Abstract

Green and roasted coffee oils are products rich in bioactive compounds, such as linoleic acid and the diterpenes cafestol and kahweol, being a potential ingredient for food and cosmetic industries. An overview of oil extraction techniques most applied for coffee beans and their influence on the oil composition is presented. Both green and roasted coffee oil extractions are highlighted. Pressing, Soxhlet, microwave, and supercritical fluid extraction were the most used techniques used for coffee oil extraction. Conventional Soxhlet is most used on a lab scale, while pressing is most used in industry. Supercritical fluid extraction has also been evaluated mainly due to the environmental approach. One of the highlighted activities in Brazilian agribusiness is the industrialization of oils due to their increasing use in the formulation of cosmetics, pharmaceuticals, and foods. Green coffee oil (raw bean) has desirable bioactive compounds, increasing the interest of private companies and research institutions in its extraction process to preserve the properties contained in the oils. Abstract

The international trade of essential oils accounted for more than US$ 10 billion in 2021, with a growing tendence. USA, China, India, France and Brazil are the main global players. Reliable and comprehensive market data, however, is hard to obtain. Apart from a few government databases dealing with import/export values, information regarding local (national/internal) production and consumption as well as market trends, when available, are for sale, and expensive. Data from the global and Brazilian markets in the last decade were gathered and discussed in this review, with highlights on the balance of trade and the main oils produced in Brazil. As a megadiverse country, opportunities for new essential oils from Brazil do exist, but several obstacles must be overcome, such as legislation compliance, quality, supply and safety assurance, so that the market can be conquered.

Abstract

Murici (Byrsonima crassifolia L., Malpighiaceae), a small, tropical fruit, very popular in Brazil, has a strong fruity, rancid cheese aroma. High resolution gas chromatography-olfactometry-mass spectrometry and aroma extract dilution analysis identified the most potent substances in its aroma as ethyl butanoate (fruity, sweet), ethyl hexanoate (fruity), 1-octen-3-ol (mushroom like), butyric acid (rancid, cheese), hexanoic acid (pungent, cheese) and phenylethyl alcohol (floral). Its pulp is mainly composed of ethyl, methyl and phenylethyl esters and by carboxylic acids, terpenoids, delta-lactones and some sulfur compounds. Seed analysis also showed a buttery and oily cheese aroma and GC-MS revealed linoleic, oleic, stearic and palmitic acids as foremost components, while the aroma was associated to the butyric and hexanoic acids present in minor amounts.

Abstract

A full account to the preparation and application of three appropriately substituted vinylcyclohexenes (2,2-dimethyl-3-vinylcyclohex-3-en-1-ol, 2,2-dimethyl-3-vinylcyclohex-3-en-1-one and 3,3-dimethyl-2-vinylcyclohexene) in thermal Diels-Alder reactions with alpha,beta-unsaturated esters (methyl tiglate and methyl angelate) is given. This approach delivered the racemic synthesis of ten octalin derivatives bearing a rearranged drimane skeleton (4 diastereomers of 1-methoxycarbonyl-6-hydroxy-1,2,5,5-tetramethyl-1,2,3,5,6,7, 8,8a-octahydronaphthalene; 1-methoxycarbonyl-6-oxo-1,2,5,5-tetramethyl-1,2,3,4,5,6,7,8-octahydronaphthalene; 2-methoxycarbonyl-6-oxo-1,2,5,5-tetramethyl-1,2,3,5,6,7,8,8a-octahydronaphthalene; 3 diastereomers of 1-methoxycarbonyl-1,2,5,5-tetramethyl-1,2,3,5,6,7,8,8a-octahydronaphthalene and 2-methoxycarbonyl-1,2,5,5-tetramethyl-1,2,3,5,6,7,8,8a-octahydronaphthalene) . Central synthetic features included preparation of enoltriflates by Stang's protocol and the successful palladium-catalyzed cross-coupling reaction (Stille reaction) of the triflate with the tri-n-butylvinylstannane. The octalins relative stereochemistry was unequivocally ascertained by spectroscopic methods and/or X-ray crystallography and these data now stand as useful tools to support the correct assignment of related natural products usually isolated in minute amounts.

Abstract

Various acid diterpenes of Copaifera cearensis Huber ex Ducke were isolated from the crude extract by flash Silica Gel/Potassium Hydroxide Chromatography. The main components were identified by GC-MS analyses. Further fractionation by reversed phase (RP) semi-preparative HPLC allowed isolation and identification of minor components and provided additional spectral data of those compounds. It was possible to detect the presence of eleven acid diterpenes in addition to a sesquiterpene acid. In addition to potassium hydroxide deposition on the silica surface, the presence of Si-OK residues, resulting from a cation exchange process, was established.

Abstract

The volatile constituents obtained from a static cryogenic headspace of Caesalpinia echinata Lam. (Leguminosae) showed E-beta-ocimene as the major compound (57.2%), beside other monoterpenes, C6 derivatives like n-hexanal and (E)-2-hexenal and nitrogen compounds such as indole and methyl anthranilate. From the essential oil of the leaves obtained by hydrodistillation in a Clevenger apparatus, (E)-3-hexen-1-ol was identified as the major constituent while phenolic compounds were the most representative class of secondary metabolites.

Abstract

The high worldwide consumption of antibiotics and their complex impurity profiles has drawn the attention of the scientific community to the development and validation of stability methods for these drugs. Amphotericin B is an antibiotic that is a natural fermentation product of bacterium Streptomyces nodosus, used as a broad-spectrum antifungal agent, and is highly unstable. For this reason, the objective of this work is the development and validation of an indicative method of stability by high performance liquid chromatography (HPLC) associated to diode array detector (DAD) for amphotericin B (AMB). To achieve this, the chromatographic profiles of acid, basic, oxidative and thermal degradation caused by AMB exposure to water and light were verified by HPLC-DAD, using an isocratic method under the following conditions: C18 chromatographic column (200 × 4.6 mm-5 µm), mobile phase composed of 65:35 of organic phase (methanol/acetonitrile in 41:18)/aqueous phase (2.5 mmol L-1 of disodium edetate, pH 5.0), flow rate of 1.0 mL min-1, injection volume of 20 µL, column temperature 30 ± 2 °C and wavelength of 383 nm. After identification of these profiles, the method was validated according to recommendations of the International Conference on Harmonization guidelines, and then, the active pharmaceutical ingredient (API) peak purity grade, percentage of drug degradation, occurrence of impurities peak and its identification by mass spectrometry (MS) with electrospray ionization (ESI), under positive ionization mode, were evaluated. It is suggested that the main degradation products were amphotericin B (2), degradation product 1 (DP1) and degradation product 2 (DP2) in acid and oxidative medium. Amphotericin B was stable in the presence of water and at 70 ± 2 °C for seven days.

Abstract

Termites are insects with a complex social organization on castes among which soldiers are the only responsible for the colony defense. The soldiers of the Nasutitermes genus use chemical defense, which comprises a mixture of mono, sesqui and mainly diterpenes. The new diterpene 2b,3a-dihydroxy-trinervita-1(15),11-diene, found in Nasutitermes macrocephalus, was isolated using preparative high performance liquid chromatography (HPLC) and identified by gas chromatography-mass spectrometry (GC-MS) and nuclear magnetic resonance (NMR) techniques. The absolute stereochemistry determined was supported by ab initio calculations and by comparison of the experimental circular dichroism data with the theoretical results. The isolated diterpene was evaluated against the bacteria Staphylococcus aureus (Gram-positive) and Escherichia coli (Gram-negative), and the fungi Candida albicans and Aspergillus brasiliensis. The isolated diterpene presented minimal inhibitory concentrations of 62.5 and 62.5 µg mL-1 against S. aureus and C. albicans, respectively.

Abstract

During a bioprospection study in the Brazilian Cerrado, samples of Bidens graveolens Mart., an endemic species, were investigated. Chemical and olfactive analyses were performed on the essential oil obtained from fresh leaves of B. graveolens collected in November 2013 (BG1) and May 2014 (BG2). A total of 60 compounds were identified in the essential oil extracted from fresh leaves, by hydrodistillation. The main constituents were identified as monoterpene hydrocarbons, ranging from 86.5% to 89.6%. Limonene was the major compound, accounting for 22.8% in BG1 and 47.7% in BG2. Thirty‐three–oxygenated monoterpenes were identified, comprising about 9% in both essential oils (EO). The attributes described by the panelists are in accordance to the sensorial profile. Myrcene, limonene, epoxy‐myrcene, and limonene dioxide were rated by all panelists. Present at a frequency of 83.3%, α‐pinene, β‐pinene, α‐pinene oxide, and carvone were picked out by five panelists. It is noteworthy that most compounds rated with the highest intensity score belonged to the oxygenated monoterpene class, considered the most expressive terpene class used in perfumery. Abstract

The latex of Himatanthus sucuuba (Spruce) Woodson, used popularly in the Amazon for the treatment of tumors, gastritis, inflammations and infections, was evaluated for cytotoxicity and antibacterial activities. The iridoid lactones, plumericin and isoplumericin were isolated from latex by bioassay fractionation and were found to be associated with DNA damage. Gallic acid exhibited the highest antimicrobial activity among the phenolic compounds isolated from the aqueous fraction. The compounds associated to cytotoxicity and antimicrobial activities could be responsible to the effects of this species used in traditional medicine.Key words: Himatanthus sucuuba, iridoids, phenolics, cytotoxicity, antibacterial.

Abstract

The ambient ionization mass spectrometry techniques: desorption electrospray ionization (DESI) and easy ambient sonic-spray ionization (EASI) were explored as fast and simple ways to directly analyze the surface of intact green Arabica coffee beans treated by the dry, semi-dry and wet post-harvest methods. Five compounds were identified, including three components of the waxy layer that covers the green coffee beans (βN-arachinoyl-5-hydroxytryptamide, βN-behenoyl- 5-hydroxytryptamide, and βN-lignoceroyl-5-hydroxytryptamide) and that are commonly related to related to stomach irritations caused by coffee beverage consumption in sensitive people. Moreover, the multivariate statistical tool principal component analysis (PCA) was employed to differentiate the coffee post-harvest methods using data from the mass spectrometry fingerprinting analyses. Extraction procedures or sample pretreatment steps were not required for DESI and EASI analyses and the results obtained suggest therefore that these techniques could be used for rapid quality control and certification processes of coffees samples. As técnicas de ionização ambiente na espectrometria de massas: ionização de dessorção por eletrospray (DESI) e ionização ambiente por spray sônico (EASI) foram exploradas como uma forma simples e rápida para analisar diretamente a superfície intacta de grãos de café arábica tratados pelas vias seca, semi-seca e úmida. Cinco substâncias foram identificadas, incluindo três componentes da camada de cera que recobre os grãos verdes de café (βN-araquidonoil-5-hidroxitriptamida, βN-behenoil-5-hidroxitriptamida e βN-lignoceroil-5-hidroxitriptamida) e geralmente associadas com irritações estomacais que a bebida do café provoca em indivíduos sensíveis. Além disso, os processos de pós-colheita empregados nos cafés puderam ser diferenciados através da ferramenta estatística multivariada análise de componentes principais (PCA) usando dados de perfil químico das amostras. Nenhum processo de extração ou preparo de amostra foi necessário nas análises de DESI e EASI e os resultados obtidos sugerem portanto a possibilidade de uso dessas duas técnicas para um rápido controle qualitativo e processos de certificação de amostras de café.

Abstract

A fruit chemical composition reflects its maturation stage. For coffee, it is also the reflex of the post-harvesting processing type, dry, semi-wet and wet. The object of this work was to verify if headspace solid phase microextraction coupled to gas chromatography (HS-SPME-GC) could be used to discriminate between samples harvested in different maturation stages and treated by different processes. With application of principal component analysis to the area of 117 compounds extracted by SPME, using divinylbenzene/Carboxen/polydimethylsiloxane fiber, it was possible to discriminate, in the roasted and ground coffee, the maturity stage and processing type used .

Abstract

Essential oils are extracted by steam distillation of plants or cold pressing of citrus fruit pericarp. They are used in food, cosmetic, personal care and pharmaceutical industries. In Brazil, oils from orange and related products contribute to near 97% to the positive commercial performance of the sector. Predatory exploitation and the availability of new sources of raw materials, with more attracting prices, changed the paradigm. Prospective studies, sustainable use of Brazilian biodiversity, domestication of exotic species with commercial relevance, the use of breeding techniques and the development of new applications for essential oils are thematic lines, usually multidisciplinary, which have been prompting the expansion of the research on essential oils. This paper presents an analysis on essential oils balance trade from 2005 to 2008 and some historical data on research and production of essential oils in Brazil.

Abstract

The chaoolmugra oil is one of the oldest medicines used for leprosy until the development of sulfas. In Brazil, it is known as sapucainha oil that is obtained from Carpotroche brasilienesis seeds (Flacourtiaceae). Chemical and medicinal studies of sapucainha oil were introduced in Brazil by Peckolt, Cole and Cardoso which showed that major ciclopentenyl fatty acids as chaulmoogric, hydnocarpic and gorlic were responsible for its bactericidal activity. In this work, a method for quantification of sapucainha oil acidic fraction by HRGC was established using methyl n-heptadecanoate as internal standard. The oil chemical stability was investigated by the use of GC-MS, IR, ¹H and 13C NMR and showed that the main degradation products belong to the cyclopentenyl moiety oxidation.

Abstract

When in blossom, jujube (Zizyphus mauritiana Lam., Rhamnaceae) emanates a strong fecal odor. The substances responsible for this scent were analysed by headspace solid-phase microextraction and gas chromatography-mass spectrometry (SPME-GC-MS) associated to standard co-injection and retention indexes, which showed benzaldehyde as major constituent. Minor benzenoids, aliphatic carboxylic acids, aldehydes, hydrocarbons and oxygenated monoterpenes were also observed. Olfactometric evaluation of the SPME using GC-sniffing port indicated skatole (3-methyl-indole) as responsible for the fecal odor and vanillin for the sweety odor. As flores de Zizyphus mauritiana Lam. (Rhamnaceae) exalam um forte odor fecal, e sua análise por micro-extração em fase sólida (SPME) associada a cromatografia gasosa- espectrometria de massas (CG-EM), co-injeção de padrões e índices de retenção revelou o benzaldeído como constituinte majoritário. Foram observados ainda benzenóides, ácidos carboxílicos, aldeídos, hidrocarbonetos alifáticos e monoterpenóides. A avaliação olfatométrica do SPME realizada por CG- olfatometria em sniffing port indicou o escatol (3-metil-indol) como o responsável pelo odor fecal das flores e a vanilina pelo seu odor adocicado.
