Abstract

Aquaculture Europe, Oceans opportunity, 4-7 October 2021, Madeira, Portugal.-- 2 pages, 1 figure, European sea bass (Dicentrarchus labrax) is an important European aquaculture species. Females, performing better than males with later maturation and larger size, are preferred. Unfortunately, in aquaculture conditions, the sex-ratio is typically biased towards males. Thus, deciphering the sex determinism system and finding routes for controlling the sexratio of this species has been a long-lasting challenge to support sea bass production and selective breeding programmes. A polygenic threshold sex determination system was demonstrated (Vandeputte et al. 2007), where the genetic sex tendency is influenced by larval rearing temperature to determine the phenotypic sex (Piferrer et al, 2005). In this study, we applied two thermal treatments during early larval stage, a low temperature protocol (16°C, LT), known to favour more balanced sex-ratios, and a high temperature masculinizing protocol (21°C, HT). Abstract

Inspired by agroecology, ecological aquaculture proposes an alternative model that uses ecology as a paradigm to develop innovative, more eco-friendly aquaculture with environmental, economic and social benefits. Integrated multi-trophic aquaculture (IMTA) is one application of this principle. Inspired by the natural trophic chain, it associates primary producers with primary or secondary consumers, providing a new source of biomass without requiring supplementary feed by recycling inorganic and organic wastes. Of these systems, land-based IMTA demonstrate several advantages, especially easier control of nutrient flows, contaminants and/or predators. This study focused on a land-based marine IMTA, combining a recirculating aquaculture system for fish consecutively with a natural marine polyculture of microalgae and oyster cultivation. The objective was to assess the ability of the microalgal polyculture both to bioremediate fish nutrients and to sustain oyster growth. For the first time in a Mediterranean climate, we confirmed the feasibility of developing a microalgae community of interest for oysters maintained by fish effluent. Despite strong variability in microalgae production, this IMTA system resulted in significant oyster growth over the experimental period of 1 month, with growth results of the same order of magnitude as natural juvenile growth. In the conditions tested, this IMTA with reduced human intervention allowed a gain in recoverable biomass of 3.7 g of oyster produced per killogramme of fish feed distributed. By transforming waste into additional biomass, IMTA offer a more promising, ecological avenue for aquaculture, based on a circular economy, which may in turn increase the social acceptability of fish farming. WOS:000475657500006; International audience; In teleosts, the regulation of hydromineral balance has a direct impact on several physiological functions, biochemical processes, and can influence behaviour, distribution and survival. As European sea bass Dicentrarchus labrax undertake seasonal migrations from seawater (SW) to brackish, estuarine and fresh water (FW) in their habitat, this study investigates their capacity to tolerate fresh water and explores intraspecific variations in physiological responses. Juvenile D. labrax were transferred from SW to FW at various ages. Freshwater-tolerant and non-tolerant phenotypes were discriminated according to behavioural and morphological characteristics. About 30% of the fish exposed to FW were identified as freshwater intolerant following FW challenges performed at different ages. Interestingly, intolerant fish exhibited the same phenotypic traits: erratic swimming, lower speed, isolation from the shoal and darker colour. Freshwater-intolerant fish were also characterised by a significant lower blood osmolality compared to tolerant fish, and significantly lower Na+/K+-ATPase α1a expression in the posterior kidney. An imbalance in ion regulatory mechanisms was further confirmed by a blood Na+/Cl− ratio imbalance observed in some freshwater-intolerant fish. The analysis of glucocorticoid and mineralocorticoid receptor expression levels in gills and kidney revealed significant differences between freshwater-intolerant and -tolerant fish in both organs, suggesting differential stress-related responses. This study clearly shows an intraspecific difference in the responses following FW transfer with a decreased renal ion uptake capacity as a major cause for freshwater intolerance.

Abstract

International audience; European sea bass has a polygenic sex determinism with temperature influences, which remains hard to predict and manipulate. To investigate the interactions between early growth and sex determination, we studied West Mediterranean domesticated sea bass from a 50♂x10♀ full factorial cross, reared as a single batch at 15°C to promote female sex determination. Temperature was increased to 25°C from 70 to 119 days post-hatching (dph) to increase growth. Fish were individually tagged at 95 dph (575 mg) with microtags and were individually weighed at 95, 115, 136, 157, 200, 256 and 325 dph, and then sexed. Parentage was recovered with 12 microsatellites; 1134 fish had appropriate pedigree, sex and growth data. Multivariate sire models were run to investigate the links between growth and sex (considered a threshold trait with an underlying sex tendency). Heritability of sex tendency was 0.39±0.12 on the underlying scale. The sex dimorphism of body weight in favour of females rose from +27% at 95 dph (BW95) to +40-45% between 115 and 325 dph, and the sex dimorphism of DGC was maximal for DGC95-115. If differential growth is seen as a consequence of phenotypic sex, growth should be corrected by a fixed sex effect. In this case, there was a positive, although not significant (rA=0.18±0.12) genetic correlation of sex tendency with DGC95-115, while it never exceeded 0.05 later than 136 dph. The genetic correlation of BW with sex tendency regularly decreased from 0.45±0.18 for BW95 to -0.20±0.17 for BW325. If differential growth is seen as a cause of phenotypic sex, no fixed sex effect should be modelled, which then led to a rA of sex tendency with DGC decreasing from 0.60±0.11 for DGC95-115 to 0.21±0.15 for DGC256-325, while the rA of BW with sex tendency also decreased from 0.72±0.10 for BW115 to 0.48±0.14 for BW325. In all cases, the genetic and environmental correlation of growth and sex tendency was maximal at the earlier stages, especially before the first reported signs of sex differentiation. This leads us to think that post-larval growth is, at least partially, a cause and not a consequence of sex determination in sea bass.
