Summary: Acute respiratory infection (ARI) represents a pervasive global health concern, contributing to significant morbidity and mortality. Our findings reveal a notably high prevalence of ARI in Ladakh. Employing a cross-sectional design, the total number of households surveyed was 401, among them, the total population was 2107; males, 1153, and females, 954. The highest incidence rate of 160.26/1000 persons was observed in the age group below 14 years, followed by those above 59 years (47.76), while the age group of 14-59 exhibited the lowest incidence rate at 21.23. To explore the factors contributing to ARI in Ladakh, we employed binary logistic regression modeling. Our analysis highlights significant associations between ARI incidence and various socioeconomic and environmental determinants. Notably, primary occupation, family type, kitchen ventilation, domestic fuel consumption, crowding, and smoking behaviors within families were found to be statistically significant determinants of ARI among the sample population of Ladakh., (Copyright © 2024 Copyright: © 2024 Indian Journal of Public Health.)

Particle classification plays a crucial role in various scientific and technological applications, such as differentiating between bacteria and viruses in healthcare applications or identifying and classifying cancer cells. This technique requires accurate and efficient analysis of particle properties. In this study, we investigated the integration of electrical and optical features through a multimodal approach for particle classification. Machine learning classifier algorithms were applied to evaluate the impact of combining these measurements. Our results demonstrate the superiority of the multimodal approach over analyzing electrical or optical features independently. We achieved an average test accuracy of 94.9% by integrating both modalities, compared to 66.4% for electrical features alone and 90.7% for optical features alone. This highlights the complementary nature of electrical and optical information and its potential for enhancing classification performance. By leveraging electrical sensing and optical imaging techniques, our multimodal approach provides deeper insights into particle properties and offers a more comprehensive understanding of complex biological systems., (© 2024. Hepatocellular carcinoma is one of the top causes of cancer-related death globally. SIRT3 belongs to the Sirtuin family of proteins, a collection of NAD+-dependent enzymes that play a role in controlling several cellular functions, including metabolism, aging, and stress response. SIRT3 expression has been discovered to be often downregulated in HCC tissues relative to normal liver tissues. Hence, SIRT3 may function as a tumor suppressor in HCC. In the present study, pharmacophore-based virtual screening of a small molecule database was performed initially, and then the screened hits were docked to the active site of SIRT3 to choose the best binding modes. One co-crystal ligand (PDB name: 1NQ) was utilized as a template to generate pharmacophore model query. A total of 0.2 million compounds from the VITAS-M Lab database were downloaded and prepared for virtual screening. Following database preparation, ligand-based virtual screening was performed using the pharmacophore query model generated in the previous phase. The compounds with the same pharmacophoric characteristics as the query at the same distance were screened. There were a total of 74 hits that matched the query model. These compounds were then docked to the SIRT3 using the standard precision protocol of the glide tool. To select hits with high binding affinities, a threshold of -8 kcal/mol was used. Based on the glide gscore, two hits were chosen. These two hits were selected to investigate the stability of the protein-ligand complex by molecular dynamics simulation. All of these findings indicate that the selected hit compounds C1 and C2 can serve as lead compounds in inhibiting the biological activity of SIRT3 requiring further detailed investigations.Communicated by Ramaswamy H. Regulation of formate flux by a key folate enzyme, MTHFD2 (methylene tetrahydrofolate dehydrogenase 2) in cancer cells remains poorly understood. Green et al. (Nature Metabolism, 2023; 5: 642-659) showed an interesting phenomenon of "folate trapping" toxicity leads to cancer cell kill using a potent inhibitor (TH9619) against the dehydrogenase and cyclohydrolase (DC) activities of cytosolic methylenetetrahydrofolate dehydrogenase 1 (cMTHFD1) and nuclear methylenetetrahydrofolate dehydrogenase 2 (nMTHFD2), but not the mitochondrial MTHFD2 (mTHFD2). But, mMTHFD2 is required for formate flow to cytosol which leads to the trapping of 10-formyl tetrahydrofolate and causes toxicity by TH9619 treatment, to kill cancer cells expressing mMTHFD2. Density functional theory calculations within the framework of generalized gradient approximation (GGA), meta-GGA, and local functionals were carried out to investigate the reactivity and catalytic activity of Ag n ( n = 15-20) clusters. Our results reveal that all the Ag n clusters in this size range, except Ag 20 , adsorb O 2 preferably in the bridged mode with enhanced binding energy as compared to the atop mode. The O 2 binding energies range from 0.77 to 0.29 in the bridged mode and from 0.36 to 0.15 eV in the atop mode of O 2 adsorption. The strong binding in the case of the bridged mode of O 2 adsorption is also reflected in the increase in O-O bond distance. Natural bond orbital charge analysis and vibrational frequency calculations reveal that enhanced charge transfer occurs to the O 2 molecule and there is significant red shift in the stretching frequency of O-O bond in the case of the bridged mode of O 2 adsorption on the clusters, thereby confirming the above results. Moreover, the simulated CO oxidation reaction pathways show that the oxidation of the CO molecule is highly facile on Ag 16 and Ag 18 clusters involving small kinetic barriers and higher heats toward CO 2 formation., Competing Interests: The authors declare no competing financial interest., (© 2022 The Authors. Cancer cells require increased levels of NADPH for increased nucleotide synthesis and for protection from ROS. Recent studies show that increased NADPH is generated in several ways. Activated AKT phosphorylates NAD kinase (NADK), increasing its activity. NADP formed, is rapidly converted to NADPH by glucose 6-phosphate dehydrogenase and malic enzymes, overexpressed in tumor cells with mutant p53. Calmodulin, overexpressed in some cancers, also increases NADK activity. Also, in IDH1/2 mutant cancer, NADPH serves as the cofactor to generate D-2 hydroxyglutarate, an oncometabolite. The present study reports the multi-technique results of the interaction of a series of bile salts, sodium cholate (NaC), sodium taurocholate (NaTC), sodium deoxycholate (NaDC), and sodium taurodeoxycholate (NaTDC) with trypsin under the experimental conditions of 25 °C and pH 7.0. The interactions between trypsin and the bile salts were characterized by the surface tension measurements and various spectroscopic techniques like UV-Visible absorption, steady-state fluorescence, and circular dichroism. The results of surface tension measurements reveal a strong interaction of trypsin (50 μM) with the increasing concentration of bile salts, being higher with the bile salt of greater hydrophobicity. The critical aggregation concentration of bile salts in the presence of trypsin (C 1 ) showed that the bile salts interact strongly with the trypsin in the order of NaTDC > NaDC > NaTC > NaC. UV-visible, steady-state fluorescence, and circular dichroism spectroscopic results confirmed significant unfolding of trypsin due to its interaction with the bile salts, the extent of which followed the same sequence as observed in the surface tension results. It could be concluded that the hydrophobic bile salts that show lower C 1 values and have less delocalized charge, are more effective in unfolding the trypsin. The study would help understand the hydrophobicity-driven unfolding of proteins aided by biological surfactants like bile salts and help devise efficient proteolytic enzyme-based detergent formulations and understand the role of such amphiphiles as antimicrobial agents., Competing Interests: Declaration of competing interest None., (Copyright © 2021. Background: We tested the antitumor effects of a modified E2F peptide substituting D-Arg for L-Arg, conjugated to penetratin (PEP) against solid tumor cell lines and the CCRF-leukemia cell line, alone and in combination with pemetrexed or with cisplatin. For in - vivo studies, the peptide was encapsulated in PEGylated liposomes (PL-PEP) to increase half-life and stability., Methods: Prostate cancer (DU145 and PC3), breast cancer (MCF7, MDA-MB-468, and 4T1), lymphoma (CCRF-CEM), and non-small cell lung cancer (NSCLC) cell lines (H2009, H441, H1975, and H2228) were treated with D-Arg PEP in combination with cisplatin or pemetrexed. Western blot analysis was performed on the NSCLC for E2F-1, pRb, thymidylate synthase, and thymidine kinase. The H2009 cell line was selected for an in - vivo study., Results: When the PEP was combined with cisplatin and tested against solid tumor cell lines and the CCRF-CEM leukemia cell line, there was a modest synergistic effect. A marked synergistic effect was seen when the combination of pemetrexed and the PEP was tested against the adenocarcinoma lung cancer cell lines. The addition of the PEP to pemetrexed enhanced the antitumor effects of pemetrexed in a xenograft of the H2009 in mice., Conclusions: The D-Arg PEP in combination with cisplatin caused synergistic cell kill against prostate, breast, lung cancers, and the CCRF-CEM cell line. Marked synergy resulted when the D-Arg PEP was used in combination with pemetrexed against the lung adenocarcinoma cell lines. A xenograft study using the PL-PEP in combination with pemetrexed showed enhanced anti-tumor effects compared to each drug alone.

The present study was designed to explore the hydrophobicity and concentration dependence of imidazolium based surface active ionic liquids (SAILs) effects on the structural-functional integrity of proteins. Specifically, we investigated the impact of SAILs viz. 1-octyl-3-methylimidazolium dodecylbenzenesulfonate ([OMIM][DBS]) and 1-dodecyl-3-methylimidazolium dodecylbenzenesulfonate ([DDMIM][DBS]) on activity, structure and stability of lysozyme. Activity measurements revealed that, in contrast to [DDMIM][DBS] that renders lysozyme either feebly active or inactive, [OMIM][DBS] significantly enhances the lysozyme activity in the concentration range of critical aggregation concentrations (CAC) to C s (SAIL saturation concentration of protein backbone) i.e., 0.5 mM-1.35 mM. Tensiometric results in agreement with turbidity measurements inferred significant composition and concentration dependence of the lysozyme-SAIL interactions. Spectroscopic investigations revealed that compared to destabilizing behaviour of [DDMIM][DBS], [OMIM][DBS] significantly enhances both conformational as well as thermal stability of lysozyme in the CAC to C s concentration regime. Altogether, results obtained do indicate that [OMIM][DBS], in the concentration regime of CAC to C s , serves as an efficient stabiliser with an ability to appreciably enhance the activity, thermal stability and overall conformational stability of lysozyme. Herein, we demonstrate that the catalytic performance of imidazolium-based surface-active ionic liquid (SAIL) micelles can be significantly enhanced through the addition of an appropriate type and amount of intelligently conceived amphiphile to form mixed micelles. Specifically, we show that the catalytic performance of 1-dodecyl-3-methyl imidazolium chloride (DDMIMCl) micelles toward the reductive degradation of rhodamine B (RhB), a carcinogenic dye extensively used in multiple industrial applications, can be appreciably boosted through addition of Brij56, a nonionic surfactant. Detailed kinetic investigations on the catalytic performance of pre- and post-micellar concentrations of DDMIMCl and its mixed micelles with Brij56 over various mole fractions, toward the reductive degradation of RhB, are presented. The data analyzed in light of Berezin's kinetic model suggest that the addition of Brij56 to DDMIMCl micelles significantly enhances their catalytic performance. The catalytic activity exhibited by the DDMIMCl-Brij56 (X Brij56 = 0.2) mixed micellar system is better than that reported for many state-of-the-art nanoparticle/homogenous catalysts. The results explained in light of Berezin's kinetic model are well supported by physico-chemical studies like conductometry, fluorimetry and dynamic light scattering. The presented results anticipate stimulation of extensive research activity for exploiting the mixed micellization approach as a novel avenue for modulating the catalytic performance of SAILs.

Actively proliferating cancer cells require sufficient amount of NADH and NADPH for biogenesis and to protect cells from the detrimental effect of reactive oxygen species. As both normal and cancer cells share the same NAD biosynthetic and metabolic pathways, selectively lowering levels of NAD(H) and NADPH would be a promising strategy for cancer treatment. Targeting nicotinamide phosphoribosyltransferase (NAMPT), a rate limiting enzyme of the NAD salvage pathway, affects the NAD and NADPH pool. Similarly, lowering NADPH by mutant isocitrate dehydrogenase 1/2 ( IDH1/2 ) which produces D-2-hydroxyglutarate (D-2HG), an oncometabolite that downregulates nicotinate phosphoribosyltransferase (NAPRT) via hypermethylation on the promoter region, results in epigenetic regulation. NADPH is used to generate D-2HG, and is also needed to protect dihydrofolate reductase, the target for methotrexate, from degradation. NAD and NADPH pools in various cancer types are regulated by several metabolic enzymes, including methylenetetrahydrofolate dehydrogenase, serine hydroxymethyltransferase, and aldehyde dehydrogenase. Thus, targeting NAD and NADPH synthesis under special circumstances is a novel approach to treat some cancers. This article provides the rationale for targeting the key enzymes that maintain the NAD/NADPH pool, and reviews preclinical studies of targeting these enzymes in cancers.

The rapid qualitative assessment of surface markers on cancer cells can allow for point-of-care prediction of patient response to various cancer drugs. Preclinical studies targeting cells with an antibody to "activated" matriptase conjugated to a potent toxin show promise as a selective treatment for a variety of solid tumors. In this paper, we implemented a novel technique for electrical detection of proteins on surfaces of cancer cells using multi-frequency microfluidic impedance cytometry. The biosensor, consists of two gold microelectrodes on a glass substrate embedded in a PDMS microfluidic channel, is used in conjugation with immuno-magnetic separation of cancer cells, and is capable of differentiating between bare magnetic beads, cancer cells and bead-cell aggregates based on their various impedance and frequency responses. We demonstrated proof-of-concept based on detection of "activated" matriptase proteins on the surface of cultured Mantle cells.

We present a novel method to rapidly assess drug efficacy in targeted cancer therapy, where antineoplastic agents are conjugated to antibodies targeting surface markers on tumor cells. We have fabricated and characterized a device capable of rapidly assessing tumor cell sensitivity to drugs using multifrequency impedance spectroscopy in combination with supervised machine learning for enhanced classification accuracy. Currently commercially available devices for the automated analysis of cell viability are based on staining, which fundamentally limits the subsequent characterization of these cells as well as downstream molecular analysis. Our approach requires as little as 20 μL of volume and avoids staining allowing for further downstream molecular analysis. Matriptase is a transmembrane serine protease, synthesized as an inactive single-chain zymogen on the endoplasmic reticulum and transported to the plasma membrane. Matriptase is activated in different epithelial and some B-cell malignancies and changes its conformation and activity is inhibited mainly by its endogenous inhibitor HAI-1. Activated matriptase plays a key role in tumor initiation as well as tumor progression, including invasiveness, and metastasis. To target the anti-mitotic toxin (monomethyl auristatin-E) to activated matriptase, a novel antibody to activated matriptase was conjugated with this toxin via a valine-citrulline-PABA linker. In a previous study, this antibody-toxin conjugate was found to be effective against triple negative breast cancer cell lines and xenografts, alone, or in combination with cisplatin (1). In this study, we examined the anti-tumor effect of the antibody toxin conjugate (ADC) against activated matriptase positive mantle cell lymphoma cell lines (JeKo-1, Maver, Mino, and Z138). This ADC was cytotoxic to these cell lines with IC 50 s between 5 and 14 μg/mL. The ADC also showed a dose dependent anti-tumor effect on the JeKo-1 xenograft in mice without toxicity.

The bacteria expressing New Delhi Metallo-β-lactamase-1 (NDM-1) can hydrolyze all β-lactam antibiotics including carbapenems, causing multi-drug resistance. The worldwide emergence and dissemination of gene bla NDM-1 (produces NDM-1) in hospital and community settings, rising problems for public health. Indeed, there is an urgent need for NDM-1 inhibitors to manage antibiotic resistance. Here, we have identified novel non-β-lactam ring-containing inhibitors of NDM-1 by applying a high-throughput virtual screening of lead-like subset of ZINC database. The screened compounds were followed for the molecular docking, the molecular dynamics simulation, and then enzyme kinetics assessment. The adopted screening procedure funnels out five novel inhibitors of NDM-1 including ZINC10936382, ZINC30479078, ZINC41493045, ZINC7424911, and ZINC84525623. The molecular mechanics-generalized born surface area and molecular dynamics (MD) simulation showed that ZINC84525623 formed the most stable complex with NDM-1. Furthermore, analyses of the binding pose after MD simulation revealed that ZINC84525623 formed two hydrogen bonds (electrostatic and hydrophobic interaction) with key amino acid residues of the NDM-1 active site. The docking binding free energy and docking binding constant for the ZINC84525623 and NDM-1 interaction were estimated to be -11.234 kcal/mol, and 1.74 × 10⁸ M -1 respectively. Steady-state enzyme kinetics in the presence of ZINC84525623 show the decreased catalytic efficiency (i.e., k cat / K m ) of NDM-1 on various antibiotics. The findings of this study would be helpful in identifying novel inhibitors against other β-lactamases from a pool of large databases. Furthermore, the identified inhibitor (ZINC84525623) could be developed as efficient drug candidates.

E2F1-3a overexpression due to amplification or to mutation or loss of the retinoblastoma gene, induces genes involved in DNA synthesis and leads to abnormal cellular proliferation, tumor growth, and invasion. Therefore, inhibiting the overexpression of one or more of these activating E2Fs is a recognized target in cancer therapeutics. In previous studies we identified by phage display, a novel 7-mer peptide (PEP) that bound tightly to an immobilized consensus E2F1 promoter sequence, and when conjugated to penetratin to increase its uptake into cells, was cytotoxic to several malignant cell lines and human prostate and small cell lung cancer xenografts. Based on molecular simulation studies that showed that the D-Arg penetratin peptide (D-Arg PEP) secondary structure is more stable than the L-Arg PEP, the L-Arg in the peptide was substituted with D-Arg. In vitro studies confirmed that it was more stable than the L- form and was more cytotoxic as compared to the L-Arg PEP when tested against the human castrate resistant cell line, DU145 and the human lung cancer H196 cell line. When encapsulated in PEGylated liposomes, the D-Arg-PEP potently inhibited growth of the DU145 xenograft in mice. Polo-like kinase-1 (PLK-1) plays a key role in cell cycle progression during mitosis. Overexpression/dysfunction of PLK-1 is directly associated with cancerous transformation and has been reported in different cancer types. Here, we employed high throughput virtual screening and molecular docking to screen Selleck's natural compound library against PLK-1 kinase domain. We have identified eight bioactive compounds (Apigenin, Dihydromyricetin, Diosmetin, Hesperidin, Hesperitin, Naringenin, Phlorizi, and Quercetin) as the potential inhibitors of PLK-1. Further investigation through Molecular Mechanics-Generalized Born Surface Area (MM-GBSA) calculations and 15 ns molecular dynamics simulation revealed that hesperidin formed the most stable complex with PLK-1 kinase domain. Altogether, our results indicate that hesperidin interacted strongly with the key residues of the PLK-1 active site (such as Leu59, Lys61, Lys82, Cys133, Asn181, Asp194, Leu59, Cys67, Ala80, Val114, Leu130, Leu132, Cys133, Leu139, Phe183, and Phe195) through hydrogen bonding and hydrophobic interactions. The Hesperidin-PLK-1 complex was stabilized by Gibb's free energy of -13.235 kcal/mol which corresponded to the binding affinity of 5.095 × 10 9  M -1 . This is the first study wherein hesperidin has been identified as a potential inhibitor of PLK-1. The antitumor effects of a novel antibody drug conjugate (ADC) was tested against human solid tumor cell lines and against human triple negative breast cancer (TNBC) xenografts in immunosuppressed mice. The ADC targeting activated matriptase of tumor cells was synthesized by using the potent anti-tubulin toxin, monomethyl auristatin-E linked to the activated matriptase-specific monoclonal antibody (M69) via a lysosomal protease-cleavable dipeptide linker. This ADC was found to be cytotoxic against multiple activated matriptase-positive epithelial carcinoma cell lines in vitro and markedly inhibited growth of triple negative breast cancer xenografts and a primary human TNBC (PDX) in vivo . Overexpression of activated matriptase may be a biomarker for response to this ADC. The ADC had potent anti-tumor activity, while the unconjugated M69 antibody was ineffective in a mouse model study using MDA-MB-231 xenografts in mice. Treatment of a human TNBC (MDA-MB-231) showed potent anti-tumor effects in combination with cisplatin in mice. Recent reports about the promising and tunable electrocatalytic activity and stability of nanoalloys have stimulated an intense research activity toward the design and synthesis of homogeneously alloyed novel bimetallic nanoelectrocatalysts. We herein present a simple one-pot facile wet-chemical approach for the deposition of high-quality bimetallic palladium-silver (PdAg) homogeneous nanoalloy crystals on reduced graphene (Gr) oxide sheets. Morphological, structural, and chemical characterizations of the so-crafted nanohybrids establish a homogeneous distribution of 1:1 PdAg nanoalloy crystals supported over reduced graphene oxide (PdAg-Gr). The PdAg-Gr nanohybrids exhibit outstanding electrocatalytic, catalytic, and electroanalytical performances. The PdAg-Gr samples were found to exhibit exceptional durability when subjected to repeated potential cycles or long-term electrolysis. In the CVs recorded for fuel cell reactions, viz. methanol oxidation reaction and oxygen reduction reaction, and for detoxification of environmental pollutants, viz. electroreduction of methyl iodide and chloroacetonitrile over PdAg-Gr with potential sweep rate of 25 mVs -1 , the peak potentials were observed to be just -0.221, -0.297, (vs Ag/AgCl, 3 M KCl) -1.508, and -1.189 V (vs Fc + /Fc), respectively. The potential of PdAg-Gr nanohybrid for simultaneous and sensitive electrochemical sensing and estimation of hydroxybenzene isomers with very low detection limits (0.05 μM for hydroquinone, 0.06 μM for catechol, 6.7 nM for 4-aminophenol, and 13.7 nM for 2-aminophenol) is demonstrated. Additionally, PdAg-Gr was observed to offer excellent solution-phase catalytic performance in bringing about the reduction of notorious environmental pollutant 4-nitrophenol to pharmaceutically important 4-aminophenol with an apparent rate constant ( k app ) of 3.106 × 10 -2 s -1 and a normalized rate constant ( k nor ) of 6.21 × 10 2 s -1 g -1 . The presented synthetic scheme besides being high yielding, low cost, and easy to carry out results in the production of PdAg-Gr nanohybrids with stability and activity significantly better than most of the nanomaterials purposefully designed and testified so far by various groups.

The modulation of properties of methylcellulose (MC) by cationic gemini surfactants with varying spacer lengths was studied employing tensiometry, rheometry and turbidimetry. Surface tension measurements anticipate that the gemini surfactant with longer spacer chain length saturates MC at lower concentrations owing to its greater hydrophobicity compared to shorter spacer analogues. Rheometric and turbidimetric measurements suggest that at very low concentrations of gemini surfactants, ion-dipole type of interactions between MC and gemini surfactants promote the extension of polymer chains which is manifested by an initial increase in the low shear viscosity and gelation temperature of MC-gemini surfactant systems, and lowering of turbidity. Such interactions were found to be stronger in case of 16-4-16 than 16-5-16, and almost absent in case of 16-6-16 surfactant system. However at concentrations above CAC, hydrophobic forces operative between MC and gemini surfactants were found to be more for 16-6-16 than that of 16-5-16 and 16-4-16. The final levelling of MC viscosity in presence of all the three gemini surfactants and the variation of gelation temperature suggests the solubilization of network junctions in the surfactant micelles. Moreover, the presence of gemini surfactant strongly affects the interaction of MC with a model hydrophobic drug rifampicin.The results highlight the importance of gemini surfactants and their spacer length in controlling the structural dynamics of MC and its effective use in pharmaceutical and food industry., (Copyright © 2016 Elsevier Ltd. Synthesis and structural characterization of hydrogels composed of sodium alginate, polyethylene oxide and acrylic acid with cyclodextrin as the hydrocolloid prepared at different pH values is presented. The hydrogels synthesized show significant variations in rheological properties, drug encapsulation capability and release kinetics. The hydrogels prepared at lower pH (pH 1) are more elastic, have high tensile strength and remain almost unaffected by varying temperature or frequency. Further, their Ibuprofen encapsulation capacity is low and releases it slowly. The hydrogel prepared at neutral pH (pH 7) is viscoelastic, thermo-reversible and also exhibits sol-gel transition on applying frequency and changing temperature. It shows highest Ibuprofen encapsulation capacity and also optimum drug release kinetics. The hydrogel prepared at higher pH (pH 12) is more viscous, has low tensile strength, is unstable to change in temperature and has fast drug release rate. The uses of thermostable starch hydrolytic biocatalysts are steadily increasing for the industrial application because of their obvious need for biocatalytic performance at elevated temperatures. The starch liquefaction and saccharification can be carried out simultaneously by the use of thermostable starch hydrolytic biocatalysts, thus minimizing the unit operations, time, and efforts. The cost factor hampers the industrialization of expensive soluble (free) enzymes for biocatalytic applications and the immobilization of enzymes offers promising alternative to the hurdle. The present investigation was aimed for immobilization of thermostable α-amylase using calcium alginate, and statistical optimization studies were carried out for enhanced biocatalytic performance. Initially, one-parameter at a time optimization studies were carried out for identification of significant factors influencing the immobilization. Furthermore, a statistical approach, response surface methodology, was applied for immobilization of α-amylase. The immobilized α-amylase in alginate microbeads showed enhanced stability to temperature and reusable property for up to seven cycles (with the retention of 50% initial activity). Finally, the kinetic behavior of free and immobilized enzyme showed the Km value of 1.2% and 2.6% (w/v) and Vmax of 1,020 and 1,030 U, respectively. The interaction of cetyltrimethylammoium bromide (CTAB) and its gemini homologue (butanediyl-1,4-bis (dimethylcetylammonium bromide), 16-4-16 with biocompatible polymer sodium alginate (SA) has been investigated in aqueous medium. Addition of K2CO3 influences viscoelastic properties of surfactant impregnated SA via competition between electrostatic and hydrophobic interactions. Viscosity of these polymer-surfactant systems increases with increase in concentration of K2CO3, and a cryogel is formed at about 0.5M K2CO3 concentration. The thermal stability of gel (5% SA+0.5M K2CO3) decreases with increase in surfactant concentration, a minimum is observed with increase in 16-4-16 concentration. The impact of surfactant addition on the alginate structure vis-à-vis its drug loading capability and release thereof was studied using Ibuprofen (IBU) as the model drug. The hydrogel with 16-4-16 exhibits higher IBU encapsulation and faster release in comparison to the one containing CTAB. In the first of its kind we herein report the results of our studies undertaken on the micellization behaviour of imidazolium based surface active ionic liquids (SAILs) to prove that their critical micelle concentration (cmc) can be estimated through ultraviolet-visible (UV-vis) spectroscopy without using any external probe. Tensiometric and spectrophotometric investigations of a series of freshly prepared SAILs viz. 1-octyl-3-methylimidazolium chloride ([OMIM][Cl]), 1-octyl-3-methylimidazolium dodecylsulphate ([OMIM][DS]), 1-octyl-3-methylimidazolium benzoate ([OMIM][Bz]), 1-octyl-3-methylimidazolium salicylate ([OMIM][Sc]), 1-octyl-3-methylimidazolium acetate ([OMIM][Ac]) are presented as a case study in support of the said claim. The cmcs estimated through spectrophotometric method were found to be close to the values estimated through tensiometry for the said SAILs. The cmcs for the investigated SAILS were found to vary in order of [OMIM][Cl]>[OMIM][Ac]>[OMIM][Bz]>[OMIM][Sc]>[OMIM][DS]. Three phase partitioning consists of precipitation of proteins due to simultaneous presence of ammonium sulphate and t-butanol. The technique has been successfully used for purification and refolding of proteins. There are however indications that the structures of proteins subjected to three phase partitioning are different from native structure of proteins. Taking several proteins, the present work examines the structural changes in proteins by comparing their thermal stabilities, secondary structure contents, surface hydrophobicities, hydrodynamic radii and solubilities in the presence of ammonium sulphate. The results show that while the nature or extent of structural changes may vary, in all the cases the changes are rather subtle and not drastic in nature. Three phase partitioning is a process in which mixing t-butanol with ammonium sulphate with a protein solution leads to the formation of three phases. Generally, the interfacial protein precipitate (formed between upper t-butanol rich and lower aqueous phase) can be easily dissolved back in aqueous buffers. In case of ovalbumin, this led to a precipitate which was insoluble in aqueous buffers. This precipitate when solubilized with 8 M urea and subjected to three phase partitioning under various conditions led to many refolded soluble conformational variants of ovalbumin. One of these showed trypsin inhibitory activity, had marginally higher β-sheet content and had higher surface hydrophobicity (both with respect to native ovalbumin). Scanning electron microscopy and Atomic force microscopy of this preparation showed a thread like structure characteristic of amyloid fibrils. The interaction of xanthine oxidase (XO), a key enzyme in purine metabolism, with cetyltrimethylammonium bromide (CTAB) and bis(cetyldimethylammonium)butane dibromide (C16C4C16Br2) has been studied using tensiometry, spectrofluorometry, spectrophotometry, and circular dichroism at pH 7.4 and 25 °C. The tensiometric profiles of CTAB and C16C4C16Br2 in the presence of XO exhibit a single break at a lower surfactant concentration termed as C1 compared to their CMC in the buffered solution and show the existence of interaction between the surfactants and the enzyme. The results of the multitechnique approach showed that, although both CTAB as well as C16C4C16Br2 interact with the XO, C16C4C16Br2 interacts more strongly than its conventional single chain counterpart. Fluorescence and absorption measurements revealed that, compared to CTAB, C16C4C16Br2 is more effective in unfolding the enzyme. Change in XO activity by the surfactants was in concurrence with the structural alterations monitored by circular dichroism and showed structural stabilization of XO at higher surfactant concentrations, consistent with the aggregation results. This stabilization has been explained in light of strong tendency of C16C4C16Br2 for micellar growth and membrane/water stabilization of proteins by membrane-like fragments provided by higher concentrations of C16C4C16Br2 . The results are related to the stronger electrostatic and hydrophobic forces in C16C4C16Br2, owing to the presence of two charged headgroups and two hydrophobic tails.

Overexpression of recombinant proteins in bacterial systems (such as E. coli) often leads to formation of inactive and insoluble ' inclusion bodies' . Protein refolding refers to folding back the proteins after solubilizing/unfolding the misfolded proteins of the inclusion bodies. Protein aggregation, a concentration dependent phenomenon, competes with refolding pathway. The refolding strategies largely aim at reducing aggregation and/or promoting correct folding. This review focuses on non-chromatographic strategies for refolding like dilution, precipitation, three phase partitioning and macro-(affinity ligand) facilitated three phase partitioning. The nanomaterials which disperse well in aqueous buffers are also discussed in the context of facilitating protein refolding. Apart from general results with these methods, the review also covers the use of non-chromatographic methods in protein refolding in the patented literature beyond 2000. The patented literature generally describes use of cocktail of additives which results in increase in refolding yield. Such additives include low concentration of chaotropic agents, redox systems, ions like SO4(2-) and Cl-, amines, carboxylic acids and surfactants. Some novel approaches like use of a "pressure window" or ionic liquids for refolding and immobilized diselenide compounds for ensuring correct -S-S- bonds pairing have also been discussed in various patents. In most of the patented literature, focus naturally has been on refolding in case of pharmaceutical proteins.

Precipitation of alpha chymotrypsin in the simultaneous presence of ammonium sulphate and t-butanol (three phase partitioning) resulted in preparations which showed self aggregation of the enzyme molecules. Precipitation with increasing amounts of ammonium sulphate led to increasing size of aggregates. While light scattering estimated the hydrodynamic diameter of these aggregates in the range of 242-1124 nm; Nanoparticle tracking analysis (NTA) gave the value as 130-462 nm. Scanning electron microscopy and gel filtration on Sephadex G-200 showed extensive aggregation in these preparations. Transmission electron microscopy showed that the aggregates had irregular shapes. All the aggregates had about 3× higher catalytic activity than the native enzyme. These aggregates did not differ in λ(max) of fluorescence emission which was around 340 nm. However, all the aggregates showed higher fluorescence emission intensity. Far-UV and near-UV circular dichroism also showed no significant structural changes as compared to the native molecule. Interestingly, HPLC gel filtration (on a hydroxylated silica column) gave 14 nm as the diameter for all preparations. Light scattering of preparations in the presence of 10% ethylene glycol also dissociated the aggregates to monomers of 14 nm. Both these results indicated that hydrophobic interactions were the driving force behind this aggregation. These results indicate: (1) Even without any major structural change, three phase partitioning led to protein molecules becoming highly prone to aggregation. (2) Different methods gave widely different estimates of sizes of aggregates. It was however possible to reconcile the data obtained with various approaches. (3) The nature of the gel filtration column is crucial and use of this technique for refolding and studying aggregation needs a rethink.

Solubilization and co-solubilization of triphenyls (TPs) viz., triphenylphosphine (TPP), triphenylphosphineoxide (TPPO), triphenylamine (TPA) and triphenylmethanol (TPM) were studied in various single and binary surfactant systems at 25 °C using UV-visible spectroscopy and HPLC. The solubilization capacities of different micelles towards TPs were found to be a function of the nature and structure of solubilizates, locus of solubilization, size of micelles and the nature of interactions between the solubilizate and micelles. The effect of surfactant mixing on the solubilization of TPs was evaluated using the Regular Solution Approach (RSA). The solubility enhancement of TPs within mixed micelles relative to that observed in single surfactant systems was explained in light of the structural micellar changes associated with the mixing of ionic and non-ionic surfactants. Moreover, kinetics of oxidation of TPP by hydrogen peroxide investigated in these surfactant systems was found to be sensitive to the nature of micelle and the locus of solubilization of TPP within the micelles., (Copyright © 2011 Elsevier Inc. Surfactants prevent the irreversible aggregation of partially refolded proteins, and they are also known to assist in protein refolding. A novel approach to protein refolding that utilizes a pair of low molecular weight folding assistants, a detergent and cyclodextrin, was proposed by Rozema and Gellman (D. Rozema, S.H. Gellman, J. Am. Chem. Soc. 117 (1995) 2373). We report the refolding of bovine serum albumin (BSA) assisted by these artificial chaperones, utilizing gemini surfactants for the first time. A combination of cationic gemini surfactants, bis(cetyldimethylammonium)pentane dibromide (C(16)H(33)(CH(3))(2)N(+)-(CH(2))(5)-N(+)(CH(3))(2)C(16)H(33)·2Br(-) designated as G5 and bis(cetyldimethylammonium)hexane dibromide (C(16)H(33)(CH(3))(2)N(+)-(CH(2))(6)-N(+)(CH(3))(2)C(16)H(33)·2Br(-) designated as G6 and cyclodextrins, was used to refold guanidinium chloride (GdCl) denatured BSA in the artificial chaperone assisted two step method. The single chain cationic surfactant cetyltrimethylammonium bromide (CTAB) was used for comparative studies. The studies were carried out in an aqueous medium at pH 7.0 using circular dichroism, dynamic light scattering and ANS binding studies. The denatured BSA was found to get refolded by very small concentrations of gemini surfactant at which the single chain counterpart was found to be ineffective. Different from the single chain surfactant, the gemini surfactants exhibit much stronger electrostatic and hydrophobic interactions with the protein and are thus effective at much lower concentrations. Based on the present study it is expected that gemini surfactants may prove useful in the protein refolding operations and may thus be effectively employed to circumvent the problem of misfolding and aggregation., (Copyright © 2011 Elsevier Inc. The interaction of the antioxidant Rutin with the radical DPPH (2,2-diphenyl-1-picrylhydrazyl) in presence of cationic (CTAB, TTAB, DTAB), non-ionic (Brij78, Brij58, Brij35), anionic (SDS) and mixed surfactant systems (CTAB-Brij58, DTAB-Brij35, SDS-Brij35) has been followed by spectrophotometric and tensiometric methods to evaluate the DPPH radical scavenging activity (RSA) of Rutin in these model self-assembled structures. The results show that the solubilization capacity of various single surfactant systems for both DPPH as well as Rutin followed the order cationics > non-ionics > anionic. The radical scavenging activity of Rutin in the solubilized form was higher within ionic micelles than in non-ionic micelles. However, the antioxidant exhibited enhanced activity for the radical in mixed cationic-non-ionic micelles compared with any of the single component micelles. In contrast, anionic-non-ionic mixed micelles modulated the activity of Rutin in-between that seen for pure anionic and non-ionic micelles only., (Copyright © 2010 Elsevier Inc. The interactions of bovine serum albumin (BSA) with cationic gemini surfactants alkanediyl-alpha,omega-bis(dimethylcetylammonium bromide) (designated as C(16)C(s)C(16)Br(2), s=4, 5, and 6) and single chain surfactant cetyltrimethylammonium bromide (CTAB) have been investigated with tensiometry, Rayleigh's scattering, fluorescence spectroscopy, and circular dichroism at physiological pH and 25 degrees C. The results of the multi-technique approach showed that the gemini surfactants interact more efficiently with the proteins than their conventional single chain counterparts and their efficiency increases with decrease in the length of the spacer. The saturation in interfacial tension occurred at a lower concentration in presence of BSA compared to CMC of the surfactants in absence of BSA and the concentration of gemini surfactants corresponding to interfacial saturation decreases with decrease in the spacer length. The interaction of bovine serum albumin (BSA) with cetyltrimethylammonium bromide (CTAB), C(16)C(4)C(16)Br(2), Brij58, and their binary mixtures has been studied using tensiometry, spectrofluorometry, and circular dichroism at physiological pH and 25 degrees C. The tensiometric profiles of CTAB and C(16)C(4)C(16)Br(2) in the presence of BSA exhibit a single break at a lower surfactant concentration termed as C(1) (concentration corresponding to saturation of the interface) compared to their critical micelle concentration (CMC) in the buffered solution. However, for Brij58, CTAB+Brij58, and C(16)C(4)C(16)Br(2)+Brij58, two breaks were observed, first at the critical aggregation concentration (CAC), corresponding to onset of interaction with BSA and the second at C(1) corresponding to saturation of the interface. The interaction of CTAB+Brij58 and C(16)C(4)C(16)Br(2)+Brij58 mixtures with the BSA solution is discussed in terms of competition between surfactant-surfactant and surfactant-BSA interactions. CTAB+Brij58 and C(16)C(4)C(16)Br(2)+Brij58 mixtures show nonideality with respect to mixed micelle formation, which is reflected in their interaction with the BSA. The interaction of CTAB+Brij58 with BSA decreases with increase in the mole fraction of CTAB in the mixture, whereas in C(16)C(4)C(16)Br(2)+Brij58 the reverse is the case. The results of the present study may prove fruitful in optimizing the properties of surfactant-protein mixtures relevant for many formulations.

Mixed surfactants may improve the performance of surfactant-enhanced solubilization of drugs and thus can serve as the tool for increased bioavalaibility, controlled drug release, and targeted delivery. Solubilization of Naproxen by micellar solutions at 25 degrees C using single and mixed surfactant systems was measured and compared. Solubilization capacity determined with spectrophotometry and tensiometry has been quantified in terms of molar solubilization ratio, micelle-water partition coefficient, and locus of solubilization. Cationic surfactants exhibited higher solubilization capacity than nonionics and anionics, the efficiency increasing with chain length. Mixing effect of surfactants on mixed micelle formation and solubilization efficiency has been discussed in light of regular solution approximation (RSA). Equimolar cationic-nonionic surfactant combinations showed better solubilization capacity than pure cationics or nonionics, which, in general, increased with increase in hydrophobic chain length. Equimolar cationic-nonionic-nonionic ternary surfactant systems exhibited intermediate solubilization efficiency between their single and binary counterparts. Use of RSA has been extended, with fair success, to predict the partition coefficient of ternary surfactant systems using data from binary mixtures. The theoretical micelle-water partition coefficients calculated from the geometric mean equation compared well with experimental values. Locus of solubilization of NAP in different micellar solutions was probed by UV-visible spectroscopy.

Mixed micelle formation and interfacial properties of aqueous binary and ternary combinations of hexadecyltrimethylammonium bromide (C(16)Br), hexadecylbenzyldimethylammonium chloride (C(16)BzCl) and polyoxyethylene (20) cetyl ether (Brij58) at 25 degrees C in 30 mM aqueous NaCl have been studied in detail employing tensiometric and fluorimetric techniques. The micellar and adsorption characteristics like composition, activity coefficients, mutual interaction parameters and free energy of micellization have been estimated using the theoretical approaches of Clint, Rosen, Rubingh, Blankschtein et al., Rubingh-Holland and Maeda. A comprehensive account of the comparative performance of these models on the selected cationic-cationic-nonionic surfactant mixtures at constant ionic strength has been presented. The Blankschtein model predicted lower synergism than from Rubingh's method because it neglects the contribution due to steric interaction between surfactant head groups of different sizes and charges. Free energy of micellization calculated using Maeda's approach, which employs interaction parameter and micellar mole fraction from Rubingh's model as inputs, shows good correlation with that calculated from commonly used phase separation model. The present study also reveals that the modified Rubingh-Holland method along with the Rosen's model can be applied to analyze the interfacial characteristics of ternary surfactant mixtures with a fair degree of success thereby widening the domain of applicability of this model.

Water solubility enhancements of polycyclic aromatic hydrocarbons (PAHs), viz., naphthalene, anthracene and pyrene, by micellar solutions at 25 degrees C using two series of surfactants, each involving two cationic and one nonionic surfactant in their single as well as equimolar binary and ternary mixed states, were measured and compared. The first series was composed of three surfactants, benzylhexadecyldimethylammonium chloride (C16BzCl), hexadecyltrimethylammonium bromide (C16Br), and polyoxyethylene(20)mono-n-hexadecyl ether (Brij-58) with a 16-carbon (C16) hydrophobic chain; the second series consisted of dodecyltrimethylammonium bromide (C12Br), dodecylethyldimethylammonium bromide (C12EBr), and polyoxyethylene(4)mono-n-dodecyl ether (Brij-30) with a 12-carbon (C12) chain. Solubilization capacity has been quantified in terms of the molar solubilization ratio, the micelle-water partition coefficient, the first stepwise association constant between solubilizate monomer and vacant micelle, and the average number of solubilizate molecules per micelle, determined employing spectrophoto-, tensio-, and flourimetric techniques. Cationic surfactants exhibited lesser solubilization capacity than nonionics in each series of surfactants with higher efficiency in the C16 series compared to the C12 series. Increase in hydrophobicity of head groups of cationics by incorporation of ethyl or benzyl groups enhanced their solubilization capacity. The mixing effect of surfactants on mixed micelle formation and solubilization efficiency has been discussed in light of the regular solution approximation (RSA). Cationic-nonionic binary combinations showed better solubilization capacity than pure cationics, nonionics, or cationic-cationic mixtures, which, in general, showed increase with increased hydrophobicity of PAHs. Equimolar cationic-cationic-nonionic ternary surfactant systems showed lower solubilization efficiency than their binary cationic-nonionic counterparts but higher than cationic-cationic ones. In addition, use of RSA has been extended, with fair success, to predict partition coefficients of ternary surfactant systems using data of binary surfactants systems. Mixed surfactants may improve the performance of surfactant-enhanced remediation of soils and sediments by decreasing the applied surfactant level and thus remediation cost.

Mixed micelle formation and interfacial properties of aqueous binary surfactant combinations of dodecyltrimethylammonium bromide (C12TAB) and tetraethyleneglycol mono-n-dodecyl ether (C12E4) at 30 degrees C in absence and presence of sodium propionate (NaPr) have been investigated. The critical micelle concentration, aggregation number, micropolarity and interfacial adsorption have been quantitatively estimated by surface tension and steady-state fluorescence measurements. The micellar and adsorption characteristics like composition, activity coefficients and mutual interaction parameters have been estimated following different theoretical treatments like that of Clint, Rubingh, Rodenas, Maeda, Blankschtein and Rosen. The analysis reveals very small mole fraction of cationic surfactant in both the mixed micelles and mixed monolayer inspite of synergism. Blankschtein's model predicts a continuous decrease in synergism due to the salt effect of NaPr; Rubingh's approach, on the contrary, indicates an increase in it above 30 mM of NaPr concentration. Aggregation number variation with NaPr indicates the same. Mixed monolayer shows better synergism compared to that in mixed micelles which increases with the addition of sodium propionate above 30 mM concentration.

Laparoscopic cholecystectomy was first performed in Buraydah Central Hospital, Al Qassim in September 1993. Since more than 125 open cholecystectomies were performed annually, there was a need to be able to deliver this type of service to the patients in our hospital. Continued audit is needed to ensure that the results with low complication rates achieved in tertiary centers are reproduced by the surgeons at district level. Our study of 340 laparoscopic cholecystectomies performed between September 21, 1993 and September 20, 1995 describes the results with incidence of complications, conversion to open procedure, mean operation time and the extend of postoperative stay in a district hospital. Our complication rates compare favorably with the results achieved in laparoscopic cholecystectomy in many centers. Data obtained by us suggests that laparoscopic cholecystectomy is safe and effective for the treatment of cholecystitis, and this technique can be introduced safely in a district general hospital in Saudi Arabia.
