Bipolar disorder (BD) is a serious mental illness with substantial common variant heritability. However, the role of rare coding variation in BD is not well established. We examined the protein-coding (exonic) sequences of 3,987 unrelated individuals with BD and 5,322 controls of predominantly European ancestry across four cohorts from the Bipolar Sequencing Consortium (BSC). We assessed the burden of rare, protein-altering, single nucleotide variants classified as pathogenic or likely pathogenic (P-LP) both exome-wide and within several groups of genes with phenotypic or biologic plausibility in BD. While we observed an increased burden of rare coding P-LP variants within 165 genes identified as BD GWAS regions in 3,987 BD cases (meta-analysis OR = 1.9, 95% CI = 1.3-2.8, one-sided p = 6.0 × 10-4), this enrichment did not replicate in an additional 9,929 BD cases and 14,018 controls (OR = 0.9, one-side p = 0.70). Although BD shares common variant heritability with schizophrenia, in the BSC sample we did not observe a significant enrichment of P-LP variants in SCZ GWAS genes, in two classes of neuronal synaptic genes (RBFOX2 and FMRP) associated with SCZ or in loss-of-function intolerant genes. In this study, the largest analysis of exonic variation in BD, individuals with BD do not carry a replicable enrichment of rare P-LP variants across the exome or in any of several groups of genes with biologic plausibility. Moreover, despite a strong shared susceptibility between BD and SCZ through common genetic variation, we do not observe an association between BD risk and rare P-LP coding variants in genes known to modulate risk for SCZ.

7. How Vulnerable Are American States to Wildfires? A Livelihood Vulnerability Assessment

Quantifying livelihood vulnerability to wildland fires in the United States is challenging because of the need to systematically integrate multidimensional variables into its analysis. We aim to measure wildfire threats amongst humans and their physical and social environment by devel-oping a framework to calculate the livelihood vulnerability index (LVI) for the top 14 American states most recently exposed to wildfires. The LVI is computed by assessing each state’s contributing factors (exposure, sensitivity, and adaptive capacity) to wildfire events. These contributing factors are determined through a set of indicator variables that are categorized into corresponding groups to produce an LVI framework. The framework is validated by performing a principal component analysis (PCA), ensuring that each selected indicator variable corresponds to the correct contributing factor. Our results indicate that Arizona and New Mexico experience the greatest livelihood vulnerability. In contrast, California, Florida, and Texas experience the least livelihood vulnerabil-ity. While California has one of the highest exposures and sensitivity to wildfires, results indicate that it has a relatively high adaptive capacity, in comparison to the other states, suggesting it has measures in place to withstand these vulnerabilities. These results are critical to wildfire managers, government, policymakers, and research scientists for identifying and providing better resiliency and adaptation measures to support states that are most vulnerable to wildfires.

This work aims to produce hybrid materials with potential applications in dye photodegradation. Therefore, hybrid films were obtained by incorporating cobalt (II, III) oxide (Co
3 O4 ) or cobalt ferrite (CoFe2 O4 ) nanoparticles (NPs) with 18 ± 1.6 nm and 26 ± 1.3 nm, respectively, into a poly 3-hydroxybutyrate (P3HB) polymeric matrix. The Co3 O4 @P3HB and CoFe2 O4 @P3HB hybrid films were fabricated by solvent casting in a ratio of 85 mg to 15 mg (P3HB-NPs). Different spectroscopic and microscopy techniques characterized the Co3 O4 and CoFe2 O4 NPs and the P3HB, Co3 O4 @P3HB and CoFe2 O4 @P3HB films. The optical band gap for Co3 O4 and CoFe2 O4 NPs was estimated from their diffuse reflectance spectra (DRS) around 2.5 eV. X-ray diffraction (XRD) of the hybrid films revealed that the nanometric sizes of the Co3 O4 and CoFe2 O4 nanoparticles incorporated into the P3HB are preserved. The magnetic hysteresis curve of CoFe2 O4 nanoparticles and CoFe2 O4 @P3HB film showed a ferromagnetic behaviour at 300 K. Transmission electron microscopy (TEM) confirmed the formation of nanocrystals, and scanning electron microscopy (SEM) provided evidence for the successful incorporation of the NPs into the P3HB matrix. The surface roughness and hydrophilicity of the hybrid films are increased compared to the P3HB film. The impact of the nanoparticles and the hybrid films on the photodegradation of methyl orange (MO) in its acidic form was studied. The photodegradation tests were carried out by direct sunlight exposure. The CoFe2 O4 @P3HB hybrid film achieved 85% photodegradation efficiency of a methyl orange solution of 20 ppm after 15 minutes of exposure to sunlight. After 30 minutes of exposure to sunlight, the nanoparticles and the hybrid films reached about 90% of the MO degradation. The results suggest that combining nanoparticles with the polymer significantly enhances photodegradation compared to isolated nanoparticles. [ABSTRACT FROM AUTHOR]- Published

In this study, we carried out an electrochemical investigation of the palladium electrodeposition process at pH 5 and 8, evaluating the kinetic parameters related to its nucleation and growth processes on a Highly Oriented Pyrolytic Graphite (HOPG) electrode from a plating bath containing 1 mM of Pd and 1 M NH
4 Cl. The voltammetric study allowed us to identify the potential values at which palladium can be electrodeposited, along with the adsorption and desorption processes of hydrogen absorbed on the deposited Pd. Analysis of the peak currents of the deposited Pd indicated diffusional control at both pH values. The evaluation of kinetic parameters, such as the number of active nucleation sites (N0 ), the nucleation rate (A), and the rate constant of the proton reduction process (kPR ), was determined via potentiostatic studies, revealing their dependence on the applied potential to the electrode. The number of active nucleation sites predicted by the nucleation model correlated well with the number of nuclei observed via Scanning Electron Microscopy (SEM). SEM images revealed that at pH 5, the Pd clusters had an average diameter of 27 nm and a height of 39 nm, while at pH 8, the clusters had an average diameter of 12.8 nm and a height of 16.6 nm. At pH 5, homogeneous and dispersed Pd clusters were obtained, while at pH 8, agglomeration of Pd clusters was observed. [ABSTRACT FROM AUTHOR]- Published

Adversity, particularly in early life, can cause illness. Clues to the responsible mechanisms may lie with the discovery of molecular signatures of stress, some of which include alterations to an individual's somatic genome. Here, using genome sequences from 11,670 women, we observed a highly significant association between a stress-related disease, major depression, and the amount of mtDNA (p = 9.00 × 10(-42), odds ratio 1.33 [95% confidence interval [CI] = 1.29-1.37]) and telomere length (p = 2.84 × 10(-14), odds ratio 0.85 [95% CI = 0.81-0.89]). While both telomere length and mtDNA amount were associated with adverse life events, conditional regression analyses showed the molecular changes were contingent on the depressed state. We tested this hypothesis with experiments in mice, demonstrating that stress causes both molecular changes, which are partly reversible and can be elicited by the administration of corticosterone. Together, these results demonstrate that changes in the amount of mtDNA and telomere length are consequences of stress and entering a depressed state. These findings identify increased amounts of mtDNA as a molecular marker of MD and have important implications for understanding how stress causes the disease.

Background: Despite a significant clinical and social burden, there is a relative scarcity of epidemiological studies on Personality Disorder (PD). Aim: To determine the current prevalence of PD and the psychosocial correlates associated with this in the Andalusian population. Method: We carried out a cross-sectional population mental-health survey in Andalusia, southern Spain. Thus, 4,518 randomly selected participants were interviewed following sampling using different standard stratification levels. We used the Spanish version of the SAPAS to estimate PD prevalence. In addition, a full battery of other instruments was utilized to explore global functionality, childhood abuse, maltreatment, threatening life events, personality traits (neuroticism, impulsivity and paranoia), medical and psychiatric comorbidities, family history of psychological problems and other potential risk factors for PD. Results: PD prevalence (10.8%; 95% CI [9.8, 11.7]) and ran two different multivariate models for PD. We obtained the highest PD prevalence in those affected by any mental disorder plus those reporting having suffered childhood abuse, particularly sexual abuse. Additional potential risk factors or correlates of PD identified were: younger age, lower levels of functioning, less social support, poorer general health, having suffered maltreatment, threatening life events, higher suicidal risk scores and higher levels of both neuroticism and impulsivity. Conclusions: This study reports PD prevalence and risk correlates in consonance with similar findings reported in other Western populations. However, longitudinal studies are needed to elicit a more thorough group of prospective determinants of PD. [ABSTRACT FROM AUTHOR]

Homoleptic zinc(II) and cadmium(II) complexes containing hybrid N-selenophosphinoylchalcogenoureato ligands, [M(
i Pr2 P(Se)NC(X)NC5 H10 -κ2 -Se,X)2 ] (M = Zn, X = S (3), X = O (4) and M = Cd, X = S (5)) and [Cd(i Pr2 P(Se)NC(O)NC5 H10 )2 -μ-O,O]2 (6), were synthesized and characterized by elemental analysis, IR and NMR spectroscopy. SCXRD molecular structures of complexes revealed bidentate ligands bonded to the metal center in a distorted tetrahedral coordination geometry for 4 and 5, while 6 exhibited a dinuclear centrosymmetric structure with a distorted trigonal bipyramidal coordination. Based on thermogravimetric analysis (TGA), the complexes (3–6) were tested as single source precursors (SSPs) to prepare metal chalcogenide thin films by aerosol-assisted chemical vapor deposition (AACVD) at 450 °C (annealing at 600 °C) and 375 °C. 4 and 6 produced good quality hexagonal MSe thin films (4F and 6F), while, remarkably, 3 and 5 deposited the corresponding ternary hexagonal MSSe thin films (3F and 5F) with S contents of 15.8% for 3F and 1.8% for 5F (EDXA). This was confirmed by X-ray photoelectron spectroscopy (XPS) and energy dispersive X-ray analysis (EDXA), from which the elemental composition was also determined. The as-deposited thin films displayed a phosphorus content between 2.0 and 4.4%, while zinc-containing precursors produced zinc-rich thin films. The surface morphologies of 3F–6F were examined by scanning electron microscopy (SEM) and atomic force microscopy (AFM), showing quasi-spherical particles in columns. The optical properties suggest good crystalline quality of as-deposited thin films. The band gaps (Eg ) of MSSe films were higher than those of MSe materials. PL spectra of Cd-containing films showed a dominant emission band with a maximum between 1.69 and 1.76 eV related to near-band-edge (NBE) emission, while Zn-containing films showed broad PL signals at ca. 2.0–2.4 eV that could be associated with structural defects. All Raman spectra showed multiple LO phonon overtones, and MSSe solid solutions (from 3 and 5) showed both expected LO scattering modes (MSe-like and MS-like phonons). This study presents an in vitro analysis of the bactericidal and cytotoxic properties of hybrid films containing nickel oxide (NiO) and nickel ferrite (NiFe
2 O4 ) nanoparticles embedded in polypropylene (PP). The solvent casting method was used to synthesize films of PP, PP@NiO, and PP@NiFe2 O4 , which were characterized by different spectroscopic and microscopic techniques. The X-ray diffraction (XRD) patterns confirmed that the small crystallite sizes of NiO and NiFe2 O4 NPs were maintained even after they were incorporated into the PP matrix. From the Raman scattering spectroscopy data, it was evident that there was a significant interaction between the NPs and the PP matrix. Additionally, the Scanning Electron Microscopy (SEM) analysis revealed a homogeneous dispersion of NiO and NiFe2 O4 NPs throughout the PP matrix. The incorporation of the NPs was observed to alter the surface roughness of the films; this behavior was studied by atomic force microscopy (AFM). The antibacterial properties of all films were evaluated against Pseudomonas aeruginosa (ATCC® : 43636™) and Staphylococcus aureus (ATCC® : 23235™), two opportunistic and nosocomial pathogens. The PP@NiO and PP@ NiFe2 O4 films showed over 90% bacterial growth inhibition for both strains. Additionally, the effects of the films on human skin cells, such as epidermal keratinocytes and dermal fibroblasts, were evaluated for cytotoxicity. The PP, PP@NiO, and PP@NiFe2 O4 films were nontoxic to human keratinocytes. Furthermore, compared to the PP film, improved biocompatibility of the PP@NiFe2 O4 film with human fibroblasts was observed. The methodology utilized in this study allows for the production of hybrid films that can inhibit the growth of Gram-positive bacteria, such as S. aureus, and Gram-negative bacteria, such as P. aeruginosa. Objective: Suicidal behavior is heritable and is a major cause of death worldwide. Two large-scale genome-wide asso-ciation studies (GWASs) recently discovered and cross- validated genome-wide significant (GWS) loci for suicide attempt (SA). The present study leveraged the genetic co-horts from both studies to conduct the largest GWAS meta- analysis of SA to date. Multi-ancestry and admixture-specific meta-analyses were conducted within groups of significant African, East Asian, and European ancestry admixtures. Methods: This study comprised 22 cohorts, including 43,871 SA cases and 915,025 ancestry-matched controls. Analytical methods across multi-ancestry and individual ancestry ad-mixtures included inverse variance-weighted fixed-effects meta-analyses, followed by gene, gene-set, tissue-set, and drug-target enrichment, as well as summary-data-based Mendelian randomization with brain expression quantitative trait loci data, phenome-wide genetic correlation, and ge-netic causal proportion analyses. Results: Multi-ancestry and European ancestry admixture GWAS meta-analyses identified 12 risk loci at p values <5×10–8. These loci were mostly intergenic and implicated DRD2, SLC6A9, FURIN, NLGN1, SOX5, PDE4B, and CACNG2. The multi-ancestry SNP-based heritability estimate of SA was 5.7% on the liability scale (SE=0.003, p=5.7×10–80). Significant brain tissue gene expression and drug set en-richment were observed. There was shared genetic variation of SA with attention deficit hyperactivity disorder, smoking, and risk tolerance after conditioning SA on both major de-pressive disorder and posttraumatic stress disorder. Genetic causal proportion analyses implicated shared genetic risk for specific health factors. © 2020, The Author(s), under exclusive licence to Springer Nature Limited.Lithium is a first-line medication for bipolar disorder (BD), but only one in three patients respond optimally to the drug. Since evidence shows a strong clinical and genetic overlap between depression and bipolar disorder, we investigated whether a polygenic susceptibility to major depression is associated with response to lithium treatment in patients with BD. Weighted polygenic scores (PGSs) were computed for major depression (MD) at different GWAS p value thresholds using genetic data obtained from 2586 bipolar patients who received lithium treatment and took part in the Consortium on Lithium Genetics (ConLi+Gen) study. Summary statistics from genome-wide association studies in MD (135,458 cases and 344,901 controls) from the Psychiatric Genomics Consortium (PGC) were used for PGS weighting. Response to lithium treatment was defined by continuous scores and categorical outcome (responders versus non-responders) using measurements on the Alda scale. Associations between PGSs of MD and lithium treatment response were assessed using a linear and binary logistic regression modeling for the continuous and categorical outcomes, respectively. The analysis was performed for the entire cohort, and for European and Asian sub-samples. The PGSs for MD were significantly associated with lithium treatment response in multi-ethnic, European or Asian populations, at various p value thresholds. Bipolar patients with a low polygenic load for MD were more likely to respond well to lithium, compared to those patients with high polygenic load [lowest vs highest PGS quartiles, multi-ethnic sample: OR = 1.54 (95% CI: 1.18–2.01) and European sample: OR = 1.75 (95% CI: 1.30–2.36)]. While our analysis in the Asian sample found equivalent effect size in the same direction: OR = 1.71 (95% CI: 0.61–4.90), this was not statistically significant. Using PGS decile comparison, we found a similar trend of association between a high genetic loading for MD and lower response to lithium. Our findings underscore the genetic contribution to lithium response in BD and support the emerging concept of a lithium-responsive biotype in BD.
