Abstract

As the percentage of antibiotic-resistant bacteria increases, alternative treatment options to deprive their pathogenicity are needed. Bacterial pathogens like Vibrio parahaemolyticus manipulate the host proteome by effector protein-mediated AMPylation. By analyzing diverse MS quantification methods in combination with different infection strategies, this thesis established a proteomic profiling approach to identify bacterially induced host AMPylation targets under physiological conditions. Da der Anteil antibiotikaresistenter Bakterien zunimmt, werden Behandlungsmöglichkeiten benötigt, um sie ihrer Pathogenität zu berauben. Bakterielle Pathogene wie V. parahaemolyticus manipulieren das Wirtsproteom durch AMPylierung. Durch die Kombination von MS-Quantifizierungsmethoden mit unterschiedlichen Infektionsstrategien wurde in dieser Arbeit ein proteomischer Profiling-Ansatz etabliert, der bakteriell induzierte Wirts-AMPylierungsziele unter physiologischen Bedingungen identifiziert.
