12. Fluorescence Correlation Spectroscopy Reveals Highly Efficient Cytosolic Delivery of Certain Penta-Arg Proteins and Stapled Peptides

Abstract

We used fluorescence correlation spectroscopy (FCS) to accurately and precisely determine the relative efficiencies with which three families of "cell-penetrating peptides" traffic to the cytosol of mammalian cells. We find that certain molecules containing a "penta-arg" motif reach the cytosol, intact, with efficiencies greater than 50%. This value is at least 10-fold higher than that observed for the widely studied cationic sequence derived from HIV Tat or polyarginine Arg8, and equals that of hydrocarbon-stapled peptides that are active in cells and animals. Moreover, we show that the efficiency with which stapled peptides reach the cytosol, as determined by FCS, correlates directly with their efficacy in cell-based assays. We expect that these findings and the associated technology will aid the design of peptides, proteins, and peptide mimetics that predictably and efficiently reach the interior of mammalian cells.

Abstract

Intrinsically disordered proteins (IDPs) do not possess well-defined three-dimensional structures in solution under physiological conditions. We develop all-atom, united-atom, and coarse-grained Langevin dynamics simulations for the IDP alpha-synuclein that include geometric, attractive hydrophobic, and screened electrostatic interactions and are calibrated to the inter-residue separations measured in recent smFRET experiments. We find that alpha-synuclein is disordered with conformational statistics that are intermediate between random walk and collapsed globule behavior. An advantage of calibrated molecular simulations over constraint methods is that physical forces act on all residues, not only on residue pairs that are monitored experimentally, and these simulations can be used to study oligomerization and aggregation of multiple alpha-synuclein proteins that may precede amyloid formation., Comment: 14 pages, 12 figures, 2 tables

Abstract

Although decades of research within the field of second language acquisition have been dedicated to investigating the impact of individual differences on second language learners’ success, longitudinal research focused on individual differences and their impact on adult second language acquisition is extremely limited. Additional longitudinal research on individual differences is necessary to further our understanding of the nature of the process of adult second language acquisition. This area of research is also critical to the U.S. Government and the Department of Defense as thousands of military service members work in language-related positions, and these service members’ maintenance of high levels of language proficiency is critical for our nation’s national security. The current study used a longitudinal design to investigate the impact of individual differences such as general cognitive ability, language aptitude, and attitude toward learning assigned second language (L2) on military service members’ language proficiency outcomes. Latent growth curve modeling (LGM) was used to model participants’ initial proficiency levels and growth trajectories, and measures of cognitive ability, language aptitude, and attitude toward learning assigned L2 were used to measure the impact of these individual differences on language proficiency outcomes. Additional variables including GPA, age, education level, number of language training hours, billet type, and sex were also included in the analyses. The results from the four phases of analyses support the conclusion that the predictive value of individual difference factors on language proficiency outcomes differ not only by DLI Language Difficulty Category, as suggested by previous research, but also by language and even language modality.

41. Multiculturalism: Beyond Food, Festival, Folklore, and Fashion

Abstract

Despite overall trends toward increasing student diversity, geographic areas in the United States vary widely in their ethnic composition. In areas where the population is predominately European American, grasping a realistic meaning of "multiculturalism" can be difficult. Often, interpretations of the concept result in a mix of classroom activities emphasizing Black History Month, illustrating how different cultures celebrate Christmas, and tasting food from different countries. At best, educators recognize the lack of true multicultural understanding; and at worst, they stereotype various cultures in terms of language, ethnicity, and traits. The intent of this article is to illustrate the necessity of tying instructional design to a valid working definition of multicultural education--one that is transformative and results in an improved quality of life for everyone. The increases of the nation's students of color reaffirm a needed commitment to educational programs that appropriately reflect the racial, ethnic, and cultural diversity within the United States and the world.
