BackgroundStable isotope labeling is a promising method for use in insect mark-capture and dispersal studies. Culicoides biting midges, which transmit several important animal pathogens, including bluetongue virus (BTV) and epizootic hemorrhagic disease virus (EHDV), are small flies that develop in various semi-aquatic habitats. Previous Culicoides dispersal studies have suffered from the limitations of other labeling techniques, and an inability to definitively connect collected adult midges to specific immature development sites.ResultsAdult C. sonorensis were successfully labeled with 13C and 15N stable isotopes as larvae developing in a semi-aquatic mud substrate in the laboratory. High and low-dose isotope treatments for both elements significantly enriched midges above the background isotope levels of unenriched controls. Enrichment had no effect on C. sonorensis survival, though a slight (~ 5 day) delay in emergence was observed, and there was no significant effect of pool size on 13C or 15N enrichment levels.ConclusionsStable isotope labeling is life-long, and does not interfere with natural insect behaviors. Stable isotope enrichment using 13C or 15N shows promise for Culicoides dispersal studies in the field. This method can be used to identify adult dispersal from larval source habitat where a midge developed. It may be possible to detect a single enriched midge in a pool of unenriched individuals, though further testing is needed to confirm the sensitivity of this method.

Differences in radiocarbon apparent ages between coexisting planktonic and benthic (B-P) foraminifers in the Santa Barbara Basin contain information about changes in the age (and thus the source) of water entering the basin. The Holocene sequence indicates a greater variability in circulation than previously detected. During the early Holocene, ca. 9 ka, B-P [sup.14]C apparent age differences fluctuated between 220 and 560 [sup.14]C yr, similar to rapid variations seen during the Younger Dryas and the last glacial episode. During the relatively stable middle Holocene (8.5-2 ka), B-P [sup.14]C apparent age differences averaged 440 [sup.14]C yr. After 2 ka, the average B-P [sup.14]C apparent age difference increased to 620 [sup.14]C yr, which has persisted until the present. The shift in B-P [sup.14]C apparent age differences likely represents a change in the strength or source of North Pacific Intermediate Water production and is concurrent with changes in regional climate patterns as well as with changes in temperature and accumulation rates measured in the Greenland Ice Sheet Project 2 ice core, suggesting large-scale changes in atmospheric circulation patterns. Deep-sea corals were discovered over a century ago, but it is only over recent years that focused efforts have been made to explore the history of the oceans using the geochemistry of their skeletal remains. They offer a promising archive of past oceanic environments given their global distribution, layered growth patterns, longevity and preservation as well as our ability to date them using radiometric techniques. This paper provides an overview of the current state-of-the-art in terms of geochemical approaches to using deep-sea coral skeletons to explore the history of the ocean. Deep-sea coral skeletons have a wide array of morphologies (e.g. solitary cup corals, branching colonial corals) and materials (calcite, aragonite and proteins). As such their biomineralization strategies are diverse, leading to complex geochemistry within coral skeletons. Notwithstanding these complications, progress has been made on developing methods for reconstructing the oceanographic environment in the past using trace elements and isotopic methods. Promising approaches within certain coral groups include clumped isotopes and Mg/Li for temperature reconstructions, boron isotopes and radiocarbon for carbon cycling, εNd, and radiocarbon for circulation studies and δ15N, P/Ca and Ba/Ca for nutrient tracer studies. Likewise there is now a range of techniques for dating deep-sea corals skeletons (e.g. U-series, radiocarbon), and determining their growth rates (e.g. radiocarbon and 210Pb). Dating studies on historic coral populations in the Atlantic, Southern Ocean and Pacific point to climate and environmental changes being dominant controls on coral populations over millennial and orbital timescales. This paper provides a review of a range of successes and promising approaches. It also highlights areas in which further research would likely provide new insights into biomineralization, palaeoceanography and distribution of past coral populations. [ABSTRACT FROM AUTHOR]

The use of stable isotope enrichment to mark mosquitoes has provided a tool to study the biology of vector species. In this study, we evaluated isotopic marking of Aedes aegypti (L.) (Diptera: Culicidae) in a laboratory setting. We determined the optimal dosage for marking adult Ae. aegypti mosquitoes with 13C and 15N. Additionally, Ae. aegypti mosquitoes were single and dually marked with 13C and 15N for up to 60 d postemergence without changes to adult body size or transgenerational marking.
