Background Transcription Enhanced Association Domain (TEAD) transcription factors family involves in many signal transduction pathways, and is necessary for cell proliferation, apoptosis and other vital activities. TEAD1-4 is abnormally expressed in many tumors, and is related to tumor progression, drug resistance, poor prognosis, etc. In this study, we assessed the effect of the TEAD family on pan-cancer. Methods We downloaded transcriptome and clinical data from 33 types of TCGA tumor samples and adjacent tissues. Then, the expression characteristics of TEAD family members TEAD1, TEAD2, TEAD3 and TEAD4 in pan-cancer were analyzed. R packet and Cox regression were used to analyze the clinical correlation. In addition, the transcription level of TEAD and immune subtypes, stem cells, immune tumor microenvironment and drug sensitivity were analyzed by multidimensional correlation. Results We found the expression of TEAD1-4 was significantly different in various tumors. The survival time of patients with high and low TEAD1-4 gene expression was significantly different. TEAD1-4 was significantly associated with immune-infiltrating subtypes of tumor microenvironment (TME), and was positively correlated with interstitial cell infiltration and tumor cell dryness. The transcriptional expression level of TEAD1-4 was also related to drug response. TEAD may play a role in the sensitivity or resistance of tumor cells to drug therapy, and may serve as a therapeutic target to overcome drug-induced resistance. Conclusions Our systematic analysis reveals TEAD gene expression and its relationship with immune invasion, TME, tumor stem cells, drug sensitivity, and prognosis in cancer patients, and it is necessary to study the role of each TEAD family member in specific cancer type. Although further laboratory validation is needed, our study confirms that TEAD family genes are promising therapeutic targets and potential prognostic indicators in cancers.

Background This study explored disparities in characteristics and mortalities among four major transmission groups on antiretroviral therapy in northwest China as well as the survival impact of each transmission route. Methods We first examined disparities in demographics and clinical characteristics of the four transmission populations. Kaplan Meier analysis was subsequently conducted to compare survival rates among all groups. At last, Cox proportional hazards regression model was employed to analyze the survival impact of a transmission route among seven main categories of survival factors associated with all-cause mortalities. Results Survival analysis showed significant differences in all-cause, AIDS- and non-AIDS-related deaths among four HIV populations (all P Conclusions Significant disparities were found in characteristics and mortalities among the four transmission groups where mortality disparities were mainly due to non-AIDS-related death. Suggestions are provided for each group to improve their survivorship.

Background: The aim of this study was to analyze and compare melanoma gene expression profiles in TCGA database through the application of different genes to explore the pathogenesis of melanoma. Furthermore, we confirmed the extent of the role of KYNU in melanoma and whether it can be a potential target for the diagnosis and treatment of melanoma. Methods: The gene expression profiles of melanoma samples were downloaded from TCGA database, and matrix files were synthesized to screen differential genes. The Kyoto Encyclopedia of Genes and Genomes (KEGG) signaling pathway analysis and GCDA broad institute were used to analyze common gene locus mutations and expression changes in melanoma, as well as methylation. In addition, the expression patterns of KYNU in melanoma were quantified by immunohistochemistry, Western blotting, qRT-PCR, software such as GEO DataSets and the Human Protein Atlas, and meta-analysis of skin diseases. KYNU was overexpressed in keratinocytes (HaCaT and HEKα) and melanoma cells (A375 and H1205-lu). CFDA-SE, Annexin V–PI double staining, and PI single staining were used to investigate the mechanism of KYNU in melanoma and its effects on melanoma proliferation, apoptosis, invasion, and migration. Results: The main signaling pathways involved in melanoma were EGF/EGFR–RAS–BRAF–MEK–ERK–CyclinD1/CDK4, Ras–PI3K–PTEN–PKB/AKT, and p14/p16 (CDKN2A)–MDM2–p53–p21–cyclinD1/CDK4/6–Rb/E2F. Moreover, MITF, KIT, CDH1. NRAS, AKT1, EGFR, TP53, KIT, and CDK4 were elevated in melanoma, whereas PTEN, cAMP, and BCL2 were reduced in melanoma. The copy number of tumor-promoting genes increased, while the copy number of tumor suppressor genes decreased. Changes in the copy number of the above tumor genes enriched in chromosomes were found through SNP gene mutations. The genes whose expression was negatively regulated by DNA methylation in melanoma included KRT18, CDK2, JAK3, BCL2, MITF, MET, CXCL10, EGF, SOX10, SOCS3, and KIT. The mutation rate of KYNU was high according to TCGA database. The KYNU level was decreased in melanoma. Overexpression of KYNU can promote changes in apoptotic BCL-2, metabolic KYN, 3-HAA, invasion and migration MMP9, E-cadherin, and other related proteins in melanoma. Fluorescence staining and flow analysis showed that a slower proliferation rate led to a stronger fluorescence intensity. In melanoma tumor cells with a low expression of KYNU, overexpression of KYNU could promote tumor cell apoptosis. IL-10 induced immunoregulatory changes in melanoma. The expression of MMP9 and AMPK decreased in A375, but the change in BCL-2 was not obvious. The expression of BCL-2 decreased significantly in H1205-lu. A375 showed cell-cycle arrest, indicating that IL-10 could slow down the cell cycle of melanoma. Conclusions: These results provide insights into the pathologic mechanisms of melanoma target genes and KYNU as a biomarker and potential therapeutic factor for melanoma.

An approximate method is introduced to predict the distribution of source reflection coefficient when transmission path with stochastic height above ground in any position. The transmission path is divided into many segments, assuming that the height of each segment is independent and identically distributed. When the height is logarithmic normal distributed, expression of mean value and variance are derived, the values are referred to as calculated value. Meanwhile, the accurate expression of source reflection coefficient is obtained based on transmission line theory, then the mean value and variance are calculated in a statistical way by calculating the source reflection coefficient thousands of times. The values are referred to as statistical value. Moreover, the comparison between the statistical value and the calculated value shows the validity and the benefit of this approach. With the number of segments increase, the approximate method is more accurate. What is more, the calculated value consists well with the statistical value in a wide frequency range. Compared with the other statistical method, the approximate method is easier and more feasible.

This paper describes a fault location method for shielded cables based on coupling characteristics that can be used to test the integrity of shielded cables nondestructively. A shield damaged coaxial cable and a wire are modelled which normally be used in aircraft cable bundles. The frequency domain coupling characteristic is affected which phase is related to the location of fault when the shield damage occurs. To infer calculate the coupling characteristics, Bethe theory is applied to a hole in the shield. Then by calculating phase of coupling signal using the signal flow network based on wave quantity, is the fault location is found. The mothed has a bandwidth of 300MHz–550MHz, a range of 1.5 m, and a resolution of 5 cm and can determine the fault location of shielded cables from measurements at a single end. The suggested method is validated by full wave simulation, and it is consistent with theoretical results, which indicate that the modeling and the method are correct. The effect of cable and mismatching losses limits the dynamic range of the measurement.

Analytical model of electromagnetic coupling between two crossing wires is calculated in this paper. Based on the wave quantity theory, the S-parameter expression of crossing wires model is derived. Compared with the full-wave method, this approach is easier and more feasible. Moreover, the comparison between theoretical and MOM simulation results shows the validity and the interest of this approach.

Using first-principles calculations, we have systematically investigated the structural, electronic and magnetic properties of blue phosphorene doped by group IV and VI atoms, including C, Si, Ge, Sn, O, S, Se and Te. All the doped systems are energetically stable. Only C, Si, Ge and O-substituted systems show the characteristics of spin polarization and the magnetic moments are all 1.0 μB. Moreover, we found that C, Si, Ge and O doped systems are indirect bandgap semiconductors, while Sn, S, Se and Te doped systems present metallic property. These results show that blue phosphorene can be used prospectively in optoelectronic and spintronic devices.

Vanadium-based alloys are considered to be one of the most promising hydrogen separation membranes due to their high hydrogen permeability. In this study, we investigate the dissolution and diffusion behaviors of hydrogen in vanadium-based binary alloys, V 15 M (where M = Al, Ti, Cr, Fe, Ni and Nb) alloys, using first-principles method based on density functional theory. The dissolution of hydrogen in V 15 M alloys is affected by both the elastic and electronic properties, but the elastic effect is the main factor. The H solution energies in the alloys follow the sequence: V Ti Nb Al Cr Ni Fe, and a smaller atom size increase the H solution energy. Therefore, the addition of alloying elements with smaller atomic sizes can reduce the solubility of hydrogen in vanadium and inhibit hydrogen embrittlement. For hydrogen diffusion, alloying elements Al, Ti and Nb can be good candidates because they have a higher diffusion coefficient. The V Ti alloy has the highest hydrogen permeability, but will have serious hydrogen embrittlement due to the increased H solubility.

STUDY DESIGN Prospective study. OBJECTIVE To analyze the most feasible choice of C1 lateral mass (C1LM) and C2 pedicle (C2P) screw in upper cervical surgeries for children younger than 6 years. SUMMARY OF BACKGROUND DATA The C1LM and C2P screw technique is a stable cervical vertebrae internal fixation method in upper cervical surgery. Some tomographic studies have indicated the feasibility of insertion of C1LM and C2P screws in children. Their results, however, varied widely, and no consensus was achieved regarding C1LM and C2P screw choices for different ages in the pediatric population, sex, and laterality. METHODS The computed tomography images of 250 patients (age 2-6 yr) were studied. The inner diameter and length for each C1LM and C2P were measured in axial view. Height was measured in sagittal view. Screw choice was considered feasible when a sample maintained an additional 0.5 mm bone cortex at the inner diameter, length, and height at the same time. Analyses with the Student t test were performed for age, sex, and laterality. The screw choice was evaluated with a feasibility percentage. Objective. To have preliminary knowledge of the current quality of the medical records in the Inner Mongolia Autonomous Region by evaluating and checking medical records, summarize and analyze existing problems, and provide guidance for future control over the quality of medical records. Method. Randomly extracting 250 medical records from 25 tertiary general hospitals in the whole region, wherein 50 copies are death medical records, 100 copies are cases with operations above class B in the surgery department and 100 copies are intractable or critical medical records in internal department. The quality of the extracted medical records in ten specialties is evaluated by ten medical associated professional specialists, and the evaluation result is reviewed by management specialists. Objective. To analyze the causes of unplanned resurgery, and provide an objective basis for the quality evaluation and improvement of the hospital's medical treatment services. Method. A retrospective analysis of 40 cases of unplanned resurgery in a hospital in 2012 was carried out. Result. The rate of occurrence of unplanned resurgery was 0.55% in the hospital in 2012; the main causes included postoperative bleeding, inconsistency between the results of the intraoperative frozen section examination and the pathological examination, which needed further consultation, and incision dehiscence. The proportions of these were 55%, 13% and 10%, respectively. Conclusion. Our department should strengthen perioperative management; the hospital should consider unplanned resurgery as the focus of medical quality management and strictly implement operation grading management and reauthorization systems to decrease the incidence of unplanned resurgery.
