Abstract

Cycles of Cdc53/Cullin1 rubylation (a.k.a NEDDylation) protect ubiquitin-E3 SCF (Skp1-Cullin1-F-box protein) complexes from self-destruction and play an important role in mediating the ubiquitination of key protein substrates involved in cell cycle progression, development, and survival. Cul1 rubylation is balanced by the COP9 signalosome (CSN), a multi-subunit derubylase that shows 1:1 paralogy to the 26S proteasome lid. The turnover of SCF substrates and their relevance to various diseases is well studied, yet, the extent by which environmental perturbations influence Cul1 rubylation/derubylation cycles per se is still unclear. In this study, we show that the level of cellular oxidation serves as a molecular switch, determining Cullin1 rubylation/derubylation ratio. We describe a mutant of the proteasome lid subunit, Rpn11 that exhibits accumulated levels of Cullin1-Rub1 conjugates, a characteristic phenotype of csn mutants. By dissecting between distinct phenotypes of rpn11 mutants, proteasome and mitochondria dysfunction, we were able to recognize the high reactive oxygen species (ROS) production during the transition of cells into mitochondrial respiration, as a checkpoint of Cullin1 rubylation in a reversible manner. Thus, the study adds the rubylation cascade to the list of cellular pathways regulated by redox homeostasis.

8. Will CARICOM countries be vulnerable to financial crises as a result of the Economic Partnership Agreement (EPA) : the Cotonou Agreement?

Abstract

Whether the EPA contains the seeds of potential financial crises in relation to the CARICOM countries, using the concepts of globalisation, volatility risk and hubris (of leadership, economic thought and a weak regulatory environment). This study seeks to establish whether financial crises may occur when the variables of globalisation, volatility risk and hubris within a weak regulatory environment coincide within the framework of the Economic Partnership Agreement between the ACP countries, (in particular the CARICOM countries) and the EU (the Cotonou Agreement). Ultimately it is the intention to assess whether there is any association between these factors to determine if when combined we can anticipate the likely occurrence of financial crises. A number of research hypotheses were derived from the review of the literature in Documents 2 and 3. The evidence presented in Document 4 did not support the null hypotheses and allowed us to conclude that there is a statistical association among globalisation, volatility risk and hubris. In conclusion, we explore the attitudes of key sectors with respect to the EPA economic framework and proffer that a likely combination of these three variables may contribute to the occurrence of financial crises. Furthermore it is the author’s contention that the Cotonou Agreement may contain the ingredients to usher in another financial crisis.

9. Natural units, numbers and numerical clusters

Abstract

Defined by Lord Kelvin as the science of measurement it is described a fundamental fact of physics. The so called `natural' units represent the unique system of units conveniently used in the realm of High Energy Physics. The system of natural units is defined by the consistent providing of the adimensional unit value to the velocity c of the eletromagnetic radiation in the vacuum, related to the Maxwell classical electromagnetic theory and to the special relativity theory, to the reduced Planck constant $\hbar\equiv\frac{h}{2\pi}$ having the dimension of action, which is the fundamental constant of the quantum mechanics as well as to the Boltzmann constant $k_B$ which has the dimension of heat capacity, being the fundamental constant associated to the statistical mechanics, $c=\hbar = k_B = 1$. Such prescription is mandatory as we investigate the ultimate constituents of matter and their dynamics, since the atoms, leptons and quarks up to the grand unification, strings, superstrings, and the supergravity theories, as well as the total unification of the interactions in the M-theory. The one-dimensional character of this phenomenology is totally featured by the use of the natural units, reflecting the fact that in order to be possible the investigation of the fundamental constituents of matter it is being necessary the resolution of lengths of the order of $10^{-16} cm$ or to reach energies of the order of at least 1 TeV. The application of this system of natural units results exceptionally adequate in the description of the Universe since the Planck scale, up to the Hubble large scale expanding Universe, resulting in the curious formation of the regular numerical clusters., Comment: 7 pages, revtex, no figures

10. Cosmological constant and spontaneous gauge symmetry breaking: the particle physics and cosmology interface charade

Abstract

We describe one of the remarkable problems of theoretical physics persevering up to the beginning of the millennium. All gaugetheories with spontaneous gauge symmetry breaking from the standard model of particle physics with the electroweak symmetry breaking at the Fermi scale, 246 GeV, up to strings, supergravity, and the M(embrane)-theory superunification with symmetry breaking starting near the Planck scale, $10^{19}$ GeV, foresee that the spontaneous symmetry breakings induce a vacuum energy at least 50 orders of magnitude larger than the stringent experimental bound $G\Lambda\lesssim 10^{-122}$ on the value of the cosmological constant $\Lambda$. This fact seems to have a universal character since it occurs from the Fermi scale up to the Planck one. It is the vacuum catastrophe., Comment: 22 pages

Abstract

Until now studies in Brazil of the pathological action of Contracaecum sp. larvae in Hoplias malabaricus have been rare. The aim of the present study was to analyze and describe histopathological changes in the stomach of H. malabaricus caused by different intensities of infection by larvae of Contracaecum sp., and the hematological effects of such infection. The mean intensity of infection by L3 larvae of Contracaecum sp. correlated positively with Hematocrit percentage (Hct%) and correlated negatively with mean corpuscular volume (MCV). These findings are possibly related to the passage of larvae from the stomach to the mesentery, potentially due to hemorrhaging t. Fish with high infection intensity had larvae of Contracaecum sp in the submucosal of the stomach, which caused destruction of Type I and Type III collagen (finer and more delicate collagen fibers which do not form beams and are permeated by fundamental substances) around the parasite. Microscopic analysis showed that crypts were arranged regularly throughout the stomach mucosal, varying in number, size and depth, depending on the region of the stomach.

Abstract

The objectives were to determine the physical and chemical characteristics of different substrates, and assess the influence of different substrates, with and without the addition of controlled-release fertilizer, on the growth and morphological quality of taperebá (Spondias macrocarpa) seedlings. The experimental design adopted was a 2 x 6 factorial scheme, with the presence and absence of 8.0 g L-1 of encapsulated controlled-release fertilizer and six substrates [Sub1= soil; Sub2= soil + carbonized rice husk (2:1 v/v); Sub3= soil + organic compos (2:1 v/v); Sub4= soil + carbonized rice husk + organic compost (1:1:1); Sub5= soil + carbonized rice husk + organic compost (2:1:1); Sub 6= soil + carbonized rice husk + organic compost (2:2:1)], with five replicates, composed of 10 seedlings (one in each container). Formulations of substrates containing soil + carbonized rice husk + organic compost (1:1:1) and soil + organic compost (2:1 v/v) had total porosity ranging from 0.88 to 0.91 m3 m-3, aeration space from 0.33 to 0.35 m3 m-3 and available water from 0.27 to 0.29 m3 m-3, hence being suitable to obtain taperebá seedlings with superior quality. The substrate Cerrado soil (Sub1) is not viable for the production of taperebá seedlings, since it has acidic pH, an undesirable chemical characteristic for the cultivation of this species. Incorporation of controlled-release fertilizer into the substrates used in the present study led to better morphological characteristics, so it is indicated as an input for fertilization in the initial stage of growth of taperebá seedlings. Abstract

EEM data recorded at different pH values was exploited by MCR-ALS in order to determine qualitative information about Brazilian red wines. In addition, the geographical traceability of wines produced in the Serra Gaúcha (Rio Grande do Sul) was carried out by DD-SIMCA considering 53 samples from the target class and 20 from other producing regions. The fluorescence signal corresponds to 9 EEMs recorded at different pH (3?11), generating four-way data. By MCR-ALS decomposition, eight factors were retrieved and related to typical chemical compounds found in red wine. In addition, the EEM pH data was used to build a one-class classification model, considering that MCR scores and all samples of the target class were properly recognised as belonging to the target class, with maximal sensitivity equal to 1. Abstract

This paper presents a methodological proposal for the teaching of exponential function, resulting from the application of a didactic sequence involving exponential function, where evidence of learning and the consolidation and application of mathematical concepts in problem solving were identified and analyzed. The Didactic Engineering of Michèle Artigue (1988) was used as a research methodology. As theoretical contributions that guided and enabled the development of the research, we chose the use of Mathematical Investigation in the classroom; Didactic Sequence in the conception of Zabala (1999); the Articulated Units of Conceptual Reconstruction proposed by Cabral (2017) and assumptions of Vygotsky's theory. A didactic sequence composed of five UARC's was elaborated to work the exponential function, with a view to minimizing the difficulties naturally imposed by the content to be explained. Microgenetic analysis of verbal interactions between teacher and students was used to analyze the results of the application. The results show that the students participating in the experiment showed evidence of learning, recorded during the process, and began to have a good understanding of the concepts and properties related to the topic, in addition to a good performance in carrying out the activities, facts that corroborate the potential of the didactic sequence proposed herein.

Abstract

Skatole and androstenone are the two main compounds responsible for the foul odour in entire male pigs' meat, known as boar taint. This study evaluated the effect of feed supplementation with inulin on the boar taint levels of 30 entire male pigs. Two months before slaughter, the animals were allocated into three groups (n = 10). The control group received a standard commercial diet. The other groups were fed the same commercial diet with 3% and 6% added inulin, respectively. Results showed that inulin addition to the feed significantly reduced skatole levels in the pigs' adipose tissue compared with the control group. The levels of androstenone were not affected by the dietary approach. Although there were differences in some parameters, the supplementation with inulin did not promote extensive changes in the meat quality parameters between the tested groups. When raising entire males, supplementation with inulin in finishing diets could be considered to reduce the boar taint perception by the consumer. [ABSTRACT FROM AUTHOR]
