12. β-Carotene 15,15’ Oxygenase-1 (BCO1) and β-Carotene 9,10’ Oxygenase-2 (BCO2) Distribution in Cells From Rat Liver and Intestine

Abstract

β-carotene-15, 15'-oxygenase 1 (BCO1) and β-carotene 9',10'-oxygenase 2 (BCO2) are the two known enzymes that metabolize carotenoids in vertebrates. The enzymes are particularly crucial both in the intestine where digestion begins and liver where nutrients are further metabolized. Although presence of both enzymes has been confirmed in liver and intestine, studies on subcellular localization of the enzymes in situ are sparse. Recent studies in liver by Shmarakov et al (2010) and Raghuvanshi (2010) show higher levels of BCO1 in the hepatic stellate cells (HSCs) which store retinoids in lipid droplets than hepatocytes which take retinoids up from chylomicron remnants in the blood. Furthermore, another recent study shows presence of stored retinoids in stellate-like cells of the small intestine (Senoo et al, 2013) indicating possible similar enzyme expression as liver. Thus this study investigated immunolocalization of BCO2 in normal rat liver and both enzymes in normal rat intestine with immunohistochemistry, immunofluorescence and western blot. BCO2 was uniformly distributed in the hepatocytes of liver with no distinct immunoreactivity in stellate cells. BCO2 showed striking colocalization with mitochondria in hepatocytes and also very minor presence in cytosol. In intestine, mucosal protein concentrations of BCO1 were highest in jejunum whereas BCO2 showed greatest presence in duodenum which tapered to the ileum. In whole intestine, immunohistochemistry showed strong BCO1 immunoreactivity in the duodenum, particularly in Brunner's glands. Both BCO1 and BCO2 showed diffuse presence along epithelia with strong immunoreactivity in endothelial cells and certain epithelial cells which warrant further investigation as intestinal retinoid storage cells.

Abstract

beta-Carotene oxygenase 2 cleaves beta-carotene asymmetrically at non-central double bonds of the polyene chain, yielding apocarotenal molecules. The hypothesis tested was that apocarotenoids are able to stimulate transcription by activating retinoic acid receptors (RARs). The effects of long- and short-chain apocarotenals and apocarotenoic acids on the activation of RARalpha and RARbeta transfected into monkey kidney fibroblast cells (CV-1) were investigated. We synthesized or purified beta-apo-8'-carotenoic acid (apo-8'-CA), beta-apo-14'-carotenoic acid (apo-14'-CA), beta-cyclocitral (BCL), beta-cyclogernanic acid (BCA), beta-ionone (BI), beta-ionylideneacetaldehyde (BIA) beta-ionylideneacetic acid (BIAA) and a C13 ketone, beta-apo-13-carotenone (C13). None of the apocarotenoids tested showed significant transactivation activity for the RARs when compared with all-trans retinoic acid (RA). The results suggest that biological effects of these apocarotenoids are through mechanisms other than activation of RARalpha and beta.
