Purpose: Currently, the significance of CDKN2A/B mutations in the pathogenesis and prognosis of acute lymphoblastic leukemia (ALL) is inconclusive. In this study, we analyzed the genetic and clinical features of children with CDKN2A/B mutations in ALL. In addition, we evaluated the expression and significance of programmed cell death protein 1 (PD-1) and programmed cell death ligand 1 (PD-L1) in serum and explored their role in the susceptibility of childhood ALL. Methods: We sequenced CDKN2A/B in the peripheral blood of 120 children with ALL and 100 healthy children with physical examination. The levels of CD4+ T, CD8+ T, and NK cells were measured by flow cytometry (FCM). Furthermore, the expression of PD-1 and PD-L1 was detected by ELISA. Results: We found 32 cases of CDKN2A rs3088440 and 11 of CDKN2B rs2069426 in 120 ALL children. Children with ALL in the CDKN2A rs3088440 were more likely to have hepatosplenomegaly (P = 0.019) and high risk (P = 0.014) than the wild group. In contrast, CDKN2B rs2069426 was more likely to develop lymph node metastasis (P = 0.017). The level of PD-L1 in the serum of ALL children was significantly higher than that of the control group, and there was no significant difference in PD-1 (P < 0.001). Additionally, children with CDKN2A rs3088440 had reduced CD8+ T cell counts than the wild group (P = 0.039). Conclusion: CDKN2A rs3088440 and CDKN2B rs2069426 may be related to the occurrence and development of ALL in Chinese children. Recently, mobile apps are rapidly emerging as an important information instrument, with the potential to boost convenience and efficiency in everyday life. The adoption of mobile apps can exert a positive influence on individuals, but also lead to adverse perceptions in different ways. The crucial issue arising is what motivates people's discontinued use of such services. Furthermore, the roles of communication overload and privacy invasion between network externalities and discontinued use intentions have not been thoroughly examined. The primary objective of this article is to investigate if negative network externalities may result in privacy invasion, communication overload and discontinued intentions, and how the underlying mechanism operates. This current research collected and evaluated data from 696 mobile app users utilizing the structural equation model (SEM) technique. The findings demonstrate that perceived critical mass and perceived complementarity positively affect the privacy invasion of mobile app users. Particularly, it was discovered that privacy invasion and communication overload mediate the association between network externalities and mobile app discontinued use intentions. This article may not only enrich the ongoing contemporary critical discussion on new information technology usage, but also offer significant theoretical and practical implications for mobile app researchers and practitioners. Objectives: To reconstruct the genomic epidemiology and evolution of MDR Salmonella Indiana in China.Methods: A total of 108 Salmonella Indiana strains were collected from humans and livestock in China. All isolates were subjected to WGS and antimicrobial susceptibility testing. Phylogenetic relationships and evolutionary analyses were conducted using WGS data from this study and the NCBI database.Results: Almost all 108 Salmonella Indiana strains displayed the MDR phenotype. Importantly, 84 isolates possessed concurrent resistance to ciprofloxacin and cefotaxime. WGS analysis revealed that class 1 integrons on the chromosome and IncHI2 plasmids were the key vectors responsible for multiple antibiotic resistance gene (ARG) [including ESBL and plasmid-mediated quinolone resistance (PMQR) genes] transmission among Salmonella Indiana. The 108 Salmonella Indiana dataset displayed a relatively large core genome and ST17 was the predominant ST. Moreover, the global ST17 Salmonella Indiana strains could be divided into five distinct lineages, each of which was significantly associated with a geographical distribution. Genomic analysis revealed multiple antimicrobial resistance determinants and QRDR mutations in Chinese lineages, which almost did not occur in other global lineages. Using molecular clock analysis, we hypothesized that ST17 isolates have existed since 1956 and underwent a major population expansion from the 1980s to the 2000s and the genetic diversity started to decrease around 2011, probably due to geographical barriers, antimicrobial selective pressure and MDR, favouring the establishment of this prevalent multiple antibiotic-resistant lineage and local epidemics.Conclusions: This study revealed that adaptation to antimicrobial pressure was possibly pivotal in the recent evolutionary trajectory for the clonal spread of ST17 Salmonella Indiana in China. Hydrochar aging may mitigate the adverse effects of direct hydrochar application on plant growth. However, little is known about the strategies for accelerating aging process and the microbiological mechanisms involved. This study explored the effects of three aging strategies, including the addition of straw and efficient-degrading microorganisms, on the aging processes of food waste-derived hydrochar, and identified the key microbial drivers. The bacterial and fungal sequencing was performed to estimate the species composition in hydrochar at different aging periods (i.e., day 1, 7, 14 and 35). The results showed that the addition of straw and microbial inoculum improved the maximum reaction temperature by 13%, and shortened the aging time by ∼30%. Three bacterial guilds, mainly including Bacillus -like species, were identified that showed significant correlations with the nitrogen and phosphorus dynamics during aging. This study demonstrates the feasibility of manipulating key microbial guilds artificially to achieve efficient harmless-treatment of hydrochar. Rare bacteria in rhizospheres from natural watermelon vs chimeric watermelon were investigated to understand the rare rhizobacteria assembling processes along with plant development and effects of rare species on functional stability. Over 80% of the total OTUs were defined as rare taxa (i.e., transient, permanent, and conditionally rare) in the rhizosphere. Among these three rarities, transient rare taxa possessed the highest richness, while the conditionally rarity displayed the largest variations along growth stages and exhibited the greatest deterministic process for assembling. Pairs of conditionally rare taxa with high asynchrony and similar functional potentials were identified in the rhizobacterial communities. This suggests the functional stability of the rhizosphere through functional redundancy. The number of pairs within the rhizosphere of chimeric plant was ~ two fold higher than that of natural watermelon, which illustrated that chimeric plants can stabilize rhizospheric functions. Objective. To investigate the protective effects and regulatory mechanisms of ferulic acid on oxidative stress-induced cardiomyocyte injury. Methods. We established a cardiomyocyte oxidative stress cell model by H2O2 treatment and a mouse heart injury model by isoprenaline infusion of male C57BL/6 mice. Ferulic acid was applied to treat oxidative stress-induced cardiomyocyte injury. DHE staining was used to detect ROS production. DNA fragmentation, TUNEL assay, and cleaved caspase-3 were used to analyze cell apoptosis. Real-time PCR and Western blotting were used to analyze miRNA and protein levels to investigate the regulatory mechanisms of ferulic acid on oxidative stress-induced cardiomyocyte injury. Results. Ferulic acid pretreatment significantly inhibited H2O2- and isoprenaline-induced oxidative stress and cell apoptosis by promoting miR-499-5p expression and inhibiting p21 expression. MiR-499-5p inhibition reversed the protective effects of ferulic acid. Further study found that ferulic acid could also attenuate isoprenaline-induced mouse heart fibrosis and cell apoptosis by reducing oxidative stress, inflammation, and apoptosis in vivo. Conclusions. We proved that ferulic acid protects cardiomyocytes from oxidative stress-induced injury by regulating the miR-499-5p/p21signaling pathway, which provides insight into the clinical application of ferulic acid in the treatment of cardiovascular diseases. We investigated the prevalence and transmission of NDM-producing Enterobacteriaceae in fecal samples of geese and environmental samples from a goose farm in southern China. The samples were cultivated on MacConkey agar plates supplemented with meropenem. Individual colonies were examined for blaNDM, and blaNDM-positive bacteria were characterized based on whole-genome sequencing (WGS) data from the Illumina and Oxford Nanopore Technologies (ONT) platforms. Of 117 samples analyzed, the carriage rates for New Delhi metallo-b-lactamase (NDM)-positive Enterobacteriaceae were 47.1, 18, and 50% in geese, inanimate environments (sewage, soil, fodder, and dust), and mouse samples, respectively. Two variants (blaNDM-1 and blaNDM-5, in 4 and 40 isolates, respectively) were found among 44 blaNDM-positive Enterobacteriaceae; these variants belonged to eight species, and Escherichia coli was the most prevalent (50%). WGS analysis revealed that blaNDM coexisted with diverse antibiotic resistance genes (ARGs). Population structure analysis showed that most E. coli and Enterobacter sp. isolates were highly heterogeneous, while most Citrobacter sp. and P. stuartii isolates possessed extremely high genetic similarities. In addition, blaNDM-5-positive ST4358/ST48 E. coli isolates were found to be clonally spread between geese and the environment and were highly genetically similar to those reported from ducks, farm environments, and humans in China. Plasmid analysis indicated that IncX3 pHNYX644-1-like (n=40) and untypeable pM2-1-like plasmids (n = 4) mediated blaNDM spread. pM2-1-like plasmids possessed diverse ARGs, including blaNDM-1, the arsenical and mercury resistance operons, and the maltose operon. Our findings revealed that the goose farm is a reservoir for NDM-positive Enterobacteriaceae. The blaNDM contamination of wild mice and the novel pM2-1-like plasmid described here likely adds to the risk for dissemination of blaNDM and associated resistance genes. Carbapenem-resistant bacteria, in particular NDM-producing Enterobacteriaceae, have become a great threat to global public. These bacteria have been found not only in hospital and community environments but also among food animal production chains, which are recognized as reservoirs for NDM-producing Enterobacteriaceae. However, the dissemination of NDM-producing bacteria in waterfowl farms has been less well explored. Our study demonstrates that the horizontal spread of blaNDM-carrying plasmids and the partial clonal spread of blaNDM-positive Enterobacteriaceae contribute to the widespread contamination of blaNDM in the goose farm ecosystem, including mice. Furthermore, we found a novel and transferable blaNDM-1-carrying multidrug resistance (MDR) plasmid that possessed multiple environmental adaptation-related genes. Grafting is a basic technique which is widely used to increase yield and enhance biotic and abiotic stress tolerance in plant production. The diversity and interactions of rhizobacterial assemblages shaped by grafting are important for the growth of their hosts but remain poorly understood. To test the hypothesis that plant grafting shapes complexity and co-occurrence of rhizobacterial assemblage, four types of plants, including ungrafted bottle gourd (B), ungrafted watermelon (W), grafted watermelon with bottle gourd rootstock (W/B), and grafted bottle gourd with watermelon rootstock (B/W), were cultivated in two soil types in a greenhouse, and the rhizosphere bacterial communities were analyzed by 16S rRNA gene high-throughput sequencing. Both the soil type and grafting significantly influenced the bacterial community composition. Grafting increased bacterial within-sample diversity in both soils. Core enriched operational taxonomic units (OTUs) in the W/B rhizosphere compared with the other three treatments (B, W, and B/W) were mainly affiliated with Alphaproteobacteria, Deltaproteobacteria, and Bacteroidetes, which are likely related to methanol oxidation, methylotrophy, fermentation, and ureolysis. Co-occurrence network analysis proved that grafting increased network complexity, including the number of nodes, edges, and modules. Moreover, grafting strengthened the structural robustness of the network in the rhizosphere, while ungrafted watermelon had the lowest network robustness. Homogeneous selection played a predominant role in bacterial community assembly, and the contribution of dispersal limitation was increased in grafted watermelon with bottle gourd rootstock. Grafting increased the diversity and transformed the network topology of the bacterial community, which indicated that grafting could improve species coexistence in the watermelon rhizosphere. Objectives: To investigate the prevalence and transmission of mcr-3 among Salmonella enterica serotype Typhimurium and 1,4,[5],12:i:-.Methods: A total of 4724 clinical Salmonella isolates were screened for the presence of mcr-3 in China during 2014-19. The clonal relationship of the mcr-3-positive isolates and their plasmid contents and complete sequence were also characterized based on WGS data from the Illumina and MinION platforms.Results: We identified 10 mcr-3-positive isolates, and all were MDR, mostly resistant to colistin, cefotaxime, ciprofloxacin, doxycycline and florfenicol. mcr-3 was co-present with blaCTX-M-55-qnrS1 on hybrid ST3-IncC-FII conjugatable plasmids (n = 6) and an ST3-IncC non-conjugatable plasmid (n = 1) and embedded into a pCHL5009T-like IncFII plasmid on the Salmonella chromosome (n = 3). Four distinctive genetic contexts surrounded mcr-3 and all but one were closely related to each other and to the corresponding region of IncFII plasmid pCHL5009T. IS15DI was most likely the vehicle for integration of mcr-3-carrying IncFII plasmids into ST3-IncC plasmids and the chromosome and for shaping the MDR regions. In addition, a phylogenetic tree based on the core genome revealed a unique Salmonella lineage (≤665 SNPs) that contained these 10 mcr-3-positive isolates and another 38 (33 from patients) mcr-3-positive Salmonella from five countries. In particular, most of the 51 mcr-3-positive isolates belonged to ST34 and harboured diverse antibiotic resistance genes (ARGs), including mcr-3-blaCTX-M-55-qnrS1, and possessed similar ARG profiles.Conclusions: Our findings revealed global clonal spread of MDR ST34 Salmonella from clinical isolates co-harbouring mcr-3 with blaCTX-M-55 and qnrS1 and a flexibility of mcr-3 co-transmittance with other ARGs mediated by mobile genetic elements. The usage of doxorubicin is hampered by its life-threatening cardiotoxicity in clinical practice. Dexrazoxane is the only cardioprotective medicine approved by the FDA for preventing doxorubicin-induced cardiac toxicity. Nevertheless, the mechanism of dexrazoxane is incompletely understood. The aim of our study is to investigate the possible molecular mechanism of dexrazoxane against doxorubicin-induced cardiotoxicity. We established a doxorubicin-induced mouse and cardiomyocyte injury model. Male C57BL/6J mice were randomly distributed into a control group (Con), a doxorubicin treatment group (DOX), a doxorubicin plus dexrazoxane treatment group (DOX+DEX), and a dexrazoxane treatment group (DEX). Echocardiography and histology analyses were performed to evaluate heart function and structure. DNA laddering, qRT-PCR, and Western blot were performed on DOX-treated cardiomyocytes with/without DEX treatment in vitro. Cardiomyocytes were then transfected with miR-17-5p mimics or inhibitors in order to analyze its downstream target. Our results demonstrated that dexrazoxane has a potent effect on preventing cardiac injury induced by doxorubicin in vivo and in vitro by reducing cardiomyocyte apoptosis. MicroRNA plays an important role in cardiovascular diseases. Our data revealed that dexrazoxane could upregulate the expression of miR-17-5p, which plays a cytoprotective role in response to hypoxia by regulating cell apoptosis. Furthermore, the miRNA and protein analysis revealed that miR-17-5p significantly attenuated phosphatase and tensin homolog (PTEN) expression in cardiomyocytes exposed to doxorubicin. Doxorubicin, as a first line chemotherapeutic agent, its usage is limited owing to cardiotoxicity. Necroptosis is a new form of programmed cell death, and recent investigations indicated that necroptosis is vitally involved in serious cardiac pathological conditions. Dexrazoxane is the only cardiac protective drug approved by FDA for anthracycline. We aimed to explore whether and how dexrazoxane regulates doxorubicin-induced cardiomyocyte necroptosis. First, doxorubicin could cause heart failure and reduce cardiomyocyte viability by promoting cell apoptosis and necroptosis in vivo and in vitro. Second, necroptosis plays an important role in doxorubicin induced cardiomyocyte injury, which could be inhibited by Nec-1. Third, dexrazoxane increased cell viability and protect heart function by decreasing both cardiomyocyte apoptosis and necroptosis after doxorubicin treatment. Forth, dexrazoxane attenuated doxorubicin-induced inflammation and necroptosis by the inhibition of p38MAPK/NF-κB pathways. In this work, ultrasound (US) irradiation assisted powder zero valent iron (ZVI) corrosion was conducted for peroxymonosulfate (PMS) activation. PMS activation activity was evaluated by Rhodamine-B (Rh B) degradation efficiency. The other US/PMS, ZVI/PMS, Fe2+/PMS and US/PMS/Fe2+ systems were performed to investigate the synergistic effects of US and ZVI on PMS activation activity. US/PMS/ZVI system performed the highest activity of Rh B degradation. 99.76% of Rh B was removed within 12 min under the optimum condition (pH = 4.5, ZVI dosage = 1 g/L, PMS = 1 mM, US power = 50 W). A comparison of US/PMS/ZVI system with PMS/ZVI and homogeneous US/PMS/Fe2+ systems for Rh B degradation was conducted, indicating that ultrasound irradiation contributed to acceleration of ZVI corrosion and removal of passive films. Besides, the electrons transformation from Fe0 to PMS made main contribution to the generation of sulfate radical, which resulted in the ability of degradation of Rh B. ZVI could be reused for five cycles with efficient activity of PMS activation. Exhaustion of ZVI led to the decrease in PMS activation at the sixth recycle. Thus, this work presents important information on understanding reuse of iron fillings for PMS/PS activation in practical application of pollution remediation. Graphical abstract Abstract Aluminum hydroxide and oxide have been widely used for decontamination due to their environmentally friendly nature and cost effectiveness. Aluminum (hydro) oxides are the main phases of aluminum-derived environment materials. Herein, the solvent-free hydrothermal synthesis of gamma-aluminum oxide (γ-Al 2 O 3) nanoparticles and phase transformation of AlOOH into γ-Al 2 O 3 are reported. Hydrothermal treatment of NH 3 ·H 2 O-induced aluminum precipitate resulted in the formation of AlOOH, which was an intermediate product of γ-Al 2 O 3. AlOOH was transformed into highly crystalline 20-nm γ-Al 2 O 3 particles through calcination at 500 °C due to dehydration. The transformation was confirmed through X-ray diffraction (XRD) and thermogravimetric (TG) analyses. The resulting γ-Al 2 O 3 had superior adsorption ability for the anionic Congo red (CR) dye than for the cationic methylene blue (MB) and malachite green (MG) dyes. The selective adsorption ability of CR instead of MB was attributed to the electrostatic attraction and hydrogen bonds between the amino group and azo double bond of CR, and between the amino group and hydroxyl group in γ-Al 2 O 3. Thus, this study investigated crystalline phase transformation into γ-Al 2 O 3 and selective adsorption capacity of CR, which provides important information regarding the synthesis of crystalline γ-Al 2 O 3 adsorbent, with selective adsorption ability for decontamination applications. Kainic acid (KA) was recently identified as an epileptogenic and neuroexcitotoxic agent that is responsible for inducing learning and memory deficits in various neurodegenerative diseases, such as Alzheimer's disease (AD). However, the mechanism by which KA acts upon AD remains unclear. To this end, we presently investigated the roles of KA in processing amyloid-β protein precursor (AβPP) and amyloid-β protein (Aβ) loads during the course of AD development and progression. Specifically, KA treatment clearly caused the upregulation of tumor necrosis factor α (TNF-α) via activation of the PI3-K/AKT, ERK1/2, and p65 pathways in glial cells. TNF-α secreted from glial cells was then found to be responsible for stimulating the expression of BACE-1 and PS1/2, which resulted in the production and deposition of Aβ in neurons. Finally, the accumulation and aggregation of Aβ lead to the cognitive decline of APP23 mice. Aim. To investigate the function of Tremella fuciformis polysaccharides (TFPS) in LPS-induced inflammation and oxidative stress of macrophages. Methods. RAW264.7 cells were pretreated with TFPS and then stimulated with 0.1 μg/ml LPS. NFκB, Akt, p38MAPK, MCP-1, and SOD-1 were analyzed by Western blotting. Cell viability was measured using MTT assays. Reactive oxygen species (ROS) production, real-time PCR, ELISA, and immunofluorescence staining were performed on RAW264.7 cells that were treated with LPS and/or TFPS to investigate the anti-inflammatory effect of TFPS. Results. LPS induced inflammation and ROS production and promoted the secretion of cytokines such as TNF-α and IL-6. LPS also enhanced the nuclear translocation of NFκB, which promoted inflammation by oxidative stress. However, pretreatment with TFPS profoundly inhibited the activation of Akt, p38MAPK, and NFκB and attenuated the expression of MCP-1 in macrophages. Meanwhile, TFPS also decreased cytokine and ROS levels and attenuated cell inflammation after treatment with LPS. Moreover, miR-155, one of the key small RNAs which regulate NFκB and inflammation in macrophages, was significantly downregulated. Conclusion. TFPS inhibits LPS-induced oxidative stress and inflammation by inhibiting miR-155 expression and NFκB activation in macrophages, which suggests that TFPS may be a potential reagent for inhibiting the development of inflammation. Companion animals including dogs are one of the important components in One Health. Parasites may cause not only diseases in pet animals but also many zoonotic diseases infecting humans. In this study, we performed a survey of intestinal parasites in fecal specimens ( n = 485) collected from outpatient pet dogs with diarrhea in Beijing, China, for the entire year of 2015 by microscopic examination (all parasites) and SSU rRNA-based nested PCR detection ( Giardia and Cryptosporidium). We observed a total of 124 (25.6%) parasite-positive specimens that contained one or more parasites, including Giardia duodenalis (12.8%), Cryptosporidium spp. (4.9%), Cystoisospora spp. (4.3%), trichomonads (4.3%), Toxocara canis (3.5%), Trichuris vulpis (0.6%), and Dipylidium caninum (0.2%). Among the 55 dog breeds, infection rates were significantly higher in border collies and bulldogs, but lower in poodles ( p < 0.05). Risk factor analysis suggested that age was negatively correlated with the infection rate ( p < 0.00001), while vaccination and deworming in the past 12 months could significantly reduce the parasite infections ( p < 0.01). Among the 62 Giardia-positive specimens, 21 were successfully assigned into assemblages using glutamate dehydrogenase ( gdh) and/or beta-giardin ( bg) genes, including assemblage D ( n = 15), C ( n = 5), and F ( n = 1). Among the 24 Cryptosporidium-positive specimens by SSU rRNA PCR, 20 PCR amplicons could be sequenced and identified as Cryptosporidium canis ( n = 20). Collectively, this study indicates that parasites are a significant group of pathogens in companion dogs in Beijing, and companion dogs may potentially transmit certain zoonotic parasites to humans, particularly those with weak or weakened immunity. Various nanotube- and graphene-based materials have been used in a wide range of applications associated with nanoconfined ions and these nanomaterial designers aim to learn more about the underlying mechanisms of ion behaviour at the nanoscale through molecular simulation. In the present work, we summarised recent progress of molecular simulation studies on the nanoconfined ionic transport behaviour in aqueous solutions. With regard to nanomaterial design, the significance of molecular simulation studies and the derived important design principles were selectively highlighted in three major applications, including separation based on nanoporous membranes, electrochemical-related energy storage and DNA nanopore sequencing. Using external stimuli to control ionic transport behavior in nanopores is an important focus of nanodevice research allowing numerous potential applications. A novel strategy was investigated to control ionic-transport resistance responses to the electric field intensity in a nanopore and regulate ionic selectivity. Molecular simulations were performed to examine the transport behaviors of K+ and Na+ in a carbon nanotube-based biomimetic nanopore under different electric fields. The responses of the modified group to the electric field intensity induced variations in ionic transport resistance, which then led to improved ionic selectivity. Object tracking is one of the most challenging problems in computer vision. Only Fast trackers can satisfy the real-time requirements and can be used in many artificial intelligence applications. Due to the impressive high-speed, correlation filters have received much attention within the field of object tracking. Recently, trackers using luminance information or color names for image description have been performed in a correlation filters framework. In this paper, we propose the usage of discriminative color descriptors to improve the tracking performance of the traditional correlation filters tracker. Discriminative color descriptors are compact and efficient. Moreover, our tracker incorporates scale estimation into the traditional correlation filters, which results in increased tracking performance. Extensive experiments demonstrate that the proposed tracker can obtain superior results compared to existing trackers using correlation filters and it is also able to outperform state-of-the-art trackers on the CVPR2013 object tracking benchmark. A magnetically recyclable nanocomposite, manganese oxide octahedral molecular sieve (OMS-2) coated Fe3O4 nanoparticles, was prepared using a facile solvent-free process with manganese salts and Fe3O4 at a low temperature. XRD, SEM, TEM, FT-IR, XPS and N2 adsorption–desorption isotherm analysis showed that in the core–shell nanocomposite, OMS-2 was homogeneously distributed on the Fe3O4 surface with similar structures to that of pure OMS-2 but a much higher surface area of 251 m2 g−1. The catalyst exhibited excellent activity toward the degradation of Acid Orange 7 (AO7), Reactive Brilliant Blue KN-R, Reactive Brilliant Red X-3B, methylene blue, methyl orange, and rhodamine B in the presence of peroxymonosulfate (PMS), with nearly complete decolorization within 15 min. The catalyst could also be easily recovered with the assistance of an external magnetic field, and could be reused for ten successive cycles without any change of structure. The loss of Mn ions in solution was lower than 0.05 mg L−1 and no Fe ions were detected. The effect of several parameters, including the OMS-2 content in the nanocomposites, the concentration of AO7, PMS and the catalyst, and solution pH, on the degradation efficiency was also discussed. Background: SIRT1, which belongs to the Sirtuin family of NAD-dependent enzymes, plays diverse roles in aging, metabolism, and disease biology. It could regulate cell survival and has been shown to be a protective factor in heart function. Hence, we verified the mechanism by which SIRT1 regulates doxorubicin induced cardiomyocyte injury in vivo and in vitro. Methods: We analyzed SIRT1 expression in doxorubicin-induced neonatal rat cardiomyocyte injury model and adult mouse heart failure model. SIRT1 was over-expressed in cultured neonatal rat cardiomyocyte by adenovirus mediated gene transfer. SIRT1 agonist resveratrol was used to treat the doxorubicin-induced heart failure mouse model. Echocardiography, reactive oxygen species (ROS) production, TUNEL, qRT-PCR, and Western blotting were performed to analyze cell survival, oxidative stress, and inflammatory signal pathways in cardiomyocytes. Results: SIRT1 expression was down-regulated in doxorubicin induced cardiomocyte injury, accompanied by elevated oxidative stress and cell apoptosis. SIRT1 over-expression reduced doxorubicin induced cardiomyocyte apoptosis with the attenuated ROS production. SIRT1 also reduced cell apoptosis by inhibition of p38MAPK phosphorylation and caspase-3 activation. The SIRT1 agonist resveratrol was able to prevent doxorubicin-induced heart function loss. Moreover, the SIRT1 inhibitor niacinamide could reverse SIRT1's protective effect in cultured neonatal rat cardiomyocytes. Abstract: LL-37/hCAP-18, as the only human cathelicidin, plays an important role in inflammation. Lipoteichoic acid (LTA) is an important bacterial component of Staphylococcus aureus, which is one of the common human pathogens for severe respiratory infection with increasing morbidity in recent years. The present study is to investigate the role of LL-37 in LTA induced inflammatory reaction in macrophages. We examined TNF-α and IL-6 production after LL-37 treatment and discussed its signal transduction pathways such as p38MAPK and Akt activation in macrophages. The expression of pro-inflammatory cytokines was analyzed by quantitative real-time RT-PCR and enzyme-linked immunosorbent assay (ELISA). The LL-37 expression was determined by Western blot and immunofluorescence staining. The results showed that LL-37 was upregulated after LTA treatment. It could inhibit LTA induced p38MAPK and Akt phosphorylation and attenuate TNF-α and IL-6 production in macrophages in some specific concentration. These results suggest that LL-37 exerts an anti-inflammatory property and attenuates the pro-inflammatory cytokine release in macrophages. [Copyright &y& Elsevier]

Due to the impressive high‐speed, correlation filters began to receive more attention. In this Letter, a real time target tracking framework based on the kernelised correlation filter is proposed. The framework includes two parts: a boosted fusion feature colour names‐histograms of oriented gradients (CN‐HOG) that consists of CNs and HOG, and a novel fragments‐based tracking algorithm that is robust to partial occlusion. Recently, an increasing number of publications concerned on the catalytic oxidation of refractory organic pollutants in wastewater dependent on the iron-based catalysts. Until now, the iron-based catalysts are diversity including Fe2+/Fe3+, ZVI, iron oxides (FeOOH, Fe 2 O 3 , Fe 3 O 4 , FeO), iron sulfides (Fe x S y) and Fe-bimetallic due to various valences and phases. A comprehensive understanding on the synthesis and applications of them as catalysts were little concerned. In this work, the methods for synthesis and control of the size, morphology, crystal phase and chemical valent properties of the iron-based catalysts were reviewed by classifying as Fe2+/Fe3+, ZVI, iron oxides, pyrite, Fe-MOF and Fe-bimetallic. Their applications in Fenton/Fenton-like oxidation, ozonation, wet air oxidation, PMS/PS activation, photocatalytic oxidation etc., for refractory organic pollutants oxidation were summarized. Generation of free radicals for oxidation of refractory organic pollutants in different systems was regarded as the main catalytic mechanisms. Refining controlled growth of size, morphology, crystal phase and chemical valence of iron-based catalysts will be prospective and challenge in synthesis of iron-based catalysts. Co-catalytic reaction will be promising for accelerating the electron transfer for generation of radicals. Nano zero-valent iron (nZVI) is widely used for decontamination. The main issues associated with nZVI are agglomeration and oxidation in the long term. In this study, the carbothermal reduction of cotton fiber was conducted for the synthesis of nZVI supported on cotton carbon fiber (nZVI/ CF) to address the agglomeration and oxidation of nZVI. Synergistic adsorption and peroxymonosulfate (PMS) activation using nZVI/CF for removing levofloxacin (LEV) are reported herein. The nZVI concentration and morphology were conveniently adjusted by soaking cotton fiber in ferric nitrate solutions of various Fe3+ concentrations. The carbothermal reduction of the cotton fiber at 900 °C contributed to the reduction of Fe3+ into nZVI. A nZVI/CF-900-0.3 system was obtained through the carbothermal reduction of cotton fiber soaked in 0.3 M ferric nitrate. Favorable adsorption of nZVI/CF-900-0.3 to LEV facilitated LEV degradation under PMS activation. Approximately 93.83% of LEV (C 0 = 20 ppm) was removed within 60 min with 0.2 g/L of the catalyst and 1 mM PMS. It was preferable to use nZVI + CF-900 to activate PMS for degrading LEV, thus confirming the favorable effect of LEV adsorption on further degradation. The nZVI/CF-900-0.3 exhibited excellent long-term stability given that it was able to activate PMS after it was stored for 6 months. ·SO 4 − played an important role in LEV degradation in the presence of PMS. A new type of economical covalent organic framework material(COF), namely resin based covalent organic framework material, was prepared by combining resin and covalent organic framework material by hydrothermal synthesis, which was based on the preparation of traditional COF material(TpBD COF). The properties of the material and covalent organic framework material were compared in the way of characterization, and the possible reaction mechanism was analyzed. The solid phase extraction separation (SPE) ability of this material for four kinds of phenolic endocrine disrupting compounds (bisphenol F, bisphenol A, octylphenol and nonylphenol) in beverage samples was investigated. The results showed that the prepared COF materials had abundant internal channels, ordered structure, large specific surface area (TpBD COF: 814.6 m2/g and resin based COF: 623.9 m2/g) and good thermal stability (pyrolysis temperature was 443 °C and 437 °C, respectively). Solid phase extraction experiments demonstrated that the two COF materials as adsorbent of solid phase extraction column had ideal adsorption separation effect and good anti-interference ability, and had strong anti-interference ability. The SPE effect was superior to the traditional solid phase extraction column. The precision RSD of this method was less than 3%. This SPE method had high recovery and could be reused (carbonated beverage: 98.18–102.18% and beverage: 98.52–101.79%), In addition, the recovery of the material did not change significantly in the 50 cycles of solid phase extraction, indicating that the material had good stability and could be reused, which could meet the requirements for the detection and analysis of trace pollutants in environmental samples. The resin based COF material prepared in this study could reduce the cost of monomer uses and provide a possibility for its industrial production. With the wide application of nuclear energy and the rapid development of mining, a great quantity of uranium (U(VI)) containing wastewater is inevitably generated. In this work, a facile environmental-friendly coprecipitation method was proposed for the green synthesis of binary layered double hydroxides (Ni–Co LDH W -7), which could be used to efficiently adsorb U(VI) from wastewater. Pseudo-second-order kinetic model and Dubinin-Radushkevich model fitted well with the adsorption of U(VI) on Ni–Co LDH W -7, the maximum adsorption capacity of U(VI) on Ni–Co LDH W -7 was 201.09 mg/g calculated by the Langmuir model. Adsorption equilibrium reached after 30 min, which is a spontaneous and endothermic process. Fourier Transform Infrared Spectrometer (FTIR) and X-ray Photoelectron Spectroscopy (XPS) measurements manifested that the favorable U(VI) extraction on Ni–Co LDH W -7 is mainly ascribed to inner-layer surface electrostatic interaction and complexation, which is dominated by abundant oxygen-containing M-OH and interlayer OH− ions. • Arylsulfatase A (ARSA) p.L300S variant is absent in either sporadic PD patients or healthy controls. • ARSA p.N352S is not significant association with PD in our Chinese cohorts. • ARSA p.L300S and p.N352S variants may not be common genetic factors for sporadic PD in Chinese Han population. Recently, the arylsulfatase A (ARSA) variant c.899 T > C (p.L300S) was identified to be segregated with Parkinson's disease (PD) in one family of Japanese descent. And the variant c.1055A > G (p.N352S) of ARSA was reported as a risk reduction factor for PD in a Japanese population. To further investigate the relationship between ARSA and PD, we screened these two loci of the ARSA gene in 407 sporadic PD patients and 471 healthy controls from a Chinese Han population. However, we did not detect the ARSA p.L300S variant in either PD patients or healthy controls. Moreover, in case-control association analysis, the p.N325S variant showed no significant association with PD. The ability to learn the reward-value and action-outcome contingencies in dynamic environment is critical for flexible adaptive behavior and development of effective pharmacological control of goal-directed behaviors represents an important challenge for improving the deficits in goal-directed behavior which may underlie seemingly disparate symptoms across psychiatric disorders. Adenosine A 2A receptor (A 2A R) is emerging as a novel neuromodulatory target for controlling goal-directed behavior for its unique neuromodulatory features: the ability to integrate dopamine and glutamate signaling, the "brake" constraint of various cognitive processes and the balanced control of goal-directed and habit actions. However, the contribution and circuit mechanisms of the striatopallidal A 2A Rs in nucleus accumbens (NAc) to control of goal-directed behavior remain to be determined. Here, we employed newly developed opto-A 2A R and the focal A 2A R knockdown strategies to demonstrate the causal role of NAc A 2A R in control of goal-directed behavior. Furthermore, we dissected out multiple distinct behavioral mechanisms underlying which NAc A 2A Rs control goal-directed behavior: (i) NAc A 2A Rs preferentially control goal-directed behavior at the expense of habit formation. (ii) NAc A 2A Rs modify the animals' sensitivity to the value of the reward without affecting the action-outcome contingency. (iii) A 2A R antagonist KW6002 promotes instrumental actions by invigorating motivation. (iv) NAc A 2A Rs facilitate Pavlovian incentive value transferring to instrumental action. Discharge of U(VI) containing wastewater causes severe damage to the environment and leads to a loss of resources. Uranium sorption on hydroxyapatite (HAP) has been studied extensively to address the abovementioned issues. In the present study, BC-HAP was recovered through phosphate sorption from wastewater, which was first reused as a potential sorbent for extracting uranium from aqueous solutions comparing to commercially available nano-HAP. The sorption behavior of uranium and its transformation on the recovered BC-HAP were investigated by conducting batch experiments as well as Fourier-transform infrared, scanning electron microscopy, and x-ray diffraction analyses. BC-HAP had superior sorption ability for uranium extraction. Autunite precipitant at nano-scale is observed after uranium sorption. Partial desorption of uranium was observed in the presence of Na 2 CO 3 and NaHCO 3. Surface complexation and phosphate dissolution precipitation contributed to the favorable uranium sorption. To characterize the colistin-resistant bacterial population in the gut and assess diversity of mcr-1 transmission within a single individual. Large numbers of isolates (>100 colonies/chicken cecum sample) were collected from nine randomly selected mcr-1 -positive chickens in China and used for comprehensive microbiological, molecular and comparative genomics analyses. Of 1273 colonies, 968 were mcr-1 positive (962 Escherichia coli , two Escherichia fergusonii , two Klebsiella pneumoniae and two Klebsiella quasipneumoniae). One to six colistin-resistant species and three to 10 E. coli pulsed-field gel electrophoresis (PFGE) clusters could be identified from each sample. Whole-genome sequencing (WGS) analysis of the representative E. coli strains revealed three to nine sequence types observed in a single chicken host. The mcr-1 genes are located in either chromosomes or plasmids of different types, including IncI2 (n =30), IncHI2 (n =14), IncX4 (n =4), p0111(n =2) and IncHI1(n =1). Strikingly, in single cecum samples, one to five Inc type plasmids harbouring mcr-1 could be identified. Great diversity was also observed for the same IncI2 plasmid within a single chicken host. In addition, up to eight genetic contexts of the mcr-1 gene occurred within a single chicken. There is extensive heterogeneity and flexibility of mcr-1 transmission in chicken gut due to bacterial species differences, distant clonal relatedness of isolates, many types and variations of mcr -positive plasmids, and the flexible genetic context of the mcr - 1 gene. Currently, a lot of uranium containing wastewater was generated in China, which required efficient treatment before being discharged. Immobilization of uranium on ZVI has attracted many concerns from researchers in the world wide. Surface oxidation and agglomeration limited the efficiency of nano zero-valent iron (nZVI) in remediation of uranium pollution. In this work, a green carbothermal reduction process was conducted to prepare environmental friendly biochar-supported ZVI (nZVI/BC). Immobilization behavior of uranium on nZVI/Cs, oxidized nZVI/Cs and Fe 3 O 4 from wastewater were investigated. The iron salt was converted into iron oxides at 700 °C, further being reduced into nZVI in the presence of ferric chloride and ferric nitrate being carbonization of starch at 800 and 900 °C. The uranium adsorption capacities were different from the carbonization temperature and iron sources, which depended on the transformation of iron species. The maximum adsorption capacities of nano-flake Fe Cl /C (1:4–900) and nanoparticles Fe N /C (1:4–900) for U(VI) were 34.82 and 55.14 mg/g. The removal efficiencies of Fe Cl /C (1:4–900) and nanoparticles Fe N /C (1:4–900) were higher than that of 44% for C–Fe 3 O 4. The SEM-mapping and XPS analysis of nZVI/Cs after U(VI) adsorption indicated that the immobilization of U(VI) was ascribed to simultaneous adsorption and reduction. Wastewater discharged from the dye production and consumption process has a high chemical oxygen demand, high chroma, and complex structure. In this study, a boat shaped flaky cobalt-based metal–organic framework (Co-MOF) was synthesized in aqueous solution by using a green one-step precipitation strategy. This strategy exhibited favorable efficiency for the removal of Congo red (CR). Furthermore, ZIF-67 with a rhombic dodecahedral shape was synthesized in anhydrous methanol solvent through a one-step precipitation strategy. The effects of the contact time, adsorbent dosage, initial CR concentration, and pH value on the adsorption of CR were also investigated. Results indicated that the adsorption of CR by Co-MOF and ZIF-67 fitted well with the Langmuir model and pseudo-second-order kinetic model. The maximum adsorption capacities obtained for Co-MOF and ZIF-67 with the Langmuir model were 1019.06 and 1044.58 mg/g at 25 °C, respectively. The obtained equilibrium time was less than 5 min. Moreover, Co-MOF and ZIF-67 had the same removal capacities for CR. The adsorption mechanism was attributed to the strong electrostatic and π–π stacking interactions of CR with Co-MOF and ZIF-67. Idiopathic pulmonary fibrosis (IPF) is a severe pulmonary interstitial pneumonia. Our study focuses on the role of PLA2 enzyme in the IPF to explore a more effective diagnosis and treatment mechanism of IPF. Transcriptome data of IPF from GEO database and bleomycin-induced pulmonary fibrosis mice were analyzed to identify PLA2 enzyme and their metabolite, lysophosphatidylcholines 18:0, in IPF. Based on PLA2G2A and PLA2G2D / PLA2G2A-associated cell death genes (PCDs), the consensus clustering analysis was used to identify the subtypes of IPF and the correlation between PLA2G2A and prognosis was analyzed. The machine learning (ML) models and artificial neural network (ANN) model was used to validate the diagnostic accuracy of PLA2s and PCDs in diagnosing IPF. The gene and protein expression of NLRP3, GSDMD, and CASP-1 was estimated in recombinant PLA2G2A protein induced MLE-12 cells. The expression of PLA2G2D, PLA2G2A, and LPC18 significantly changed in IPF. Furtherly, PLA2G2A has a significant correlation with poor patient prognosis, which could predict the 2 or 3-years mortality rates of IPF. Two subtypes of IPF patients, identified based on PCDs, showed significant different immunoinfiltration. Recombinant PLA2G2A protein could induce the pyrotosis in the MLE-12 cell. The generalized linear model and ANN model of PLA2s or PCDs accurate diagnosis IPF. [Display omitted] • An aqueous two-phase system(ATPs) for separating thorium was established. • ATPs have anti-interference and selectivity for the separation of thorium. • Mechanisms of thorium separation by an aqueous two-phase system are elaborated. • High selectivity separation of thorium was proved in ATPs. As a potential nuclear fuel, the recovery of thorium with high efficiency, high selectivity and high extraction capacity from thorium-containing wastewater is considered to be one of the effective ways to reduce pollution and solve the shortage of thorium resources. However, the problem of efficient and selective separation of thorium from the complex environment has not been completely solved. Herein, a functional ligand called arsenazo (ASA), polyethylene glycol, and sodium sulphate were used to construct an aqueous two-phase system (ATPs) for the selectivity extraction and separation of thorium from mining wastewater. Furthermore, the complex agent, pH value, temperature, equilibrium time, coexisting substances, and other conditions for the extraction and separation of thorium were optimized, indicating that 98.7 % of thorium was efficiently and precisely separated from the acid-leaching solution to the pH of 2–3 or 5–6 with 0.08 g/L of ASA within 10 min, and the interference with multiple coexisting ions and organic substances was avoided. Importantly, the analysis of water quality parameters, thermogravimetry, infrared spectroscopy, and ultraviolet–visible spectroscopy revealed the coordination relationship between thorium ions and complex agents, indicating that the extraction of thorium in the aqueous two-phase system was a cation exchange mechanism. More importantly, SEM, TEM, EDX, and XRD verified that thorium was efficiently and selectively separated by a sulfone acid ligand-aqueous two-phase system, providing quantitative information on thorium-containing product distribution of relatively high accuracy and avoiding the interference with coexisting substance. Therefore, this work provided theoretical guidance on treating and reusing thorium in wastewater and enriched the content of functional ligand-aqueous two-phase extraction systems in green production technology. Myocardial ischemia/reperfusion injury (MIRI) is a prevalent condition associated with numerous critical clinical conditions. miR‐322 has been implicated in MIRI through poorly understood mechanisms. Our preliminary analysis indicated potential interaction of CREB‐binding protein (CBP), a transcriptional coactivator and acetyltransferase, with HIF‐1α/β‐catenin, which might regulate miR‐322 expression. We, therefore, hypothesized that CBP/HIF‐1α/β‐catenin/miR‐322 axis might play a role in MIRI. Rat cardiomyocytes subjected to oxygen‐glucose deprivation /reperfusion (OGD/R) and Langendorff perfused heart model were used to model MIRI in vitro and in vivo, respectively. We used various techniques such as CCK‐8 assay, transferase dUTP nick end labeling staining, western blotting, RT‐qPCR, chromatin immunoprecipitation (ChIP), dual‐luciferase assay, co‐immunoprecipitation (Co‐IP), hematoxylin and eosin staining, and TTC staining to assess cell viability, apoptosis, and the levels of CBP, HIF‐1α, β‐catenin, miR‐322, and acetylation. Our results indicate that OGD/R in cardiomyocytes decreased CBP/HIF‐1α/β‐catenin/miR‐322 expression, increased cell apoptosis and cytokines, and reduced cell viability. However, overexpression of CBP or miR‐322 suppressed OGD/R‐induced cell injury, while knockdown of HIF‐1α/β‐catenin further exacerbated the damage. HIF‐1α/β‐catenin bound to miR‐322 promoter to promote its expression, while CBP acetylated HIF‐1α/β‐catenin for stabilization. Overexpression of CBP attenuated MIRI in rats by acetylating HIF‐1α/β‐catenin to stabilize their expression, resulting in stronger binding of HIF‐1α/β‐catenin with the miR‐322 promoter and subsequent increased miR‐322 levels. Abstract Currently, 18-crown-6-like graphene nanopore has emerged and received much theoretical and experimental attention. Inspired by the multilayer oxygen-containing structure of the K+ channel (KcsA), we adopted molecular dynamics to investigate the selectivity of K+/Na+ for a series of multilayer 18-crown-6-like graphene nanopores. The results demonstrated that the nanopores of the multilayer graphene could facilitate the selectivity of K+/Na+ compared to the monolayer graphene. A proper combination of layer number and interlayer spacing could achieve a high selectivity. The spatial distribution of fluid molecules and ionic hydration microstructures indicated that the easier dehydration of K+ could lead to the more uniformity of K+ pathways in the central region of the nanopore and in turn be helpful in increasing the selectivity of K+. Meanwhile, the increased unsuitability for the pore wall oxygen atoms to compensate the partial dehydrated Na+ could enhance the resistance of Na+. These findings provide useful insights to the further design of 18-crown-6-like and other crown-ether-like graphene nanopore-based nanodevices as sensors and ion separators. Transgenic Bacillus thuringiensis (Bt) rice extends significant protection against insect pests and meets the increasing demands for food and energy. Many studies have been conducted investigating the impacts of Bt rice to the agricultural ecosystem, but much less attention has been given to efforts attempting to determine how the presence of Bt rice influences and shapes the microbial community, especially the active microbes. Stable isotope probing and high-throughput sequencing were employed to explore the active microbes involved in Bt-containing straw decomposition. Compared to its near isoline, the Bt straw contained higher contents of total N, total P, total K, lignin, cellulose, and Cry1Ab toxin protein. These chemical differences did not affect the decomposition rate but significantly changed the active microbial decomposer communities. During the decomposition of Bt-containing straw, fungi were more affected than bacteria. Agromyces, Terrabacter, Microbacterium, Glycomyces, and Kribbella were the most representative unique (existed only in the Bt treatments and appeared at the early stage) bacterial genera, and Trichoderma was the most representative unique fungal genus in the Bt straw decomposition. By using similarity index calculation and function prediction, the significant differences between Bt straw and non-Bt straw treatments were found to be transient for both microbial taxa and functional traits. These results suggested that Bt rice has a significant but transient impact on soil microbes in terms of microbial straw decomposition. [ABSTRACT FROM AUTHOR]

Hericium erinaceus, a mushroom species commonly known as Yamabushitake in Japan, is known to have a stimulatory effect on neurotrophic factors, such as brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF). Hericenone C, a meroterpenoid with palmitic acid as the fatty acid side chain, is reported to be one such stimulant. However, according to the structure of the compound, the fatty acid side chain seems highly susceptible to lipase decomposition, under in vivo metabolic conditions. To study this phenomenon, hericenone C from the ethanol extract of the fruiting body was subjected to lipase enzyme treatment and observed for changes in the chemical structure. The compound formed after the lipase enzyme digestion was isolated and identified using LC-QTOF-MS combined with 1H-NMR analysis. It was found to be a derivative of hericenone C without its fatty acid side chain and was named deacylhericenone. Interestingly, a comparative investigation of the neuroprotective properties of hericenone C and deacylhericenone showed that the BDNF mRNA expression in human astrocytoma cells (1321N1) and the protection against H2O2-induced oxidative stress was considerably higher in the case of deacylhericenone. Microglial hyperactivation of the NOD‐, LRR‐, and pyrin domain‐containing 3 (NLRP3) inflammasome contributes to the pathogenesis of Parkinson's disease (PD). Recently, neuronally expressed NLRP3 was demonstrated to be a Parkin polyubiquitination substrate and a driver of neurodegeneration in PD. However, the role of Parkin in NLRP3 inflammasome activation in microglia remains unclear. Thus, we aimed to investigate whether Parkin regulates NLRP3 in microglia. We investigated the role of Parkin in NLRP3 inflammasome activation through the overexpression of Parkin in BV2 microglial cells and knockout of Parkin in primary microglia after lipopolysaccharide (LPS) treatment. Immunoprecipitation experiments were conducted to quantify the ubiquitination levels of NLRP3 under various conditions and to assess the interaction between Parkin and NLRP3. In vivo experiments were conducted by administering intraperitoneal injections of LPS in wild‐type and Parkin knockout mice. The Rotarod test, pole test, and open field test were performed to evaluate motor functions. Immunofluorescence was performed for pathological detection of key proteins. Overexpression of Parkin mediated NLRP3 degradation via K48‐linked polyubiquitination in microglia. The loss of Parkin activity in LPS‐induced mice resulted in excessive microglial NLRP3 inflammasome assembly, facilitating motor impairment, and dopaminergic neuron loss in the substantia nigra. Accelerating Parkin‐induced NLRP3 degradation by administration of a heat shock protein (HSP90) inhibitor reduced the inflammatory response. Parkin regulates microglial NLRP3 inflammasome activation through polyubiquitination and alleviates neurodegeneration in PD. DNA damage and defective DNA repair are extensively linked to neurodegeneration in Parkinson's disease (PD), but the underlying molecular mechanisms remain poorly understood. Here, we determined that the PD-associated protein DJ-1 plays an essential role in modulating DNA double-strand break (DSB) repair. Specifically, DJ-1 is a DNA damage response (DDR) protein that can be recruited to DNA damage sites, where it promotes DSB repair through both homologous recombination and nonhomologous end joining. Mechanistically, DJ-1 interacts directly with PARP1, a nuclear enzyme essential for genomic stability, and stimulates its enzymatic activity during DNA repair. Importantly, cells from PD patients with the DJ-1 mutation also have defective PARP1 activity and impaired repair of DSBs. In summary, our findings uncover a novel function of nuclear DJ-1 in DNA repair and genome stability maintenance, and suggest that defective DNA repair may contribute to the pathogenesis of PD linked to DJ-1 mutations. Myocardial ischemia/reperfusion injury (MIRI) is a major cause of heart failure after myocardial infarction. It has been reported that miR‐322 is involved in MIRI progression, while the molecular mechanism of miR‐322 in regulating MIRI progression needs to be further probed. MIRI cell model was established by oxygen‐glucose deprivation/reoxygenation (OGD/R). Cell viability was assessed using MTS assay. Flow cytometry and terminal deoxynucleotidyl transferase‐mediated dUTP nick‐end labeling staining were employed to analyze cell apoptosis. In addition, the interactions between miR‐322, Smad7/Smurf2, hypoxia‐inducible factor alpha (HIF‐1α), and β‐catenin were verified by dual‐luciferase reporter gene assay. Our results displayed that miR‐322 was significantly downregulated in OGD/R‐treated H9c2 cells, and its overexpression resulted in increased cell viability and reduced the apoptosis. Smurf2 and Smad7 were identified as the direct targets of miR‐322. Smad7 knockdown or Smurf2 knockdown increased OGD/R‐treated H9c2 cell viability and suppressed the apoptosis. Meanwhile, miR‐322 mimics abolished the mitigating effect of Smad7 or Smurf2 overexpression on MIRI. In addition, the Smad3/β‐catenin pathway was identified as the downstream pathway of Smurf2/Smad7. Moreover, it was found that HIF‐1α interacted with the miR‐322 promoter, and β‐catenin interacted with the HIF‐1α promoter to form a loop. HIF‐1α‐induced upregulated miR‐322 activated the Smad3/β‐catenin pathway by targeting Smurf2 and Smad7 to improve MIRI; meanwhile, β‐catenin/HIF‐1α formed a positive feedback loop to continuously improve MIRI. Background: Inflammasome activation has a pathogenic role in Parkinson's disease (PD). Up-regulated expressions of inflammasome adaptor apoptosis-associated speck-like protein containing a CARD (ASC) and assembly of ASC specks have been observed in postmortems of human PD brains and experimental PD models. Extracellular ASC specks behave like danger signals and sustain prolonged inflammasome activation. However, the contribution of ASC specks in propagation of inflammasome activation and pathological progression in PD has not been fully established. Methods: Herein, we used human A53T mutant α-synuclein preformed fibrils (PFFs)-stimulated microglia in vitro and unilateral striatal stereotaxic injection of PFFs-induced mice model of PD in vivo, to investigate the significance of ASC specks in PD pathological progression. Rotarod and open-field tests were performed to measure motor behaviors of indicated mice. Changes in the molecular expression were evaluated by immunofluorescence and immunoblotting (IB). Intracellular knockdown of the ASC in BV2 cells was performed using si-RNA. Microglial and neuronal cells were co-cultured in a trans-well system to determine the effects of ASC knockdown on cytoprotection. Results: We observed a direct relationship between levels of ASC protein and misfolded α‑synuclein aggregates in PD mice brains. ASC specks amplified NLRP3 inflammasome activation driven by α-synuclein PFFs stimulation, which aggravated reactive microgliosis and accelerated α‑synuclein pathology, dopaminergic neurodegeneration and motor deficits. Endogenous ASC knockdown suppressed microglial inflammasome activation and neuronal α‑synuclein aggregation. Conclusions: In conclusion, our study elucidated that ASC specks contribute to the propagation of inflammasome activation-associated α‑synuclein pathology in PD, which forms the basis for targeting ASC as a potential therapy for PD. Rhizosphere microbiomes contribute considerably to host‐plant fitness; however, limited information is available on their responses to abiotic stresses. Stevia (Stevia rebaudiana Bertoni) is a crop sensitive to both nitrogen and water limitations; thus, stevia was employed to explore plant and rhizobacteria responses to nitrogen and/or water stresses. Water stress specifically enhanced root biomass, whereas nitrogen deficiency caused a general plant growth inhibition and foliar steviol glycosides content surge. In contrast, the quantitative abundances of overall rhizobacteria were more sensitive to water than nitrogen deficiency. Furthermore, 128 responsive bacterial taxa within four discrete ecological categories were screened, mainly members of Proteobacteria , Bacteroidetes, and Chloroflexi phyla. Bacterial taxa motivated by nitrogen/water stress prevailingly displayed a random phylogenetic distribution, while those suppressed taxa exhibited a phylogenetically conserved ecological strategy. Moreover, the quantitative abundance of some water stress-motivated genera, such as Labilithrix and Myxobacterium, were actively correlated with stimulated root growth, while others, such as Steroidobacter and Chthonomonas , which were co-motivated by nitrogen and water stress, were positively linked to steviol glycoside synthesis. This study unveiled the adaptive assembly of rhizobacterial communities to nitrogen and/or water stresses and identified the potential core microorganisms regulating steviol glycoside synthesis and stevia tolerance. Background: The clinical utility of the quantitative flow ratio (QFR), a novel angiography-based index for the functional assessment of coronary stenoses, has recently been demonstrated in patients undergoing percutaneous coronary intervention (PCI).Objectives: This study aimed to ascertain whether the beneficial outcomes of QFR guidance for lesion selection during PCI is affected by diabetes status.Methods: This substudy from the FAVOR III China trial, in which diabetes was one of the prespecified factors for stratified randomization, compared clinical outcomes of QFR-guided vs angiography-guided PCI lesion selection according to the presence of diabetes. The primary endpoint was the 1-year risk of major adverse cardiac events (MACE) (a composite of all-cause death, myocardial infarction, or ischemia-driven revascularization).Results: Among 3,825 patients enrolled, 1,295 (33.9%) had diabetes, 347 (26.8%) of whom were treated with insulin. Baseline characteristics were well balanced between treatment arms in both diabetic and nondiabetic patients. Compared with standard angiography-based lesion selection, the QFR-guided strategy consistently reduced the risk of 1-year MACE in both diabetic patients (6.2% vs 9.6%; HR: 0.64; 95% CI: 0.43-0.95) and nondiabetic patients (5.6% vs 8.3%; HR: 0.66; 95% CI: 0.49-0.89) (Pinteraction = 0.88).
