31. Insects associated with chicken manure in a breeder poultry farm of Central Chile

Abstract

Insects are common in poultry facilities and different biosecurity measures are enforced to prevent their spread due to the fact that they may carry pathogenic agents. Therefore, it is of utmost importance to know what insects are commonly present in poultry sheds to optimize control protocols. Since information on this subject is scarce, this investigation aimed to determine the main insect taxa present in chicken manure on a major poultry farm in Central Chile. Samples of hen manure were collected at a poultry farm in the Region of Valparaíso, Chile, from areas adjacent to feeding lines and water reservoirs. Samples were chilled, transported to the laboratory and processed for taxonomic classification of both adult and immature stages of insects. Results indicated a marked colonization of the beetleAlphitobius diaperinusand of the dipteransFanniasp., compared to other six families of insects that were also determined. About 94% of the insects found in chicken manure were present in samples from areas adjacent to water reservoirs. Therefore, leaks from water supply devices become a critical point of control of these entomological poultry pests that have been reported to carry a variety of viral, bacterial and eukaryotic parasites. &nbsp, Los insectos son muy comunes en las instalaciones de la industria avícola y diferentes medidas de bioseguridad se aplican para evitar su propagación debido al hecho que pueden acarrear agentes patógenos. Por lo tanto, es de suma importancia saber qué insectos comúnmente están presentes en los galpones avícolas para optimizar los protocolos de control. Ya que la información sobre este tema es escasa, el objetivo de la presente investigación fue determinar los principales grupos taxonómicos de insectos presentes en el guano de una granja avícola industrial en la zona central de Chile. Las muestras de guano fueron recolectadas en una granja avícola en la Región de Valparaíso, Chile, de áreas adyacentes a las líneas de alimentación y depósitos de agua. Las muestras fueron refrigeradas, transportadas al laboratorio y procesadas para la clasificación taxonómica de los adultos y estados inmaduros de insectos. Los resultados indican una colonización marcada por el escarabajoAlphitobius diaperinusy por los dípterosFanniasp. en relación con otras seis familias de insectos que se determinaron. Alrededor del 94% de los insectos encontrados en el guano estaban presentes en las muestras procedentes de las zonas adyacentes a las reservas de agua. Por lo tanto, las fugas de los dispositivos de suministro de agua se han convertido en un punto crítico de control de estas plagas entomológicas de las aves de corral, las cuales han sido reportadas como portadoras de una gran variedad de virus, bacterias y parásitos eucariotas. &nbsp

Abstract

Salmonella entericaserovar Enteritidis (SE) remains an important enteropathogen in the poultry industry and public health. Due to the limited effectiveness of control measures, lytic bacteriophages have shown a potential use as biocontrollers of SE in birds. The aim of this study was to determine the effectiveness of prophylactic therapy with phages to control intestinal and reproductive tract colonization of SE in laying hens. 22-week old Hy-Line Brown hens free ofSalmonella, were treated with a mixture of three bacteriophages (1011PFU/dose/phage) and challenged with 2.4 x 108CFU of SE, 24 hours post phage treatment. On day 10 post challenge, hens were euthanatized, and individual samples of cecum, ovary and oviduct, were analyzed using qualitative and quantitative bacteriology. Eggs laid during the experience were collected and processed to detect SE. The incidence ofSalmonellain ceca was similar between positive control and treated groups (96.67%) and cecal bacterial counts did not present significant differences between them (P > 0.05). In reproductive tissues, phagetherapy was able to slightly reduce the SE count in the ovary (P 0.05). This lytic activity of phages observed in ovaric tissue, encourages further efforts to elucidate the real contribution of bacteriophages as SE biocontrollers in laying hens. &nbsp, Salmonella entericaserotipo Enteritidis (SE) continúa siendo un importante enteropatógeno para la industria avícola y para la Salud Pública. Debido a la limitada efectividad de las medidas de control, los bacteriófagos líticos han mostrado un uso potencial como biocontroladores de SE en aves. El propósito de este estudio fue determinar la efectividad de una terapia profiláctica con fagos para el control de la colonización de SE en el tracto intestinal y reproductivo de gallinas de postura. Gallinas Hy-Line Brown, de 22 semanas de edad, libres deSalmonella, fueron tratadas con una mezcla de tres bacteriófagos (1011UFP/dosis/fago) y desafiadas con 2,4 x 108UFC deSE, veinticuatro horas postratamiento. Al día 10 postdesafío, las gallinas se sometieron a eutanasia, obteniéndose muestras individuales de ciegos, ovario y oviducto, las cuales fueron analizadas por bacteriología cualitativa y cuantitativa. Se recolectaron los huevos puestos durante la experiencia y se procesaron para detectar SE. La proporción deSalmonellaEnteritidis en los ciegos fue similar entre los grupos control positivo y tratado (96,67%) y el recuento bacteriano cecal tampoco presentó diferencias significativas (P > 0,05). En los tejidos reproductivos, la administración de fagos no disminuyó la incidencia de infección (P > 0,05), pero fue capaz de reducir levemente los recuentos de SE en ovario (P 0,05). La actividad lítica de los fagos demostrada en tejido ovárico, aunque escasa, sugiere realizar mayores esfuerzos para dilucidar la real contribución de los bacteriófagos como biocontroladores de SE en gallinas de postura. &nbsp

Abstract

Antarctic and Southern Ocean science is vital to understanding natural variability, the processes that govern global change and the role of humans in the Earth and climate system. The potential for new knowledge to be gained from future Antarctic science is substantial. Therefore, the international Antarctic community came together to ‘scan the horizon’ to identify the highest priority scientific questions that researchers should aspire to answer in the next two decades and beyond. Wide consultation was a fundamental principle for the development of a collective, international view of the most important future directions in Antarctic science. From the many possibilities, the horizon scan identified 80 key scientific questions through structured debate, discussion, revision and voting. Questions were clustered into seven topics: i) Antarctic atmosphere and global connections, ii) Southern Ocean and sea ice in a warming world, iii) ice sheet and sea level, iv) the dynamic Earth, v) life on the precipice, vi) near-Earth space and beyond, and vii) human presence in Antarctica. Answering the questions identified by the horizon scan will require innovative experimental designs, novel applications of technology, invention of next-generation field and laboratory approaches, and expanded observing systems and networks. Unbiased, non-contaminating procedures will be required to retrieve the requisite air, biota, sediment, rock, ice and water samples. Sustained year-round access to Antarctica and the Southern Ocean will be essential to increase winter-time measurements. Improved models are needed that represent Antarctica and the Southern Ocean in the Earth System, and provide predictions at spatial and temporal resolutions useful for decision making. A co-ordinated portfolio of cross-disciplinary science, based on new models of international collaboration, will be essential as no scientist, programme or nation can realize these aspirations alone.
