línea cinco cuestionarios con respuestas abiertas a lo largo de dos semestres de clases presenciales y se realizó un análisis de contenido. Encontramos que las y los estudiantes valoraron las clases presenciales, se sintieron felices, más concentrados en lo académico y contentos con la vida social. Empero, consideraron que las clases en línea son favorables porque les permiten ahorrar dinero y tiempo en traslados a la escuela. Se concluye que en ambas modalidades el estudiantado aprendió a reconocer y destacar las ventajas y desventajas de cada una.

Abstract

Three hybrid populations (F1) of Coffea arabica were evaluated under field and laboratory conditions, derived from sources carrying the SH1 coffee leaf rust (CLR) resistance gene and the CX.2385 line, obtained from the Caturra × Timor Hybrid CIFC-1343. The results obtained under controlled conditions and analyzed using survival curves allowed to estimate the probable times (p < 0.05) for the development of symptoms associated with CLR in the plants of populations evaluated. Phenotypic variation was observed as a defense response to Hemileia vastatrix infection, and plants with incomplete resistance to CLR were identified via an evaluation using the increasing lesions scale. The plants with incomplete resistance exhibited a delay in the development of the incubation period and an absence of the development of the dormancy period. Data suggest that when resistance genes in the sources are defeated by compatible strains, their recombination can give rise to new levels of resistance in the progeny. Additionally, the detached leaf methodology is recommended as an alternative to preselect genotypes with resistance to CLR, thus reducing the number of plants that are finally planted for field evaluations.
