Nonlinear, i.e., multi-photon microscopy is a powerful technique for imaging deep into biological tissues. Its penetration depth can be increased further using adaptive optics. In this work, we present a fast, feedback-based adaptive-optics algorithm, termed C-DASH, for multi-photon imaging through multiply-scattering media. C-DASH utilises complex-valued light shaping (i.e., joint shaping of amplitude and phase), which offers several advantages over phase-only techniques: it converges faster, it delivers higher image quality enhancement, it shows a robust performance largely insensitive to the axial position of the correction plane, and it has a higher ability to cope with, on the one hand, scattering media that vary over time and, on the other hand, scattering media that are partially absorbing. Furthermore, our method is practically self-aligning. We also present a simple way to implement C-DASH using a single reflection off a phase-only spatial light modulator. We provide a thorough characterisation of our method, presenting results of numerical simulations as well as two-photon excited fluorescence imaging experiments.

Adaptive optics in combination with multi-photon techniques is a powerful approach to image deep into a specimen. Remarkably, virtually all adaptive optics schemes today rely on wavefront modulators which are reflective, diffractive, or both. This, however, can pose a severe limitation for applications. Here, we present a fast and robust sensorless adaptive optics scheme adapted for transmissive wavefront modulators. We study our scheme in numerical simulations and in experiments with a novel, optofluidic wavefront shaping device which is transmissive, refractive, polarisation-independent and broadband. We demonstrate scatter correction of two-photon-excited fluorescence images of microbeads as well as brain cells and benchmark our device against a liquid-crystal spatial light modulator. Our method and technology could open new routes for adaptive optics in scenarios where previously the restriction to reflective and diffractive devices may have staggered innovation and progress.

Optical tweezers are tools made of light that enable contactless pushing, trapping, and manipulation of objects ranging from atoms to space light sails. Since the pioneering work by Arthur Ashkin in the 1970s, optical tweezers have evolved into sophisticated instruments and have been employed in a broad range of applications in life sciences, physics, and engineering. These include accurate force and torque measurement at the femtonewton level, microrheology of complex fluids, single micro- and nanoparticle spectroscopy, single-cell analysis, and statistical-physics experiments. This roadmap provides insights into current investigations involving optical forces and optical tweezers from their theoretical foundations to designs and setups. Adaptive optics (AO) is a powerful tool to increase the imaging depth of multiphoton scanning microscopes. For highly scattering tissues, sensorless wavefront correction techniques exhibit robust performance and present a straight-forward implementation of AO. However, for many applications such as live-tissue imaging, the speed of aberration correction remains a critical bottleneck. Dynamic Adaptive Scattering compensation Holography (DASH) -- a fast-converging sensorless AO technique introduced recently for scatter compensation in nonlinear scanning microscopy -- addresses this issue. DASH has been targeted at highly turbid media, but to-date it has remained an open question how it performs for mild turbidity, where limitations imposed by phase-only wavefront shaping are expected to impede its convergence. In this work, we study the performance of DASH across different turbidity regimes, in simulation as well as experiments. We further provide a direct comparison between DASH and a novel, modified version of the Continuous Sequential Algorithm (CSA) which we call Amplified CSA (a-CSA).

We present a method to measure the optical torque applied to particles of arbitrary shape held in an optical trap, inferred from the change of angular momentum of light induced by the particle. All torque components can be determined from a single interference pattern recorded by a camera in the back focal plane of a high-NA condenser collecting all forward scattered light. We derive explicit expressions mapping the measured complex field in this plane to the torque components. The required phase is retrieved by an iterative algorithm, using the known position of the optical traps as constraints. In this paper, we study the mathematical imaging problem of optical diffraction tomography (ODT) for the scenario of a microscopic rigid particle rotating in a trap created, for instance, by acoustic or optical forces. Under the influence of the inhomogeneous forces the particle carries out a time-dependent smooth, but complicated motion described by a set of affine transformations. The rotation of the particle enables one to record optical images from a wide range of angles, which largely eliminates the "missing cone problem" in optics. This advantage, however, comes at the price that the rotation axis in this scenario is not fixed, but continuously undergoes some variations, and that the rotation angles are not equally spaced, which is in contrast to standard tomographic reconstruction assumptions. In the present work, we assume that the time-dependent motion parameters are known, and that the particle's scattering potential is compatible with making the first order Born or Rytov approximation. We prove a Fourier diffraction theorem and derive novel backprojection formulae for the reconstruction of the scattering potential, which depends on the refractive index distribution inside the object, taking its complicated motion into account. This provides the basis for solving the ODT problem with an efficient non-uniform discrete Fourier transform.

Optical and acoustical trapping has been established as a tool for holding and moving microscopic particles suspended in a liquid in a contact-free and non-invasive manner. Opposed to standard microscopic imaging where the probe is fixated, this technique allows imaging in a more natural environment. This paper provides a method for estimating the movement of a transparent particle which is maneuvered by tweezers, assuming that the inner structure of the probe is not subject to local movements. The mathematical formulation of the motion estimation shows some similarities to Cryo-EM single particle imaging, where the recording orientations of the probe need to be estimated.

Objects at finite temperature emit thermal radiation with an outward energy-momentum flow, which exerts an outward radiation pressure. At room temperature, a cesium atom scatters on average less than one of these blackbody radiation photons every 10^8 years. Thus, it is generally assumed that any scattering force exerted on atoms by such radiation is negligible. However, atoms also interact coherently with the thermal electromagnetic field. In this work, we measure an attractive force induced by blackbody radiation between a cesium atom and a heated, centimeter-sized cylinder which is orders of magnitude stronger than the outward directed radiation pressure. Using atom interferometry, we find that this force scales with the fourth power of the cylinder`s temperature. The force is in good agreement with that predicted from an ac Stark shift gradient of the atomic ground state in the thermal radiation field. This observed force dominates over both gravity and radiation pressure, and does so for a large temperature range.

Blackbody radiation around hot objects induces ac Stark shifts of the energy levels of nearby atoms and molecules. These shifts are roughly proportional to the fourth power of the temperature and induce a force decaying with the third power of the distance from the object. We explicitly calculate the resulting attractive blackbody optical dipole force for ground state hydrogen atoms. Surprisingly, this force can surpass the repulsive radiation pressure and actually pull the atoms against the radiation energy flow towards the surface with a force stronger than gravity. We exemplify the dominance of the "blackbody force" over gravity for hydrogen in a cloud of hot dust particles. This overlooked force appears relevant in various astrophysical scenarios, in particular, since analogous results hold for a wide class of other broadband radiation sources.

Light forces induced by scattering and absorption in elastic dielectrics lead to local density modulations and deformations. These perturbations in turn modify light propagation in the medium and generate an intricate nonlinear response. We generalise an analytic approach where light propagation in one-dimensional media of inhomogeneous density is modelled as a result of multiple scattering between polarizable slices. Using the Maxwell stress tensor formalism we compute the local optical forces and iteratively approach self-consistent density distributions where the elastic back-action balances gradient- and scattering forces. For an optically trapped dielectric we derive the nonlinear dependence of trap position, stiffness and total deformation on the object's size and field configuration. Generally trapping is enhanced by deformation, which exhibits a periodic change between stretching and compression. This strongly deviates from qualitative expectations based on the change of photon momentum of light crossing the surface of a dielectric. We conclude that optical forces have to be treated as volumetric forces and that a description using the change of photon momentum at the surface of a medium is inappropriate.

We demonstrate coherent anti-Stokes Raman scattering (CARS) microscopy in a wide-field setup with nanosecond laser pulse excitation. In contrast to confocal setups, the image of a sample can be recorded with a single pair of excitation pulses. For this purpose the excitation geometry is specially designed in order to satisfy the phase matching condition over the whole sample area. The spectral, temporal and spatial sensitivity of the method is demonstrated by imaging test samples, i.e. oil vesicles in sunflower seeds, on a nanosecond timescale.
