4. Anaerobic Dynamic Membrane Bioreactor Development to Facilitate Organic Waste Conversion to Medium-Chain Carboxylic Acids and Their Downstream Recovery

- Abstract
Platform chemicals such as medium-chain carboxylic acids (MCCAs) can be produced from organic waste streams via chain elongation in anaerobic mixed-culture bioreactors. A product recovery system is needed to collect MCCAs from the bioreactor effluent. Membrane-based liquid-liquid extraction, the most commonly used product recovery approach, requires suspended solids removal from the bioreactor effluent to avoid membrane fouling. An anaerobic dynamic membrane bioreactor (AnDMBR) was developed to evaluate MCCA production from brewery and prefermented food waste and to produce a permeate with low suspended solids to facilitate downstream product recovery. The AnDMBR employed an inexpensive stainless-steel mesh as the support material for the development of a biofilm or dynamic membrane, which was responsible for solids-liquid separation. The AnDMBR produced a permeate quality with an average total suspended solids (TSS) concentration of 0.12 g L-1, while the average bioreactor TSS concentration was two orders of magnitude higher (21.6 g L-1). A maximum solids removal efficiency of ≥99% was achieved and good permeate quality was sustained for over 200 days without fouling control or cleaning the support material. In addition to solids-liquid separation, the dynamic membrane was responsible for a substantial fraction of the biological activity of the AnDMBR. The relative activity of Clostridiales, as determined by 16S rRNA sequencing, correlated with MCCA production and was higher in the dynamic membrane (20.0 ± 4.9%) than in the suspended biomass (5.2 ± 2.7%) in the bioreactor. This observation was consistent with MCCA production data as the permeate MCCA concentrations were significantly (p = 8.2 × 10-5) higher than that in the bioreactor, suggesting that the dynamic membrane biofilm contributed to chain elongation.

- Abstract
Kinetic-Family-Drawings were obtained from 50 kindergarten or first grade children with delayed perceptual and/or motor development and 50 controls to investigate the effectiveness of this instrument as a measure of socio-emotional dimensions of high risk low achievers. Each child was asked to draw each member of his family (including himself) actively doing something. Drawings were scored on the criteria of isolation, bodily concerns, and rivalry. Results indicated that Ss having delayed development were differentiated from controls by a greater sense of isolation and bodily concern. Rivalry was not a significant discriminator. (Author/DB)

- Abstract
OBJECTIVE: An understanding of the etiologic heterogeneity of colorectal cancer (CRC) is critical for improving precision prevention, including individualized screening recommendations and the discovery of novel drug targets and repurposable drug candidates for chemoprevention. Known differences in molecular characteristics and environmental risk factors among tumors arising in different locations of the colorectum suggest partly distinct mechanisms of carcinogenesis. The extent to which the contribution of inherited genetic risk factors for CRC differs by anatomical subsite of the primary tumor has not been examined. DESIGN: To identify new anatomical subsite-specific risk loci, we performed genome-wide association study (GWAS) meta-analyses including data of 48 214 CRC cases and 64 159 controls of European ancestry. We characterised effect heterogeneity at CRC risk loci using multinomial modelling. RESULTS: We identified 13 loci that reached genome-wide significance (p<5×10-8) and that were not reported by previous GWASs for overall CRC risk. Multiple lines of evidence support candidate genes at several of these loci. We detected substantial heterogeneity between anatomical subsites. Just over half (61) of 109 known and new risk variants showed no evidence for heterogeneity. In contrast, 22 variants showed association with distal CRC (including rectal cancer), but no evidence for association or an attenuated association with proximal CRC. For two loci, there was strong evidence for effects confined to proximal colon cancer. CONCLUSION: Genetic architectures of proximal and distal CRC are partly distinct. Studies of risk factors and mechanisms of carcinogenesis, and precision prevention strategies should take into consideration the anatomical subsite of the tumour.

- Abstract
To further dissect the genetic architecture of colorectal cancer (CRC), we performed whole-genome sequencing of 1,439 cases and 720 controls, imputed discovered sequence variants and Haplotype Reference Consortium panel variants into genome-wide association study data, and tested for association in 34,869 cases and 29,051 controls. Findings were followed up in an additional 23,262 cases and 38,296 controls. We discovered a strongly protective 0.3% frequency variant signal at CHD1. In a combined meta-analysis of 125,478 individuals, we identified 40 new independent signals at P < 5 × 10 −8 , bringing the number of known independent signals for CRC to ~100. New signals implicate lower-frequency variants, Krüppel-like factors, Hedgehog signaling, Hippo-YAP signaling, long noncoding RNAs and somatic drivers, and support a role for immune function. Heritability analyses suggest that CRC risk is highly polygenic, and larger, more comprehensive studies enabling rare variant analysis will improve understanding of biology underlying this risk and influence personalized screening strategies and drug development. © 2018, This is a U.S. government work and not under copyright protection in the U.S.; foreign copyright protection may apply.

- Abstract
Quantifying the genetic correlation between cancers can provide important insights into the mechanisms driving cancer etiology. Using genome-wide association study summary statistics across six cancer types based on a total of 296,215 cases and 301,319 controls of European ancestry, here we estimate the pair-wise genetic correlations between breast, colorectal, head/neck, lung, ovary and prostate cancer, and between cancers and 38 other diseases. We observed statistically significant genetic correlations between lung and head/neck cancer (rg = 0.57, p = 4.6 x 10-8), breast and ovarian cancer (rg = 0.24, p = 7 x 10-5), breast and lung cancer (rg = 0.18, p =1.5 x 10-6) and breast and colorectal cancer (rg = 0.15, p = 1.1 x 10-4). We also found that multiple cancers are genetically correlated with non-cancer traits including smoking, psychiatric diseases and metabolic characteristics. Functional enrichment analysis revealed a significant excess contribution of conserved and regulatory regions to cancer heritability. Our comprehensive analysis of cross-cancer heritability suggests that solid tumors arising across tissues share in part a common germline genetic basis.Copyright © 2019, The Author(s).

- Abstract
BACKGROUND: Previous genome-wide association studies (GWAS) have identified 42 loci (P < 5 × 10-8) associated with risk of colorectal cancer (CRC). Expanded consortium efforts facilitating the discovery of additional susceptibility loci may capture unexplained familial risk. METHODS: We conducted a GWAS in European descent CRC cases and control subjects using a discovery-replication design, followed by examination of novel findings in a multiethnic sample (cumulative n = 163 315). In the discovery stage (36 948 case subjects/30 864 control subjects), we identified genetic variants with a minor allele frequency of 1% or greater associated with risk of CRC using logistic regression followed by a fixed-effects inverse variance weighted meta-analysis. All novel independent variants reaching genome-wide statistical significance (two-sided P < 5 × 10-8) were tested for replication in separate European ancestry samples (12 952 case subjects/48 383 control subjects). Next, we examined the generalizability of discovered variants in East Asians, African Americans, and Hispanics (12 085 case subjects/22 083 control subjects). Finally, we examined the contributions of novel risk variants to familial relative risk and examined the prediction capabilities of a polygenic risk score. All statistical tests were two-sided. RESULTS: The discovery GWAS identified 11 variants associated with CRC at P < 5 × 10-8, of which nine (at 4q22.2/5p15.33/5p13.1/6p21.31/6p12.1/10q11.23/12q24.21/16q24.1/20q13.13) independently replicated at a P value of less than .05. Multiethnic follow-up supported the generalizability of discovery findings. These results demonstrated a 14.7% increase in familial relative risk explained by common risk alleles from 10.3% (95% confidence interval [CI] = 7.9% to 13.7%; known variants) to 11.9% (95% CI = 9.2% to 15.5%; known and novel variants). - Abstract
Quantifying the genetic correlation between cancers can provide important insights into the mechanisms driving cancer etiology. Using genome-wide association study summary statistics across six cancer types based on a total of 296,215 cases and 301,319 controls of European ancestry, here we estimate the pair-wise genetic correlations between breast, colorectal, head/neck, lung, ovary and prostate cancer, and between cancers and 38 other diseases. We observed statistically significant genetic correlations between lung and head/neck cancer (rg = 0.57, p = 4.6 × 10-8), breast and ovarian cancer (rg = 0.24, p = 7 × 10-5), breast and lung cancer (rg = 0.18, p =1.5 × 10-6) and breast and colorectal cancer (rg = 0.15, p = 1.1 × 10-4). We also found that multiple cancers are genetically correlated with non-cancer traits including smoking, psychiatric diseases and metabolic characteristics. Functional enrichment analysis revealed a significant excess contribution of conserved and regulatory regions to cancer heritability. Our comprehensive analysis of cross-cancer heritability suggests that solid tumors arising across tissues share in part a common germline genetic basis.

- Abstract
To further dissect the genetic architecture of colorectal cancer (CRC), we performed whole-genome sequencing of 1,439 cases and 720 controls, imputed discovered sequence variants and Haplotype Reference Consortium panel variants into genome-wide association study data, and tested for association in 34,869 cases and 29,051 controls. Findings were followed up in an additional 23,262 cases and 38,296 controls. We discovered a strongly protective 0.3% frequency variant signal at CHD1. In a combined meta-analysis of 125,478 individuals, we identified 40 new independent signals at P < 5 × 10-8, bringing the number of known independent signals for CRC to ~100. New signals implicate lower-frequency variants, Krüppel-like factors, Hedgehog signaling, Hippo-YAP signaling, long noncoding RNAs and somatic drivers, and support a role for immune function. Heritability analyses suggest that CRC risk is highly polygenic, and larger, more comprehensive studies enabling rare variant analysis will improve understanding of biology underlying this risk and influence personalized screening strategies and drug development.

- Abstract
Quantifying the genetic correlation between cancers can provide important insights into the mechanisms driving cancer etiology. Using genome-wide association study summary statistics across six cancer types based on a total of 296,215 cases and 301,319 controls of European ancestry, here we estimate the pair-wise genetic correlations between breast, colorectal, head/neck, lung, ovary and prostate cancer, and between cancers and 38 other diseases. We observed statistically significant genetic correlations between lung and head/neck cancer (rg = 0.57, p = 4.6 × 10−8), breast and ovarian cancer (rg = 0.24, p = 7 × 10−5), breast and lung cancer (rg = 0.18, p =1.5 × 10−6) and breast and colorectal cancer (rg = 0.15, p = 1.1 × 10−4). We also found that multiple cancers are genetically correlated with non-cancer traits including smoking, psychiatric diseases and metabolic characteristics. Functional enrichment analysis revealed a significant excess contribution of conserved and regulatory regions to cancer heritability. Our comprehensive analysis of cross-cancer heritability suggests that solid tumors arising across tissues share in part a common germline genetic basis.

Розглянуто задачі багатокритеріальної оптимізації. Відомі методи розв’язання цих задач узагальнено на випадок, коли вагові коефіцієнти, що враховують відносну важливість окремих критеріїв, визначені нечітко. Обґрунтовано процедуру побудови функцій приналежності нечітких чисел, заданих наборами інтервалів можливих значень, що використовує лінеаризовану обчислювальну схему методу найменших квадратів. Для описів нечітких чисел вибрано функції приналежності (L-R)-типу. Запропоновано метод розв’язування нечіткої задачі багатокритеріальної оптимізації для скаляризованого критерію. Технологія розв’язування задачі зводить її до дробово-нелінійної задачі математичного програмування. Описано збіжну ітераційну процедуру знаходження оптимального плану. Розглянуто альтернативний метод розв’язування нечіткої задачі на основі формування Парето-множини недомінуючих варіантів. Для розв’язання цієї задачі запропоновано процедуру порівняння нечітких чисел з використанням теоретико-ймовірнісної апроксимації їх функцій приналежності. The problems of multi-criteria optimization are considered. Known methods for solving these problems are generalized to the case when weights that take into account the relative importance of particular criteria are not clearly defined. The procedure for constructing membership functions of fuzzy numbers, given by sets of intervals of possible values, using a linearized computation of least squares methods is substantiated. In this case, for the description of fuzzy numbers, the membership functions of (L-R)-type were chosen. A method for solving a fuzzy multi-criteria optimization problem for a scalarized criterion is proposed. The technology of solving the problem reduces it to a linear fractional problem of mathematical programming. A converging iterative procedure for finding the optimal plan is described. An alternative method for solving the original fuzzy problem based on the formation of a Pareto-set of non-dominated options is considered. To solve this problem, a procedure has been proposed for comparing fuzzy numbers using a probability-theoretic approximation of their membership functions.

Fuzzy set theory is an effective alternative to probability theory in solving many problems of studying processes and systems under conditions of uncertainty. The application of this theory is especially in demand in situations where the system under study operates under conditions of rapidly changing influencing parameters or characteristics of the environment. In these cases, the use of solutions obtained by standard methods of the probability theory is not quite correct. At the same time, the conceptual, methodological and hardware base of the alternative fuzzy set theory is not sufficiently developed. The paper attempts to fill existing gaps in the fuzzy set theory in some important areas. For continuous fuzzy quantities, the concept of distribution density of these quantities is introduced. Using this concept, a method for calculating the main numerical characteristics of fuzzy quantities, as well as a technology for calculating membership functions for fuzzy values of functions from these fuzzy quantities and their moments is proposed. The introduction of these formalisms significantly extends the capabilities of the fuzzy set theory for solving many real problems of computational mathematics. Using these formalisms, a large number of practical problems can be solved: fuzzy regression and clustering, fuzzy multivariate discriminant analysis, differentiation and integration of functions of fuzzy arguments, state diagnostics in a situation where the initial data are fuzzy, methods for solving problems of unconditional and conditional optimization, etc. The proof of the central limit theorem for the sum of a large number of fuzzy quantities is obtained. This proof is based on the characteristic functions of fuzzy quantities introduced in the work and described at the formal level. The concepts of independence and dependence for fuzzy quantities are introduced. The method for calculating the correlation coefficient for fuzzy numbers is proposed. Typical problems of the theory of statistical hypothesis testing are considered. All these problems belong to the same object area and are formulated in a single system of axioms and assumptions using a common linguistic thesaurus. However, different approaches are used to solve each of these problems and a unique solution method is developed. In this regard, the work proposes a unified methodological approach for formulating and solving these problems. The mathematical basis of the approach is the theory of continuous linear programming (CLP), which generalizes the known mathematical apparatus of linear programming for the continuous case. The mathematical apparatus of CLP allows passing from a twopoint description of the solution of the problem in the form {0; 1} to a continuous one on the segment [0; 1]. Theorems justifying the solution of problems in terms of CLP are proved. The problems of testing a simple hypothesis against several equivalent or unequal alternatives are considered. To solve all these problems, a continuous function is introduced that specifies a randomized decision rule leading to continuous linear programming models. As a result, it becomes possible to expand the range of analytically solved problems of the theory of statistical hypothesis testing. In particular, the problem of making a decision based on the maximum power criterion with a fixed type I error probability, with a constraint on the average risk, the problem of testing a simple hypothesis against several alternatives for given type II error probabilities. The method for solving problems of statistical hypothesis testing for the case when more than one observed controlled parameter is used to identify the state is proposed. [ABSTRACT FROM AUTHOR]

- Abstract
The underlying genetic cause of colorectal cancer (CRC) can be identified for 5-10% of all cases, while at least 20% of CRC cases are thought to be due to inherited genetic factors. Screening for highly penetrant mutations in genes associated with Mendelian cancer syndromes using next-generation sequencing (NGS) can be prohibitively expensive for studies requiring large samples sizes. The aim of the study was to identify rare single nucleotide variants and small indels in 40 established or candidate CRC susceptibility genes in 1,046 familial CRC cases (including both MSS and MSI-H tumor subtypes) and 1,006 unrelated controls from the Colon Cancer Family Registry Cohort using a robust and cost-effective DNA pooling NGS strategy. We identified 264 variants in 38 genes that were observed only in cases, comprising either very rare (minor allele frequency <0.001) or not previously reported (n=90, 34%) in reference databases, including six stop-gain, three frameshift, and 255 non-synonymous variants predicted to be damaging. We found novel germline mutations in established CRC genes MLH1, APC, and POLE, and likely pathogenic variants in cancer susceptibility genes BAP1, CDH1, CHEK2, ENG, and MSH3. For the candidate CRC genes, we identified likely pathogenic variants in the helicase domain of POLQ and in the LRIG1, SH2B3, and NOS1 genes and present their clinicopathological characteristics. Using a DNA pooling NGS strategy, we identified novel germline mutations in established CRC susceptibility genes in familial CRC cases. Further studies are required to support the role of POLQ, LRIG1, SH2B3 and NOS1 as CRC susceptibility genes.

A method for solving the fractional nonlinear optimization problem has been proposed. It is shown that numerous inventory management tasks, on the rational allocation of limited resources, on finding the optimal paths in a graph, on the rational organization of transportation, on control over dynamical systems, as well as other tasks, are reduced exactly to such a problem in cases when the source data of a problem are described in terms of a probability theory or fuzzy math. We have analyzed known methods for solving the fractional nonlinear optimization problems. The most efficient among them is based on the iterative procedure that sequentially improves the original solution to a problem. In this case, every step involves solving the problem of mathematical programming. The method converges if the region of permissible solutions is compact. The obvious disadvantage of the method is the uncontrolled rate of convergence. The current paper has proposed a method to solve the problem, whose concept echoes the known method of fractional-linear optimization. The proposed technique transforms an original problem with a fractional-rational criterion to the typical problem of mathematical programming. The main advantage of the method, as well its difference from known ones, is the fact that the method is implemented using a single-step procedure for obtaining a solution. In this case, the dimensionality of a problem is not a limiting factor. The requirements to a mathematical model of the problem, which narrow the region of possible applications of the devised procedure, imply: 1) the components of the objective function must be separable functions; 2) the indicators for the power of all nonlinear terms of component functions should be the same. Another important advantage of the method is the possibility of using it to solve the problem on unconditional and conditional optimization. The examples have been considered. [ABSTRACT FROM AUTHOR]

- Abstract
Genetic susceptibility to colorectal cancer is caused by rare pathogenic mutations and common genetic variants that contribute to familial risk. Here we report the results of a two-stage association study with 18,299 cases of colorectal cancer and 19,656 controls, with follow-up of the most statistically significant genetic loci in 4,725 cases and 9,969 controls from two Asian consortia. We describe six new susceptibility loci reaching a genome-wide threshold of P<5.0E-08. These findings provide additional insight into the underlying biological mechanisms of colorectal cancer and demonstrate the scientific value of large consortia-based genetic epidemiology studies.

Для вузла комп’ютерної мережі запропоновано метод вирішення задачі визначення порядку передачі сукупності пакетів із урахуванням відомого розподілу динаміки зайнятості елементів мережі. Для розв’язання задачі запропоновано критерій — максимальна тривалість доставки пакета, що мінімізується. Запропоноване завдання редукується до розв’язку сукупності двоіндексних задач призначення. Виконано обчислення оцінки доцільності використання методу оптимізації порядку передачі пакетів. Виграш, одержуваний при оптимізації порядку передачі пакетів, зростає зі збільшенням числа повідомлень, що передаються, і підвищенням рівня варіабельності довжини черги пакетів, що очікують обслуговування в проміжних вузлах. Отримано співвідношення для обчислення рівня варіабельності довжин черг. Використовуючи імітаційну модель вузла мережі було побудовано графіки, що показують виграш застосування методу оптимізації порядку передачі повідомлень у вузлах мережі для різної кількості черг., For the network node it was suggested a method, which solves the problem of the packets aggregation transfer order optimization with the known distribution of network elements busy dynamics. To solve the problem it was proposed the criterion - maximum packet delivery duration, which is minimized. It was shown, that the complicated problem reduces to a set of two-indexed assignment problems. The estimation of expediency of the assessment for packet transfer order optimization method usage was done. The gain, obtained using packet transfer order optimization, increases with the number of transferred packets and with an increasing the level of the variability of packets order length, that are waiting for service in intermediate nodes. The equations for calculating the level of variability of the order lengths are received. Using the simulation model, the graphs, which show the gain of the application of the messages transfer order optimization in the network nodes for the different number of orders, were constructed., Для узла компьютерной сети предложен метод решения задачи определения порядка передачи совокупности пакетов с учетом известного распределения динамики занятости элементов сети. Для решения задачи предложен критерий — максимальная продолжительность доставки пакета, которая минимизируется. Предложенная задача редуцируется к решению совокупности двухиндексных задач назначения. Выполнено вычисление оценки целесообразности использования метода оптимизации порядка передачи пакетов. Выигрыш, получаемый при оптимизации порядка передачи пакетов, растет с увеличением числа передаваемых сообщений и повышением уровня вариабельности длины очереди пакетов, ожидающих обслуживания в промежуточных узлах. Получены соотношения для вычисления уровня вариабельности длин очередей. Используя имитационную модель узла сети были построены графики, показывающие выигрыш применения метода оптимизации порядка передачи сообщений в узлах сети для различного числа очередей.
