Abstract Acetylcholine regulates various cognitive functions through broad cholinergic innervation. However, specific cholinergic subpopulations, circuits and molecular mechanisms underlying recognition memory remain largely unknown. Here we show that Ngfr + cholinergic neurons in the substantia innominate (SI)/nucleus basalis of Meynert (nBM)-medial prefrontal cortex (mPFC) circuit selectively underlies recency judgements. Loss of nerve growth factor receptor (Ngfr −/− mice) reduced the excitability of cholinergic neurons in the SI/nBM-mPFC circuit but not in the medial septum (MS)-hippocampus pathway, and impaired temporal order memory but not novel object and object location recognition. Expression of Ngfr in Ngfr −/− SI/nBM restored defected temporal order memory. Fiber photometry revealed that acetylcholine release in mPFC not only predicted object encounters but also mediated recency judgments of objects, and such acetylcholine release was absent in Ngfr −/− mPFC. Chemogenetic and optogenetic inhibition of SI/nBM projection to mPFC in ChAT-Cre mice diminished mPFC acetylcholine release and deteriorated temporal order recognition. Impaired cholinergic activity led to a depolarizing shift of GABAergic inputs to mPFC pyramidal neurons, due to disturbed KCC2-mediated chloride gradients. Finally, potentiation of acetylcholine signaling upregulated KCC2 levels, restored GABAergic driving force and rescued temporal order recognition deficits in Ngfr −/− mice. Thus, NGFR-dependent SI/nBM-mPFC cholinergic circuit underlies temporal order recognition memory.

IntroductionMagnesium (Mg) is a crucial macronutrient for plants. Understanding the molecular responses of plants to different levels of Mg supply is important for improving cultivation practices and breeding new varieties with efficient Mg utilization.MethodsIn this study, we conducted a comprehensive transcriptome analysis on tobacco (Nicotiana tabacum L.) seedling leaves to investigate changes in gene expression in response to different levels of Mg supply, including Mg-deficient, 1/4-normal Mg, normal Mg, and 4×-normal Mg, with a particular focus on Mg deficiency at 5, 15 and 25 days after treatment (DAT), respectively.ResultsA total of 11,267 differentially expressed genes (DEGs) were identified in the Mg-deficient, 1/4-normal Mg, and/or 4×-normal Mg seedlings compared to the normal Mg seedlings. The global gene expression profiles revealed potential mechanisms involved in the response to Mg deficiency in tobacco leaves, including down-regulation of genes–two DEGs encoding mitochondria-localized NtMGT7 and NtMGT9 homologs, and one DEG encoding a tonoplast-localized NtMHX1 homolog–associated with Mg trafficking from the cytosol to mitochondria and vacuoles, decreased expression of genes linked to photosynthesis and carbon fixation at later stages, and up-regulation of genes related to antioxidant defenses, such as NtPODs, NtPrxs, and NtGSTs.DiscussionOur findings provide new insights into the molecular mechanisms underlying how tobacco responds to Mg deficiency.

Abstract Background Boron (B) is an essential micronutrient for plants. Inappropriate B supply detrimentally affects the productivity of numerous crops. Understanding of the molecular responses of plants to different B supply levels would be of significance in crop improvement and cultivation practices to deal with the problem. Results We conducted a comprehensive analysis of the transcriptome and proteome of tobacco seedlings to investigate the expression changes of genes/proteins in response to different B supply levels, with a particular focus on B deficiency. The global gene and protein expression profiles revealed the potential mechanisms involved in the responses of tobacco to B deficiency, including up-regulation of the NIP5;1-BORs module, complex regulation of genes/proteins related to cell wall metabolism, and up-regulation of the antioxidant machinery. Conclusion Our results demonstrated that B deficiency caused severe morphological and physiological disorders in tobacco seedlings, and revealed dynamic expression changes of tobacco genes/proteins in response to different B supply levels, especially to B deficiency, thus offering valuable insights into the molecular responses of tobacco to B deficiency.

Abstract Background Chronic pain was associated with a higher risk of mental disorders (e.g., depression and anxiety). However, the role of 24-h movement behaviors in the association remains unclear. Methods A total of 72,800 participants with accelerometer data and free of mental disorders from the UK Biobank were analyzed. The compositional mediation model and isotemporal substitution model were used to explore the associations between chronic pain, 24-h movement behaviors, and the incidence of overall mental disorders, depression, and anxiety. Results With a median follow-up of 13.36 years, participants with chronic pain had a higher rate of incident overall mental disorders (hazard ratio (HR): 1.281, 95% confidence interval (CI): 1.219 to 1.344), anxiety (HR: 1.391, 95% CI: 1.280 to 1.536), and depression (HR: 1.703, 95% CI: 1.551 to 1.871). Increased sedentary behavior (SB) and reduced moderate-to-vigorous physical activity (MVPA) caused by chronic pain both increased the risk of mental disorders. Twenty-four-hour movement behaviors explained the relationship between chronic pain and overall mental disorders, depression, and anxiety by 10.77%, 5.70%, and 6.86%, respectively. Interaction effects were found between MVPA and chronic pain when predicting the incidence of depression and between MVPA, sleep (SLP), and chronic pain when predicting the incidence of mental disorders. People with chronic pain would recommend at least 0.5 h per day of MVPA and 7 h per day of SLP and restricting SB below 11.5 h per day. Conclusions Twenty-four-hour movement behaviors played a significant mediating role in the association between chronic pain and mental disorders. Individuals with chronic pain should engage in more MVPA, less sedentary behavior, and have 7-h sleep per day.

Silica nanoparticles (SNPs) induce reproductive toxicity through ROS production, which significantly limits their application. The protective effects of taurine (Tau) against SNP-induced reproductive toxicity remain unexplored. So this study aims to investigate the impact of Tau on SNP-induced porcine ovarian granulosa cell toxicity. In vitro, granulosa cells were exposed to SNPs combined with Tau. The localization of SNPs was determined by TEM. Cell viability was examined by CCK-8 assay. ROS levels were measured by CLSM and FCM. SOD and CAT levels were evaluated using ELISA and qPCR. Cell apoptosis was detected by FCM, and pro-inflammatory cytokine transcription levels were measured by qPCR. The results showed that SNPs significantly decreased cell viability, while increased cell apoptosis and ROS levels. Moreover, SOD and CAT were decreased, while IFN-α, IFN-β, IL-1β, and IL-6 were increased after SNP exposures. Tau significantly decreased intracellular ROS, while it increased SOD and CAT compared to SNPs alone. Additionally, Tau exhibited anti-inflammatory effects and inhibited cell apoptosis. On the whole, these findings suggest that Tau mitigates SNP-induced cytotoxicity by reducing oxidative stress, inflammatory response, and cell apoptosis. Tau may be an effective strategy to alleviate SNP-induced toxicity and holds promising application prospects in the animal husbandry and veterinary industry.

BackgroundPrevious studies using meta-analysis to explore the relationship between air pollution exposure and ischemic stroke (IS) mostly focus on particulate matter-related themes, few include gaseous pollutants in the study, and subgroup analyses of factors such as different lag days, seasons, and genders are rarely been reported.ObjectiveTo quantitatively evaluate the relationships between short-term exposures to 6 common air pollutants, including fine particulate matter (PM2.5), inhalable particulate matter (PM10), carbon monoxide (CO), nitrogen dioxide (NO2), sulfur dioxide (SO2), and ozone (O3), and the incidence of IS.MethodsA systematic search was conducted to collect literature studying the 6 common air pollutants and IS published up to May 1, 2022 in 6 databases (China Journal Full-text Database, China Biology Medicine Disc, PubMed, Cochrane Library, Web of Science, and Embase). Literature quality evaluation was performed using the Newcastle-Ottawa Scale. Stata 16.0 software was used to conduct meta-analysis including heterogeneity test, combined effect size, meta-regression, subgroup analysis, sensitivity analysis, and publication bias test.ResultsA total of 33 articles were qualified for inclusion. The total number of samples included in the literature was 7195631. The meta-analysis results showed that short-term exposures to PM2.5 (OR=1.0082, 95%CI: 1.0049−1.0116), PM10 (OR=1.0017, 95%CI: 1.0008−1.0026), CO (OR=1.0328, 95%CI: 1.0231−1.0426), NO2 (OR=1.0150, 95%CI: 1.0079−1.0222), SO2 (OR=1.0158, 95%CI: 1.0078-1.0238), and O3 (OR=1.0017, 95%CI: 1.0003−1.0032) were associated with an increased risk of IS. PM10 and O3 increased the risk of IS in both lag0 and lag1, while PM2.5, CO, NO2, and SO2 all showed an associated increased risk of IS only in lag0. The results of sensitivity analysis showed stable results for all pollutants studied, and there was no publication bias in the literature on the association of the remaining five pollutants with IS incidence except for the PM2.5-related literature.ConclusionShort-term exposures to PM2.5, PM10, CO, NO2, SO2, and O3 may increase the incidence of IS, with this risk showing the most significant level on the day of IS onset.

Alkali-activated slag (AAS) is a promising cementing material for winter construction due to its continuous hydration at sub-zero temperature. In order to obtain a higher mechanical strength of AAS mortar in winter construction, the most efficient alkali activator should be selected. Potassium silicate and sodium silicate as alkaline activators of AAS mortar possess a high strength and hydration rate at ordinary temperature. Potassium silicate and sodium silicate as alkali activators showed different properties because of different alkali cations, so the effect of alkali cation on the performance of AAS mortar was studied at sub-zero temperature. The mechanical properties of potassium-silicate-activated AAS (K-AAS) mortar and sodium-silicate-activated AAS (N-AAS) mortar were thoroughly compared at the ambient temperatures of −10 °C, 0 °C, and 20 °C. The compressive and flexural strength of K-AAS mortar was increased by 130.4% and 72.3% at the age of 1 day and increased by 49% and 33.7% at the age of 28 days at the ambient temperature of −10 °C compared with N-AAS mortar. In order to reveal the mechanism behind the influence of different alkali cations on the mechanical properties of AAS mortar, the hydration heat, hydration products, and pore structures of AAS were characterized. The hydration heat exhibited that the rate of heat release and the cumulative heat release of K-AAS are higher than N-AAS, indicating the higher reaction degree of K-AAS. DTG/TG and ATR-FTIR spectra showed that K-AAS generated more gel product C(-A)-S-H compared with N-AAS at the same curing temperature. The MIP results exhibited that the porosity of K-AAS was lower than N-AAS. Finally, the mechanism explanation of the effect of alkali cation on the performance of AAS mortar was proposed. To sum up, potassium silicate should be selected as an alkali activator of AAS in winter construction.

Rural folk custom ecotourism is one of the most unique tourism projects today, which relies on the unique landscape of the country for architectural planning and tourism. In the article, the architectural planning and design based on the protective development of rural folk custom ecotourism was studied. Based on this, firstly, the characteristics and development background of rural tourism were expatiated, and the theoretical research background of current landscape planning and design was analyzed; then the principles and methods of planning and designing of rural folk custom ecotourism landscape architecture were analyzed, and the functions of architecture and space use were described in detail, so as to meet the need for protective and sustainable development; in addition, through the analysis and planning of specific projects, the concrete methods of architectural planning and design were discussed and the localization of architectural planning and design was achieved. Background With the rapid progression of aging in China, the challenge of multimorbidity has become a significant concern for both public health and clinical practice. Nocturnal sleep status, including sleep duration and quality, is crucial for regulating body metabolism and physiological functions in the elderly. However, current research on the relationship between nocturnal sleep status and multimorbidity was limited to specific regions or the middle-aged population. The association between nocturnal sleep status and prevalence of multimorbidity among community-dwelling older adults remains ambiguous. Objective To investigate the association between nocturnal sleep status and prevalence of multimorbidity among community-dwelling older adults. Methods In April 2023, a total of 11 917 community-dwelling older adults from the Chinese Longitudinal Healthy Longevity and Happy Family Study (CLHLS-HF, wave 2018) were selected as the participants. Multivariate Logistic regression was used to explore the association of sleep duration and sleep quality with the prevalence of multimorbidity, represented by odds ratios (OR) and 95% confidence intervals (95%CI). Additionally, restrictive cubic splines (RCS) based on Logistic regression analysis were utilized to analyze the dose-response relationship between sleep duration and prevalence of multimorbidity. Results Of the 11 917 community-dwelling older adults, with an average age of (84.4±11.4) years ranged from 65.0 to 117.0 years, 6 477 were females (54.35%) and 5 440 were males (45.65%). Multivariate Logistic regression results after adjusting for covariates such as gender, age, region, and years of education, indicated that compared to those with 6-8 h of nocturnal sleep duration, older adults with shorter sleep duration (
