Mosquitoes belonging to the genus Anopheles are the only vectors of human malaria. Anopheles gibbinsi has been linked to malaria transmission in Kenya, with recent collections in Zambia reporting the mosquito species exhibiting zoophilic and exophilic behavioral patterns with occasional contact with humans. Given the paucity of genetic data, and challenges to identification and molecular taxonomy of the mosquitoes belonging to the Anopheles genus; we report the first complete mitochondrial genome of An. gibbinsi using a genome skimming approach. An Illumina Novaseq 6000 platform was used for sequencing, the length of the mitochondrial genome was 15401 bp, with 78.5% AT content comprised of 37 genes. Phylogenetic analysis by maximum likelihood using concatenation of the 13 protein coding genes demonstrated that An. marshallii was the closest relative based on existing sequence data. This study demonstrates that the skimming approach is an inexpensive and efficient approach for mosquito species identification and concurrent taxonomic rectification, which may be a useful alternative for generating reference sequence data for evolutionary studies among the Culicidae.

Abstract Currently, there are increasing concerns about the possibility of a new epidemic due to emerging reports of Mayaro virus (MAYV) fever outbreaks in areas of South and Central America. Haemagogus mosquitoes, the primary sylvan vectors of MAYV are poorly characterized and a better understanding of the mosquito’s viral transmission dynamics and interactions with MAYV and other microorganisms would be important in devising effective control strategies. In this study, a metatranscriptomic based approach was utilized to determine the prevalence of RNA viruses in field-caught mosquitoes morphologically identified as Haemagogus janthinomys from twelve (12) forest locations in Trinidad, West Indies. Known insect specific viruses including the Phasi Charoen-like and Humaiata-Tubiacanga virus dominated the virome of the mosquitoes throughout sampling locations while other viruses such as the avian leukosis virus, MAYV and several unclassified viruses had a narrower distribution. Additionally, assembled contigs from the Ecclesville location suggests the presence of a unique uncharacterized picorna-like virus. Mapping of RNA sequencing reads to reference mitochondrial sequences of potential feeding host animals showed hits against avian and rodent sequences, which putatively adds to the growing body of evidence of a potentially wide feeding host-range for the Haemagogus mosquito vector.

In the Americas, the expansion in incidence of arboviral infections including Mayaro virus (MAYV) has drawn attention to the resurgence of viruses associated with understudied arthropods. Mosquitoes belonging to the genus Haemagogus are generally geographically restricted to the forests of Central and South America and the Caribbean and are the known sylvan vectors for yellow fever virus and emerging MAYV. With an established population in Jamaica, Haemagogus equinus has been reported to be well-adapted to oviposition in artificial containers close to human populations. Its role in arboviral transmission however is not fully understood. Given the dearth of genetic information and the difficulty in morphologically identifying cryptic features in species belonging to this genus, we report the first mitochondrial genome of Hg. equinus. Using a genome skimming approach, two Hg. equinus mosquito specimens were sequenced using the Illumina Novaseq 6000 platform. A representative mitogenome of 16,471 bp, 80.7% AT and 37 genes was assembled using NOVOplasty. Phylogenetic analysis placed Hg. equinus in the Albomaculatus section of the Haemagogus subgenus supporting previously described taxonomic studies.
