Aims/Introduction Periodontal disease, a chronic inflammation induced by bacteria, is closely linked with diabetes mellitus. Many complications associated with diabetes are related to epigenetic changes. However, the exact epigenetic changes whereby diabetes affects periodontal disease remain largely unknown. Thus, we sought to investigate the role of diabetes‐dependent epigenetic changes of gingival tissue in the susceptibility to periodontal disease. Materials and Methods We studied the effect of streptozotocin‐induced diabetes in minipigs on gingival morphological and epigenetic tissue changes. Accordingly, we randomly divided six minipigs into two groups: streptozotocin‐induced diabetes group, n = 3; and non‐diabetes healthy control group, n = 3. After 85 days, all animals were killed, and gingival tissue was collected for histology, deoxyribonucleic acid methylation analysis and immunohistochemistry. Results A diabetes mellitus model was successfully created, as evidenced by significantly increased blood glucose levels, reduction of pancreatic insulin‐producing β‐cells and histopathological changes in the kidneys. The gingival tissues in the diabetes group presented acanthosis of both gingival squamous epithelium and sulcular/junctional epithelium, and a significant reduction in the number and length of rete pegs. Deoxyribonucleic acid methylation analysis showed a total of 1,163 affected genes, of which 599 and 564 were significantly hypermethylated and hypomethylated, respectively. Immunohistochemistry staining showed that the hypomethylated genes – tumor necrosis factor‐α and interleukin‐6 – were positively expressed under the junctional epithelium area in the diabetes group. Conclusions Diabetes mellitus induces morphological and epigenetic changes in periodontal tissue, which might contribute to the increased susceptibility of periodontal diseases in patients with diabetes., This is the first study investigating the effect of diabetes on periodontal tissue from an epigenetic perspective. Using a streptozotocin‐induced diabetes model in pigs, we showed that induction of diabetes results in significant changes in the methylation status of multiple genes in gingival tissue.

The rabies virus is one of the most neurotropic of all viruses infecting mammals. During the terminal phases of infection, the virus spreads to peripheral tissues, including the skin. The external skin of the nose, called the nasal planum, is a sensory organ where numerous nerve bundles and terminal nerves are distributed. Therefore, the nasal planum is expected to serve as a postmortem diagnostic material. However, the distribution of rabies virus antigens in the nasal planum in rabid animals has not yet been studied. In this study, the nasal planum was obtained from 45 rabid dogs. In all rabid dogs, the viral antigen was detected in the peripheral nerve tissues, Merkel cells, and squamous cells. The viral antigen in the epidermis exhibited three patterns: first, a diffuse positive pattern from the basal layer to the squamous layer; second, a reticular positive pattern along the cell membrane in the squamous layer; and third, a basal layer pattern of the epidermis. In the dermis, viral antigens were detected more often in lamellated corpuscles just beneath the rete pegs. These results suggest that the nasal planum could serve as a useful alternative source for postmortem diagnosis in rabies endemic countries.

Skin is the largest organ in the body of man and animal where interdigitation of the dermal papillae with epidermal pegs enhances structural stability of the skin especially where frequent pressure is applied. Cane rats are robust rodents with stocky bodies. They are only found in Africa and only a single species known as Thryonomys has been found to exist. This work was designed to establish the nature of the interdigitating structural disposition of the epidermal-dermal interface in T. swinderianus (Greater cane rat) which may account for the fragility and easy tear of their skin. A total of twenty adult male greater cane rats were used. Three samples sized 1cm to 2cm were taken from each of 14 different body sites for histological evaluation for rete pegs. These pegs were generally few in six regions with the following average per light microscopic field: head (5.5±3.7), neck (1.0±1.7), forelimb (5.6±3.7), trunk (4.3±4.2), tail (1.0±1.7) and the hindlimb (6.3±5.3). The neck and the tail parts of the body have the least concentration of this structure compared to other regions evaluated. This few in number of rete pegs may account for the fragility of the skin which makes it difficult to restrain or handle without damage during research work procedures and management during domestication. Cadmium is a toxic, heavy industrial metal that causes serious environmental health hazards. Cadmium containing compounds have been classified as known human carcinogens and epidemiological data show causal associations to different types of cancers. This study designed to assess the carcinogenic effects of Cadmium on the gingival tissue of rats. Material and methods; Sixteen male albino Rats (age: 8 to 10 weeks and about 150 to 180 g body weight) from Animal Center of Faculty of Medicine, Mansoura University. The animals were divided into two equal groups, group I (control) and group II (cadmium chloride) rats were given 2 ml dose of a solution containing 10 mg/kg body weight of monohydrated cadmium chloride orally by gavage needle for 24 days. By the end of experiments, gingival epithelium around the teeth were carefully dissected, processed and stained using Haematoxyline & Eosin and immunohistochemical evaluation by iNOS. Statistical analysis was done to observe the statistical significant differences between the Cd group and control group in relation to dysplastic changes and iNOS immunoreaction. Results: the gingival tissue of Cd group revealed several dysplastic changes as, drop shape rete pegs, loss of polarity of basal cells, nuclear hyperchromatism, pleomorphism and increased mitotic pattern. a statistical significant difference regarding iNOS immunoreaction was found between control group and Cd group. conclusion to this study Cadmium chloride at low concentration and longtime of exposure can cause dysplastic changes in gingival tissue of rats.

This study aims to examine the composition of lining and masticatory mucosa at the pre- and post-soft tissue augmentation procedures with a volume-stable cross-linking collagen matrix (VCMX) in humans. In 12 patients, single implant sites were augmented with a VCMX. Biopsies were obtained including masticatory (MM) and lining (LM) mucosa before augmentation and at 12 weeks post-augmentation procedures. Rete pegs density (RPD), length (RPL), and blood vessel density (BVD) were histomorphometrically analyzed at both time points. Picrosirius red staining under polarized light microscopy was used to evaluate collagen fiber organization. The effects of time and tissue type were evaluated by ANOVA with repeated measures. Regeneration of large acute and chronic wounds is a concern worldwide. The present study evaluates wound healing competence of a completely human-origin, extracellular matrix (ECM)-based skin substitute/graft. It comprises cell-less amniotic membrane (AM), clinical-grade fibrin (FIB), and hyaluronic acid (HA) termed as AMFIBHA. The use of large-area third-degree rabbit burn wounds evaluated the product efficiency. The AMFIBHA induces hemostasis and permits suture-less positioning on the wound bed. In wet wounds, the AMFIBHA degrades and release biologically active molecules and guide cell migration, proliferation, and regeneration. The study demonstrated the effectiveness of this wound care product in terms of epithelial-dermal regeneration with angiogenesis. The study assessed injury-associated inflammation and different wound healing markers after 28 days of experiment and compared with both positive and negative controls-treated wounds. The regeneration of mature epidermis and dermis with rete pegs and hair follicle-like structure was evident upon a single application. The active involvement of host cells resulted in supple tissue formation. The ECM organization of AMFIBHA-treated tissue resulted in re-gain of mechanical properties comparable to native skin after 56 days. These guided regenerative outcomes reveal a promising translational value of the novel AMFIBHA skin substitute as an off-the-shelf product for clinical use.

Dowling–Degos disease (DDD) is a rare genodermatosis with autosomal dominant inheritance. It is characterized by reticulate pigmentation of flexures, comedo-like follicular papules, and perioral pitted scars. Here, we report a 3-year-old female child presenting with hyperpigmentation of the external genitalia, intergluteal fold, and armpits with follicular keratotic papules over the neck. Histopathological examination showed hyperkeratosis, irregular acanthosis, papillomatosis, and basal layer pigmentation in the epidermis with elongated and branched rete pegs and relatively more melanin concentration in the tips of the rete pegs. The diagnosis of DDD was made. This case is reported due to the unusual early onset and involvement of external genitalia.

Introduction: double lip is one of lip abnormalities that may be congenital or acquired. It consists of a fold of excess tissue on the mucosal side of the lip. It causes functional and aesthetic problems to affected patientsAim of the work : The present study was performed to compare clinically and histopathologically between redundant tissue in double lip and normal labial mucosa.Subjects and methods: Seventeen patients with congenital non syndromic double lip were selected. Redundant tissues were removed surgically together with part of adjacent normal mucosa as part of the surgical manipulation. Hematoxylin and eosin staining was done to compare both tissues microscopically. Expression of ki-67 and p53 was evaluated in both tissue by immunohistochemistryResults: Clinically, redundant tissue appeared as thick extra fold with normal mucosal color and soft consistency analogous to normal labial mucosa. This redundant tissue was removed surgically without any complications. Histopathologically, normal labial mucosa revealed non keratinized stratified squamous epithelium and delicate underlying connective tissue. Excess tissue exhibited parakeratosis, acanthosis, elongation of rete pegs. The underlying connective tissue was fibrotic with inflammatory cell infiltration. In normal labial mucosa, ki-67 was detected only in parabasal layer of epithelium whereas it was localized in parabasal and superficial layers of epithelium in redundant tissue. No expression of p53 was detected in both normal and excess tissue. For both ki-67 and p53, the difference in expression between normal and redundant tissue was significant.Conclusion : Redundant tissue is a benign hyperplastic tissue that require surgical removal. So surgical excision here is the treatment of choice when excess tissue causes functional and aesthetic problems. Moreover, the high proliferation of redundant tissue may necessitate the surgical excision of this tissue.

Background Odontogenic Keratocysts (OKCs) are known for their rapid growth and tendency to invade the adjacent tissues. Its high recurrence rate (2.5%–62%) has attracted many kinds of research to identify new parameters to predict recurrence that would enable better treatment outcomes for such patients. The present study aims to correlate Sub-Epithelial Hyalinization (SEH) that has not been explored in an Asian population to date and other histopathologic features of OKC to its recurrence propensity. Materials & Methods A total of 60 OKCs were analyzed for histopathologic correlation of thickness of lining, complete/incomplete lining, corrugated surface, ortho/para keratinization, intercellular edema, reversed polarity, basilar hyperplasia, the palisading arrangement of basal cells, presence/absence of rete pegs, folding of the epithelium, epithelial/connective tissue separation, SEH, basal offshoots, daughter cysts and inflammation with recurrence to arrive at the most significant histologic feature predicting recurrence. Results The most significant histologic parameter differentiating recurrent and non-recurrent OKCs was the presence of SEH (p = 0.004), incomplete lining epithelium (p ​= ​0.023), and a corrugated surface (p ​= ​0.049). Several other histologic parameters evaluated did not statistically correlate with recurrence, this can be attributed to the smaller sample size considered in the present study. Conclusion SEH is a reliable histologic parameter to predict recurrence in OKCs. The presence of SEH is indicative of a higher recurrence potential in OKCs. Evaluation of histological parameters and their correlation with recurrence in OKCs on a larger sample size could validate the results of the current study undertaken and potentially unravel more insights on predicting recurrence. BACKGROUND Multiple lasers have been used for the treatment of cafe au lait macules (CALMs) with various results. Objective tools to predict therapeutic efficacy of CALMs treatment is lacking. OBJECTIVE To determine whether reflectance confocal microscopy (RCM) characteristics correlate with CALMs response to laser treatment. MATERIALS AND METHODS All CAMLs underwent RCM examination of length and density of dermal papillae followed by 3 sessions of Q-switched alexandrite laser (QSAL). A visual analog scale was used to assess clinical treatment efficacy. RESULTS Forty-three patients were included, 22 had CALMs with irregular borders and 21 with smooth borders. Cafe au lait macules with irregular border had shorter rete pegs and less papillae (p < .05) on RCM compared with smooth border CAMLs and responded better to QSAL treatment (2.32 vs 1.10). CONCLUSION Reflectance confocal microscopy measurement of length and density of papillae were inversely correlated with treatment response. Reflectance confocal microscopy may be a useful tool to predict CALMs response to laser treatment.

Rete pegs are finger-like structures that are formed during the development and wound healing process of the skin and oral mucosa, and they provide better mechanical resistance and nutritional supply between the epithelium and dermis. An increasing number of studies have shown that rete pegs have physiological functions, such as resisting bacterial invasion, body fluid loss, and other harmful changes, which indicate that rete pegs are important structures in natural skin and oral mucosa. Although a great deal of progress has been made in scaffold materials and construction methods for tissue-engineered skin and oral mucosa in recent years, construction of the oral mucosa with functional rete pegs remains a major challenge. In this review, we summarized current research on the progress on formation of rete pegs in human oral mucosa as well as its molecular basis and regulatory mechanism, which might provide new ideas for functional construction of tissue-engineered skin and oral mucosa.

Background The present report describes a case of pseudocowpox virus (PCPV) infection in a seven-year-old female bison euthanized due to a history of declining condition and sores on the vulva and udder. Case presentation External examination revealed multifocal, raised, keratinized plaques (0.5–2 cm) covering the skin of the ventral surface of the tail, perineum, caudoventral abdomen, udder, both inguinal recesses, and the medial aspects of both thighs. No significant gross lesions were present in the reminder of the tissues examined. Histopathological examination of the affected skin showed moderate epidermal hyperplasia with rete pegs, marked parakeratotic hyperkeratosis with crusts of degenerate neutrophils and cell debris, and few epithelial cells undergoing ballooning degeneration with occasional eosinophilic intracytoplasmic inclusion bodies (3–5 μm Bollinger body). Negative staining electron microscopy from skin revealed typical Parapoxvirus (PPV) particles, which were also confirmed by real-time PCR (Ct =18.6). Metagenomic analysis of the skin samples revealed only poxviruses. The bison parapox B2L envelope gene clustered with other parapox sequences identified from ruminants. Conclusions This is the first report of PCPV virus infection in an American bison. Identification of novel susceptible hosts of parapox viruses sheds light on the viral evolution and highlights the importance of potential economic impact of this disease to the bison industry.

The objectives of this study were to clinically and histologically assess the capacity of bilaminar subepithelial connective tissue grafts (SCTGs) alone or in combination with gingivoplasty (Gv) to increase the keratinized gingiva width (KGW) in contralateral mandibular sites lacking KG (10 patients, 42 sites). The effects of Gv timing (1 vs 2 months) and technique (blade vs laser) were also evaluated. SCTGs alone resulted in mean KGW increase of 0.1 to 0.7 mm. Laser-Gv significantly increased KGW by an additional 1.9 mm at 4 months postabrasion as opposed to 0.9 mm achieved with blade-Gv. Histologically, laser-treated sites displayed parakeratinization with more pronounced rete pegs than observed in blade-abraded sites.

Background: The extent of involvement of cervical lymph nodes is known to be the most important prognosticator in oral squamous cell carcinoma (SCC) that significantly affects the survival rate of patients. The clinical, radiological and pathological factors that can predict cervical lymph node metastasis are yet to be ascertained clearly, which poses a challenge for the surgeon to determine the extent of neck dissection. Aim: This study aims to identify the clinical and histopathologic predictors of lymph node metastasis among patients with oral SCC and to devise a scoring system based on those predictors to aid in better clinical decision-making regarding the extent of neck dissection. Setting: Malabar Cancer Centre, a specialized tertiary cancer care center in Kerala, India. Methods: A retrospective review of 160 patient records and biopsy slides collected and preserved between June 2014 and May 2016. Conclusion: The clinicopathologic parameters such as site of cancer (P = 0.03), histologic differentiation (P = 0.03), shape of rete pegs (P = 0.002), pattern of invasion (P = 0.0001) and depth of invasion >3 mm (P = 0.016) were significantly associated with the risk of lymph node metastasis. The risk score devised based on these predictors serves as an efficient tool in aiding clinical decision-making regarding the extent of neck dissection.

OBJECTIVES To evaluate the histologic and volumetric changes of gingival tissues following grafting with collagen-based matrices at labial aspect of teeth in canines. MATERIALS AND METHODS Gingival augmentation was performed in the mandibular incisor area using two types of xenogeneic cross-linked collagen matrices (CCMs), bovine CCM for BCCM group and porcine CCM for PCCM group, whereas the contralateral sides remained untreated (B-control group and P-control group). Descriptive histology, histometric and volumetric analyses were performed after 12 weeks. For statistical comparison between each test group and respective control group, paired t test was used for histometric analysis, and repeated-measured analysis of variance was used for volumetric analysis (p < 0.05). RESULTS An increased number of rete pegs and an enhanced formation of new blood vessels were observed at both grafted sites compared to the corresponding control sites. There was statistically significant gain of horizontal thickness only in BCCM group (1.36 ± 0.27 mm vs. 1.26 ± 0.34 mm; p < 0.05) compared to the B-control groups. CONCLUSION BCCM was effective for gingival augmentation in terms of horizontal thickness at the labial aspect of teeth at 12 weeks post-surgery.

Automated identification of healing progression in lower limb wounds is an important clinical challenge since such wounds fail to heal in an orderly and timely manner. Selection of suitable target features and employment of appropriate analytical tool are critical for the precise monitoring of the healing progression through automated diagnostic method. The present study aims at automated detection of healing trend based on characteristics of epithelial rete pegs, which is considered as an important parameter for monitoring and hence healing progression of epithelial maturation. To this aim, the applicability of various segmentation methods namely, watershed, marker-controlled watershed, k-means clustering and active contour method and comprehensive characterization of rete-pegs are studied and compared. Among them active contour provided better performance in terms of non-linear objective assessments including performance metrics like-peak-signal-to-noise-ratio, processing speed and Mean Square Error. Further, support vector machine is trained for classification of the rete-peg features at different time points of clinically diagnosed non-healing wounds treated with a specific protocol. Kappa scoring tests inter-rater agreement between applied segmentation protocol and ground truth images. The overall accuracy of the system has been found to be 95%. It is thus concluded that with the aid of the proposed segmentation and classification method an automated detection system can be developed for precise monitoring of lower limb wound healing progression.

Verruciform xanthoma (VX), a papillary or wart-like lesion of the mucosa, is histopathologically characterized by papillary projection of the epithelium and an aggregation of foam cells in the lamina propria. Here, we describe a case of VX in the posterior mandibular gingiva, initially suspected to be a benign lesion based on clinical findings and cytology prior to an excisional biopsy. The patient was a 62-year-old man who had visited a local dentist approximately 1 year earlier, presenting with a white lesion in the left posterior mandibular gingiva that resisted removal by scraping. The lesion was left untreated as there were no subjective symptoms. Thereafter, the surface of the lesion roughened and the patient was referred to our department for a comprehensive examination. A circumscribed, granular mass, 15-mm in diameter, with a red and white surface was observed in the left posterior mandibular buccal gingiva. Exfoliative cytology was performed. The diagnosis was a class III lesion. Excisional biopsy was performed under local anesthesia. Histopathological examination led to a diagnosis of VX. At 1 year postoperatively, the patient is making satisfactory progress without recurrence. Verruciform xanthoma is difficult to diagnose preoperatively, and is commonly resected under a clinical diagnosis of papilloma or benign tumor. A benign lesion was also initially suspected in the present case and cytological analysis performed to confirm absence of malignancy. The lesion could not be diagnosed as VX preoperatively. Verruciform xanthoma can be over-diagnosed based solely on cytological examination because it often involves cellular atypia reflecting its characteristic extension of rete pegs and keratinization of surface cells, indicating the need for care in arriving at a definitive diagnosis.

AimsTo demonstrate and characterise distinct subepithelial compact nerve endings (CNE) at the human corneoscleral limbus.MethodsTen fresh human donor corneoscleral discs (mean age, 67 years) and 26 organ-cultured corneoscleral rims (mean age, 59 years) were studied. All samples were subjected to enzyme histochemical staining related to endogenous acetylcholinesterase present in nerve tissue and H&E staining. Whole-mount en face imaging with NanoZoomer digital pathology microscope and serial cross-section imaging with light microscope were undertaken.ResultsNerves entering the corneoscleral limbus and peripheral cornea terminate under the epithelium as enlarged multiloculated and multinucleated ovoid structures within a 2 mm zone. They are closely associated with the rete pegs of the limbal palisades and the limbal epithelial crypts, often located within characteristic stromal invaginations of these structures. Their numbers ranged from 70 to 300 per corneoscleral rim. The size ranged from 20 to 100 µm. They had one or more nerve connections and were interconnected to other similar endings and to the limbal nerve plexus.ConclusionHuman corneoscleral limbus demonstrates a population of nerve terminals resembling CNE with distinct morphological features. They are closely associated with the limbal stem cell niches, suggesting a potential contribution to the niche environment.

Purpose To further study the mechanism of epithelization on the fascia side of the flap after surgical incision and the treatment of the negative pressure therapy. Methods With the patients' informed consent, parts of tissue samples were obtained from a 51-year-old diabetic patient who was suffering lower extremity ulcers. The samples were processed with hematoxylin and eosin (HE) staining and Masson trichrome staining. The keratin 19, keratin 15 and carcino-embryonic antigen (CEA) were immunohistochemically detected. Results The results of HE staining showed that the specimen was divided into two regions, newborn area and original epithelial area. There were more inflammatory cells infiltrating in the dermis in the newborn epithelial area, compared with the original epithelial area. Cells in newborn epithelial area were more active and many dinuclear and polynuclear cells were observed in newborn epithelial area. But there were more cuticular layers and obvious rete pegs in original epithelial area. In addition, the cells with keratin 19 and CEA positive were found around hair follicle, while keratin 15 was negative. Masson trichrome staining showed that there was a lot of de novo collagen in newborn epithelial area. Conclusion Epidermal cells on the fascia side of the flap could be derived from the stem cells. Negative pressure wound therapy would attract not only cells but also other elements such as growth factors, cytokines, some nutrients and extracellular matrix. With the formation of the appropriate microenvironment after debridement, the migrated cells can grow, differentiate and spread, eventually leading to the epithelization on the fascia side of the flap in diabetic foot.

Oral epithelial adhesion to the lamina propria underlies the physiologic function of the oral mucosa and contributes to resisting bacterial invasion, preventing body fluid loss, and maintaining routine chewing; thus, understanding the factors that positively influence oral epithelial adhesion is a research topic of great interest. Rete pegs contribute to oral epithelial adhesion by enlarging the contact areas, whereas integrins are the major molecules that mediate epithelial cell adhesion to the basement membrane. Keratinocyte growth factor (KGF) can promote both rete peg elongation in the skin and the expression of integrins in various cell types. Herein, we tested the effects of submucosal injection of KGF in the ventral surfaces of rat tongues on oral epithelial adhesion. The data confirmed that topical injection of KGF elevated the adhesive forces, elongated the rete pegs, and increased the abundance of integrins, KGF, and KGF receptor on the rat tongue ventral surface. However, HYD-1 (Lys-Ile-Lys-Met-Val-Ile-Ser-Trp-Lys-Gly), an integrin antagonist, inhibited the KGF-enhanced epithelial adhesion and rete peg elongation. Moreover, KGF promoted the expression of integrin subunits α6, β4, α3, and β1 in human immortalized oral epithelial cells in 2- and 3-dimensional culture systems. In vitro cell attachment assays demonstrated that HYD-1 inhibited the adhesion of human immortalized oral epithelial cells on Matrigel. Strikingly, the expression of integrins, KGF, and KGFR in human masticatory mucosae with longer rete pegs was more abundant than that in the lining mucosae with shorter rete pegs. In addition, rete peg lengths were positively correlated with the expression levels of integrins, KGF, and KGF receptor. These findings indicate that KGF strengthens oral epithelial adhesion and rete peg elongation via integrins.

Background: Sex hormones play an important role in periodontal tissue integration. Oral hormonal contraceptive pills (OCPs) were developed over 50 years ago. Different studies reported higher progression rate of gingival inflammation in females using OCPs. The present study was performed to evaluate histologically the influence of oral contraceptives on periodontal tissues.Materials and Methods: A total of thirty healthy female Wistar albino rats aging between 10 to 12 weeks and weighing 230 – 260 gm were enrolled in this study. The animals were divided into two groups, fifteen rats each. Group I (Control group), rats receiving no medication. Group II (Study group), rats received Microcept® monophasic combined oral contraceptive tablets for 36 days. Histological examination of the peridontium was accomplished using light microscopy. Results: Light microscopic examination of the control group revealed that the gingival epithelium was parakeratinized stratified squamous epithelium with long pointed rete pegs. Pyknotic nuclei were seen in the keratinous layer. Concerning the study group, the gingival epithelium revealed orthokeratinzed stratified squamous epithelium with short and blunt rete pegs and absence of nuclei in keratinous layer. Dentin and cementum showed external resorption.Conclusion: The current study showed that oral contraceptives exhibited histological changes on the periodontium which can be correlated to different clinical findings.

Introduction: The morphology of the epithelium of the oral lips comprised keratinized external epithelium (anteriorly) and nonkeratinized or sometimes parakeratinized mucous membrane epithelium (posteriorly). Knowledge of morphometry of the lip lining helps in deciding the best site for choosing graft for its better uptake during several dermal grafting procedures following trauma or tumor excision following craniofacial cancers or cosmetic procedures. Materials and Methods: Ten human male cadavers were procured at the Department of Anatomy King George's Medical University, Lucknow, Uttar Pradesh. The rectangle-shaped skin specimen through the right commissure of the lip which included the skin, mucocutaneous junction, and mucosa was stained with hematoxylin and eosin stain. A total of 30 slides were prepared. Thus, the readings were obtained for three regions, respectively, with the help of CAT-CAM E-series HD cameras which were installed in a light microscope. Results: Thickness of skin (epidermis + dermis) of the lip ranged from 756 μm to 1068 μm among males. Epidermal thickness increases on moving from the cutaneous region to the mucosa region of the lip. The lowest contribution of the stratum corneum in thickness of the epidermis was observed in the vermillion region, while the highest contribution was observed in the skin region. It was found to be absent in the mucosa region of the lip. Rete pegs at the dermoepidermal junction was found to be maximum in the vermillion region and minimum in the skin region. Its depth increased as we move from the skin to the mucosa region of the lip. Depth of the dermis was found to be maximum in the skin region, while minimum in the vermillion region. It ranged between 291 μm and 693 μm. Conclusion: Care should be taken while using dermal fillers in lip augmentation surgeries, especially in the vermillion region due to its close proximity to musculature in the core of the lip.

Odontogenic keratocysts (OKCs) have a diagnostic thin epithelial lining characterised by a linear epithelial connective tissue interface generally lacking inflammatory changes, basal palisading of the nuclei and a wavy parakeratotic layer on the surface. This typical epithelium may convert to a thicker non-keratinizing one with rete pegs and a relatively flat surface after operative decompression. The aim was to characterize this type of epithelial change by immunohistochemistry for bcl2, keratin17, 10 and 19. Eleven out of 33 archived OKCs demonstrated an altered epithelium related to previous biopsy, decompressing drainage or inflammation. The typical basal bcl2 staining was lost in 10/11 cases; transepithelial CK17 was lost or markedly reduced in 9/11 cases. CK10 displayed a segmental upper layer staining in OKCs, and its loss or partial loss in the altered epithelium did not differ from negative areas of OKCs. CK19 displayed various staining patterns in the altered epithelium from lost to maintained in a patchy transepithelial distribution, the latter of which did not differ from the typical OKC staining pattern. Three of four non-keratinizing epithelial linings with basal palisading displayed immunostaining reminiscent of typical OKC epithelium. The lack of a typical epithelium is not sufficient to exclude the diagnosis of OKC if the sampling is not generous (e.g. biopsy), and the presence of non-keratinizing epithelium with basal palisading and an immunophenotype characteristic of OKC (basal bcl2, patchy or diffuse CK17 and upper layer CK10 positivity) may be consistent with the OKC diagnosis even in the absence of typical epithelial lining.

Clear cell acanthoma (CCA), also known as pale cell acanthoma, represents a rare benign epidermal tumor with strong predilection for the lower extremities of middle-aged individuals and no frank gender preference. The etiology of CCA is poorly understood, although a localized psoriasiform reaction is favored. Herein, we report on the clinicopathologic and immunohistochemical features, and HPV status of an apparent example of oral CCA. A 58-year-old female presented with a well-circumscribed, asymptomatic, exophytic, sessile and erythematous nodule of the right hard palate, measuring 0.7 cm in greatest dimension. Microscopically, the lesion featured parakeratosis and acanthosis with neutrophilic microabscesses and broad elongated rete pegs. In areas, spinous epithelial cells exhibited pale or clear cytoplasm without nuclear pleomorphism, mitoses or cytologic atypia. The supporting connective tissue revealed mild chronic inflammation with few scattered neutrophils and numerous capillary vessels. PAS histochemical stain with and without diastase disclosed the presence of cytoplasmic glycogen in the pale cells. The majority of glycogen-rich epithelial cells stained strongly for EMA and were negative for D2-40. Ki-67 immunostaining was confined only to the basal cell layer of the epithelium. A diagnosis of CCA was rendered. The lesion was negative for human papillomavirus (HPV) infection, as assessed by HPV-DNA PCR using the MY09/11 primers for the L1 conserved region, thus HPV infection does not appear to contribute to the pathogenesis of oral CCA. In conclusion, we report an intraoral example of CCA in order to raise awareness about this entity.

Background Basement membrane remodeling is an indispensable factor for oral mucosal rete peg formation, but how the basement membrane is remodeled remains unclear. Our previous study indicated that keratinocyte growth factor induces the assembly of podosomes, which are dynamic organelles critical for matrix remodeling in human immortalized oral epithelial cells. This study explores podosome formation and its role in basement membrane remodeling during murine oral mucosal rete peg formation. Methods Perinatal murine palatal tissue slices were obtained from embryonic day 17.5 (E 17.5) to postnatal day 10.5 (P 10.5) BALB/c mice. Rete peg formation was observed by hematoxylin and eosin (HE) staining. Proteolysis of the basement membrane was detected by immunofluorescence staining. The assembly of podosomes and their correlation with basement membrane proteolysis were investigated by laser scanning confocal microscopy. Results The shape of basal layer keratinocytes at the sites of emerging rete pegs changed from typically polygonal to spindle-shaped. Basement membrane proteolysis, indicated by decreased type IV collagen (Col IV) staining, was detected during rete peg formation. Classical markers for podosomes, including cortactin/Tks5, WASP, and matrix metalloproteinase foci, were easily observed at the spindle-shaped cells. Podosomes were visible in regions where there was a significant decrease in Col IV staining. Conclusions These observations indicated that podosomes form at the front of the emerging rete peg and may play a pivotal role in basement membrane remodeling during rete peg formation.

EoE in children presents with four main symptoms. Most common symptoms exhibited by our clinic population are dysphagia (D) and abdominal pain (AP). Despite similar treatments, we found in an earlier study that the outcomes between these two groups were different. Therefore, we investigated if there exist any histological differences between these groups that could further our knowledge of EoE. Aim. To compare esophageal histology in detail, apart from the eosinophil count, between EoE-D and EoE-AP. Method. Biopsies of patients with EoE-D and EoE-AP were reevaluated for 10 additional histological criteria, in addition to the eosinophil count. Results. Both groups had 67 patients; peak mean eosinophil was 33.9 and 31.55 for EoE-D and EoE-AP (p<0.05). Eosinophilic microabscesses, superficial layering of eosinophils, and epithelial desquamation were twice as common and significant in EoE-D group than EoE-AP. Eosinophil distribution around rete pegs was also significantly higher in EoE-D group. The remaining criteria were numerically higher in EoE-D, but not significant, with the exception of rete peg elongation. Conclusion. EoE-D patients have significantly higher eosinophils compared to EoE-AP, and the level of inflammation as seen from eosinophil microabscesses, superficial layering, desquamation, and the distribution around rete pegs is significantly higher.

Aims Histological and histochemical analysis of retraction pocket of pars tensa of tympanic membrane in children. Identification of morphological abnormalities in comparison with a healthy tympanic membrane as it is described in standard textbook. Identification of signs typical for cholesteatoma and support for a retraction theory of cholesteatoma formation. Study design A prospective study analysing 31 samples of retraction pockets taken during surgery. Departments University Hospital, Children's Medical Centre Methods Samples of retraction pockets were processed by a standard process for light microscopy, stained by haematoxylin-eosin. Van Gieson's stain was used for differential staining of collagen, Verhoeff's stain for elastic fibre tissues, Alcian blue for acidic polysaccharides and PAS (Periodic Acid Schiff) method for basement membrane polysaccharides. Results The following findings were observed in the samples of retraction pockets: hyperkeratosis (100%), hypervascularisations (100%), subepithelial fragmented elastic fibres (96%), myxoid changes (87%), subepithelial inflammatory infiltration (84%), rete pegs (71%), papilomatosis (71%), intraepithelial inflammatory cellularizations, (48%), intraepithelial spongiosis (16%) and parakeratosis (3%). No basement membrane continuity interruptions were observed. Thickness of retraction pocket, thickness of epidermis, occurrence of rete pegs and frequency of fragmented elastic fibres was higher in a Grade III stage RP than Grade II stage RP (according to Charachon). Conclusion Morphological abnormalities in the structure of retraction pockets in comparison with a healthy tympanic membrane were described. The changes are typical for a structure of cholesteatoma (these changes are common in matrix and perimatrix), supporting retraction theory of its origin. Our observations show that it is inflammation that probably plays a key role in the pathogenesis of retraction pocket. The frequency of some of the changes increases with the stage of retraction pocket (II–III according to Charachon). Basement membrane continuity interruptions are not typical for retraction pockets.

Equine sarcoids are the most common equine skin tumors distributed world-wide which affects horses, donkeys and mules. Bovine papillomavirus (BPV) is causally associated with equine sarcoid with genetic, fly vectors and skin trauma identified as potential predisposing factors for development of the disease. The methods of transmission are contamination of the environment with subsequent trauma; infection of the skin direct or indirect transmission from affected animals; biting, rubbing, fomites or management practices and face flies. Dermatophytis, chronic skin rubbing, equine papilomatosis, squamous cell carcinoma, fibroma, fibrosarcoma, melanoma and granulation tissue are the common differential diagnoses of equine sarcoids. Clinical examination, histopathology, detection of BPVDNA by PCR, electron microscope and immunohistochemistry are the diagnostic technique of equine sarcoid. The histopathological characteristics of sarcoids, found that the classic epithelial changes of hyperkeratosis, hyperplasia, elongated rete pegs and picket fence. Treatment of equine sarcoids are challenging for the veterinarians due to the variable clinical presentation of the lesions and the frequent local recurrences. Both surgical and non-surgical techniques are used for the treatment of equine sarcoids with variable success rates. None of the current treatment techniques for equine sarcoid have been proven to be 100% successful. Awareness enhancement to make equine owners, prevention of fly contact, use of breeding from those individuals that are ‘immune’, high genetic resistance, early diagnosis and treatment of any confirmed lesions are the most valuable prevention methods for equine sarcoids. Natural aging process is reflected by gradual changes in the structure of the skin. These changes become very marked in old age. The changes in the epidermis and dermis as age advances is reflected externally as wrinkling, dryness, loss of elasticity, thinning and tendency towards purpura on minor injury. So the study is done in skin specimens by grouping the individuals in 4 age groups namely Group A (3-20yrs), Group B (21-50yrs), Group C (51-65yrs and Group D (>65yrs). The specimens were stained with Haematoxylin and Eosin stain and the changes in the thickness of the epidermis, number of Rete Pegs and the Dermo-Epidermal junction were observed. The changes in the skin appendages namely the sweat glands and the sebaceous glands were also noted. Verhoeff Van Geison Stain was done and the changes in the density and thickness of the elastic fibers and collagen fibers were noted. As the average life expectancy is increasing, the aging of skin presents a growing problem for the dermatologists. Human aging is characterized by a number of disorders like Epidermolysis bullosa and Phemphigous Vulgaris affecting the structure of the skin. So it is necessary to study the normal changes that occur in the skin as age advances which predisposes to various disorders. The study will be done among Indian population.

Background Major risk factors for oral squamous cell carcinoma (SCC) are tobacco smoking, a betel quid chewing habit, and heavy alcohol consumption. However, around 15% of oral SCCs cannot be explained by these risk factors. Although oral SCC associated with dental implants is quite rare, there has been a recent gradual accumulation of reports about it. Here, we report a case of primary peri-implant oral intra-epithelial neoplasia/carcinoma in situ (OIN/CIS) in a woman without the major risk factors for oral SCC. Case presentation A 65-year-old woman was referred to our clinic with a tumor in the right lower gingiva. She had no history of tobacco smoking and only drank socially. Ten years previously, mandibular right posterior teeth had been replaced with an implant-supported porcelain-fused-to-metal restoration in a dental clinic. About 7 years later, she noticed swelling on the lingual side of the gingiva around the implant-supported restoration, and was eventually referred to our clinic with the suspicion of a neoplasia around the dental implant. The upper part of the implant body was exposed on the implant corresponding to the first molar of the right side of the mandible; this was associated with painless, elastic soft, and relatively well circumscribed gingival swelling on the lingual site. A panoramic radiograph showed slight vertical bone resorption around the implants. An incisional biopsy was conducted under the suspicion of neoplasia. Pathological microscopic examination of the biopsy specimen revealed thickened squamous epithelia with slight nuclear atypism and disorders of the epithelial rete pegs. Immunohistochemical findings showed positive staining for keratin 17 and a negative staining mosaic pattern for keratin 13. High p53, p63, and Ki-67 reactivity was also observed. From these findings, OIN/CIS of the gingiva was pathologically diagnosed, and a wide local excision with rim resection of the mandible, including the implants, was performed. The pathological findings for the resected specimen were same as those for the biopsy specimen. After 1 year of follow-up, there was no evidence of recurrence. Conclusion In this case, prolonged peri-implant mucositis or peri-implantitis may have been a plausible risk factor for carcinogenesis.

Worldwide, there are over 450,000 new cases of oral cancer reported each year. Late-stage diagnosis remains a significant factor responsible for its high mortality rate (>50%). In-vivo non-invasive rapid imaging techniques, that can visualise clinically significant changes in the oral mucosa, may improve the management of oral cancer. We present an analysis of features extracted from oral images obtained using our hand- held wide-field Optical Coherence Tomography (OCT) instrument. The images were analyzed for epithelial scattering, overall tissue scattering, and 3D basement membrane topology. The associations between these three features and disease state (benign, pre-cancer, or cancer), as measured by clinical assessment or pathology, were determined. While scattering coefficient has previously been shown to be sensitive to cancer and dysplasia, likely due to changes in nuclear and cellular density, the addition of basement membrane topology may increase diagnostic ability- as it is known that the presence of bulbous rete pegs in the basement membrane are characteristic of dysplasia. The resolution and field-of-view of our oral OCT system allowed analysis of these features over large areas of up to 2.5mm x 90mm, in a timely fashion, which allow for application in clinical settings.

Verrucous lesions of the penis are rare and difficult to classify. Superficial biopsy in any of the verrucous lesions shows only hyperkeratosis, acanthosis, and papillomatosis. Therefore, an adequate biopsy is a must to show the rete pegs that extend into the deeper tissues. Deeper biopsies are always required for an accurate diagnosis. These nonneoplastic lesions of the penis need to be differentiated from cancer which is associated with high morbidity. We report a case of a verrucous lesion of the penis in a 38-year-old male.

Herein, we report the histopathology of angiokeratoma characterised by non-invasive, proliferative, ectatic vascular malformations accompanying lysosomal dilation in the canine skin. Two cutaneous angiokeratomas were diag - nosed in a six-year-old spayed female Pekinese dog. Physical examination of the skin revealed two small erythematous papules on the axillary and abdominal regions. The masses were approximately 2-3 mm in diameter, well-circumscribed, purple to red in colour, and slightly elevated above the skin surface. The two small masses had similar histopathological features, characterised by exophytic proliferation of a mixture of multiple vascular channels resembling a cavernous hae - mangioma on the superficial dermis. The entire lesion was well circumscribed and the overlying epidermal hyperplasia formed a collarette. Ultrastructural analysis with transmission electron microscopy revealed electron-lucent lysosomal dilatation in the keratinocytes of the irregular hyperplastic epithelial trabeculae like rete pegs.

The aim of this study was to explore the clinical value of the porcine acellular dermal xenograft (ADX) in combination with autologous split-thickness skin and pure autologous split-thickness skin grafting applied in deep full-thickness burns and scar wounds. A total of 30 patients with deep burns were randomly divided into experimental and control groups following escharectomy. The patients were separately treated with porcine acellular dermal xenograft (ADX) in combination with autologous split-thickness skin and pure autologous split-thickness skin graft. The wound healing was observed routinely and the scores were evaluated using Vancouver scar scale at different times following transplant surgery. The samples of cograft regions and the control group (pure transplant split-thickness skin autograft) were observed using light microscopy and electron microscopy, and the follow-up results were recorded. No conspicuous rejections on the cograft wound surface were observed. Compared with the control group, the cograft wounds were smooth, presented no scar contracture and exhibited good skin elasticity and recovery of the joint function. The cografted skin combined well and displayed a clear and continuous basal membrane, as well as gradually combined skin structure, a mature stratum corneum, downward extended rete pegs, a mainly uniform dermal collagen fiber structure, regular alignment, and fewer blood capillaries. Clear desmosome cograft regions were identified among heckle cells, as well as a clear and continuous basal membrane. The cografted skin of the combined split-thickness autograft and the acellular heterologous (porcine) dermal matrix showed an improved shape and functional recovery compared with the pure split-thickness skin autograft. The combination of the meshed ADX and the split-thickness skin autograft applied in deep full-thickness burns and scar wounds may induce tissue regeneration via dermis aiming. This method also has superior shape and functional recovery, and has an extensive clinical application value.

Dolphins are considered superior in their swimming capabilities when compared to technologies of nautical engineering. Accordingly, the specialized adaptations of their morphological design for drag reduction, thrust production, energy economy, and maneuverability must be highly effective. The heavy interdigitation of the skin layers by epidermal rete pegs and long dermal papillae and its high rate of sloughing may be adaptations to reduce drag. The blubber layer results of merging of the dermal fiber system with the subcutaneous fat layer.

The spatial organization of stem cells into a niche is a key factor for growth and continual tissue renewal during development, sustenance, and regeneration. Stratified epithelia serve as a great context to study the spatial aspects of the stem cell niche and cell lineages by organizing into layers of different cell types. Several types of stratified epithelia develop morphologies with advantageous, protruding structures where stem cells reside, such as rete pegs and palisades of Vogt. Here, multistage, spatial cell lineage models for epithelial stratification are used to study how the stem cell niche influences epithelial morphologies. When the stem cell niche forms along a rigid basal lamina, relatively regular morphologies are maintained. In contrast, stem cell niche formation along a free-moving basal lamina may prompt distorted epithelial morphologies with stem cells accumulating at the tips of fingerlike structures that form. The correspondence between our simulated morphologies and developmental stages of the human epidermis is also explored. Overall, our work provides an understanding of how stratified epithelia may attain distorted morphologies and sheds light on the importance of the spatial aspects of the stem cell niche.

Trichodinid ciliophorans are opportunistic parasites of many species of fish, amphibians, and molluscs, but yet never reported in association with lesions in birds. Postmortem and histopathological evaluation of a commercial adult Toulouse gander and female goose, and a wild Mallard drake revealed the presence of severe pathological parasitic colonization of their reproductive tracts. Histopathological findings included moderate to severe granulocytic inflammation, acanthosis, accentuation of the rete pegs, and proliferative hyperplastic squamous metaplasia of the mucosa of the ejaculatory ducts and groove, sulcus spermaticus, glandular part of the phallus (cavum penis), and oviduct in association with large numbers of ciliated protozoa anchored to the tissues or free in the lumen. These protozoa had characteristic morphological features analogous to the family of Trichodinidae. The source of this parasitism could not be determined. To our knowledge, this is the first report of trichodinosis associated with pathology in birds.

Hypertrophic scar (HSc) is a fibroproliferative disorder that occurs following deep dermal injury. Lack of a relevant animal model is one barrier toward better understanding its pathophysiology. Our objective is to demonstrate that grafting split-thickness human skin onto nude mice results in survival of engrafted human skin and murine scars that are morphologically, histologically, and immunohistochemically consistent with human HSc. Twenty nude mice were xenografted with split-thickness human skin. Animals were euthanized at 30, 60, 120, and 180 days postoperatively. Eighteen controls were autografted with full-thickness nude mouse skin and euthanized at 30 and 60 days postoperatively. Scar biopsies were harvested at each time point. Blinded scar assessment was performed using a modified Manchester Scar Scale. Histologic analysis included hematoxylin and eosin, Masson's trichrome, toluidine blue, and picrosirius red staining. Immunohistochemistry included anti-human human leukocyte antigen-ABC, α-smooth muscle actin, decorin, and biglycan staining. Xenografted mice developed red, shiny, elevated scars similar to human HSc and supported by blinded scar assessment. Autograft controls appeared morphologically and histologically similar to normal skin. Xenografts survived up to 180 days and showed increased thickness, loss of hair follicles, adnexal structures and rete pegs, hypercellularity, whorled collagen fibers parallel to the surface, myofibroblasts, decreased decorin and increased biglycan expression, and increased mast cell density. Grafting split-thickness human skin onto nude mice results in persistent scars that show morphologic, histologic, and immunohistochemical consistency with human HSc. Therefore, this model provides a promising technique to study HSc formation and to test novel treatment options.

A case of an epulis in an 8-month-old boy is reported. The tumor was localized on the mandibular alveolar ridge and, according to the parents, was a recurrence of a congenital tumor excised when the boy was 2 months old. Microscopically, it was composed of many spindled or ovoid cells with vesicular nuclei and non-granular eosinophilic cytoplasm, and covered by acanthotic parakeratinized squamous epithelium with broad rete pegs. Immunohistochemically, the spindled and ovoid cells were intensely positive for vimentin and neuron-specific enolase, and negative for S-100 protein. The final diagnosis was spindle cell epulis. The possible correlation of this lesion with congenital granular cell epulis is discussed.

Introduction: Localized juvenile spongiotic gingival hyperplasia (LJSGH) is a recently described uncommon and distinctive form of inflammatory hyperplasia. Treatment of this condition has varied from surgical excision to no treatment followed by spontaneous remission. This case report demonstrates successful management of the lesion using a conservative treatment approach.Case Presentation: A 14-year-old male with negative medical and dental histories presented with LJSGH. The vivid red lesions involved the papillary and marginal gingiva of all maxillary anterior teeth. Initially, the lesion developed as a single red patch-like area involving the maxillary left cuspid. Subsequently, the lesion displayed a linear pattern of spread to involve successively more teeth in the anterior sextant. Biopsy revealed hyperplastic elongated epithelial rete pegs, atrophy of the overlying stratified squamous epithelium with mild spongiosis, neutrophilic exocytosis, and a highly vascular connective tissue with a dominant ...

Background: Since the use of laminin-5 as a marker of invasiveness has been proposed by several authors, our objective was to compare the expression of this protein in pseudocarcinomatous hyperplasia and squamous cell carcinoma (SCC). Methods: Sixty-four paraffin-embedded skin biopsy samples with diagnosis of epidermal hyperplasia (non-pseudocarcinomatous), pseudocarcinomatous hyperplasia, actinic keratosis/carcinoma in situ, microinvasive and frankly invasive SCC were obtained for immunohistochemical study. Results: Adjacent normal epithelium and epidermal hyperplasia (non-pseudocarcinomatous) showed no staining. In pseudocarcinomatous hyperplasia, laminin-5 was positive, at least focally, in 14 of 16 (87.5%) samples and was concentrated in peripheral cells of elongated rete pegs and in migrating cells in dermis. In samples of microinvasive carcinoma (n = 7), the expression was observed in all cases and was concentrated in the leading edge of the tumor. All cases (n = 21) of frankly invasive SCC showed cells expressing laminin-5, at least focally. Well-differentiated areas of the tumor presented a pattern of expression in peripheral cells of tumor nests while a diffuse pattern of expression was observed in less differentiated areas. The present investigation describes the genetic characterization of BPV-1 and its pathological features of cutaneous warts in cattle. A total of 7 clinical cases of cutaneous warts on various positions of cattle were studied at Mukteswar, Uttarakhand. The cases were confirmed by histopathology and molecular method. Grossly, dome shaped lesions were observed on nostril, around the eye, shoulder and neck region whereas single long cylindrical and dome shaped protruding growths were observed in teats. Histopathological study showed presence of hyperkeratinization, basket wave appearance, parakeratosis, koilocytosis, rete pegs formation in epidermal layer and fibroblast proliferation in dermal layer and pathologically diagnosed as fibropapilloma. In teat warts, the characteristic pathological lesions were hyperkeratinization, digit like proliferation, parakeratosis, keratohylinization with koilocytes which were diagnosed as papillomas. Two clinical cases of warts on teat revealed hyperplasia with mild cornifi- cation and thickening of epidermal layer. Immunohistochemistry, performed by using monoclonal mouse antibody against E2 protein of BPVs, revealed positive immunolabeling in the nucleus, cytoplasm and cytoplasmic membrane of basal, spinosum and granulosum layer. In PCR, 5 clinical samples were positive for BPV and based on sequence analysis it was identified as BPV-1. The phylogenetic analysis of the L1 gene placed the present BPV under a cluster of delta-papillomavirus. In conclusion, the confirmatory detection of BPV-1 in particular herd and its genetic characterization help in molecular epidemiology study and to set appropriate therapeutic and control measures.

Removal of necrotic tissue is essential for wound-bed preparation for the treatment of chronic wounds. Enzymatic debridement appears to be most useful in the removal of eschar when surgical techniques cannot be utilized. The purpose of this study is to determine the efficacy of papain–urea chlorophyllin ointment compared to moist wound care using an infected, acute, porcine full thickness wound model. Twenty full thickness wounds were created on the dorsum of domestic pigs. Following hemostasis, the wounds were contaminated with wound-isolated bacteria then treated with test articles and dressed with moisture-retentive dressing. All the wounds were evaluated 1, 4, 8, 11, 14,18, and 21 days postsurgery for epithelialization and biopsies were obtained for microscopic evaluation. The results showed no adverse effect in reepithelialization in acute wounds when using papain–urea chlorophyllin "ointment compared to moist wound care, TEWL measurements also showed no adverse affects from the treatment. Microscopic examination reveals an increase in the number of keratinocytes present in the epidermis of the treated pigs as well as an increase in the depth of rete pegs. New blood vessel formation was increased at all examined time points in the papain–urea chlorophyllin ointment treated pigs, which was verified with CD31 and von Willebrand Factor staining. VEGF was increased after 1 week in the wound fluid from treated wounds compared to moist control. While the reepithelialization rate does not appear to be inhibited by papain–urea-chlorophyllin combination in acute wounds, the healing appears to be more complete based on the number of keratinocytes present in the epidermis and the extensive rete peg formation. The wounds also appear to have enhanced vascularization which is vital in supplying nutrients to the newly healed wound.
