The UK’s Defence Science and Technology Laboratory (Dstl) is partnering with the US Naval Research Laboratory (NRL) on a joint mission to launch miniature sensors that will advance space weather measurement and modelling capabilities. The Coordinated Ionospheric Reconstruction Cubesat Experiment (CIRCE) comprises two 6U cube-satellites that will be launched into a near-polar low earth orbit (LEO), targeting 500 km altitude, in 2021. The UK contribution to CIRCE is the In situ and Remote Ionospheric Sensing (IRIS) suite, complementary to NRL sensors, and comprising three highly miniaturised payloads provided to Dstl by University College London (UCL), University of Bath, and University of Surrey/Surrey Satellite Technology Ltd (SSTL). One IRIS suite will be flown on each satellite, and incorporates an ion/neutral mass spectrometer, a tri-band global positioning system (GPS) receiver for ionospheric remote sensing, and a radiation environment monitor. From the US, NRL have provided two 1U Triple Tiny Ionospheric Photometers (Tri-TIPs) on each satellite (Nicholas et al., 2019), observing the ultraviolet 135.6 nm emission of atomic oxygen at night-time to characterize the two-dimensional distribution of electrons.

Serosurveys are a key resource for measuring severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) population exposure. A growing body of evidence suggests that asymptomatic and mild infections (together making up over 95% of all infections) are associated with lower antibody titers than severe infections. Antibody levels also peak a few weeks after infection and decay gradually. We developed a statistical approach to produce estimates of cumulative incidence from raw seroprevalence survey results that account for these sources of spectrum bias. We incorporate data on antibody responses on multiple assays from a postinfection longitudinal cohort, along with epidemic time series to account for the timing of a serosurvey relative to how recently individuals may have been infected. We applied this method to produce estimates of cumulative incidence from 5 large-scale SARS-CoV-2 serosurveys across different settings and study designs. We identified substantial differences between raw seroprevalence and cumulative incidence of over 2-fold in the results of some surveys, and we provide a tool for practitioners to generate cumulative incidence estimates with preset or custom parameter values. While unprecedented efforts have been launched to generate SARS-CoV-2 seroprevalence estimates over this past year, interpretation of results from these studies requires properly accounting for both population-level epidemiologic context and individual-level immune dynamics.

Flare frequency distributions represent a key approach to addressing one of the largest problems in solar and stellar physics: determining the mechanism that counter-intuitively heats coronae to temperatures that are orders of magnitude hotter than the corresponding photospheres. It is widely accepted that the magnetic field is responsible for the heating, but there are two competing mechanisms that could explain it: nanoflares or Alfv\'en waves. To date, neither can be directly observed. Nanoflares are, by definition, extremely small, but their aggregate energy release could represent a substantial heating mechanism, presuming they are sufficiently abundant. One way to test this presumption is via the flare frequency distribution, which describes how often flares of various energies occur. If the slope of the power law fitting the flare frequency distribution is above a critical threshold, $\alpha=2$ as established in prior literature, then there should be a sufficient abundance of nanoflares to explain coronal heating. We performed $>$600 case studies of solar flares, made possible by an unprecedented number of data analysts via three semesters of an undergraduate physics laboratory course. This allowed us to include two crucial, but nontrivial, analysis methods: pre-flare baseline subtraction and computation of the flare energy, which requires determining flare start and stop times. We aggregated the results of these analyses into a statistical study to determine that $\alpha = 1.63 \pm 0.03$. ABSTRACT: The environmental regulation of continental fish farming in Brazil is closely linked to land occupation and use. Areas used for this activity are mostly located in permanent preservation areas (PPA), close to rivers and springs, demonstrating the necessity of regulating this activity for its compliance with public policy. Santa Catarina is a leading state in fish farming and one of the first to regulate the use of PPAs. The feasibility of a proposal for environmental compensation to regularize the use of PPAs in Santa Catarina for fish farming; was therefore, assessed. The results showed that 74% of the fish farms analyzed need to regularize the occupation of production structures in PPAs. Regarding the proposed index of compensation for use in PPAs, the environmental compensation factor (ECF), through evaluation of the individual maps and subsequent integrated evaluation, all the fish farms identified with environmental liabilities in occupation had ratios of 1:2 and 1:3, and this index is a viable alternative for environmental regularization of the activity. It was further proposed that ECF be applied to small fish farms of up to four fiscal modules and that they have a standard minimum occupancy profile in relation to the total area of the property, as assessed in the present study.

Many take Exodus 19:3-6 to set out the uniqueness of Israel’s relationship with God (v. 5ab) and the particular role and responsibility Israel has in relation to the rest of the world (v. 6). This raises an important question. Was choosing Israel God’s main priority, or was his intention, from the start, to redeem the whole world, with the choice of Israel part of that wider purpose? This article argues that the translation of the clause in verse 5c, introduced by the Hebrew particle כִּי (_ki_), is relevant to that discussion. In the context of the use of כִּי in the wider OT, it discusses the main interpretations of the כִּי clause in Exodus 19:5c – asseverative, causal, concessive, and explanatory. It concludes in favour of a causal understanding and points to the implications for an understanding of the mission theology of the OT.

Archaeological remains of brachyurans (e.g. crabs) are often overlooked as potential paleoenvironmental and paleoclimatical proxies in contrast to other marine and terrestrial invertebrates such as mollusks and landsnails. The potential for fine-scale regional paleoclimate reconstruction based upon these organisms' behavioral ecology has yet to be examined. We present novel isotopic analyses of the remains of a semiterrestrial freshwater crab (Potamon potamios) endemic to southwest Asia recovered from the archaeological site of Khirbat al-Mudayna al-’Aliya (KMA). KMA lies on a southern tributary of the Wadi al-Mujib in the Hashemite Kingdom of Jordan, approximately 40 km east of the Dead Sea. Excavations here recovered architecture, artifacts, and ecofacts dating to a single-period occupation during the early Iron Age (∼1000 cal BCE). Oxygen and carbon isotopes from these brachyuran remains were analyzed in conjunction with a two-year isotopic and morphometric study of the modern potomonautid population near the archaeological site and at nearby wadi systems to assess whether the isotopic values of modern crab populations correlated with variables such as temperature, including isotopic study of the pools in which crabs were found. In turn, these data could be used to reconstruct paleoclimate or paleoenvironment in the archaeological population. The high correlation in oxygen isotope values between crab carapaces and the water of the pools in which they were sampled suggest that variability in isotopic values of crab specimens recovered from KMA is tracking ancient human capture of these organisms across potentially different pools. Further, intra-year variability in isotopic values in modern crab carapaces as well as the similarity of isotopic values across wadi systems with different environments suggest that P. potamios remains cannot be used as reliable paleoenvironmental indicators. The implications of this research will have significance for archaeologists and other researchers in the study of paleoenvironment and paleoecology, and bears upon human mobility, resource acquisition, and human-nonhuman animal relationships.

Clinical trials for Alzheimers disease (AD) are slower to enroll study participants, take longer to complete, and are more expensive than trials in most other therapeutic areas. The recruitment and retention of a large number of qualified, diverse volunteers to participate in clinical research studies remain among the key barriers to the successful completion of AD clinical trials. An advisory panel of experts from academia, patient-advocacy organizations, philanthropy, non-profit, government, and industry convened in 2020 to assess the critical challenges facing recruitment in Alzheimers clinical trials and develop a set of recommendations to overcome them. This paper briefly reviews existing challenges in AD clinical research and discusses the feasibility and implications of the panels recommendations for actionable and inclusive solutions to accelerate the development of novel therapies for AD.

Antidepressant drugs (ADDs) are one of the most extensively used pharmaceuticals globally. They act at particularly low therapeutic concentrations to modulate monoamine neurotransmission, which is one of the most evolutionary conserved pathways in both humans and animal species including invertebrates. As ADDs are widely detected in the aquatic environment at low concentrations (ng/L to low µg/L), their potential to exert drug-target mediated effects in aquatic species has raised serious concerns. Amitriptyline (AMI) is the most widely used tricyclic ADD, while monoamines, the target of ADDs, are major bioregulators of multiple key physiological processes including feeding, reproduction and behaviour in molluscs. However, the effects of AMI on feeding, reproduction and mating behaviour are unknown in molluscs despite their ecological importance, diversity and reported sensitivity to ADDs. To address this knowledge gap, we investigated the effects of environmentally relevant concentrations of AMI (0, 10, 100, 500 and 1000 ng/L) on feeding, reproduction and key locomotor behaviours, including mating, in the freshwater gastropod, Biomphalaria glabrata over a period of 28 days. To further provide insight into the sensitivity of molluscs to ADDs, AMI concentrations (exposure water and hemolymph) were determined using a novel extraction method. The Fish Plasma Model (FPM), a critical tool for prioritization assessment of pharmaceuticals with potential to cause drug target-mediated effects in fish, was then evaluated for its applicability to molluscs for the first time. Disruption of food intake (1000 ng/L) and reproductive output (500 and 1000 ng/L) were observed at particularly low hemolymph levels of AMI, whereas locomotor behaviours were unaffected. Importantly, the predicted hemolymph levels of AMI using the FPM agreed closely with the measured levels. The findings suggest that hemolymph levels of AMI may be a useful indicator of feeding and reproductive disruptions in wild population of freshwater gastropods, and confirm the applicability of the FPM to molluscs for comparative pharmaceutical hazard identification.

25. Lymph node dissection in lung cancer surgery: a comparison between robot-assisted vs. video-assisted thoracoscopic approach

BackgroundTNM staging is the most important prognosticator for non-small cell lung cancer (NSCLC) patients. Staging has significant implications for the treatment modality for these patients. Lymph node dissection in robot-assisted thoracoscopic (RATS) surgery remains an area of ongoing evaluation. In this study, we aim to compare lymph node dissection in RATS and VATS approach for lung resection in NSCLC patients.MethodsWe retrospectively compiled a database of 717 patients from July 31, 2015–July 7, 2022, who underwent either a wedge resection, segmentectomy or lobectomy. We analysed the database according to lymph node dissection. The database was divided into RATS (n = 375) and VATS (n = 342) procedures.ResultsThe mean number of lymph nodes harvested overall with RATS was 6.1 ± 1.5 nodes; with VATS approach, it was 5.53 ± 1.8 nodes. The mean number of N1 stations harvested was 2.66 ± 0.8 with RATS, 2.36 ± 0.9 with VATS. RATS approach showed statistically higher lymph node dissection rates compared to VATS (p = 0.002). Out of the 375 RATS procedures, 26 (6.4%) patients undergoing a RATS procedure were upstaged from N0/N1 staging to N2. N0/N1–N2 upstaging was reported in 28 of 342 (8.2%) patients undergoing a VATS procedure. The majority of upstaging was seen in N0–N2 disease: 19 of 375 (5%) for RATS and 23 of 342 (6.7%) for VATS.ConclusionsWe conclude that in RATS procedures, there is a higher rate of lymph node dissection compared to VATS procedures. Upstaging was mostly seen in N0–N2 disease, this was observed at a higher rate with VATS procedures.

Recent research into critical pedagogy supports the implementation of performance-based practices into the classroom. This qualitative research explored the pragmatic ways in which youth, ages 8-17, are taught in Canada's, specifically Ontario's, education system on topics of difference and power. Through semi-structured interviews, four elementary and high school educators described their experiences using performance-based teaching in the classroom. A thematic analysis revealed that teachers found including such practices empowered students and influenced their understanding of their own identities as well as the systemic oppression of marginalized groups.

35. HARMONI : advancing science with the next-generation integral field spectrograph

We start by investigating the noise behaviour of Teledyne HxRG infrared detectors. We look at data from KMOS H2RG darks, along with data from a MOONS engineering grade H4RG detector. From these we determine read noise distributions for the HxRG detectors, and identify a number of other non-uniform noise contributions. We then incorporate this information, along with a pre-existing HxRG noise generation tool, into HSIM. We then use this new advanced detector systematics (ADS) mode to produce simulations investigating the point source sensitivities, spatial variation of noise, reproducibility of recovered point source flux, and signal-to-noise ratio (SNR) for a number of different noise components. We determine from this that these additional detector noise effects provide non-trivial contributions to the noise profile of the detectors. In particular we find pink noise to be problematic, as it is a spatially-varying noise component that has a large impact on the overall SNR. We recommend careful use of the detector reference pixels to correct this, accounting for the potential for bad pixels within these. We also look at the impact of an under-illuminated spectrograph entrance slit on telluric correction. We model the SINFONI 250x125 mas H-band line spread function (LSF) and vary the level of the input illumination. From this we determine an expected reduction in LSF width of 14%. We then use observations of CD-23 13701 and Callisto taken with SINFONI with adaptive optics (AO) correction to confirm a result of 14.0+/-2.1% when fitted using molecfit. We attempt four methods of telluric correction; directly using CD-23 13701 as a telluric standard star, a synthetic spectrum created from molecfit run on CD-23 13701, this synthetic spectrum convolved with the appropriate LSF, and a synthetic spectrum created from molecfit run on the Callisto data. We perform this analysis on multiple spaxels within the Callisto data, and determine average residual sum of squares of 0.43, 0.34, 0.30, and 0.29 for the four methods. We determine from this that inadequate knowledge of the LSF leads to systematically worse telluric correction. Provided the LSF is known, the level of correction is similar between using a telluric standard star or the science target itself, with the remaining differences partly explained by variations in the atmospheric profile between the observations. We end with testing a science case of HARMONI; recovering spatially resolved kinematics from high redshift galaxies. We use the NUTFB simulation to create a galaxy at an effective redshift of 1.44. We then produce a number of simulations using HSIM to determine how well we can recover the rotation curve and star formation rate (SFR), varying the simulated observation time and spectral resolution. We determine that 1 hour of HARMONI observation is sufficient to recover the kinematics to the point at which we become limited by effects within the galaxy. We also investigate the impact of the AO point spread function (PSF), particularly showing the effects of the PSF wings smearing out the rotation curve, as well as differences in the kinematics as a result of PSF elongation.

36. Extracellular trafficking of Wnt signals in gastric cancer

Wnt proteins are secreted glycoproteins which signal in a tissue to regulate multiple cellular processes, such as cell differentiation, migration, and proliferation. However, post-translational modifications result in Wnt ligands being hydrophobic in nature. Thus, their ability to freely diffuse in the aqueous extracellular environment is restricted, and alternative mechanisms of transport have been proposed. In this thesis, I investigate and characterise the use of signalling filopodia - termed cytonemes - in the intercellular transport of Wnt3 ligands by gastric cancer cells, which display overactivated Wnt/β-catenin signalling. Additionally, I identify the membrane scaffolding protein Flotillin-2 (Flot2), which is overexpressed in gastric cancers, as a novel positive regulator of Wnt cytoneme formation and consequently proliferation. Mechanistically, I show that Flot2 is required for the intracellular transport, membrane localisation and thus signalling of the Wnt co-receptor Ror2; a known regulator of Wnt cytonemes. In parallel, I show that Flot2 also has a function in transducing signals in the Wnt- receiving cell. Here, Flot2 co-localises with the Wnt co-receptor Lrp6 and is involved in its endocytic uptake. Additionally, Flot2 knockdown results in the perinuclear accumulation of Lrp6 and its absence from recycling endosomes. Therefore, I suggest Flot2 may also be involved in the endosomal transport of Lrp6 following internalisation. Finally, following my observed co-localisations of both Ror2 and Lrp6 with Flot2, I found that these Wnt co-receptors co-localise with one another, as well as the cognate Wnt receptor Frizzled 7, in Flot2 microdomains. Expression of a mutant Ror2 missing its cysteine-rich domain, however, causes loss of co-localisation with Lrp6 and perturbed Wnt/β-catenin signalling. Together, these findings led me to propose a model whereby Frizzled 7, Ror2 and Lrp6 all interact and form one large complex, which I have termed the Wnt Receptor Supercomplex (WRS). I hypothesise that these receptors may interact, even in the absence of Wnt ligands, to regulate one another's binding affinities for either Wnt/β-catenin or Wnt/PCP ligands. Here, I propose that flotillin microdomains provide the scaffold necessary for these interactions.

Background: Restoration of blood supply is a desired goal for the treatment of acute ischemic stroke. However, the restoration often leads to cerebral ischemia-reperfusion injury (CIR/I), which greatly increases the risk of non-neural organ damage. In particular, the acute kidney injury might be one of the most common complications. Aims: The study aimed to understand the damage occurred and the potential molecular mechanisms. Methods: The study was explored on the CIR/I rats generated by performing middle cerebral artery occlusion/reperfusion (MCAO/Reperfusion). The rats were evaluated with injury on the brains, followed by the non-neural organs including kidneys, livers, colons and stomachs. They were examined further with histopathological changes, and gene expression alterations by using RT-qPCR of ten aquaporins (Aqps) subtypes including Aqp1~Aqp9 and Aqp11. Furthermore, the Aqps expression profiles were constructed for each organ and analyzed by performing Principle Component Analysis. In addition, immunohistochemistry was explored to look at the protein expression of Aqp1, Aqp2, Aqp3 and Aqp4 in the rat kidneys. Results: There was a prominent down-regulation profile in the MCAO/Reperfusion rat kidneys. The protein expression of Aqp1, Aqp2, Aqp3 and Aqp4 was decreased in the kidneys of the MCAO/Reperfusion rats. We suggested that the kidney was in the highest risk to be damaged following the CIR/I. Down-regulation of Aqp2, Aqp3 and Aqp4 was involved in the acute kidney injury induced by the CIR/I.

The sheer volume of financial statements makes it difficult for humans to access and analyze a business's financials. Robust numerical reasoning likewise faces unique challenges in this domain. In this work, we focus on answering deep questions over financial data, aiming to automate the analysis of a large corpus of financial documents. In contrast to existing tasks on general domain, the finance domain includes complex numerical reasoning and understanding of heterogeneous representations. To facilitate analytical progress, we propose a new large-scale dataset, FinQA, with Question-Answering pairs over Financial reports, written by financial experts. We also annotate the gold reasoning programs to ensure full explainability. We further introduce baselines and conduct comprehensive experiments in our dataset. The results demonstrate that popular, large, pre-trained models fall far short of expert humans in acquiring finance knowledge and in complex multi-step numerical reasoning on that knowledge. Our dataset -- the first of its kind -- should therefore enable significant, new community research into complex application domains. As SARS-CoV-2 continues to spread and vaccines are rolled-out, the "double burden" of disparities in exposure and vaccination intersect to determine patterns of infection, immunity, and mortality. Serology provides a unique opportunity to measure prior infection and vaccination simultaneously. Leveraging algorithmically-selected residual sera from two hospital networks in the city of San Francisco, cross-sectional samples from 1,014 individuals from February 4-17, 2021 were each tested on two assays (Ortho Clinical Diagnostics VITROS Anti-SARS-CoV-2 and Roche Elecsys Anti-SARS-CoV-2), capturing the first year of the epidemic and early roll-out of vaccination. We estimated, using Bayesian estimation of infection and vaccination, that infection risk of Hispanic/Latinx residents was five times greater than of White residents aged 18-64 (95% Credible Interval (CrI): 3.2-10.3), and that White residents over 65 were twice as likely to be vaccinated as Black/African American residents (95% CrI: 1.1-4.6). We found that socioeconomically-deprived zipcodes had higher infection probabilities and lower vaccination coverage than wealthier zipcodes. While vaccination has created a 'light at the end of the tunnel' for this pandemic, ongoing challenges in achieving and maintaining equity must also be considered.
