Objective: This study aimed to investigate the efficacy of Bacillus-based probiotics supplemented at two different levels to modulate the productive performance, egg quality, tibia traits, and specific cecal bacteria counts of Hy-Line Brown layers from 25 to 37 weeks of age., Methods: A total of 216 twenty-five-week-old hens were randomly distributed into 3 experimental diets with 12 replicates of 6 birds per cage. Diets included basal diet supplemented with 0 (CON), 3×108 (PRO1), or 3×109 (PRO2) colony-forming unit (CFU) of the test probiotic containing Bacillus subtilis PB6, Bacillus subtilis FXA, and Bacillus licheniformis G3 per kilogram of feed., Results: Improved egg weights and mass at 29 weeks; and feed intake at 31 weeks (p<0.10) were noticed with the probiotic-supplemented PRO1 and PRO2 diets. Considering egg quality, the shell thickness, Haugh units, and yolk color were improved; but yolk cholesterol was lowered (p<0.05) with PRO1 and PRO2 diets at 29 weeks. At both 33 and 37 weeks, the egg-breaking strength, shell color and thickness, albumen height, Haugh units, and yolk color were improved; but yolk cholesterol was similarly lowered (p<0.05) with the PRO1 and PRO2 diets. Improved tibia Ca, ash, weights, and density; and raised cecal counts of Bifidobacteria and Lactobacilli (p<0.05) were noticed with PRO1 and PRO2 diets. Improved tibia P but reduced Clostridia counts (p<0.10) were also observed with the PRO1 and PRO2 diets., Conclusion: Probiotic supplementation of Bacillus subtilis PB6, Bacillus subtilis FXA, and Bacillus licheniformis G3 at 3×108 CFU/kg of feed is adequate to significantly improve egg quality, lower yolk cholesterol, enhance several tibia traits, and raise the populations of beneficial cecal bacteria. Modest improvements in several productive parameters and tibia P but reduced Clostridia were also observed; and could warrant further investigation of probiotic effects beyond the current test period.

In this paper, we introduce the concepts of sensitivity and specificity to mathematically describe the accuracy of the peer review process. Sensitivity refers to the probability that the final decision for a manuscript would be acceptance, provided the manuscript meets the journal standards required for publication (i.e., true positive rate). Specificity refers to the probability that the final decision would be rejection, provided the work does not meet the standards required for publication (i.e., true negative rate). Therefore, in the peer review process, sensitivity measures the ability to correctly accept manuscripts that meet the required standards (true positives) and specificity measures the ability to correctly reject manuscripts that do not meet those quality standards required for publication (true negatives). Sensitivity and specificity values can inform the editor under what conditions the outcome of a peer review process becomes more precise and, therefore, if this does not occur, when the editor must improve the analysis involved in processing the information received from reviewers' reports. Sensitivity and specificity understood in this way can promote the ethical conduct of peer review processes and improve the validity of manuscript editorial decisions.

Brain signalling pathways involved in subclinical anxiety and depressed mood can be modulated via the gut brain axis (GBA), providing the potential for diet and dietary components to affect mood. We investigated behavioural, physiological and gut microbiome responses to the Lacticaseibacillus rhamnosus strain HN001 (LactoB HN001™), which has been shown to reduce postpartum anxiety and depression, and a milk fat globule membrane-enriched product, Lipid 70 (Surestart TM MFGM Lipid 70), which has been implicated in memory in stress-susceptible Wistar Kyoto rats. We examined behaviour in the open field, elevated plus maze and novel object recognition tests in conjunction with the expression of host genes in neuro-signalling pathways, and we also assessed brain lipidomics. Treatment-induced alterations in the caecal microbiome and short-chain fatty acid (SCFA) profiles were also assessed. Neither ingredient induced behavioural changes or altered the brain lipidome (separately or when combined). However, with regard to brain gene expression, the L. rhamnosus HN001 + Lipid 70 combination produced a synergistic effect, reducing GABAA subunit expression in the amygdala ( Gabre , Gat3 , Gabrg1 ) and hippocampus ( Gabrd ). Treatment with L. rhamnosus HN001 alone altered expression of the metabotropic glutamate receptor ( Grm4 ) in the amygdala but produced only minor changes in gut microbiota composition. In contrast, Lipid 70 alone did not alter brain gene expression but produced a significant shift in the gut microbiota profile. Under the conditions used, there was no observed effect on rat behaviour for the ingredient combination. However, the enhancement of brain gene expression by L. rhamnosus HN001 + Lipid 70 implicates synergistic actions on region-specific neural pathways associated with fear, anxiety, depression and memory. A significant shift in the gut microbiota profile also occurred that was mainly attributable to Lipid 70.

Floating objects play a pivotal role in pelagic ecosystems by serving as shelters, meeting points, cleaning stations, nurseries, and feeding grounds. The abundance of these objects is increasing globally in the form of flotsam, plastics, discarded or lost fishing gear, and fish aggregating devices (FADs) deployed by commercial fisheries. However, it is difficult to measure how often and in what ways fish interact with floating objects in pelagic environments. Dolphinfish (Coryphaena hippurus) is prevalent among the fish species that associate with floating objects, but the extent to which dolphinfish utilize them is unclear. This study applies existing knowledge of FAD-associated dolphinfish diving behavior to identify periods of potential association with floating objects in a remote telemetry dataset of 23 fish with a total of 678 days at liberty spanning two distinct regions within the Eastern Pacific Ocean. Fish inhabiting waters off the western coast of Baja California Sur, Mexico spent significantly more time exhibiting behavior indicative of association with floating objects than those off the coast of Oaxaca, Mexico. When not exhibiting this behavior, dolphinfish in both regions occupied similar vertical habitats, with western Baja fish utilizing more of the water column than Oaxaca fish. Observed regional differences in behavior were coincident with regional differences in size (Oaxaca fish fork lengths ranged from 103 to 118 cm (mean = 110 cm), while Baja fish ranged from 85 to 106 cm (mean = 93 cm)). Although larger fish in the Baja region displayed behavior consistent with smaller Baja fish, future studies should investigate whether the observed regional differences are due to (i) size, (ii) sex, (iii) oceanography, or (iv) availability of floating objects. Dolphinfish are an important mid-trophic level predator and potentially sustainable fishery resource. Understanding their behavior and use of floating objects is of both ecological and economic importance-particularly in the context of expanding international FAD-based fisheries. Our study suggests dolphinfish spend a large amount of their time exhibiting potential floating object associated behavior, and this could influence their population structure and growth., Competing Interests: The authors have declared that no competing interests exist.

Laboratory mice are commonly euthanised with carbon dioxide (CO 2 ); however, there is ample evidence that this gas is aversive. Previous work suggests that sedation achieved via injection with benzodiazepines prior to CO 2 administration could reduce aversive behaviours during euthanasia. We explored the potential of using a voluntarily ingested sedative (tiletamine-zolazepam, Zoletil ® ) prior to euthanasia. Male and female C57BL/6 mice were allocated into one of the five experimental groups, which differed in the dose of Zoletil: 0, 10, 20, 40, 80 or 100 mg/kg. A dose of 20 mg/kg was found to achieve mild sedation prior to euthanasia; mice which received this dose numerically reared and walked on the cage lid less, and showed ataxia, immobility and recumbency for longer than mice that received a lower dose. During euthanasia, mice that received 20 mg/kg showed fewer aversive responses to CO 2 . Doses of 40 to 100 mg/kg were associated with signs of moderate to severe sedation, but resulted in an incomplete intake of the sedative, which made the interpretation of the aversiveness to CO 2 difficult. Voluntary oral administration of a sedative is an effective, affordable, and easy way to minimize the stress of mice to euthanasia with CO 2 .
