Pseudomonas spp. are commonly related to spoilage of dairy products. Some species belonging to this genus, in addition to multiplying at cold temperatures, produce hydrolytic enzymes responsible for dairy products spoilage. The AprX metalloprotease is one of the proteases produced by Pseudomonas. This enzyme, encoded by the aprX gene, is heat-resistant and able to maintain its activity even after heat treatments commonly used in the dairy industry, such as pasteurization and UHT (Ultra-high Temperature) treatment, resulting in loss of sensory characteristics and age gelation. Some strains of Pseudomonas can also produce heat-resistant lipolytic enzymes. These enzymes catalyze the hydrolysis of triacylglycerols, resulting in the release of fatty acids associated with spoilage in many foods. These acids contribute to the strong, rancid, soapy flavor of dairy products. Thus, the objective of this work was to highlight, through a literature review, the challenges faced by the dairy industry against proteases and lipases produced by the genus Pseudomonas. Algumas espécies desse gênero, além de se multiplicarem em temperaturas de refrigeração, produzem enzimas hidrolíticas responsáveis pela deterioração do leite e seus derivados. A metaloprotease AprX é uma das proteases produzidas por Pseudomonas. Essa enzima, codificada pelo gene aprX, é resistente ao calor e capaz de manter sua atividade mesmo após tratamentos térmicos comumente utilizados na indústria de laticínios, como pasteurização e tratamento UHT (Ultra-high Temperature), resultando em perdas das características sensoriais e em problemas como a gelificação. Algumas cepas de Pseudomonas também podem produzir enzimas lipolíticas termorresistentes. Essas enzimas catalisam a hidrólise de triacilgliceróis, resultando na liberação de ácidos graxos associados à deterioração de muitos alimentos. Esses ácidos contribuem para o sabor forte, rançoso e de sabão de produtos lácteos. Conselho Nacional de Desenvolvimento Científico e Tecnológico Biofilme é uma comunidade microbiana contida em uma matriz polimérica e aderida a uma superfície. Essa estrutura é responsável por garantir maior resistência de células bacterianas imersas nessa matriz a agentes antimicrobianos. Uma alternativa para controlar a ocorrência de biofilmes em ambientes de processamento de alimentos é o uso de agentes surfactantes. Os biossurfactantes têm atraído a atenção como substitutos dos surfactantes sintéticos, exibindo entre diversas qualidades, maior biodegradabilidade, baixa toxicidade e baixa concentração crítica micelar (CMC). O objetivo desse trabalho foi avaliar a formação de biofilme de Pseudomonas fluorescens em aço inoxidável, em contato com leite integral UHT, por 120 h, em temperatura de 7 °C. E avaliar o efeito dos biossurfactantes ramnolipídeo e surfactina e do surfactante sintético cloreto de benzalcônio (CB) em concentrações abaixo, próxima ou igual e acima da CMC, além de avaliar o tempo de contato, na redução do biofilme pré-formado. Cupons de aço inoxidável AISI 304 # 4, foram incubados em poços de placas de poliestireno, com inóculo inicial de 10 4 UFC mL -1 de P. fluorescens 07A em leite integral UHT, e incubados a 7 °C. Foi realizada quantificação de células aderidas a cada 24 h. A aplicação dos agentes surfactantes foi realizada sobre os biofilmes pré-formados por 120 h. Além disso, antes e após os tratamentos, os cupons de aço inoxidável foram observados por Microscopia Confocal de Varredura a Laser. Constatou-se que a cepa de P. fluorescens estudada foi capaz de formar biofilme em temperatura de refrigeração em contato com leite integral UHT, após 72 h de incubação. Os biossurfactantes não foram tão eficazes quanto o surfactante sintético estudado. O aumento da concentração de surfactina não aumentou a redução de células do biofilme, no entanto o aumento do tempo de contato demonstrou efeito positivo sobre a redução. Ramnolipídeo foi capaz de reduzir aproximadamente 1 ciclo log de UFC cm -2 de células viáveis, na CMC; aumento do tempo de contato também demonstrou efeito. Para ambos os biossurfactantes a interação entre concentração e tempo de contato foi não significativa (P > 0,01); enquanto para o surfactante sintético CB a interação foi significativa (P < 0,01), mostrando que o aumento do tempo de contato e da concentração desse agente garantem maior redução do número de células viáveis no biofilme. A partir dos resultados obtidos verificou-se que P. fluorescens pode formar biofilmes em superfície utilizada no processamento de alimentos em temperatura de refrigeração com a presença de resíduos de leite. No entanto, surfactantes sintéticos ainda demostram melhor efeito do que os biossurfactantes sobre biofilme. Biofilm is a microbial community contained in a polymer matrix and adhered to a surface. Biosurfactants have garnered attention as substitutes for synthetic surfactants, exhibiting among several qualities, greater biodegradability, low toxicity and low critical micelle concentration (CMC). The objective of this study is to evaluate the formation of Pseudomonas fluorescens biofilm in stainless steel, in contact with UHT whole milk, for 120 h at a temperature of 7 ° C. And to evaluate the effect of the biosurfactants rhamnolipid and surfactin and the synthetic surfactant benzalkonium chloride (BC) in concentrations below, near or equal to and above the CMC, besides evaluating the contact time, in reducing the preformed biofilm. Stainless steel coupons AISI 304 # 4 were incubated in polystyrene plate wells with initial inoculum of 10 4 CFU mL -1 of P. fluorescens 07A in UHT whole milk and incubated at 7 ° C. Quantification of adhered cells were performed every 24 h. The application of the surfactants was performed after the biofilm had been preformed for 120 h. In addition, before and after the treatments, the stainless steel coupons were observed by Confocal Laser Scanning Microscopy. It was verified that the strain of P. fluorescens studied was able to form biofilm at refrigeration temperature in contact with UHT whole milk, after 72 h of incubation. The biosurfactants were not as effective as the synthetic surfactant studied. The increased surfactin concentration did not increase the reduction of biofilm cells, however, the increase in contact time showed a positive effect on the reduction. Ramnolipid was able to reduce approximately 1 log cycle of CFU cm -2 of viable cells in CMC; the increased contact time was also effective. For both biosurfactants the interaction between concentration and contact time was not significant (P > 0,01); while for the synthetic surfactant BC the interaction was significant (P < 0,01), showing that increased contact time and concentration of this agent provides a greater reduction in the number of viable cells in the biofilm. From the results obtained it was verified that P. fluorescens can form biofilms on surfaces used in food processing at refrigeration temperature with the presence of milk residues. However, synthetic surfactants still have a better effect than biosurfactants on biofilm. more...
