Gehirnintrinsische Zytodegeneration triggert eine Umverteilung von Aquaporin 4 mit sekund��rer Immunzellinvasion Als einer der Hauptregulatoren der Wasserhom��ostase im zentralen Nervensystem wird vermutet, dass Aquaporin 4 (AQP4) eine ma��gebliche Rolle an der Ausbildung neuroinflammatorischer L��sionen hat. Nach der Entdeckung von AQP4 als Autoantigen der Neuromyelitis optica (NMO) ist dessen Rolle am Krankheitsgeschehen in den Fokus der Forschung gelangt. Die vorliegende Arbeit hat das Ziel, Ver��nderungen in der Expression und Verteilung von AQP4 im Kontext von neuroinflammatorischen Tiermodellen, Patienten mit Multipler Sklerose (MS) und pathologischen Prozessen der NMO zu untersuchen. Weibliche C57/BL6 M��use wurden in 4 Gruppen unterteilt. Die Kontrollgruppe wurde mit normalem Nagetierfutter gef��ttert. In der zweiten Gruppe wurde 5 Wochen lang mit 0,25% Cuprizone (Cup) gef��ttert, um einen neurodegenerativen, metabolischen Schaden zu erzeugen. In der dritten Gruppe wurde ��ber eine Injektion von MOG33-55 Experimentelle Autoimmune Encephalomyelitis (EAE) induziert, um eine Autoimmunreaktion zu erzeugen. Die vierte Gruppe erhielt beide Behandlungen (Cup-EAE) und wurde somit 3 Wochen mit Cup und 2 Wochen mit Standardfutter gef��ttert, bevor EAE induziert wurde. Immunhistochemische und Immunfluoreszenz-F��rbungen von Gehirngewebe wurden konventionell und mit neuartigen digitalen histologischen Methoden untersucht. Blutserum aus allen Versuchsgruppen wurde mittels ELISA auf AQP4-Autoantik��rper untersucht. Zus��tzlich wurden post mortem Gehirnschnitte verstorbener Patienten mit progessiver MS analysiert. An Vorderhirnl��sionen der Versuchstiere im Cuprizone-Modell fand sich eine generalisierte Expressionssteigerung von AQP4 in der grauen und vor allem wei��en Substanz. Dieser Effekt konnte auch in L��sionen von PP/SP-MS Patienten nachgewiesen werden. In der grauen Substanz zeigte sich zudem eine generalisierte Umverteilung von AQP4. In der unmittelbaren Umgebung inflammatorischer Gef����e im Cup-EAE Modell fand sich ein fokaler Verlust von AQP4. Diese L��sionen zeigten eine ausgepr��gte Zellinfiltration, unter anderem mit neutrophilen Granulozyten. Die Immunzellinfiltration wurde nach l��ngerer vorangehender Cuprizone-Behandlung gr����er. In den Blutsera der Versuchstiere konnte keine humorale Immunantwort in Form von AQP4-IgG nachgewiesen werden. Die vorliegende Arbeit legt nahe, dass eine prim��r metabolische Sch��digung des Vorderhirns zu einer generalisierten Expressionssteigerung von AQP4 und einem Verlust der AQP4-Polarisation f��hrt. Es fanden sich Hinweise, dass die Invasion peripherer Immunzellen ins Vorderhirn unabh��ngig vom Auftreten von Autoantik��rpern zu einem fokalen Verlust von AQP4 an postkapill��ren Venolen f��hrt. Zusammengefasst unterst��tzen die Ergebnisse dieser Arbeit die sog. ���inside-out��� Hypothese einer prim��r neurodegenerativen Pathogenese der MS. Es konnte gezeigt werden, dass sich das Cup-EAE Modell neben der Untersuchung pathologischer Aspekte der MS auch zur Untersuchung der seronegativen NMO-spectrum-disorders eignet. Zuk��nftige Studien sollten das Cup-EAE Tiermodell noch tiefergehender charakterisieren und prim��r neurodegenerative Prozesse als vielversprechenden Angriffspunkt neuer MS- und NMO-Therapeutika in den Fokus setzen.

Objective: Space for personality development as well as for the development of critical, creative and interdisciplinary thinking is rarely found in medical curricula in Germany. To be prepared for the challenges of modern medicine, future physicians need a visionary mindset. The aim of this study is to determine the need for teaching such content among medical students in the context of visionary elective curricula and to examine these with regard to the desired topics and organizational structure. Methods: This is a cross-sectional study with 236 medical students from all semesters of the Ludwig-Maximilians-University Munich. The survey consists of 50 questions and includes single choice, multiple choice, matrix questions, open-ended questions and Likert scales. Responses were examined using descriptive statistics and compared parametrically in sub-aspects. Results: Three-quarters of respondents would like to see curricular content on interdisciplinary interfaces with other disciplines. A suitable framework for this is seen by 87% of the respondents in a visionary elective curriculum. Students would like to see a broad range of specific content such as global health, politics, business, and computer science. The majority of respondents would like to see 1 unit of instruction per week and would participate in an appropriate program. Such an offering would promote creative (53.6%), critical (63.7%), and interdisciplinary thinking (69.0%) and train to become better physicians (87%). Conclusion: Participants in this study are positive toward the introduction of visionary content in medical school. Faculties should build visionary elective curricula according to the graduate profile requirements of the new NKLM 2.0 to make medical education sustainable., Zielsetzung: Platz f��r Pers��nlichkeitsbildung sowie f��r die Entwicklung kritischen, kreativen und interdisziplin��ren Denkens findet sich nur selten in medizinischen Curricula in Deutschland. Um auf die Herausforderungen der modernen Medizin vorbereitet zu sein, brauchen angehende ��rztinnen/��rzte eine vision��re Geisteshaltung. Ziel der Studie ist es, den Bedarf f��r die Vermittlung solcher Inhalte unter Medizinstudierenden im Rahmen von vision��ren Wahlcurricula zu ermitteln und diese hinsichtlich der gew��nschten inhaltlichen und organisatorischen Struktur zu untersuchen. Methodik: Es handelt sich um eine Querschnittstudie mit 236 Medizinstudierenden aus allen Semestern der Ludwig-Maximilians-Universit��t M��nchen. Die Umfrage besteht aus 50 Fragen und beinhaltet Single-Choice, Multiple Choice, Matrixfragen, offene Fragen und Likert-Skalen. Die Antworten wurden mittels deskriptiver Statistik untersucht und in Teilaspekten parametrisch verglichen. Ergebnisse: Dreiviertel der Befragten w��nschen sich curriculare Inhalte zu interdisziplin��ren Schnittstellen mit anderen Fachrichtungen. Ein geeigneter Rahmen daf��r wird von 87% der Befragten in einem vision��ren Wahlcurriculum gesehen. Die Studierenden w��nschen sich ein breites Angebot spezifischer Inhalte wie Global Health, Politik, Wirtschaft und Informatik. Die Mehrheit der Befragten w��nscht sich einen Zeitumfang von 1 Unterrichtseinheit pro Woche und w��rde an einem entsprechenden Programm teilnehmen. Ein solches Angebot w��rde das kreative (53,6%), kritische (63,7%) und interdisziplin��re Denken (69,0%) f��rdern und zu besseren ��rztinnen/��rzten ausbilden (87%). Schlussfolgerung: Die Teilnehmenden dieser Studie sind gegen��ber der Einf��hrung vision��rer Inhalte im Medizinstudium positiv eingestellt. Fakult��ten sollten entsprechend der Anforderungen des Absolventenprofils des neuen NKLM 2.0 vision��re Wahlcurricula aufbauen, um die Ausbildung von ��rztinnen und ��rzten zukunftsf��hig zu machen.

Probing the quantum world with macroscopic objects has been a core challenge for research in physics during the past decades. Proposed systems to reach this goal include hybrid devices that couple a nanomechanical resonator to a single spin qubit. In particular, the coherent actuation of a macroscopic mechanical oscillator by a single electronic spin would open perspectives in the creation of arbitrary quantum states of motion.In this manuscript, we investigate a hybrid system coupling a nanomechanical oscillator and a single electronic spin of a NV defect in magnetic interaction. We focus on the parametric interaction case, when the mechanical motion modulates the qubit energy, and in particular when the driven qubit and mechanical oscillators evolves on similar timescales. In that situation a synchronization of the qubit dynamics onto the mechanical motion is observed. The phenomenon is first explored on a test experiment where mechanical motion is replaced by a parametrically coupled RF field. It allows to establish the main properties of the phenomenon, which is subsequently investigated on the core experiment. It consists of a NV defect attached at the vibrating extremity of a silicon carbide nanowire, immersed in a strong magnetic field gradient. The bidimensional character of the nanowire deformations is responsible for novel vectorial signatures in the synchronization, which can also be viewed as a phononic Mollow triplet as observed in early quantum electrodynamics experiments. We finally explore the robustness of the synchronization against the Brownian motion of the resonator and demonstrate the possibility to protect the qubit against this additional decoherence source by applying a small coherent mechanical drive.; L'exploration du monde quantique au moyen d'objets macroscopiques constitue l'un des défis centraux de ces dernières décennies pour la recherche en physique. Parmi les systèmes proposés pour atteindre cet objectif, les systèmes hybrides, qui couplent un résonateur nanomécanique à un qubit unique, font figure de paradigme.L'excitation cohérente d'un oscillateur mécanique macroscopique par un unique spin électronique ouvrirait en particulier de nouvelles perspectives pour la création d'états quantiques arbitraires du mouvement.Dans ce manuscrit, nous considérons un système hybride constitué d'un oscillateur nanomécanique et du spin électronique d'un unique centre NV, couplés entre eux par une interaction magnétique. Nous nous concentrons sur le cas d'une interaction paramétrique où la vibration mécanique module l'énergie du qubit, et plus précisément sur le cas où le qubit ainsi forcé et l'oscillateur mécanique évoluent sur des échelles de temps comparables.Dans cette situation, nos observations montrent une synchronisation de la dynamique du qubit sur l'oscillation mécanique. Le phénomène est dans un premier temps abordé par une expérience-test qui remplace le mouvement mécanique par un champ radiofréquence en couplage paramétrique avec le spin. This thesis makes a contribution to autonomous service robots, centered around two aspects. The first is modeling decision making in the face of incomplete information on top of diverse basic skills of a service robot. Second, based on such a model, it is investigated, how to transfer complex decision-making knowledge into the system. Interactive learning, naturally from both demonstrations of human teachers and in interaction with objects, yields decision-making models applicable by the robot.
