Abstract: Observations of the millimeter sky contain valuable information on a number of signals, including the blackbody cosmic microwave background (CMB), Galactic emissions, and the Compton-$y$ distortion due to the thermal Sunyaev-Zel'dovich (tSZ) effect. Extracting new insight into cosmological and astrophysical questions often requires combining multi-wavelength observations to spectrally isolate one component. In this work, we present a new arcminute-resolution Compton-$y$ map, which traces out the line-of-sight-integrated electron pressure, as well as maps of the CMB in intensity and E-mode polarization, across a third of the sky (around 13,000 sq.~deg.). We produce these through a joint analysis of data from the Atacama Cosmology Telescope (ACT) Data Release 4 and 6 at frequencies of roughly 93, 148, and 225 GHz, together with data from the \textit{Planck} satellite at frequencies between 30 GHz and 545 GHz. We present detailed verification of an internal linear combination pipeline implemented in a needlet frame that allows us to efficiently suppress Galactic contamination and account for spatial variations in the ACT instrument noise. These maps provide a significant advance, in noise levels and resolution, over the existing \textit{Planck} component-separated maps and will enable a host of science goals including studies of cluster and galaxy astrophysics, inferences of the cosmic velocity field, primordial non-Gaussianity searches, and gravitational lensing reconstruction of the CMB., Comment: The Compton-y map and associated products will be made publicly available upon publication of the paper. 2. Attention Deficit Hyperactivity Disorder: Is There an App for That? Suitability Assessment of Apps for Children and Young People With ADHD

Abstract: BackgroundAttention-deficit/hyperactivity disorder (ADHD) is a complex highly comorbid disorder, which can have a huge impact on those with ADHD, their family, and the community around them. ADHD is currently managed using pharmacological and nonpharmacological interventions. However, with advances in technology and an increase in the use of mobile apps, managing ADHD can be augmented using apps specifically designed for this population. However, little is known regarding the suitability and usability of currently available apps. ObjectiveThe aim of this study was to explore the suitability of the top 10 listed apps for children and young people with ADHD and clinicians who work with them. It is hypothesized that mobile apps designed for this population could be more suitably designed for this population. MethodsThe top 10 listed apps that are specifically targeted toward children and young people with ADHD in the United Kingdom were identified via the Google Play (n=5) and iTunes store (n=5). Interviews were then undertaken with 5 clinicians who specialize in treating this population and 5 children and young people with ADHD themselves, to explore their opinions of the 10 apps identified and what they believe the key components are for apps to be suitable for this population. ResultsFive themes emerged from clinician and young people interviews: the accessibility of the technology, the importance of relating to apps, addressing ADHD symptoms and related difficulties, age appropriateness, and app interaction. Three additional themes emerged from the clinician interviews alone: monitoring symptoms, side effects and app effect on relationships, and the impact of common comorbid conditions. The characteristics of the apps did not appear to match well with the views of our sample. ConclusionsThese findings suggest that the apps may not be suitable in meeting the complex needs associated with this condition. Further research is required to explore the value of apps for children and young people with ADHD and their families and, in particular, any positive role for apps in the management of ADHD in this age group. A systematic review on how technology can be used to engage this population and how it can be used to help them would be a useful way forward. This could be the platform to begin exploring the use of apps further.

Abstract: We present weak gravitational lensing measurements of a sample of 157 clusters within the Kilo Degree Survey (KiDS), detected with a $>5\sigma$ thermal Sunyaev-Zel'dovich (SZ) signal by the Atacama Cosmology Telescope (ACT). Using a halo-model approach we constrain the average total cluster mass, $M_{\rm WL}$, accounting for the ACT cluster selection function of the full sample. We find that the SZ cluster mass estimate $M_{\rm SZ}$, which was calibrated using X-ray observations, is biased with $M_{\rm SZ}/M_{\rm WL} = (1-b_{\rm SZ}) = 0.65\pm 0.05$. Separating the sample into six mass bins, we find no evidence of a strong mass-dependency for the mass bias, $(1-b_{\rm SZ})$. Adopting this ACT-KiDS SZ mass-calibration would bring the Planck SZ cluster count into agreement with the counts expected from the {\it Planck} cosmic microwave background $\Lambda$CDM cosmological model, although it should be noted that the cluster sample considered in this work has a lower average mass $M_{\rm SZ, uncor} = 3.64 \times 10^{14} M_{\odot}$ compared to the Planck cluster sample which has an average mass in the range $M_{\rm SZ, uncor} = (5.5-8.5) \times 10^{14} M_{\odot}$, depending on the sub-sample used., Comment: 12 pages, 7 figures

Abstract: We present cosmological constraints from a gravitational lensing mass map covering 9400 sq. deg. reconstructed from CMB measurements made by the Atacama Cosmology Telescope (ACT) from 2017 to 2021. In combination with BAO measurements (from SDSS and 6dF), we obtain the amplitude of matter fluctuations $\sigma_8 = 0.819 \pm 0.015$ at 1.8% precision, $S_8\equiv\sigma_8({\Omega_{\rm m}}/0.3)^{0.5}=0.840\pm0.028$ and the Hubble constant $H_0= (68.3 \pm 1.1)\, \text{km}\,\text{s}^{-1}\,\text{Mpc}^{-1}$ at 1.6% precision. A joint constraint with CMB lensing measured by the Planck satellite yields even more precise values: $\sigma_8 = 0.812 \pm 0.013$, $S_8\equiv\sigma_8({\Omega_{\rm m}}/0.3)^{0.5}=0.831\pm0.023$ and $H_0= (68.1 \pm 1.0)\, \text{km}\,\text{s}^{-1}\,\text{Mpc}^{-1}$. These measurements agree well with $\Lambda$CDM-model extrapolations from the CMB anisotropies measured by Planck. To compare these constraints to those from the KiDS, DES, and HSC galaxy surveys, we revisit those data sets with a uniform set of assumptions, and find $S_8$ from all three surveys are lower than that from ACT+Planck lensing by varying levels ranging from 1.7-2.1$\sigma$. These results motivate further measurements and comparison, not just between the CMB anisotropies and galaxy lensing, but also between CMB lensing probing $z\sim 0.5-5$ on mostly-linear scales and galaxy lensing at $z\sim 0.5$ on smaller scales. We combine our CMB lensing measurements with CMB anisotropies to constrain extensions of $\Lambda$CDM, limiting the sum of the neutrino masses to $\sum m_{\nu} < 0.13$ eV (95% c.l.), for example. Our results provide independent confirmation that the universe is spatially flat, conforms with general relativity, and is described remarkably well by the $\Lambda$CDM model, while paving a promising path for neutrino physics with gravitational lensing from upcoming ground-based CMB surveys., Comment: 32 pages, 17 figures, replaced with version accepted in ApJ (Feb 2024). Abstract: We present new measurements of cosmic microwave background (CMB) lensing over $9400$ sq. deg. of the sky. These lensing measurements are derived from the Atacama Cosmology Telescope (ACT) Data Release 6 (DR6) CMB dataset, which consists of five seasons of ACT CMB temperature and polarization observations. We determine the amplitude of the CMB lensing power spectrum at $2.3\%$ precision ($43\sigma$ significance) using a novel pipeline that minimizes sensitivity to foregrounds and to noise properties. To ensure our results are robust, we analyze an extensive set of null tests, consistency tests, and systematic error estimates and employ a blinded analysis framework. The baseline spectrum is well fit by a lensing amplitude of $A_{\mathrm{lens}}=1.013\pm0.023$ relative to the Planck 2018 CMB power spectra best-fit $\Lambda$CDM model and $A_{\mathrm{lens}}=1.005\pm0.023$ relative to the $\text{ACT DR4} + \text{WMAP}$ best-fit model. From our lensing power spectrum measurement, we derive constraints on the parameter combination $S^{\mathrm{CMBL}}_8 \equiv \sigma_8 \left({\Omega_m}/{0.3}\right)^{0.25}$ of $S^{\mathrm{CMBL}}_8= 0.818\pm0.022$ from ACT DR6 CMB lensing alone and $S^{\mathrm{CMBL}}_8= 0.813\pm0.018$ when combining ACT DR6 and Planck NPIPE CMB lensing power spectra. These results are in excellent agreement with $\Lambda$CDM model constraints from Planck or $\text{ACT DR4} + \text{WMAP}$ CMB power spectrum measurements. Our lensing measurements from redshifts $z\sim0.5$--$5$ are thus fully consistent with $\Lambda$CDM structure growth predictions based on CMB anisotropies probing primarily $z\sim1100$. We find no evidence for a suppression of the amplitude of cosmic structure at low redshifts, Comment: 45+22 pages, 50 figures. v2 matches with published version in ApJ. Abstract: Cosmological data in the next decade will be characterized by high-precision, multi-wavelength measurements of thousands of square degrees of the same patches of sky. By performing multi-survey analyses that harness the correlated nature of these datasets, we will gain access to new science, and increase the precision and robustness of science being pursued by each individual survey. However, effective application of such analyses requires a qualitatively new level of investment in cross-survey infrastructure, including simulations, associated modeling, coordination of data sharing, and survey strategy. Abstract: We introduce and validate a delensing framework for the Simons Observatory (SO), which will be used to improve constraints on inflationary gravitational waves (IGWs) by reducing the lensing noise in measurements of the $B$-modes in CMB polarization. SO will initially observe CMB by using three small aperture telescopes and one large-aperture telescope. While polarization maps from small-aperture telescopes will be used to constrain IGWs, the internal CMB lensing maps used to delens will be reconstructed from data from the large-aperture telescope. Since lensing maps obtained from the SO data will be noise-dominated on sub-degree scales, the SO lensing framework constructs a template for lensing-induced $B$-modes by combining internal CMB lensing maps with maps of the cosmic infrared background from Planck as well as galaxy density maps from the LSST survey. We construct a likelihood for constraining the tensor-to-scalar ratio $r$ that contains auto- and cross-spectra between observed $B$-modes and the lensing $B$-mode template. We test our delensing analysis pipeline on map-based simulations containing survey non-idealities, but that, for this initial exploration, does not include contamination from Galactic and extragalactic foregrounds. We find that the SO survey masking and inhomogeneous and atmospheric noise have very little impact on the delensing performance, and the $r$ constraint becomes $\sigma(r)\approx 0.0015$ which is close to that obtained from the idealized forecasts in the absence of the Galactic foreground and is nearly a factor of two tighter than without delensing. Abstract: We use gravitational lensing of the cosmic microwave background (CMB) to measure the mass of the most distant blindly-selected sample of galaxy clusters on which a lensing measurement has been performed to date. In CMB data from the the Atacama Cosmology Telescope (ACT) and the Planck satellite, we detect the stacked lensing effect from 677 near-infrared-selected galaxy clusters from the Massive and Distant Clusters of WISE Survey (MaDCoWS), which have a mean redshift of $ \langle z \rangle = 1.08$. There are no current optical weak lensing measurements of clusters that match the distance and average mass of this sample. We detect the lensing signal with a significance of $4.2 \sigma$. We model the signal with a halo model framework to find the mean mass of the population from which these clusters are drawn. Assuming that the clusters follow Navarro-Frenk-White density profiles, we infer a mean mass of $\langle M_{500c}\rangle = \left(1.7 \pm 0.4 \right)\times10^{14}\,\mathrm{M}_\odot$. We consider systematic uncertainties from cluster redshift errors, centering errors, and the shape of the NFW profile. These are all smaller than 30% of our reported uncertainty. Abstract: We present new arcminute-resolution maps of the Cosmic Microwave Background temperature and polarization anisotropy from the Atacama Cosmology Telescope, using data taken from 2013-2016 at 98 and 150 GHz. The maps cover more than 17,000 deg$^2$, the deepest 600 deg$^2$ with noise levels below 10 $\mu$K-arcmin. We use the power spectrum derived from almost 6,000 deg$^2$ of these maps to constrain cosmology. The ACT data enable a measurement of the angular scale of features in both the divergence-like polarization and the temperature anisotropy, tracing both the velocity and density at last-scattering. From these one can derive the distance to the last-scattering surface and thus infer the local expansion rate, $H_0$. By combining ACT data with large-scale information from WMAP we measure $H_0 = 67.6 \pm 1.1$ km/s/Mpc, at 68% confidence, in excellent agreement with the independently-measured Planck satellite estimate (from ACT alone we find $H_0 = 67.9 \pm 1.5$ km/s/Mpc). The $\Lambda$CDM model provides a good fit to the ACT data, and we find no evidence for deviations: both the spatial curvature, and the departure from the standard lensing signal in the spectrum, are zero to within 1$\sigma$; the number of relativistic species, the primordial Helium fraction, and the running of the spectral index are consistent with $\Lambda$CDM predictions to within $1.5 - 2.2\sigma$. We compare ACT, WMAP, and Planck at the parameter level and find good consistency; we investigate how the constraints on the correlated spectral index and baryon density parameters readjust when adding CMB large-scale information that ACT does not measure. Abstract: We present the temperature and polarization angular power spectra of the CMB measured by the Atacama Cosmology Telescope (ACT) from 5400 deg$^2$ of the 2013-2016 survey, which covers $>$15000 deg$^2$ at 98 and 150 GHz. For this analysis we adopt a blinding strategy to help avoid confirmation bias and, related to this, show numerous checks for systematic error done before unblinding. Using the likelihood for the cosmological analysis we constrain secondary sources of anisotropy and foreground emission, and derive a "CMB-only" spectrum that extends to $\ell=4000$. At large angular scales, foreground emission at 150 GHz is $\sim$1% of TT and EE within our selected regions and consistent with that found by Planck. Using the same likelihood, we obtain the cosmological parameters for $\Lambda$CDM for the ACT data alone with a prior on the optical depth of $\tau=0.065\pm0.015$. $\Lambda$CDM is a good fit. The best-fit model has a reduced $\chi^2$ of 1.07 (PTE=0.07) with $H_0=67.9\pm1.5$ km/s/Mpc. We show that the lensing BB signal is consistent with $\Lambda$CDM and limit the celestial EB polarization angle to $\psi_P =-0.07^{\circ}\pm0.09^{\circ}$. We directly cross correlate ACT with Planck and observe generally good agreement but with some discrepancies in TE. Abstract: We construct cosmic microwave background lensing mass maps using data from the 2014 and 2015 seasons of observations with the Atacama Cosmology Telescope (ACT). These maps cover 2100 square degrees of sky and overlap with a wide variety of optical surveys. The maps are signal dominated on large scales and have fidelity such that their correlation with the cosmic infrared background is clearly visible by eye. We also create lensing maps with thermal Sunyaev-Zel'dovich contamination removed using a novel cleaning procedure that only slightly degrades the lensing signal-to-noise ratio. The cross-spectrum between the cleaned lensing map and the BOSS CMASS galaxy sample is detected at $10$-$\sigma$ significance, with an amplitude of $A=1.02 \pm 0.10$ relative to the Planck best-fit LCDM cosmological model with fiducial linear galaxy bias. Abstract: We present new constraints on anisotropic birefringence of the cosmic microwave background polarization using two seasons of data from the Atacama Cosmology Telescope covering $456$ square degrees of sky. The birefringence power spectrum, measured using a curved-sky quadratic estimator, is consistent with zero. Our results provide the tightest current constraint on birefringence over a range of angular scales between $5$ arcminutes and $9$ degrees. We improve previous upper limits on the amplitude of a scale-invariant birefringence power spectrum by a factor of between $2$ and $3$. Assuming a nearly-massless axion field during inflation, our result is equivalent to a $2\,\sigma$ upper limit on the Chern-Simons coupling constant between axions and photons of $g_{\alpha\gamma}<4.0\times 10^{-2}/H_I$ where $H_I$ is the inflationary Hubble scale., Comment: 18 pages, 3 figures, Accepted for publication in PRD

Abstract: We measured the cross-correlation between galaxy weak lensing data from the Kilo Degree Survey (KiDS-1000, DR4) and cosmic microwave background (CMB) lensing data from the Atacama Cosmology Telescope (ACT, DR4) and the Planck Legacy survey. We used two samples of source galaxies, selected with photometric redshifts, (0.1 < zB < 1.2) and (1.2 < zB < 2), which produce a combined detection significance of the CMB lensing and weak galaxy lensing cross-spectrum of 7.7σ. With the lower redshift galaxy sample, for which the cross-correlation was detected at a significance of 5.3σ, we present joint cosmological constraints on the matter density parameter, ωm, and the matter fluctuation amplitude parameter, σ8, marginalising over three nuisance parameters that model our uncertainty in the redshift and shear calibration as well as the intrinsic alignment of galaxies. We find our measurement to be consistent with the best-fitting flat ΛCDM cosmological models from both Planck and KiDS-1000. We demonstrate the capacity of CMB weak lensing cross-correlations to set constraints on either the redshift or shear calibration by analysing a previously unused high-redshift KiDS galaxy sample (1.2 < zB < 2), with the cross-correlation detected at a significance of 7σ. This analysis provides an independent assessment for the accuracy of redshift measurements in a regime that is challenging to calibrate directly owing to known incompleteness in spectroscopic surveys.

Abstract: Optimal analyses of many signals in the cosmic microwave background (CMB) require map-level extraction of individual components in the microwave sky, rather than measurements at the power spectrum level alone. To date, nearly all map-level component separation in CMB analyses has been performed exclusively using satellite data. In this paper, we implement a component separation method based on the internal linear combination (ILC) approach which we have designed to optimally account for the anisotropic noise (in the 2D Fourier domain) often found in ground-based CMB experiments. Using this method, we combine multi-frequency data from the Planck satellite and the Atacama Cosmology Telescope Polarimeter (ACTPol) to construct the first wide-area, arcminute-resolution component-separated maps (covering approximately 2100 sq. deg.) of the CMB temperature anisotropy and the thermal Sunyaev-Zel'dovich (tSZ) effect sourced by the inverse-Compton scattering of CMB photons off hot, ionized gas. Our ILC pipeline allows for explicit deprojection of various contaminating signals, including a modified blackbody approximation of the cosmic infrared background (CIB) spectral energy distribution. The cleaned CMB maps will be a useful resource for CMB lensing reconstruction, kinematic SZ cross-correlations, and primordial non-Gaussianity studies. The tSZ maps will be used to study the pressure profiles of galaxies, groups, and clusters through cross-correlations with halo catalogs, with dust contamination controlled via CIB deprojection. Abstract: We present a weak lensing detection of filamentary structures in the cosmic web, combining data from the Kilo-Degree Survey, the Red Cluster Sequence Lensing Survey and the Canada-France-Hawaii Telescope Lensing Survey. The line connecting luminous red galaxies with a separation of $3 - 5\, h^{-1}\text{Mpc}$ is chosen as a proxy for the location of filaments. We measure the average weak lensing shear around $\sim$11,000 candidate filaments selected in this way from the Sloan Digital Sky Survey. After nulling the shear induced by the dark matter haloes around each galaxy, we report a $3.4\,\sigma$ detection of an anisotropic shear signal from the matter that connects them. Abstract: We use gravitational lensing of the cosmic microwave background (CMB) to measure the mass of the most distant blindly selected sample of galaxy clusters on which a lensing measurement has been performed to date. In CMB data from the the Atacama Cosmology Telescope and the Planck satellite, we detect the stacked lensing effect from 677 near-infrared-selected galaxy clusters from the Massive and Distant Clusters of WISE Survey (MaDCoWS), which have a mean redshift of zñ = 1.08. There are currently no representative optical weak lensing measurements of clusters that match the distance and average mass of this sample. We detect the lensing signal with a significance of 4.2s. We model the signal with a halo model framework to find the mean mass of the population from which these clusters are drawn. Assuming that the clusters follow Navarro–Frenk–White (NFW) density profiles, we infer a mean mass of M500cñ = (1.7 + 0.4) ´ 1014 M*. We consider systematic uncertainties from cluster redshift errors, centering errors, and the shape of the NFW profile. These are all smaller than 30% of our reported uncertainty. This work highlights the potential of CMB lensing to enable cosmological constraints from the abundance of distant clusters populating ever larger volumes of the observable universe, beyond the capabilities of optical weak lensing measurements.

21. Observing the dark universe with weak gravitational lensing

Abstract: We are now entering into an era of huge cosmological data sets, with observations testing the current paradigm to high accuracy. By combining different probes of cosmology, we can access new information about our Universe and investigate the systematic effects that plague our ability to measure cosmology accurately. In this thesis we use weak gravitational lensing as a tool to measure the large scale structure in the Universe. First, we present the theoretical framework in which this thesis is set. We then detail the methodology required to measure the cross-correlation between galaxy weak lensing and cosmic microwave background (CMB) weak lensing data sets. In particular we highlight the impact of the complicated nature of galaxy weak lensing data and investigate ways to improve the signal-to-noise of the cross-correlation measurement using simulations. We measure the CMB lensing/galaxy weak lensing power spectrum using the latest CMB lensing data from the Atacama Cosmology Telescope (ACT) and galaxy weak lensing data from the Kilo Degree Survey (KiDS), making a detection at 5σ significance. We compare our measured cross-spectrum to the cosmological parameters inferred from the Planck and KiDS experiments. We then present fits for the mat- ter density and the amplitude of matter fluctuations, marginalising over the galaxy weak lensing systematic biases. We forecast the expected signal-to-noise using the next data release from ACT and use simulated fields to investigate the impact on estimating cosmological parameters with that data set. Finally, we measure the masses of clusters selected from ACT CMB data via the Sunyaev-Zel’dovich effect using weak gravitational lensing data from KiDS, making a 20σ detection of the lensing signal. We present an estimate of the mass bias for a set of stacked clusters as well as the highest signal-to-noise individual clusters. We also describe tests of systematic effects and analysis choices.

Abstract: We present weak-lensing measurements using the first-year data from the Hyper Suprime-Cam Strategic Survey Program on the Subaru telescope for eight galaxy clusters selected through their thermal Sunyaev-Zel'dovich (SZ) signal measured at 148 GHz with the Atacama Cosmology Telescope Polarimeter experiment. The overlap between the two surveys in this work is 33.8 square degrees, before masking bright stars. The signal-to-noise ratio of individual cluster lensing measurements ranges from 2.2 to 8.7, with a total of 11.1 for the stacked cluster weak-lensing signal. We fit for an average weak-lensing mass distribution using three different profiles, a Navarro-Frenk-White profile, a dark-matter-only emulated profile, and a full cosmological hydrodynamic emulated profile. We interpret the differences among the masses inferred by these models as a systematic error of 10\%, which is currently smaller than the statistical error. We obtain the ratio of the SZ-estimated mass to the lensing-estimated mass (the so-called hydrostatic mass bias $1-b$) of $0.74^{+0.13}_{-0.12}$, which is comparable to previous SZ-selected clusters from the Atacama Cosmology Telescope and from the {\sl Planck} Satellite. Abstract: We present the tomographic cross-correlation between galaxy lensing measured in the Kilo Degree Survey (KiDS-450) with overlapping lensing measurements of the cosmic microwave background (CMB), as detected by Planck 2015. We compare our joint probe measurement to the theoretical expectation for a flat $\Lambda$CDM cosmology, assuming the best-fitting cosmological parameters from the KiDS-450 cosmic shear and Planck CMB analyses. We find that our results are consistent within $1\sigma$ with the KiDS-450 cosmology, with an amplitude re-scaling parameter $A_{\rm KiDS} = 0.86 \pm 0.19$. Adopting a Planck cosmology, we find our results are consistent within $2\sigma$, with $A_{\it Planck} = 0.68 \pm 0.15$. We show that the agreement is improved in both cases when the contamination to the signal by intrinsic galaxy alignments is accounted for, increasing $A$ by $\sim 0.1$. This is the first tomographic analysis of the galaxy lensing -- CMB lensing cross-correlation signal, and is based on five photometric redshift bins. We use this measurement as an independent validation of the multiplicative shear calibration and of the calibrated source redshift distribution at high redshifts. We find that constraints on these two quantities are strongly correlated when obtained from this technique, which should therefore not be considered as a stand-alone competitive calibration tool., Comment: 14 pages, 11 figures

Abstract: We present the cluster mass-richness scaling relation calibrated by a weak lensing analysis of >18000 galaxy cluster candidates in the Canada-France-Hawaii Telescope Lensing Survey (CFHTLenS). Detected using the 3D-Matched-Filter cluster-finder of Milkeraitis et al., these cluster candidates span a wide range of masses, from the small group scale up to $\sim10^{15} M_{\odot}$, and redshifts 0.2 $\lesssim z\lesssim$ 0.9. The total significance of the stacked shear measurement amounts to 54$\sigma$. We compare cluster masses determined using weak lensing shear and magnification, finding the measurements in individual richness bins to yield 1$\sigma$ compatibility, but with magnification estimates biased low. This first direct mass comparison yields important insights for improving the systematics handling of future lensing magnification work. In addition, we confirm analyses that suggest cluster miscentring has an important effect on the observed 3D-MF halo profiles, and we quantify this by fitting for projected cluster centroid offsets, which are typically $\sim$ 0.4 arcmin. We bin the cluster candidates as a function of redshift, finding similar cluster masses and richness across the full range up to $z \sim$ 0.9. We measure the 3D-MF mass-richness scaling relation $M_{200} = M_0 (N_{200} / 20)^\beta$. We find a normalization $M_0 \sim (2.7^{+0.5}_{-0.4}) \times 10^{13} M_{\odot}$, and a logarithmic slope of $\beta \sim 1.4 \pm 0.1$, both of which are in 1$\sigma$ agreement with results from the magnification analysis. We find no evidence for a redshift-dependence of the normalization. Abstract: Isoform selective histone deacetylase (HDAC) inhibitors are urgently required to investigate HDAC isoform phenotypes and provide targeted therapeutics. Class 1 HDACs (1, 2 and 3) are recruited into large multi-subunit co-repressor complexes for maximal activity. However, little attention has been paid to targeting the disruption of these protein-protein interactions (PPIs) and so represents a novel approach to develop more specific therapeutics and chemical probes. This thesis describes the design, synthesis and evaluation of stapled peptide analogues of the SMRT-DAD protein. A systematic, structure based approach was taken to design the second generation stapled peptides containing phosphotyrosine residues. These were shown to be super activators of the HDAC3:SMRT-DAD corepressor complex. The synthesis of a hydroxamic acid amino acid is also described and an improvement on the current synthetic scheme is discussed. A novel hydroxamic acid containing histone H4 tail peptide was then prepared which was shown to be an inhibitor of the HDAC1:MTA1 corepressor complex with an IC50 of 366 nM. A crystal structure of this inhibitor peptide bound in the active site of HDAC1 was also obtained. The challenging synthesis of a series of histone H3 (1-21) tail sequence peptides has also been described. NMR experiments were carried out using these peptides to study both the demethylase and deacetylase activities of the ternary LSD1:CoREST:HDAC1. The deacetylase activity of HDAC1 has been shown to be much faster than the demethylase activity of LSD1. In order to obtain a crystal structure of the ternary LSD1:CoREST:HDAC1 complex and to determine the distance between the LSD1 and the HDAC1 components, three dual peptide inhibitors based on SNAIL have been designed and synthesised. These crystallisation experiments and biological studies are in progress.

Abstract: Targeting the lysine deacetylase activity of class I histone deacetylases (HDACs) is potentially beneficial for the treatment of several diseases including human immunodeficiency virus (HIV) infection, Alzheimer's disease, and various cancers. It is therefore important to understand the function and mechanism of action of these enzymes. Class I HDACs act as catalytic components of seven large, multiprotein corepressor complexes. Different HDAC corepressor complexes have specific, nonredundant roles in the cell. It is likely that their specific functions are at least partly influenced by the substrate specificity of the complexes. To address this, we developed chemical tools to probe the specificity of HDAC complexes. We assessed a library of acetyl-lysine-containing substrate peptides and hydroxamic acid-containing inhibitor peptides against the full range of class I HDAC corepressor complexes. The results suggest that site-specific HDAC corepressor complex activity is driven in part by the recognition of the primary amino acid sequence surrounding a particular lysine position in the histone tail.

Abstract: We report a novel divinyltriazine linker for the stapling of two cysteine residues to form macrocyclic peptides from their unprotected linear counterparts. The stapling reaction occurred rapidly under mild conditions on a range of unprotected peptide sequences. The resulting constrained peptides displayed greater stability in a serum stability assay when compared to their linear counterparts.

A new class of compounds called stapled peptides have been developed that mimicα-helix secondary structures involved in PPIs. These conformationally constrained peptides also show favorable physicochemical properties and so are promising lead compounds for drug discovery. This review focuses on the design aspects of hydrocarbon constrainedα-helical proteomimetics and provides examples in which they target biologically relevant PPIs.
