Abstract

Animal fibres from South American camelids and other fibre or wool bearing species provide important products for use by the human population. The contemporary context includes the competition with petrocarbon-based artificial fibres and concern about excessive persistence of these in the natural environment. Animal fibres present highly valuable characteristics for sustainable production and processing as they are both natural and renewable. On the other hand, their use is recognised to depend on availability of appropriate quality and quantity, the production of which is underpinned by a range of sciences and processes which support development to meet market requirements. This collection of papers combines international experience from South and North America, China and Europe. The focus lies on domestic South American camelids (alpacas, llamas) and also includes research on sheep and goats. It considers latest advances in sustainable development under climate change, breeding and genetics, reproduction and pathology, nutrition, meat and fibre production and fibre metrology. Publication of this book is supported by the Animal Fibre Working Group of the European Federation of Animal Science (EAAP). ‘Advances in Fibre Production Science in South American Camelids and other Fibre Animals’ addresses issues of importance to scientists and animal breeders, textile processors and manufacturers, specialised governmental policy makers and students studying veterinary, animal and applied biological sciences.

20. Fleece variation in alpaca (Vicugna pacos): a two-locus model for the Suri/Huacaya phenotype

Abstract

Abstract Background Genetic improvement of fibre-producing animal species has often induced transition from double coated to single coated fleece, accompanied by dramatic changes in skin follicles and hair composition, likely implying variation at multiple loci. Huacaya, the more common fleece phenotype in alpaca (Vicugna pacos), is characterized by a thick dense coat growing perpendicularly from the body, whereas the alternative rare and more prized single-coated Suri phenotype is distinguished by long silky fibre that grows parallel to the body and hangs in separate, distinctive pencil locks. A single-locus genetic model has been proposed for the Suri-Huacaya phenotype, where Huacaya is recessive. Results Two reciprocal experimental test-crosses (Suri × Huacaya) were carried out, involving a total of 17 unrelated males and 149 unrelated females. An additional dataset of 587 offspring of Suri × Suri crosses was analyzed. Segregation ratios, population genotype frequencies, and/or recombination fraction under different genetic models were estimated by maximum likelihood. The single locus model for the Suri/Huacaya phenotype was rejected. In addition, we present two unexpected observations: 1) a large proportion (about 3/4) of the Suri animals are segregating (with at least one Huacaya offspring), even in breeding conditions where the Huacaya trait would have been almost eliminated; 2) a model with two different values of the segregation ratio fit the data significantly better than a model with a single parameter. Conclusions The data support a genetic model in which two linked loci must simultaneously be homozygous for recessive alleles in order to produce the Huacaya phenotype. The estimated recombination rate between these loci was 0.099 (95% C.L. = 0.029-0.204). Our genetic analysis may be useful for other species whose breeding system produces mainly half-sib families.

Abstract

AbstractThe aim of this work was to assess the interaction between MC1Rand ASIPgenes in determining coat colour variation in black and brown Peruvian alpacas. Skin biopsies were collected from 25 black alpacas and 15 brown alpacas. Genomic DNA was isolated, amplified and sequenced. Black alpacas were dominant homozygous (E/E)or heterozygous (E/e) at MC1Rlocus while brown alpacas were heterozygous at the locus. All the black animals were heterozygous for the non-functional alleles at ASIP locus (aH– aΔ57– aht). On the contrary, brown alpacas were dominant homozygous (A/A) or heterozygous (A/aht) at this locus. When the combined genotypes at MC1Rand ASIPwere considered together, alpacas with black coats had four possible genotype combinations namely aH/aΔ57– E/E, aH/aht– E/E, aH/aΔ57– E/eand aH/aht– E/e. The brown alpacas were found to have the A/aht– E/eand the A/A – E/egenotype. Our results show that the two genes interact to sinergically modulate the amount of melanin synthetised in alpaca fibre. In black coat alpacas in fact, the expression of non-functional ASIP results in the failure of pheomelanin production while the functional MC1R allows the synthesis almost exclusively of eumelanin. On the other hand, the brown coat phenotype is produced when the animal possesses at least one functional copy of the gene at both ASIPand MC1Rloci. In fact, MC1R binds both ASIP and α-MSH allowing a mixed melanogenesis characterised by the synthesis of eumelanin and pheomelanin granules in a similar ratio.HIGHLIGHTSThe income of thousands of Peruvian alpaca farmers depends mainly on the sale of fibre in high demand on the global textile market.A wide variety of colours exists, therefore this fibre is preferred by the ecologically sustainable industry for organic textile products.Our results provide information for selection breeding program of coloured alpaca helpful to improve the condition of welfare of Andean populations.

Abstract

The heritability and the phenotypic and genetic correlations of down weight (DW), down fibre diameter (DFD), and coefficient of variation of the down fibre diameter (CVDFD) of Chinese Alashan Left Banner White Cashmere goat were estimated on 1375 one-year-old animals, born in 2009, 2011 and 2013 and bred at the Station for Livestock Improvement of Alashan (Left Banner, Inner Mongolia, P.R. China). For all traits, significant effects were for sex, cohort and sex–cohort interaction (p < .001). The heritability for DFD and CVDFD was high, 0.41 ± 0.08 and 0.52 ± 0.06, respectively. Heritability for the DW was low (0.12 ± 0.03). Phenotypic correlation calculated by Pearson’s coefficient showed that DFD is positively correlated with both CVDFD (0.29 ± 0.07) and DW (0.20 ± 0.05). The phenotypic correlation between CVDFD and DW was negative (−0.11 ± 0.06). The genetic correlations between DW and CVDFD and between DFD and CVDFD were both high and positive (0.63 ± 0.16 and 0.39 ± 0.1, respectively) while the DW showed a negative genetic correlation with DFD (−0.27 ± 0.2). Our results suggest that the selection for reducing DFD and its CVDFD is possible and a genetic progress can be achieved quickly in the Chinese Alashan Left Banner White Cashmere goat.
