Understanding the impact of wood vinegar on the growth of cherry radish is indispensable for use in crop production and environmental safety. Our study explored the regulation of rhizosphere microbial abundance and activity by wood vinegar, as well as the relationship between microbial community and growth factors in-depth and systematically. Bacterial communities at the phylum and genus levels were significantly changed after wood vinegar treatment. Application of 200-fold diluted wood vinegar significantly boosted Actinobacteriota and Firmicutes abundances by 40.88% and 126.67%, respectively, while Proteobacteria was promoted in carbon-rich soil. Fungi positively responded to cherry radish root traits and were correlated with aboveground biomass and fruit production. The fungi that correlated with photosynthesis included Albifimbria, Allomyces, Calcarisporiella, Clonostachys, Fusarium, Fusicolla, Knufia, Nigrospora, Paraconiothyrium, Preussia, Talaromyces, and Mortierellomycota. Wood vinegar treatment significantly affected the composition and abundance of soil bacterial and fungal communities in cherry radish rhizosphere, while simultaneously enhancing photosynthetic efficiency (e.g., Pn: 80.45% and Tr: 56.75%) and resulting in a 44.91% increase in crop yield. The promotion of cherry radish growth by wood vinegar may be attributed to the stimulation of soil microorganisms that degraded aromatic compounds and drove nitrogen cycling. This study provided novel insights into the significant promotion of cherry radish growth using wood vinegar diluted 200 times and identified potential microbial targets for agricultural applications. Generation of crops with low phytic acid (myo‐inositol‐1,2,3,4,5,6‐hexakisphosphate (InsP6)) is an important breeding direction, but such plants often display less desirable agronomic traits. In this study, through ethyl methanesulfonate‐mediated mutagenesis, we found that inositol 1,3,4‐trisphosphate 5/6‐kinase 4 (ITPK4), which is essential for producing InsP6, is a critical regulator of salt tolerance in Arabidopsis. Loss of function of ITPK4 gene leads to reduced root elongation under salt stress, which is primarily because of decreased root meristem length and reduced meristematic cell number. The itpk4 mutation also results in increased root hair density and increased accumulation of reactive oxygen species during salt exposure. RNA sequencing assay reveals that several auxin‐responsive genes are down‐regulated in the itpk4‐1 mutant compared to the wild‐type. Consistently, the itpk4‐1 mutant exhibits a reduced auxin level in the root tip and displays compromised gravity response, indicating that ITPK4 is involved in the regulation of the auxin signaling pathway. Through suppressor screening, it was found that mutation of Multidrug Resistance Protein 5 (MRP5)5 gene, which encodes an ATP‐binding cassette (ABC) transporter required for transporting InsP6 from the cytoplasm into the vacuole, fully rescues the salt hypersensitivity of the itpk4‐1 mutant, but in the itpk4‐1 mrp5 double mutant, InsP6 remains at a very low level. These results imply that InsP6 homeostasis rather than its overall amount is beneficial for stress tolerance in plants. Collectively, this study uncovers a pair of gene mutations that confer low InsP6 content without impacting stress tolerance, which offers a new strategy for creating "low‐phytate" crops. The water yield of aquifers increases the risk of water inflow, threatens the safe production of coal mines, and even causes geological disasters and construction hazards. To predict water yield quickly and accurately, multiple composite factors are used to invert unit water inflow rates to judge water yield grade. Taking the typical representative of north China-type coal fields as an example, six factors are selected: aquifer thickness, the radius of influence, normalized drawdown, permeability coefficient, the core rate of drilling holes, and the proportion of clay thickness to the thickness of the lower group. The whale optimization algorithm (WOA)–convolutional neural network (CNN)–support vector machine (SVM) model is established with the unit water inflow rate as the forecast target, and different models are selected for comparison. The water yield zoning map is obtained by bringing the borehole data into the model for prediction. The findings indicate that the root mean square error and average absolute error of the composite predictive model models are 0.0318 and 0.0268, respectively, and the model outperforms alternative models. Photorespiration is an energetically costly metabolic pathway in plants that responds to environmental stresses. The molecular basis of the regulation of the photorespiratory cycle under stress conditions remains unclear. Here, we discovered that FERONIA (FER) regulates photorespiratory flow under salt stress in Arabidopsis (Arabidopsis thaliana). FER mutation results in hypersensitivity to salt stress, but disruption of ferredoxin-dependent glutamate synthase 1 (GLU1), an enzyme that participates in the photorespiratory pathway by producing glutamate, greatly suppresses fer-4 hypersensitivity to salt stress primarily due to reduced glycine yield. In contrast, disrupting mitochondrial serine hydroxymethyltransferase1 (SHM1), which is supposed to increase glycine levels by hampering the conversion of glycine to serine in the photorespiratory cycle, aggravates fer-4 hypersensitivity to salt stress. Biochemical data show that FER interacts with and phosphorylates SHM1, and this phosphorylation modulates SHM1 stability. Additionally, the production of proline and its intermediate △1-pyrroline-5-carboxylate (P5C), which are both synthesized from glutamate, also contributes to fer-4 hypersensitivity to salt stress. In conclusion, this study elucidates the functional mechanism of FER in regulating salt tolerance by modulating photorespiratory flux, which greatly broadens our understanding of how plants adapt to high salinity., Competing Interests: Conflict of interest statement. To prepare recombinant human retinol binding protein 4 (RBP4) by using the baculovirus expression system and to detect its immunogenicity, the fusion DNA fragment of secretory signal peptide SS64 and human RBP4 gene was subcloned into a baculovirus transfer vector pFastBac-dual(pFBd), and the corresponding recombinant transfer plasmid was transformed into E. coli strain DH10bac, after transposition recombinant shuttle bacmid was screened out. The logarithmic phase Sf9 cells were transfected with the recombinant bacmid and then the recombinant baculovirus containing hRBP4 expression box were generated. After amplification of recombinant baculovirus, the recombinant baculovirus seeds were obtained. To express human RBP4, logarithmic phase Sf9 cells were infected with the virus seeds and SDS-PAGE and Western blotting were used to detect and identify the expression. Finally, to prepare a batch of RBP4 protein, logarithmic phase Sf9 cells in suspension culture were infected with recombinant baculovirus seeds and the supernatant was harvested after 120 hours post-infection for purification. Finally for preparation of polyclonal antibody and evaluation of immunogenicity, the recombinant hRBP4 from insect cells and from E. coli were immunized rabbits. Restriction enzyme digestion and sequencing confirmed that the recombinant baculovirus transfer plasmid was constructed correctly, and subsequently recombinant RBP4-bacmid was generated successfully. SDS-PAGE and Western blotting analysis suggested that human RBP4 protein was highly expressed in Sf9 cells with the molecular weight of approximately 23 kDa. The recombinant RBP4 protein could be secreted into the medium efficiently, and the expression level was calculated amount of 100 mg/L. Finally the rabbit antiserum was harvested after recombinant RBP4 immunization, therein the titer of antiserum against baculovirus recombinant RBP4 is 1:100 000 whereas the titer of antiserum against E. coli recombinant RBP4 is only 1:10 000. Overall, human RBP4 was high efficiently expressed successfully with good antigenicity in baculovirus system, and high affinity antiserum was obtained. A solid foundation was laid for the next step of the preparation of human serum RBP4 detection kit.
