Amomum is a pungent and aromatic plant genus that contains 150–180 species, where Southeast Asia is the center of endemism, with Indonesia indigenous to 24 breeds. These species are used as spices and traditional medicine for the treatment of various diseases. This paper aims to provide Amomum species summarized data regarding phytochemistry and biological activities. Several studies have been carried out on the fruits, seeds, roots, rhizomes, and leaves of Amomum species from 1999 to 2024, as approximately 127 metabolites were isolated as flavonoid, diterpenoid, diarylheptanoid, monoterpenoid, sesquiterpenoid, phenylpropanoid, phenolic, and steroid groups. Besides cytotoxicity, antioxidant, and anti-inflammatory potentials; it also has an owed tendency for use as a chemical marker. The extracts and compounds obtained from the Amomum species were evaluated for biological activities, including cytotoxicity, antioxidant, anticancer, antiproliferative, anti-inflammatory, antifungal, antimicrobial, neuroprotective, platelet antiaggregation, and antidiabetic properties. Tsaoko arilon (neolignane) had antiproliferative and cytotoxic activity, with the highest reactions considered as lead compounds for further development. The findings highlighted the significance of using compounds from the Amomum genus in traditional medicine and the discovery of new medicines. Therefore, the results supported the concept of utilizing Amomum species as a potential source for producing biologically active compounds.

ABSTRACT. Sesquiterpenoids are a class of terpenoid compounds with a remarkable diversity of structures and biological activities. Sesquiterpenoids are primarily found in higher plants, such as the Meliaceae family's Aglaia genus. Aglaia cucullata is a species of Aglaia that is still rarely explored and can potentially contain sesquiterpenoid compounds with cytotoxic activity. Hence, the research intended to isolate sesquiterpenoids from the n-hexane extract of A. cucullata stem bark and evaluate their cytotoxic effect against A549 lung cancer cells. Compounds 1 and 2 were isolated and purified from n-hexane extracts utilizing various chromatographic techniques. The structure of compounds 1 and 2 was determined by analyzing various spectroscopic methods (IR, MS, and NMR) and comparing them to previously reported spectral data. Compound 1 was identified as an isodaucane-type sesquiterpenoid, 10-hydroxy-6,10-epoxy-7(14)-isodaucane, and was first reported in Meliaceae family. Compound 2 was confirmed as an eudesmane-type sesquiterpenoid, eudesm-4(15)-ene-1β,6α-dihydroxy, and was first reported in Aglaia cucullata. Cytotoxic activity of 1 and 2 were investigated in vitro against A549 lung cancer cells using the PrestoBlue assay and resulted in inactive and low cytotoxicity with IC50 values of 292.77 and 90.55 μM, respectively. ABSTRACT. Limonoid group belongs to triterpenoid that has undergone further oxidation accompanied by the loss of four carbon atoms to form a furan ring in the chain. This limonoid compound is often found in the Chisocheton genus. Limonoid compounds have been known as compounds with high structural variations and this makes limonoids have diverse and interesting activities, including cytotoxic and anticancer. In the course of our continuing study for limonoid constituents that have cytotoxic activity against cancer cell lines, methanol extract of stem bark from the Chisocheton pentandrus plant provides significant activity. The methanol extract was separated using various chromatographic techniques in the normal and reverse stationary phase to produce four azadirone-type limonoid compounds (1-4). The elucidation structure of 1-4 was determined using spectroscopic methods including, UV-Visible, IR, and 1D-NMR as well as optical rotation. All four known compounds were established as trichilenone acetate (1), toonaciliaton C (2), 11α-acetoxiazadiron (3), and 16β-hydroxydisobinine (4). The cytotoxicity of compounds 1-4 was assessed by examination using the resazurin method, which showed that compound 4 was the promising constituent against the MCF-7 cells with an IC50 value of 43.1 µM and was stronger than its positive control. Aim – This study investigates the effect of Family-Supportive Supervisor Behaviour (FSSB), work-life balance, job satisfaction, motivation, and job performance among married private employees in Indonesia. Methodology – Data collection was carried out online using Goole form. This study used a sample of 206 private employees who were married and worked for at least 3 years in Indonesia. Data was analysed using Structural Equational Model (SEM) with Lisrel 8.80 software. Findings – The result showed that FSBB, job satisfaction, and motivation, have a positive effect on job performance. FSSB also has a positive effect on motivation. Work-life balance and motivation also positively influence job satisfaction. Another finding showed that work-life balance has no effect on the performance of married private employees. Based on testing the indirect relationship, there are two mediating roles whose results are significant, namely motivation mediates the relationship between FSSB on job performance. In addition, job satisfaction mediates the relationship between motivation and job performance. Limitations – This research still has some limitations that need to be improved in the future. First, the survey only focuses on private sector employees. Second, this study uses respondents who are private employees, married, but limited to a minimum working period of 3 years and a minimum education of bachelor’s degrees. Third, based on regional distribution, most of the respondents work on the island of Java, especially in the Greater Jakarta area. Fourth, the variables used in this study only focus on five variables, namely FSSB, work-life balance, motivation, job satisfaction, and performance. Originality/value – The study contributes to literature that the family-supportive supervisor behaviours can increase motivation and job performance especially for among private employees who are married. When motivation increases, it affects satisfaction and job performance. Companies can also implement a work-life balance as an effort to increase job satisfaction.

Background: Ramie (Boehmeria nivea) has been reported to have a wide variety of secondary metabolite compounds in its vegetative organ that have distinct organic activities. Regarding the chemical composition of flowers, there have been no study considering their pharmacological effects. Purpose: This research aims to determine profile metabolites contained in B. nivea flowers employing a metabolomics approach. Study Design: This research was designed to obtain a metabolite database from five dissolved fractions of ramie flower using a Gas Chromatography-Mass Spectrometry (GC–MS/MS) instrument. This study includes metabolite profiles from each fraction of the spectrum obtained, which are then investigated for potential bioactivity based on previously reported studies. Metabolite profiles were developed using Principal Component Analysis (PCA) to obtain information on clustering of the compounds and determination of marker compounds from each fraction analyzed. Methods: The active compounds of the flowers were separated utilizing the maceration method with ethanol solvent and multilevel fractionation using n-hexane, dichloromethane, ethyl acetic acid derivation, butanol, and water employing a GC–MS/MS approach. Principal Component Analysis (PCA) was optimized with a total represented data diversity range of 60 % - 65 %. This allowed for the identification of characteristic compounds and similarities in compound substances accross the fractions. Result: GC–MS/MS interpretation showed the dominant compounds contained in each fraction of n-hexane, dichloromethane, n-butanol, ethyl acetate, and water are 9,12-Octadecadienoic acid, ethanol, 2-phenoxy-, butyl ester, and hexanedioic acid, bis(2-ethylhexyl) ester. PCA clustering showed combined similarity of the compound for ethyl acetate fraction with butanol, and also butanol with n-hexane. Meanwhile, the dichloromethane and water fractions have very small content similarities. The score plot indicated marker compounds of each fraction are: ethanol, 2-butoxy; hexane, 2,2,3-trimethyl-; phthalic acid, in (2-propylpentyl) ester; n-hexadecanoic acid, and Hexanedioic acid, bis(2- ethylhexyl) ester. Conclusions: The abundance of compounds in the fractions shows potential for application as a source of new drug candidates. In addition, there are still many compounds that are unknown in structures or biological activities which could lead to further research to provide potential clues towards the bioprospecting of the plant. Steroids are one of the secondary metabolite groups that are abundant in many organisms. In plants, this type of compound is called phytosterols. Phytosterols have been widely known to show many potential bioactivities such as anti-inflammatory, induced apoptosis, cytotoxic, anti-diabetic, angiogenic, and antioxidant. One of the sources of phytosterol compounds is the genus Aglaia. As the largest genus in the Meliaceae family, the genus Aglaia contains many bioactive compounds, including steroids. This research reported the isolation, structural determination, and cytotoxic activity of steroid compounds from the stem bark of Aglaia cucullata, one of the members of the Aglaia genus. Pure isolated steroid was obtained after maceration of dried stem bark with ethanol and partitioned based on difference polarity, continued by column chromatography. Spectroscopic methods, including HRMS, FTIR, 1D and 2D NMR, were used for structural determination. The compound structure identified as stigmast-5-en-3β-ol-3β-oleate was first isolated from this species. MCF-7 breast cancer cell, B16-F10 melanoma cell, and CV-1 normal fibroblast kidney cell were used to evaluate its cytotoxicity. Stigmast-5-en-3β-ol-3β-oleate displayed low cytotoxicity against those two cancer cells and a normal cell.

The Aglaia species, which contains triterpenoids, is the most numerous in the Meliaceae family. The A. cucullata species, of which there are only a few known examples, has received scant research attention. This investigation aims to identify triterpenoids in an n-hexane preparation of A. cucullata stem bark and evaluate their effects against the A549 lung cancer cell line. Five dammarane-type triterpenoids were isolated from the A. cucullata trunk bark, which is (1) (20S)-20-hydroxydammar-24-en-3-one, (2) cabraleone, (3) cabralealactone, (4) eichlerianic acid, and (5) (+)-fouquierol. Their chemical structures were determined using infrared, high-resolution mass spectrometry, and nuclear magnetic resonance, as well as through data comparison of the reported compounds. Compound 1 was priorly separated from the Aglaia genus, compounds 2–4 were first isolated from the A. cucullata species, and compound 5 has been reportedly isolated from the Meliaceae family and the Aglaia genus. All substances were tested for their lethal potential against the A549 lung cancer cell type. A seco structure in the A ring of dammarane-type triterpenoid might play an important part in the lethal activity of component 4, which showed the greatest activity with an IC50 value of 32.17 µM against the A549 lung cancer cell line.

Chisocheton is a plant of the Meliaceae family which has been known as a source of limonoids, triterpenoids, steroids, alkaloids and phenolics. This plant is the second largest in the Meliaceae, with 53 species widely distributed in tropical and subtropical regions including Indonesia. From this genus, compounds that have interesting activities have been found, including anti-inflammatory, antimalarial, cytotoxic, antitumor and anticancer. One of the species that has the potential to find new compounds is Chisocheton lasiocarpus because there are still few phytochemical studies. This study aims to inform the structural elucidation of one of the compounds from the stem bark of C. lasiocarpus, namely morelloflavone and its cytotoxic potential against breast cancer cells using the in silico method against ER-α receptors (PDB code: 3ERD), ER-β (PDB code: 1QKM). ) and HER-2 (GDP code: 3PP0).

Natural compounds have demonstrated good biological activity when combined with certain amino acids. For example, a glycine-conjugated glycyrrhetinic acid exhibits heightened efficiency against MCF7 cancer cells. Consequently, a molecular modeling analysis is conducted to construct glycine-conjugated α-mangostins and investigate their potential. According to pharmacophore modeling using the ligand-based drug design technique, only two glycine-conjugated α-mangostins conform to the pharmacophore features. The docking simulation results show that the Am1Gly conjugate can interact with the estrogen receptor-α (ERα) with a binding energy of −10.91 kcal/mol. This interaction is further supported by molecular dynamics simulations performed over a 200 ns timeframe. Based on molecular dynamics modeling using the MMPBSA method, the binding affinity of Am1Gly (ΔGTotal = −48.79 kcal/mol) is determined. The results of this analysis indicate that Am1Gly might function as an antagonist to estrogen receptors.

Nocardiotide-A (cWIWLVA) is a cyclic peptide with significant cytotoxicity against several cancer cells. The present research aimed to design a radiopeptide based on nocardiotide-A analogues to be labeled by technetium-99m targeting SSTR2, which is the most widely expressed receptor in several types of human cancers and used as radiopeptide target. Nocardiotide-A analogues were individually designed by replacing valine at the lead compound with lysine, arginine, histidine, asparagine, and glutamine, and this was simulated by molecular dynamics using AMBER18. A molecular docking using AutoDock 4.2 was performed and evaluated to understand the effect of chelation of technetium-99m on 99mTc-HYNIC-EDDA and 99mTc-HYNIC-tricine on the binding affinity of nocardiotide-A analogues. The molecular dynamics simulation confirmed that the designed nocardiotide-A-based peptides were stable in the binding pocket of SSTR2 for 200 ns. Moreover, the nocardiotide-A-based radiopeptides are able to interact with residues Q102, D122, Q126, and N276 by building hydrogen bonds, which are essential binding residues in SSTR2. The molecular docking simulation revealed that the best docking parameter is exhibited by 99mTc/EDDA/HYNIC-cWIWLNA and 99mTc/tricine/HYNIC-cWIWLNA with a binding free energy of –12.59 kcal/mol and –8.96 kcal/mol, respectively. Taken together, nocardiotide-A-based radiopeptides are prospective to be further developed for cancer imaging targeting SSTR2.

Marine environment is known as a source of potential steroids with multiple biological activities, one of which is an anticancer agent. Atactodea striata are one of the seashells distributed in Indonesia located in the Kei Islands, Southeast Maluku. During the course of our continuing search for biologically active substances from Indonesia seashells, seven steroids have been isolated from the n-hexane fraction of A. striata and they were identified as 7β-hydroxy-sitosterol (1), campesterol (2), β-sitosterol (3), cholesterol (4), 5α,8α-epidioxycholest-6-en-3-β-ol (5), 7-keto-cholesterol (6), and 7α-hydroxy-cholesterol (7). The structure was identified by spectroscopic methods including 2D NMR techniques, FTIR, HRTOFMS, and chemical shift comparison with previously reported spectral data. Compounds 1-7 were evaluated for their cytotoxic effects against MCF-7 breast cancer cells and showed weak or no anticancer activity.

Sesquiterpenoids are derivatives of terpenoid compound that have a diverse skeleton and broad spectrum of biological activities, particularly anticancer activity. This research reported the isolation and elucidation structure of sesquiterpenoid from Chisocheton lasiocarpus stem bark, as well as their cytotoxic activity. Four sesquiterpenoid compounds, namely eudesm-4(15)-ene-1β,6β-diol (1), allo-aromadendrane-10α,14-diol (2), allo-aromadendrane-10β,14-diol (3) and guaianediol (4), were isolated for the first time from Chisocheton lasiocarpus stem bark. The chemical structure of all compounds was identified by spectroscopic analysis, including IR, MS, 1H-NMR, 13C-NMR and DEPT, and compared with previous reported spectral data. The cytotoxic activity of the isolated compounds against MCF-7 breast cancer line were examined by using the resazurin method. The results showed compound 1 shown the highest cytotoxic activity with IC50 108.08 ± 0.58 µM.

Catechins are a type of flavonoid known for their beneficial functions as antioxidants and antibacterials. Recent research indicates the antioxidant potential of catechins on the skin. Catechin and epigallocatechin are reported to have significant potential in preventing ageing. Epigallocatechin gallate, gallocatechin gallate, and epigallocatechin can inhibit hyperpigmentation processes. Additionally, catechins exhibit potential in UV protection and inflammation inhibition in acne. Consequently, catechins are now being used in the cosmetics industry, with formulations containing catechins as the active ingredient developed to produce various products such as soap, sunscreen, creams, etc. Herein, this paper reviews the antioxidant potential of catechins for use in cosmetic formulations and the current status of clinical trials of catechins in cosmetics.

Bacicyclin is a cyclic hexapeptide with antibacterial activity against Enterococcus faecalis and Staphylococcus aureus with minimum inhibition concentration (MIC) values of 8 and 12 µM, respectively. Studies on a reversed sequence of bacicyclin were conducted to investigate how the reversed peptide sequence affects its biological properties. A reversed-bacicylin, cyclo-(Gly-Leu-Val-Ile-Ala-Phe), was successfully synthesized by constructing the linear precursor on 2-chlorotrityl chloride resin using a Fmoc-based strategy. The HATU/HOAt reagent was applied in all peptidic bond formations, and the desired linear hexapeptide (82% yield) was cleaved off the resin using a mixture of trifluoroacetic acid:dichloromethane (2:8). The linear peptide was cyclized using 1-[bis(dimethylamino)methylene]-1H-1,2,3-triazolo[4,5-b]pyridinium-3-oxidehexa-fluorophosphate (HATU) as a coupling agent and diisopropylethylamine (DIPEA) as the base in a very dilute solution (0.001 M) in dichloromethane, then purified by octadecyl silica gel (ODS) column chromatography to obtain the reversed-bacicyclin (43.7% yield). The purity of the cyclic product was analyzed using analytical RP-HPLC (tR = 20.01 min), and characterized by HR-TOF-MS, 1H-NMR, and 13C-NMR. The biological activity of the reversed-bacicyclin is much lower compared to bacicyclin, indicating that the amino acid sequence of the cyclopeptide dictates the antibacterial activity. This finding gives additional information on the relationship between peptide sequence and biological properties.

The diversity of structures of triterpenoids provides a variety of interesting pharmacological effects, one of them being anticancer. Sources of triterpenoids are found in Dysoxylum excelsum, it is still rarely studied, and it potentially contains triterpenoids with cytotoxic activity. Therefore, the research aimed to isolate the triterpenoids from the stem bark of D. excelsum and determine their cytotoxic efficacy against the breast cancer cells of MCF-7. The dried stem bark of D. excelsum was macerated using methanol and then fractionated with n-hexane and ethyl acetate to obtain the corresponding fractions. Two triterpenoids, 3α,7α-dihydroxyapotirucalla-14,24-Z-dien-26-oic acid (1) and masticadienonic acid or 3-oxo-tirucalla-7,24-Z-dien-26-oic acid (2) were isolated in the first place from the Dysoxylum genus. Compounds 1 and 2 were assayed for their cytotoxic potency anti- MCF-7 using the MTT assay and gave moderate IC50 values of 60.87 and 72.86 µM, respectively. Based on their analogs, partial structure modification of the compounds might increase the activity; thus, it can be further utilized as better lead compounds for anticancer drugs candidate.

The n-hexane fraction from the stem bark of Lansium domesticum Corr. Cv. Kokosan afforded 3 sesquiterpenoids, namely eudesm-4(15),7-dien-1β-ol (1), eudesm-4(15)-ene-1β,6α-diol (2), and octahydro-4-hydroxy-3a-methyl-7-methylene-a-(1-methylethyl)-1H-indene-1-methanol (3). These three compounds were discovered in the Lansium genus for the first time. Their chemical structures were determined based on data generated from various spectroscopic methods, including one- and two-dimensional NMR, as well as mass spectroscopy. Sesquiterpenoid compounds (1-3) have also been evaluated on MCF-7 breast cancer cell lines and the compound 1 showed the strongest activity with an IC50 value of 17.97 µg/mL while compounds 2 and 3 showed moderate and weak activity with IC50 values of 121.65 and 201.57 µg/mL, respectively. The implication of the findings of these compounds is as an illustration that one of these sesquiterpenoids has potential as an anticancer with the presence of double bond which played important role in the cytotoxic activity that can be studied for new drug discovery.

A new limonoid, belonging to the havanensin-type, called pentandricine E (1), alongside four other known types, i.e., trichilenone acetate (2), toonaciliatones C (3), 11α-acetoxyazadirone (4) and ceramicine I (5), were found in Chisocheton pentandrus stem bark. The structures of the isolated 1-5 were identified by NMR spectroscopy and HRTOFMS. Pentandricine E (1) displayed the highest cytotoxicity toward the MCF-7 breast cancer line with an IC50 value of 63.27 ± 0.21 mM.

Epimerisation is basically a chemical conversion that includes the transformation of an epimer into another epimer or its chiral partner. Epimerisation of amino acid is a side reaction that sometimes happens during peptide synthesis. It became the most avoided reaction because the process affects the overall conformation of the molecule, eventually even altering the bioactivity of the peptide. Epimerised products have a high similarity of physical characteristics, thus making it difficult for them to be purified. In regards to amino acids, epimerisation is very important in keeping the chirality of the assembled amino acids unchanged during the peptide synthesis and obtaining the desirable product without any problematic purification. In this review, we report several factors that induce epimerisation during peptide synthesis, including how to characterise and affect the bioactivities. To avoid undesirable epimerisation, we also describe several methods of suppressing the process.

Peptide compounds play a significant role in medicinal chemistry as they can inhibit the activity of species that cause malaria. This literature review summarizes the isolation of antimalarial peptides, the synthesis method with the detailed structure and sequences of each peptide, and discusses the biological activity of the isolated and synthesized compounds. The synthetic routes and reactions for cyclic and linear antimalarial peptides are systematically highlighted in this review including preparing building blocks, protection and deprotection, coupling and cyclization reactions until the target compound is obtained. Based on the literature data and the results, this review’s aim is to provide information to discover and synthesize more antimalarial peptide for future research.

Aglaia is the largest genus of the Meliaceae family which contains terpenoid compounds. This type of compounds showed a diverse structures and biological activities that can be found in natural resources. Aglaia elaeagnoidea is a species from Aglaia genus that only has a few previous research. This study was aimed to isolate and determine the chemical structure of terpenoid compounds from the ethyl acetate extract of A. elaeagnoidea stem bark. Ethyl acetate extracts were separated and purified by various chromatographic techniques to obtain compounds 1-5. Compounds 1-5 were identified their chemical structures by spectroscopic methods (IR, MS, and NMR) and comparison with previous reported spectral data. Compounds 1 and 2 were identified as eudesmane-type sesquiterpenoids, 5-epi-eudesm-4(15)-ene-1β,6β-diol (1) and 6α-Hydroxy-eudesm-4(15)-en-1-one (2). Compounds 3-5 were identified as dammarane-type triterpenoids, 20S,24S-epoxy-25-hydroxydammarane-3-one (3), 20S,24S-epoxydammarane-3α,25-diol (4), and 3α-epi-cabraleahydroxy lactone (5). These compounds are first time reported from this plant. Compounds 1-5 were tested for cytotoxicity against HeLa cervical cancer cell and DU145 prostate cancer cell and as a result, compound 4 (20S,24S-epoxydammarane-3α,25-diol) showed the stronger activity compared to other compounds.

Epitope-based peptide vaccine can elicit T-cell immunity against SARS-CoV-2 to clear the infection. However, finding the best epitope from the whole antigen is challenging. A peptide screening using immunoinformatics usually starts from MHC-binding peptide, immunogenicity, cross-reactivity with the human proteome, to toxicity analysis. This pipeline classified the peptides into three categories, i.e., strong-, weak-, and non-binder, without incorporating the structural aspect. For this reason, the molecular detail that discriminates the binders from non-binder is interesting to be investigated. In this study, five CTL epitopes against HLA-A*02:01 were identified from the coarse-grained molecular dynamics-guided immunoinformatics screening. The strong binder showed distinctive activities from the non-binder in terms of structural and energetic properties. Furthermore, the second residue from the nonameric peptide was most important in the interaction with HLA-A*02:01. By understanding the nature of MHC-peptide interaction, we hoped to improve the chance of finding the best epitope for a peptide vaccine candidate.

Four sesquiterpenoid derivatives, i.e., 4β,10α-dihydroxyaromadendrane (1), caryophyllenol-II (2), senecrassidiol (3), and clovane-2β,9α-diol (4) have been isolated from the stem bark of Aglaia simplicifolia. The chemical structures of compounds 1-4 were determined based on spectroscopic data, including one and two-dimensional NMR and mass spectroscopy. In addition, these sesquiterpenoids 1-4, were also tested for their cytotoxic activity against B16-F10 melanoma skin cancer cell lines through in vitro assay. Among the isolated compounds 1-4, compound 1 showed the highest activity with an IC50 value of 44.8 μg/mL, suggesting the presence of a cyclopropane ring that plays an essential role in cytotoxic activity against B16-F10 melanoma skin cancer cell lines.

Amomum compactum Soland Ex Maton is one of the Zingiberaceae family plants which is the endemic plants from West Java, Indonesia. This study was aimed to determine the chemical structure of flavonoid compounds from n-hexane extract of A.compactum Sol. Ex Maton roots. Dried powder of the roots was extracted consecutively with n-hexane, ethyl acetate, and methanol solvents. Three flavonoids, 5-hydroxy-3,7,4`-trimethoxy kaempferol (1), 5-hydroxy-3,7,3',4'-tetra methoxy kaempferol (2) and 4'-hydroxy-3,5,7-trimethoxy kaempferol (3), have been isolated from the roots of A. compactum Sol. Ex Maton. The chemical structures of compounds 1-3 were identified by spectroscopy data including infrared 1D-NMR, 2D-NMR and HRTOF-MS as well as by comparison with previously reported spectral data. Compounds 1-3 were isolated from this plant for the first time and showed free radical DPPH scavenging activity.

Limonoids (tetranortriterpenoids) are triterpenoid compounds that lose four terminals in their structural framework. These compounds have a wide variety of structures and interesting activities including anti-inflammatory, anti-cancer, anti-tumor and anti-malarial properties. The purpose of this study was to find limonoid compounds from the Indonesian Chisocheton plant, and one of which is Chisocheton macrophyllus. The dried and powdered seeds of C. macrophyllus (4.5 kg) were extracted with methanol and partitioned successively with n-hexane, ethyl acetate and n-butanol. Evaporation of each extract resulted in the crude extracts of n-hexane (346.6 g), ethyl acetate (60.8 g) and n-butanol (14.6 g). The n-hexane fraction was subjected to a silica gel vacuum-liquid chromatography (VLC) column packed with silica gel 60 using gradient of n-hexane, ethyl acetate and methanol (10% stepwise) to afford thirteen fractions (A-M). Fraction F (4.4 g) was subjected to silica gel column chromatography using gradient of n-hexane and ethyl acetate (5% stepwise). Subfraction F5 (1.2 g) was chromatographed on a column of silica gel eluted with n-hexane: CH2Cl2: EtOAc (2:7.5:0.5) to give compound 1 (19.7 mg) and fraction H (1.8 g) was subjected to silica gel column chromatography using gradient of n-hexane and ethyl acetate (5% stepwise) as eluting solvent to give 2 (12.0 mg). Chemical structures of 1 and 2 were elucidated by spectroscopic methods and determined as 6α-acetoxyepoxyazadiradione (1) and Dysobinin(2). Dysobinin (2) showed weak cytotoxic activity against MCF-7 breast cancer cells with an IC50 value of 228.15 μM

One of the common diseases of shallot is moler disease, caused by Fusarium oxysporum. This disease causes some damage to the plant. The attack mechanism of Fusarium oxysporum is by penetrating, colonizing, and multiplying in the root area, then parasitizing and inhibiting the process of carrying water to whole parts of the plant. Biological control of moler diseases using fungal antagonist has developed considerably in recent years. Fungal antagonists play a significant role in controlling plant pathogens and moler diseases caused by Fusarium oxysporum, such as Trichoderma harzianum and Acremonium variecolor. A series of experiments in field conducted to assess the comparison of ability between Trichoderma harzianum strain MGQ2 and Acremonium variecolor strain 130360 in effectively controling moler in shallot.

Cyclopurpuracin is a cyclooctapeptide isolated from the methanol extract of Annona purpurea seeds with a sequence of cyclo-Gly-Phe-Ile-Gly-Ser-Pro-Val-Pro. In our previous study, the cyclisation of linear cyclopurpuracin was problematic; however, the reversed version was successfully cyclised even though the NMR spectra revealed the presence of a mixture of conformers. Herein, we report the successful synthesis of cyclopurpuracin using a combination of solid- and solution-phase synthetic methods. Initially, two precursors of cyclopurpuracin were prepared, precursor linear A (NH2-Gly-Phe-Ile-Gly-Ser(t-Bu)-Pro-Val-Pro-OH) and precursor linear B (NH-Pro-Gly-Phe-Ile-Gly-Ser(t-Bu)-Pro-Val-OH, and various coupling reagents and solvents were trialled to achieve successful synthesis. The final product was obtained when precursors A and B were cyclised using the PyBOP/NaCl method, resulting in a cyclic product with overall yields of 3.2% and 3.6%, respectively. The synthetic products were characterised by HR-ToF-MS, 1H-NMR, and 13C-NMR, showing similar NMR profiles to the isolated product from nature and no conformer mixture. The antimicrobial activity of cyclopurpuracin was also evaluated for the first time against S. aureus, E. coli, and C. albicans, showing weak activity with MIC values of 1000 µg/mL for both synthetic products, whereas the reversed cyclopurpuracin was more effective with an MIC of 500 µg/mL.

Chisocheton balansae C.DC., is one of the Meliaceae family plants which is the endemic plants from Soputan Mountain, North Sulawesi, Indonesia. This study was aimed to determine the chemical structure of flavan-3-ol compounds from ethyl acetate extract of C. balansae C.DC stembark. Dried powder of C. balansae C.DC stem bark was extracted consecutively with n-hexane, ethyl acetate, and methanol solvents. Four flavan-3-ol compounds, named catechin (1), epicatechin (2), epigallocatechin-3-O-gallate (3), and epicatechin-3-O-gallate (4) were successfully isolated from ethyl acetate extract. The chemical structure of these isolates was determined by spectroscopic methods (1H-NMR, 13C-NMR, DEPT, and 2D-NMR) and comparison with previous reported spectral data. These compounds are first time reported from this plant.

Three aromadendrane-type sesquiterpenoids, spathulenol (1), 4β,10α-dihydroxyaromadendrane (2), and 4α,10α-dihydroxyaromadendrane (3) were isolated from the stem bark of Aglaia harmsiana (Meliaceae). Compound 3 was isolated for the first time from Aglaia genus. The chemical structures of isolated compounds were elucidated by various spectroscopic methods, including one and two-dimensional NMR, as well as mass spectroscopy analysis. These sesquiterpenoids 1-3 were evaluated for their cytotoxic activity against MCF-7 breast cancer cell lines. The IC50 value of compound 1-3 were 31.65 ± 0.1, 8.41 ± 0.04 and 2.80 ± 0.02 µM, respectively. Among the aromadendrane-type sesquiterpenoids, compounds 2 and 3, which do not have a double bond, showed higher activity than compound 1. Whereas, compound 3 showed the strongest activity indicate that α configuration of hydroxyl group increases the cytotoxic activity.

Lansium domesticum Corr. is a member of the Meliaceae family that is widely spread in tropical and subtropical region of Asia and America. Traditionally, the fruit of this plant has been consumed because of its sweet taste. However, the fruit peels and the seeds of this plant have been rarely utilized. The previous chemical investigation of this plant showed the presence of secondary metabolites with many biological activities, including cytotoxic triterpenoid. Triterpenoids is a class of secondary metabolites which contain thirty carbon atoms in the main skeleton. The high modification of this type of compound, including the ring opening, highly oxygenated carbons, and the degradation of its carbon chain to give the nor-triterpenoid structure, is responsible for its cytotoxic activity. In this paper, we isolated and elucidated the chemical structure of two new onoceranoid triterpenes, kokosanolides E (1) and F (2), from the fruit peels of L. domesticum Corr., along with a new tetranortriterpenoid, kokosanolide G (3), from the seeds of L. domesticum Corr. The structural determination of compounds 1–3 was undertaken through FTIR spectroscopic analysis, 1D and 2D NMR, mass spectrometry, as well as through a comparison of the chemical shifts of the partial structures of compounds 1–3 with the literature data. The cytotoxic properties of compounds 1–3 were tested against MCF-7 breast cancer cells using the MTT assay. Moderate activity was shown by compounds 1 and 3, with IC50 values of 45.90 and 18.41 μg/mL, respectively, while compound 2 showed no activity (IC50 168.20 μg/mL). For the onoceranoid-type triterpene, the high symmetrical structure of compound 1 is presumably the reason for its better cytotoxic activity compared with that of compound 2. Compound 3 showed moderate activity, mainly because of the presence of the furan ring, which, based on the literature, gives better cytotoxic activity in a tetranortriterpenoid-type structure. The findings of three new triterpenoid compounds from L. domesticum indicate the significant value of this plant as a source of new compounds.

Resin glycoside is a type of secondary metabolite isolated commonly from the Convolvulaceae family. It consists of oligosaccharides conjugated to organic acids with a larger percentage having a macrocyclic structure. The resin glycosides reported in this review is classified mostly based on the number of sugar units constructing the structure, which is correlated to the biological properties of the compounds. According to preliminary reviews, the protocols to isolate the compounds are not straightforward and require a special technique. Additionally, the structural determination of the isolated compounds needs to minimize the structure for the elucidation to become easier. Even though resin glycosides have a complicated structural skeleton, several total syntheses of the compounds have been reported in articles published from 2010 to date. This review is an update on the prior studies of the resin glycosides reported in 2010 and 2017. The review includes the classification, isolation techniques, structural determination, biological properties, and total synthesis of the resin glycosides.

Protein is one of the essential macronutrients required by all living things. The breakdown of protein produces monomers known as amino acids. The concept of conjugating natural compounds with amino acids for therapeutic applications emerged from the fact that amino acids are important building blocks of life and are abundantly available; thus, a greater shift can result in structural modification, since amino acids contain a variety of sidechains. This review discusses the data available on amino acid–natural compound conjugates that were reported with respect to their backgrounds, the synthetic approach and their bioactivity. Several amino acid–natural compound conjugates have shown enhanced pharmacokinetic characteristics, including absorption and distribution properties, reduced toxicity and increased physiological effects. This approach could offer a potentially effective system of drug discovery that can enable the development of pharmacologically active and pharmacokinetically acceptable molecules.

Two new dammarane-type triterpenoid fatty acid ester derivatives, 3β-oleate-20S-hydroxydammar-24-en (1) and 3β-oleate-20S,24S-epoxy-25-hydroxydammarane (2) with a known dammarane-type triterpenoid compound, such as 20S-hydroxydammar-24-en-3-on (3), were isolated from the stem bark of Aglaiaelliptica (C.DC.) Blume. The chemical structures were determined by spectroscopic methods, including FTIR, NMR (one and two-dimensional), and HRESITOF-MS analysis, as well as chemical derivatization and comparison with previous literature. Furthermore, the synthetic analog resulting from transesterification of 1 and 2 also obtained 3β,20S-dihydroxy-dammar-24-en (4) and 20S,24S-epoxy-3β,25-dihydroxydammarane (5), respectively. The cytotoxic effect of all isolated and synthetic analog compounds was evaluated using PrestoBlue reagent against MCF-7 breast cancer cell and B16-F10 melanoma cell lines. The 20S-hydroxydammar-24-en-3-on (3) showed the strongest activity against MCF-7 breast cancer and B16-F10 melanoma cell, indicating that the ketone group at C-3 in 3 plays an essential role in the cytotoxicity of dammarane-type triterpenoid. On the other hand, compounds 1 and 2 had very weak cytotoxic activity against the two cell lines, indicating the presence of fatty acid, significantly decreasing cytotoxic activity. This showed the significance of the discovery to investigate the essential structural feature in dammarane-type triterpenoid, specifically for the future development of anticancer drugs.

In the course of our continuing search for anticancer compounds from Chisocheton species, three steroids, stigmast-5-en-3β-ol (1), stigmast-5-en-3β-ol-3-O-β-D-glucopyranoside (2) and stigmast-5,22-dien-3β-ol-3-O-β-D-glucopyranoside (3), were obtained from the stembark of Chisocheton celebicus. The structures of compound 1-3 were identified with spectroscopic data including IR, 1D-NMR, 2D-NMR and TOF-MS, as well as by comparing with those spectral data previously. Compounds 1-3, were evaluated for their cytotoxic effects against P-388 murine leukemia cells and displayed the cytotoxicity activity with IC50 values of 12.45 ± 0.050, 52.27 ± 0.031 and 62.52 ± 0.076 µg/mL, respectively.

Sesquiterpenoid is a class of terpenoid compounds that have the most abundant diversity of structures and biological activitiesthat can be found in natural resources. Tropical plants are main source of sesquiterpenoid compounds such as Aglaiagenus belong to Meliaceae family. A. harmsianais a species from Aglaiathat only has few previous researchs. Therefore, the purpose of this study was to isolate and determine the structure of sesquiterpenoid compounds from stem barkA. harmsianaalong withtheir cytotoxic activity against MCF-7 breast cancer cells. The isolation process begins by extracting powder from A. harmsianastembark using n-hexane, ethyl acetate and methanol. All extracts were evaluated for their cytotoxic activity against MCF-7 breast cancer cells, and n-hexane extractsshowed significant cytotoxic activitywith IC50values of 117.86 µg/mL. Therefore, n-hexane extracts were further separated and purified by various chromatographic techniquesto obtain compounds 1and 2. Compounds 1and 2were elucidated their chemical structures by spectroscopic methods includingIR, NMR, and MS as well as bycomparison of data with literatures and identified ascaryophyllene-typesesquiterpenoids, β-caryophylleneoxide (1) and senecrassidiol (2). Compounds 1and 2were submitted for cytotoxic eveluation on MCF-7 breast cancer cells and as a result, β-caryophyllene oxide (1) showed the stronger activity compared to senecrassidiol (2). These finding indicatedthat the cytotoxic activity of caryophyllene-typesesquiterpenoid areinfluenced by the presence of double bonds and configuration of methyl groups.

[Leu]6-aureobasidin K 1 is an analog of AbK, in which aIle residue at the sixth position of AbK is replaced by Leu. [Leu]6-AbK 1 was successfully synthesized using a combination of solid- and solution-phase that has a difference with the previous method applied for the synthesis of other aureobasidin analogues, [2S,3S-Hmp]-AbL 2. The linear precursor was prepared by using solid-phase method with Fmoc strategy, in which an ester bond was constructed on the resin. The cyclic product was obtained from a solution-phase reaction of the linear precursor using HATU reagent. The crude of the cyclic peptide was purified using a column chromatography technique to obtain [Leu]6-AbK 1 with a percent yield of 5.31% that is higher compared to previous similar method for synthesis of [2S,3S-Hmp]-AbL 2. The purity level of cyclic product was analyzed using analytical RP-HPLC (tR = 6.49 min) and analysis of structure using 1H-NMR.

Peptida antioksidan merupakan kelompok peptida yang berperan penting karena dapat menetralkan radikal bebas, sehingga dapat mencegah dan mengobati penyakit kronis. Salah satu senyawa peptida antioksidan alami yang telah ditemukan peneliti sebelumnya adalah senyawa tetrapeptida PAGY (Pro-Ala-Gly-Tyr) yang diisolasi dari gelatin kulit ikan amur sturgeon (Acipenser schrenckii) dan dilaporkan memiliki aktivitas antioksidannya dengan IC50 5.38 mg/mL dalam uji DPPH dan 0,008 mg/mL dalam uji ABTS berturut-turut. Kelompok kami telah berhasil mensintesis PAGY bersama dengan analognya yakni PSGY, PFFY, PAFY, dan PAIY dengan menggunakan metode sintesis peptida fase padat (SPPS). Pengujian aktivitas antioksidan pada senyawa hasil sintesis menunjukkan bahwa PSGY memiliki aktivitas antioksidan lebih baik dari PAGY. Pencarian analog tetrapeptida antioksidan yang lebih baik hingga saat ini masih terus dilakukan. Pada penelitian ini telah berhasil disintesis analog tetrapeptida lainnya PADY (Pro-Ala-Asp-Tyr) dengan metode sintesis peptida fase padat menggunakan strategi Fmoc/t-Bu pada resin 2-klorotritilklorida dilanjutkan dengan pengujian aktivitas antioksidannya. HATU/HOAT digunakan sebagai reagen pengkopling dalam sintesis PADY. Pemurnian krud PADY dilakukan menggunakan RP-HPLC preparatif sehingga diperoleh PADY murni seberat 14.7 mg (12.6%). Penentuan struktur peptida hasil sintesis dianalisis dengan menggunakan spektroskopi 1H-NMR dan TOF-MS. Pada pengujian aktivitas antioksidan dengan metode DPPH, PADY hasil sintesis memberikan nilai IC50 sebesar 1.850 mg/mL, yang mengindikasikan bahwa PADY menunjukkan aktivitas antioksidan yang lebih rendah daripada PAGY hasil sintesis peneliti sebelumnya. Antioxidant peptide is a class of peptides that play an important in neutralizing free radicals, therefore this compound can be used to prevent and treat chronic diseases. One of the natural antioxidant peptides reported by previous researcher is PAGY (Pro-Ala-Gly-Tyr), which is isolated from amur sturgeon fish (Acipenser schrenckii) gelatin that showed antioxidant activity with IC50 5.38 and 0.008 mg/mL using DPPH and ABTS assay, respectively. Our group has successfully synthesized PAGY, along with its analogues of PSGY, PFFY, PAFY, and PAIY using solid phase peptide synthesis method (SPPS). Antioxidant assay on synthesised compounds showed that PSGY has better antioxidant activity than PAGY. The search on the analogues of the antioxidant tetrapeptide was continued. From this study, a tetrapeptide analogue PADY (Pro-Ala-Asp-Tyr) has been successfully synthesised by solid phase peptide synthesis method with Fmoc/t-Bu strategy on 2-chlorotrityl chloride resin and tested for its antioxidant activity. HATU and HOAt reagents were used as the coupling reagent for the synthesis of PADY. The resulting PADY peptide solid was then purified using preparative RP-HPLC yielding PADY of 14.7 mg (12.6%). Characterisation of the synthesized compound was analysed by 1H-NMR and TOF-MS. On the antioxidant assay using DPPH method, PADY showed IC50 value of 1.850 mg/mLindicating a lower activity than the synthetic PAGY. Cyclic peptides have been widely reported to have therapeutic abilities in the treatment of cancer. This has been proven through in vitro and in vivo studies against breast, lung, liver, colon, and prostate cancers, among others. The multitude of data available in the literature supports the potential of cyclic peptides as anticancer agents. This review summarizes the findings from previously reported studies and discusses the different cyclic peptide compounds, the sources, and their modes of action as anticancer agents. The prospects and future of cyclic peptides will also be described to give an overview on the direction of cyclic peptide development for clinical applications.

Antimicrobial peptides (AMPs) are interesting compounds owing to their ability to kill several pathogens. In order to identify new AMPs, c-PLAI analogues were synthesized and evaluated together with their linear precursors for their antimicrobial properties against two Gram-positive bacteria (Staphylococcus aureus and Bacillus cereus), two Gram-negative bacteria (Escherichia coli and Klebsiella pneumoniae), and two fungal strains (Candida albicans and Trichophyton mentagrophytes). The new c-PLAI analogues were prepared through a combination of solid- and solution-phase syntheses, as previously employed for the synthesis of c-PLAI. The antimicrobial activity tests showed that the synthetic parent peptide c-PLAI was inactive or weakly active towards the bioindicators employed in the assay. The tests also indicated that cyclic c-PLAI analogues possessed enhanced antimicrobial properties against most of the bacteria and fungi tested. Furthermore, this study revealed that analogues containing cationic lysine residues displayed the highest activity towards most bioindicators. A combination of lysine and aromatic residues yielded analogues with broad-spectrum antimicrobial properties.

A new onoceranoid triterpenes, namely 3-hydroxy-8,14-secogammacera-7,14-dien-21-one (1), has been isolated from the fruit peels of Lansium domesticum Corr. cv kokossan. The structure of 1 was determined on the basis of spectroscopic data including infrared, 1D and 2D-NMR, as well as high resolution mass spectroscopy analysis. Compound 1 showed a weak activity against MCF-7 breast cancer cell lines.

Crop cabbage worm (Crocidolomia pavonana) is one of the pests that has a bad effect on the cabbage production in Indonesia. One of the efforts that has been applied to control this pest is by using bayrusil. However, the bayrusil can cause negative effects such as poisoning and environmentally polluting effects. To prevent the undesired effects, it has been developed the use of environmentally friendly insecticide and one of them is the use of insecticidal peptide. Tripsin-modulating oostatic factor (TMOF) and ist analogues is one of the peptides that has an insecticidal activity. The aim of this research is to synthesise a linear tetrapeptide (DPAP) an analogues of TMOF and to test insecticidal activities against crop cabbage worm. A DPAP has been synthesised by Solid Phase Peptide Synthesis on 2-chlorotrytil chloride resin. Fmoc strategy was applied on the synthesis and a combination DIC/Oxime was employed in the coupling reaction.The tetrapeptidyl resin was cleaved by TFA:water:EDT (90:5:5). The peptide was purified by reverse-phase column chromatography and characterized by TOF ES-MS spectroscopy. A linear peptide was biologically tested towards the C. pavonana, showing percentage of mortality values at 1000 ppm.

A new propylcholesterol-type steroid, namely (22E,24S)-24-propylcholest-5en-3α-acetate (1), has been isolated from the stembark of Aglaia angustifolia (Miq.). The structure of 1 was determined on the basis of spectroscopic data including 1D- and 2D-NMR as well as high resolution mass spectroscopy analysis. Compound 1 showed weak activity against the MCF-7 breast cancer cell line.

Two dammarane-type triterpenoids, 20S,24S-epoxy-3α,25-dihydroxydammarane (1) and 3α-acetyl-20S,24S-epoxy-3α,25-dihydroxydammarane (2), have been isolated from the stembark of Aglaia argentea. The chemical structure of compounds (1 and 2) were identified by spectroscopic evidences including UV, IR, 1D-NMR, 2D-NMR and MS as well as by comparing with previously reported spectral data. Those compounds were isolated from this plant for first time. Compounds (1 and 2) showed cytotoxic activity against P-388 murine leukemia cells with IC50 values of 23.96 and 8.14 mM, respectively. Four dammarane-type triterpenoid compounds, dammar-24-en-3a-ol (1), 3-epi-cabraleahydroxy lactone (2), (E)-25-hydroperoxydammar-23-en-3b,20-diol (3), and dammar-24-en-3β,20-diol (4), were isolated from the methanolic extract of the stembark of Aglaia argentea. Compounds 1-4 were isolated for first time from this plant. The structure of isolated compounds were elucidated by spectroscopic methods including one and two-dimensional NMR as well as mass spectrometric analysis. Compounds, 1-4, along with a known synthetic analog, 20-hydroxy-dammar-24-en-3-on (5), were evaluated their cytotoxic activity against P-388 murine leukemia cells in vitro. The IC50 values of compounds, 1-5 were 9.09 ± 0.10, 68.53 ± 0.08, 5.89 ± 0.08, 22.40 ± 0.11, and 11.53 ± 0.08 µg/mL, respectively. Among the dammarane-type triterpenoids, compounds 1, 3, 4 and 5 having opened side chain showed the stronger activity, wheres compound 2 with cyclic side chain showed weak or no activity. In addition, compound 3 showed strongest activity, indicate that hydroperoxy group at side chain increase cytotoxic activity.

Two mixtures of flavanoid compounds (1 and 2), a mixture of catechin (1a) and epicatechin (1b), and a mixture of gallocatechin (2a) and epigallocatechin (2b), were isolated from the active fraction of the stem bark of Aglaia elliptica methanol extract. The chemical structure of the compounds was identified with spectroscopic data, including UV, IR, NMR (1H, 13C, DEPT 135°, HMQC, HMBC, 1H-1H COSY), and MS, and additionally compared with previously reported spectral data. All compounds were evaluated for their cytotoxic effects against P-388 murine leukemia cells. Compound 2 showed cytotoxicity against the P-388 murine leukemia cell, with an IC50 value of 7.79 µg/mL, but compound 1 was found not to be active (more than 100 µg/mL).

The Aglaia genus belong to Meliceae family is unique plant species because the presence of rocaglate and rocaglamide which is so far isolated only from Aglaia genus, indicate that type of this compound as a chemical marker for the genus of Aglaia. This type of compound known to have strong activity, such as insecticide and cytotoxic. This study describe the isolation, structure elucidation, and cytotoxic activity of an isolated rocaglate compound. Dried stembark of A. argentea extracted with methanol and partition between n-hexane, ethyl acetate, and n-butanol, respectively The extracts were tested against P-388 murine leukemia cells and the ethyl acetat showed strongest activity with IC50 value of 15.5 mg/mL. The ethyl acetate then was separated and purified with chromatography technique to obtain isolated compound 1. The chemical structure of isolated compounds were elucidated by spectroscopic methods including one and two-dimensional NMR as well as high-resolution mass spectrometric analysis and identified as a methyl rocaglate. Compound 1 showed strong cytotoxic activity with an IC50 value of < 0.1 μg/mL.

Three cytotoxic steroids, stigmasterol (1), stigmast-5-en-3b-ol (2) and b-sitosterol-3-O-acetate (3) were isolated from the stem bark of Chisocheton cumingianus. The chemical structures of those compounds were identified based on spectroscopic data and by comparison with those data previously reported. All of the compounds isolated were evaluated for their cytotoxic effects against P-388 murine leukemia cells in vitro. Compounds 1-3 showed cytotoxicity activity against P-388 murine leukemia cells with IC50values of 12.4, 60.8, and ˃ 100 mg/mL, respectively.

Tetrapeptide, OH-Pro-Ala-Gly-Tyr-NH2, was successfully synthesised on 2-chlorotrityl chloride resin by taking advantage of PyBOP (benzotriazol-1-yl-oxytripyrrolidinophosphonium hexafluorophosphate) as coupling reagent. The selection of the peptide as the target of synthesis was due to its interesting bioactivity as antioxidant. The synthesis was undertaken with Fmoc strategy, where Fmoc-proline was added onto the resin at the first place. It is known from the literature that proline can resist from rasemisation when it was attached on the resin at the first time. Fmoc deprotection step was carried out by employing 20% piperidine in DMF and the reaction mixture was shaken for 30 minutes. Once the proline attached, the next step was to attach amino acids, alanine (Ala), glycine (Gly) and tyrosine(Tyr), subsequently onto the resin until tetrapeptidyl resin was constructed on the resin. Hydroxyl group of Tyr was protected with t-butyl, which is TFA-labiled. Coupling reaction was undertaken by mixing the amino acid and PyBOP in a mixture of dichloromethane and DMF (1:1) and in the presence of basic DIPEA. Resin cleavage step was carried out by using 95% TFA in water, where t-butyl protecting group on the side chain of Tyr was cleaved at the same time. The analytical RP-HPLC of the final product showed a single peak at 21.9 minutes (20-90% of acetonitrile in water with 0.1% of TFA during 30 minutes), indicating that each coupling has given a good coupling performance and resulting in a pure product. A tetrapeptide, an analog of trypsin-modulating oostatic factor (TMOF), has been synthesised by using a method of solid-phase peptide synthesis on 2-chlorotrityl chloride resin. Fmoc strategy was applied on the synthesis. The formation of peptide bond was facilitated by DIC/oxime as coupling reagent. After all of amino acids were attached on the resin, tetrapeptidyl resin was added by a mixture of TFA:water:EDT(90:5:5) in dichloromethane to cleave the peptide.Crude peptide was purified by reverse-phase column chromatography and the purified peptide was analysed by TOF ES-MS spectroscopy.

Kalanchoe is the biggest genera of Crassulaceae family and distributed in tropical and subtropical regions. This genera is found to be a rich source of biologically active natural products such as triterpenes, flavonoids and steroids. As a part of our continuing search for novel insecticidal compounds from Indonesian Kalanchoe plants, we examined Kalanchoe daigremontiana collected from Bandung region, West Java, Indonesia. The methanolic extract of the dried leaves of K. daigremontiana was concentrated and extracted with methylene chloride The methylene chloride extract exhibited an insecticidal activity toward silkworms. The methylene chloride extract was partitioned between n-hexane and methanol containing 10% water. The active lower layer was extracted with ethyl acetate. By using the insecticidal activity to follow the separations, the ethyl acetate fraction was separated by combination of column chromatography on Kieselgel 60 and ODS to afford two insecticidal bufadienolides 1 and 2. The structures of these compounds were elucidated based on spectroscopic analysis (UV, IR, NMR, 2D-NMR) and comparison with those related data previously reported. In this paper, the isolation, structural elucidation, and insecticidal activities against the third instar larvae of silkworm will be described.
