Plantain (Plantago lanceolata L.) is a forage that produces secondary metabolites with one, aucubin, known to inhibit soil nitrification. This study aimed to quantify the exudation of aucubin and catalpol by plantain root systems in a hydroponic experiment; evaluate the effect of aucubin on nitrogen (N) leaching and nitrous oxide (N2O) emissions from urine applied to plantain and ryegrass-white clover swards; and compare N losses from a ryegrass-white clover sward after urine from cows fed plantain and cows fed ryegrass-white clover was applied. Nitrate ( NO 3 − ) leaching and N2O losses were measured in a lysimeter experiment. Catalpol, but not aucubin, was exudated by plantain roots. N2O emissions were decreased by plantain swards and by ryegrass-white clover swards to which aucubin was also applied. Aucubin had no effect on NO 3 − leaching. Urine from cows grazing plantain had no effect on N2O emissions, and N leaching when compared to the urine from cows grazing ryegrass-white clover with the same N content. We conclude the plantain sward and the aucubin applied to the ryegrass-white clover sward decreased N2O emissions via mechanisms in the soil separate from the decreased emissions resulting from the lower N concentration of urine derived from cows grazing plantain. The first law of black hole mechanics, which relates the change of energy to the change of entropy and other conserved charges, has been the main motivation for probing the thermodynamic properties of black holes. In this work, we investigate the thermodynamics of Kerr Taub-NUT AdS black holes. We present geometric Komar definitions for the black hole charges, that by construction satisfy the Smarr formula. Further, by a scaling argument based on Euler’s theorem, we establish the first law for the Kerr Taub-NUT AdS black holes. The corresponding first law includes variations in the cosmological constant, NUT charges and angular momenta. The key new ingredient in the construction are the independent variations of both angular momenta, the black hole and Misner string angular momenta. Employing the Brown-York quasi-local charge definitions we show that our expression for the mass and spin coincide with our generalized Komar expressions. We indicate the relevance of these results to the thermodynamics of rotating AdS black holes, including the proper choice of time-like Killing vector to produce the correct thermodynamic mass. Nontrivial diffeomorphisms act on the horizon of a generic 4D black holes and create distinguishing features referred to as soft hair. Amongst these are a leftâ€"right pair of Virasoro algebras with associated charges that reproduce the Bekensteinâ€"Hawking entropy for Kerr black holes. In this paper we show that if one adds a negative cosmological constant, there is a similar set of infinitesimal diffeomorphisms that act non-trivially on the horizon. The algebra of these diffeomorphisms gives rise to a central charge. Adding a boundary counterterm, justified to achieve integrability, leads to well-defined central charges with c L = c R. Force-Free Electrodynamics for black holes in Anti de Sitter is considered. We present new, energy extracting solutions of Force-Free Electrodynamics in Anti de Sitter-Near Horizon Extremal Kerr and Super-Entropic Near Horizon Extremal Kerr geometries. The relevant equations of motion are derived from an action for force-free plasma surrounding spinning black holes with generic asymptotics. We consider the energy flux of electrodynamic fields in rotating frames to argue that the correct measure for energy extraction is the energy flux measured by a rotating observer in the near horizon region. Background: The use of hypoallergenic derivatives is considered beneficial to promote the safety and efficacy of allergen-specific immunotherapy. We aimed to assess the efficacy of reduced and alkylated (R/A) Pru p 3, a hypoallergenic folding variant of the major peach allergen, in subcutaneous immunotherapy (SCIT) using a murine model of peach allergy.Methods and Results: After sensitization with Pru p 3, BALB/c mice received SCIT with Pru p 3 or R/A Pru p 3 and were challenged with Pru p 3. SCIT with Pru p 3, but not with R/A Pru p 3, suppressed anaphylaxis upon the challenge significantly. SCIT with Pru p 3 did not suppress Pru p 3-specific IgE and IgG1 production, but enhanced IgG2a production. In contrast, SCIT with R/A Pru p 3 suppressed IgE and IgG1 production, but enhanced IgG2a production only moderately. The therapeutic efficacy of SCIT with Pru p 3 was associated with induction of IL-10 and IFN-γ.Conclusion: Hypoallergenic folding variant of Pru p 3 is not likely an efficacious therapeutic component in SCIT of peach allergy. Holographic dualities between certain gravitational theories in four and five spacetime dimensions and 2D conformal field theories (CFTs) have been proposed based on hidden conformal symmetry exhibited by the radial Klein-Gordon (KG) operator in a so-called near-region limit. In this paper, we examine hidden conformal symmetry of black rings and black strings solutions, thus demonstrating that the presence of hidden conformal symmetry is not linked to the separability of the KG-equation (or the existence of a Killing-Yano tensor). Further, we will argue that these classes of non-extremal black holes have a dual 2D CFT. New revised monodromy techniques are developed to encompass all the cases we consider. A new solution of four-dimensional vacuum General Relativity is presented. It describes the near horizon region of the extreme (maximally spinning) binary black hole system with two identical extreme Kerr black holes held in equilibrium by a massless strut. This is the first example of a non-supersymmetric, near horizon extreme binary black hole geometry of two uncharged black holes. The black holes are co-rotating, their relative distance is fixed, and the solution is uniquely specified by the mass. Asymptotically, the geometry corresponds to the near horizon extreme Kerr (NHEK) black hole. The binary extreme system has finite entropy. [ABSTRACT FROM AUTHOR]

Pru p 3 is the major peach allergen in the Mediterranean area. It frequently elicits severe reactions, limiting its study in humans, raising the need for animal models to investigate the immunological mechanisms involved. However, no anaphylaxis model exists for Pru p 3. We aimed to develop a model of peach anaphylaxis by sensitising mice with Pru p 3 in combination with lipopolysaccharide (LPS) as an adjuvant. Four groups of mice were sensitised intranasally: untreated; treated with Pru p 3; treated with LPS; treated with Pru p 3 + LPS. After sensitisation mice were intraperitoneally challenged with Pru p 3 and in vivo and in vitro parameters were evaluated. Only mice in the Pru p 3 + LPS group showed anaphylaxis symptoms, including a decrease in temperature. Determination of in vitro parameters showed a Th2 response with an increase of Pru p 3-specific IgE and IgG1. Moreover, at the cellular level, we found increased levels of IgE and IgG1 secreting Pru p 3-specific cells and a proliferative CD4+ T-cell response. These results demonstrate that Pru p 3-specific anaphylaxis can be generated after nasal sensitisation to Pru p 3 in combination with LPS. Amyotrophic lateral sclerosis is a progressive fatal neurodegenerative disease caused by loss of motor neurons and characterized neuropathologically in almost all cases by nuclear depletion and cytoplasmic aggregation of TDP-43, a nuclear RNA-binding protein (RBP). We identified ELAVL3 as one of the most downregulated genes in our transcriptome profiles of laser captured microdissection of motor neurons from sporadic ALS nervous systems and the most dysregulated of all RBPs. Neuropathological characterizations showed ELAVL3 nuclear depletion in a great percentage of remnant motor neurons, sometimes accompanied by cytoplasmic accumulations. These abnormalities were common in sporadic cases with and without intermediate expansions in ATXN2 and familial cases carrying mutations in C9orf72 and SOD1. Depletion of ELAVL3 occurred at both the RNA and protein levels and a short protein isoform was identified, but it is not related to a TDP-43-dependent cryptic exon in intron 3. Strikingly, ELAVL3 abnormalities were more frequent than TDP-43 abnormalities and occurred in motor neurons still with normal nuclear TDP-43 present, but all neurons with abnormal TDP-43 also had abnormal ELAVL3. In a neuron-like cell culture model using SH-SY5Y cells, ELAVL3 mislocalization occurred weeks before TDP-43 abnormalities were seen. We interrogated genetic databases, but did not identify association of ELAVL3 genetic structure with ALS. Taken together, these findings suggest that ELAVL3 is an important RBP in ALS pathogenesis acquired early and the neuropathological data suggest that it is involved by loss of function rather than cytoplasmic toxicity. Background: Amoxicillin (AX) is nowadays the β‐lactam that more frequently induces immediate allergic reactions. Nevertheless, diagnosis of AX allergy is occasionally challenging due to risky in vivo tests and non‐optimal sensitivity of in vitro tests. AX requires protein haptenation to form multivalent conjugates with increased size to be immunogenic. Knowing adduct structural features for promoting effector cell activation would help to improve in vitro tests. We aimed to identify the optimal structural requirement in specific cellular degranulation to AX using well‐precised nanoarchitectures of different lengths. Method: We constructed eight Bidendron Antigens (BiAns) based on polyethylene glycol (PEG) linkers of different lengths (600–12,000 Da), end‐coupled with polyamidoamine dendrons that were terminally multi‐functionalized with amoxicilloyl (AXO). In vitro IgE recognition was studied by competitive radioallergosorbent test (RAST) and antibody–nanoarchitecture complexes by transmission electron microscopy (TEM). Their allergenic activity was evaluated using bone marrow‐derived mast cells (MCs) passively sensitized with mouse monoclonal IgE against AX and humanized RBL‐2H3 cells sensitized with polyclonal antibodies from sera of AX‐allergic patients. Results: All BiAns were recognized by AX‐sIgE. Dose‐dependent activation responses were observed in both cellular assays, only with longer structures, containing spacers in the range of PEG 6000–12,000 Da. Consistently, greater proportion of immunocomplexes and number of antibodies per complex for longer BiAns were visualized by TEM. Conclusions: BiAns are valuable platforms to study the mechanism of effector cell activation. These nanomolecular tools have demonstrated the importance of the adduct size to promote effector cell activation in AX allergy, which will impact for improving in vitro diagnostics. • HS-SPME/GC × GC–ToFMS was used for determination of polar death-related VOCs. • First use of zwitterionic polymeric ionic liquids (PIL) for extraction of polar VOCs. • Coatings' extraction efficiency has the following order: PDMS/DVB < PIL < PA < DVB/CAR/PDMS. • PA and PIL provided a good balance representativeness of headspace composition/extraction efficiency. • The 19 polar VOCs were capture from headspace and determined at ng level. Decomposition of bodies generates several types of polar volatile organic compounds (VOCs), whose types, patterns and ratios change during the various stages of decomposition and, therefore, their determination has huge potential to provide useful information to disclose events related to the time of death, or body surrounding environment. As sample preparation is a mandatory key-point in a method development, this research aims to develop a simple, accurate and rapid approach to study death-related polar VOCs based on headspace solid-phase microextraction (HS-SPME) combined with comprehensive two-dimensional gas chromatography-time of flight mass spectrometry (GC × GC-ToFMS) analysis. The performance of zwitterionic PIL-based fibers (containing a [VIm+C 9 COO− monomer and a [(VIm) 2 C1 2 2+-2Br− crosslinker), tailored for polar compounds, was evaluated for a set of 19 analytes associated with the unique odour created by decomposing bodies, and it was compared to the commercially-available fibers: divinylbenzene/carboxen/poly(dimethylsiloxane) – DVB/CAR/PDMS, poly(dimethylsiloxane)/divinylbenzene – PDMS/DVB and polyacrylate (PA). Fibers with absorptive-type mechanism, such as PA and PIL, showed the best results in the balance of the parameters studied, being able to detect analytes at ng level and providing a profile representative of the headspace composition, thus they may represent a useful tool to respond to current challenges in forensic taphonomy. The reproducibility (with relative standard deviation lower than 18%, depending on the analyte) and relative recoveries (higher than 99.1%) were similar and acceptable for both fibers. The zwitterionic PIL, with ca. 4 times smaller film thickness than PA, still has potential to have the best performance, supported by the efforts to obtain thicker sorbent coatings. Mechanisms underlying motor neuron degeneration in amyotrophic lateral sclerosis (ALS) are yet unclear. Specific deletion of the ER-component membralin in astrocytes manifested postnatal motor defects and lethality in mice, causing the accumulation of extracellular glutamate through reducing the glutamate transporter EAAT2. Restoring EAAT2 levels in membralin KO astrocytes limited astrocyte-dependent excitotoxicity in motor neurons. Transcriptomic profiles from mouse astrocytic membralin KO motor cortex indicated significant perturbation in KEGG pathway components related to ALS, including downregulation of Eaat2 and upregulation of Tnfrsf1a. Changes in gene expression with membralin deletion also overlapped with mouse ALS models and reactive astrocytes. Our results shown that activation of TNF receptor (TNFR1)-NFκB pathway known to suppress Eaat2 transcription was upregulated with membralin deletion. Further, reduced membralin and EAAT2 levels correlated with disease progression in spinal cord from SOD1-mutant mouse models, and reductions in membralin/EAAT2 were observed in human ALS spinal cord. Importantly, overexpression of membralin in SOD1G93A astrocytes decreased TNFR1 levels and increased EAAT2 expression, and improved motor neuron survival. Importantly, upregulation of membralin in SOD1G93A mice significantly prolonged mouse survival. Together, our study provided a mechanism for ALS pathogenesis where membralin limited glutamatergic neurotoxicity, suggesting that modulating membralin had potentials in ALS therapy. The article focuses on the quantitative study of antisense RNA foci related to nucleoli and TDP-43 mislocalization C9ORF72 amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) (C9-ALS/FTD).

Hexanucleotide repeat expansions in C9orf72 are the most common genetic cause of amyotrophic lateral sclerosis (C9 ALS). The main hypothesized pathogenic mechanisms are C9orf72 haploinsufficiency and/or toxicity from one or more of bi-directionally transcribed repeat RNAs and their dipeptide repeat proteins (DPRs) poly-GP, poly-GA, poly-GR, poly-PR and poly-PA. Recently, nuclear import and/or export defects especially caused by arginine-containing poly-GR or poly-PR have been proposed as significant contributors to pathogenesis based on disease models. We quantitatively studied and compared DPRs, nuclear pore proteins and C9orf72 protein in clinically related and clinically unrelated regions of the central nervous system, and compared them to phosphorylated TDP-43 (pTDP-43), the hallmark protein of ALS. Of the five DPRs, only poly-GR was significantly abundant in clinically related areas compared to unrelated areas (p < 0.001), and formed dendritic-like aggregates in the motor cortex that co-localized with pTDP-43 (p < 0.0001). While most poly-GR dendritic inclusions were pTDP-43 positive, only 4% of pTDP-43 dendritic inclusions were poly-GR positive. Staining for arginine-containing poly-GR and poly-PR in nuclei of neurons produced signals that were not specific to C9 ALS. We could not detect significant differences of nuclear markers RanGap, Lamin B1, and Importin β1 in C9 ALS, although we observed subtle nuclear changes in ALS, both C9 and non-C9, compared to control. The C9orf72 protein itself was diffusely expressed in cytoplasm of large neurons and glia, and nearly 50% reduced, in both clinically related frontal cortex and unrelated occipital cortex, but not in cerebellum. In summary, sense-encoded poly-GR DPR was unique, and localized to dendrites and pTDP43 in motor regions of C9 ALS CNS. This is consistent with new emerging ideas about TDP-43 functions in dendrites. Molecular and cellular mechanisms underlying the peripheral conditioning lesion remain unsolved. We show here that injection of a chemical demyelinating agent, ethidium bromide, into the sciatic nerve induces a similar set of regeneration-associated genes and promotes a 2.7-fold greater extent of sensory axon regeneration in the spinal cord than sciatic nerve crush. We found that more severe peripheral demyelination correlates with more severe functional and electrophysiological deficits, but more robust central regeneration. Ethidium bromide injection does not activate macrophages at the demyelinated sciatic nerve site, as observed after nerve crush, but briefly activates macrophages in the dorsal root ganglion. This study provides a new method for investigating the underlying mechanisms of the conditioning response and suggests that loss of the peripheral myelin may be a major signal to change the intrinsic growth state of adult sensory neurons and promote regeneration. We study scattering coefficients in black hole spacetimes using analytic properties of complexified wave equations. For a concrete example, we analyse the singularities of the Teukolsky equation and relate the corresponding monodromies to scattering data. These techniques, valid in full generality, provide insights into complex-analytic properties of greybody factors and quasinormal modes. • A NGFR100W knockin mouse model of Hereditary Sensory Autonomic Neuropathy V (HSAN V). • Homozygous mutant mice showed severe,deficits in peripheral sensory structure and function. • Heterozygous mutant mice developed progressive degeneration of small sensory fibers. • Heterozygous mutant mice showed no apparent structural/functional changes in the brain. • HSAN V is likely resulted from reduced secretion of NGF. The R100W mutation in nerve growth factor is associated with hereditary sensory autonomic neuropathy V in a Swedish family. These patients develop severe loss of perception to deep pain but with apparently normal cognitive functions. To better understand the disease mechanism, we examined a knockin mouse model of HSAN V. The homozygous mice showed significant structural deficits in intra-epidermal nerve fibers (IENFs) at birth. These mice had a total loss of pain perception at ∼2 months of age and often failed to survive to adulthood. Heterozygous mutant mice developed a progressive degeneration of small sensory fibers both behaviorally and functionally: they showed a progressive loss of IENFs starting at the age of 9 months accompanied with progressive loss of perception to painful stimuli such as noxious temperature. Quantitative analysis of lumbar 4/5 dorsal root ganglia revealed a significant reduction in small size neurons, while analysis of sciatic nerve fibers revealed the heterozygous mutant mice had no reduction in myelinated nerve fibers. Significantly, the amount of NGF secreted from mouse embryonic fibroblasts were reduced from both heterozygous and homozygous mice compared to their wild-type littermates. Interestingly, the heterozygous mice showed no apparent structural alteration in the brain: neither the anterior cingulate cortex nor the medial septum including NGF-dependent basal forebrain cholinergic neurons. Accordingly, these animals did not develop appreciable deficits in tests for brain function.
