[Image: see text] Hit-to-lead studies employ a variety of strategies to optimize binding to a target of interest. When a structure for the target is available, hypothesis-driven structure–activity relationships (SAR) are a powerful strategy for refining the pharmacophore to achieve robust binding and selectivity characteristics necessary to identify a lead compound. Recrafting the three-dimensional space occupied by a small molecule, optimization of hydrogen bond contacts, and enhancing local attractive interactions are traditional approaches in medicinal chemistry. Ring size, however, is rarely able to be leveraged as an independent variable because most hits lack the symmetry required for such a study. Our discovery that the cyclic oligomeric depsipeptide ent-verticilide inhibits mammalian cardiac ryanodine receptor calcium release channels with submicromolar potency provided an opportunity to explore ring size as a variable, independent of other structural or functional group changes. We report here that ring size can be a critical independent variable, suggesting that modest conformational changes alone can dramatically affect potency.

Recently published syntheses, reactions and characterizations of unusual unsaturated ring strained Group 4 metallocene metallacycles like metalla-cyclocumulenes, -cycloallenes and -cycloalkynes with different ring size are updated for the last three years. There exist for some of these metallacycles, depending on the ring size, 7-, 5- and 4-membered compounds. The new results for these metallacycles are summarized here and considered in addition to the former published results. Additionally, several compounds of this type were now characterized by new reactions. For a better understanding of these compounds, some spectroscopical methods as well as theoretical calculations were published. Despite of these all-C-metallacycles, only in some cases the syntheses and reactions for the corresponding hetero-metallacycles were published too. Examples for these metallaheterocyclic compounds will not be considered in this article. All these unusual ring strained compounds have a great potential for a lot of interesting synthetic applications in the future. Additionally, they are very interesting from the theoretical point of view.

An underside binding site was recently identified in the transmembrane domain of smoothened receptor (SMO). Herein, we report efforts in the exploration of new insights into the interactions between the ligand and SMO. The hydantoin core in the middle of the parent compound was found to be highly conservative in chirality, ring size, and substituents. On each benzene at two ends, a plethora of variations, particularly halogen substitutions, were introduced and investigated. Analysis of the structure‐activity relationship revealed miscellaneous halogen effects. The ligands with double halogen substituents exhibit remarkably enhanced potency, providing promising candidates that potentially overcome the common drug resistance and useful heavy‐atom labeled chemical tools for co‐crystallization studies of SMO., A series of diaryl hydantoin analogs were synthesized and evaluated in the inhibition of Hedgehog pathway by targeting the smoothened receptor. Double halogen substituents introduced at two ends of the molecule greatly enhanced the potency.

Here, we report the introduction of internally catalyzed amide bonds to obtain covalent adaptable polyamide networks that rely on the dissociation equilibrium between dicarboxamides and imides. While amide bonds are usually considered to be robust and thermally stable, the present study shows that their dynamic character can be activated by a smart choice of available building blocks without the addition of any external catalyst or other additives. Hence, a range of polyamide-based dynamic networks with variable mechanical and viscoelastic properties have been obtained in a systematic study, using a straightforward curing process of dibasic ester and amine compounds. Since the dissociation process involves a cyclic imide formation, the correlation between ring size and the thermomechanical viscosity profile was studied for five- to seven-membered ring intermediates, depending on the chosen dibasic ester monomer. This resulted in a marked temperature response with activation energies in the range of 116-197 kJ mol-1, yielding a sharp transition between elastic and viscous behavior. Moreover, the ease and versatility of this chemistry platform were demonstrated by selecting a variety of amines, resulting in densely cross-linked dynamic networks with Tg values ranging from -20 to 110 °C. With this approach, it is possible to design amorphous polyamide networks with an acute temperature response, allowing for good reprocessability and, simultaneously, high resistance to irreversible deformation at elevated temperatures.

The structural and bonding properties of Bi and Ac complexes with cyclen-based chelating ligands have been studied using relativistic DFT calculations in conjunction with TZ2P all-electron basis sets. Besides the parent cyclen ligand, the study has covered its extensions with pyridine-type (Lpy), carboxylate (DOTA, DOTPA), picolinate (MeDO2PA) and phosphonate (DOTMP) pendant arms. The effect of the cyclen ring size has been probed by increasing it from [12]aneN4 to [16]aneN4. Additional extensions in the DOTA complexes included the H2O ligand at the 9th coordination site as well as the p-SCN-Bn substituent (a popular linker to the targeting vector). The study focuses on the complex stability, the nature of bonding and the differences between Ac and Bi in the complexes. The metal–ligand interactions have been analysed by the Extended Transition State method combined with Natural Orbitals of Chemical Valence theory and Quantum Theory of Atoms in Molecules models.

We report a comprehensive study on the molecular conformation and dynamics of very large poly(ethylene oxide) rings in the melt: (i) for all rings, independent of the ring size, by small angle neutron scattering we observe a crossover from a strong Q-dependence at intermediate Q to a Q − 2 dependence at higher Q. Constructing a generic model including a crossover from Gaussian statistics at short distances to more compact structures at larger distances, we find the crossover at a distance along the ring of N e , 0 = 45 ± 2.5 monomers close to the entanglement distance in the linear counterpart. This finding is clear evidence for the predicted elementary loops building the ring conformation. (ii) The radius of gyration R g ( N ) follows quantitatively the result of numerous simulations. However, other than claimed, the crossover to mass fractal statistics does occur around N ≅ 10 N e , 0, but up to N ≅ 44 N e , 0, the relation R g ( N ) ∼ N 0.39 holds. The self-similar ring dynamics was accessed by pulsed field gradient-NMR and neutron spin echo spectroscopy: we find three dynamic regimes for center of mass diffusion starting (i) with a strong subdiffusive domain ⟨ r c o m 2 ( t ) ⟩ ∼ t α ( 0.4 ≤ α ≤ 0.65 ), (ii) a second subdiffusive region ⟨ r c o m 2 ( t ) ⟩ ∼ t 0.75 that (iii) finally, crosses over to Fickian diffusion. The internal dynamics at scales below the elementary loop size is well described by the ring Rouse motion. At larger scales, the dynamics is self-similar and follows very well the predictions of scaling models with a preference for the fractal loopy globule model. Finally, we note that the key results were previously published in the form of two letters [Kruteva et al., ACS Macro Lett. 9, 507–511 (2020) and Kruteva et al., Phys. Rev. Lett. 125, 238004 (2020)].

Feroniellin analogs isolated from Feroniella lucida possess a furanocoumarin skeleton connected to monoterpenic five- to seven-membered ethereal rings by an ether linkage and exhibit a broad spectrum of biological activities. In this contribution, we intended to establish a "ring-size-divergent" synthetic strategy for the monoterpenic five- to seven-membered ethereal rings through the chemical sythesis of feroniellins. The short and comprehensive synthesis of feroniellins was achieved in only two steps from easily available bergamottin based on the "ring-size-divergent" strategy. In addition, these syntheses resulted in revision of the proposed structures for feroniellins A and B and the determination of all the absolute configurations of feroniellins; their preliminary anti-inflammatory activities were investigated as well.

Cyclic peptide diversity has been broadened by elaborating the A3-macrocyclization to include various di-amino carboxylate components with different Ne-amine substituents. Triple-bond reduction provided new cyclic peptide macrocycles with Z-olefin and completely saturated structures. Moreover, cyclic azasulfurylpeptides were prepared by exchanging the propargylglycine (Pra) component for an amino sulfamide surrogate. Examination of such diversity-oriented methods on potent cyclic azapeptide modulators of the cluster of differentiation 36 receptor (CD36) identified the importance of the triple bond as well as the Ne-allyl lysine and azaPra residues for high CD36 binding affinity. Cyclic azapeptides which engaged CD36 effectively reduced pro-inflammatory nitric oxide and downstream cytokine and chemokine production in macrophages stimulated with a Toll-like receptor-2 agonist. Studying the triple bond and amine components in the multiple-component A3-macrocyclization has given a diverse array of macrocycles and pertinent information to guide the development of ideal CD36 modulators with biomedical potential for curbing macrophage-driven inflammation.

A reinvestigation into the macrocyclooligomerization (MCO) of a tetradepsipeptide is reported, uncovering a paradox in which the MCO of depsipeptide monomers can produce “impossible” ring sizes: a 12-atom chain produced the expected 24-membered ring, alongside unexpected 18- and 30-membered cyclic oligomeric depsipeptides (CODs). We report an alternative preparation of authentic 18- and 36-membered macrocycles for this case using a stepwise synthesis that provides definitive analytical characterization for each ring size. Our investigation yields a recharacterization and reassignment of two macrocycles originally reported in this MCO series, along with updated yields and isothermal titration calorimetry data after implementation of new critical protocols for purification and subsequent analysis. Initial studies to probe this mechanistic conundrum are described.

Inhibition of the immunoproteasome (iCP) offers new opportunities in the treatment of cancer, autoimmune disorders and neurodegenerative diseases. Inspired by the success of boronic acids as proteasome inhibitors we have complied a virtual library of commercially available 5- and 6-membered borocycles and performed a structure based virtual screening against the chymotrypsin-like (β5i) subunit of the iCP. The top scored docking poses were visually inspected to select compounds for experimental testing. Six compounds with 5-membered ring and another six compounds with 6-membered ring were subjected to biochemical tests. All compounds exhibited detectable inhibitory activity at 100 µM concentration and these are the first reported cyclic boronic acid inhibitors of the iCP. Structural variations including the ring size and the substitution of the borocyles and the substitution pattern of the attached aromatic ring resulted in no major variation of the inhibitory activity. We propose that the evaluation of larger cycling boronic acid libraries is needed to fully elucidate the potential of these structures.

The redox state of the metallomonooxygenases is finely tuned by imposing specific coordination environments on the metal center to reduce the activation energy for the generation of active-oxygen species and subsequent substrate oxygenation reactions. In this study, copper(II) complexes supported by a series of linear tetradentate ligands consisting of a rigid 6-, 7-, or 8-membered cyclic diamine with two pyridylmethyl (-CH2Py) side arms (L6Pym2, L7Pym2, and L8Pym2) are employed to examine the effects of the coordination environment on the reactivity of their acylperoxide adduct complexes. The UV-vis and electron paramagnetic resonance spectroscopic data indicate that the ligand-field splitting between the dx2-y2 and dz2 orbitals of the starting copper(II) complexes increase with an increase of the ring size of the diamine moiety (L6Pym2 → L7Pym2 → L8Pym2). In the reaction of these copper(II) complexes with m-chloroperbenzoic acid (m-CPBA), the L6Pym2 complex gives a stable m-CPBA adduct complex, whereas the L7Pym2 and L8Pym2 complexes are immediately converted to the corresponding m-chlorobenzoic acid (m-CBA) adducts, indicating that the reactivity of the copper(II) acylperoxide complexes largely depends on the coordination environment induced by the supporting ligands. Density functional theory (DFT) calculations on the m-CPBA adduct complexes show that the ligand-field-splitting energy increases with an increase of the ring size of the diamine moiety, as in the case of the starting copper(II) complexes, which enhances the reactivity of the m-CPBA adduct complexes. The reasons for such different reactivities of the m-CPBA adduct complexes are evaluated by using DFT calculations.

Here, we report a facile method to synthesize a series of macrocycles with different conformations. The planar macrocycle dimer (1), twisted macrocycle trimer (2) and "figure-eight" tetramer (3) are clearly elucidated by X-ray single-crystal analysis, in which the electron-rich phenanthrene units offer the possibility of supramolecular assembly. As expected, in the solid state, 1 and 3 assemble into a columnar stack and an interlocking dimer, respectively, via π-π interactions between the phenanthrene units. Compared to the rigid conformation of dimer 1, the structure of tetramer 3 is more flexible due to its enlarged ring size. 3 can deform from a figure-eight into a boat-shaped geometry to host a planar electron-deficient guest using its electron-rich phenanthrene units. When assembled with spherical electron-deficient C60 , interestingly, 3 further undergoes a conformational transformation from a figure-eight to a belt shape in order to host C60 . These supramolecular assembly behaviors of 3 demonstrate that it is an adaptable macrocyclic host for both planar molecules and fullerenes.

Here, we report a method for the one-pot ribosomal synthesis of macrocyclic depsipeptides. This method is based on a Ser-Pro-Cys-Gly (SPCG) motif discovered by in vitro selection of peptides for the function of self-acylation in the presence of a thioester acyl donor, which forms an O-acyl isopeptide bond via intramolecular S-to-O acyl transfer. Ribosomal synthesis of linear peptides containing the SPCG motif and a backbone "acyl donor" thioester at a downstream position results in spontaneous conversion to the corresponding cyclic depsipeptides (CDPs) in a nearly independent manner of ring size and sequence context. Mutational analysis of the SPCG motif revealed that the P and G residues are dispensable to some extent, but the arrangement of residues in SXCX is crucial for efficient acyl transfer, e.g., CPSG is much less efficient. Finally, one-pot ribosomal synthesis of macrocyclic depsipeptides with various ring sizes and sequences has been demonstrated. This synthetic method can facilitate the ribosomal construction of highly diverse CDP libraries for the discovery of de novo bioactive CDPs.

Ruthenium-catalyzed cycloadditions to form five-, six-, and seven-membered rings are summarized, including applications in natural product total synthesis. Content is organized by ring size and reaction type. Coverage is limited to processes that involve formation of at least one C-C bond. Processes that are stoichiometric in ruthenium or exploit ruthenium as a Lewis acid (without intervention of organometallic intermediates), ring formations that occur through dehydrogenative condensation-reduction, σ-bond activation-initiated annulations that do not result in net reduction of bond multiplicity, and photochemically promoted ruthenium-catalyzed cycloadditions are not covered.

Ring rolling technology is widely used in the production of GH4738 aerospace rings, but the ring produced by ring rolling often has the phenomenon of uneven stress and unqualified strength. To solve these problems, the thick-walled ring bulge forming technology was proposed. In this paper, a 3D-coupled thermo-mechanical FE model of the GH4738 rectangular ring bulging process was developed, which is verified to be in good agreement with the experimental process. Based on this model, the thermal parameter distribution and evolution of GH4738 rectangular ring during the bulging process were investigated systematically. Then, the evolution of the minimum bulging displacement (BD) required for rings with the change of ring size was obtained by comparing the stress distribution of rings during the bulging process in seven cases. All these provide a theoretical basis for the selection of optimal BD in practical production. Finally, the influence of the bulging process on the microstructure and mechanical properties of GH4738 alloy ring was analyzed by experimental means, and it was verified that the bulging process can play a good role in improving the strength of the ring from the perspective of the average geometrically necessary dislocation (GND) density. The experimental results indicate that with the increase of BD, the grain size of the ring does not change that much, but the average GND density increases gradually, and the yield strength and tensile strength of the ring increase significantly.

A Series of new tacrine analogs were designed, synthesized, characterized by respective spectral data and evaluated for cholinesterase inhibitory activity to be useful in Alzheimer’s disease. Most of the synthesized compounds showed good in vitro inhibitory activities toward acetyl cholinesterase (AChE) and butyrylcholinesterase (BuChE) enzymes. Among the compounds, 6i, 6o and 6r with increased saturated carboxylic ring size attached to the pyridine moiety and having 3,4-dihydroxy, 3,4,5-trimethoxy substituents on the aromatic ring attached at the stereogenic center have shown equal potency to that of tacrine with IC50values 0.65 ± 0.06, 1.32 ± 0.02 and 0.85 ± 0.05, 1.65 ± 0.12 and 0.92 ± 0.03, 1.91 ± 0.12 μM against AChE and BuChE, respectively. Standard drug tacrine exhibited IC50 values of 0.47 ± 0.02 and 0.65 ± 0.08, while Donepezil showed IC50 0.71 ± 0.06 and 0.31 ± 0.04 μM against AChE and BuChE, respectively. Docking studies of all the molecules disclosed close hydrogen bond interactions with the binding site.

A variety of cyclen and hexacyclen derivatives decorated with (S)-1-phenylethyl side chains or (S)-pyrrolidine units have been prepared via a reductive approach from the corresponding cyclic peptoids containing N-(S)-(1-phenylethyl)glycine and L-proline residues. Spectroscopic and DFT studies on their Na+ complexes show that point chirality and ring size play a crucial role in controlling the structural dynamism of 1,2-diaminoethylene units and pendant arms. The detection of highly symmetric C4- and C3-symmetric metalated species demonstrates that a full understanding of the relationship between the structure and conformational properties of peraza-macrocyclic metal complexes is possible.

Bioactive flax cyclic octa- and nona-peptides containing single methionine (Met) and its oxidized forms were treated under mild alkaline conditions to perform regio-selective epimerization. Cyclic peptide epimerization at the Met α-proton in a single chemical step has not been reported previously. The epimerization rate varies among Met oxidation states and ring size. These d-amino isomers along with the developed Met alkylation strategy will enable an approach to novel chemical functionalization of biomolecules. The amino acid configurations were confirmed by Marfey derivatizations, and cytotoxicity studies show the difference among the isomers. These d-amino analogs can act as a potential biomarker in plant protein processing and biomedical applications.

One-pot sequential p-hydroxybenzylation/oxidative dearomatization/spiroannulation has been designed for the efficient construction of tetrahydrofuran containing spiro-cyclohexadienones. This reaction proceeds through the p-hydroxybenzylation of 1,3-diketones with p-hydroxybenzyl alcohol via quinone methide formation followed by oxidative dearomatization/spiroannulation with suitable alcohols. The Friedel–Crafts alkylation of spiro[4.5]cyclohexadienones with indoles provided a broad array of highly diastereoselective C-3 alkylated spirocycles and cyclohepta[b]indoles depending upon the ring size of the fused cyclic ketones.

Triazolate-based pillarplexes, supramolecular organometallic complexes (SOCs) featuring a tubular pore, are introduced. Following a macrocycle-templated synthesis strategy, a hybrid imidazolium/triazole cyclophane of defined ring size was used as ligand precursor, serving also as a supramolecular element in the subsequent SOC self-assembly. The new pillarplexes in part behave similar to previously reported pyrazolate-based congeners, e.g., regarding luminescence or host–guest properties. In contrast, for the Au(I) complex, an unprecedented shape-adaptive behaviour is observed in the solid-state, caused by the additional nitrogen atoms being present in the rim and rationalised by DFT calculations. Additionally, complementary hydrogen bonding between the cationic complexes is observed, proving that the rim modification not only modulates the flexibility of the cavitand but is also a tool to introduce further functionality into the SOC.

Depending on the ring size in the Reformatsky reagent and the nature of substituents in arylmethylidene fragment, the reactions of Reformatsky reagents obtained from methyl esters of 1-bromocycloalkanecarboxylic acids and zinc with N′-(arylmethylidene)-2-oxo-2H-chromene-3-carbohydrazides afforded methyl 1-{3-[2-(arylmethylidene)hydrazinylcarbonyl]-2-oxochroman-4-yl}cycloalkanecarboxylates or methyl 1-{3-[(3-aryl-1-oxo-2-azaspiro[3.5]nonan-2-yl)carbamoyl]-2-oxochroman-4-yl}cyclohexanecarboxylates. The resulting compounds exhibit antinociceptive activity.

The currently limited portfolio of volatile organoselenium compounds used for atomic layer deposition (ALD) has been extended by designing and preparing a series of four-, five- and six-membered cyclic silylselenides. Their fundamental properties were tailored by alternating the ring size, the number of embedded Se atoms and the used peripheral alkyl chains. In contrast to former preparations based on formation of sodium or lithium selenides, the newly developed synthetic method utilizes a direct and easy reaction of elemental selenium with chlorosilanes. Novel 2,2,4,4-tetraisopropyl-1,3,2,4-diselenadisiletane, which features good trade-off between chemical/thermal stability and reactivity, has been successfully used for gas-to-solid phase reaction with MoCl5 affording MoSe2. A thorough characterization of the as-deposited 2D MoSe2 flakes revealed its out-of-plane orientation and high purity. Hence, the developed four-membered cyclic silylselenide turned out to be well-suited Se-precursor for ALD of MoSe2. Pro depozici atomárních vrstev byla navržena a připravena série čtyř-, pěti- a šestičlenných cyklických silylselenidů. Nově vyvinutá metoda syntézy využila přímou a jednoduchou reakci selenu s chlorsilany. Vyvinuté čtyřčlenné cyklické silylselenidy se ukázaly jako vhodné prekurzory pro ALD sloučeniny MoSe2.

In order to further investigate the importance of the conformation of the ring I side chain in aminoglycoside antibiotic binding to the ribosomal target several derivatives of paromomycin were designed with conformationally locked side chains. By changing the size of the appended ring between O-4′ and C-6′ used to restrict the motion of the side chain, the position of the C-6′ hydroxy group was fine tuned to probe for the optimal conformation for inhibition of the ribosome. While the changes in orientation of the 6′-hydroxy group cannot be completely dissociated from the size and hydrophobicity of the conformation-restricting ring, overall, it is apparent that the preferred conformation of the ring I side chain for interaction with A1408 in the decoding A site of the bacterial ribosome is an ideal gt conformation, which results in the highest antimicrobial activity as well as increased selectivity for bacterial over eukaryotic ribosomes.

The copolymerization of an ε-substituted ε-lactone, menthide (MI), and a range of nonsubstituted lactones (6-, 7-, 8-, and 9-membered rings) was investigated in order to determine the factors that affect the sequencing of the MI copolymers. Analysis by quantitative 13C NMR spectroscopy showed the copolymerization of MI with a nonsubstituted lactone of ring size 7 or less produced a randomly sequenced copolymer, as a consequence of the smaller lactone polymerizing first and undergoing rapid transesterification as MI was incorporated. Conversely, copolymerization with larger ring lactones (ring size 8 and above) produced block-like copolymers as a consequence of MI polymerizing initially, which does not undergo rapid transesterification side reactions during the incorporation of the second monomer. Terpolymerizations of a small ring lactone, macrolactone, and menthide demonstrated methods of producing lactone terpolymers with different final sequences, depending on when the small ring lactone was injected into the reaction mixture.\ud \ud

DAMASCENOLIDETM [1, 4-(4-methylpent-3-en-1-yl)furan-2(5H)-one], which has a citrus-like odor, is an important aroma component of roses. We have previously reported on the synthesis and odor evaluation of 24 analogs of 1 as part of our new aroma compound developing project. To accumulate more information on structure–odor relationships, 10 more promising analogs such as dimethylated and cyclopropanated analogs were synthesized and subjected to odor evaluation. As a result, it was found that dimethylation of the furanone ring affected the odor. It was also found that cyclopropanation of 1 affected the odor, whereas cyclopropanation of the double bond isomer of 1 did not significantly affect the odor. The effects on the odor caused by ring size expansion and replacement of the side chain were also investigated.
