Background Antiarrhythmic drugs are widely used to treat patients with atrial fibrillation (AF), but the mechanisms conveying their variable effectiveness are not known. Recent data suggested that paired like homeodomain-2 transcription factor (PITX2) might play an important role in regulating gene expression and electrical function of the adult left atrium (LA). Objectives After determining LA PITX2 expression in AF patients requiring rhythm control therapy, the authors assessed the effects of Pitx2c on LA electrophysiology and the effect of antiarrhythmic drugs. Methods LA PITX2 messenger ribonucleic acid (mRNA) levels were measured in 95 patients undergoing thoracoscopic AF ablation. The effects of flecainide, a sodium (Na+)-channel blocker, and d,l-sotalol, a potassium channel blocker, were studied in littermate mice with normal and reduced Pitx2c mRNA by electrophysiological study, optical mapping, and patch clamp studies. PITX2-dependent mechanisms of antiarrhythmic drug action were studied in human embryonic kidney (HEK) cells expressing human Na channels and by modeling human action potentials. Results Flecainide 1 μmol/l was more effective in suppressing atrial arrhythmias in atria with reduced Pitx2c mRNA levels (Pitx2c+/–). Resting membrane potential was more depolarized in Pitx2c+/– atria, and TWIK-related acid-sensitive K+ channel 2 (TASK-2) gene and protein expression were decreased. This resulted in enhanced post-repolarization refractoriness and more effective Na-channel inhibition. Defined holding potentials eliminated differences in flecainide’s effects between wild-type and Pitx2c+/– atrial cardiomyocytes. More positive holding potentials replicated the increased effectiveness of flecainide in blocking human Nav1.5 channels in HEK293 cells. Computer modeling reproduced an enhanced effectiveness of Na-channel block when resting membrane potential was slightly depolarized. Conclusions PITX2 mRNA modulates atrial resting membrane potential and thereby alters the effectiveness of Na-channel blockers. Lithium-ion cells are currently the dominant technology in the market for battery-powered electric vehicles. Their biggest downside is their degradation over lifetime and usage, limiting the main vehicle functions such as the driving power and range. Moreover, the battery aging state has a big influence on the expected residual value of electric vehicles. The speed of the degradation depends on specific operating conditions such as temperature, SOC level and current. These stress factors are influenced by the usage behaviour of the vehicle driver as well as possible tuning measures. The aim of this study is to investigate the impact of typical driver behaviour combined with tuning measures on battery aging and the resulting residual value. A fully configurable, longitudinal simulation model is presented for the analysis that covers the main components of an electric drivetrain and takes battery aging into account. The model allows us to simulate various possible driver characteristics and tuning measures. Furthermore, the affiliated regression model allows the establishment of a link between the battery aging induced by driver behaviour and the residual value. First results show that our approach is promising and indicate that driver behaviour has a big influence on battery aging and thus the residual value, this being further reinforced by tuning measures. In addition, there is no linear correlation between the aging induced by driver behaviour and the residual value.

INTRODUCTION: Hemodynamic parameters in zebrafish receive increasing attention because of their important role in cardiovascular processes such as atherosclerosis, hematopoiesis, sprouting and intussusceptive angiogenesis. To study underlying mechanisms, the precise modulation of parameters like blood flow velocity or shear stress is centrally important. Questions related to blood flow have been addressed in the past in either embryonic or ex vivo-zebrafish models but little information is available for adult animals. Here we describe a pharmacological approach to modulate cardiac and hemodynamic parameters in adult zebrafish in vivo. MATERIALS AND METHODS: Adult zebrafish were paralyzed and orally perfused with salt water. The drugs isoprenaline and sodium nitroprusside were directly applied with the perfusate, thus closely resembling the preferred method for drug delivery in zebrafish, namely within the water. Drug effects on the heart and on blood flow in the submental vein were studied using electrocardiograms, in vivo-microscopy and mathematical flow simulations. RESULTS: Under control conditions, heart rate, blood flow velocity and shear stress varied less than ± 5%. Maximal chronotropic effects of isoprenaline were achieved at a concentration of 50 μmol/L, where it increased the heart rate by 22.6 ± 1.3% (n = 4; p < 0.0001). Blood flow velocity and shear stress in the submental vein were not significantly increased. Sodium nitroprusside at 1 mmol/L did not alter the heart rate but increased blood flow velocity by 110.46 ± 19.64% (p = 0.01) and shear stress by 117.96 ± 23.65% (n = 9; p = 0.03). DISCUSSION: In this study, we demonstrate that cardiac and hemodynamic parameters in adult zebrafish can be efficiently modulated by isoprenaline and sodium nitroprusside. Together with the suitability of the zebrafish for in vivo-microscopy and genetic modifications, the methodology described permits studying biological processes that are dependent on hemodynamic alterations.

Especially in grid areas with a high photovoltaic penetration, storage systems can increase the level of self-sufficiency for consumers using privately owned photovoltaic systems as well as offload the grid in times of high photovoltaic generation in order to reduce necessary grid reinforcement. A further application scenario is provision of control power for stabilizing the grid frequency. Storage systems can fulfill single individual abovementioned tasks as well as a combination of several ones, which may result in a variety of different load curves for the storage system. As the aging behavior of the battery cells is strongly influenced by load profiles, the storage application will severely influence lifetime of storage systems. In order to investigate the aging behavior dependent on the application mode, the load curves resulting from the different applications given above are raised for a battery aging simulation. This study presents aging behavior for lithium-ion cells with NMC technology. As a result, the deterioration rate of this cell type is determined dependent on the different application modes. On the basis of these outcomes, conclusions regarding the aging behavior for the different applications are drawn.
