Biodegradation, particularly via enzymatic degradation, has emerged as an efficient and eco-friendly solution for Poly (ethylene terephthalate) (PET) pollution. The production of PET hydrolases plays a role in the large-scale enzymatic degradation. However, an effective variant, 4Mz, derived from Thermobifida fusca cutinase (Tfu_0883), was previously associated with a significant reduction in yield when compared to the wild-type enzyme. In this study, a novel cell membrane protection strategy was developed to enhance the yield of 4Mz. This approach increased the yield of 4Mz by 18.2-fold from shaken flasks to 3-L bioreactors, reaching a yield of 3.1 g·L -1 , the highest yield of a PET hydrolase described thus far. In addition, the raw culture broth from 4Mz was applied directly for the enzymatic degradation of PET bottles, achieving a 91.2 % degradation rate. Background: Breast cancer is one of the leading causes of cancer deaths in women. Early diagnosis offers the best hope for a cure. Ductal carcinoma in situ is considered a precursor of invasive ductal carcinoma of the breast. In this study, we carried out microRNA sequencing from 7 ductal carcinoma in situ (DCIS), 6 infiltrating ductal carcinomas (IDC Stage IIA) with paired normal, and 5 unpaired normal breast tissue samples., Methods: We have deployed miRge for microRNA analysis, DESeq for differential expression analysis, and Cytoscape for competing endogenous RNA network investigation., Results: Here, we identified 76 miRNAs that were differentially expressed in DCIS and IDC. Additionally, we provide preliminary evidence of miR-365b-3p and miR-7-1-3p being overexpressed, and miR-6507-5p, miR-487b-3p, and miR-654-3p being downregulated in DCIS relative to normal breast tissue. We also identified a miRNA miR-766-3p that was overexpressed in earlystage IDCs. The overexpression of miR-301a-3p in DCIS and IDC was confirmed in 32 independent breast cancer tissue samples., Conclusion: Higher expression of miR-301a-3p is associated with poor overall survival in The Cancer Genome Atlas Breast Cancer (TCGA-BRCA) dataset, indicating that it may be associated with DCIS at high risk of progressing to IDC and warrants deeper investigation., (Copyright© Bentham Science Publishers; For any queries, please email at epub@benthamscience.net.)

Poly (ethylene terephthalate) (PET) is a widely used type of general plastic that produces a significant amount of waste due to its non-degradable properties. We propose a novel directional-path modification (DPM) strategy, involving positive charge amino acid introduction and binding groove remodeling, and apply it to Thermobifida fusca cutinase to enhance PET degradation. The highest value of PET degradation (90%) was achieved in variant 4Mz (H184S/Q92G/F209I/I213K), exhibiting values almost 30-fold that of the wild-type. We employed molecular docking, molecular dynamics simulations, and QM/MM MD for the degradation process of PET, accompanied by acylation and deacylation. We found that the distance of nucleophilic attack was reduced from about 4.6 Å in the wild type to 3.8 Å in 4Mz, and the free energy barrier of 4Mz dropped from 14.3 kcal/mol to 7.1 kcal/mol at the acylation which was the rate-limiting step. Subsequently, the high efficiency and universality of the DPM strategy were successfully demonstrated in LCC, Est119, and BhrPETase enhancing the degradation activity of PET. Finally, the highest degradation rate of the pretreated commercial plastic bottles had reached to 73%. Density functional theory (DFT) calculations are carried out to investigate the effect of point defects on acetylene hydrogenation reaction over Ni(111) surface with three different defect concentrations (DC = 0.0500, 0.0625, and 0.0833), compared with the perfect Ni(111) surface. The adsorptions of C 2 species and H atoms and the mechanism of acetylene hydrogenation via the ethylene pathway are systematically analyzed. The results indicate that the existence of defects will make C 2 species and H atoms more inclined to adsorb near the defects. Introducing an appropriate amount of point defect concentration can enhance the catalytic activity and ethylene selectivity of Ni. In this work, DC = 0.0625 Ni(111) surface has the highest catalytic activity and selectivity of ethylene. This work provides useful theoretical information on the effect of defects on acetylene hydrogenation and is helpful for the design of Ni and related metal catalysts with defects.

We examined the effects of phosphorus (P) levels on photosynthetic and P/Fe traits of soybean under the stress of low Fe and their genotypic differences, to provide a theoretical basis for rational application of P and Fe fertilizer. Six P-efficient and six P-inefficient soybean varieties screened in the early stage were used as experimental materials. Four treatments of P:Fe ratio were set, including 0:30, 30:30, 150:30 and 300:30 (μmol·L -1 ). We measured chlorophyll fluorescence traits and P-Fe utilization efficiency in soybean. A stepwise regression equation was established with seed weight per plant. Pathway analysis was performed, with the response of P-efficient and P-inefficient soybean genotypes to different P:Fe treatments being comprehensively evaluated by factor scores. The results showed significant main and interactive effects of genotype and P:Fe on the relative electron transfer rate of photosystem Ⅱ (ETR) at beginning of flowering stage (R1), the proportion of the energy absorbed by photosystem Ⅱ dissipated into heat (NPQ) at R1 stage, and proportion of energy absorbed by photosystem Ⅱ devoted to the photochemical reaction ( q L) at R1 stage. Results of canonical correlation analysis showed a negative correlation between P utilization efficiency of seed at full maturity stage (R8) and photosynthetic rate at R1 stage of P-efficient genotypes. Seed Fe utilization efficiency of P-inefficient genotypes at R8 stage was positively correlated with NPQ at R1 stage, but negatively correlated with q L at R1 stage. The actual photochemical efficiency of PSⅡ ( Φ PSⅡ ) at R1 stage was negatively correlated with P-efficient genotypes, but positively correlated with P-inefficient genotypes, which indicated that Φ PSⅡ at R1 stage was an important indicator for identifying soybean genotypes with different P efficiency under stress of low Fe. The comprehensive performance of P-efficient soybean genotypes decreased first and then increased with P level, while P-inefficient soybean genotypes increased first and then decreased. The inflection point of both genotypes appeared in P:Fe of 30:30. Thus, P:Fe ratio of 30:30 could be used as a threshold to identify soybean genotypes with different P efficiency under stress of low Fe. In conclusion, P fertilizer application should be equal to or greater than 1:1 (P:Fe) when planting P-efficient soybean genotypes in low Fe area, while P fertilizer application should not exceed 1:1 (P:Fe) when planting P-inefficient soybean genotypes.

Background: Sorghum , the C4 dry-land cereal, important for food, fodder, feed and fuel, is a model crop for abiotic stress tolerance with smaller genome size, genetic diversity, and bio-energy traits. The heat shock proteins/chaperonin 60s (HSP60/Cpn60s) assist the plastid proteins, and participate in the folding and aggregation of proteins. However, the functions of HSP60s in abiotic stress tolerance in Sorghum remain unclear., Methods: Genome-wide screening and in silico characterization of SbHSP60s were carried out along with tissue and stress-specific expression analysis., Results: A total of 36 HSP60 genes were identified in Sorghum bicolor . They were subdivided into 2 groups, the HSP60 and HSP10 co-chaperonins encoded by 30 and 6 genes, respectively. The genes are distributed on all the chromosomes, chromosome 1 being the hot spot with 9 genes. All the HSP60s were found hydrophilic and highly unstable. The HSP60 genes showed a large number of introns, the majority of them with more than 10. Among the 12 paralogs, only 1 was tandem and the remaining 11 segmental, indicating their role in the expansion of SbHSP60s . Majority of the SbHSP60 genes expressed uniformly in leaf while a moderate expression was observed in the root tissues, with the highest expression displayed by SbHSP60-1 . From expression analysis, SbHSP60-3 for drought, SbHSP60-9 for salt, SbHSP60-9 and 24 for heat and SbHSP60-3, 9 and SbHSP10-2 have been found implicated for cold stress tolerance and appeared as the key regulatory genes., Conclusion: This work paves the way for the utilization of chaperonin family genes for achieving abiotic stress tolerance in plants., (© 2021 Bentham Science Publishers.)

Background: Production of biofuels from lignocellulosic crop biomass is an alternative to reduce greenhouse gas emissions. The biofuel production involves collecting biomass, breaking down cell wall components followed by the conversion of sugars to ethanol. The lingo-cellulosic biomass comprises 40-50% cellulose, 20-30% hemicellulose, and 10-25% lignin. Sorghum is a widely adapted energy crop for biofuel production. Biomass with low lignin, high cellulose, and high hemicellulose contents are exploited to attain maximum biofuel production efficiency. Resistance to lodging, pest, disease, and abiotic stresses related to cell wall components is well documented, and quantitative trait loci were identified to understand these traits' genetic correlation. Selection for reduced lignin and increased cellulose content in stover can increase the ethanol yield. The Genome-Wide Association Studies (GWAS) is a complementary approach to evaluating the marker and phenotype associations among large diversity panels. Single nucleotide polymorphisms were scanned to identify loci associated with the traits of interest. In this study, the GWAS was performed on 245 sorghum minicore genotypes to analyze agronomic traits (days to 50%flowering, fresh biomass yield, dry biomass yield) and cell wall components (cellulose, hemicellulose, and lignin). Further, in-silico validation of the candidate genes was performed in a global gene expression data from large-scale RNA sequencing studies in sorghum available in the NCBI GEO database was used., Objective: The objectives of this study are to evaluate native variations in biofuel related agronomic traits and stalk cell wall components and to identify significant SNPs or loci related to the cell wall components., Methods: In this article, an association mapping panel, comprising of 245 sorghum minicore germplasm accessions, was evaluated during two post rainy seasons of 2013 and 2014, and observations were recorded on the whole plot- for days to 50% flowering, fresh biomass yield (tha -1 , and dry biomass yield (tha -1 ). The biomass of sun-dried plants from both seasons was collected separately, chopped, dried, and ground to powder. The cellulose, hemicellulose, and lignin contents were determined in the powdered. The content of each of these three components in sorghum was expressed in percent of dry matter. The data on agronomic traits and composition analysis was subjected to Analysis of Variance. For the current study, we remapped the raw GBS data with the sorghum assembly version v3.1. A total of 27,589 SNPs were obtained with a minor allele frequency (MAF) >1% and missing data <50%. The GWAS was performed in a single minicore population using FarmCPU, in R software. The synteny positions of the identified significant SNPs between sorghum and other model crop species viz., maize, switchgrass, and Arabidopsis were represented using CIRCOS software for traits viz., dry biomass yield, cellulose, hemicellulose, and lignin. The transcriptome dataset from where sorghum gene atlas studies of grain, sweet, and bioenergy sorghums are available through NCBI's Gene Expression Omnibus (GEO) under accession number GSE49879, was used to cross-validate the identified SNPs for cellulose, hemicellulose, and lignin through GWAS., Results: High broad-sense heritability was exhibited for all the traits in individual seasons along with significant genotype × environment interaction across seasons except lignin. Association mapping with a P < 1×10 -4 revealed genomic regions associated with the- (i) agronomic traits (days to 50% flowering, fresh and dry biomass), and (ii) biochemical traits (cellulose, hemicellulose, and lignin) associated with biofuels production, in individual seasons. Twelve significant SNPs for flowering time, 30 fresh biomass yields, and 24 for dry biomass yield, 25 for cellulose, 7 for hemicellulose, and 21 for lignin were identified. CIRCOS plot was constructed to identify and analyze similarities and differences while comparing the sorghum genome with different crops. For cellulose high similarity of >80% was observed for all sorghum gene sequences with the maize homologs. The overall similarity of sorghum homologs with foxtail millet was >65%, for Arabidopsis from 30.6% to 48.6%, and rice from 28.2% to 92.8%. SNPs for hemicellulose displayed maximum similarity to foxtail millet followed by maize. The sequence similarity of lignin SNPs in sorghum was highest with the maize genome followed by Arabidopsis. Both rice and foxtail millet showed >55% similarity to the sorghum genome., Conclusion: This study reports large variability for agronomic and biofuel traits in the sorghum minicore collection with high heritability. The genetic architecture of cell wall components using the GWAS approach was studied and candidate genes for each component were annotated. These results give a better understanding of the genetic basis of the sorghum cell wall composition. Invasive lobular carcinoma of the breast (ILC) is strongly estrogen-driven and represents a unique context for estrogen receptor (ER) signaling. In ILC, ER controls the expression of the Wnt ligand WNT4, which is critical for endocrine response and anti-estrogen resistance. However, signaling mediated by WNT4 is cell type- and tissue-specific, and has not been explored in ILC. We utilized reverse phase protein array (RPPA) to characterize ER and WNT4-driven signaling in ILC cells and identified that WNT4 mediates downstream mTOR signaling via phosphorylation of S6 Kinase. Additionally, ER and WNT4 control levels of MCL-1, which is associated with regulation of mitochondrial function. In this context, WNT4 knockdown led to decreased ATP production and increased mitochondrial fragmentation. WNT4 regulation of both mTOR signaling and MCL-1 were also observed in anti-estrogen resistant models of ILC. We identified that high WNT4 expression is associated with similar mTOR pathway activation in ILC and serous ovarian cancer tumors, suggesting that WNT4 signaling is active in multiple tumor types. The identified downstream pathways offer insight into WNT4 signaling and represent potential targets to overcome anti-estrogen resistance for patients with ILC.

Background: Alternative medicine is available for those diseases which cannot be treated by conventional medicine. Ayurveda and herbal medicines are important alternative methods in which the treatment is done with extracts of different medicinal plants. This work is concerned with the evaluation of anti-stress bioactive compounds from the ethanolic root extract of Hemidesmus indicus., Methods: Gas chromatography and Mass Spectrum studies are used to identify the compounds present in the ethanolic extract based on the retention time, area. In order to perform docking studies, Vasopressin model is generated using modeling by Modeller 9v7. Vasopressin structure is developed based on the crystal structure of neurophysin-oxytocin from Bos taurus (PDB ID: 1NPO&#95;A) collected from the PDB data bank. Using molecular dynamics simulation methods, the final predicted structure is obtained and further analyzed by verifying 3D and PROCHECK programs, confirmed that the final model is reliable. The identified compounds are docked to vasopressin for the prediction of anti-stress activity using GOLD 3.0.1 software., Results: The predicted model of Vasopressin structure is stabilized and confirmed that it is a reliable structure for docking studies. The results indicated ARG4, THR7, ASP9, ASP26, ALA32, ALA 80 in Vasopressin are important determinant residues in binding as they have strong hydrogen bonding with phytocompounds. Among the 21 phytocompounds identified and docked, molecule Deoxiinositol, pentakis- O-(trimethylsilyl) showed the best docking results with Vasopressin., Conclusion: The identified compounds were used for anti-stress activity by insilico method with Vasopressin which plays an important role in causing stress and hence selected for inhibitory studies with phytocompounds. The phytocompounds are inhibiting vasopressin through hydrogen bodings and are important in protein-ligand interactions. The small heat shock proteins (sHsps/Hsp20s) are the molecular chaperones that maintain proper folding, trafficking and disaggregation of proteins under diverse abiotic stress conditions. In the present investigation, a genome-wide scan revealed the presence of a total of 47 sHsps in Sorghum bicolor (SbsHsps), distributed across 10 subfamilies, the major subfamily being P (plastid) group with 17 genes. Chromosomes 1 and 3 appear as the hot spot regions for SbsHsps, and majority of them were found acidic, hydrophilic, unstable and intron less. Interestingly, promoter analysis indicated that they are associated with both biotic and abiotic stresses, as well as plant development. Sorghum sHsps exhibited 15 paralogous and 20 orthologous duplications. Expression analysis of 15 genes selected from different subfamilies showed high transcript levels in roots and leaves implying that they are likely to participate in the developmental processes. SbsHsp genes were highly induced by diverse abiotic stresses inferring their critical role in mediating the environmental stress responses. Gene expression data revealed that SbsHsp-02 is a candidate gene expressed in all the tissues under varied stress conditions tested. Porcupine O- acyltransferase (PORCN) is considered essential for Wnt secretion and signaling. However, we observed that PORCN inhibition does not phenocopy the effects of WNT4 knockdown in WNT4-dependent breast cancer cells. This suggests a unique relationship between PORCN and WNT4 signaling. To examine the role of PORCN in WNT4 signaling, here we overexpressed WNT4 or WNT3A in breast cancer, ovarian cancer, and fibrosarcoma cell lines. Conditioned media from these lines and co-culture systems were used to assess the dependence of Wnt secretion and activity on the critical Wnt secretion proteins PORCN and Wnt ligand secretion (WLS) mediator. We observed that WLS is universally required for Wnt secretion and paracrine signaling. In contrast, the dependence of WNT3A secretion and activity on PORCN varied across the cell lines, and WNT4 secretion was PORCN-independent in all models. Surprisingly, WNT4 did not exhibit paracrine activity in any tested context. Absent the expected paracrine activity of secreted WNT4, we identified cell-autonomous Wnt signaling activation by WNT4 and WNT3A, independent of PORCN or Wnt secretion. The PORCN-independent, cell-autonomous Wnt signaling demonstrated here may be critical in WNT4-driven cellular contexts or in those that are considered to have dysfunctional Wnt signaling., (© 2019 Rao et al.)

A wide range of research has promised new tools for forecasting infectious disease dynamics, but little of that research is currently being applied in practice, because tools do not address key public health needs, do not produce probabilistic forecasts, have not been evaluated on external data, or do not provide sufficient forecast skill to be useful. We developed an open collaborative forecasting challenge to assess probabilistic forecasts for seasonal epidemics of dengue, a major global public health problem. Sixteen teams used a variety of methods and data to generate forecasts for 3 epidemiological targets (peak incidence, the week of the peak, and total incidence) over 8 dengue seasons in Iquitos, Peru and San Juan, Puerto Rico. Forecast skill was highly variable across teams and targets. While numerous forecasts showed high skill for midseason situational awareness, early season skill was low, and skill was generally lowest for high incidence seasons, those for which forecasts would be most valuable. A comparison of modeling approaches revealed that average forecast skill was lower for models including biologically meaningful data and mechanisms and that both multimodel and multiteam ensemble forecasts consistently outperformed individual model forecasts. Leveraging these insights, data, and the forecasting framework will be critical to improve forecast skill and the application of forecasts in real time for epidemic preparedness and response. Over the past two decades, colloidal particles with a variety of shapes, sizes, and compositions have been synthesized and characterized successfully. One of the most important applications for colloidal building blocks is to engineer functional structures as mechanical, electrical, and optical metamaterials. However, complex interaction dynamics between the building blocks as well as sophisticated structure-property relationships make it challenging to design structures with predictable target properties. In this paper, we implement an inverse material design framework using Genetic Algorithm (GA)-based techniques to streamline the design of colloidal structures based on target properties. We investigate spherical particles as well as colloidal molecules of different sizes and shapes and evaluate a Geometric Landscape Accessibility parameter that identifies the size of feasible domains within the geometric phase space of each structure. Considering target photonic properties, our GA-assisted framework is further utilized to identify sets of building blocks and structures that lead to various target values for the size of the photonic band gaps. The proposed framework in this study will provide new insight for predictive computational material design approaches and help establish more efficient ways of understanding structure-property relations in sub-micrometer-scale materials.

Background: Although cigarette smoking (CS) is by far the most important risk factor of chronic obstructive pulmonary disease (COPD), repeated and sustained infections are clearly linked to disease pathogenesis and are responsible for acute inflammatory flares (i.e. COPD exacerbations). We have previously identified Fatty Acid Binding Protein 5 (FABP5) as an important anti-inflammatory protein in primary airway epithelial cells., Results: In this study we found decreased FABP5 mRNA and protein levels in peripheral blood mononuclear cells (PBMCs) of COPD patients, especially among those who reported episodes of COPD exacerbations. Using wildtype (WT) and FABP5 -/- mice, we examined the effects of FABP5 on CS and infection-induced inflammatory responses. Similarly to what we saw in airway epithelial cells, infection increased FABP5 expression while CS decreased FABP5 expression in mouse lung tissues. CS-exposed and P. aeruginosa-infected FABP5 -/- mice had significantly increased inflammation as shown by increased lung histopathological score, cell infiltration and inflammatory cytokine levels. Restoration of FABP5 in alveolar macrophages using a lentiviral approach attenuated the CS- and bacteria-induced pulmonary inflammation. And finally, while P. aeruginosa infection increased PPARγ activity, CS or FABP5 knockdown greatly reduced PPARγ activity., Conclusions: These findings support a model in which CS-induced FABP5 inhibition contributes to increased inflammation in COPD exacerbations. It is interesting to speculate that the increased inflammation is a result of decreased PPARγ activity.

Radiodermatitis is a painful side effect for cancer patients undergoing radiotherapy. Irradiation of the skin causes inflammation and breakdown of the epidermis and can lead to significant morbidity and mortality in severe cases, as seen in exposure from accidents or weapons such as "dirty bombs" and ultimately leads to tissue fibrosis. However, the pathogenesis of radiodermatitis is not fully understood. Using a mouse model of radiodermatitis, we showed that the Transient Receptor Potential Melastatin 2 (TRPM2) ion channel plays a significant role in the development of dermatitis following exposure to ionizing radiation. Irradiated TRPM2-deficient mice developed less inflammation, fewer severe skin lesions and decreased fibrosis when compared to wild type mice. The TRPM2-deficient mice also showed a faster recovery period as seen by their increased weight gain post irradiation. Finally, TRPM2-deficient mice exhibited lower systemic inflammation with a reduction in inflammatory cytokines present in the serum. These findings suggest that TRPM2 may be a potential therapeutic target for reducing the severity of radiodermatitis.

Recently, Ni-Sn intermetallic compounds (IMCs) with unique geometric structures have been proved to be selective catalysts for acetylene hydrogenation to ethylene, but the origin of the selectivity remains unclear. In this work, a density functional theory (DFT) study has been carried out to investigate the mechanism of acetylene hydrogenation on six surfaces of Ni-Sn IMCs, and the geometric effects towards ethylene selectivity were revealed. Two key parameters (adsorption energy and the hydrogenation barrier of ethylene), which determine the ethylene selectivity, were studied quantitatively. The adsorption sites for C2Hy (y = 2, 3, 4) can be classified into three types: Type 1 (Ni3Sn(111) and Ni3Sn2(101)-2) with Ni trimers, Type 2 (Ni3Sn(001) and Ni3Sn2(001)) with Ni monomers, and Type 3 (Ni3Sn2(101) and Ni3Sn2(001)-2) with reconstructed metal trimers. The adsorption energy (Ead) decreases following the order: Type 1 > Type 3 > Type 2, which indicates that the adsorption strength depends significantly on site ensemble: a more isolated Ni site would facilitate the desorption of ethylene. However, the surface roughness mainly dominates the hydrogenation barrier of ethylene. Either low or high roughness decreases the interactions between H and C2H4 (Eint), resulting in an enhanced energy barrier for over-hydrogenation of C2H4 (Ea,hydr); while moderate roughness benefits Eint and lowers Ea,hydr. The selectivity to ethylene is denoted as ΔEa = Ea,hydr - |Ead|, thus depending on the interplay of site ensemble effects and surface roughness. From this point of view, Ni3Sn(001) and Ni3Sn2(101) surfaces with well-isolated Ni ensembles and low (or high) surface roughness exhibit decreased |Ead| and increased Ea,hydr, giving rise to excellent selectivity to ethylene. This work provides significant understanding of the origin of ethylene selectivity in terms of geometric effects, which gives helpful instruction for the design and preparation of intermetallic catalysts for acetylene semi-hydrogenation.

Late embryogenesis abundant (LEA) proteins, the space fillers or molecular shields, are the hydrophilic protective proteins which play an important role during plant development and abiotic stress. The systematic survey and characterization revealed a total of 68 LEA genes, belonging to 8 families in Sorghum bicolor. The LEA-2, a typical hydrophobic family is the most abundant family. All of them are evenly distributed on all 10 chromosomes and chromosomes 1, 2, and 3 appear to be the hot spots. Majority of the S. bicolor LEA (SbLEA) genes are intron less or have fewer introns. A total of 22 paralogous events were observed and majority of them appear to be segmental duplications. Segmental duplication played an important role in SbLEA-2 family expansion. A total of 12 orthologs were observed with Arabidopsis and 13 with Oryza sativa. Majority of them are basic in nature, and targeted by chloroplast subcellular localization. Fifteen miRNAs targeted to 25 SbLEAs appear to participate in development, as well as in abiotic stress tolerance. Promoter analysis revealed the presence of abiotic stress-responsive DRE, MYB, MYC, and GT1, biotic stress-responsive W-Box, hormone-responsive ABA, ERE, and TGA, and development-responsive SKn cis-elements. This reveals that LEA proteins play a vital role during stress tolerance and developmental processes. Using microarray data, 65 SbLEA genes were analyzed in different tissues (roots, pith, rind, internode, shoot, and leaf) which show clear tissue specific expression. qRT-PCR analysis of 23 SbLEA genes revealed their abundant expression in various tissues like roots, stems and leaves. Higher expression was noticed in stems compared to roots and leaves. Majority of the SbLEA family members were up-regulated at least in one tissue under different stress conditions. The SbLEA3-2 is the regulator, which showed abundant expression under diverse stress conditions. Present study provides new insights into the formation of LEAs in S. bicolor and to understand their role in developmental processes under stress conditions, which may be a valuable source for future research., Competing Interests: The authors have declared that no competing interests exist.

To investigate the role of fatty acid-binding protein 5 (FABP5) in infectious diseases, FABP5-deficient mice were challenged with Listeria monocytogenes, a facultative intracellular bacterial pathogen. Interestingly, FABP5-deficient animals were able to clear the infection within 3 days whereas control wild-type (WT) animals showed comparatively higher bacterial burdens in the liver and spleen. Sections of infected tissues showed an increase in inflammatory foci in WT mice compared to FABP5-deficient mice. FABP5-deficient mice had lower circulating inflammatory cytokines and increased inducible nitric oxide synthase production. FABP5-deficient mouse bone marrow-derived macrophages produced higher levels of nitrite anion than their WT counterparts in response to various stimuli. Additionally, in contrast to FABP5-/- mice, transgenic mice overexpressing FABP5 in myeloid cells (LysM-Cre driven) showed decreased survival rates and increased bacterial burden and inflammatory cytokines. Overall, these findings suggest that increased FABP5 levels correlate with a higher L. monocytogenes bacterial burden and elevated subsequent inflammation., (© 2019 The Author(s) Published by S. Intermetallic compounds (IMCs) have shown excellent catalytic performance toward the selective hydrogenation of acetylene, but the theoretical understanding on this reaction over Ni-based IMCs is rather limited. In this work, the adsorptions of the C 2 species, Bader charge, projected density of states (PDOS) and the reaction pathways were calculated by the density functional theory (DFT) method to investigate the mechanism and selectivity for the acetylene hydrogenation on the (111) surface of Ni n Ga (n = 1, 3) IMCs, with a comparative study on the pristine Ni(111) surface. The results indicate that the adsorption energy of acetylene increased along with the Ni/Ga ratio, therefore a feasible acetylene adsorption on the Ga-rich surface guaranteed a low effective barrier, leading to the best activity for the NiGa(111) surface among three surfaces. Bader charge analysis shows that electrons transferred from Ga atoms to Ni atoms and further delivered to C 2 species, decreasing the adsorption capacity of C 2 species on NiGa(111) in comparison with those on Ni(111) and Ni 3 Ga(111). The reaction pathway of acetylene hydrogenation to ethylene via vinyl or even over-hydrogenation to ethane via ethyl is more favorable than the pathway involving the ethylidene intermediate on all surfaces. Moreover, the ethylene selectivity has a positive correlation with the gallium content by comparing the desorption barrier with the hydrogenation barrier of ethylene, thus the NiGa(111) surface also exhibits the best selectivity. Therefore, the NiGa(111) surface demonstrates to be an excellent reaction facet for the semihydrogenation of acetylene, which agreed with the experimental findings, and would provide helpful instructions for designing and preparing highly-selective and noble-substitute catalysts of alkyne semihydrogenation.

Trisomy 21 (Down Syndrome, DS) is the most common chromosomal anomaly. Although DS is mostly perceived as affecting cognitive abilities and cardiac health, individuals with DS also exhibit dysregulated immune functions. Levels of pro-inflammatory cytokines are increased, but intrinsic alterations of innate immunity are understudied in DS. Furthermore, elevated Reactive Oxygen Species (ROS) are well documented in individuals with DS, further exacerbating inflammatory processes. Chronic inflammation and oxidative stress are often precursors of subsequent tissue destruction and pathologies, which affect a majority of persons with DS. Together with ROS, the second messenger ion Ca 2+ plays a central role in immune regulation. TRPM2 (Transient Receptor Potential Melastatin 2) is a Ca 2+ -permeable ion channel that is activated under conditions of oxidative stress. The Trpm2 gene is located on human Chromosome 21 (Hsa21). TRPM2 is strongly represented in innate immune cells, and numerous studies have documented its role in modulating inflammation. We have previously found that as a result of suboptimal cytokine production, TRPM2 -/- mice are highly susceptible to the bacterial pathogen Listeria monocytogenes (Lm). We therefore used Lm infection to trigger and characterize immune responsiveness in the DS mouse model Dp10(yey), and to investigate the potential contribution of TRPM2. In comparison to wildtype (WT), Dp10(yey) mice show an increased resistance against Lm infection and higher IFNγ serum concentrations. Using a gene elimination approach, we show that these effects correlate with Trpm2 gene copy number, supporting the notion that Trpm2 might promote hyperinflammation in DS., (Copyright © 2017. Gene mapping of inherited ophthalmic diseases such as congenital cataracts, retinal degeneration, glaucoma, age-related macular degeneration, myopia, optic atrophy, and eye malformations has shed more light on the disease pathology, identified targets for research on therapeutics, earlier detection, and treatment options for disease management and patient care. This article details the different approaches to gene identification for both Mendelian and complex eye disorders.

A genome-wide scanning of Sorghum bicolor resulted in the identification of 25 SbHsf genes. Phylogenetic analysis shows the ortholog genes that are clustered with only rice, representing a common ancestor. Promoter analysis revealed the identification of different cis-acting elements that are responsible for abiotic as well as biotic stresses. Hsf domains like DBD, NLS, NES, and AHA have been analyzed for their sequence similarity and functional characterization. Tissue specific expression patterns of Hsfs in different tissues like mature embryo, seedling, root, and panicle were studied using real-time PCR. While Hsfs4 and 22 are highly expressed in panicle, 4 and 9 are expressed in seedlings. Sorghum plants were exposed to different abiotic stress treatments but no expression of any Hsf was observed when seedlings were treated with ABA. High level expression of Hsf1 was noticed during high temperature as well as cold stresses, 4 and 6 during salt and 5, 6, 10, 13, 19, 23 and 25 during drought stress. This comprehensive analysis of SbHsf genes will provide an insight on how these genes are regulated in different tissues and also under different abiotic stresses and help to determine the functions of Hsfs during drought and temperature stress tolerance.

Background: Intercontinental migratory waterfowl are the primary vectors for dispersion of H5N1 viruses and have been implicated in several zoonotic epidemics and pandemics. Recent investigations have established that with a single mutation, the virus gains the ability to transmit between humans. Consequently, there is a heightened urgency to identify innovative approaches to proactively mitigate emergent epidemics. Accordingly, a novel methodology combining temporo-geospatial epidemiology and phylogeographic analysis of viral strains is proposed to identify critical epicenters and epidemic pathways along with high risk candidate regions for increased surveillance., Results: Epidemiological analysis was used to identify 91,245 candidate global infection transmission pathways between 22 high risk waterfowl species. Dominant infection pathways (25,625 and 54,500 in summering and wintering zones) were identified through annotation using phylogeographical data computed from the phylogram of 2417 H5N1 HA isolates (from GISAID EpiFlu database). Annotation of infection pathways in turn delineated 23 influential clades out of 130 clades in the phylogram., Conclusions: The phylogeographic analyses provides strong cross-validation of epidemic pathways and identifies the dominant pathways for use in other epidemiological and prophylactic studies. The temporo-geospatial characteristics of infection transmission provides corroborating, but novel evidence for rapid genesis of H5N1 lineages in S.E. Asia. The proposed method pinpoints several regions, particularly in the southern hemisphere, as candidates for increased surveillance.

Objective: The aim was to assess the accuracy of three-dimensional anatomical reductions achieved by open method of treatment in cases of displaced unilateral mandibular subcondylar fractures using preoperative (pre op) and postoperative (post op) computed tomography (CT) scans., Materials and Methods: In this prospective study, 10 patients with unilateral sub condylar fractures confirmed by an orthopantomogram were included. A pre op and post op CT after 1 week of surgical procedure was taken in axial, coronal and sagittal plane along with three-dimensional reconstruction. Standard anatomical parameters, which undergo changes due to fractures of the mandibular condyle were measured in pre and post op CT scans in three planes and statistically analysed for the accuracy of the reduction comparing the following variables: (a) Pre op fractured and nonfractured side (b) post op fractured and nonfractured side (c) pre op fractured and post op fractured side. P < 0.05 was considered as significant., Results: Three-dimensional anatomical reduction was possible in 9 out of 10 cases (90%). The statistical analysis of each parameter in three variables revealed (P < 0.05) that there was a gross change in the dimensions of the parameters obtained in pre op fractured and nonfractured side. When these parameters were assessed in post op CT for the three variables there was no statistical difference between the post op fractured side and non fractured side. The same parameters were analysed for the three variables in pre op fractured and post op fractured side and found significant statistical difference suggesting a considerable change in the dimensions of the fractured side post operatively., Conclusion: The statistical and clinical results in our study emphasised that it is possible to fix the condyle in three-dimensional anatomical positions with open method of treatment and avoid post op degenerative joint changes. CT is the ideal imaging tool and should be used on a regular basis for cases of condylar fractures.

Introduction: The objective is to analyze proinflammatory cytokines [interleukin-1β (IL-1β), interleukin-6 and tumor necrosis factor-α] and sphingomyelinase in women with bacterial vaginosis (BV), cervicitis and vaginitis., Methods: From January 2009 to June 2010, human immunodeficiency virus (HIV)-negative, nonpregnant, married women, living with husband, aged 20 to 40 years were recruited from a slum at Hyderabad, India, after taking written consent. One hundred forty-six women including 61 women with BV, 47 women with intermediate flora and 38 women with normal vaginal flora were evaluated for local proinflammatory cytokines and sphingomyelinase. Cervicitis and vaginitis were also analyzed by scoring white blood cells in the cervix and vaginal smears, respectively., Results: Of the 146 women, 50.7% had cervicitis and 19.5% had vaginitis. IL-1β, tumor necrosis factor-α and interleukin-6 levels were significantly high in women with cervicitis (P < 0.001) and vaginitis (P < 0.001) and IL-1β in BV (P < 0.005), intermediate flora (P < 0.05) when compared with normal women. High vaginal pH was associated with IL-1β. Neutral sphingomelinase showed an inverse association (P < 0.05) with cervicitis. Acid sphingomelinase directly correlated with IL-1β although not significantly., Conclusions: This study shows proinflammatory response not only in BV but also in women with vaginitis and cervicitis. These conditions are likely to be important in promoting the transmission of HIV-1 and should be the focus of HIV prevention strategies.

The chemopreventive effect of ethanol root extract of Indigofera aspalathoides was evaluated in N-nitrosodiethylamine-induced experimental liver tumor in mice. Pretreatment with ethanol root extract (100 mg/kg, p.o.) for three weeks significantly reduced the impact of N-nitrosodiethylamine toxicity (50 mg/kg, i.p.) on the serum markers of liver damage, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase and total bilirubin. Protective effect was reconfirmed the elevated serum total protein levels were significantly restored towards normalization by the extracts, and this was confirmed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Histological examination of the liver of animals treated with ethanol root extract of Indigofera aspalathoides showed the reduction of necrosis. These results suggest that ethanol root extract of Indigofera aspalathoides possess protective effect against N-nitrosodiethylamine-induced hepatocellular carcinogenesis.

Alzheimer's disease (AD) is a devastative neurodegenerative disorder which needs adequate studies on effective treatment options. The extracts of plants and their effect on the amelioration of AD symptoms have been extensively studied. This paper summarizes the mechanisms like acetylcholinesterase (AChE) inhibition, modification of monoamines, antiamyloid aggregation effect, and antioxidant activity which are actively entailed in the process of amelioration of AD symptoms. These effects are induced by extracts of a few plants of different origin like Yizhi Jiannao, Moringa oleifera (Drumstick tree), Ginkgo Biloba (Ginkgo/Maidenhair tree), Cassia obtisufolia (Sicklepod), Desmodium gangeticum (Sal Leaved Desmodium), Melissa officinalis (Lemon Balm), and Salvia officinalis (Garden sage, common sage).

IL-18 modulates immune functions by inducing IFN-γ production and promoting Th1 immune responses. In the present study, we amplified and cloned the sequence (582 bp) encoding full-length bovine IL-18 from PBMC stimulated with PHA. The nucleotide and the deduced amino acid sequence of Bos indicus IL-18 showed an identity of 86-98% compared with IL-18 sequences of other ruminants. The insert was subcloned into a pET 32a vector and expressed in Escherichia coli as a fusion protein and the matured protein was obtained by caspase I treatment. The specificity of these proteins was confirmed by western blotting. The biological activity of the purified protein was analyzed by its ability to induce IFN-γ production in PBMC measured by ELISA and qPCR.

Alzheimer's disease is a neurodegenerative disorder with unclear etiology for a few decades. Many animal models employed to study the etiology of the disease and test the efficacy of a drug could give limited understanding of these events. Introduction of aluminum salts into aged New Zealand rabbit brain could demonstrate neurofibrillary tangle formation in 1965. This outstanding contribution substantiated the role of aluminum in Alzheimer's disease in turn becoming the basis further molecular studies in rabbits. In this review, various animal models (transgenic mice, rats, rabbits, zebrafish) used to study the pathology of the disease and to test the efficacy of a drug have been summarized. It also focuses on the growing need to unravel the molecular underpinnings of the disease progression.

We present PEACE, a stand-alone tool for high-throughput ab initio clustering of transcript fragment sequences produced by Next Generation or Sanger Sequencing technologies. It is freely available from www.peace-tools.org. Installed and managed through a downloadable user-friendly graphical user interface (GUI), PEACE can process large data sets of transcript fragments of length 50 bases or greater, grouping the fragments by gene associations with a sensitivity comparable to leading clustering tools. Once clustered, the user can employ the GUI's analysis functions, facilitating the easy collection of statistics and allowing them to single out specific clusters for more comprehensive study or assembly. Using a novel minimum spanning tree-based clustering method, PEACE is the equal of leading tools in the literature, with an interface making it accessible to any user. It produces results of quality virtually identical to those of the WCD tool when applied to Sanger sequences, significantly improved results over WCD and TGICL when applied to the products of Next Generation Sequencing Technology and significantly improved results over Cap3 in both cases. In short, PEACE provides an intuitive GUI and a feature-rich, parallel clustering engine that proves to be a valuable addition to the leading cDNA clustering tools.

Deoxyribonucleic acid (DNA) damage has been implicated in ageing and neurodegenerative disorders including Alzheimer's disease (AD) for a few decades. Although it is an established finding, yet there are limited studies on DNA damage. In both nucleus and mitochondria, DNA damage is primarily free radical mediated. It has been proven that mitochondrial DNA is more vulnerable to damage compared to the nuclear DNA. A few studies summarized in this review throw light on the mechanisms of free radical mediated DNA damage and impairment of DNA repair mechanisms in AD. There is a growing need to initiate studies on DNA damage and repair and unravel the molecular underpinnings entailed in the etiopathogenesis of the disease. The outcome of such studies substantiates the corner stone streamlined to employ therapeutic strategies.

Background: Alzheimers disease (AD) is a devastative neurodegenerative disorder. Our studies for the first time provided substantial animal model and tattered the etiology of the disease at a molecular level., Methods: In this study DNA was isolated from Hippocampus (H), Midbrain (M) and Frontal Cortex (Fc) of control and aluminium maltolate (Al-M) treated aged New Zealand rabbit brain. DNA damage has been studied using Agarose gel electrophoresis, Ethidium Bromide (EtBr) binding and Melting temperature techniques., Results: Al-M treated aged New Zealand rabbit's H and M showed higher DNA damage compared to corresponding controls, where as Fc showed mild DNA damage compared to corresponding controls., Conclusions: This study tangibly provides substantial molecular level understanding of the disease in turn providing an adequate platform to streamline potential therapeutic strategies., Keywords: Alzheimer's disease; Aluminium maltolate; Animal model; DNA damage.

Humanity is facing an increasing number of highly virulent and communicable diseases such as avian influenza. Researchers believe that avian influenza has potential to evolve into one of the deadliest pandemics. Combating these diseases requires in-depth knowledge of their epidemiology. An effective methodology for discovering epidemiological knowledge is to utilize a descriptive, evolutionary, ecological model and use bio-simulations to study and analyze it. These types of bio-simulations fall under the category of computational evolutionary methods because the individual entities participating in the simulation are permitted to evolve in a natural manner by reacting to changes in the simulated ecosystem. This work describes the application of the aforementioned methodology to discover epidemiological knowledge about avian influenza using a novel eco-modeling and bio-simulation environment called SEARUMS. The mathematical principles underlying SEARUMS, its design, and the procedure for using SEARUMS are discussed. The bio-simulations and multi-faceted case studies conducted using SEARUMS elucidate its ability to pinpoint timelines, epicenters, and socio-economic impacts of avian influenza. This knowledge is invaluable for proactive deployment of countermeasures in order to minimize negative socioeconomic impacts, combat the disease, and avert a pandemic.

Successive petroleum ether, benzene, chloroform, ethyl acetate and methanol extracts of Chukrasia tabularis leaves were tested for their phytochemical constituents, antibacterial and antifungal activity. The ethyl acetate and methanol extracts were found to be most effective against most of the tested organisms.

In this study, we have examined whether tumor grade and morphometric nuclear features can predict the outcome of treatment by orchiectomy in patients with stage D2 prostate cancer. Two outcome groups based on duration of survival postorchiectomy were examined, a bad outcome group of 63 patients who died from prostate cancer within 12 months and a good outcome group of 34 patients who survived beyond 5 years. Tumors were histologically classified as well (17%), moderate (17%), or poorly differentiated (66%). Tumor grade and patient outcome were significantly associated (Mann-Whitney test; P < 0.005), with 76% of poorly differentiated tumors in the bad outcome group, and 65% of well-differentiated tumors in the good outcome group. Using discriminant function analysis, tumor grade correctly predicted outcome in 70% of cases. A statistically significant difference was also detected in nuclear shape values between the two outcome groups (P < 0.05) and histological grades (P < 0.05). Using discriminant function analysis, 51% of cases were correctly classified into outcome groups using nuclear shape factors, a figure which rose to 65% when all nuclear morphometric features were used. This demonstrates that nuclear morphometric features are of no clinical value in predicting the outcome of treatment in stage D2 disease. Furthermore, these evaluations cannot select patients who might be spared orchiectomy on the basis of a predicted poor response. However, nuclear shape and variance measurements of benign glandular epithelial cells within cancerous prostates were significantly different from those of malignant cells (P < 0.005). We conclude that, while video image analysis of prostatic nuclear shape can reliably discriminate between benign and malignant cells, nuclear morphometric features are of minimal prognostic value in men with stage D2 prostate cancer treated by androgen ablation.

A rapid, sensitive and selective method has been developed for the determination of palladium with 5,6-dimethyl-1,3-indanedione-2-oxime in acetate buffer (pH 5.5). Beer's law is obeyed over the range 0.15-4.17 mug/ml palladium(II). The molar absorptivity at 370 nm is 2.98 x 10(4) 1.mole(-1).cm(-1). The method has been applied for the determination of palladium in synthetic mixtures corresponding to PtIr and Oakay alloys. An interesting feature of the system is the abnormal shape of the Job and molar-ratio plots obtained.

This study was designed to compare the prognostic potential of tumor grade and ploidy status in patients with stage D2 prostate cancer. Two outcome groups were selected on the basis of survival after orchiectomy: a bad outcome group consisting of 66 patients who died of the disease within 12 months and a good outcome group comprising 37 patients who survived beyond 5 years. Tumors were classified histologically as well (17%), moderately (17%) or poorly (66%) differentiated. Tumor grade was a significant predictor of outcome, with 76% of poorly differentiated tumors in the bad outcome group and 65% of well differentiated tumors in the good outcome group (p less than 0.005). Deoxyribonucleic acid (DNA) ploidy analysis was performed on formalin fixed, paraffin embedded samples of the primary tumor to yield 97 final tracings that were classified using set criteria for DNA ploidy status. Over-all, 54% of the tumors were nondiploid (33% aneuploid and 21% tetraploid) and the remaining 46% were diploid. DNA ploidy status was a significant indicator of outcome (p less than 0.001), with 64% of diploid tumors in the good outcome group and 88% of the nondiploid tumors in the poor outcome group. Tetraploid tumors behaved no differently from other nondiploid tumors. We conclude that DNA ploidy status and tumor grading are significant independent predictors of outcome after orchiectomy and when combined yield important additional prognostic information.

Reports of Melon et al. [7] and Hodgkinson [4] regarding the efficacy of succinic acid or its derivative (salt) in reducing the endogenous synthesis of oxalate are conflicting. The possible application of succinic acid as a potential inhibitor of oxalate synthesis, and the biochemical alterations resulting therein, has been studied in 10 idiopathic stone-forming patients (6 primary stone-formers and 4 recurrent stone-formers) and in 21 normal adult rats. In rats no significant changes in oxalate excretion were observed during the 4 weeks of succinic acid administration (500 mg/100 g BW/day) and 2 weeks therafter. In the clinical trial of 3 months, where stone-formers were given 20 g/day of succinic acid, the post-therapy values of urinary oxalate excretion showed a singificant drop ( less than 0.001) as compared with those of pretherapy--recurrent stone-formers showing a more marked decrease (63%) than primary stone-formers (49%).

After the resection of a localized lung carcinoma, a 65-year-old veteran developed jejunal perforation and pneumonia, both of which subsequently were shown to be caused by infection with Strongyloides stercoralis. The patient recovered after jejunal resection and treatment with thiabendazole. Several unusual aspects of the case are discussed.

A new solvent system 1% and 2% acetone in hexane, was developed for chromatography on partially inactivated Florisil to facilitate the separation of isomers of endosulfan and its metabolites from fish tissues under tropical conditions. By using the colorimetric method of Maitlen et al. (1963), the minimum detectable limit for each isomer of endosulfan in various fish tissues is 5 micro g. With this solvent system, the 2 isomers present in the technical material can be separated, so their toxicity to fish can be assessed separately.

Chromosome studies were conducted on 117 men with primary infertility (54 with oligospermia and 63 with azoospermia) and 25 fertile volunteers who acted as controls. No chromosomal abnormalities were observed except for nine cases of Klinefelter's syndrome. From this study, it is apparent that chromosomal abnormalities other than Klinefelter's syndrome are not etiologic factors in the infertility of oligo- or azoospermic men.
