The few-view image reconstruction problem is one of the challenging research areas in industrial Computed Tomography(CT). On the one hand, acquiring enough data for reconstruction leads to a longer scanning time, which may not be applicable in industrial CT. On the other hand, reconstructing under-sampled data leads to artifacts and inaccurate image analysis. To get a usable reconstruction for image analysis, from the insufficient data caused by quick cone beam CT scans, we use a Total Variation (TV) regularized optimization problem. A split Bregman implementation is used to solve the TV regularized CT reconstruction problem. To evaluate the quality of the few-view reconstruction produced by the split Bregman, we perform gray value analysis and porosity analysis. Alexander Rhode investigates performance-oriented measures of Contracting Authorities in public tenders conducted within the EU. He finds that Contracting Authorities can improve their performance and attract more suppliers by publishing (as precise as possible) starting prices in the beginning of a tender. First, he reports that compared with private-sector negotiations, starting prices do not create entry barriers in public procurement. Second, he finds that increased numerical precision of starting prices is linearly correlated with better performance and a higher number of bids. In public procurement, suppliers tend to attribute increased credibility to precise starting prices which reduces their (perceived) entry risks.

Die Fluoreszenz in situ Hybridisierung (FISH) ist eine Nachweismethode für Mikroorganismen und stellt eine vielversprechende Alternative zum konventionellen, langsamen Kulturnachweis beziehungsweise zu anderen schnellen molekularen Detektionsmethoden ohne Möglichkeit einer Lebend/Tot- Differenzierung dar. In dieser Arbeit sollte das Potenzial von FISH für den Nachweis lebender pathogener Bakterien in Lebensmitteln evaluiert werden. Für eine Reihe wichtiger Zoonoseerreger wie S. enterica, thermophile Campylobacter spp., pathogene Yersinien oder L. monocytogenes wurden FISH-Sonden, die auf die 16S oder 23S rRNA gerichtet sind, entworfen und getestet. Die neu entwickelten Sonden waren bereits bekannten FISH-Sonden hinsichtlich ihrer Spezifität und Sensitivität häufig deutlich überlegen. Umfangreiche thermodynamische Anpassungen und der gezielte Einsatz von Kompetitoren und Nukleotidanaloga erlaubten den spezifischen Simultannachweis vieler unterschiedlicher Erregergruppen in Gegenwart nahverwandter Bakterien. Die Kombination von FISH mit dem direct viable count (DVC-FISH) erwies sich als zuverlässige Methode zur Lebend/Tot-Unterscheidung. Mit Hilfe eines FISH-Tests auf mRNA-Ebene, der durch den Einsatz eines Sondengemisches auch Zielsequenzen mit geringer Abundanz erfassen kann, konnte zusätzlich die Transkription von Antibiotikaresistenzgenen auf Einzelzellebene detektiert werden. Die Kugelmühle FastPrep-24 und mit Einschränkungen der Stomacher erwiesen sich als geeignete Homogenisierungssysteme zur Probenvorbereitung, um lebende Bakterien aus einer festen Lebensmittelmatrix herauszulösen. Nach einer ein- bis zweitägigen kulturellen Voranreicherung erreichten die entwickelten FISH-Tests eine vergleichbare Sensitivität wie die deutlich zeitaufwändigeren Goldstandardmethoden. Eine kulturunabhängige Anreicherung gelang durch mehrstufige Vorfiltrationen und nichtfluoreszierende Membranfilter. Die Zugabe eines Markerbakteriums ermöglichte gleichzeitig die Quantifizierung der Bakterienlast. FISH repräsentiert eine Brückentechnologie, die die Geschwindigkeit anderer Schnellnachweise mit der Eigenschaft der kulturellen Methoden zur Unterscheidung von lebenden und toten Bakterien verbindet. Um FISH als eine anerkannte Technik in der Lebensmittelmikrobiologie zu etablieren, müssen jedoch die Automatisierung und Standardisierung dieser Nachweismethode weiter vorangetrieben werden., Fluorescence in situ hybridization (FISH) is a detection method for microorganisms and represents a promising alternative to the slow conventional culture techniques or to other rapid molecular methods without the capacity to distinguish dead from viable bacteria. In this work, the potential of FISH for the detection of viable pathogenic bacteria in food was evaluated. FISH probes targeting the 16S or 23S rRNA were designed and tested for various important zoonotic agents like S. enterica, thermophilic Campylobacter spp., pathogenic Yersinia spp. and L. monocytogenes. In comparison to previously published probes, specificities and sensitivities of the newly developed FISH probes often proved to be superior. Due to thorough thermodynamic adaptations and the accurate use of competitors and nucleotide analogues, the specific simultaneous detection of multiple different pathogens in the presence of closely related bacteria was possible. The combination of FISH with the direct viable count (DVC-FISH) was demonstrated to be a suitable method to differentiate dead from viable bacteria. Furthermore, a FISH assay targeting mRNAs with a low abundance via a probe mixture enabled the detection of the transcription of antibiotic resistance genes. The ball mill FastPrep-24 and the Stomacher, with some limitations, proved to be appropriate homogenization tools for sample preparation to separate and obtain viable bacteria from a solid food matrix. After one or two days of cultural enrichment, the newly developed FISH-assays reached similar sensitivities as the much more time- consuming gold standard methods. Multistage prefiltration procedures and non- fluorescent membrane filters allowed the culture-independent enrichment. The addition of marker bacteria enabled the concurrent quantification of the bacterial load. FISH is a bridge technology, which combines the test speed of other rapid methods with the ability of the culture methods to differentiate between viable and dead bacteria. However, automation and standardization of this technique have to be promoted to establish FISH as a recognized standard method in food microbiology.

In dieser Arbeit sollte die Rolle des humanen Staufen1-Proteins hinsichtlich seiner Interaktion mit dem akzessorischen Protein Rec der endogenen humanen Retroviren der Familie HERV-K(HML-2) sowie dem funktionell verwandten Protein Rev des exogenen Retrovirus HIV-1 näher charakterisiert werden. Diese drei Proteine wirken maßgeblich am Kernexport ungespleißter sowie unvollständig gespleißter viraler RNAs mit. Die Kriterien, die zur Einwanderung dieser Proteine in die Stressgranula führen, sowie die beteiligten Proteinregionen sollten genauer beleuchtet werden. In diesen Untersuchungen zeigte sich, dass virale genomische RNAs nur im Falle des Rec-Proteins einen förderlichen Effekt auf die Lokalisation in den Stressgranula hatten. Der Knockdown von endogenem Staufen1 hatte ebenfalls nur einen negativen Effekt auf die Einwanderung von Rec. Analog dazu zeigte sich bei der Überexpression von Staufen1 eine verbesserte Einwanderung von Rec in die Stressgranula. Weder virale genomische RNAs noch die Expressionsstärke von Staufen1 beeinflussten dagegen die Einwanderung von Rev in die Stressgranula. Deletionsmutanten von Rev, die zur Interaktion mit Staufen1, zur Oligomerisierung oder zur Bindung an die Ziel-RNAs nicht mehr fähig waren, fanden sich trotzdem in den Stressgranula. Insgesamt war die Rekrutierung von Rev in die Stressgranula stärker als der Transport von Rec in diese Granula. Im Gegensatz zu Rec konnte Rev auch in den Prozessierungskörperchen beobachtet werden. Die Anzahl an Unterschieden legt nahe, dass Rec und Rev durch unterschiedliche Vorgänge in die Stressgranula gelangen. Trotz einiger Gemeinsamkeiten ist es denkbar, dass Staufen1 auch im Replikationsmechanismus dieser Retroviren recht unterschiedliche Funktionen wahrnimmt. Das Protein Staufen1 wird ebenfalls in großen Mengen in die Stressgranula rekrutiert. Die N-terminale Domäne mit der nicht mehr funktionsfähigen, aber konservierten dsRNA-bindenden Proteindomäne RBD2 ist für diese Einwanderung maßgeblich verantwortlich. Die Domäne RBD3 mit der stärksten Bindeaktivität an dsRNAs trägt in einem schwächeren Ausmaß ebenfalls zur Einwanderung in die Stressgranula bei. Stressgranula treten im Zuge von viralen Infektionen häufig auf und können von vielen Viren manipuliert werden. HIV-1 scheint dazu die inhibierende Wirkung von Staufen1 zu nutzen. Die genaue Rolle der Stressgranula und des in diesen enthaltenen Proteins Staufen1 während der Replikation von HIV-1, HERV-K(HML-2) und anderen Viren bleibt eine bislang nur teilweise beantwortete Frage mit möglicherweise therapeutischer Relevanz in der Zukunft., The role of the human protein Staufen1 regarding its interaction with the accessory protein Rec of the endogenous retroviruses of the HERV-K(HML-2) family and the functionally related protein Rev of the exogenous retrovirus HIV-1 was further characterized in this work. These three proteins are decisively involved in the nuclear export of unspliced and incompletely spliced viral RNAs. The criteria that lead to the recruitment of these proteins in the stress granules as well as the involved protein domains should be examined more closely. The experiments presented in this work illustrated that viral genomic RNAs enhance only the localization of Rec in the stress granules. The knockdown of endogenous Staufen1 had only a negative effect on the transport of Rec into the stress granules. Correspondingly, the Staufen1-overexpression resulted in an increased transport of Rec into the stress granules. In contrast to these findings, neither viral genomic RNAs nor the expression level of Staufen1 had an influence on the recruitment of Rev into the stress granules. Deletion mutants of Rev which were incapable to interact with Staufen1, to oligomerize or to bind to the target RNAs were still found in the stress granules. All in all, the recruitment of Rev into the stress granules was stronger than the transport of Rec in these granules. In contrast to Rec, Rev could also be observed in the processing bodies. The number of differences between Rec and Rev suggests that these proteins are recruited into the stress granules by different processes. Despite several similarities, it is conceivable that Staufen1 may serve different functions in the replication mechanism of these retroviruses as well. The protein Staufen1 is also recruited into the stress granules in large amounts. The N-terminal domain with the nonfunctional, but conserved dsRNA-binding domain RBD2 is crucial for this migration into the stress granules. The domain RBD3 with the strongest affinity for dsRNAs is also participating to a minor degree in the recruitment of Staufen1 into the stress granules. Stress granules occur frequently during viral infections and can be manipulated by various viruses. In case of HIV-1, this manipulation is mediated by taking advantage of the inhibitory capacities of Staufen1. The precise role of the stress granules and the therein included Staufen1 during the replication of HIV-1, HERV-K(HML-2) and other viruses remains to be an incompletely answered question with potential therapeutic relevance for the future.

The gadolinium(III) trifluoroacetates ((CH3)2NH 2)[Gd(CF3COO)4] (1), ((CH3) 3NH)[Gd(CF3 COO)4(H2O)] (2), Gd(CF3COO)3(H2O)3 (3) as well as Gd2(CF3COO)6(H2O) 2(phen)3 · C2H5OH (4) (phen = 1,10-phenanthroline) were synthesized and structurally characterized by X-ray crystallography. These compounds crystallize in the space group P1 (No. 2, Z = 2) (1, 2 and 4) and P 21/c (No. 14, Z = 4) (3), respectively, with the following lattice constants 1: a = 884.9(2), b = 1024.9(2), c = 1173.1(2) pm, α = 105.77(2), β = 99.51(2), γ = 107.93(2)°; 2: a = 965.1(1), b = 1028.6(1), c = 1271.3(2) pm, α = 111.83(2), β = 111.33(2), γ = 90.44(2)°; 3: a = 919.6(2), b = 1890.6(4), c = 978.7(2) pm, β = 113.94(2)°; 4: a = 1286.7(8), b = 1639.3(8), c = 1712.2(9) pm, α = 62.57(6), β = 84.13(5), γ = 68.28(5)°. The compounds consist of Gd3+ ions which are bridged by carboxylate groups either to chains (1 and 2) or to dimers (3 and 4). In addition to the Gd3+ dimers, compound (4) also contains monomeric Gd3+ units. The magnetic behaviour of 2 and 3 was investigated in a temperature range of 1.77 to 300 K.
