Abstract: The spin structure functions of the proton and the deuteron were measured during the EG4 experiment at Jefferson Lab in 2006. Data were collected for longitudinally polarized electron scattering off longitudinally polarized NH$_3$ and ND$_3$ targets, for $Q^2$ values as small as 0.012 and 0.02 GeV$^2$, respectively, using the CEBAF Large Acceptance Spectrometer (CLAS). This is the archival paper of the EG4 experiment that summaries the previously reported results of the polarized structure functions $g_1$, $A_1F_1$, and their moments $\overline \Gamma_1$, $\overline \gamma_0$, and $\overline I_{TT}$, for both the proton and the deuteron. In addition, we report on new results on the neutron $g_1$ extracted by combining proton and deuteron data and correcting for Fermi smearing, and on the neutron moments $\overline \Gamma_1$, $\overline \gamma_0$, and $\overline I_{TT}$ formed directly from those of the proton and the deuteron. Our data are in good agreement with the Gerasimov-Drell-Hearn sum rule for the proton, deuteron, and neutron. Furthermore, the isovector combination was formed for $g_1$ and the Bjorken integral $\overline \Gamma_1^{p-n}$, and compared to available theoretical predictions. All of our results provide for the first time extensive tests of spin observable predictions from chiral effective field theory ($\chi$EFT) in a $Q^2$ range commensurate with the pion mass. They motivate further improvement in $\chi$EFT calculations from other approaches such as the lattice gauge method., Comment: 33 pages. 26 figures. We used regression analyses to identify baseline clinical features, including CRP value in those randomized to testing, associated with bacterial, viral or mixed infections, defined by the presence of bacterial and viral pathogens in sputum, detected by culture or polymerase chain reaction (PCR), respectively.Results: Of 386 participants with baseline sputum samples, 79 (20.5%), 123 (31.9%), and 91 (23.6%) had bacterial, viral/atypical, and mixed bacterial/viral/atypical pathogens identified, respectively. Increasing sputum purulence assessed by color chart was associated with increased odds of finding bacterial and mixed (bacterial and viral/atypical) pathogens in sputum (area under the ROC curve (AUROC) for bacterial pathogens =0.739 (95% CI: 0.670, 0.808)). Elevated CRP was associated with increased odds of finding bacterial pathogens and mixed pathogens but did not significantly increase the AUROC for predicting bacterial pathogens over sputum color alone (AUROC for combination of sputum color and CRP = 0.776 (95% CI: 0.708, 0.843), p for comparison of models = 0.053). We found no association between the presence of sputum pathogens and other clinical or demographic features.Conclusion: Sputum purulence was the best predictor of sputum bacterial pathogens and mixed bacterial viral/atypical pathogens in patients with COPD exacerbations in our study. Abstract: Measuring Deeply Virtual Compton Scattering on the neutron is one of the necessary steps to understand the structure of the nucleon in terms of Generalized Parton Distributions (GPDs). Neutron targets play a complementary role to transversely polarized proton targets in the determination of the GPD $E$. This poorly known and poorly constrained GPD is essential to obtain the contribution of the quarks' angular momentum to the spin of the nucleon. DVCS on the neutron was measured for the first time selecting the exclusive final state by detecting the neutron, using the Jefferson Lab longitudinally polarized electron beam, with energies up to 10.6 GeV, and the CLAS12 detector. The extracted beam-spin asymmetries, combined with DVCS observables measured on the proton, allow a clean quark-flavor separation of the imaginary parts of the GPDs $H$ and $E$., Comment: 7 pages, 6 figures

Abstract: The new experimental measurements of beam spin asymmetry were performed for the deeply virtual exclusive $\pi^0$ production in a wide kinematic region with the photon virtualities $Q^2$ up to 8 GeV$^2$ and the Bjorken scaling variable $x_B$ in the valence regime. The data were collected by the CEBAF Large Acceptance Spectrometer (CLAS12) at Jefferson Lab with longitudinally polarized 10.6 GeV electrons scattered on an unpolarized liquid-hydrogen target. Sizable asymmetry values indicate a substantial contribution from transverse virtual photon amplitudes to the polarized structure functions.The interpretation of these measurements in terms of the Generalized Parton Distributions (GPDs) demonstrates their sensitivity to the chiral-odd GPD $\bar E_T$, which contains information on quark transverse spin densities in unpolarized and polarized nucleons and provides access to the proton's transverse anomalous magnetic moment. Abstract: The polarized cross section ratio $\sigma_{LT'}/\sigma_{0}$ from hard exclusive $\pi^{-} \Delta^{++}$ electroproduction off an unpolarized hydrogen target has been extracted based on beam-spin asymmetry measurements using a 10.2 GeV / 10.6 GeV incident electron beam and the CLAS12 spectrometer at Jefferson Lab. The study, which provides the first observation of this channel in the deep-inelastic regime, focuses on very forward-pion kinematics in the valence regime, and photon virtualities ranging from 1.5 GeV$^{2}$ up to 7 GeV$^{2}$. The reaction provides a novel access to the $d$-quark content of the nucleon and to $p \rightarrow \Delta^{++}$ transition generalized parton distributions. A comparison to existing results for hard exclusive $\pi^{+} n$ and $\pi^{0} p$ electroproduction is provided, which shows a clear impact of the excitation mechanism, encoded in transition generalized parton distributions, on the asymmetry.

Abstract: Deeply virtual Compton scattering (DVCS) allows one to probe Generalized Parton Distributions (GPDs) describing the 3D structure of the nucleon. We report the first measurement of the DVCS beam-spin asymmetry using the CLAS12 spectrometer with a 10.2 and 10.6 GeV electron beam scattering from unpolarised protons. The results greatly extend the $Q^2$ and Bjorken-$x$ phase space beyond the existing data in the valence region and provide over 2000 new data points measured with unprecedented statistical uncertainty, setting new, tight constraints for future phenomenological studies., Comment: Revised Figure 4 and discussion around the number of effective ANNs after Bayesian reweighting

Abstract: A multidimensional extraction of the structure function ratio $\sigma_{LT'}/\sigma_{0}$ from the hard exclusive $\vec{e} p \to e^\prime n \pi^+$ reaction above the resonance region has been performed. The study was done based on beam-spin asymmetry measurements using a 10.6 GeV incident electron beam on a liquid-hydrogen target and the CLAS12 spectrometer at Jefferson Lab. The measurements focus on the very forward regime ($t/Q^{2}$ $\ll$ 1) with a wide kinematic range of $x_{B}$ in the valence regime (0.17 $<$ $x_{B}$ $<$ 0.55), and virtualities $Q^{2}$ ranging from 1.5 GeV$^{2}$ up to 6 GeV$^{2}$. The results and their comparison to theoretical models based on Generalized Parton Distributions demonstrate the sensitivity to chiral-odd GPDs and the directly related tensor charge of the nucleon. In addition, the data is compared to an extension of a Regge formalism at high photon virtualities. It was found that the Regge model provides a better description at low $Q^{2}$, while the GPD model is more appropriate at high $Q^{2}$., Comment: arXiv admin note: text overlap with arXiv:2007.15677

Abstract: We report results of $\Lambda$ hyperon production in semi-inclusive deep-inelastic scattering off deuterium, carbon, iron, and lead targets obtained with the CLAS detector and the Continuous Electron Beam Accelerator Facility 5.014~GeV electron beam. These results represent the first measurements of the $\Lambda$ multiplicity ratio and transverse momentum broadening as a function of the energy fraction~($z$) in the current and target fragmentation regions. The multiplicity ratio exhibits a strong suppression at high~$z$~and~an enhancement at~low~$z$. The measured transverse momentum broadening is an order of magnitude greater than that seen for light mesons. This indicates that the propagating entity interacts very strongly with the nuclear medium, which suggests that propagation of diquark configurations in the nuclear medium takes place at least part of the time, even at high~$z$. The trends of these results are qualitatively described by the Giessen Boltzmann-Uehling-Uhlenbeck transport model, particularly for the multiplicity ratios. Abstract: This article presents a collection of simulation studies using the ECCE detector concept in the context of the EIC's exclusive, diffractive, and tagging physics program, which aims to further explore the rich quark-gluon structure of nucleons and nuclei. To successfully execute the program, ECCE proposed to utilize the detecter system close to the beamline to ensure exclusivity and tag ion beam/fragments for a particular reaction of interest. Preliminary studies confirmed the proposed technology and design satisfy the requirements. The projected physics impact results are based on the projected detector performance from the simulation at 10 or 100 fb^-1 of integrated luminosity. Additionally, a few insights on the potential 2nd Interaction Region can (IR) were also documented which could serve as a guidepost for the future development of a second EIC detector.

Abstract: We report the first measurements of deep inelastic scattering spin-dependent azimuthal asymmetries in back-to-back dihadron electroproduction, where two hadrons are produced in opposite hemispheres along the z-axis in the center-of-mass frame, with the first hadron produced in the current-fragmentation region and the second in the target-fragmentation region. The data were taken with longitudinally polarized electron beams of 10.2 and 10.6 GeV incident on an unpolarized liquid-hydrogen target using the CLAS12 spectrometer at Jefferson Lab. Observed non-zero $\sin\Delta\phi$ modulations in $ep \rightarrow e'p\pi^+X$ events, where $\Delta\phi$ is the difference of the azimuthal angles of the proton and pion in the virtual photon and target nucleon center-of-mass frame, indicate that correlations between the spin and transverse momenta of hadrons produced in the target- and current-fragmentation regions may be significant. The measured beam-spin asymmetries provide a first access in dihadron production to a previously unobserved leading-twist spin- and transverse-momentum-dependent fracture function. The fracture functions describe the hadronization of the target remnant after the hard scattering of a virtual photon off a quark in the target particle and provide a new avenue for studying nucleonic structure and hadronization., Comment: 7 pages, 6 figures

Abstract: We performed feasibility studies for various measurements that are related to unpolarized TMD distribution and fragmentation functions. The processes studied include semi-inclusive Deep inelastic scattering (SIDIS) where single hadrons (pions and kaons) were detected in addition to the scattered DIS lepton. The single hadron cross sections and multiplicities were extracted as a function of the DIS variables $x$ and $Q^2$, as well as the semi-inclusive variables $z$, which corresponds to the momentum fraction the detected hadron carries relative to the struck parton and $P_T$, which corresponds to the transverse momentum of the detected hadron relative to the virtual photon. The expected statistical precision of such measurements is extrapolated to accumulated luminosities of 10 fb$^{-1}$ and potential systematic uncertainties are approximated given the deviations between true and reconstructed yields., Comment: 12 pages, 9 figures, to be submitted in joint ECCE proposal NIM-A volume

Abstract: We performed feasibility studies for various single transverse spin measurements that are related to the Sivers effect, transversity and the tensor charge, and the Collins fragmentation function. The processes studied include semi-inclusive deep inelastic scattering (SIDIS) where single hadrons (pions and kaons) were detected in addition to the scattered DIS lepton. The data were obtained in {\sc pythia}6 and {\sc geant}4 simulated e+p collisions at 18 GeV on 275 GeV, 18 on 100, 10 on 100, and 5 on 41 that use the ECCE detector configuration. Typical DIS kinematics were selected, most notably $Q^2 > 1 $ GeV$^2$, and cover the $x$ range from $10^{-4}$ to $1$. The single spin asymmetries were extracted as a function of $x$ and $Q^2$, as well as the semi-inclusive variables $z$, and $P_T$. They are obtained in azimuthal moments in combinations of the azimuthal angles of the hadron transverse momentum and transverse spin of the nucleon relative to the lepton scattering plane. The initially unpolarized MonteCarlo was re-weighted in the true kinematic variables, hadron types and parton flavors based on global fits of fixed target SIDIS experiments and $e^+e^-$ annihilation data. The expected statistical precision of such measurements is extrapolated to 10 fb$^{-1}$ and potential systematic uncertainties are approximated given the deviations between true and reconstructed yields. The impact on the knowledge of the Sivers functions, transversity and tensor charges, and the Collins function has then been evaluated in the same phenomenological extractions as in the Yellow Report. The impact is found to be comparable to that obtained with the parameterized Yellow Report detector and shows that the ECCE detector configuration can fulfill the physics goals on these quantities., Comment: 22 pages, 22 figures, to be submitted to joint ECCE proposal NIM-A volume

Abstract: The ECCE detector has been recommended as the selected reference detector for the future Electron-Ion Collider (EIC). A series of simulation studies have been carried out to validate the physics feasibility of the ECCE detector. In this paper, detailed studies of heavy flavor hadron and jet reconstruction and physics projections with the ECCE detector performance and different magnet options will be presented. The ECCE detector has enabled precise EIC heavy flavor hadron and jet measurements with a broad kinematic coverage. Abstract: Exclusive heavy quarkonium photoproduction is one of the most popular processes in EIC, which has a large cross section and a simple final state. Due to the gluonic nature of the exchange Pomeron, this process can be related to the gluon distributions in the nucleus. The momentum transfer dependence of this process is sensitive to the interaction sites, which provides a powerful tool to probe the spatial distribution of gluons in the nucleus. Recently the problem of the origin of hadron mass has received lots of attention in determining the anomaly contribution $M_{a}$. The trace anomaly is sensitive to the gluon condensate, and exclusive production of quarkonia such as J/$\psi$ and $\Upsilon$ can serve as a sensitive probe to constrain it. In this paper, we present the performance of the ECCE detector for exclusive J/$\psi$ detection and the capability of this process to investigate the above physics opportunities with ECCE., Comment: 11 pages, 14 figures, 1 table

Abstract: The recently approved Electron-Ion Collider (EIC) will provide a unique new opportunity for searches of charged lepton flavor violation (CLFV) and other new physics scenarios. In contrast to the $e \leftrightarrow \mu$ CLFV transition for which very stringent limits exist, there is still a relatively large discovery space for the $e \to \tau$ CLFV transition, potentially to be explored by the EIC. With the latest detector design of ECCE (EIC Comprehensive Chromodynamics Experiment) and projected integral luminosity of the EIC, we find the $\tau$-leptons created in the DIS process $ep\to \tau X$ are expected to be identified with high efficiency. A first ECCE simulation study, restricted to the 3-prong $\tau$-decay mode and with limited statistics for the Standard Model backgrounds, estimates that the EIC will be able to improve the current exclusion limit on $e\to \tau$ CLFV by an order of magnitude., Comment: 11 pages, 8 figures, to be submitted to NIM

Abstract: We describe the design and performance the calorimeter systems used in the ECCE detector design to achieve the overall performance specifications cost-effectively with careful consideration of appropriate technical and schedule risks. The calorimeter systems consist of three electromagnetic calorimeters, covering the combined pseudorapdity range from -3.7 to 3.8 and two hadronic calorimeters. Abstract: The Electron-Ion Collider (EIC) is a cutting-edge accelerator facility that will study the nature of the "glue" that binds the building blocks of the visible matter in the universe. The proposed experiment will be realized at Brookhaven National Laboratory in approximately 10 years from now, with detector design and R&D currently ongoing. Notably, EIC is one of the first large-scale facilities to leverage Artificial Intelligence (AI) already starting from the design and R&D phases. The EIC Comprehensive Chromodynamics Experiment (ECCE) is a consortium that proposed a detector design based on a 1.5T solenoid. The EIC detector proposal review concluded that the ECCE design will serve as the reference design for an EIC detector. Herein we describe a comprehensive optimization of the ECCE tracker using AI. The work required a complex parametrization of the simulated detector system. Our approach dealt with an optimization problem in a multidimensional design space driven by multiple objectives that encode the detector performance, while satisfying several mechanical constraints. We describe our strategy and show results obtained for the ECCE tracking system. The AI-assisted design is agnostic to the simulation framework and can be extended to other sub-detectors or to a system of sub-detectors to further optimize the performance of the EIC detector., Comment: 16 pages, 18 figures, 2 appendices, 3 tables

Abstract: The Electron Ion Collider (EIC) is the next generation of precision QCD facility to be built at Brookhaven National Laboratory in conjunction with Thomas Jefferson National Laboratory. There are a significant number of software and computing challenges that need to be overcome at the EIC. During the EIC detector proposal development period, the ECCE consortium began identifying and addressing these challenges in the process of producing a complete detector proposal based upon detailed detector and physics simulations. In this document, the software and computing efforts to produce this proposal are discussed; furthermore, the computing and software model and resources required for the future of ECCE are described.

Abstract: Rationale: Common genetic variants have been associated with idiopathic pulmonary fibrosis (IPF). Objectives: To determine functional relevance of the 10 IPF-associated common genetic variants we previously identified. Methods: We performed expression quantitative trait loci (eQTL) and methylation quantitative trait loci (mQTL) mapping, followed by co-localization of eQTL and mQTL with genetic association signals and functional validation by luciferase reporter assays. Illumina multi-ethnic genotyping arrays, mRNA sequencing, and Illumina 850k methylation arrays were performed on lung tissue of participants with IPF (234 RNA and 345 DNA samples) and non-diseased controls (188 RNA and 202 DNA samples). Abstract: BackgroundIdiopathic pulmonary fibrosis (IPF) is a progressive, fatal fibrotic interstitial lung disease. Few circulating biomarkers have been identified to have causal effects on IPF.MethodsTo identify candidate IPF-influencing circulating proteins, we undertook an efficient screen of circulating proteins by applying a two-sample Mendelian randomisation (MR) approach with existing publicly available data. For instruments, we used genetic determinants of circulating proteins which reside cis to the encoded gene (cis-single nucleotide polymorphisms (SNPs)), identified by two genome-wide association studies (GWASs) in European individuals (3301 and 3200 subjects). We then applied MR methods to test if the levels of these circulating proteins influenced IPF susceptibility in the largest IPF GWAS (2668 cases and 8591 controls). We validated the MR results using colocalisation analyses to ensure that both the circulating proteins and IPF shared a common genetic signal.ResultsMR analyses of 834 proteins found that a 1 sd increase in circulating galactoside 3(4)-l-fucosyltransferase (FUT3) and α-(1,3)-fucosyltransferase 5 (FUT5) was associated with a reduced risk of IPF (OR 0.81, 95% CI 0.74-0.88; p=6.3×10-7 and OR 0.76, 95% CI 0.68-0.86; p=1.1×10-5, respectively). Sensitivity analyses including multiple cis-SNPs provided similar estimates both for FUT3 (inverse variance weighted (IVW) OR 0.84, 95% CI 0.78-0.91; p=9.8×10-6 and MR-Egger OR 0.69, 95% CI 0.50-0.97; p=0.03) and FUT5 (IVW OR 0.84, 95% CI 0.77-0.92; p=1.4×10-4 and MR-Egger OR 0.59, 95% CI 0.38-0.90; p=0.01). FUT3 and FUT5 signals colocalised with IPF signals, with posterior probabilities of a shared genetic signal of 99.9% and 97.7%, respectively. Further transcriptomic investigations supported the protective effects of FUT3 for IPF.ConclusionsAn efficient MR scan of 834 circulating proteins provided evidence that genetically increased circulating FUT3 level is associated with reduced risk of IPF.

Abstract: The genetic make-up of an individual contributes to the susceptibility and response to viral infection. Although environmental, clinical and social factors have a role in the chance of exposure to SARS-CoV-2 and the severity of COVID-191,2, host genetics may also be important. Identifying host-specific genetic factors may reveal biological mechanisms of therapeutic relevance and clarify causal relationships of modifiable environmental risk factors for SARS-CoV-2 infection and outcomes. We formed a global network of researchers to investigate the role of human genetics in SARS-CoV-2 infection and COVID-19 severity. Here we describe the results of three genome-wide association meta-analyses that consist of up to 49,562 patients with COVID-19 from 46 studies across 19 countries. We report 13 genome-wide significant loci that are associated with SARS-CoV-2 infection or severe manifestations of COVID-19. Several of these loci correspond to previously documented associations to lung or autoimmune and inflammatory diseases3-7. They also represent potentially actionable mechanisms in response to infection. Mendelian randomization analyses support a causal role for smoking and body-mass index for severe COVID-19 although not for type II diabetes. The identification of novel host genetic factors associated with COVID-19 was made possible by the community of human genetics researchers coming together to prioritize the sharing of data, results, resources and analytical frameworks. This working model of international collaboration underscores what is possible for future genetic discoveries in emerging pandemics, or indeed for any complex human disease.

Abstract: We report on variability and correlation studies using multiwavelength observations of the blazar Mrk 421 during the month of February, 2010 when an extraordinary flare reaching a level of $\sim$27~Crab Units above 1~TeV was measured in very-high-energy (VHE) $\gamma$-rays with the VERITAS observatory. This is the highest flux state for Mrk 421 ever observed in VHE $\gamma$-rays. Data are analyzed from a coordinated campaign across multiple instruments including VHE $\gamma$-ray (VERITAS, MAGIC), high-energy (HE) $\gamma$-ray (Fermi-LAT), X-ray (Swift}, RXTE, MAXI), optical (including the GASP-WEBT collaboration and polarization data) and radio (Mets\"ahovi, OVRO, UMRAO). Light curves are produced spanning multiple days before and after the peak of the VHE flare, including over several flare `decline' epochs. The main flare statistics allow 2-minute time bins to be constructed in both the VHE and optical bands enabling a cross-correlation analysis that shows evidence for an optical lag of $\sim$25-55 minutes, the first time-lagged correlation between these bands reported on such short timescales. Limits on the Doppler factor ($\delta \gtrsim 33$) and the size of the emission region ($ \delta^{-1}R_B \lesssim 3.8\times 10^{13}\,\,\mbox{cm}$) are obtained from the fast variability observed by VERITAS during the main flare. Analysis of 10-minute-binned VHE and X-ray data over the decline epochs shows an extraordinary range of behavior in the flux-flux relationship: from linear to quadratic to lack of correlation to anti-correlation. Abstract: We report on a multi-band variability and correlation study of the TeV blazar Mrk 421 during an exceptional flaring activity observed from 2013 April 11 to 2013 April 19. The study uses, among others, data from GASP-WEBT, Swift, NuSTAR, Fermi-LAT, VERITAS, and MAGIC. The large blazar activity, and the 43 hours of simultaneous NuSTAR and MAGIC/VERITAS observations, permitted variability studies on 15 minute time bins, and over three X-ray bands (3-7 keV, 7-30 keV and 30-80 keV) and three very-high-energy (>0.1 TeV, hereafter VHE) gamma-ray bands (0.2-0.4 TeV, 0.4-0.8 TeV and >0.8 TeV). We detected substantial flux variations on multi-hour and sub-hour timescales in all the X-ray and VHE gamma-ray bands. The characteristics of the sub-hour flux variations are essentially energy-independent, while the multi-hour flux variations can have a strong dependence on the energy of the X-ray and the VHE gamma rays. The three VHE bands and the three X-ray bands are positively correlated with no time-lag, but the strength and the characteristics of the correlation changes substantially over time and across energy bands. Our findings favour multi-zone scenarios for explaining the achromatic/chromatic variability of the fast/slow components of the light curves, as well as the changes in the flux-flux correlation on day-long timescales. We interpret these results within a magnetic reconnection scenario, where the multi-hour flux variations are dominated by the combined emission from various plasmoids of different sizes and velocities, while the sub-hour flux variations are dominated by the emission from a single small plasmoid moving across the magnetic reconnection layer., Comment: 46 pages, 17 figures, 9 tables, accepted for publication in ApJ Supplements

42. Genetically increased circulating FUT3 level leads to reduced risk of Idiopathic Pulmonary Fibrosis: a Mendelian Randomisation Study.

Abstract: Idiopathic pulmonary fibrosis (IPF) is a progressive, fatal fibrotic interstitial lung disease. Few circulating biomarkers have been identified to have causal effects on IPF.To identify candidate IPF-influencing circulating proteins, we undertook an efficient screen of circulating proteins by applying a two-sample Mendelian randomisation (MR) approach with existing publicly available data. For instruments we used genetic determinants of circulating proteins which reside cis to the encoded gene (cis-SNPs), identified by two genome-wide association studies (GWASs) in European individuals (3301 and 3200 subjects). We then applied MR methods to test if the levels of these circulating proteins influenced IPF susceptibility in the largest IPF GWAS (2668 cases and 8591 controls). We validated the MR results using colocalization analyses to ensure that both the circulating proteins and IPF shared a common genetic signal.MR analyses of 834 proteins found that a one sd increase in circulating FUT3 and FUT5 was associated with a reduced risk of IPF (OR: 0.81, 95%CI: 0.74-0.88, p=6.3×10-7, and OR: 0.76, 95%CI: 0.68-0.86, p=1.1×10-5). Sensitivity analyses including multiple-cis SNPs provided similar estimates both for FUT3 (inverse variance weighted [IVW] OR: 0.84, 95%CI: 0.78-0.91, p=9.8×10-6, MR-Egger OR: 0.69, 95%CI: 0.50-0.97, p=0.03) and FUT5 (IVW OR: 0.84, 95%CI: 0.77-0.92, p=1.4×10-4, MR-Egger OR: 0.59, 95%CI: 0.38-0.90, p=0.01) FUT3 and FUT5 signals colocalized with IPF signals, with posterior probabilities of a shared genetic signal of 99.9% and 97.7%. Further transcriptomic investigations supported the protective effects of FUT3 for IPF.An efficient MR scan of 834 circulating proteins provided evidence that genetically increased circulating FUT3 level is associated with reduced risk of IPF.

Abstract: To examine whether lifelong genetically increased serum urate levels, a potent antioxidant, contribute to MS susceptibility using Mendelian randomization (MR). This 2-sample MR study included 25 independent genetic variants strongly associated with serum urate levels in a genome-wide association study meta-analysis of 140,949 individuals. Effects on the risk of MS were assessed with summary statistics from 3 large-scale MS genetic data sets totaling 61,667 MS cases and 86,806 controls from the International MS Genetic Consortium. Multiple sensitivity analyses were performed to evaluate the assumptions of MR and remove potentially pleiotropic variants. Using inverse-variance weighted MR, we found no evidence for a causal effect of serum urate level on the risk of MS in any of the cohorts (MS1: OR 0.99 per each mg/dL unit increase in urate, 95% CI 0.89-1.08, p = 0.76; MS2: OR = 0.99, 95% CI 0.89-1.11, p = 0.90; MS3: OR = 1.00, 95% CI 0.98-1.2, p = 0.91). Pleiotropy robust MR methods yielded consistent estimates. This MR study does not support a clinically relevant causal effect of serum urate levels on the risk of MS.

Abstract: Yielding behavior is well known in attractive colloidal suspensions. Adhesive non-Brownian suspensions, in which the interparticle bonds are due to finite-size contacts, also show yielding behavior. We use a combination of steady-state, oscillatory and shear-reversal rheology to probe the physical origins of yielding in the latter class of materials, and find that yielding is not simply a matter of breaking adhesive bonds, but involves unjamming from a shear-jammed state in which the micro-structure has adapted to the direction of the applied load. Comparison with a recent constraint-based rheology model shows the importance of friction in determining the yield stress, suggesting novel ways to tune the flow of such suspensions., Comment: 8 pages, 6 figures
