Abstract

Streetwalkers have unique transactions with the urban environment. They must be aware of the shifting environmental stimuli and react accordingly in order to remain in business and avoid arrests. The transactions and schemata of streetwalkers are explored in light of various sociocultural factors by use of police arrest records, field observations, survey results from arrested prostitutes, a multiple regression, and a multidimensional scaling technique. Women of different races are found to be using similar street crime strategies, however their schemata differ. Black and White streetwalkers appear to work at different times, and sometimes places, during the day. The interrelationships between behavior, space, and time are apparent and reveal the dynamic, multifaceted nature of criminal transactions.

Abstract

The 2-amine-9H-purine scaffold was identified as a weak bromodomain template and was developed via iterative structure based design into a potent nanomolar ligand for the bromodomain of human BRD9 with small residual micromolar affinity toward the bromodomain of BRD4. Binding of the lead compound 11 to the bromodomain of BRD9 results in an unprecedented rearrangement of residues forming the acetyllysine recognition site, affecting plasticity of the protein in an induced-fit pocket. The compound does not exhibit any cytotoxic effect in HEK293 cells and displaces the BRD9 bromodomain from chromatin in bioluminescence proximity assays without affecting the BRD4/histone complex. The 2-amine-9H-purine scaffold represents a novel template that can be further modified to yield highly potent and selective tool compounds to interrogate the biological role of BRD9 in diverse cellular systems.

Abstract

The goat αS1-casein is a main milk phosphoprotein of 199 aminoacid residues long. The αS1-casein gene (CSN1S1) contains 19 exons, ranging in size from 24 to 385 bp (for a total of 1138 bp) in the coding region, spread over about 17,5 kb transcription unit (Leroux et al., 1992). The goat αS1-casein exhibits both qualitative and quantitative remarkable variation arising from genetic polymorphism in the encoding gene. In particular, two alleles, CSN1S1 01 and 02, associated with a null content of the corrisponding protein, were detected (Martin et al., 1999), characterized by large DNA deletion (Cosenza et al., 2002) and insertion (Martin et al., 1999) events, respectively. Recently, by means of SDS-PAGE and CSN1S1 gene sequence analysis of goat individual milk and DNA sample, another allele (CSN1S1 N), associated with an apparently null amount of this protein fraction in the milk, was identified (Ramunno et al., 2002). This allele is characterized by the citosine deletion at position 23 in the 98th exon of the gene, likewise to CSN1S1 F allele (associated to a low content of αS1- casein) and, contrary to this one, for the absence of two insertions, 11 and 3 bp base pair in length, in the downstream intron. The present work reports the results of a preliminary analysis of mRNA transcribed from the N allele at goat CSN1S1 locus compared with the one transcribed from a homozygote individual for the CSN1S1 A allele.

Abstract

Purpose: To ivestigate in vitrothe receptivity to the resin bonding process of Er:YAG laser-prepared dentin surfacecompared to that of tooth cavities prepared with a conventional diamond bur. Materials and Methods: Fifty extracted human teeth free of caries were divided into two preparation groups of 25 teeth each. An Er:YAG laser was used to prepare cavities in one group, and a conventional diamond bur was used in the other. In each preparation group, 13 teeth were filled with the composite Diamond Crown and 12 with Z100 MP. The dentin/resin interface properties of the two different restorative compounds were investigated by visible light micro-Raman spectroscopy (uRS) and scanning electron microscopy (SEM). Results: The measured Raman spectra showed typical resonance peaks of dentin. Resin contribution to the Raman spectra were also detected and were used to monitor the interface properties of the boundary region. In both mechanically and laser prepared samples, the diffusion depth of the Diamond Crown composite in dentin was limited to ca 10um. Deeper diffusion was observed at the boundary of dentin and Z100 MP. Conclusion: Under the conditions of this study, there were no appreciable differences between mechanically and laser prepared samples in term of dentin/resin interface characteristics. The obteined results again confirmed the validity of micro-Raman spectroscopy as a noninvasive method of examining the interaction between dentin surface and biomaterials. Knowledge thus gained is valuable in planning new dental materials to be employed specifically when laser technology is used.

Abstract

Petrosaspongiolide M (PM) is an anti-inflammatory marine metabolite that displays a potent inhibitory activity toward group II and III secretory phospholipase A2 (PLA2) enzymes. The details of the mechanism, which leads to a covalent adduct between PLA2 and γ-hydroxybutenolide-containing molecules such as PM, are still a matter of debate. In this paper the covalent binding of PM to bee venom PLA2 has been investigated by mass spectrometry and molecular modeling. The mass increment observed for the PM-PLA2 adduct is consistent with the formation of a Schiff base by reaction of a PLA2 amino group with the hemiacetal function (masked aldehyde) at the C-25 atom of the PM γ-hydroxybutenolide ring. Proteolysis of the modified PLA2 by the endoprotease LysC followed by HPLC MS analysis allowed us to establish that the PLA2 α-amino terminal group of the lle-1 residue was the only covalent binding site for PM. The stoichiometry of the reaction between PM and PLA2 was also monitored and results showed that even with excess inhibitor, the prevalent product is a 1:1 (inhibitor:enzyme) adduct, although a 2:1 adduct is present as a minor component. The 2:1 adduct was also characterized, which showed that the second site of reaction is located at the ε-amino group of the Lys-85 residue. Similar results in terms of the reaction profile, mass increments, and location of the PLA2 binding site were obtained for manoalide, a paradigm for irreversible PLA2 inhibitors, which suggests that the present results may be considered of more general interest within the field of anti-inflammatory sesterterpenes that contain the γ-hydroxybutenolide pharmacophore. Finally, a 3D model, constrained by the above experimental results, was obtained by docking the inhibitor molecule into the PLA2 binding site through AFFINITY calculations. The model provides an interesting insight into the PM-PLA2 inhibition process and may prove useful in the design of new anti-inflammatory agents that target PLA2 secretory enzymes.

Abstract

Cavities in human teeth were fabricated by means of Er:YAG laser and compared with samples processed by using conventional drill. The cavities of lased and unlased samples were cured with two different composite resins (Bis-GMA vs PEX) to investigate differences in their adhesion properties. Morphological characterization of the sample surfaces by SEM analysis was performed. The same samples were then examined by micro-Raman spectroscopy by using He-Ne laser visible light excitation, in order to characterize the dentin/resin interfaces. The Raman spectra exhibited typical peaks of dentine. Resin contributions to the Raman spectra were also detected and have been used to monitor the interface properties of the boundary region. The dependence of the Raman spectrum on spatial position has been monitored. In both lased and unlased samples the PEX diffusion in dentine is limited in a region of order of 10 micrometer depth. A larger interdiffusion region is observed at the boundary of dentine and Bis-GMA resin.
