This study examined the inclusion teachers' attitudes towards inclusive education in the public schools of Liloan District, Cebu Province Division. A descriptive-correlational design was utilized to collect data from purposively sampled 30 elementary teacher respondents through the M STATIC structured questionnaire. Most teachers were experienced females aged 34-43 years, married with some graduate studies. They had 1-5 years of teaching experience in inclusion yet only 1-2 inclusive education training sessions. Results found teachers generally supported inclusive philosophies and recognized social benefits but had concerns regarding training, resources, and support. While philosophically positive, worries existed about the support and resources needed in the classrooms. Pearson's r correlations and one-way ANOVA found no significant relationships between demographic profiles and attitudes. Based on these findings, a Teacher Inclusion Support Plan was recommended and customized for each school to enhance the long-term implementation of high-quality inclusion practices through ongoing, evidence-driven capacity building and professional development.

The relevance of the study lies in the authors consideration of the problem of hidden double taxation in the current tax legislation of the Russian Federation. The authors have identified the problem that is observed in practice, namely its latent manifestation in various forms, leading to an increase in the tax burden on business entities and citizens. The main method of the study is an analytical review of the problem of latent double taxation within the framework of the current Tax Code of the Russian Federation. The purpose of the study is to substantiate specific examples demonstrating the presence of binary taxation, which, although not an obvious violation of the legislation, but creates significant economic costs and injustice for taxpayers, including the taxation of wages, VAT, excise duties and patent system of taxation, revealing the mechanisms through which there is a hidden double taxation of the same income. The result of the study is a proposal to solve the problem of double taxation within a comprehensive approach, including the possibility of simplifying tax rules, developing a more transparent taxation system, reducing the tax burden of taxpayers, as well as revising the structure of taxation and a clearer definition of taxation principles to prevent hidden double taxation. The key conclusion of the study is the thesis that the introduction of open and clear taxation mechanisms will improve the position of taxpayers and create a more sustainable tax environment in Russia. Further research should focus on combating double taxation, which requires careful analysis and reform of the tax system to ensure its transparency and fairness.

The Vermont STEM Leadership Institute (VSTEM) was designed to provide professional learning and leadership opportunities for K-12 educators teaching primarily in high-need schools. The fundamental premise of the program was to actively engage teachers in constructivist curriculum and pedagogy coupled with authentic scientific research experiences within the context of local environments or "places." This study investigated changes in content and pedagogical content knowledge that teachers exhibited in their science teaching practice over the course of their program participation. Data analysis revealed that teachers' science content knowledge and pedagogical content knowledge were enhanced by VSTEM program participation with moderate to strong indications about place-based education, project-based learning, and the importance of engaging students in authentic scientific research. The study found that participants learned new content-specific teaching strategies and implemented standards-based units and lessons that aligned with constructivist theories of teaching and learning.

This article presents approaches for enhancing engineering-geological and geotechnical research for the restoration and reconstruction projects of cultural heritage objects in St. Petersburg’s historical center. According to the Venice Charter, an interdisciplinary approach is recommended for developing a preservation program for cultural heritage objects. The authors consider the underground space as a multi-component system, where soils are a host medium for underground water with specific composition, complex physicochemical and biochemical conditions, the presence and vigorous activity of microorganisms, gases of various origins, and underground bearing and enclosing structures. The last of these components is crucial for the long-term stability of historical and cultural objects, as they interact in complex with water-saturated soils and microorganisms. This approach is not to be found in Russian cultural heritage preservation regulations. In engineering-geological and geotechnical research, multi-component underground space should be studied at the regional, local, and object levels. Assessing redox conditions in the underground environment is crucial for understanding the state of its components. Moreover, this can trigger hazardous processes such as changes in the stress–strain state and unconsolidation of the soil layer due to the generation of low-solubility gases and biocorrosion of ancient and modern building materials, causing premature destruction. Recommendations are provided for carrying out engineering-geological and geotechnical research based on cultural heritage objects, considering the multi-component underground space and features of its geoenvironmental state due to the long-term contamination and transformation of its components.

Background: The global impact of the coronavirus disease 2019 (COVID-19) pandemic has strained healthcare workers (HCWs) and services worldwide. As the pandemic evolves, there is a need to assess the patterns, clinical manifestations, and outcomes among HCWs, especially in low-middle income countries where challenges such as inadequate infection prevention and control practices, personal protective equipment, and low diagnostic capacity persist. Objective: To determine the pattern, clinical manifestations, laboratory characteristics, and outcomes of COVID-19 infection among HCWs in a specialty unit in Ibadan, Nigeria. Materials and Methods: A retrospective cohort study was conducted on 107 HCWs over 8 weeks during a COVID-19 outbreak among HCWs in a specialty unit in Ibadan, Nigeria. Data on demographics, clinical characteristics, and laboratory findings were collected using a structured proforma. Severe-acute-respiratory-syndrome-related coronavirus laboratory tests followed the World Health Organization recommendations, with nasopharyngeal swabs obtained for real-time reverse transcription–polymerase chain reaction. Results: Among the HCWs, a 49% COVID-19 positivity rate was found, with cases peaking in the third week. Nurses demonstrated the highest infection rate (36.5%). Fatigue and myalgia were predominant symptoms with 51.9% (P < 0.001) of COVID-19 HCWs reporting both symptoms. Conclusion: The high prevalence of asymptomatic cases highlights the potential role of HCWs as sources of infection for COVID-19, emphasizing the need for stringent infection control measures and continuous support for the well-being of healthcare professionals.

In response to the SARS-CoV-2 pandemic, a cancer research education program at the University of Nebraska Medical Center designed for Native American middle school, high school and undergraduate students adapted activities to be delivered online. There are considerable challenges to adapting in-person science programming to online delivery that can impact overall effectiveness. These challenges are further exacerbated when the cognate student population also faces significant disparities in health, wealth, and educational outcomes. We encountered both disadvantages and advantages in transitioning programming to online virtual formats. Challenges faced in delivering our programming during the pandemic included varied online accessibility, peripheral stressors, and disconnection to places and people. Despite these challenges, we found several benefits in remote delivery, some of which have alleviated barriers to program participation for Native American students. Some successes achieved by transitioning to fully remote programming included increased program reach, sustainability, and cultural relevancy. In this overview of the implementation of four online programs at the middle school, high school, and undergraduate levels, we highlight the challenges and successes experienced. Through this program description, we aim to provide insight into potential strategies to improve program delivery designed for Native American students during the SARS-CoV-2 pandemic and beyond.

The article presents the results of an analysis of perceptions of migrants in press releases and regulatory documents of law enforcement and civil government agencies. We considered these texts within the framework of a "soft" constructionist approach, as a tool for problematizing the social process and one of the key ways of producing the discourse of power. The purpose of the study was to use a combination of quantitative content analysis and discourse analysis to identify the "equivalence chains" that give meanings to the nodal sign "migration" in the texts of power. The objectives of the study were to collect a text array relevant to the topic and determine the social context in which the migrant is placed: the spaces in which they are present, the actions of which they are the subject and object. In most of the 548 texts reviewed, the migrant is presented as a suspicious, impersonal object of state paternalism. The most extensive category of social actions in which a migrant acts as a subject are crimes, mainly related to violation of the regime of stay in the Russian Federation. The most common actions committed against a migrant are inspections and raids, as well as various kinds of punishment. The subject of action against a migrant in regional government texts is not the Main Directorate for Migration Affairs of the Ministry of Internal Affairs of the Russian Federation, but the Main Directorate for Public Order Maintenance, the Prosecutor's Office of the Russian Federation, and the courts. The article proposes a hypothesis according to which law enforcement agencies use press releases simultaneously as a way to problematize migration and maintain public attention to it. Having monopolized the problem, the Ministry of Internal Affairs, the Prosecutor's Office, and the courts use it to repeatedly "sell" this issue to the public through the only tool available to them - symbolic and physical violence

BackgroundPosttraumatic stress disorder (PTSD) and major depressive disorder (MDD) are commonly reported co-occurring mental health consequences of psychological trauma exposure. The disorders have high genetic overlap. Trauma is a complex phenotype but research suggests that trauma sensitivity has a heritable basis. We investigated whether sensitivity to trauma in those with MDD reflects a similar genetic component in those with PTSD.MethodsGenetic correlations between PTSD and MDD in individuals reporting trauma and MDD in individuals not reporting trauma were estimated, as well as with recurrent MDD and single-episode MDD, using genome-wide association study (GWAS) summary statistics. Genetic correlations were replicated using PTSD data from the Psychiatric Genomics Consortium and the Million Veteran Program. Polygenic risk scores were generated in UK Biobank participants who met the criteria for lifetime MDD (N = 29 471). We investigated whether genetic loading for PTSD was associated with reporting trauma in these individuals.ResultsGenetic loading for PTSD was significantly associated with reporting trauma in individuals with MDD [OR 1.04 (95% CI 1.01-1.07), Empirical-p = 0.02]. PTSD was significantly more genetically correlated with recurrent MDD than with MDD in individuals not reporting trauma (rg differences = ~0.2, p < 0.008). Participants who had experienced recurrent MDD reported significantly higher rates of trauma than participants who had experienced single-episode MDD (χ2 > 166, p < 0.001).ConclusionsOur findings point towards the existence of genetic variants associated with trauma sensitivity that might be shared between PTSD and MDD, although replication with better powered GWAS is needed. Our findings corroborate previous research highlighting trauma exposure as a key risk factor for recurrent MDD.

Visuospatial processing speed underlies several cognitive functions critical for successful completion of everyday tasks, including driving and walking. While it is widely accepted that visuospatial processing speed peaks in early adulthood, performance across the lifespan remains incompletely characterized. Additionally, there remains a lack of paradigms available to assess visuospatial processing speed in unsupervised web-based testing environments. To address these gaps, we developed a novel visuospatial processing speed (VIPS) task adapted from two tests sensitive to visuospatial processing speed declines in older adults, the Useful Field of View paradigm and the PERformance CEntered Portable Test. The VIPS task requires participants to make a central orientation discrimination and complete a simultaneous peripheral visual search task. Data were collected from 86 in-lab volunteers (18-30 years) to compare performance to traditional neuropsychological measures. Consistent with previous literature, performance on the novel VIPS task significantly correlated with measures of selective attention, executive functioning, visual speed, and working memory. An additional 4395 volunteers (12-62 years) were recruited on TestMyBrain.org to establish lifespan trajectories of visuospatial processing speed and associations with functional disability. VIPS task performance peaked in the early 20's, and steadily decreased such that thresholds doubled in 60-year-olds relative to 20-year-olds (817 ms vs. 412 ms). VIPS task performance significantly correlated with self-reported cognitive functioning deficits broadly across the lifespan but was specifically related to mobility issues in middle-age. These findings have important implications for early detection of cognitive decline and provide insights into potential early intervention targets for younger and middle-aged adults.

Introduction Polymethyl methacrylate (PMMA) is a common depot system in the treatment of chronic osteomyelitis. However, a lot of its shortcomings do not allow us to consider it ideal. Purpose of the work was to study in vivo the effectiveness of a polymer hydrogel containing an antibiotic for chronic osteomyelitis of the tibia in a rabbit model and compare it with PMMA Materials and methods The study was performed on the lower leg of 25 mature Chinchilla rabbits. A model of chronic osteomyelitis of the tibia was created. A methicillin-sensitive strain of Staphylococcus aureus (MSSA), highly active against cefazolin, was chosen as an infectious agent. Surgical debridement started 21 days after the clinical, laboratory, radiological and microbiological confirmation of the diagnosis, the technique for all animals was the same. The experimental group (n = 11) was treated by implantation of a polymer hydrogel, the comparison group (n = 11) with PMMA, and the control group (n = 3) had no implanted substance. In the postoperative period, monitoring of the local status, weight and body temperature of the animals, microbiological and radiological studies were carried out. Animals were taken out of the experiment by stages. Biopsies were sent for bacteriological and histomorphometric studies. Statistical comparison of the groups was performed using the Mann ‑ Whitney, Kruskell ‑ Wallis and Tukey criteria, descriptive statistics were used for the control group. Results In the experimental group, in all cases, postoperative wounds healed in a timely manner, the levels of WBC and CRP significantly (p = 0.040) decreased from 14 and 21 days, respectively. Microbiologically, the growth of microflora from the wound discharge and biopsy specimens was not detected; radiographic progression of chronic osteomyelitis was not observed; histomorphometry revealed a significant (p = 0.002) effective relief of the inflammatory process. In the comparison group, systemic antibiotic therapy was required from postoperative day 7. Levels of inflammatory markers decreased less effectively than in the experimental group. MSSA was verified from wound discharge and biopsy specimens at almost every follow-up time-point. X-rays and histomorphometry (p = 0.001), on average, detected exacerbation of osteomyelitis. In the control group, systemic therapy did not give positive dynamics. Discussion A comparative analysis showed that the hydrogel impregnated with an antibacterial agent, unlike PMMA, reliably arrests chronic osteomyelitis without auxiliary systemic antibiotic therapy and does not cause material-associated bone resorption. The clinical and laboratory picture is fully consistent with the data of microbiology, radiology and histomorphometry. Conclusion Hydrogel impregnated with an antibiotic reliably and effectively stops chronic osteomyelitis compared to PMMA.

The Alexander Column as a compositional center of the architectural ensemble of Palace Square in Saint Petersburg, Russia, has always been a matter of concern for both the public and specialists due to progressive deterioration of its granite shaft caused by crack formation. The article examines previous studies related to the inspection and restoration of the column's shaft and other parts above ground level, as well as reasons for crack initiation and propagation in the column. An analysis was performed on the anomalies in the Fennoscandian Shield and the structural-tectonic conditions at the Montferrand quarry site, revealing the presence of faults and circular features within the studied area. The research considers N.Hast's measurements of excess tectonic stresses in anomaly zones (southeastern Finland), which acted horizontally and resulted in the development of tensile cracks within the granite massif and later in the column’s shaft after its installation. The most dangerous type of deformation for the Alexander Column is its tilt in the northeast direction, recorded in 1937 and 2000. The article analyzes the construction features of the column's foundations and additional underground elements, as well as soil and groundwater characteristics based on archival data. The contamination history of the underground space is taken into account, and an analogy-based method is used to assess the engineering-geological and hydrogeological conditions of the underground load-bearing structures within the placement zone of the Alexander Column and the New Hermitage buildings. The results of visual observations on the nature of deterioration and deformation of the pavement around the monument, as well as its pedestal, indicating the development of uneven settlement of the foundation, are presented. The article concludes with general recommendations for organizing and implementing comprehensive monitoring to forecast the deformation dynamics of the Alexander Column.

Abstract Background Uterine leiomyosarcomas (uLMS) are aggressive tumours with poor prognosis and limited treatment options. Although immune checkpoint blockade (ICB) has proven effective in some ‘challenging‐to‐treat’ cancers, clinical trials showed that uLMS do not respond to ICB. Emerging evidence suggests that aberrant PI3K/mTOR signalling can drive resistance to ICB. We therefore explored the relevance of the PI3K/mTOR pathway for ICB treatment in uLMS and explored pharmacological inhibition of this pathway to sensitise these tumours to ICB. Methods We performed an integrated multiomics analysis based on TCGA data to explore the correlation between PI3K/mTOR dysregulation and immune infiltration in 101 LMS. We assessed response to PI3K/mTOR inhibitors in immunodeficient and humanized uLMS patient‐derived xenografts (PDXs) by evaluating tumour microenvironment modulation using multiplex immunofluorescence. We explored response to single‐agent and a combination of PI3K/mTOR inhibitors with PD‐1 blockade in humanized uLMS PDXs. We mapped intratumoural dynamics using single‐cell RNA/TCR sequencing of serially collected biopsies. Results PI3K/mTOR over‐activation (pS6high) associated with lymphocyte depletion and wound healing immune landscapes in (u)LMS, suggesting it contributes to immune evasion. In contrast, PI3K/mTOR inhibition induced profound tumour microenvironment remodelling in an ICB‐resistant humanized uLMS PDX model, fostering adaptive anti‐tumour immune responses. Indeed, PI3K/mTOR inhibition induced macrophage repolarisation towards an anti‐tumourigenic phenotype and increased antigen presentation on dendritic and tumour cells, but also promoted infiltration of PD‐1+ T cells displaying an exhausted phenotype. When combined with anti‐PD‐1, PI3K/mTOR inhibition led to partial or complete tumour responses, whereas no response to single‐agent anti‐PD‐1 was observed. Combination therapy reinvigorated exhausted T cells and induced clonal hyper‐expansion of a cytotoxic CD8+ T‐cell population supported by a CD4+ Th1 niche. Conclusions Our findings indicate that aberrant PI3K/mTOR pathway activation contributes to immune escape in uLMS and provides a rationale for combining PI3K/mTOR inhibition with ICB for the treatment of this patient population.

The National Institutes of Health and the National Science Foundation have made a compelling call to action not only to strengthen the pipeline of available STEM-trained talent, but in addition to foster students who are members of populations currently under-represented in science. furthermore, the scientific community must not only increase the accessibility of STEM-related education, but also implement and test evidence-based practices. Presented here, we detail the proceedings of a hands-on, science-focused informal learning opportunity aimed at educating an underrepresented population in cancer biology. Fifteen undergraduate and graduate student volunteer instructors from the University of Nebraska at Omaha and the University of Nebraska Medical Center engaged with 89 high school students, mostly Native American, in an informal learning event called "Cancer Biology and You Day." Throughout the event, students completed two independent lessons focusing on breast cancer and skin cancer and demonstrated strong learning gains associated with the lessons as assessed by KWL charts. Exit surveys of the students indicated high levels of satisfaction with the event, and positive attitudes associated with considering a career in science/research were evident in survey responses. Overall, we report the event as a success and outline how similar experiences may be achieved.

Growing research suggests that posttraumatic stress disorder (PTSD) may be a risk factor for poor cardiovascular health, and yet our understanding of who might be at greatest risk of adverse cardiovascular outcomes after trauma is limited. In this study, we conducted the first examination of the individual and synergistic contributions of PTSD symptoms and blood pressure genetics to continuous blood pressure levels. We harnessed the power of the Psychiatric Genomics Consortium-PTSD Physical Health Working Group and investigated these associations across 11 studies of 72,224 trauma-exposed individuals of European (n = 70,870) and African (n = 1,354) ancestry. Genetic contributions to blood pressure were modeled via polygenic scores (PGS) for systolic blood pressure (SBP) and diastolic blood pressure (DBP) that were derived from a prior trans-ethnic blood pressure genome-wide association study (GWAS). Results of trans-ethnic meta-analyses revealed significant main effects of the PGS on blood pressure levels [SBP: β = 2.83, standard error (SE) = 0.06, p < 1E-20; DBP: β = 1.32, SE = 0.04, p < 1E-20]. Significant main effects of PTSD symptoms were also detected for SBP and DBP in trans-ethnic meta-analyses, though there was significant heterogeneity in these results. When including data from the largest contributing study - United Kingdom Biobank - PTSD symptoms were negatively associated with SBP levels (β = -1.46, SE = 0.44, p = 9.8E-4) and positively associated with DBP levels (β = 0.70, SE = 0.26, p = 8.1E-3). However, when excluding the United Kingdom Biobank cohort in trans-ethnic meta-analyses, there was a nominally significant positive association between PTSD symptoms and SBP levels (β = 2.81, SE = 1.13, p = 0.01); no significant association was observed for DBP (β = 0.43, SE = 0.78, p = 0.58). Blood pressure PGS did not significantly moderate the associations between PTSD symptoms and blood pressure levels in meta-analyses. Additional research is needed to better understand the extent to which PTSD is associated with high blood pressure and how genetic as well as contextual factors may play a role in influencing cardiovascular risk.

The objective was to explore the dynamics and duration of antibiotic elution in samples based on polymer hydrogel and PMMA. Material and methods The samples impregnated with vancomycin, rifampicin and cefazolin at various concentrations were placed in phosphate-buffered saline and incubated at 37 °C. The medium was completely replaced at 1, 3, 7, 14, 21 and 28 days. Spectrophotometry was used to measure concentration of drugs in solution and the release profiles. The median and 95 % CI were employed to statistically describe data obtained from 5 parallel studies. Results Concentrations of the antibiotics eluted from the polymer hydrogel were 7 times greater than those released from PMMA on day 1; 15 times greater on days 2 and 3; 6.6 times greater on day 7 and 3 times or more in the following days of observation. The rate of antibiotic release from hydrogel volumes also differed markedly. Discussion The drug release was more than 70% of the total amount for all hydrogel samples in contrast to PMMA with elution not exceeding 10 %. Despite the fact that a burst release was observed with 80 % of the antibiotic released in the first 5 days as seen in the case of bone cement, the concentration of the drug eluted from hydrogels was several times higher and exceeded the MIC throughout the observation period. The release of the antimicrobial agent from hydrogels was caused by diffusion of the particles from the entire volume of the matrix demonstrating an important advantage over PMMA with the potential being limited by surface depletion. Conclusion At this point, we have shown the possibility of creating potential depot systems based on unsaturated PVA derivatives with controlled release of antibiotics and characteristics being superior to PMMA.

Background: Antibiotic residue in food products and the resulting antibiotic-resistant bacteria represent a significant global public health threat. The misuse of antibiotics is a primary contributor to this issue. This study investigated the knowledge, attitudes, and practices (KAP) regarding antibiotic use among cage fish farmers on Ghana’s Volta Lake. Method: We conducted a cross-sectional survey with 91 cage fish farmers across three scales: small, medium, and large. A semi-structured questionnaire complemented by personal observations provided comprehensive data. We used several statistical methods for analysis: Pearson Chi-Square and Spearman correlation tests to examine relationships and trends among variables, logistic regression to analyze variable interactions, and Cronbach’s alpha to check internal consistency. Additionally, Kendall’s coefficient was used to rank challenges, utilizing STATA and SPSS for these calculations. Results: The survey revealed that 58.55% of cage fish farmers earn an average of 10,000 USD annually, with 35.16% having over 16 years of experience. From the survey, all sampled populations admitted to antibiotic applications in their farming operation. Knowledge of antibiotic types was mainly influenced by peers (46.15%), with tetracycline being the most recognized and used. There was a significant reliance on the empirical use of antibiotics, with 52.75% of farmers using them based on personal experience and 40.66% without a prescription. When initial treatments failed, 41.76% of the farmers would change or combine drugs. Older farmers (over 51 years) and those with tertiary education demonstrated significantly better KAP scores regarding antibiotic use. Strong correlations were also found among knowledge, attitudes, and practices in antibiotic usage. Conclusions: The findings indicate a need for improved education on antibiotic use among fish farmers to reduce misuse and enhance awareness of the potential consequences. This study provides foundational data for designing interventions to address these issues in the context of cage fish farming on Volta Lake.

Purpose To evaluate the results of a comparative analysis of the in vitro antimicrobial activity of polymer hydrogels and PMMA impregnated with antibiotics against test cultures of the leading pathogens of bone infection. Materials and methods A comparative analysis of the antimicrobial activity of the polymer hydrogel and PMMA impregnated with antibiotics was carried out. The bactericidal efficacy and prolongation of action of the following microbe-antibiotic pairs were assessed: MSSA – gentamicin, MSSE – cefazolin, MRSA and MRSE – vancomycin, A. baumannii – tobramycin. The duration of the study is 7 days. Statistical comparison of groups was carried out using the Mann-Whitney and Wilcoxon tests; the median and 95 % CI were used to describe the data. Results The obtained indicators of the zone of inhibition of the growth of microorganisms around the polymer hydrogel were several times higher than the results of PMMA (p < 0.05 for all periods). The antimicrobial activity of hydrogels in all cases lasted more than 7 days. Hydrogels impregnated with vancomycin showed the lowest rate of contraction of the inhibition zone. The best antimicrobial effect was demonstrated by the compared samples with cefazolin in relation to MSSE, the maximum index of the zone of suppression of which, on average, for the hydrogel was 29.3 mm, PMMA – 22.3 mm. Discussion Samples from polymer hydrogel impregnated with antibiotics demonstrated prolonged pronounced effective antimicrobial activity against the leading pathogens of orthopedic infections in comparison with PMMA. The bactericidal efficacy of the polymer hydrogel significantly exceeded the activity of PMMA already from the first day of the study (p = 0.002). The action of the hydrogel throughout the study was uniform without abrupt changes or dips (p > 0.05), in contrast to bone cement, whose activity against all test cultures significantly decreased (p = 0.042) on the 2nd day of the study. Conclusion Polymeric biodegradable hydrogels impregnated with antibiotics have a greater antimicrobial potential compared to PMMA. Uniform, slow and controlled release of the drug and effective inhibition of the growth of microorganisms, as well as prolonged action show that these properties may turn hydrogels into an effective depot system.

BackgroundEducation years, as a measure of cognitive reserve, have been shown to affect the progression of Alzheimer’s disease (AD), both pathologically and clinically. However, inconsistent results have been reported regarding the association between years of education and intermediate structural changes in AD-vulnerable brain regions, particularly when AD risk factors were not considered during the preclinical phase.ObjectiveThis study aimed to examine how Aβ deposition and APOE ε4 carrier status moderate the relationship between years of education and cortical volume in AD-vulnerable regions among cognitively normal older adults.MethodsA total of 121 participants underwent structural MRI, [18F] flutemetamol PET-CT imaging, and neuropsychological battery assessment. Multiple regression analysis was conducted to examine the interaction between years of education and the effects of potential modifiers on cortical volume. The associations between cortical volume and neuropsychological performance were further explored in subgroups categorized based on AD risk factors.ResultsThe cortical volume of the left lateral occipital cortex and bilateral fusiform gyrus demonstrated a significant differential association with years of education, depending on the presence of Aβ deposition and APOE ε4 carrier status. Furthermore, a significant relationship between the cortical volume of the bilateral fusiform gyrus and AD-nonspecific cognitive function was predominantly observed in individuals without AD risk factors.ConclusionAD risk factors exerted varying influences on the association between years of education and cortical volume during the preclinical phase. Further investigations into the long-term implications of these findings would enhance our understanding of cognitive reserves in the preclinical stages of AD.
