2. Red blood cell count and cystatin C as the specific biomarkers for diabetic retinopathy from diabetes mellitus: a case–control study.

Diabetic retinopathy (DR) is a common microvascular complication of diabetes mellitus (DM). However, the blood physiological indicators for this eye disease are less reported. Here, we systemically investigated healthy controls, DR patients and DM patients from 2014 to 2022 in Sichuan Provincial People’s Hospital (Chengdu, China). We found the leukocyte count, urea, and direct bilirubin in the DR patients were significantly increased (P < 0.001) compared with the controls. The ratios of aspartate aminotransferase/alanine transaminase, total bilirubin, indirect bilirubin, and red cell distribution width were lower in the DR patients than in the controls (P < 0.001). Notably, Red blood cell count (mean [SD] = 4.54 [0.67] 10E12/L, OR = 0.75) was significantly decreased (P < 0.001), and cystatin C level (mean [SD] = 1.42 [1.13] mg/L, OR = 1.90) was significantly increased in the DR patients compared with the controls (P < 0.001). However, these two indicators were not significantly different between the DM patients and the controls (P = 0.87 and P = 0.14, respectively). 3. Parallel Evolution at the Regulatory Base-Pair Level Contributes to Mammalian Interspecific Differences in Polygenic Traits.

Parallel evolution occurs when distinct lineages with similar ancestral states converge on a new phenotype. Parallel evolution has been well documented at the organ, gene pathway, and amino acid sequence level but in theory, it can also occur at individual nucleotides within noncoding regions. To examine the role of parallel evolution in shaping the biology of mammalian complex traits, we used data on single-nucleotide polymorphisms (SNPs) influencing human intraspecific variation to predict trait values in other species for 11 complex traits. We found that the alleles at SNP positions associated with human intraspecific height and red blood cell (RBC) count variation are associated with interspecific variation in the corresponding traits across mammals. These associations hold for deeper branches of mammalian evolution as well as between strains of collaborative cross mice. While variation in RBC count between primates uses both ancient and more recently evolved genomic regions, we found that only primate-specific elements were correlated with primate body size. We show that the SNP positions driving these signals are flanked by conserved sequences, maintain synteny with target genes, and overlap transcription factor binding sites. This work highlights the potential of conserved but tunable regulatory elements to be reused in parallel to facilitate evolutionary adaptation in mammals. [ABSTRACT FROM AUTHOR]

4. Preoperative blood-based nutritional biomarkers as significant prognostic factors after intravesical BCG therapy in patients with non-muscle-invasive bladder cancer.

The aim of this study was to investigate the prognostic role of blood-based nutritional biomarkers, including red blood cell (RBC count), hemoglobin (Hb), total protein (TP), albumin, the serum albumin to globulin ratio (AGR) and the prognostic nutritional index (PNI) in patients who underwent intravesical treatment for non-muscle invasive bladder cancer (NMIBC). A total of 501 NMIBC patients who received intravesical Bacillus Calmette-Guerin (BCG) treatment following transurethral resection of bladder tumor (TURBT) were included. The optimal cutoff values for these nutrition-based indicators were determined using receiver operating characteristic curve analysis. We observed a significantly higher recurrence-free survival (RFS) rate in patients with elevated levels of RBC count, Hb, TP, and albumin. Cox univariate and multivariate Cox regression analyses demonstrated that serum albumin (P = 0.002, HR = 0.51, 95%CI: 0.33–0.78), RBC count (P = 0.002, HR = 0.50, 95%CI: 0.32–0.77), TP (P = 0.028, HR = 0.62, 95%CI: 0.41–0.95), Hb (P = 0.004, HR = 0.53, 95%CI: 0.33–0.84), AGR (P = 0.003, HR = 0.46, 95%CI: 0.27–0.76) and PNI (P = 0.019, HR = 0.56, 95%CI: 0.35–0.91) were significant independent factors predicting RFS. These cost-effective and convenient blood-based nutritional biomarkers have the potential to serve as valuable prognostic indicators for predicting recurrence in NMIBC patients undergoing BCG-immunotherapy. Introduction: Many studies have shown that the functional adaptation of immigrants to high-altitude is closely related to oxygen transport, inflammatory response and autonomic nervous system. However, it remains unclear how human attention changes in response to hypoxia-induced neurophysiological activity during high-altitude exposure. Methods: In the present study, we analyzed the relationship between hypoxic-induced neurophysiological responses and attention networks in 116 immigrants (3,680 m) using an attention network test to simultaneously record electroencephalogram and electrocardiogram in combination with specific routine blood markers. Results: Our analysis revealed that red blood cells exert an indirect influence on the three attention networks, mediated through inflammatory processes and heart rate variability. Discussion: The present study provides experimental evidence for the role of a neuroimmune pathway in determining human attention performance at high- altitude. Our findings have implications for understanding the complex interactions between physiological and neurocognitive processes in immigrants adapting to hypoxic environments. [ABSTRACT FROM AUTHOR]

6. Observational and Genetic Evidence for Bidirectional Effects Between Red Blood Cell Traits and Diastolic Blood Pressure.

BACKGROUND Previous studies have found associations of red blood cell (RBC) traits (hemoglobin and RBC count) with blood pressure; whether these associations are causal is unknown. METHODS We performed cross-sectional analyses in the Lifelines Cohort Study (n = 167,785). Additionally, we performed bidirectional 2 sample Mendelian randomization (MR) analyses to explore the causal effect of the 2 traits on systolic (SBP) and diastolic blood pressure (DBP), using genetic instrumental variables regarding hemoglobin and RBC identified in UK Biobank (n = 350,475) and International Consortium of Blood Pressure studies for SBP and DBP (n = 757,601). RESULTS In cross-sectional analyses, we observed positive associations with hypertension and blood pressure for both hemoglobin (odds ratio [OR] = 1.18, 95% confidence interval [CI]: 1.16–1.20 for hypertension; B = 0.11, 95% CI: 0.11–0.12 for SBP; B = 0.11, 95% CI: 0.10–0.11 for DBP, all per SD) and RBC (OR = 1.14, 95% CI: 1.12–1.16 for hypertension; B = 0.11, 95% CI: 0.10–0.12 for SBP; B = 0.08, 95% CI: 0.08–0.09 for DBP, all per SD). MR analyses suggested that higher hemoglobin and RBC cause higher DBP (inverse-variance weighted B = 0.11, 95% CI: 0.07–0.16 for hemoglobin; B = 0.07, 95% CI: 0.04–0.10 for RBC, all per SD). Reverse MR analyses (all per SD) suggested causal effects of DBP on both hemoglobin (B = 0.06, 95% CI: 0.03–0.09) and RBC (B = 0.08, 95% CI: 0.04–0.11). No significant effects on SBP were found. 7. Paired logistic regression analysis of influencing factors for vascular crisis after repair of oral and maxillofacial tumors with free tissue flaps

Objective To investigate the influencing factors of vascular crisis after oral and maxillofacial tumor free tissue flap transplantation. Methods A retrospective analysis was performed on all patients who underwent free tissue flap transplantation and developed vascular crisis in the surgical ward of head and neck cancer in a grade A specialized hospital. Forty-six possible influencing factors were collected using 1:1 matching according to surgeons, operation time, sex and age of patients from patients without vascular crisis from 2015-2020 in this ward during the same period. SPSS 26.0 statistical software was used for univariate and multivariate logistic regression analyses of the data. Results A total of 158 patients were enrolled, including 79 in the crisis group and 79 in the pairing group. Univariate analysis was performed for each variable. Paired logistic regression analysis showed that only postoperative blood potassium (P = 0.048, OR = 3.118, 95% CI: 1.008-9.641) and preoperative and postoperative red blood cell count differences (P = 0.004, OR = 4.53, 95% CI: 1.609-12.750) were statistically significant. Conclusion High blood potassium levels and red blood cell count differences before and after surgery were risk factors for vascular crisis.

To describe the characteristics of multiple myeloma (MM) by retrospectively analyzing data from patients with MM in hematology, nephrology, and orthopedic departments, we selected inpatients diagnosed with MM for the first time who were admitted to the hematology department of Taizhou Hospital of Zhejiang Province between January 1, 2017, and June 30, 2021 ( n = 110 ) and those admitted to the nephrology ( n = 48 ) and orthopedic ( n = 42 ) departments of Taizhou Hospital of Zhejiang Province between January 1, 2010, and June 30, 2021. For patients with MM initially diagnosed in the nephrology or orthopedic departments, age- and sex-matched patients without MM were randomly selected in a 1:1 ratio. Conditional logistic regression models were used to assess the risk of MM in hospitalized patients in the nephrology and orthopedic departments. This study included 200 patients, with a median age of 65 years. There were differences among patients in the three departments with respect to fracture, degree of anemia, proteinuria levels, red blood cell (RBC) count, hemoglobin levels, total protein (TP) levels, albumin levels, and laboratory indicators of renal function. TP levels > 68.45 g/L were independent risk factors for patients in the nephrology department (odds ratio (OR) = 1.01, 95% confidence interval (CI) = 1.001–1.024, P = 0.04), and RBC count < 3.79 × 1012/L was an independent risk factor for orthopedic inpatients (OR = 0.49, 95% CI 0.268–0.913, P = 0.02). Therefore, nephrologists should pay attention to the TP level of hospitalized patients to facilitate the early identification of MM in patients with chronic kidney disease. Orthopedic surgeons should pay attention to RBC counts in patients with fractures, and patients with low RBC counts should avoid unnecessary surgery and move to specialist care as early as possible. 9. Machine learning-based model for predicting the esophagogastric variceal bleeding risk in liver cirrhosis patients.

Background: Liver cirrhosis patients are at risk for esophagogastric variceal bleeding (EGVB). Herein, we aimed to estimate the EGVB risk in patients with liver cirrhosis using an artificial neural network (ANN). Methods: We included 999 liver cirrhosis patients hospitalized at the Beijing Ditan Hospital, Capital Medical University in the training cohort and 101 patients from Shuguang Hospital in the validation cohort. The factors independently affecting EGVB occurrence were determined via univariate analysis and used to develop an ANN model. Results: The 1-year cumulative EGVB incidence rates were 11.9 and 11.9% in the training and validation groups, respectively. A total of 12 independent risk factors, including gender, drinking and smoking history, decompensation, ascites, location and size of varices, alanine aminotransferase (ALT), γ-glutamyl transferase (GGT), hematocrit (HCT) and neutrophil-lymphocyte ratio (NLR) levels as well as red blood cell (RBC) count were evaluated and used to establish the ANN model, which estimated the 1-year EGVB risk. The ANN model had an area under the curve (AUC) of 0.959, which was significantly higher than the AUC for the North Italian Endoscopic Club (NIEC) (0.669) and revised North Italian Endoscopic Club (Rev-NIEC) indices (0.725) (all P < 0.001). Decision curve analyses revealed improved net benefits of the ANN compared to the NIEC and Rev-NIEC indices. Conclusions: The ANN model accurately predicted the 1-year risk for EGVB in liver cirrhosis patients and might be used as a basis for risk-based EGVB surveillance strategies. [ABSTRACT FROM AUTHOR]

- Abstract
Ceftriaxone, a widely used antibiotic, is one of the most common drugs to cause drug-induced immune hemolytic anemia. In this report, we describe the effect of ceftriaxone on red blood cell parameters (low red blood cell count, low hematocrit, and high erythrocyte index values) in two pediatric patients without clinical symptoms of hemolytic anemia. Although automated hematology analyzers have helped to detect incorrect results, a peripheral blood smear examination was necessary for recognizing the erythrocyte agglutinins caused by ceftriaxone. Serological testing was not possible, but the resulting drug-induced antibodies mimicked cold agglutinins in the first patient and warm agglutinins in the second patient. Timely reactions and corresponding laboratory procedures prevented potential complications due to drug administration. Abstract Background Gestational Diabetes Mellitus (GDM) is an underlying cause of maternal and newborn morbidity and mortality all around the world. Timely diagnosis of GDM plays an important role in reducing its adverse consequences and burden. This study aimed to determine diagnostic accuracy of multiple indicators in complete blood count (CBC) test for early prediction of GDM. Methods In this prospective cohort study, the data from 600 pregnant women was analyzed. In the study sample, the two-step approach was utilized for the diagnosis of GDM at 24–28 weeks of gestation. We also used the repeated measures of hemoglobin (Hb), hematocrit (Hct), fasting blood sugar (FBS) and red blood cell count (RBC) in the first and early second trimesters of pregnancy as the longitudinal multiple indicators for early diagnosis of GDM. The classification of pregnant women to GDM and non-GDM groups was performed using a statistical technique based on the random-effects modeling framework. Results Among the sample, 49 women (8.2%) were diagnosed with GDM. 13. Hepatotoxic effect of lead and hepatoprotective effect of Hydrilla verticillata on hepatic transcriptional and physiological response in edible fish Labeo rohita.

Lead tops the list of developmental toxicants released by industry into the environment according to Commission for Environmental Corporation. Hydrilla verticillata is an aquatic weed and rich source of variable nutrients and chemical constituents like saponins, vitamins, minerals, antioxidants, amino acids, detoxifying agents, etc. This weed is used in a beneficial way to detoxify lead in fish model. The fish were treated with sublethal concentration of lead to induce liver damage and fed with supplementary feed containing 20% of hydrilla dry powder. The present work revealed that lead accumulation is highly toxic to liver. Lead toxicity increased the expressions of cytochrome P450 1 A (CYP1A) and cytochrome P450 3 A (CYP3A) when compared to control. The metal binding stress proteins, heat shock proteins (Hsp60 & Hsp70) and metallothionein were also upregulated due to lead toxicity. Lead intoxicated fish exhibited reduced δ-aminolevulinic acid dehydratase activity in the blood, leading to reduced red blood cell count and changes in cell morphology. The lead toxicity also decreases the level of liver marker enzymes alanine transaminase, aspartate transaminase in serum. In hydrilla supplemented protective group, there was amelioration of these changes. Administration of supplementary feed to the lead intoxicated protective group significantly decreased the expressions of CYP1A, CYP3A, Hsp60, Hsp70 and metallothionein genes. The red blood cell count in lead intoxicated fish was maintained almost normal due to the protective action of Hydrilla verticillata in the diet. The observed increase in the enzymes, which further confirmed the protective effect of Hydrilla verticillata against lead induced toxicity. [ABSTRACT FROM AUTHOR]

MetS was diagnosed using a modified version of the Chinese Diabetes Society 2004 criteria. Demographic and clinical characteristics were stratified by MetS and compared. Logistic regression analyses were performed to calculate the odds ratios (ORs) of myometrial invasion.Results: The prevalence of MetS was 18.8% (89/473). RBC and Hb levels were significantly higher in EEC patients with MetS. RBC and Hb values increased linearly with the number of MetS components. The OR of the MetS risk increased across the RBC quartiles (OR=2.212, 95% confidence interval [Cl]=1.125-4.350, P˂0.05). Among EEC patients with MetS, the OR of deep myometrial invasion increased with the decrease of the RBC quartiles (P=0.037). Similarly, in EEC patients with obesity and hyperglycemia or with ≥ 2 MetS criteria, the lowest RBC level (Q1) was a risk factor for deep myometrial invasion compared with the reference RBC group (Q4, P˂0.05). However, this trend was not evident in EEC patients without MetS and its components.Conclusion: EEC with MetS and its components had higher RBC and Hb levels. The association between the erythrocyte parameters and myometrial invasion differed with MetS. Abstract Background Cardiac morphogenesis is a dynamic and complex process that involves a complex interaction of many genetic and environmental factors. Ventricular septal defects (VSDs) are the commonest congenital heart defects (CHDs), accounting for ~ 40% of all cardiac malformations. In Pakistan, the prevalence of VSD is increasing (4–6 in 1000 live births). In the current study, we aimed to determine the pattern of different hematological parameters and various risk factors in VSDs in local pediatric patients. We recruited the clinically diagnosed VSD children (n = 125) from various hospitals. The diagnosis was made based on echocardiography, size, number, and exact location of the defect. Hematological parameters, chemical pathological assays, and liver function analysis were performed. The blood group distribution and various risk factors were also assessed. The statistical analysis was done using the SPSS (IBM statistics version 22) software. Results The results showed that for RBCs, 20% of patients in category of 0 to 3 months are above normal range; for WBCs, 33.3% of patients are above normal range in category of 4–5 years and 12–14 years. For hemoglobin, highest percentage of patients was observed below normal range; 30% of patients in category of 0 to 3 months, 40% of patients in category 4–9 months, 35.2% of patients in category of 10 months–3 years, and 33.3% of patients in category of 12–14 years were below normal range. For platelet count, 5.66% patients were below normal range and 16.9% were above normal range. For prothrombin time (PT) and activated partial thromboplastin time (APTT) more than 90% patients were in normal range value. Elevated ALP level and significantly lower albumin levels were observed. In age range of 13–14 years, 50% patients were below range for both calcium and serum creatinine. The prevalence of cousin marriages was 62.3%, about 60% mothers used antibiotics during pregnancy, B + ve and O + ve had the highest frequencies, and most of the patients were seen in age group of 2–35 months. Conclusion All tested parameters show divergence from normal values their predictive capabilities of VSDs. To the best of our knowledge, the present study is the first to report data on hematological parameters and demographic risk factor associated with VSDs, in the Pakistani children. This data may have implication on the characterization and diagnosis of VSDs as well as on the assessment of related risk factors.

Background: Gestational Diabetes Mellitus (GDM) is an underlying cause of maternal and newborn morbidity and mortality all around the world. Timely diagnosis of GDM plays an important role in reducing its adverse consequences and burden. This study aimed to determine diagnostic accuracy of multiple indicators in complete blood count (CBC) test for early prediction of GDM.Methods: In this prospective cohort study, the data from 600 pregnant women was analyzed. In the study sample, the two-step approach was utilized for the diagnosis of GDM at 24-28 weeks of gestation. We also used the repeated measures of hemoglobin (Hb), hematocrit (Hct), fasting blood sugar (FBS) and red blood cell count (RBC) in the first and early second trimesters of pregnancy as the longitudinal multiple indicators for early diagnosis of GDM. The classification of pregnant women to GDM and non-GDM groups was performed using a statistical technique based on the random-effects modeling framework.Results: Among the sample, 49 women (8.2%) were diagnosed with GDM. In the first and early second trimester of pregnancy, the mean HcT, Hb and FBS of women with GDM was significantly higher than non-GDMs (P < 0.001). The concurrent use of multiple longitudinal data from HcT, Hb, RBC and FBS in the first and early second trimester of pregnancy resulted in a sensitivity, specificity and area under the curve (AUC) of 87%, 70% and 83%, respectively, for early prediction of GDM.Conclusions: In general, our findings showed that the concurrent use of repeated measures data on Hct, Hb, FBS and RBC in the first and early second trimester of pregnancy might be utilized as an acceptable tool to predict GDM earlier in pregnancy. Purpose: Exercise can lead to various hematological changes and it may be effects on some blood variables.The purpose of present study was to investigate the effects of resistance training with different order onhematological changes (include HGB, HCT, RBC, WBC, MCV, MCH, MCHC, RDW, PLT and MPV) in overweightwomen (BMI= 25-30 Kg/m2).Methods: Thirty-one overweight students (Weight: 71.91 ± 8.28 kg and Height: 160 ± 0.05 cm) wereobjectively selected and randomly divided into three groups: the experimental group 1 (perform exercisesfrom large to small muscles, n= 10), the experimental group 2 (perform exercises from small to large muscles,n= 11) and the control group (without any training program, n= 10). Resistance training consists of 6 weeks(3 sessions per week). The hematological variables were measured in resting conditions before experimentand again after completion of the training period.Results: Findings using by one-way ANOVA revealed there were no significant differences in any of thehematologic variables levels between three groups.Conclusion: In general, results showed that medium-term resistance training program and so changing thesequence of performance of resistance exercise – none of them – do not have any effect on hematologicresponses.

Background: Oral squamous cell carcinoma (OSCC) is the second most common cancer in India and is more prevalent in South Asian countries. It affects any age group depending upon the risk factors. They are mainly caused due to tobacco and its related products but sometimes may be idiopathic. There are very few studies reporting the association of anemia with OSCC. Materials and Method: A Retrospective study was conducted in the Oral Oncology and Oral Medicine Department of Saveetha Dental College from June 2019 to March 2020. Case records of 55 patients who were clinically and histopathologically proven as OSCC were reviewed. 15 patients were included in the final study. The data was evaluated and descriptive analysis was done to assess correlation and association. Results: Predominant age group of study population was 61-70 years (40%), female predilection, most common site left buccal mucosa (13.3%) and right buccal vestibule (13.3%), moderately differentiated OSCC (66.1%) and T1N0M0 (20%), T1N1M0 (20%) was more commonly reported. No tobacco habits or other risk factors were present in the study group. The RBC and Hemoglobin was reduced and moderately differentiated carcinoma was associated with decreased hematological parameters. Conclusion: Anemia can be a predisposing risk factor for OSCC. 19. Adjusted preoperative variables to predict perioperative red blood cell transfusion in coronary artery bypass grafting.

Background: The variables for predicting blood transfusion perioperatively are not completely clear in coronary artery bypass grafting (CABG) patients. Objectives: To construct a comprehensive model to predict perioperative RBC transfusion in patients undergoing isolated CABG using adjusted preoperative variables. Methods: Perioperative data of 1253 patients who underwent isolated CABG by the same surgical team were collected from April 2018 to March 2019. Logistic regression analyses were used to establish equations to construct two models for predicting intraoperative and postoperative RBC transfusions, respectively. All significant variables included in the two models were combined to form a comprehensive model to predict perioperative RBC transfusion. Area under the receiver operating characteristic (ROC) curve (AUC) was used to evaluate the discriminatory power of the models. Results: The total RBC transfusion rate for CABG patients during hospitalization was 29.05%. The rate of intraoperative and postoperative RBC transfusions was 6.9% and 26.7%, respectively. Eight variables in a total of 30 risk factors constituted the intraoperative prediction model, 12 variables constituted the postoperative prediction model, and 13 variables for the combined model. The AUC of the three models were 0.87, 0.82, and 0.83, respectively, demonstrating moderate discriminatory power for RBC transfusion during the intraoperative, postoperative, and perioperative periods. Conclusion: The comprehensive model combined with all variables of predicting intraoperative and postoperative RBC transfusion is feasible for predicting perioperative RBC transfusion. Objective: To explore the effects of different blood transfusion methods on autoimmune hemolytic anemia (AIHA). Methods: 64 cases of children with autoimmune hemolytic anemia diagnosed and treated in the hematology department from January 2017 to December 2018 of our hospital were selected as the research subjects and were divided into observation group and control group with 32 cases in each groups accorded to the different blood transfusion methods. The observation group were given washed erythrocyte infusion treatment, and the control group were given non-wash erythrocyte (suspended red blood cell) infusion treatment, and the transfusion effects were recorded in the both groups. Results: 4 weeks post-treatment, the total effective rate in the observation group was significantly higher than that in the control group (100.0% vs. 87.5%, P<0.05). The red blood cell count and hemoglobin at 4 weeks post-treatment in two groups were significantly higher than pretherapy, and the observation group was significantly higher than the control group (P<0.05). The oxygen intake, mechanical ventilation, and hospital stay of the observation group were significantly shorter than those of the control group (P<0.05). The incidence of allergic reactions, fever reactions, and purpura in the observation group was significantly lower than that in the control group (3.1 % vs. 21.9 %, P<0.05). Conclusions: The washing red blood cell infusion for children with autoimmune hemolytic anemia can promote the body's red blood cell count and hemoglobin to return to normal, reduce the occurrence of adverse reactions, improve the treatment effect and promote the rehabilitation of children. Abstract Objectives Clinical laboratory reference intervals used in a specific area should be derived from the local population as they are influenced by many factors. The purpose of this quantitative cross sectional study was to establish hematological reference intervals for healthy adults in Asmara and to determine whether the currently used reference interval do represent the adult population in the city. In addition, the established reference intervals were compared to findings from similar studies conducted in selected countries in Africa. Results There was a significant difference between males and females in the reference intervals for erythrocyte count, hemoglobin, hematocrit, mean cell volume, mean cell hemoglobin, mean cell hemoglobin concentration and differential white blood cell count. All the evaluated hematological analytes were found to be higher in males than in females except for platelet count. The out of range percentage for the parameters extends from 3.5 to 46.7%; with red blood cell count having the lowest while mean cell volume having the highest out of range percentage. The results indicated that the currently used reference interval does not represent the population in Asmara and are different from those obtained elsewhere in Africa.

Background: Manipulation of training variables such as intensity, volume and training session’s frequency could affect the exercise compatibility and dynamicity these carry out different physiological adaptations. The purpose of this study is determining the effect of two types high intensity training (HIIT) on some hem rheological factors in young overweight and obese women. Methods: 24 girls with age 25/75±1/18 y, height: 161/29± 4/23cm, weight 74/51±6/27 kg, fat 30/70± 2/23%, BMI 28/66±1/89 kg/m2 voluntarily participated in this study and randomly divided into three experimental HIIT1 group (n=8), HIIT2 group (n=8) and control group (n=8). The subjects of the group HIIT1 in each session running 30 seconds in four sets with 19 to 20 the rate of perceived exertion (RPE) Borg scale and two minutes of rest between sets. HIIT2 group running four sets of 60 s with the same intensity and four minutes rest between sets, training protocol performed three times a week and as long as four weeks. 48 hours before and 48 hours after exercise, blood samples were taken. For analyzing data, use Mixed between-within Subjects analysis of variance in level of (p>0.05). Results: Results showed in levels of RBC , MCV, MCH , hematocrit, hemoglobin wasn’t significantly variation and also difference between groups was not significant (p>0.05). Background: Iron deficiency in polycythaemia vera (PV) may impact the validity of the haematocrit (HCT), since HCT is red blood cell count (RBC) × mean corpuscular volume (MCV). Objectives: To investigate (a) the effect of microcytosis on HCT, (b) the erythrocyte sedimentation rate (ESR) as a possible additional diagnostic marker for PV. Material and methods: This study included 182 subjects: 39 with PV, 27 with essential thrombocythemia (ET) and 116 suspected of myeloproliferative neoplasm (MPN) with a secondary cause for either thrombocytosis or erythrocytosis. Results: Patients with PV had significantly lower ratio of MCV and serum ferritin compared to MPN suspects. A good correlation of RBC versus HCT was found for PV and MPN subjects when individuals with microcytosis were excluded (R2 =.87 in PV and R2 =.82 in MPN suspects). We found a specificity of 98% and a sensitivity of 37% for ESR <2 mm in the diagnosis of PV. Conclusion: The RBC may more precisely reflect the total red cell mass and accordingly the hypercoagulable state of the PV patient, which is integrated in the ESR. A combination of RBC and ESR is proposed as a novel tool to substitute the Hb concentration and the HCT in the diagnosis of PV. [ABSTRACT FROM AUTHOR]

28. Effect of cold agglutinins on red blood cell parameters in a trauma patient: a case report.

The presence of cold agglutinins (CAs) in samples intended for complete blood count (CBC) using automated haematology analysers might cause serious preanalytical errors. In this report we describe the case of a 90-year old female patient admitted to the Emergency department following trauma injuries. A blood testing on admission revealed surprisingly low red blood cell count (0.99 x 1012/L), low haematocrit (0.102 L/L) which did not correlate with haemoglobin concentration (100 g/L), and high erythrocytes indices (mean corpuscular haemoglobin, 101 pg; mean corpuscular haemoglobin concentration, 980 g/L). In the second sample, after repeated collection, almost equal results were observed. Blood smear examination under the microscope revealed clusters of erythrocytes. Cold agglutinins presence was suspected and, in order to get valid results, sample was warmed to 37 °C. Correction of CBC was observed. Furthermore, we performed some additional analysis to confirm the presence of CAs in this patient. The aim of this report was to present the laboratory findings in a case of CAs and propose a laboratory procedure for whole blood samples with suspected CAs. [ABSTRACT FROM AUTHOR]

In this study, to inhibit the activation of T lymphocytes formed in the experimental arthritis, tolerogenic dendritic cells were aimed to be obtained by the genetical modification of dendritic cells with CTLA4-KDEL overexpression in endoplasmic reticulum. Then, RA created animals treated with tolDCs and the effect of treatment on blood parameters were investigated. Dogs are of immense social, psychological and economic importance in Nigeria and are severely affected by African trypanosomosis. However, the prevalence of canine African trypanosomosis (CAT) in Nigeria is underreported and the identification of the parasites relies mostly on basic morphological characteristics under the microscope, which could be misleading. The present study was carried out to determine the prevalence and characterize trypanosomes isolated from dogs in South east Nigeria. A cross-sectional survey was carried out to determine the prevalence and molecular identification of trypanosomes in dogs in Enugu North Senatorial Zone (ENSZ), South east Nigeria. Dogs (n = 450) were randomly sampled, their blood collected and some characteristics such as sex, breed, sampling location, season and age duly noted. The blood samples were screened for trypanosomosis using standard trypanosome detection techniques. Trypanosome-positive blood samples were spotted on FTA® cards for molecular identification using nested Tubulin-PCR, ITS-PCR, TgsGP-PCR, and DNA sequencing. Some hematological parameters of the dogs such as packed cell volume (PCV), total leucocyte count (TLC), red blood cell count (RBC) were also determined. Of the 450 dogs sampled, 51 dogs were positive for trypanosomes with a prevalence rate of 11.3% (95% CI = 0.087–0.146). Trypanosoma brucei was the predominant trypanosome species infecting dogs in the study area. T. congolense, T. evansi, and T. vivax were also identified. The prevalence of canine trypanosomosis was significantly associated with season (χ2 = 13.821, df = 1, P = 0.0001) and the sampling location (χ2 = 6.900, df = 2, P = 0.032) while sex, breed, and age were not. This study determined haematological changes in Wistar rats experimentally infected with local strains of Trypanosoma congolense and Trypanosoma brucei brucei. Forty-five Wistar rats between 10 – 12 weeks old weighing between 210 – 240 g were used. The Wistar rats were randomly divided into four groups (A, B, C and D), with the infected groups (B, C and D) having 10 rats each, while the uninfected control group (A) had 15 rats. Packed cell volume (PCV) is commonly used to assess and monitor red blood cell count in animals, but the results can be altered if inappropriate ratios of anticoagulant/blood are used. The purpose of this study was to determine the effect of ideally filled, overfilled, and underfilled K3 ethylenediaminetetraacetic acid (EDTA) tubes with various volumes of healthy dog blood on centrifuged PCV. Six milliliters of blood was obtained from 94 blood donors each. Initial distribution was injected into two nonheparinized microhematocrit tubes. The remainder was instilled into 1.3 mL K3 EDTA spray-dried tubes as 1.5 mL, 1.3 mL, 0.75 mL, 0.5 mL, and 0.25 mL aliquots. Normality was determined using the D’agostino–Pearson method and by visual examination of histograms. Data were analyzed using a repeated-measures analysis of variance with post hoc testing using Tukey’s test. There is a statistically significant decrease in the PCV between all groups with progressive underfilling of tubes (P < .0001). The closest difference is between 1.5 and 1.3 mL (P = .0138). Our study suggested that underfilling K3 EDTA tubes significantly and negatively influences the PCV in healthy dogs. Using underfilled K3 EDTA tubes result in a lower PCV compared with directly filled microhematocrit tubes without anticoagulant.

The study sought to execute a systematic review and meta-analysis to describe the toxicological implications associated with exposures of humans and laboratory animals to Spent Crankcase Oil (SCO). Databases like PubMed, Scopus, Science Direct, Google Scholar, Web of Science, and PlosOne were searched systematically for all data that assessed the effects of SCO on humans and animals. For each parameter involved in the meta-analysis (those with extractable data), mean, standard deviation, the sample size was extracted for both exposure groups and control. This was then used to compute the standardized mean difference (SMD). Statistical analysis and forest plots were done with RevMan 5.3 software. Twenty-eight (28) studies fulfilled the pre-specified criteria for eligibility. Fourteen (14) of the studies were used for the meta-analysis, which included a total of 1243 subjects from different human epidemiological occupational exposure studies and animal experimental studies. The meta-analysis revealed that SCO exposure caused a significant reduction in the body weight of animals (n = 5, SMD; − 1.2; 95% CI; (− 1.78, − 0.67), p = 0.0001, I(2) = 22%), and in the red blood cell count (n = 5, SMD; − 1.28; 95% CI; (− 2.18, − 0.38, p = 0.02); I(2) = 78%) and haemoglobin (n = 4, SMD; − 1.12, 95% CI; (− 2.71, 0.46); p = 0.16; I(2) = 89%) in animal models. While there was a significant elevation of the aspartate amino transferase (AST) (n = 6, SMD; 0.76; 95%CI; (0.41, 1.11), p = 0.0001, I(2) = 89%), alkaline phosphatase (ALP) (n = 5, SMD; 1.92; 95% CI; (0.02, 3.83), p = 0.05, I(2) = 92%), and creatinine (n = 4, SMD = 1.56; 95% CI; (0.05, 3.07), p = 0.04, I(2) = 90%) concentrations in comparison to the control. On the other hand, there was a non-significant effect on the alanine amino transferase (ALT) (n = 5, SMD; 1.13; 95% CI; (− 0.37, 2.62); p = 0.14; I(2) = 92%), urea (n = 4, SMD; 1.23; 95% CI; (− 1.18, 3.65), p = 0.32, I(2) = 94%), packed cell volume (PCV) (n = 5, SMD; 0.10; 95% CI; (− 0.36, 0.56), p = 0.67; I(2) = 47%); and the haemoglobin (n = 6; SMD; − 0.74; 95% CI; (− 1.73, 0.26), p = 0.15; I(2) = 89%) concentrations. Oxidative stress, heavy metals bioaccumulation, immunotoxic, genotoxic, and carcinogenic effects were also in the list of findings. The toxicological implications associated with SCO exposure points to the need for immediate establishment of policies that regulate the disposal of spent crankcase oil in the environment. Ploidy level affects both the cell size and metabolic rate (MR) of organisms. The present study aimed to examine whether ploidy levels cause differences in cell surface area (SA), MR and metabolic scaling. The resting MR (RMR), red blood cell SA (SARBC), red blood cell count (RBCC), gill SA (GSA), and ventilation frequency (VF) were measured in diploid and triploid common carp with different body masses (M). The results showed that both M and ploidy level affected the RMR, GSA, VF, and SARBC, with interactions between M and ploidy level. The triploids had larger SARBC but lower RBCC than those of the diploids. The SARBC increased weakly but significantly with increasing M, by an exponent of 0.043, in the triploids but did not increase in the diploids. The RMR of the triploids and diploids scaled with M, by exponents of 0.696 and 1.007, respectively. The RMR was higher in the triploids than the diploids. The GSA scaled with M, with an exponent of 0.906 in the triploids and an exponent of 1.043 in the diploids. The VF scaled with M by an exponent of − 0.305 in the triploids but showed no correlation with M in the diploids. The larger SARBC and RMR and smaller scaling exponents of both the GSA and VF of the triploids were consistent with the finding that the bR was smaller in the triploids than in the diploids. This suggests that the ploidy-induced changes of SA and SA scaling affect the metabolic scaling of fish.

No Brasil, as infecções por hemoparasitas representam uma das principais causas de busca por assistência veterinária em cães. Infecções por bactérias da ordem Rickettsiales que incluem Ehrlichia sp. e Anaplasma sp. tem uma grande participação nessas buscas. Foi objetivo deste estudo elucidar aspectos epidemiológicos regionais e hematológicos de infecções por Anaplasma sp. e Ehrlichia sp, muitas vezes assintomáticas, em cães no município de Jataí-GO. Os dados deste estudo foram baseados em amostras de sangue de cães atendidos no Hospital Veterinário da Universidade Federal de Jataí, em Jataí, Goiás, entre os anos de 2005 e 2017. A observação microscópica da presença de mórulas de Ehrlichia sp. e Anaplasma sp. foi utilizada como critério de positividade. 5.015 animais foram atendidos nesse período e 691 (13,78%) deles foram positivos para a presença destes hemoparasitas. Entre esses animais, 9,23% (463/5015) possuíam mórulas de Ehrlichia sp., e 4,37% (219/5015) apresentavam mórulas típicas de Anaplasma sp., enquanto 0,18% (9/5015) possuíam mórula de ambas as espécies. As fêmeas representaram 57,89% (400/691) dos animais positivos e os machos 42,11% (291/691). 35,02% (242/691) dos animais tinham menos de 1 ano de vida, 54,13% (374/691) tinham entre 1 e 8 anos, e 10,85% (75/691) tinham mais de 8 anos. As fêmeas, com menos de um ano de idade e coinfectadas, apresentaram as alterações mais significativas nos aspectos hematológicos, hematócrito (29%), contagem de hemácias (3 milhões/mm3), contagem de plaquetas (40.000/mm3) e contagem total de leucócitos (16.000 mil/mm3), evidenciando anemia e trombocitopenia. Animais infectados com Anaplasma sp. e Ehrlichia sp., ao mesmo tempo, apresentaram trombocitopenia mais intensa, evidenciada em machos entre 1 e 8 anos (30.000/mm3). As infecções por Anaplasma sp. causaram uma diminuição severa na média da contagem de glóbulos vermelhos, evidenciada em machos (3 milhões/mm3) e fêmeas com menos de 1 ano (3,79 milhões/mm3).

Two preparations of iron supplements have been developed with vitamins, Irovit-1 and Irovit-2 containing 1 mg/mL and 2 mg/mL elemental iron respectively in complex form. Clinical trials of Irovit-1 and Irovit-2 have been conducted randomly on selected anemic males and females from urban population of Karachi, an improvement in hemoglobin levels were estimated for the treatments with Irovit-1 (10 mg Fe/day) and Irovit-2 (20 mg Fe/day). Both preparations have been found to be effective in increasing hemoglobin, Hematocrit and red blood cell count. The mean hemoglobin levels were observed increased 0.73 ± 0.25 g/dL and 1.10 ± 0.30 g/dL, after 5 weeks by oral intake of Irovit-1 and Irovit-2, respectively.

Objectives: Clinical laboratory reference intervals used in a specific area should be derived from the local population as they are influenced by many factors. The purpose of this quantitative cross sectional study was to establish hematological reference intervals for healthy adults in Asmara and to determine whether the currently used reference interval do represent the adult population in the city. In addition, the established reference intervals were compared to findings from similar studies conducted in selected countries in Africa. Results: There was a significant difference between males and females in the reference intervals for erythrocyte count, hemoglobin, hematocrit, mean cell volume, mean cell hemoglobin, mean cell hemoglobin concentration and differential white blood cell count. All the evaluated hematological analytes were found to be higher in males than in females except for platelet count. The out of range percentage for the parameters extends from 3.5 to 46.7%; with red blood cell count having the lowest while mean cell volume having the highest out of range percentage. The results indicated that the currently used reference interval does not represent the population in Asmara and are different from those obtained elsewhere in Africa. [ABSTRACT FROM AUTHOR]

Body condition and haematological indices provide powerful information when assessing wildlife health. Reference intervals for these indices can facilitate wildlife management, and would benefit initiatives to save the Critically Endangered geometric tortoise (Psammobates geometricus). We collected data from 126 geometric tortoises to establish baseline values reflecting variation over four seasons (spring 2000 to winter 2001) and among three groups (female, male and juvenile). We measured body condition index (BCI; mass to shell volume), blood urea nitrogen (BUN), plasma chloride, packed cell volume (PCV), haemoglobin concentration (Hb) and red blood cell count (RBC), and used PCV, Hb and RBC to calculate erythrocyte indices. BCI correlated poorly with haematological measures but had a strong inverse relationship with BUN. BCI did not vary among groups, but all groups had low condition indices in autumn, the end of the dry season. High BUN and chloride values in autumn indicate dehydration, particularly in males. Males had the highest PCV, Hb and RBC values, especially during summer and autumn when they moved long distances, likely pursuing mates. Female and juvenile indices were similar, except that female Hb was higher than juvenile Hb, possibly to meet female energy needs associated with their large size and reproductive metabolism. Low Hb levels in winter coincided with low temperatures and reduced movements. Our results illustrate how intrinsic and extrinsic factors influence the physiology of geometric tortoises and provide reference intervals to monitor their health. [ABSTRACT FROM AUTHOR]

This experiment was conducted to investigate the effect of coat colour on heat stress among West African Dwarf sheep. A total number of twenty-four West African Dwarf sheep with different coat colour types were used for the experiment. Rectal temperature (RT), respiratory rate (RR) and pulse rate(PR) were taken on each animal before sunrise and when they returned from grazing in the afternoon during late dry season ( January-March) and early rainy season (April June ). Blood samples were collected for evaluation of packed cell volume (PCV), red blood cell count (RBC), white blood cell count (WBC), plasma sodium (Na+) and potassium (K+). Coat colour had significant (P0.05) affected by variation in coat colour, sex and season. From the results obtained, it can be concluded that coat colour type had significant effect on heat stress in WestAfrican Dwarf sheep.

Background: Cervicofacial cellulitis is a severe infection and is responsible for a major disruption of host homeostasis. The aim of this work was to describe the haematological and biochemical variations of cervico-facial cellulitis. Methods: This was a cross-sectional study conducted from 1 January to 31 December 2020. All patients hospitalized for cervico-facial cellulitis of dental origin were included in the study. The usual parameters of descriptive statistics were estimated for each variable. Results: Our sample consisted of 166 patients. The average age of patients with cervicofacial cellulitis in our study was 39.9 ± 15.39 years. Red blood cell count was low in 55.5% of patients and hyperleukocytosis was noted in 89.16% of patients. Platelets were normal in 43.14% of patients and 78.43% of patients had a low haematocrit (haemodilution). Anemia was noted in 64.7% of patients. In addition, 61.0% of patients had elevated uricemia and 31.17% had elevated creatinemia. Conclusion: Cellulitis is still a common condition in less privileged environments. Its diagnosis can be guided by clinical and biological findings. Consistent education of the population on oral health, and the control of biological disorders that result from it, are sufficient to greatly reduce its prevalence and the appearance of complicated forms.

43. Enteral Vitamin B12 Supplementation Is Effective for Improving Anemia in Patients Who Underwent Total Gastrectomy

Background: This study aimed to investigate the efficacy of enteral supplementation of vitamin B12 for vitamin B12 deficiency in patients who underwent total gastrectomy for gastric cancer. Methods: The study enrolled 133 patients who underwent total gastrectomy for gastric cancer at Kochi Medical School. Clinical data were obtained to investigate associations between vitamin B12 supplementation and vitamin B12 levels. Vitamin B12 deficiency was defined as serum vitamin B12 less than 200 pg/mL. Baseline characteristics and changes in hematological variables, including vitamin B12 levels, were examined. Results: Vitamin B12 deficiency was present in 71.4% of the 133 patients. Vitamin B12 levels at 3, 6, and 12 months after enteral supplementation were 306 pg/mL, 294 pg/mL, and 367 pg/mL, respectively, which were all significantly higher than those before supplementation (p < 0.001 for all comparisons). The median red blood cell count at 3, 6, and 12 months after enteral supplementation were 380 × 104/mm3, 394 × 104/mm3, and 395 × 104/mm3, respectively, which were all significantly higher than those before supplementation (p = 0.020, p = 0.001, and p = 0.003, respectively). Vitamin B12 levels at 3, 6, and 12 months after supplementation were significantly higher in patients supplemented enterally than those supplemented parenterally (p < 0.001 for all comparisons). Conclusions: Vitamin B12 deficiency was found in 71.4% of postoperative patients who underwent total gastrectomy for gastric cancer, and enteral vitamin B12 supplements might be effective to improve anemia in these patients.

The aim of this study was to determine the biochemical and haematological profiles of Kalahari Red goats reared in the tropical environment. The Kalahari Red goats were imported into Nigeria from South Africa by the Federal University of Agriculture Abeokuta, Nigeria. The study was carried out on fifty animals of both sexes. The animals were managed under semi-intensive system where they were allowed to go out and graze on paddocks during some part of the day and brought back to the pens in the evening. The goats were fed with concentrate and grazed on sown pastures of Chloris gayana and Stylosanthes amata. Water Rwas given ad-libitum. The serum biochemical analysis was determined using Randox analysis kits. The sex of each animal was noted and recorded. The packed cell volume, haemoglobin concentration, red blood cell count, white blood cell count, neutrophils, lymphocytes, monocytes, eosinophils and basophils were determined as haematological parameters while total protein, albumin, globulin, sodium, potassium, calcium and phosphorus were determined in serum analysis. The management, haematological, serum biochemical and reproductive parameters of rabbits reared in the 3 zones (Okigwe, Orlu and Owerri) of Imo State were investigated. Data for this study were collected through questionnaire, on-the-farm assessment and fluid sample (blood and semen) from the rabbits. The hutch/cage type of housing is the most prevalent in Imo State, constituting 73.58 %. Most rabbit farmers in Imo State (45.28 %) combine concentrate and forage in rabbit feeding, 47.17 % of the rabbit house/environment were fairly clean, 30.19 % dirty and 22.64 % were clean. Rabbits with ages ranging from 5 – 8 months recorded average live weight of 1.91 kg, while annual average kindling and litter size of rabbit does reared in Imo State were 5.67 and 7.43, respectively; with average litter survival rate of 60.16 % between parturition and weaning. Haematological analysis of the adult rabbits recorded 27.80 %, 9.56 g/dl and 3.40 x 106/ml in packed cell volume, haemoglobin concentration and red blood cell count, respectively, which were lower than normal reference values. Total serum protein, globulin and urea values were lower than normal reference values recording 4.44 g/dl, 1.57 g/dl and 16.56 mg/dl, respectively. Semen parameters obtained from adult rabbit bucks were inferior to semen parameters of adult rabbit bucks reared in similar geographical locations and environment, but under standard management practices. Poor bioavailability of inorganic trace elements used in broiler nutrition and their subsequent release into the environment via poultry excereta is major cause of biological and environmental concern. Hence, the need to explore the use of Chelated trace minerals in broiler diets. This study was carried out to evaluate the effect of supplemental inorganic and chelated Cu, Zn and Mn on haematology and serum biochemistry of broiler chickens. A 42-day feeding trial was conducted using a total of 300 Arbor acre broiler chickens. The birds were allocated to five dietary groups. Each group had six replicates containing 10 birds. High feed cost resulting from the use of conventional feed ingredients represents a major challenge in broiler production in South-East Nigeria. Feed constitutes 70% of the cost of production of broiler chickens hence, the replacement of unconventional feed ingredients holds the key to sustainable poultry production. EnglishAnemia can be defined as a reduction in one or more of the principal parameters of red blood cells (hemoglobin concentration, hematocrit, or red blood cell count) below the minimum values required for adequate tissue oxygenation. It is a very frequent entity in the general population. The diagnostic strategy to distinguish between the multiple causes of anemia starts with an adequate medical record and complete physical examination. A diagnosis is made based on the signs and symptoms associated with anemic syndrome in addition to laboratory test data: complete blood count with the main red blood cell indices, including the reticulocyte count, and a comprehensive biochemical study. From a practical point of view, it is useful to classify anemias based on the morphological characteristics of red blood cells, information provided by automated counts and supplemented with peripheral blood smears.

Summary Thyroid storm (TS) is a life-threatening condition that may suffer thyrotoxic patients. Therapeutic plasma exchange (TPE) is a rescue approach for TS with acute hepatic failure, but it should be initiated with careful considerations. We present a 55-year-old male patient with untreated Graves’ disease who developed TS. Severe hyperthyroidism and refractory atrial fibrillation with congestive heart failure aggregated to multiple organ failure. The patient was recovered by intensive multimodal therapy, but we had difficulty in introducing TPE treatment considering the risk of exacerbation of congestive heart failure due to plasma volume overload. In addition, serum total bilirubin level was not elevated in the early phase to the level of indication for TPE. The clinical course of this patient instructed delayed elevation of bilirubin until the level of indication for TPE in some patients and also demonstrated the risk of exacerbation of congestive heart failure by TPE. Learning points: Our patient with thyroid storm could be diagnosed and treated promptly using Japan Thyroid Association guidelines for thyroid storm. Delayed elevation of serum bilirubin levels could make the decision of introducing therapeutic plasma exchange difficult in cases of thyroid storm with acute hepatic failure. The risk of worsening congestive heart failure should be considered carefully when performing therapeutic plasma exchange.

A 12-week feeding trial was conducted to evaluate the effects of replacing fish meal by chicken haemoglobin powder in largemouth bass diets. Four isonitrogenous (48%) and isolipidic (12%) diets were formulated to replace 0% (control), 9.80%, 19.61% and 29.41% of fish meal with chicken haemoglobin powder. Each diet was fed to triplicate groups of fish (initial weight: 49.50 ± 0.07 g) twice daily. The fish specific growth rate was significantly reduced when diet replacement level was up to 19.61%, which may be related to the feed intake and apparent digestibility coefficient of protein and amino acids. Meanwhile, the feed efficiency ratio and protein efficiency ratio were not significantly decreased until the replacement level up to 29.41%. The activity of CCP was significantly reduced when 19.61% of fish meal was replaced. Meanwhile, the activity of lysozyme and serum protein content was significantly altered in fish fed with the diets up to 29.41% replacement. Additionally, the red blood cell count and haemoglobin content was significantly decreased when the replacement level was up to 19.61%. In overall, the diet with 9.80% of chicken haemoglobin powder was more suitable for largemouth bass.
