11. FishChoice 2.0: Information on health benefits / risks and sustainability for seafood consumers

Abstract

Since seafood is a significant source of nutrients with known health benefits, its consumption is promoted as a healthy food choice. However, seafood can also contain potentially hazardous environmental pollutants. In the context of the ECsafeSEAFOOD FP7 project, FishChoice (www.fishchoice.eu) was developed as a communication tool to help to the consumers to take the most appropriate decisions on their seafood consumption habits. FishChoice relies on scientific information that allows calculating, on an individual basis, intakes of nutrients and pollutants derived from seafood consumption. In the framework of the EU-H2020 funded SEAFOODTOMORROW project, an optimized version of the online tool has been released. FishChoice is available in 25 EU languages with a customized list of seafood species per EU country, considering specific (national) consumption habits. The list of nutrients has been extended according to the latest EFSA recommendations, while pollutants data incorporate results from recent studies. The sustainability of seafood consumption has been also implemented, providing recommendations to help preserve the marine environment. Finally, FishChoice is suitable not only for consumers, but also health professionals, schools and academia, as well as the industrial sector and public health providers.

Abstract

International audience; Background: Resistance to thyroid hormone alpha (RTH alpha) is a rare genetic disease due to mutations in the THRA gene, which encodes thyroid hormone receptor alpha 1 (TR alpha 1). Since its first description in 2012, 46 cases of RTH alpha have been reported worldwide, corresponding to 26 different mutations of TR alpha 1. RTH alpha patients share some common symptoms with hypothyroid patients, without significant reduction in thyroid hormone level. The high variability of clinical features and the absence of reliable biochemical markers make the diagnosis of this disease difficult. Some of these mutations have been recently modeled in mice. Methods: In our study, we used four different mouse models heterozygous for frameshift mutations in the Thra gene. Two of them are very close to human mutations, while the two others have not yet been found in patients. We characterized the metabolic phenotypes of urine and plasma samples collected from these four animal models using an untargeted nuclear magnetic resonance (NMR)-based metabolomic approach. Results: Multivariate statistical analysis of the metabolomic profiles shows that biofluids of mice that carry human-like mutations can be discriminated from controls. Metabolic signatures associated with Thra mutations in urine and plasma are stable over time and clearly differ from the metabolic fingerprint of hypothyroidism in the mouse. Conclusion: Our results provide a proof-of-principle that easily accessible NMR-based metabolic fingerprints of biofluids could be used to diagnose RTH alpha in humans.

Abstract

International audience; Understanding the transcriptional function of thyroid hormone receptors implies a precise analysis of their interactions with chromatin and other protein components of the cells. We present here two protocols that are routinely used in our laboratory. The first co-immunoprecipitation procedure allows addressing the capacity of proteins to form stable multiprotein complexes with TRs in cells. The chromatin affinity purification enables us to define the sites occupied by TRs on chromatin. In this case the lack of high quality antibodies is circumvented by introducing an N-terminal tag in TR, with unspecific affinity for immunoglobulins.

Abstract

ATLAS work package 8 & 9 presentationat ATLAS 3rd General Assembly. ATLAS Work Packages 8 and 9 are dedicated to the communication and dissemination of knowledge outputs from the ATLAS project. Collectively the WPs work to ensure optimal communication, management and effective knowledge transfer of ATLAS outputs, to different end users (academia, industry, policy makers and wider society), resulting in the widest exploitation and impact possible. To achieve this, an ATLAS Dissemination and Exploitation Plan (D9.1) has been developed, implemented and is regularly updated. An ATLAS dissemination portfolio (D9.2) has been created to facilitate the promotion of the ATLAS project. A Data Management Plan has been developed which specifies how ATLAS will improve and maximise access to and re-use of research data generated by the project (D8.1). Looking forward, we aim to improve linkages with other WPs focusing on science-policy and science-industry engagements, extracting knowledge outputs and analysing their potential applications. An outreach educational programme will be developed to target public end users, ensuring the greatest impact from ATLAS and its legacy.
