Pesquisa do Estado do Amazonas This study presents possibilities of pedagogical practices that favor the students' understanding of the environmental impacts in the production of solid waste. This ability to understand is amplified when it addresses the discussions and in the study room of the global context but does not study the expansion to the local reality of teaching. In this sense, the present work aims to stimulate a critical-reflexive attitude of the students of the Education of Young and Adults (EJA) about the impacts generated by the production and destination of Solid Waste (RS) in the environment. To this end, this research had as target audience, students of the Youth and Adult Education (EJA) modality, from a public school, located in the Abial neighborhood, Municipality of Tef??/AM. The action was studied as a research-logical methodological approach, through the participation of teacher-students in order to achieve learning. In this sense, as activities developed, their activities provide moments of educational dialogue, moments of activation in each, establishing a dialogic-reflective thought. Therefore, it was necessary to contextualize the problem of solid waste with the reality experienced by the students, relating it to social, economic, and environmental issues. How data collection techniques and instruments were applied and photos. From the analysis of the results, it was possible to observe that the pedagogical activities stimulate a critical-reflective attitude of the students regarding the production and disposal of solid waste. In a sustainable sense, political societies are public policies that guarantee favorable conditions for education and generally dispose of waste, such as selective garbage collection. Este estudo apresenta possibilidades de pr??ticas pedag??gicas que favorecem a compreens??o dos educandos quanto aos impactos ambientais na produ????o dos res??duos s??lidos. Essa capacidade de compreens??o ?? ampliada quando h?? discuss??es e reflex??es em sala de aula abordando o contexto global, mas n??o deixando de trazer para a realidade local do educando. Nesse sentido, o presente trabalho tem como objetivo estimular a atitude cr??tica-reflexiva dos educandos da Educa????o de Jovens e Adultos (EJA) acerca dos impactos causados pela produ????o e destina????o de Res??duos S??lidos (RS) no ambiente. Para tal, esta pesquisa teve como p??blico-alvo, educandos da modalidade de Educa????o de Jovens e Adultos (EJA), de uma escola p??blica, localizada no bairro Abial, Munic??pio de Tef??/AM. A pesquisa-a????o foi adotada como abordagem metodol??gica, caracterizada pela participa????o de professora-alunos a fim de alcan??ar um aprendizado pautado na pr??tica cr??tico-reflexiva. Nesse sentido, as atividades desenvolvidas propiciaram momentos de di??logos, participa????o ativa dos educandos, momentos de expressarem suas ideias em cada atividade, estabelecendo um pensamento dial??gico-reflexivo. Para tanto, fez-se necess??rio contextualizar a problem??tica dos res??duos s??lidos com a realidade vivenciada pelos educandos, relacionando ?? quest??o social, econ??mica e ambiental. Como t??cnicas e instrumentos para a coleta de dados foram aplicados: question??rio semiestruturado, discuss??es coletivas, observa????es e registros ??udios visuais: filmagens e fotos. 13. La posición de siembra y la aplicación de Azospirillum brasilense influyen en los caracteres agronómicos de la yuca

The management of the cassava crop is what determines the productive potential, being in many cases neglected, as it is considered a rustic crop that responds poorly to agricultural inputs. The objective of this work was to evaluate the development of cassava as a function of the position of the manioc in combination with Azospirillum brasilense. The experimental design used was completely randomized, with a 2x2 factorial design, one factor being the positioning [horizontal (PH) and vertical (PV)] and the other the application of A. brasilense [with (+Azos) and without (-Azos)], with 20 replicates per treatment. At 165 days after planting (DAP) the variables: stem diameter (SD), number of shoots (NS), main shoot height (MSH) and leaflet length (LL) were evaluated at an interval of 30 days. At 285 DAP in the final collection, the weight of the part area (WPA), the number of commercial roots (NCR) and the total production (TP) were evaluated. Multivariate principal component (PC) analyzes were performed. The first two PC retain >88% of the data variability on most of the measured variables. Morphological development is stimulated from 165 DAP to 285 DAP, thus generating the highest productivity of cassava in the association of PH+Azos. The horizontal planting position is the best option in the cassava crop, and together with the fixation and better availability of N by A. brasilense, they promote the best performance of the crop. El manejo del cultivo de yuca es lo que determina el potencial productivo, siendo en muchos casos descuidado, por considerarse un cultivo rústica que responde mal los insumos agrícolas. El objetivo de este trabajo fue evaluar el desarrollo de la yuca en función de la posición de la yuca en combinación con Azospirillum brasilense. El diseño experimental utilizado fue completamente al azar, con un diseño factorial 2x2, siendo un factor el posicionamiento [horizontal (PH) y vertical (PV)] y el otro la aplicación de A. brasilense [con (+Azos) y sin (-Azos)], con 20 repeticiones por tratamiento. A los 165 días después de la siembra (DDS) se evaluaron las variables: diámetro del tallo (DT), número de brotes (NB), altura del brote principal (ABP) y longitud del folíolo (LF) con un intervalo de 30 días. A los 285 DDS en la colecta final se evaluó el peso de la parte área (PPA), el número de raíces comerciales (NRC) y la producción total (PT). Se realizaron análisis multivariados de componentes principales (PC). Los dos primeros CP conservan >88 % de la variabilidad de los datos en la mayoría de las variables medidas. Se estimula el desarrollo morfológico desde los 165 DDS hasta los 285 DDS, generando así la mayor productividad de yuca en la asociación de PH+Azos. La posición de siembra horizontal es la mejor opción en el cultivo de yuca, y junto con la fijación y mejor disponibilidad de N por parte de A. brasilense, promueven el mejor comportamiento del cultivo. O manejo da cultura da mandioca é o que determina o potencial produtivo, sendo em muitos casos negligenciado, por ser considerada uma cultura rústica que responde pouco aos insumos agrícolas. O objetivo deste trabalho foi avaliar o desenvolvimento da mandioca em função da posição da maniva em combinação com Azospirillum brasilense. O delineamento experimental utilizado foi inteiramente casualizado, com desenho fatorial 2x2, sendo um fator o posicionamento [horizontal (PH) e vertical (PV)] e outro a aplicação do A. brasilense [com (+Azos) e sem (-Azos)], com 20 repetições por tratamento. Aos 165 dias após plantio (DAP) foram avaliadas em intervalo de 30 dias as variáveis: diâmetro do caule (DC), número de brotos (NB), altura do broto principal (ABP) e comprimento de folíolos (CF). Aos 285 DAP na coleta final foi avaliado o peso da parte área (PPA), o número de raízes comerciais (NR) e a produção total (PT). Análises multivariada de componentes principais (CP) foi realizado. Os primeiros dois CP retem > 88% da variabilidade dos dados na maioria das variáveis mesuradas. O desenvolvimento morfológico é estimulado a partir dos 165 DAP até os 285 DAP, gerando assim, a maior produtividade da mandioca na associação do PH+Azos. A posição horizontal de plantio é a melhor opção na cultura da mandioca e junto com a fixação e melhor disponibilidade de N pelo A. brasilense promovem o melhor desempenho da cultura.

RESUMO Objetivo: Descrever o IMPACTO-MR, um estudo brasileiro de plataforma nacional em unidades de terapia intensiva focado no impacto das infecções por bactérias multirresistentes relacionadas à assistência à saúde. Métodos: Descrevemos a plataforma IMPACTO-MR, seu desenvolvimento, critérios para seleção das unidades de terapia intensiva, caracterização da coleta de dados, objetivos e projetos de pesquisa futuros a serem realizados na plataforma. Resultados: Os dados principais foram coletados por meio do Epimed Monitor System® e consistiram em dados demográficos, dados de comorbidades, estado funcional, escores clínicos, diagnóstico de internação e diagnósticos secundários, dados laboratoriais, clínicos e microbiológicos e suporte de órgãos durante a internação na unidade de terapia intensiva, entre outros. De outubro de 2019 a dezembro de 2020, 33.983 pacientes de 51 unidades de terapia intensiva foram incluídos no banco de dados principal. Conclusão: A plataforma IMPACTO-MR é um banco de dados clínico brasileiro de unidades de terapia intensiva focado na pesquisa do impacto das infecções por bactérias multirresistentes relacionadas à assistência à saúde. Essa plataforma fornece dados para o desenvolvimento e pesquisa de unidades de terapia intensiva individuais e ensaios clínicos observacionais e prospectivos multicêntricos. ABSTRACT Objective: To describe the IMPACTO-MR, a Brazilian nationwide intensive care unit platform study focused on the impact of health care-associated infections due to multidrug-resistant bacteria. Methods: We described the IMPACTO-MR platform, its development, criteria for intensive care unit selection, characterization of core data collection, objectives, and future research projects to be held within the platform. Results: The core data were collected using the Epimed Monitor System® and consisted of demographic data, comorbidity data, functional status, clinical scores, admission diagnosis and secondary diagnoses, laboratory, clinical, and microbiological data, and organ support during intensive care unit stay, among others. From October 2019 to December 2020, 33,983 patients from 51 intensive care units were included in the core database. Conclusion: The IMPACTO-MR platform is a nationwide Brazilian intensive care unit clinical database focused on researching the impact of health care-associated infections due to multidrug-resistant bacteria. This platform provides data for individual intensive care unit development and research and multicenter observational and prospective trials.

Foram selecionadas 245 vacas Holandesas de alta produção leiteira, que não apresentaram em 308 dias de lactação, nenhuma enfermidade como: mastite, deslocamento de abomaso, pneumonia, hipocalcemia, retenção de placenta, metrite, diarreia e cetose, que pudesse interferir nos resultados mensurados. Os dados da pedometria (produção de leite, número de passos, número de vezes que o animal deitou e porcentagem de tempo em que ele permaneceu deitado) foram obtidos diariamente e então, calculada uma média semanal. As afecções podais foram avaliadas e classificadas. A úlcera de sola e a dermatite digital foram as de maior incidência com 64,86% e 75% respectivamente, sendo estas associadas aos fatores de risco como o concreto abrasivo, umidade e sobrecarga de peso sobre os dígitos. Nas variáveis pedométricas, a produção de leite foi considerada neste estudo, um fator de risco para desenvolvimento de enfermidades podais. Houve diferença entre o número de passos e quantidade de vezes que os animais deitaram, sendo os animais com lesão aqueles que apresentaram menor atividade pedométrica. Estas variáveis foram adequadas para a detecção precoce de vacas susceptíveis as lesões podais. This study aimed at the early identification of infectious and secondary laminitis lesions in high production Holstein cows, based on changes in pedometry. A total of 245 dairy cows with no milk production in 308 days of lactation showed no disease such as: mastitis, abomasum displacement, pneumonia, hypocalcemia, placenta retention, metritis, diarrhea and ketosis, which could interfere with the results measured. The pedometer data (milk production, number of steps, number of times the animal lay down and percentage of time in which it remained lying down) were obtained daily and then, calculated a weekly average. The foot conditions were evaluated and classified. Soil ulcer and digital dermatitis were the highest incidence with 64.86% and 75%, respectively, being associated with risk factors such as abrasive concrete, moisture and weight overload on the digits. In the pedometer variables, milk production was considered in this study, a risk factor for the development of pediatric diseases. There was a difference between the number of steps and the number of times the animals lay, and the animals with injuries were those that presented lower pedometer activity. Esta pesquisa investigou a contribuição que o patrimônio cultural edificado exerce na qualidade visual da paisagem urbana a partir da abordagem na área da Percepção Ambiental. Adotou como estudo de caso áreas dos centros históricos de três cidades com diferentes graus de preservação do patrimônio cultural edificado: Piratini, a mais preservada, resultado de legislação urbanística pioneira; São José do Norte, onde o patrimônio cultural edificado foi mais destruído ou descaracterizado e Porto Alegre, onde o patrimônio cultural edificado foi parcialmente preservado. A investigação foi operacionalizada por meio da utilização de métodos qualitativos e quantitativos em duas etapas de investigação. A primeira teve por objetivo reunir subsídios para definir a área de estudo em cada uma das três cidades, e por meio da aplicação da técnica de mapas mentais a usuários dos centros históricos, foram identificadas as imagens fortes positivas e negativas das edificações e espaços urbanos (locais, ruas e praças). A segunda etapa teve como objetivo principal testar as hipóteses desta pesquisa por meio da aplicação de questionários acompanhados de cenas urbanas com diferentes níveis de homogeneidade, escolhidas a partir de critérios estabelecidos a fim de responder os objetivos da investigação. As informações obtidas pelos questionários foram analisadas quantitativamente por meio de freqüências e testes não paramétricos tais como tabulações cruzadas e testes de correlação Spearman. Os resultados obtidos permitiram chegar a conclusões sobre o importante papel que o patrimônio cultural edificado exerce na qualidade visual da paisagem urbana. Esta relevância foi verificada na avaliação da aparência visual das edificações analisadas individualmente pelos seus atributos formais e simbólicos e as suas relações no contexto urbano. Importantes correlações foram encontradas entre a avaliação estética da aparência visual das cenas urbanas e atributos formais de volumetria, fachadas e coberturas. As conclusões desta pesquisa reúnem subsídios relativos às questões da estética urbana, do patrimônio cultural edificado e do planejamento urbano que podem contribuir substancialmente para as políticas públicas nas áreas da preservação do patrimônio cultural, do planejamento urbano e ambiental e na implementação de planos e ações de gestão municipal visando qualificar visualmente a paisagem urbana. The study investigates the aesthetic contributions of historical buildings on the visual quality of the urban historical centers based on the Environmental Perception approach. The research focused on urban scenes of three cities which present different degrees of historical heritage preservation: Piratini, which is the most preserved among them due to existent legislation; São José do Norte, where most of it historical heritage was destroyed or damaged; and Porto Alegre, where historical heritage was partially preserved. The investigation was operationalised through the application of qualitative and quantitative methods, in two stages. On the first stage, the definition of the study area in each city was carried out through the application of mental maps to users of the historical centers, which allowed the identification of strongest positive and negative images. On the second stage, questionnaires were applied in order to verify the research hypotheses. The data gathered through questionnaires was analyzed by means of non parametric statistics, such as qui square test and Spearman correlation tests. Results allowed to conclude about the important role historical buildings play on the visual quality of the urban scene, verified through the evaluation of visual appearance of the buildings individually analyzed according to their formal and symbolic attributes and their relation with the urban context. Important correlations were found between the aesthetic evaluation of the visual appearance of the urban scenes and the formal attributes volume try, facades and roofs. Conclusions provide substantial support to contribute to urban heritage preservation, environmental and urban planning and implementation of plans and actions for the municipal government in order to achieve visual quality in urban areas located in historical centers.

Considering that resilience is the result of a process that occurs during a period of time in which the body goes through and surpasses an adverse situation, with changes and success, because of some mechanisms, called protection factors, that stimulate people to confront risks, assist them to endure adversities and to manage the consequences of the psychological damages that had been overtaken, in this dissertation the main objective was to understand the process of resilience in the behavior of employees who had lived the acquisition operation, in 2000, of the public institution, where they worked, by a private institution, in which they still work until today, that is, 2009, developing their work activities. This is a descriptive, qualitative and interpretative research, with sectional temporal cut and longitudinal perspective, once it describes the depositions grouped in four points of inflection, that compose the resilience process, which are: factors of risk, vulnerability, internal factors of protection - coping - and external factors of protection - buffers - and, after that, they had been interpreted by the content analysis method according to the feelings that emerged in the participants of this research, after the acquisition operation period of time which they passed through. The data came from primary origin, collected by means of half-structuralized interview, which followed a preset script to unmask the occult or manifest intentions of these research participants' behaviors. The results had pointed that those who had been part of the selected sample for this research, by the snow ball technique, had demonstrated indications of resilient behavior and today, in 2009, they are fortified in detriment of the acquisition process, that happened in 2000, that is, in detriment of the adverse situation that they lived and faced due to protection factors: religiosity, self-knowledge and art. Considerando que a resiliência é o resultado de um processo ocorrido ao longo de um tempo em que o corpo enfrenta e ultrapassa uma situação aversiva, com mudanças e sucessos, devido aos mecanismos, denominados de fatores de proteção, que estimulam o enfrentamento dos riscos, auxiliam as pessoas a suportarem as adversidades e a administrarem as consequências dos danos psicossociais que foram ultrapassados, optou-se, nesta dissertação, como objetivo principal, compreender o processo de resiliência no comportamento dos funcionários que vivenciaram a operação de aquisição no ano de 2000 da instituição pública, onde trabalhavam, por uma instituição privada, na qual permanecem até hoje, ou seja, 2009, desenvolvendo suas atividades de trabalho. A pesquisa é do tipo descritiva, qualitativa e interpretativa, corte temporal seccional, com perspectiva longitudinal, pois descreve os depoimentos agrupados em quatro pontos de inflexão, que compõem o processo de resiliência, quais sejam: fatores de risco, vulnerabilidade, fatores de proteção internos - coping - e fatores de proteção externos - buffers - e, em seguida, foram interpretados, por intermédio da análise de conteúdo, de acordo com os sentimentos que emergiram nas pessoas que participaram desta pesquisa, após o período que compreendeu a operação de aquisição pela qual passaram. Os dados são de origem primária, coletados por meio de entrevista semiestruturada, a qual seguiu um roteiro pré-estabelecido para desvendar os propósitos ocultos ou manifestos dos comportamentos das pessoas que fizeram parte desta pesquisa. Os resultados apontaram que aqueles que fizeram parte da amostra, selecionada pela técnica snow ball para esta pesquisa, demonstraram indícios de comportamento resiliente e hoje, em 2009, se encontram fortalecidos em detrimento do processo de aquisição, ocorrido no ano 2000, ou seja, da situação adversa vivenciada e enfrentada por eles devido aos fatores de proteção: religiosidade, autoconhecimento e arte. 84 f

Salmonella spp. are widespread in nature; however, human infections occur mainly through ingestion of contaminated food, specially poultry and eggs. In Brazil, the Ministry of Agriculture (MAPA) oversees food production in general, with the goal of preventing transmission of pathogens through the food chain. In 2004, MAPA initiated a program to monitor and control levels of Salmonella in poultry during slaughter. This study analyzes isolates from MAPA's program for β-lactam resistance and the resistance genes involved, as well as the geographic distributions of potentially clonal populations of resistant isolates within Brazil. Initially, 1,939 Salmonella spp. isolated between 2004 and 2011 were examined. These isolates were tested for antimicrobial susceptibility, and 100 isolates resistant or intermediate to ampicillin and ceftriaxone were screened initially for the presence of blaSHV, blaTEM, blaOXA, blaPSA, blaCMY-1, and blaCMY-2 genes. There were 55 isolates whose resistance genes were not identified by this panel and these isolates are the subject of this report. These 55 isolates were differentiated into 31 distinct ribogroups, with multiple β-lactam resistance genes, including AmpC blaCMY, blaTEM, blaCTX-M-1, blaCTX-M-2, blaCTX-M-8, and blaCTX-M-14. Isolates carrying variants of blaCTX-M were identified in three geographic regions. Salmonella carrying particular genetic variants of blaCTX-M and belonging to the same ribogroup were identified from multiple poultry slaughtering facilities. In some instances, these presumptive clonal-related isolates were from facilities over 300 miles apart, indicating potential clonal spread between two geographic regions. This is the first report of blaCTX-M-1 and blaCTX-M-14 in Salmonella in Brazil. [ABSTRACT FROM AUTHOR]

- Abstract
This study evaluated the breath CH4 excretion and concentration of M. smithii in intestinal microbiota of schoolchildren from 2 slums. One hundred and eleven children from a slum near a sanitary landfill, 35 children of a slum located away from the sanitary landfill, and 32 children from a high socioeconomic level school were included in the study. Real-time PCR was performed to quantify the M. smithii nifH gene and it was present in the microbiota of all the participating children, with higher P<0.05 concentrations in those who lived in the slum near the landfill (3.16×107 CFU/g of feces), comparing with the children from the slum away from the landfill (2.05×106 CFU/g of feces) and those from the high socioeconomic level group (3.93×105 CFU/g of feces). The prevalence of children who present breath methane was 53% in the slum near the landfill, 31% in the slum further away from the landfill and, 22% in the high socioeconomic level group. To live near a landfill is associated with higher concentrations of M. smithii in intestinal microbiota, comparing with those who live away from the landfill, regardless of their socioeconomics conditions.

In human medicine, RDW has been widely used as a predictor and biomarker of various diseases, such as acute mesenteric ischemia. In equines, this ischemia has been associated with vermintic colic, a potentially fatal syndrome triggered by the larvae of the nematodes Strongylus vulgaris. However, there are few studies in equine medicine on RDW and its relation to S. vulgaris infection, as well as the standardization of RDW-CV in this species. In the first phase, the objective was to compare the values of RDW-CV in equine horses of the healthy Quarter Horses and in horses attended in a Veterinary Hospital School. A total of 176 horses were evaluated, 95 of which were healthy, reared on 27 farms in the region of Presidente Prudente, São Paulo, and 81 horses hospitalized. Blood samples were evaluated to obtain the RDW index, from automated cell counter. The RDW values in hospitalized horses (18.6%) were significantly higher (p = 0.0006) than those observed in healthy horses (17.8%). The RDW-CV value (17,8%) can be used as a reference for horses of Quarter Horse breed. It is suggested that further studies are needed to evaluate the use of RDW as a biomarker or predictor of several other diseases in horses. For the second phase of the study, it was hypothesized that RDW-CV could also be used as a biomarker for Strongylus vulgaris infection. We evaluated 160 healthy horses (Quarter Horse and Quarter Horse mixed), at least two years old and of both sexes. The ELISA showed a positive result in 80% of the samples; however, the co-culture and the specific PCR were negative for S. vulgaris, despite the high number of animals (91.25%) positive for egg counts (> 50 epg). No evidence of association was observed between RDW-CV and ELISA. These results are discussed and, despite the lack of association, further investigations are needed to evaluate the role of RDW as a biomarker/predictor of vermintic arteritis in horses with colic. No entanto, existem poucos estudos na medicina equina sobre o RDW e sua relação com a infecção por S. vulgaris, bem como a padronização do RDW-CV nesta espécie. A hanseníase é uma complexa doença dermato-neurológica, crônica, de causa infecciosa que afeta a pele e os nervos periféricos, especialmente durante os episódios imunoinflamatórios agudos conhecidos com reações tipo 1/RT1 e tipo 2/RT2. Não existe modelo experimental para hanseníase e as lesões de pele têm sido extensivamente usadas para desvendar os multifacetados mecanismos imunopatológicos associados com a doença. Este estudo investigou a expressão in situ de duas distintas populações celulares que apresentam importante papel imunorregulatório: células Treg (Treg) e mastócitos (MC) em pele normal e diversas doenças cutâneas com ênfase nas reações hansênicas RT1 e RT2. Para o estudo de Tregs foram utilizadas 154 biópsias cutâneas de 114 participantes de três grupos: 1. Hanseníase (n=74), 56 RT1 (28-biópsias pareadas do mesmo paciente sem reação/durante reação, 28 biópsias únicas de RT1), 18 RT2 (12 biópsias pareadas sem reação/durante reação, 6 biópsias únicas de RT2); 2. Dermatoses: (n=29) doenças cutâneas não infecciosas e infecciosas. Controles Normais: fragmentos de peles obtidos de mamoplastias eletivas em mulheres saudáveis. Imunomarcação dupla CD25+ Foxp3+ de células Treg foi realizada em plataforma automatizada. Quantificação das células Treg duplo positivas foram feitas sem conhecimento da condição clínica do paciente (valores expressos em mm2 ). Para o estudo dos MC 80 biópsias de 3 grupos foram investigadas: 40 pacientes com hanseníase recém diagnosticados não tratados (18 sem reação, 11 RT1, 11 RT2), 29 pacientes com outras dermatoses e 11 biópsias de pele normal. Quantificação de MC intactos e desgranulados corados por azul de toluidina/mm2 ; imunomarcação streptavidina-biotina-peroxidase foi empregada para detectar MC triptase/try+ e chimase/chy+ e a densidade ótica (mediana da densidade ótica) foi avaliada. Resultados: Estudo das Tregs: Nenhuma célula Treg CD25+ Foxp3+ foi identificada em nenhuma das 11 amostras de pele normal, enquanto um número variável de Tregs foi identificado nas diversas doenças cutâneas (p0.8). Estudo dos Mastócitos: lesões cutâneas de origem infecciosa e não infecciosa mostraram números aumentados de mastócitos desgranulados do que intactos tanto na hanseníase como nas outras dermatoses quando comparados com pele normal. Os números de mastócitos (MC) desgranulados foram maiores do que intactos, independente da forma de hanseníase (do polo tuberculoide/TT ao lepromatoso/LL), independente da ocorrência de reações hansênicas (lesão reacional/sem reação) e independente do tipo de reação (RT1/RT2). Número e densidade de mastócito triptase positivo (MC try+ ) estão aumentados em relação aos quimase positivo (MC chy+ ) na hanseníase, em pacientes com e sem reação, particularmente na RT2, mas não nas outras dermatoses. Conclusões: aumento nas Treg detectados durante RT1 sugerem papel supressor dessas células em eventos associados à resposta imune celular exacerbada, responsáveis pela RT1. Expressão diferencial das subpopulações de MC try+ e chy+ foi observada na hanseníase em relação a outras doenças cutâneas e pele normal. Entretanto, nem a forma da hanseníase, nem a ocorrência de reação hansênica, estava associada a mudanças nas subpopulações de MC nas lesões sugerindo que o processo infeccioso pelo Mycobacterium leprae per se direciona a expressão de MC nas lesões cutâneas da hanseníase. Leprosy is a complex, chronic, infectious dermato-neurological disease that affects the skin and peripheral nerves especially during acute immune-inflammatory episodes known as type 1/T1R and type 2/T2R reactions. There is no experimental model for leprosy and leprosy skin lesions have been extensively used to unravel its multifaceted immunopathological mechanisms.This study investigated in situ expression of two distinct cell populations with important immunoregulatory roles: T regulatory (Treg) cells and mast cells (MC) in diverse skin diseases with emphasis on leprosy T1R and T2R. For the Treg cell study, 154 skin biopsies from 114 participants belonging to 3 groups were investigated: 1. Leprosy (n=74), 56 T1R (28-paired biopsies reactionfree/reactional from the same patient, 28 single reactional biopsy), 18 T2R (12 pairedreaction-free/reactional lesions, 6 single reactional biopsy); 2. Dermatoses: (n=29) noninfectious and cutaneous infectious diseases; 3. Normal controls: skin fragment of mammoplasty from healthy females that had cosmetic surgery. Double immunohistochemical detection of Treg cells was performed with automated platform for CD25 and Foxp3 staining. Quantifications of double immunostained Treg cells was performed (values expressed by mm2 ) blinded to the participants’ clinical status. For the mast cell study 80 skin biopsies from 3 groups were investigated: 40 newly diagnosed untreated leprosy patients (18 reaction-free, 11 T1R, 11 T2R), 29 patients with other dermatoses (the same as for Treg study) and 11 normal skins. Toluidine blue stained intact and degranulated MC counts/mm2 ; streptavidin-biotin-peroxidase immunostaining was used to detect tryptase/try+ and chymase/chy+ MCs and their density (median optical density) was evaluated. Results: Treg study: Not one CD25+ Foxp3+ Treg cell was seen in any of the 11 normal skin sections while variable numbers were detected in skin diseases (p0.8). Mast cell study: Infectious and non-infectious skin lesions showed higher numbers of degranulated than intact MC both for leprosy and other dermatoses, compared to normal skin. The numbers of degranulated MC were higher than intact MC regardless of the leprosy form (from tuberculoid/TT to lepromatous/LL), regardless of the occurrence of leprosy reactions (reactional and reaction-free) and regardless of the type of reaction (T1R/T2R). Try+ MC numbers and density were higher than chy+ MC in leprosy, in reaction-free and reactional lesions, particularly in T2R, but not in other dermatoses. Conclusions: Higher Treg numbers seen in T1R suggest Treg role in suppressing the exacerbated cell-mediated phenomenon that causes T1R. Differential expression/ of try+ and chy+ MC subsets was seen in leprosy compared to other skin diseases and to normal skin. However, neither leprosy form nor leprosy reaction was associated with MC changes in lesions suggesting that the Mycobacterium leprae infectious process per se dictates MC expression in leprosy skin lesions.

- Abstract
Aim: To analyze small intestinal bacterial overgrowth in school-aged children and the relationship between hydrogen and methane production in breath tests., Methods: This transversal study included 85 children residing in a slum and 43 children from a private school, all aged between 6 and 10 years, in Osasco, Brazil. For characterization of the groups, data regarding the socioeconomic status and basic housing sanitary conditions were collected. Anthropometric data was obtained in children from both groups. All children completed the hydrogen (H(2)) and methane (CH(4)) breath test in order to assess small intestinal bacterial overgrowth (SIBO). SIBO was diagnosed when there was an increase in H(2) ≥ 20 ppm or CH(4) ≥ 10 ppm with regard to the fasting value until 60 min after lactulose ingestion., Results: Children from the slum group had worse living conditions and lower nutritional indices than children from the private school. SIBO was found in 30.9% (26/84) of the children from the slum group and in 2.4% (1/41) from the private school group (P = 0.0007). Greater hydrogen production in the small intestine was observed in children from the slum group when compared to children from the private school (P = 0.007). A higher concentration of hydrogen in the small intestine (P < 0.001) and in the colon (P < 0.001) was observed among the children from the slum group with SIBO when compared to children from the slum group without SIBO. Methane production was observed in 63.1% (53/84) of the children from the slum group and in 19.5% (8/41) of the children from the private school group (P < 0.0001). Methane production was observed in 38/58 (65.5%) of the children without SIBO and in 15/26 (57.7%) of the children with SIBO from the slum. Colonic production of hydrogen was lower in methane-producing children (P = 0.017)., Conclusion: Children who live in inadequate environmental conditions are at risk of bacterial overgrowth and methane production. Hydrogen is a substrate for methane production in the colon.
