The Diaprepes root weevil (DRW), Diaprepes abbreviatus, is a broadly polyphagous invasive pest of agriculture in the southern United States and the Caribbean. Its genome was sequenced, assembled, and annotated to study genomic correlates of specialized plant-feeding and invasiveness and to facilitate the development of new methods for DRW control. The 1.69 Gb D. abbreviatus genome assembly was distributed across 653 contigs, with an N50 of 7.8 Mb and the largest contig of 62 Mb. Most of the genome was comprised of repetitive sequences, with 66.17% in transposable elements, 5.75% in macrosatellites, and 2.06% in microsatellites. Most expected orthologous genes were present and fully assembled, with 99.5% of BUSCO genes present and 1.5% duplicated. One hundred and nine contigs (27.19 Mb) were identified as putative fragments of the X and Y sex chromosomes, and homology assessment with other beetle X chromosomes indicated a possible sex chromosome turnover event. Genome annotation identified 18,412 genes, including 43 putative horizontally transferred (HT) loci. Notably, 258 genes were identified from gene families known to encode plant cell wall degrading enzymes and invertases, including carbohydrate esterases, polysaccharide lyases, and glycoside hydrolases (GH). GH genes were unusually numerous, with 239 putative genes representing 19 GH families. The Asian citrus psyllid, Diaphorina citri Kuwayama, 1908 (Hemiptera: Liviidae), is a phloem-feeding insect pest of Citrus L. ( Sapindales: Rutaceae), Murraya J. Koenig ex L. (Sapindales: Rutaceae), and other Rutaceae, native to the Indian subcontinent and transmits the citrus greening pathogens (huanglongbing). We herein evaluate the occurrence of D. citri in Puerto Rico at a fine scale and the potential invasion history of the Caribbean from the native range. We found that the genetic diversity of D. citri in Puerto Rico, using the mitochondrial genetic marker CO1, indicated that the Caribbean haplotype is unique and more related to Southwest Asia haplotypes than other groups. Results then suggest that the invasion of the Caribbean did not occur from the United States. Also, we found that isothermality was the abiotic variable that influenced D. citri species distribution model performance the most in current and future climate change scenarios. Given the data available from the Global Biodiversity Information Facility (GBIF) for present D.citri reported occurrence, future projections suggest that climate change might promote an expansion of the pest to other Caribbean islands and Latin American countries. Still, it might elicit a range contraction across the Cordillera Central in Puerto Rico, possibly promoting citriculture. Regarding pest management strategies, we must consider a holistic approach to understanding D. citri invasion and resilience in novel habitats by including broader and more robust genetic population structure studies and if its current distribution reflects projected models across the neotropics., (© The Author(s) 2023. Worker division of labor is a defining trait in social insects. Many species are characterized by having behavioral flexibility where workers perform non-typical tasks for their age depending on the colony's needs. Worker division of labor and behavioral flexibility were examined in the little fire ant Wasmannia auropunctata (Roger, 1863), for which age-related division of labor has been found. Young workers perform nursing duties which include tending of brood and queens, and colony defense, while older workers forage. When nurses were experimentally removed from the colony, foragers were observed carrying out nursing and colony defense duties, yet when foragers were removed nurses did not forage precociously. We also administered juvenile hormone analog, methoprene, to workers. When methoprene was applied, foragers increased their nursing and defense activities while nurses became mainly idle. The behavioral flexibility of foragers of the little fire ant may be evidence of an expansion of worker's repertoires as they age; older workers can perform tasks they have already done in their life while young individuals are not capable of performing tasks ahead of time. Urinalysis is a rapid, simple, inexpensive, and reliable test that documents urine abnormalities reflecting various types of renal, hormonal, or metabolic diseases. Urinalysis could assist proper monitoring of the health of dolphins under human care; however, normal baseline values for dolphin urinalysis have not been reported, to our knowledge. We sampled urine from 193 common bottlenose dolphins ( Tursiops truncatus), living under human care in 24 Caribbean dolphinariums, by voluntary free-catch and analyzed the urine for chemical and microscopic variables using multi-agent dry reagent chemistry dipstick test strips, dedicated pH reagent strips, and unstained sediment slides. Most urine was clear, pale yellow to dark yellow, and had a fishy odor. Dipstick glucose, bilirubin, ketones, and nitrites were negative in all dolphins. The urine pH was acidic ( x ¯ ± SD; 5.88 ± 0.58) and specific gravity (SG) was 1.035 ± 0.008. Most animals had 0-2 red blood cells and white blood cells per 40× field, and were negative for proteins. On microscopic sediment, 42.7% of samples had few-to-many squamous epithelial cells; hyaline and epithelial casts were observed only rarely. Crystals were observed in 36.6% of the samples; most were calcium oxalate dihydrate (48.2%) and amorphous urates (42.4%). The values obtained in our study can be used as a reference for health monitoring of dolphins in dolphinariums, and to monitor renal conditions and function in dolphins being rehabilitated or under human care.

Background: The Africanized honey bee is one of the most spectacular invasions in the Americas. African bees escaped from apiaries in Brazil in 1956, spread over Americas and by 1994 they were reported in Puerto Rico. In contrast to other places, the oceanic island conditions in Puerto Rico may mean a single introduction and different dynamics of the resident European and new-coming Africanized bees.To examine the genetic variation of honey bee feral populations and colonies from different locations in Puerto Rico, we used eight known polymorphic microsatellite loci., Results: In Puerto Rico, gAHB population does not show any genetic structure (Fst = 0.0783), and is best described as one honey bee population, product of hybridization of AHB and EHB. The genetic variability in this Africanized population was similar to that reported in studies from Texas. We observed that European private allele frequencies are high in all but one locus. This contrasts with mainland Africanized populations, where European allele frequencies are diminished. Two loci with European private alleles, one on Linkage Group 7, known to carry two known defensiveness Quantitative Trait Loci (QTLs), and the other on Linkage Group 1, known to carry three functionally studied genes and 11 candidate genes associated with Varroa resistance mechanisms were respectively, significantly greater or lower in European allele frequency than the other loci with European private alleles., Conclusions: Genetic structure of Puerto Rico gAHB differs from mainland AHB populations, probably representing evolutionary processes on the island.

Oceanic islands have reduced resources and natural enemies and potentially affect life history traits of arriving organisms. Among the most spectacular invasions in the Western hemisphere is that of the Africanized honeybee. We hypothesized that in the oceanic island Puerto Rico, Africanized bees will exhibit differences from the mainland population such as for defensiveness and other linked traits. We evaluated the extent of Africanization through three typical Africanized traits: wing size, defensive behavior, and resistance to Varroa destructor mites. All sampled colonies were Africanized by maternal descent, with over 65% presence of European alleles at the S-3 nuclear locus. In two assays evaluating defense, Puerto Rican bees showed low defensiveness similar to European bees. In morphology and resistance to mites, Africanized bees from Puerto Rico are similar to other Africanized bees. In behavioral assays on mechanisms of resistance to Varroa, we directly observed that Puerto Rican Africanized bees groomed-off and bit the mites as been observed in other studies. In no other location, Africanized bees have reduced defensiveness while retaining typical traits such as wing size and mite resistance. This mosaic of traits that has resulted during the invasion of an oceanic island has implications for behavior, evolution, and agriculture.
