Abstract The efficacy and safety of venetoclax in newly diagnosed pediatric acute myeloid leukemia (AML) are not well-established as they are in adults. Children newly diagnosed with AML were recommended for induction therapy with venetoclax and chemotherapy or hypomethylating agents (HMAs) as per for the ChiCTR1900027146 trial. Venetoclax was administered at a consistent dose of 200 mg/m2/day for 28 days, with adjustments when used concurrently with azoles. The study measured both the remission rates and the safety assessments of venetoclax. We enrolled 45 newly diagnosed pediatric patients with AML. The complete remission rates were 94.7% in the low/middle-risk group and 80.8% in the high-risk group; MRD-negative rates were 52.6% and 38.5% in the low/middle-risk group and high-risk group, respectively. Venetoclax based combination therapy was well tolerated by the majority of patients. The median duration of venetoclax dosing was 18 days (range 9–28), with hematological toxicity and infection being the most common adverse events. Venetoclax-based induction regimens demonstrated a high response rate and safety profile in newly diagnosed pediatric AML cases. This underscores the significance of venetoclax as a viable treatment option for untreated AML, extending beyond its role as salvage therapy for refractory/relapsed AML.

Acute myeloid leukemia (AML), which has distinct genetic abnormalities, has unique clinical and biological features. In this study, the incidence, clinical characteristics, induction treatment response, and outcomes of a large cohort of Chinese AML pediatric patients treated according to the BCH-AML 2005 protocol were analyzed. RUNX1-RUNX1T1 was the most common fusion transcript, followed by the CBFβ-MHY11 and KMT2A rearrangements. FLT3-ITD and KIT mutations are associated with unfavorable clinical features and induction responses, along with KMT2A rearrangements, DEK-NUP214, and CBF-AML. The 5-year event-free survival (EFS) and overall survival (OS) rates of our cohort were 53.9 ± 3.7% and 58.5 ± 3.6%, with the best survival found among patients with CBFβ-MYH11 and the worst survival among those with DEK-NUP214. In addition, we found that patients with FLT3-ITD mutation had adverse outcomes and that KIT mutation had a negative impact on OS in RUNX1-RUNX1T1+ patients. Furthermore, the risk classification and response to treatment after each induction block also influenced the prognosis, and HSCT after first remission could improve OS in high-risk patients. Not achieving complete remission after induction 2 was found to be an independent prognostic factor for OS and EFS. These findings indicate that genetic abnormalities could be considered stratification factors, predict patient outcomes, and imply the application of targeted therapy.

Background Sedation techniques can ease anxiety during medical procedures for children. Our previous report on Chinese sedation practices for non-invasive procedures in 2018 is outdated due to the rapid development of sedation services. This study provides an updated report on sedation practices for non-invasive procedures in China.Methods This is a cross-sectional study. Questionnaires were sent to tertiary maternity and children’s hospitals nationwide through the WeChat Mini Program. The survey questioned the location and caseloads of hospitals providing sedation services, support facilities, contraindications, fasting practices, sedation regimens, monitoring practices, staff structure, certification requirements for sedation providers and quality control data.Results Procedural sedation for non-invasive procedures were provided in 88 of 114 hospitals. These hospitals are located across the country except Heilongjiang province and the Tibet Autonomous Region. Compared with previous reports, significant increases were found in the number of hospitals providing sedation services, dedicated sedation rooms and recovery rooms and full-time sedation providers. Most hospitals advocated the 2-4-6 rule for pre-sedation fasting. Dexmedetomidine was the most used first-choice sedative. Anaesthesiologists remain the primary sedation providers, but nurses are also important. The most mentioned qualification requirements for sedation providers were a professional title of attending doctor, ≥5 years of working experience in paediatric anaesthesia and paediatric advanced life support certification. Sedation service records were used in 83 hospitals, but only 42 and 39 recorded success rates and adverse events, respectively.Conclusions Sedation services for non-invasive procedures are available in most areas of China. More hospitals now provide sedation services and full-time sedation providers. Supporting facilities and sedation regimens have improved. Non-anaesthesiologist sedation providers are important at current stage, developing training programmes for them may be necessary. Attention should be focused on quality control and improvement of sedation services.

Abstract Background This study reported height prediction and longitudinal growth changes in Chinese pediatric patients with acute myeloid leukemia (AML) during and after treatment and their associations with outcomes. Methods Changes in 88 children with AML in percentages according to the growth percentile curve for Chinese boys/girls aged 2–18/0–2 years for body mass index (BMI), height, and weight from the time of diagnosis to 2 years off therapy were evaluated. The outcomes of AML were compared among patients with different BMI levels. Results The proportion of underweight children (weight 75th percentile) after 2 years of drug withdrawal. Unhealthy BMI at the initial diagnosis and during intensive chemotherapy leads to poorer outcomes. For height, all patients were in the range of genetic height predicted based on their parents’ height at final follow-up. Conclusions Physicians should pay more attention to the changes in height and weight of children with AML at these crucial treatment stages and intervene in time.

Drought stress during sorghum emergence significantly affects seedling establishment, adversely affecting both emergence and population growth. This study investigates drought tolerance mechanisms during sorghum germination by analyzing physiological changes and transcriptomic data from two lines: W069 (drought tolerant) and W040 (drought sensitive). Under drought conditions, a phenotypic analysis revealed that W069 exhibited longer shoots and roots than W040. Additionally, physiological data indicated higher osmotic substance and lower malondialdehyde levels in W069. Using Kyoto Encyclopedia of Genes and Genome analyses, we identified three key pathways (starch and sucrose metabolism, phenylpropanoid biosynthesis, and phytohormone signaling) as pivotal in the drought response during seed germination in sorghum plants. Expression profiling revealed that most drought tolerance-related genes in the three key pathways were expressed at higher levels in the drought-tolerant cultivar W069, possibly explaining its greater stress tolerance. These findings enhance our understanding of drought-responsive gene networks in sorghum seed germination, offering potential target genes and strategies for enhancing drought tolerance in this crop.

TTMV::RARA is a recently reported fusion gene associated with acute promyelocytic leukemia (APL), caused by the integration of torque teno mini virus (TTMV) genomic fragments into the second intron of the RARA gene. Currently, there have been only six documented cases, with clinical presentations showing significant variability. Although initial responses to all-trans retinoic acid (ATRA) treatment may be observed in patients with TTMV::RARA-APL, the overall prognosis remains unfavorable among infrequent reported cases. This article presents a pediatric case that manifested as PML::RARA-negative APL with central nervous system involvement at onset. The patient experienced both intramedullary and extramedullary relapse one year after undergoing allogeneic hematopoietic stem cell transplantation. Upon identification as TTMV::RARA-APL and subsequent administration of two rounds of ATRA-based treatment, the patient rapidly developed multiple RARA ligand-binding domain mutations and demonstrated extensive resistance to ATRA and various other therapeutic interventions. Additionally, the patient experienced ARID1A mutant clone expansion and progressed MYC-targeted gene activation. This case represents the first documentation of extramedullary involvement at both the initial diagnosis and relapse stages, emphasizing the intricate clinical features and challenges associated with the rapid accumulation of multiple ATRA-resistant mutations in TTMV::RARA-APL, characterizing it as a distinct and complex sub-entity of atypical APL.

Abstract Various plants, including sorghum (Sorghum bicolor L.), are exposed to waterlogging; however, little is known about the effects of waterlogging at different growth stages on sorghum. A pot experiment was conducted using two sorghum hybrids, Jinuoliang 01 (JN01) and Jinza 31 (JZ31), to investigate the effects of waterlogging at different growth stages on the photosynthesis enzyme activity, chlorophyll content, malondialdehyde (MDA) content, photosynthetic parameters, dry matter accumulation, and grain yield. The experiment was conducted using waterlogging treatments implemented at the five-leaf stage (T1), flowering stage (T2), and filling stage (T3), using standard management (no waterlogging) as a control (CK). The adverse effects of waterlogging on sorghum growth varied with the waterlogging timing, with the maximum impact at T1, followed by T2 and T3. JZ31 was more sensitive to waterlogging compared to JN01. Waterlogged conditions inhibited the photosynthetic enzyme activity and reduced the chlorophyll content and photosynthesis, ultimately lowering the biomass yield and grain yield. The maximum yield loss was observed with the T1 waterlogging treatment; the grain yield of JN01 and JZ31 decreased by 52.01–54.58% and 69.52–71.97%, respectively, compared with CK. Furthermore, the decline in grain yield in T1 was associated with reducing grain number per panicle. These findings indicate that sorghum is sensitive to waterlogging at the five-leaf stage and JZ31 is more sensitive to waterlogging than JN01, which may provide a basis for selecting genotypes and management measures to cope with waterlogging in sorghum.

Abstract Background Little is known about DNMT3A expression and its prognostic significance in childhood B cell acute lymphoblastic leukemia (B-ALL). Methods We determined DNMT3A mRNA expression in 102 children with B-ALL. Correlations with relapse-free survival (RFS) and common clinical characteristics were analyzed. DNMT3A was stably knocked out by CRISPR/Cas9 gene editing technology in Reh and 697 B-ALL cell lines. Cell proliferation activity after treated with daunorubicin (DNR) was determined by CCK8 assay in DNMT3A KO Reh and 697 cell lines. Results DNMT3A expression in B-ALL patients who were in continuous complete remission (CCR) was higher than in those who got relapse (P = 0.0111). Abstract Background Dasatinib and imatinib are the recommended tyrosine kinase inhibitors (TKIs) for treating pediatric Philadelphia-positive acute lymphoblastic leukemia (Ph + ALL), and the one which has been approved indication in China is imatinib. Recently, clinical demand for Ph + ALL treatment is becoming unmet gradually with the increasing resistance of imatinib. There are some studies reporting the better efficacy and comparative safety of dasatinib compared with imatinib, but no economic comparison has been published. This study aims to supplement economic evidence by comparing the cost-effectiveness between imatinib and dasatinib in treating pediatric patients with Ph+ ALL in China, and to help clinical rational drug use via multi-dimensional value assessment. Methods A decision tree model combined with a 10-year Markov model were established based on the disease progression. The parameters were collected from published literatures and our hospital’s electronic medical records. From the health system perspective, the incremental cost-effectiveness ratio (ICER) between the two treatment groups was calculated through cost-effectiveness analysis and then compared with the willingness-to-pay (WTP) threshold. The set WTP threshold in this study was 1 times per capita gross domestic product (GDP) of China, as recommended by the World Health Organization. Direct medical costs and quality-adjusted life years (QALYs) were calculated and discounted at 5%. The sensitivity analyses were conducted to assess the uncertainty and robustness of the results. Results The total costs were CNY 1,020,995.35 and CNY 1,035,788.50 in imatinib group and dasatinib group during the 10-year simulation, and the total QALYs were 2.59 and 4.84. Compared with the imatinib treatment group, the ICER was around CNY 6,575.78/ QALY, which was less than the set threshold CNY 70,892/ QALY. The sensitive analyses indicated the robustness of the results. Conclusions The cost-effectiveness analysis shows the potential cost-effective advantages of adding dasatinib comparing with adding imatinib for pediatric Ph + ALL patients in China under the set WTP threshold, which indicates that those patients could achieve more QALYs by paying acceptable fee.

Allogeneic hematopoietic stem cell transplantation (HSCT) is an effective treatment for acute myeloid leukemia (AML). Pediatric patients with AML who relapse after HSCT have an extremely poor prognosis. We performed a retrospective study of pediatric patients diagnosed with AML from August 2015 to October 2019 who were treated with HSCT. Kaplan–Meier analyses were used to evaluate overall survival (OS), event-free survival (EFS), and cumulative recurrence rate (CRR). Cox regression analysis was used to determine the association between the baseline characteristics and relapse. A total of 37 pediatric patients met the inclusion criteria. Twenty-eight (75.7%) patients survived, and 9 (24.3%) patients died. The OS rates of AML patients treated with HSCT were 89.2% ± 5.1%, 75.7% ± 7.1%, and 75.7% ± 7.1% at 1, 3, and 5 years, respectively, and the CRRs were 11.4% ± 5.4%, 24.7% ± 7.7%, and 33.1% ± 10.4% at 1, 3, and 5 years after HSCT, respectively; four of nine children who relapsed after transplantation died. Induction with etoposide rather than homoharringtonine and fungal infections could be high-risk factors for recurrence after transplantation. The association between homoharringtonine-based induction therapy and a low recurrence rate persisted after adjusting for age, sex, risk stratification, fusion genes, and fungal infections. This study clarifies the clinical features and poor prognosis of post-transplant relapse in pediatric AML and indicates the urgent need for effective therapy for patients who relapse after HSCT.

Simulation localization and mapping(SLAM) is a popular research problem in the field of driverless cars, but there are still some difficult problems to solve. The conventional SLAM algorithm does not take into account the dynamic objects in the environment, resulting in problems such as low bit pose estimation. In this study, we provide a deep learning-based SLAM scheme. In order to solve the problem of inaccurate feature point extraction in a dynamic environment, this paper adopts image pyramid to distribute feature points uniformly and extracts feature points by using the adaptive thresholding method. To address the problem of incomplete dynamic object mask segmentation, an improved Mask R-CNN network was proposed to improve the integrity of the mask edges by adding an edge detection end to the Mask R-CNN network. To address the problem of incomplete dynamic feature point rejection, this study uses a motion consistency detection algorithm to detect dynamic feature points, and uses the remaining static features for bit pose estimation. The experimental results on the TUM R-GBD dataset show that the absolute trajectory error of the SLAM algorithm in this study is reduced by 93.2% on average compared to the ORB-SLAM2 algorithm. Compared to the Dyna-SLAM algorithm, the absolute trajectory error of the algorithm in this study was reduced by 36.6% on average.

ObjectivesBlinatumomab was shown to be safe and effective for consolidation therapy in B-cell acute lymphoblastic leukemia (B-ALL). This study aimed to investigate the effectiveness and safety of blinatumomab in pediatric B-ALL patients in a real-world setting.MethodsThis was a retrospective, observational study that included patients who initiated blinatumomab treatment between October 1, 2020 and June 20, 2022. Patients with B-ALL diagnosis, age below 18 years, and at least one blinatumomab treatment cycle were included. Treatment-related toxicities were assessed.ResultTotally 23 pediatric patients were included in this study, with a median age of 6 years (range, 2 to 11 years). Blinatumomab therapy was applied for MRD-positive (disease ≥0.01%, n = 3) or chemotherapy-ineligible (n = 20) B-ALL cases. The median follow-up time was 9 months, and all evaluable patients achieved complete molecular remission with undetectable MRD. Four relapsed B-ALL cases proceeded to hematopoietic stem cell transplantation (HSCT) without further bridging therapy, while the others underwent maintenance chemotherapy after blinatumomab treatment. Grade ≥3 febrile neutropenia, white blood cell decrease and seizure were observed in 57%, 48% and 4.3% of patients, respectively. One case discontinued therapy due to neurologic toxicities. The complete mitochondrial genome of the lizard, Teratoscincus przewalskii, which belongs to the family Sphaerodactylidae was determined based on Illumina data in this study. The result showed that the closed double-stranded circular mitogenome was 16,779 bp in total length (GenBank accession number: MW491837) with 44.07% GC. The complete mitochondrial genome consisted of 13 protein-coding genes, 22 transfer RNA genes, two ribosomal genes, and one noncoding control region. Phylogenetic analysis using mitochondrial genomes suggested that T. przewalskii was most closely related to its congener T. keyserlingii. This work provides valuable molecular information for further research on species identification and molecular evolution.

Abstract Introduction There is a known association between primary mediastinal germ cell tumor (PMGCT) and hematologic malignancy that is not linked to treatment. They are exceptionally rare entities with a low morbidity and a poor prognosis. Case presentation An 11‐year‐old boy presented with an anterior mediastinal mass diagnosed as a malignant germ cell tumor on the basis of an excisional biopsy. He was found to have acute myeloid leukemia (AML) two years after the chemotherapy for his germ cell tumor. The clinical course was very aggressive with a survival time of only 1 week after diagnosis of AML associated with PMGCT. Conclusion AML associated with PMGCT needs to be diagnosed correctly. Relevant examinations should be carried out in patients with PMGCTs during and after chemotherapy, and long‐term follow‐up is still necessary to reduce the risk of morbidity and mortality.

Seed germination is the sensitive period to salt stress. Calcium chloride (CaCl2) has been proved as an effective priming agent which can promote the sorghum germination under salt stress. However, there are few reports on CaCl2 priming to improve the salt tolerance during seed germination. The present study investigated the effects of CaCl2 priming on sorghum germination, antioxidant metabolism, osmotic regulation and ion balance under salt stress (150 mM NaCl). The results revealed that the salt stress inhibited the elongation of mesocotyl and root and reduced the germination rate of sorghum. While CaCl2 priming significantly promoted the elongation of mesocotyl and root, and increased the germination rate of sorghum under salt stress. CaCl2 priming notably increased the content of osmotic substances in mesocotyl and root of sorghum under salt stress, and increased the relative water content in these tissues. CaCl2 priming decreased Na+ content and increased K+, Ca2+ contents and the K+/ Na+ in mesocotyl and root, such effects might be induced by up-regulating the expression of NHX2, NHX4, SOS1, AKT1, AKT2, HKT1, HAK1, and KUP. CaCl2 priming reduced the antioxidant enzymes activities and related gene expression compared with untreated sorghum seeds under salt stress. In short, CaCl2 priming improved sorghum germination by enhancing osmotic regulation and ion balance instead of antioxidant enzyme activity. However, the molecular mechanisms of Ca2+ signaling induced by CaCl2 priming in association with the enhanced germination in primed sorghum seeds under salt stress need to be addressed in future studies.

When using deep learning networks for dynamic feature rejection in SLAM systems, problems such as a priori static object motion leading to disturbed build quality and accuracy and slow system runtime are prone to occur. In this paper, based on the ORB-SLAM2 system, we propose a method based on improved YOLOv5 networks combined with geometric constraint methods for SLAM map building in dynamic environments. First, this paper uses ShuffleNetV2 to lighten the YOLOv5 network, which increases the improved network’s operation speed without reducing the accuracy. At the same time, a pyramidal scene parsing network segmentation head is added to the head part of the YOLOv5 network to achieve semantic extraction in the environment, so that the improved YOLOv5 network has both target detection and semantic segmentation functions. In order to eliminate the objects with low dynamic features in the environment, this paper adopts the method of geometric constraints to extract and eliminate the dynamic features of the low dynamic objects. By combining the improved YOLOv5 network with the geometric constraint method, the robustness of the system is improved and the interference of dynamic targets in the construction of the SLAM system map is eliminated. The test results on the TUM dataset show that, when constructing a map in a dynamic environment, compared with the traditional ORB-SLAM2 algorithm, the accuracy of map construction in a dynamic environment is significantly improved. The absolute trajectory error is reduced by 97.7% compared with ORB-SLAM2, and the relative position error is reduced by 59.7% compared with ORB-SLAM2. Compared with DynaSLAM for dynamic scenes of the same type, the accuracy of map construction is slightly improved, but the maximum increase in keyframe processing time is 94.7%.

The wetland ecosystem is an important organic carbon pool on earth, and rich carbon storage and high carbon density are present in wetland vegetation and soil. However, large areas of wetlands have been drained, the area of wetlands has shrunk, and the degree of fragmentation has intensified due to rapid urbanization in the past 20 years. Enhancing ecological carbon sequestration capacity is a key task of carbon peaking and neutrality, and studying the carbon sequestration function of urban wetland is urgently needed. In this work, the wetland in Wuhan was taken as the main research object. The landscape pattern index of Wuhan wetland was calculated and the evolution characteristics of landscape pattern were analyzed using the remote sensing image interpretation data from 2000 to 2020. CASA model was used to estimate the carbon sequestration of wetlands and explore the spatial and temporal distribution pattern of carbon sequestration. Spearman correlation was used to study the effects of different landscape pattern parameters on carbon sequestration. Results indicate the following: (1) From 2000 to 2020, the total wetland area in Wuhan has been reduced by 158.99 km2, among which the area of lakes, reservoirs and marshes decreased by 60.65 km2, 55.51 km2 and 41.77 km2, respectively. The density of wetland patches and the degree of fragmentation and irregularity increased, and the LPI, aggregation degree and diversity decreased. (2) In the past 20 years, the total carbon sequestration amount of wetland in Wuhan first decreased and then increased, among which the carbon sequestration in wetland was the lowest in 2010 (571,900 t). The amount of carbon sequestration in 2020 was the same as that in 2000, reaching 800,000 t. (3) A larger wetland area means more concentrated and denser distribution, more complex shape, more diverse type, and stronger carbon sequestration capacity for Wuhan wetland. On the contrary, the distribution of wetlands becomes more fragmented and regular, and the disordered planning leads to the reduction in wetland types in a unit area, which will decrease the carbon sink capacity of urban wetlands.

Abstract Background Moderate and deep sedation are well-established techniques in many developed countries, and several guidelines have been published. However, they have received attention in China only in recent years. The aim of this study is to investigate current paediatric sedation practices in tertiary children’s hospitals and tertiary maternity and children hospitals in China. Methods All tertiary children’s hospitals and tertiary maternity and children hospitals registered with the National Health Commission of the People’s Republic of China were invited to participate in an electronic survey, which included information on the sedation caseload, facility availability, staff structure, clinical skill requirements for sedation providers, fasting guidelines, patient-monitoring practices, and choice of sedatives. Results Fifty-eight of the 63 hospitals that completed the survey (92.1%) provided moderate and deep sedation. Dedicated sedation rooms and post-sedation recovery rooms were found in 14 (24.1%) and 19 (32.8%) hospitals, respectively. Sedation for non-invasive procedures was primarily performed by anaesthesiologists (69.0%); however, 75.9% of the sedation providers had not received paediatric basic or advanced life-support training. Children were asked to fast from clear liquids for at least 2 h in 44.8% of hospitals and up to 6 h in 5.2% of hospitals; they were asked to fast from solid food/milk for at least 4 h in 27.6% of hospitals and more than 8 h in 1.7% of hospitals. The most commonly used sedative in all groups was chloral hydrate. For rescue, propofol was the most widely used sedative, particularly for children older than 4 years. Conclusions Moderate and deep sedation practices vary widely in tertiary children’s hospitals and tertiary maternity and children hospitals in China. Optimised practices should be established to improve the quality of moderate and deep sedation.

Abstract Interference is the main performance-limiting factor in most wireless networks. Protocol interference model is extensively used in the design of wireless networks. However, the setting of interference range, a crucial part of the protocol interference model, is rather heuristic and remains an open problem. In this paper, we use the stochastic geometry and the direct approach to obtain the associated feasibility distributions. After that, we use the binary hypothesis testing to achieve the Bayes risk under binomial point process (BPP) and Poisson point process (PPP), respectively. According to the first derivative of the Bayes risk, we provide the equation to achieve the optimal interference range for minimum Bayes risk. We extend the method proposed by Wildman et al. to a more general situation. Furthermore, we show that for infinite PPP, those two methods converge to the same results. Several numerical results for wireless networks under BPP, finite PPP, and infinite PPP are given. Simulation results show that in the finite wireless network, the BPP method performs better than the PPP method.

Osteoporosis is the most common bone metabolic disease. Abnormal osteoclast formation and resorption play a fundamental role in osteoporosis pathogenesis. Recent researches have greatly broaden our understanding of molecular mechanisms of osteoporosis. However, the molecular mechanisms of key mRNAs and lncRNAs, and their interactions leading to osteoporosis are still not entirely clear. The purpose of this work is to study the key mRNAs and lncRNAs, and their interactions involved in bone mineral homeostasis and osteoclastogenesis. Systematic analyses such as differential expression analysis, GO and KEGG analysis, and PPI network construction revealed that up-regulated mRNAs were significantly enriched in inflammation-related pathways. Moreover, we observed that the down-regulated proteins, including JDP2, HADC4, HDAC5, CDYL2, ACADVL, ACSL1 and BRD4, were key components in the down-regulated PPI network, indicating that the downregulation of histone deacetylases and cofactors, such as, HDAC4, HDAC5 and JDP2 may be critical regulators in osteoclastogenesis. In addition, we also highlighted one lncRNA, RP11-498C9.17, was highly correlated with epigenetic regulators, such as HDAC4, MORF4L1, HMGA1 and DND1, indicating that the lncRNA RP11-498C9.17 may also be an epigenetic regulator. In conclusion, our integrative analysis reveals key mRNAs and lncRNAs, involved in bone mineral homeostasis and osteoclastogenesis, which not only broaden our insights into lncRNAs in bone mineral homeostasis and osteoclastogenesis, but also improve our understanding of molecular mechanism.

Abstract Background Acute lymphoblastic leukemia (ALL) contains cytogenetically distinct subtypes that respond differently to cytotoxic drugs. Therefore, subtype classification is important and indispensable in ALL diagnosis. In our previous study, we identified some marker genes in childhood ALL by means of microarray technology and, furthermore, detected the relative expression levels of 57 marker genes and built a comparatively convenient and cost-effective classifier with a prediction accuracy as high as 94% based on the advanced fragment analysis (AFA) technique. Methods A more convenient improved AFA (iAFA) technique with one-step multiplex RT-PCR and an anti-contamination system was developed to detect 57 marker genes for ALL. Results The iAFA assay is much easier and more convenient to perform than the previous AFA assay and has a prediction accuracy of 95.29% in ALL subtypes. The anti-contamination system could effectively prevent the occurrence of lab DNA contamination. We also showed that marker gene expression profiles in pediatric ALL revealed 2 subgroups with different outcomes. Most ALL patients (95.8%) had a good-risk genetic profile, and only 4.2% of ALL patients had a poor-risk genetic profile, which predicted an event-free survival (EFS) of 93.6 ± 1.3% vs 18.8 ± 9.8% at 5 years, respectively (P

When building a map of a dynamic environment, simultaneous localization and mapping systems have problems such as poor robustness and inaccurate pose estimation. This paper proposes a new mapping method based on the ORB-SLAM2 algorithm combined with the YOLOv5 network. First, the YOLOv5 network of the tracing thread is used to detect dynamic objects of each frame, and to get keyframes with detection of dynamic information. Second, the dynamic objects of each image frame are detected using the YOLOv5 network, and the detected dynamic points are rejected. Finally, the global map is constructed using the keyframes after eliminating the highly dynamic objects. The test results using the TUM dataset show that when the map is constructed in a dynamic environment, compared with the ORB-SLAM2 algorithm, the absolute trajectory error of our algorithm is reduced by 97.8%, and the relative positional error is reduced by 59.7%. The average time consumed to track each image frame is improved by 94.7% compared to DynaSLAM. In terms of algorithmic real-time performance, this paper’s algorithm is significantly better than the comparable dynamic SLAM map-building algorithm DynaSLAM.

Sorghum [Sorghum bicolor (L.) Moench] seed germination is sensitive to salinity, and seed priming is an effective method for alleviating the negative effects of salt stress on seed germination. However, few studies have compared the effects of different priming agents on sorghum germination under salt stress. In this study, we quantified the effects of priming with distilled water (HP), sodium chloride (NaCl), potassium chloride (KCl), calcium chloride (CaCl2), and polyethylene glycol (PEG) on sorghum seed germination under 150 mM NaCl stress. The germination potential, germination rate, germination index, vigor index, root length, shoot length, root fresh weight, shoot fresh weight, root dry weight, and shoot dry weight were significantly reduced by salt stress. Different priming treatments alleviated the germination inhibition caused by salt stress to varying degrees, and 50 mM CaCl2 was the most effective treatment. In addition, the mitigation effect of priming was stronger on root traits than on shoot traits. Mitigation efficacy was closely related to both the type of agent and the concentration of the solution. Principal component analysis showed that all concentrations of CaCl2 had higher scores and were clearly distinguished from other treatments based on their positive effects on all germination traits. The effects of the other agents varied with concentration. The priming treatments were divided into three categories based on their priming efficacy, and the 50, 100, and 150 mM CaCl2 treatments were placed in the first category. The 150 mM KCl, 10% PEG, HP, 150 mM NaCl, 30% PEG, and 50 mM KCl treatments were placed in the second category, and the 100 mM NaCl, 100 mM KCl, 20% PEG, and 50 mM NaCl treatments were least effective and were placed in the third category. Choosing appropriate priming agents and methods for future research and applications can ensure that crop seeds germinate healthily under saline conditions.

Genomic regions with repeated sequences are unstable and prone to rapid DNA diversification. However, the role of tandem repeats within the coding region is not fully characterized. Here, we have identified a new hypervariable C-type lectin gene family with different numbers of tandem repeats (Rlecs; R means repeat) in oriental river prawn (Macrobrachium nipponense). Two types of repeat units (33 or 30 bp) are identified in the second exon, and the number of repeat units vary from 1 to 9. Rlecs can be classified into 15 types through phylogenetic analysis. The amino acid sequences in the same type of Rlec are highly conservative outside the repeat regions. The main differences among the Rlec types are evident in exon 5. A variable number of tandem repeats in Rlecs may be produced by slip mispairing during gene replication. Alternative splicing contributes to the multiplicity of forms in this lectin gene family, and different types of Rlecs vary in terms of tissue distribution, expression quantity and response to bacterial challenge. These variations suggest that Rlecs have functional diversity. The results of experiments on sugar binding, microbial inhibition and clearance, regulation of antimicrobial peptide gene expression and prophenoloxidase activation indicate that the function of Rlecs with the motif of YRSKDD in innate immunity is enhanced when the number of tandem repeats increases. Our results suggest that Rlecs undergo gene expansion through gene duplication and alternative splicing, which ultimately leads to functional diversity.

Waterlogging stress is one of the most important abiotic stresses limiting sorghum growth and development. Consequently, the responses of sorghum to waterlogging must be monitored and studied. This study investigated changes in the leaf water status, xylem exudation rate, leaf anatomical structure, leaf temperature and photosynthetic performance. Waterlogging-tolerant (Jinuoliang 01, abbreviated JN01) and waterlogging-sensitive (Jinza 31, abbreviated JZ31) sorghum cultivars were planted in pots. The experiment was carried out using a split block design with three replications. Waterlogging stress was imposed at the sorghum five-leaf stage. The leaf free water content (FWC) and relative water content (RWC) decreased under the waterlogged condition. The leaf thickness was thinner under the waterlogged condition, and the main changes occurred in the upper epidermal and mesophyll cells. Gas exchange parameters and the xylem exudation rate were also restrained by waterlogging; however, greater responses of these parameters were observed in JZ31. JZ31 had a higher leaf-air temperature difference (ΔT) than JN01. We found that changes in ΔT were always consistent with changes in the RWC and the gas exchange parameters. ΔT was significantly associated with the leaf RWC, photosynthetic rate (Pn) and transpiration rate (Tr). The results suggest that ΔT may be an indicator reflecting the water status in leaves and can be used to evaluate the tolerance of sorghum to waterlogging.

Root function plays a vital role in maintaining crop production. However, the role of deep roots in yield production and their effects on photosynthetic performance in sorghum remain unclear. This study aimed to provide theoretical supports for establishing highly efficient root systems of sorghum to achieve more yield under certain conditions. In this study, two sorghum (Sorghum bicolor L. Moench) cultivars, Jiza127 and Jiza305, were cultivated in soil columns as experimental materials. Three treatments (no roots removed, CK; roots removed at 30 cm underground, R30; roots removed at 60 cm underground, R60) were carried out under drought conditions during the filling stage. The root bleeding intensity, endogenous substances in the root bleeding sap, photosynthetic characteristics, dry matter accumulation, and yield were measured. The results showed that R30 and R60 significantly reduced yield in both sorghum cultivars, and the effect of R30 on yield was greater than that of R60. The contributions of roots below 30 cm to the yield of both sorghum hybrids were notably higher than those below 60 cm. R30 significantly reduced the dry matter weights (DMWs) of leaves, stems, sheaths, and panicles. R60 significantly reduced the DMW of panicles but had no significant effect on the DMWs of leaves and stems. R30 significantly reduced the photosynthetic level and PSII reaction center activity; however, the effect of R60 was not significant. Although both R30 and R60 significantly reduced root activity and the soluble sugar, amino acid, gibberellin (GA3), and abscisic acid (ABA) contents of the root bleeding sap, some of the above indicators in R60 were significantly higher than those in R30 during the filling stage, indicating that the deeper roots (below 30 cm) had a critical regulatory effect on the physiological processes of the aerial parts in sorghum, which resulted in a stronger effect on yield, especially under drought conditions. In brief, the deep roots of sorghum played a key role in yield production, but the roots in different soil depths regulated yield production in different ways. Our results indicate that deep roots of sorghum deserve consideration as a potential trait for yield improvement especially under drought conditions.
