25 páginas, 6 figuras, 2 tablas, Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele., This work was funded by a grant (EADB) from the EU Joint Programme – Neurodegenerative Disease Research. - Abstract
Sodium borosilicate glass fibers pulled from the melt were plastic coated "on-line" to preserve the fiber strength. By the use of tapered nozzles within the coating applicator the coating could be made concentric around the fiber, and a 40-μm concentric coating was found to be sufficient for the fiber to withstand a mild abrasion test with no deterioration in strength. Strength measurements made in ambient conditions showed that the fiber had a uniform breaking strain of 4% elongation. The same fiber measured at liquid nitrogen temperatures had a breaking strain of 14%, even after extensive handling, which is close to the theoretical glass cohesive strength.

- Abstract
We have defined a DNA sequence that behaves as an RNA polymerase II termination signal by using the human HeLa cell transient expression system. Surprisingly, this sequence is tripartite, including part of the coding region of the sea urchin H2A histone gene together with two separate sequences in the 3' flanking region of the gene. We demonstrate that this signal functions both in its normal gene environment and also when placed within the human alpha-globin gene. However, we have failed to detect a discrete 3' terminus. Rather, our data indicate the presence of an extremely heterogeneous series of nonpolyadenylated RNAs. These heterogeneous nonpolyadenylated RNAs are stable when transcribed from the intact histone gene but are highly unstable within the human alpha-globin gene. This provides evidence for the role of poly(A) in the stability of mRNA.

- Abstract
1. There is considerable disagreement in the literature concerning the manner in which eggs are fertilized and reach the exterior in chaetognaths, even in studies on the same species. This study makes use of observations on populations of living animals maintained in the laboratory, and of material fixed at known stages of the egg-laying process, and provides photographic evidence to illustrate the points enumerated below.2. There is a single duct, serving both as an oviduct and sperm duct in the female reproductive system of the chaetognath Sagitta hispida, which runs from the gonopore anteriorly along the outer border of each ovary.3. Following fertilization eggs slowly pass through the germinal epithelium (crescent) through pores much smaller than their own diameter into this duct, and displace the sperm in it.4. The eggs, which become cylindrically compressed, are extruded very rapidly from the duct to the exterior by a combination of muscular forces, and deposited in a mucous matrix onto a surface to which the animal is attached.

Collection and culture of the large lobate ctenophore Mnemiopsis mccradyiMayer is described, including the requirements for successful development of larvae. Particular attention must be given to the collection of these delicate animals, the handling and provision of live microzooplankton of suitable size for the larvae, and the provision of food densities for the adults which neither stimulate “wasteful” feeding nor limit their growth. Although these ctenophores will ingest detritus and algal cells in high concentration, they lost weight at the same rate as starved individuals unless provided with living zooplankton. Under optimum conditions, specimens would lay eggs within 13 days of their own birth. By the 17th day they laid eggs daily, and had produced an average of 8,000 eggs within 23 days after birth. The maximum number of eggs laid by a single wild individual within 24 h after being brought into the laboratory was 10,000. Their high fecundity, rapid generation time, and ability to self-fertilize help to explain their sudden appearance in bloom proportions at periods of high food concentration in the environment, often referred to in the literature.

28. Food Habits of the Southern Channel Catfish (Ictalurus Lacustris Punctatus) in the Des Moines River, Iowa

The stomach contents of 912 channel catfish (769 containing food) taken in a short section of the Des Moines River from September, 1940, to October, 1941, are analyzed. The physical and biotic characteristics of the study area are described; a partial list of the fishes present together with comments on their importance and relative abundance is included. The channel catfish is omnivorous, as is revealed by a review of the pertinent literature and by this study. A wide variety of organisms is eaten (some 50 families of insects alone are represented–these are listed). Insects and fish serve as staple foods, plant seeds are taken in season, and various other items are eaten in limited numbers. The principal groups of foods (insects, fish, plants, and miscellaneous) are analyzed volumetrically, by frequency of occurrence, and numerically. In the area studied, catfish grow at a rate of about 4 inches a year during the first 3 years of life (determined by length-frequency analysis). These natural size groups are utilized to establish the relationship between size and food habits. Young fish feed almost exclusively on aquatic insect larvae–chiefly midges, blackflies, mayflies, and caddis flies. In fish from 4 to 12 inches long insects continue to make up the bulk of the food, but at progressively greater size larger insects (mayflies and caddis flies) are eaten with increasing frequency and dipterans are of less importance than in the smaller size group; small fish and plant seeds become significant items of diet. In catfish more than 12 inches long, fish and large insects are of major importance, but many seeds and other items are taken. Although fewer insects are eaten by larger fish, those taken are more diversified and include a higher percentage of terrestrial forms. Plant foods, chiefly of terrestrial origin, show the most striking seasonal trends of any of the food organisms. Seeds of the American elm in particular are taken in great numbers in May and June. Terrestrial insects are seasonal in appearance, being eaten at times of the principal flights. Aquatic dipterans are consumed consistently and in large numbers in the spring but progressively less frequently during the summer and fall; Trichoptera, however, are infrequently eaten in the spring but are of increased incidence in the summer and fall. These trends reflect changes in the numbers of organisms available. The numbers of the various species of forage fishes consumed are closely correlated with their relative abundance in the area. Several organisms which have been reported as important foods by other investigators (filamentous algae, microcrustaceans, crayfish, mollusks) are taken in insignificant quantity here, probably because these are rare in the Des Moines River. In periods of low, relatively clear water, forage fish are eaten in sharply increased numbers–a reflection of their greater vulnerability. Feeding appears to be most active during the period from sundown until about midnight. Between 50° and 94° F., temperature does not seem to inhibit feeding, but in the winter the fish rarely feed. The available evidence indicates that adults do not feed during the breeding season. In normal years reproduction of channel catfish is highly successful in prairie rivers. Efforts to increase production should be directed toward improvement of food supply and shelter facilities.

- Abstract
Bottom trawling by the U. S. Fish and Wildlife Service motor vessel Cisco in Lake Superior in 1952–1953 revealed a large population of a tiny whitefish, Coregonus (Prosopium) coulteri, which has been reported previously only from northwestern North America. The hiatus in range, from Lake Superior to the Columbia River basin, is the greatest known for a North American freshwater fish. Although minor structural differences characterize the disjunct populations of the pygmy whitefish, these are not deemed worthy of nomenclatorial recognition. Comparisons with related species indicate that the pygmy whitefish is distinctive in the small size, large scales, few vertebrae, few pyloric caeca, and in other characters. The pygmy whitefish is widely distributed in Lake Superior, especially in semi-protected bays, such as Keweenaw Bay which yielded about 68 percent of the 1,623 specimens caught. The bathymetric range was 10 to 49 fathoms, with maximum abundance at the 25- to 39-fathom interval. Average length of fish increased progressively with water depth, chiefly because the number of yearlings declined from 100 percent at 10–14 fathoms to none at 45–49 fathoms. The average total length of pygmy whitefish caught by trawling was 3.4 inches (range 1.2 to 5.7). Extraordinarily slow growth was revealed by the examination of scales. Two fish from Keweenaw Bay, both nearing the end of their eighth growing season, were only 5.4 inches long. Compared to Keweenaw Bay, growth rate was about the same near Laughing Fish Point, faster in the Apostle Islands (and in Bull and McDonald Lakes, Montana), and slower in Siskiwit Bay, Isle Royale. Females grew more rapidly than males after the second year and had a longer life span. All male pygmy whitefish were mature at the age of 2 years and a total length of 3.6 or more inches. Most females were mature at 3 years and 4.2 inches; all older females were mature. Mean egg production was 362 (range, 93 to 597) per fish and 26 per gram of total weight for fish from 3.4 to 5.9 inches long. Spawning in 1953 occurred sometime in November or December. Crustacea (principally ostracods and amphipods–copepods in the young) occurred in 106 of 112 pygmy whitefish stomachs and made up 77 percent of the total food volume. When available, fish eggs appear to be important in the diet. Other cold-water fishes–cottids, ninespine sticklebacks, smelt, and four species of coregonines–were the most frequent associates of the pygmy whitefish. Lake trout and trout-perch were also taken with it at the same stations or in the same trawl hauls. Its closest relative in Lake Superior, the round whitefish, was not an ecological associate.

1. The first description is provided of the act of copulation of a planktonic chaetognath which leads to successful insemination and fertilization. It consists of a behavioral sequence in which partners maintain contact at their head ends while engaging in violent movements which result in the transfer of spermatophores.2. Acts of self-insemination in the same species are also described, as well as experiments in which self-insemination and successful fertilization are induced.3. The consequences of attachment of spermatophores to various parts of the body are described and compared to observations on the benthic genus Spadella.4. Experiments showed 2% of batches of eggs laid by individuals isolated prior to maturity produced hatchings, suggesting that self-fertilization, although uncommon, does occur naturally.5. Experiments indicated that one insemination was effective for only a single batch of eggs.6. The similarities between copulation in Spadellaand Sagittaare discussed and the possibility that self-fertilization presents a short-term survival mechanism for populations at low density is suggested.

H. van Thiel demonstrated only 16 years ago that the text of Longus's novel rests on two manuscripts, Laur. conv. sopp. 627 (s. xiii2) and Vat. gr. 1348 (s. xvi1). The descriptinevertheless yield historical information, and so it is as well to establish their relationships. Three scholars who knew them at first hand have tried: A. Kairis in his edition (Athens 1932), H. Dörrie in De Longi Achillis Tatii Heliodori memoria(Göttingen 1935), and E. Vilborg in his edition of Achilles Tatius (Gothenburg 1955). None of them arrived at the truth. Perhaps the same applies to me, because I have contented myself with collating at most book i and checking a few passages elsewhere; but I can give reasons for my disagreement.I will first set out my stemma. As everyone's symbols conflict, I will use none except F for the Laurentianus and V for the Vaticanus, but in my discussion I will abbreviate the shelfmarks. Relationships that I do not discuss are adequately treated by Kairis, Dörrie, or Vilborg.

Studies have been carried out on the use of octamer oligonucleotides tailed with different base analogues as primers in cycle sequencing reactions. 5-Nitroindole tails improved the performance as primers of a number of octamers. A tail length of three or four 5-nitroindole residues significantly increased the sequencing signal intensity for almost all primers. The use of incomplete libraries of tailed octamer primers for primer walking is discussed.

THE chaetognaths have long been known as active carnivores1, though knowledge of their feeding behaviour is limited to qualitative observations on the nature of their food, and mode of operation of their mouthparts. Nothing is known of their rate of food intake. The main, reason for the lack of information on this and almost every aspect of their physiology is stated2to be their small size and inability to withstand removal from their natural environment for more than a few hours. Such information would be of interest since chaetognaths are very common among the plankton, and are usually the most important plankton carnivore.

Four manuscripts of Achilles Tatius break off at i 10.3 t??? ??µas??, and a fifth originally did so. All five begin with Longus, where they descend from Vat. gr. 1348. This manuscript too has Achilles Tatius after Longus, but complete, so that they may seem unlikely to descend from it in Achilles Tatius. According to the latest editor, E. Vilborg (Gothenburg 1955), they do not descend from it: they avoid three of its errors (p. lxii, lxvi). Expectation and evidence coincide, then, and that should be that.Every fragmentary manuscript, however, goes back eventually to a complete one, and whether three errors will bear the weight of a stemma depends on others.

THE complete cycle of development from egg to egg for a pelagic species of chaetognath has not been achieved previously1in the laboratory. Dallot2maintained Sagitta setosa and Murikami3,4S. crassa long enough to observe cyclic reproductive changes. Murikami reared larvae for several days as I did5with S. hispida. Otherwise, the life span of most chaetognaths brought into the laboratory has been measured in hours6. Extended studies of live chaetognaths would be useful because these are among the commonest marine macroplanktonic carnivores and, as such, must play a major part in the trophodynamics of the sea. They have also frequently been used as “indicator species” for water masses7.

Type 2 diabetes mellitus is a heterogeneous inherited disorder characterized by chronic hyperglycemia resulting from pancreatic b -cell dysfunction and insulin resistance. Although the pathogenic mechanisms are not fully un-derstood, manifestation of the disease most likely requires interaction between both environmental and genetic factors. In the search for such susceptibility genes, we have performed a genomewide scan in 58 multiplex families (comprising 440 individuals, 229 of whom were affected) from the Botnia region in Finland. Initially, linkage between chromosome 12q24 and impaired insulin secretion had been reported, by Mahtani et al., in a subsample of 26 families. In the present study, we extend the initial genomewide scan to include 32 additional families, update the affectation status, and fine map regions of interest, and we try to replicate the initial stratification analysis. Gestational diabetes mellitus (GDM) affects more than 16 million pregnancies annually worldwide and is related to an increased lifetime risk of Type 2 diabetes (T2D). The diseases are hypothesized to share a genetic predisposition, but there are few GWAS studies of GDM and none of them is sufficiently powered to assess whether any variants or biological pathways are specific to GDM. We conducted the largest genome-wide association study of GDM to date in 12,332 cases and 131,109 parous female controls in the FinnGen Study and identified 13 GDM-associated loci including 8 novel loci. Genetic features distinct from T2D were identified both at the locus and genomic scale. Our results suggest that the genetics of GDM risk falls into two distinct categories - one part conventional T2D polygenic risk and one part predominantly influencing mechanisms disrupted in pregnancy. Loci with GDM-predominant effects map to genes related to islet cells, central glucose homeostasis, steroidogenesis, and placental expression. - Abstract
Tuberous sclerosis (TSC) is a relatively common hamartoma syndrome caused by mutations in either of two genes, TSC1 and TSC2. Here we report comprehensive mutation analysis in 224 index patients with TSC and correlate mutation findings with clinical features. Denaturing high-performance liquid chromatography, long-range polymerase chain reaction (PCR), and quantitative PCR were used for mutation detection. Mutations were identified in 186 (83%) of 224 of cases, comprising 138 small TSC2 mutations, 20 large TSC2 mutations, and 28 small TSC1 mutations. A standardized clinical assessment instrument covering 16 TSC manifestations was used. Sporadic patients with TSC1 mutations had, on average, milder disease in comparison with patients with TSC2 mutations, despite being of similar age. They had a lower frequency of seizures and moderate-to-severe mental retardation, fewer subependymal nodules and cortical tubers, less-severe kidney involvement, no retinal hamartomas, and less-severe facial angiofibroma. Patients in whom no mutation was found also had disease that was milder, on average, than that in patients with TSC2 mutations and was somewhat distinct from patients with TSC1 mutations. Although there was overlap in the spectrum of many clinical features of patients with TSC1 versus TSC2 mutations, some features (grade 2-4 kidney cysts or angiomyolipomas, forehead plaques, retinal hamartomas, and liver angiomyolipomas) were very rare or not seen at all in TSC1 patients. Thus both germline and somatic mutations appear to be less common in TSC1 than in TSC2. The reduced severity of disease in patients without defined mutations suggests that many of these patients are mosaic for a TSC2 mutation and/or have TSC because of mutations in an as-yet-unidentified locus with a relatively mild clinical phenotype.
