Abstract: The "Annual Disability Statistics Compendium," is a summary of statistics about people with disabilities and about the government programs which serve them. The Compendium, presents key overall statistics on topics including the prevalence of disability, employment among persons with disabilities, rates of participation in disability income and social insurance programs, and other statistics. It is a compilation of data from multiple sources, such as the Social Security Administration, Veterans Benefits Administration, and frequently, the U.S. Census Bureau's American Community Survey, among others. A companion "Annual Report" is available, providing graphic representations of key findings.

Abstract: Abstract We report a determination of the CKM angle ϕ 3, also known as γ, from a combination of measurements using samples of up to 711 fb −1 from the Belle experiment and up to 362 fb −1 from the Belle II experiment. We combine results from analyses of B + → DK +, B + → Dπ +, and B + → D * K + decays, where D is an admixture of D 0 and D ¯ 0 $$ {\overline{D}}^0 $$ mesons, in a likelihood fit to obtain ϕ 3 = (75.2 ± 7.6) ° . We also briefly discuss the interpretation of this result.

Abstract: Abstract We report measurements of the e + e − → B B ¯ $$ B\overline{B} $$ , B B ¯ ∗ $$ B{\overline{B}}^{\ast } $$ , and B ∗ B ¯ ∗ $$ {B}^{\ast }{\overline{B}}^{\ast } $$ cross sections at four energies, 10653, 10701, 10746 and 10805 MeV, using data collected by the Belle II experiment. We reconstruct one B meson in a large number of hadronic final states and use its momentum to identify the production process. In the first 2 – 5 MeV above B ∗ B ¯ ∗ $$ {B}^{\ast }{\overline{B}}^{\ast } $$ threshold, the e + e − → B ∗ B ¯ ∗ $$ {B}^{\ast }{\overline{B}}^{\ast } $$ cross section increases rapidly. This may indicate the presence of a pole close to the threshold.

4. Disturbed function of TBL1X has a differential effect on T3-regulated gene expression in two human liver cell models

Abstract: Background: Mutations in TBL1X, part of the NCOR1/SMRT corepressor complex, were identified in patients with hereditary X-linked central congenital hypothyroidism and associated hearing loss. The role of TBL1X in thyroid hormone (TH) action, however, is incompletely understood. The aim of the present study was to investigate the role of TBL1X on T3-regulated gene expression in two human liver cell models. Methods: A human hepatoma cell line (HepG2) wherein TBL1X was downregulated using siRNAs, and human-induced pluripotent stem cell-derived hepatocytes (iHeps) generated from individuals with a TBL1X N365Y mutation. Both cell types were treated with increasing concentrations of T3. The expression of T3-regulated genes was measured by qPCR. Results: KLF9, CPT1A, and PCK1 mRNA expression were higher upon T3 stimulation in the HepG2 cells with decreased TBL1X expression compared to controls, while DIO1 mRNA expression was lower. Hemizygous TBL1X N365Y iHeps exhibited decreased expression of CPT1A, G6PC1, PCK1, FBP1, and ELOVL2 compared to cells with the heterozygous TBL1X N365Y allele, but KLF9 and HMGCS2 expression was unaltered. Conclusion: Downregulation of TBL1X in HepG2 cells and the TBL1X N365Y variant in iHeps have differential effects on T3-regulated gene expression. This suggests that TBL1X may play a gene context role in TH action.

Abstract: Abstract We present a study of Ξ c 0 → Ξ 0 π 0 $$ {\Xi}_c^0\to {\Xi}^0{\pi}^0 $$ , Ξ c 0 → Ξ 0 η $$ {\Xi}_c^0\to {\Xi}^0\eta $$ , and Ξ c 0 → Ξ 0 η ′ $$ {\Xi}_c^0\to {\Xi}^0{\eta}^{\prime } $$ decays using the Belle and Belle II data samples, which have integrated luminosities of 980 fb −1 and 426 fb −1, respectively. We measure the following relative branching fractions B Ξ c 0 → Ξ 0 π 0 / B Ξ c 0 → Ξ − π + = 0.48 ± 0.02 stat ± 0.03 syst , B Ξ c 0 → Ξ 0 η / B Ξ c 0 → Ξ − π + = 0.11 ± 0.01 stat ± 0.01 syst , B Ξ c 0 → Ξ 0 η ′ / B Ξ c 0 → Ξ − π + = 0.08 ± 0.02 stat ± 0.01 syst $$ {\displaystyle \begin{array}{c}\mathcal{B}\left({\Xi}_c^0\to {\Xi}^0{\pi}^0\right)/\mathcal{B}\left({\Xi}_c^0\to {\Xi}^{-}{\pi}^{+}\right)=0.48\pm 0.02\left(\textrm{stat}\right)\pm 0.03\left(\textrm{syst}\right),\\ {}\mathcal{B}\left({\Xi}_c^0\to {\Xi}^0\eta \right)/\mathcal{B}\left({\Xi}_c^0\to {\Xi}^{-}{\pi}^{+}\right)=0.11\pm 0.01\left(\textrm{stat}\right)\pm 0.01\left(\textrm{syst}\right),\\ {}\mathcal{B}\left({\Xi}_c^0\to {\Xi}^0{\eta}^{\prime}\right)/\mathcal{B}\left({\Xi}_c^0\to {\Xi}^{-}{\pi}^{+}\right)=0.08\pm 0.02\left(\textrm{stat}\right)\pm 0.01\left(\textrm{syst}\right)\end{array}} $$ for the first time, where the uncertainties are statistical (stat) and systematic (syst). By multiplying by the branching fraction of the normalization mode, B Ξ c 0 → Ξ − π + $$ \mathcal{B}\left({\Xi}_c^0\to {\Xi}^{-}{\pi}^{+}\right) $$ , we obtain the following absolute branching fraction results B Ξ c 0 → Ξ 0 π 0 = 6.9 ± 0.3 stat ± 0.5 syst ± 1.3 norm × 10 − 3 , B Ξ c 0 → Ξ 0 η = 1.6 ± 0.2 stat ± 0.2 syst ± 0.3 norm × 10 − 3 , B Ξ c 0 → Ξ 0 η ′ = 1.2 ± 0.3 stat ± 0.1 syst ± 0.2 norm × 10 − 3 , $$ {\displaystyle \begin{array}{c}\mathcal{B}\left({\Xi}_c^0\to {\Xi}^0{\pi}^0\right)=\left(6.9\pm 0.3\left(\textrm{stat}\right)\pm 0.5\left(\textrm{syst}\right)\pm 1.3\left(\operatorname{norm}\right)\right)\times {10}^{-3},\\ {}\mathcal{B}\left({\Xi}_c^0\to {\Xi}^0\eta \right)=\left(1.6\pm 0.2\left(\textrm{stat}\right)\pm 0.2\left(\textrm{syst}\right)\pm 0.3\left(\operatorname{norm}\right)\right)\times {10}^{-3},\\ {}\mathcal{B}\left({\varXi}_c^0\to {\Xi}^0{\eta}^{\prime}\right)=\left(1.2\pm 0.3\left(\textrm{stat}\right)\pm 0.1\left(\textrm{syst}\right)\pm 0.2\left(\operatorname{norm}\right)\right)\times {10}^{-3},\end{array}} $$ where the third uncertainties are from B Ξ c 0 → Ξ − π + $$ \mathcal{B}\left({\Xi}_c^0\to {\Xi}^{-}{\pi}^{+}\right) $$ . The asymmetry parameter for Ξ c 0 → Ξ 0 π 0 $$ {\Xi}_c^0\to {\Xi}^0{\pi}^0 $$ is measured to be α Ξ c 0 → Ξ 0 π 0 = − 0.90 ± 0.15 stat ± 0.23 syst $$ \alpha \left({\Xi}_c^0\to {\Xi}^0{\pi}^0\right)=-0.90\pm 0.15\left(\textrm{stat}\right)\pm 0.23\left(\textrm{syst}\right) $$ .

Abstract: Abstract We present the result of a search for the charged-lepton-flavor violating decay τ − → μ − μ + μ − using a 424 fb −1 sample of data recorded by the Belle II experiment at the SuperKEKB e + e − collider. The selection of e + e − → τ + τ − events is based on an inclusive reconstruction of the non-signal tau decay, and on a boosted decision tree to suppress background. We observe one signal candidate, which is compatible with the expectation from background processes. We set a 90% confidence level upper limit of 1.9 × 10 −8 on the branching fraction of the τ − → μ − μ + μ − decay, which is the most stringent bound to date.

Abstract: Abstract We present measurements of the branching fractions of eight B ¯ 0 $$ {\overline{B}}^0 $$ → D (*)+ K − K S ∗ 0 $$ {K}_{(S)}^{\left(\ast \right)0} $$ , B − → D (*)0 K − K S ∗ 0 $$ {K}_{(S)}^{\left(\ast \right)0} $$ decay channels. The results are based on data from SuperKEKB electron-positron collisions at the Υ(4S) resonance collected with the Belle II detector, corresponding to an integrated luminosity of 362 fb −1. The event yields are extracted from fits to the distributions of the difference between expected and observed B meson energy, and are efficiency-corrected as a function of m(K − K S ∗ 0 $$ {K}_{(S)}^{\left(\ast \right)0} $$ ) and m(D (*) K S ∗ 0 $$ {K}_{(S)}^{\left(\ast \right)0} $$ ) in order to avoid dependence on the decay model. These results include the first observation of B ¯ 0 $$ {\overline{B}}^0 $$ → D + K − K S 0 $$ {K}_S^0 $$ , B − → D* 0 K − K S 0 $$ {K}_S^0 $$ , and B ¯ 0 $$ {\overline{B}}^0 $$ → D* + K − K S 0 $$ {K}_S^0 $$ decays and a significant improvement in the precision of the other channels compared to previous measurements. The helicity-angle distributions and the invariant mass distributions of the K − K S ∗ 0 $$ {K}_{(S)}^{\left(\ast \right)0} $$ systems are compatible with quasi-two-body decays via a resonant transition with spin-parity J P = 1− for the K − K S 0 $$ {K}_S^0 $$ systems and J P = 1+ for the K − K* 0 systems. We also present measurements of the branching fractions of four B ¯ 0 $$ {\overline{B}}^0 $$ → D (*)+ D s − $$ {D}_s^{-} $$ , B − → D (*)0 D s − $$ {D}_s^{-} $$ decay channels with a precision compatible to the current world averages.

Abstract: Abstract We present an analysis of the process e + e − → π + π − Υ(nS) (where n = 1, 2, or 3) reconstructed in 19.6 fb −1 of Belle II data during a special run of the SuperKEKB collider at four energy points near the peak of the Υ(10753) resonance. By analyzing the mass distribution of the π + π − Υ(nS) system and the Born cross sections of the e + e − → π + π − Υ(nS) process, we report the first observation of Υ(10753) decays to the π + π − Υ(1S) and π + π − Υ(2S) final states, and find no evidence for decays to π + π − Υ(3S). Possible intermediate states in the π + π − Υ(1S, 2S) transitions are also investigated, and no evidence for decays proceeding via the π ∓ Z b ± $$ {\pi}^{\mp }{Z}_b^{\pm } $$ or f 0(980)Υ(nS) intermediate states is found. We measure Born cross sections for the e + e − → π + π − Υ(nS) process that, combined with results from Belle, obtain the mass and width of Υ(10753) to be (10756.6 ± 2.7 ± 0.9) MeV/c 2 and (29.0 ± 8.8 ± 1.2) MeV, respectively. The relative ratios of the Born cross sections at the Υ(10753) resonance peak are also reported for the first time.

Abstract: Abstract One of the most vital parameters to achieve sustainability in any field is encompassing the Occupational Health and Safety (OHS) of the workers. In mining industry where heavy earth moving machineries are largely employed, ergonomic hazards turn out to be significant OHS hazards causing Musculoskeletal Disorders (MSDs) in the operators. Nevertheless, the Indian mining industry lacks a comprehensive technique of OHS risk assessment, especially for ergonomic hazards that cause MSDs. This research appraises ergonomic hazards and develops Fuzzy Musculoskeletal-disorders Index (FMI) model to evaluate ergonomic-related MSDs. Work process and work tool ergonomic risk factors were identified through literature review and directives recommended by experts. Work posture was evaluated using RULA. The data-collecting approach was implemented using participatory ergonomic and design science principles. The FMI results show average MSDs score of 3.69, indicating high to extremely high risk. Surface plots show that combined work tool and work process was the most sensitive factors to MSDs risk compared to other two combinations. A two-sample t-test validated the FMI. The findings should help safety experts and managers develop effective OHS management plans and programmes for the sustainability of Indian mining industry.

12. Machine learning workflow for edge computed arrhythmia detection in exploration class missions

Abstract: Abstract Deep-space missions require preventative care methods based on predictive models for identifying in-space pathologies. Deploying such models requires flexible edge computing, which Open Neural Network Exchange (ONNX) formats enable by optimizing inference directly on wearable edge devices. This work demonstrates an innovative approach to point-of-care machine learning model pipelines by combining this capacity with an advanced self-optimizing training scheme to classify periods of Normal Sinus Rhythm (NSR), Atrial Fibrillation (AFIB), and Atrial Flutter (AFL). 742 h of electrocardiogram (ECG) recordings were pre-processed into 30-second normalized samples where variable mode decomposition purged muscle artifacts and instrumentation noise. Seventeen heart rate variability and morphological ECG features were extracted by convoluting peak detection with Gaussian distributions and delineating QRS complexes using discrete wavelet transforms. The decision tree classifier’s features, parameters, and hyperparameters were self-optimized through stratified triple nested cross-validation ranked on F1-scoring against cardiologist labeling. The selected model achieved a macro F1-score of 0.899 with 0.993 for NSR, 0.938 for AFIB, and 0.767 for AFL. The most important features included median P-wave amplitudes, PRR20, and mean heart rates. The ONNX-translated pipeline took 9.2 s/sample. This combination of our self-optimizing scheme and deployment use case of ONNX demonstrated overall accurate operational tachycardia detection.

Abstract: Abstract Missions into Deep Space are planned this decade. Yet the health consequences of exposure to microgravity and galactic cosmic radiation (GCR) over years-long missions on indispensable visceral organs such as the kidney are largely unexplored. We performed biomolecular (epigenomic, transcriptomic, proteomic, epiproteomic, metabolomic, metagenomic), clinical chemistry (electrolytes, endocrinology, biochemistry) and morphometry (histology, 3D imaging, miRNA-ISH, tissue weights) analyses using samples and datasets available from 11 spaceflight-exposed mouse and 5 human, 1 simulated microgravity rat and 4 simulated GCR-exposed mouse missions. We found that spaceflight induces: 1) renal transporter dephosphorylation which may indicate astronauts’ increased risk of nephrolithiasis is in part a primary renal phenomenon rather than solely a secondary consequence of bone loss; 2) remodelling of the nephron that results in expansion of distal convoluted tubule size but loss of overall tubule density; 3) renal damage and dysfunction when exposed to a Mars roundtrip dose-equivalent of simulated GCR.

Abstract: Abstract We report results from a study of B ± → DK ± decays followed by D decaying to the CP-even final state K + K − and CP-odd final state K S 0 π 0 $$ {K}_S^0{\pi}^0 $$ , where D is an admixture of D 0 and D ¯ 0 $$ {\overline{D}}^0 $$ states. These decays are sensitive to the Cabibbo-Kobayashi-Maskawa unitarity-triangle angle ϕ 3. The results are based on a combined analysis of the final data set of 772 × 106 B B ¯ $$ B\overline{B} $$ pairs collected by the Belle experiment and a data set of 198 × 106 B B ¯ $$ B\overline{B} $$ pairs collected by the Belle II experiment, both in electron-positron collisions at the Υ(4S) resonance. We measure the CP asymmetries to be A $$ \mathcal{A} $$ CP+ = (+12.5 ± 5.8 ± 1.4)% and A $$ \mathcal{A} $$ CP− = (−16.7 ± 5.7 ± 0.6)%, and the ratios of branching fractions to be R $$ \mathcal{R} $$ CP+ = 1.164 ± 0.081 ± 0.036 and R $$ \mathcal{R} $$ CP− = 1.151 ± 0.074 ± 0.019. The first contribution to the uncertainties is statistical, and the second is systematic. The asymmetries A $$ \mathcal{A} $$ CP+ and A $$ \mathcal{A} $$ CP− have similar magnitudes and opposite signs; their difference corresponds to 3.5 standard deviations. From these values we calculate 68.3% confidence intervals of (8.5 ° < ϕ 3 < 16.5 ° ) or (84.5 ° < ϕ 3 < 95.5 ° ) or (163.3 ° < ϕ 3 < 171.5 ° ) and 0.321 < r B < 0.465.

Abstract: Objective An experiment was conducted to study the effect of supplementing DL methionine (DL Met) at graded concentrations on performance, carcass variables, immune responses and antioxidant variables in broiler chicken fed folic acid (FA) fortified (4 mg/kg) low-methionine diet. Methods A basal diet (BD) without supplemental DL Met, but with higher level (4 mg/kg) of FA and a control diet (CD) with the recommended concentration of methionine (Met) were prepared. The BD was supplemented with DL Met at graded concentrations (0%, 10%, 20%, 30%, 40%, and 50% supplemental DL Met of CD). Each diet was fed ad libitum to 10 replicates of 5 broiler male chicks in each from 1 to 42 d of age. Results Body weight gain (BWG) reduced, and feed conversion ratio (FCR) increased in broilers fed low-Met BD. At 30% and 20% inclusion of DL met, the BWG and FCR, respectively were similar to those fed the CD. Similarly, supplementation of 10% DL Met to the BD significantly increased ready to cook meat yield and breast meat weight, which were similar to those of the CD fed broilers. Lipid peroxidation reduced, the activity of antioxidant enzymes (GSHPx and GSHRx) in serum increased and lymphocyte proliferation increased with increased supplemental DL Met level in the BD. The concentrations of total protein and albumin in serum increased with DL Met supplementation to the BD. Conclusion Based on the data, it can be concluded that supplemental Met can be reduced to less than 50% in broiler chicken diets (4.40, 3.94, and 3.39 g/kg, respectively in pre-starter, starter and finisher phases) containing 4 mg/kg FA.

Abstract: Abstract We report a search for the charged-lepton flavor violation in Υ(2S) → ℓ ∓ τ ± (ℓ = e, μ) decays using a 25 fb −1 Υ(2S) sample collected by the Belle detector at the KEKB e + e − asymmetric-energy collider. We find no evidence for a signal and set upper limits on the branching fractions ( B $$ \mathcal{B} $$ ) at 90% confidence level. We obtain the most stringent upper limits: B $$ \mathcal{B} $$ (Υ(2S) → μ ∓ τ ± ) < 0.23 × 10 −6 and B $$ \mathcal{B} $$ (Υ(2S) → e ∓ τ ± ) < 1.12 × 10 −6.

21. Southern Weddell Sea surface freshwater flux modulated by icescape and atmospheric forcing

Abstract: Sea ice formation dominates surface salt forcing in the southern Weddell Sea. Brine rejected in the process of sea ice production results in the production of High Salinity Shelf Water (HSSW) that feeds the global overturning circulation and fuels the basal melt of the adjacent ice shelf. The strongest sea ice production rates are found in coastal polynyas, where steady offshore winds promote divergent ice movement during the freezing season. We used the Finite Element Sea ice–ice shelf–Ocean Model (FESOM) forced by output from the regional atmospheric model COSMO-CLM (CCLM) with 14 km horizontal resolution to investigate the role of polynyas for the surface freshwater flux of the southern Weddell Sea (2002–2017). The presence of stationary icescape features (i.e., fast-ice areas and grounded icebergs) can influence the formation of polynyas and, therefore, impact sea ice production. The representation of the icescape in our model is included by prescribing the position, shape and temporal evolution of a largely immobile ice mélange formed between the Filchner–Ronne Ice Shelf (FRIS) and a major grounded iceberg based on satellite data. We find that 70 % of the ice produced on the continental shelf of the southern Weddell Sea is exported from the region. While coastal polynyas cover 2 % of the continental shelf area, sea ice production within the coastal polynyas accounts for 17 % of the overall annual sea ice production (1509 km3). The largest contributions come from the Ronne Ice Shelf and Brunt Ice Shelf polynyas and polynyas associated with the ice mélange. Furthermore, we investigate the sensitivity of the polynya-based ice production to the (i) representation of the icescape and (ii) regional atmospheric forcing. Although large-scale atmospheric fields determine the sea ice production outside polynyas, both the treatment of the icescape and the regional atmospheric forcing are important for the regional patterns of sea ice production in polynyas. The representation of the ice mélange is crucial for the simulation of polynyas westward/eastward of it, which are otherwise suppressed/overestimated. Compared to using ERA-Interim reanalysis as an atmospheric forcing data set, using CCLM output reduces polynya-based ice production over the eastern continental shelf due to weaker offshore winds, yielding a more realistic polynya representation. Our results show that the location and not just the strength of the sea ice production in polynyas is a relevant parameter in setting the properties of the HSSW produced on the continental shelf, which in turn affects the basal melting of the Filchner–Ronne Ice Shelf.

Abstract: We have previously demonstrated that deletion of activin receptor-like kinase 1 (Alk1) or endoglin in a fraction of endothelial cells (ECs) induces brain arteriovenous malformations (bAVMs) in adult mice upon angiogenic stimulation. Here, we addressed three related questions: (1) could Alk1- mutant bone marrow (BM)-derived ECs (BMDECs) cause bAVMs? (2) is Alk1- ECs clonally expended during bAVM development? and (3) is the number of mutant ECs correlates to bAVM severity? For the first question, we transplanted BM from PdgfbiCreER;Alk12f/2f mice (EC-specific tamoxifen-inducible Cre with Alk1-floxed alleles) into wild-type mice, and then induced bAVMs by intra-brain injection of an adeno-associated viral vector expressing vascular endothelial growth factor and intra-peritoneal injection of tamoxifen. For the second question, clonal expansion was analyzed using PdgfbiCreER;Alk12f/2f;confetti+/- mice. For the third question, we titrated tamoxifen to limit Alk1 deletion and compared the severity of bAVM in mice treated with low and high tamoxifen doses. We found that wild-type mice with PdgfbiCreER;Alk12f/2f BM developed bAVMs upon VEGF stimulation and Alk1 gene deletion in BMDECs. We also observed clusters of ECs expressing the same confetti color within bAVMs and significant proliferation of Alk1- ECs at early stage of bAVM development, suggesting that Alk1- ECs clonally expanded by local proliferation. Tamoxifen dose titration revealed a direct correlation between the number of Alk1- ECs and the burden of dysplastic vessels in bAVMs. These results provide novel insights for the understanding of the mechanism by which a small fraction of Alk1 or endoglin mutant ECs contribute to development of bAVMs.

Abstract: Allogeneic hematopoietic cell transplantation (HCT) is used to treat many blood-based disorders and malignancies, however it can also result in serious adverse events, such as the development of acute graft-versus-host disease (aGVHD). This study aimed to develop a donor-specific epigenetic classifier to reduce incidence of aGVHD by improving donor selection. Genome-wide DNA methylation was assessed in a discovery cohort of 288 HCT donors selected based on recipient aGVHD outcome; this cohort consisted of 144 cases with aGVHD grades III-IV and 144 controls with no aGVHD. We applied a machine learning algorithm to identify CpG sites predictive of aGVHD. Receiver operating characteristic (ROC) curve analysis of these sites resulted in a classifier with an encouraging area under the ROC curve (AUC) of 0.91. To test this classifier, we used an independent validation cohort (n = 288) selected using the same criteria as the discovery cohort. Attempts to validate the classifier failed with the AUC falling to 0.51. These results indicate that donor DNA methylation may not be a suitable predictor of aGVHD in an HCT setting involving unrelated donors, despite the initial promising results in the discovery cohort. Our work highlights the importance of independent validation of machine learning classifiers, particularly when developing classifiers intended for clinical use.

26. Exploring the similarities between risk factors triggering depression in humans and elevated in-cage 'inactive but awake' behavior in laboratory mice

Abstract: IntroductionDepression is a human mental disorder that can also be inferred in non-human animals. This study explored whether time spent inactive but awake (“IBA”) in the home-cage in mice was further triggered by risk factors similar to those increasing vulnerability to depression in humans (early life stress, genetic predispositions, adulthood stress).MethodsEighteen DBA/2 J and 18 C57BL/6 J females were tested, of which half underwent as pups a daily maternal separation on post-natal days 2–14 (early-life stress “ELS”) (other half left undisturbed). To assess the effect of the procedure, the time the dams from which the 18 subjects were born spent active in the nest (proxy for maternal behavior) was recorded on post-natal days 2, 6, 10 and 14 for 1 h before separation and following reunion (matched times for controls), using live instantaneous scan sampling (total: 96 scans/dam). For each ELS condition, about half of the pups were housed post-weaning (i.e., from 27 days old on average) in either barren (triggering IBA and depression-like symptoms) or larger, highly enriched cages (n = 4–5 per group). Time mice spent IBA post-weaning was observed blind to ELS treatment using live instantaneous scan sampling in two daily 90-min blocks, two days/week, for 6 weeks (total: 192 scans/mouse). Data were analyzed in R using generalized linear mixed models.ResultsThe dams were significantly more active in the nest over time (p = 0.016), however with no significant difference between strains (p = 0.18), ELS conditions (p = 0.20) and before/after separation (p = 0.83). 27. Inhibition of malaria and babesiosis parasites by putative red blood cell targeting small molecules

Abstract: BackgroundChemotherapies for malaria and babesiosis frequently succumb to the emergence of pathogen-related drug-resistance. Host-targeted therapies are thought to be less susceptible to resistance but are seldom considered for treatment of these diseases.MethodsOur overall objective was to systematically assess small molecules for host cell-targeting activity to restrict proliferation of intracellular parasites. We carried out a literature survey to identify small molecules annotated for host factors implicated in Plasmodium falciparum infection. Alongside P. falciparum, we implemented in vitro parasite susceptibility assays also in the zoonotic parasite Plasmodium knowlesi and the veterinary parasite Babesia divergens. We additionally carried out assays to test directly for action on RBCs apart from the parasites. To distinguish specific host-targeting antiparasitic activity from erythrotoxicity, we measured phosphatidylserine exposure and hemolysis stimulated by small molecules in uninfected RBCs.ResultsWe identified diverse RBC target-annotated inhibitors with Plasmodium-specific, Babesia-specific, and broad-spectrum antiparasitic activity. The anticancer MEK-targeting drug trametinib is shown here to act with submicromolar activity to block proliferation of Plasmodium spp. in RBCs. Some inhibitors exhibit antimalarial activity with transient exposure to RBCs prior to infection with parasites, providing evidence for host-targeting activity distinct from direct inhibition of the parasite.ConclusionsWe report here characterization of small molecules for antiproliferative and host cell-targeting activity for malaria and babesiosis parasites. This resource is relevant for assessment of physiological RBC-parasite interactions and may inform drug development and repurposing efforts.

Abstract: Objective: Congenital hypothyroidism (CH) is an inborn thyroid hormone (TH) deficiency mostly caused by thyroidal (primary CH) or hypothalamic/pituitary (central CH) disturbances. Most CH newborn screening (NBS) programs are thyroid-stimulating-hormone (TSH) based, thereby only detecting primary CH. The Dutch NBS is based on measuring total thyroxine (T4) from dried blood spots, aiming to detect primary and central CH at the cost of more false-positive referrals (FPRs) (positive predictive value (PPV) of 21% in 2007–2017). An artificial PPV of 26% was yielded when using a machine learning-based model on the adjusted dataset described based on the Dutch CH NBS. Recently, amino acids (AAs) and acylcarnitines (ACs) have been shown to be associated with TH concentration. We therefore aimed to investigate whether AAs and ACs measured during NBS can contribute to better performance of the CH screening in the Netherlands by using a revised machine learning-based model. Methods: Dutch NBS data between 2007 and 2017 (CH screening results, AAs and ACs) from 1079 FPRs, 515 newborns with primary (431) and central CH (84) and data from 1842 healthy controls were used. A random forest model including these data was developed. Results: The random forest model with an artificial sensitivity of 100% yielded a PPV of 48% and AUROC of 0.99. Besides T4 and TSH, tyrosine, and succinylacetone were the main parameters contributing to the model’s performance. Conclusions: The PPV improved significantly (26–48%) by adding several AAs and ACs to our machine learning-based model, suggesting that adding these parameters benefits the current algorithm.

Abstract: Abstract African Americans have a significantly higher risk of developing chronic kidney disease, especially focal segmental glomerulosclerosis -, than European Americans. Two coding variants (G1 and G2) in the APOL1 gene play a major role in this disparity. While 13% of African Americans carry the high-risk recessive genotypes, only a fraction of these individuals develops FSGS or kidney failure, indicating the involvement of additional disease modifiers. Here, we show that the presence of the APOL1 p.N264K missense variant, when co-inherited with the G2 APOL1 risk allele, substantially reduces the penetrance of the G1G2 and G2G2 high-risk genotypes by rendering these genotypes low-risk. These results align with prior functional evidence showing that the p.N264K variant reduces the toxicity of the APOL1 high-risk alleles. These findings have important implications for our understanding of the mechanisms of APOL1-associated nephropathy, as well as for the clinical management of individuals with high-risk genotypes that include the G2 allele.

Abstract: A compact single-layer, polarization-insensitive, and angularly stable frequency selective surface (FSS) based multi-stop band filter is reported for shielding three useful frequency bands covering 1.82–2.86 GHz Industrial, scientific, and medical (ISM), 3.52–4.06 GHz Worldwide Interoperability for Microwave Access (WiMAX), and 7.42–8.72 GHz (satellite downlink), centered at 2.4 GHz, 3.6 GHz, and 8.1 GHz, respectively. The proposed unit cell (15 mm × 15 mm) contains four equal-sized square-headed dumbbell (SHD) shaped resonators surrounded by two square ring resonators. The outer and inner square rings offer the first and second stop bands, while the SHD resonators provide the third stop band. A highly polarization-insensitive response is realized owing to the four-fold symmetry in the proposed structure. The unique arrangements of the SHD resonators help to realize higher angular stability under transverse electric (TE), transverse magnetic (TM), and diagonally polarized incident electromagnetic (EM) waves for incidence angles up to 80°, 80°, and 70°, respectively. A detailed analysis in terms of equivalent circuit and parametric variation is carried out to illustrate the higher to lower frequency band ratio. A prototype is fabricated and tested through a proper measurement setup to validate its performance, and it shows good agreement with the simulated results. The proposed FSS unit cell offers better angular stability under diagonally polarised incident waves, attenuation level, minimum higher to lower frequency band ratio, good fractional bandwidth, and compactness. So the designed multi-stopband FSS can be considered a potential candidate for shielding EM radiation across the useful bands

Abstract: Abstract Hydrothermal carbonization (HTC) is a promising technology for waste valorisation and nutrient recovery to achieve sustainability. HTC converts organic waste into hydrochar, a carbon‐rich solid with numerous surface functionalities that can be used for energy and wastewater treatment. In this review, we highlight the potential of hydrochar‐based technology for improving the performance of anaerobic digestion (AD) systems and downstream applications of nutrient‐laden hydrochar. We identify knowledge gaps in hydrochar production, performance in AD systems and nutrient recovery, including the need for larger‐scale production facilities, multielement adsorption studies, and computational modelling. Techno‐economic analysis and life cycle assessment of hydrochar applications are critical to evaluating the commercial viability of this technology. Overall, hydrochar‐based technology offers a sustainable solution for waste management and resource recovery, with potential socioeconomic benefits for developing economies. The deployment of hydrochar‐based technology will directly address key issues highlighted in the United Nations' Sustainable Development Goals such as Clean water and sanitation (SDG 6); Zero hunger (SDG 2); and Climate action (SDG 13) thereby contributing to a more sustainable future.

36. Assessing Salinity Tolerance in Pinto Bean Varieties: Implications for Sustainable Agriculture

Abstract: Salinity is an abiotic stress restricting agricultural crop production globally, in which salts inhibit plants’ ability to absorb water and nutrients. Pinto beans (Phaseolus vulgaris L.) are very important in human nutrition and are sensitive to salinity. The objective of this study was to assess the salinity tolerance of six pinto bean varieties by evaluating the effect of different salt types on germination and growth. In the germination experiment, varieties were arranged in a randomized complete block design with five replications and three saline solutions (NaCl, CaCl2, MgSO4·7H2O) at 0, 0.05 M, 0.1 M, and 0.15 M concentrations each. For the greenhouse experiment, saline solutions with the same EC (5 dS m−1), control (distilled water), and six pinto bean varieties were organized in a Complete Random Design with 10 replicates. The results demonstrated that germination percentage, speed of germination, and hypocotyl length decreased as salt concentrations increased. Othello’s vegetative and reproductive parameters were significantly higher compared to the other varieties under saline conditions; its early maturity may have enabled it to perform better under salt stress. In addition to soil and water management, selecting salt-tolerant crops and varieties is essential to maintaining agricultural sustainability in regions undergoing salinization.

Abstract: Trypanosoma brucei causes African trypanosomiasis in humans. Infection with T. brucei elicits a potent pro-inflammatory immune response within infected human hosts, and this response is thought to at least be partially due to Toll-like receptor (TLR) activation. In response to stimulation by lipopolysaccharide and other pathogen antigens, TLR4 translocates to lipid rafts, which induces the expression of pro-inflammatory genes. However, cholesterol efflux is acknowledged as anti-inflammatory due to promoting lipid raft disruption. In this study, we wanted to assess the impact of T. brucei “ghosts”, which are non-viable T. brucei essentially devoid of intracellular contents, in stimulating macrophage TLR4 translocation to lipid rafts, and whether promoting cholesterol efflux in macrophages incubated with T. brucei ghosts attenuates TLR4-target gene expression. When cultured macrophages were exposed to T. brucei ghosts, we observed an increase in lipid raft TLR4 protein content, which suggests certain surface molecules of T. brucei serve as ligands for TLR4. However, pretreating macrophages with cholesterol acceptors before T. brucei ghost exposure decreased lipid raft TLR4 protein content and the expression of pro-inflammatory TLR4-target genes. Taken together, these results imply that macrophage cholesterol efflux weakens pro-inflammatory responses which occur from T. brucei infection via increasing macrophage lipid raft disruption.

Abstract: Abstract We present a search for the lepton-flavor-violating decays B s 0 $$ {B}_s^0 $$ → ℓ ∓ τ ± , where ℓ = e, μ, using the full data sample of 121 fb −1 collected at the Υ(5S) resonance with the Belle detector at the KEKB asymmetric-energy e + e − collider. We use B s 0 B ¯ s 0 $$ {B}_s^0{\overline{B}}_s^0 $$ events in which one B s 0 $$ {B}_s^0 $$ meson is reconstructed in a semileptonic decay mode and the other in the signal mode. We find no evidence for B s 0 $$ {B}_s^0 $$ → ℓ ∓ τ ± decays and set upper limits on their branching fractions at 90% confidence level as B $$ \mathcal{B} $$ ( B s 0 $$ {B}_s^0 $$ → e ∓ τ ± ) < 14 × 10 −4 and B $$ \mathcal{B} $$ ( B s 0 $$ {B}_s^0 $$ → μ ∓ τ ± ) < 7.3 × 10 −4. Our result represents the first upper limit on the B s 0 $$ {B}_s^0 $$ → e ∓ τ ± decay rate.

Abstract: Abstract We report the first measurement of the inclusive e + e − → b b ¯ $$ b\overline{b} $$ → D s ± $$ {D}_s^{\pm } $$ X and e + e − → b b ¯ $$ b\overline{b} $$ → D 0 / D ¯ 0 $$ {\overline{D}}^0 $$ X cross sections in the energy range from 10.63 to 11.02 GeV. Based on these results, we determine σ(e + e − → B s 0 B ¯ s 0 $$ {B}_s^0{\overline{B}}_s^0 $$ X) and σ(e + e − → B B ¯ $$ B\overline{B} $$ X) in the same energy range. We measure the fraction of B s 0 $$ {B}_s^0 $$ events at Υ(10860) to be f s = ( 22.0 − 2.1 + 2.0 $$ {22.0}_{-2.1}^{+2.0} $$ )%. We determine also the ratio of the B s 0 $$ {B}_s^0 $$ inclusive branching fractions B $$ \mathcal{B} $$ ( B s 0 $$ {B}_s^0 $$ → D 0 / D ¯ 0 $$ {\overline{D}}^0 $$ X)/ B $$ \mathcal{B} $$ ( B s 0 $$ {B}_s^0 $$ → D s ± $$ {D}_s^{\pm } $$ X) = 0.416 ± 0.018 ± 0.092. The results are obtained using the data collected with the Belle detector at the KEKB asymmetric-energy e + e − collider.

Abstract: Abstract We measure the cross section of e + e − → η c J/ψ at the Υ(nS)(n = 1–5) on-resonance and 10.52 GeV off-resonance energy points using the full data sample collected by the Belle detector with an integrated luminosity of 955 fb −1. We also search for double charmonium production in e + e − → η c J/ψ via initial state radiation near the η c J/ψ threshold. No evident signal of the double charmonium state is found, but evidence for the e + e − → η c J/ψ process is found with a statistical significance greater than 3.3σ near the η c J/ψ threshold. The average cross section near the threshold is measured and upper limits of cross sections are set for other regions.

Abstract: Transducin β-like 1 X-linked receptor 1 (TBL1XR1) is a WD40 repeat-containing protein and part of the corepressor complex SMRT/NCoR that binds to the thyroid hormone receptor (TR). We recently described a mutation in TBL1XR1 in patients with Pierpont syndrome. A mouse model bearing this Tbl1xr1 mutation (Tbl1xr1Y446C/Y446C) displays several aspects of the Pierpont phenotype. Although serum thyroid hormone (TH) concentrations were unremarkable in these mice, tissue TH action might be affected due to the role of TBL1XR1 in the SMRT/NCoR corepressor complex. The aim of the present study was to evaluate tissue TH metabolism and action in a variety of tissues of Tbl1xr1Y446C/Y446C mice. We studied the expression of genes involved in TH metabolism and action in tissues of naïve Tbl1xr1Y446C/Y446C mice and wild type (WT) mice. In addition, we measured deiodinase activity in liver (Dio1 and Dio3), kidney (Dio1 and Dio3) and BAT (Dio2). No striking differences were observed in the liver, hypothalamus, muscle and BAT between Tbl1xr1Y446C/Y446C and WT mice. Pituitary TRα1 mRNA expression was lower in Tbl1xr1Y446C/Y446C mice compared to WT, while the mRNA expression of Tshβ and the positively T3-regulated gene Nmb were significantly increased in mutant mice. Interestingly, Mct8 expression was markedly higher in WAT and kidney of mutants, resulting in (subtle) changes in T3-regulated gene expression in both WAT and kidney. In conclusion, mice harboring a mutation in TBL1XR1 display minor changes in cellular TH metabolism and action. TH transport via MCT8 might be affected as the expression is increased in WAT and kidney. The mechanisms involved need to be clarified.

Abstract: Thyroid hormone (TH) is indispensable for brain development in utero and during the first 2–3 years of life, and the negative effects of TH deficiency on brain development are irreversible. Detection of TH deficiency early in life by neonat al screening allows early treatment, thereby preventing brain damage. Inborn shortage of TH, also named congenital hypothyroidism (CH), can be the result of defective thyroid gland development or TH synthesis (primary or thyroidal CH (CH-T)). Primary CH is characterized by low blood TH and elevated thyroi d-stimulating hormone (TSH) concentrations. Less frequently, CH is due to insufficient stimulation of the thyroid gland because of disturbed hypothalamic or pituitary function (central CH). Central CH is characterized by low TH concentrations, while TSH is normal, lo w or slightly elevated. Most newborn screening (NBS) programs for CH are primarily TSH based and thereby do not detect central CH. Only a few NBS programs worldwide aim to detect both forms of CH by different strategies. In the Netherlands, we have a uni que T4–TSH–thyroxine-binding globulin (TBG) NBS algorithm for CH, which enables the detection of primary and central CH. Although the necessity of central CH detection by NBS is still under debate, it has been shown that most central CH patients have moderate-to-severe hyp othyroidism instead of mild and that early detection of central CH by NBS probably improves its clinical outcome and clinical care for central CH patients with multiple pituitary hormone deficiency. We are therefore convinced that detection of central CH by NBS is of utmost importance.

Abstract: Abstract Charged-lepton-flavor-violation is predicted in several new physics scenarios. We update the analysis of τ lepton decays into a light charged lepton (ℓ = e ± or μ ± ) and a vector meson (V 0 = ρ 0, ϕ, ω, K *0, or K ¯ $$ \overline{K} $$ *0) using 980 fb −1 of data collected with the Belle detector at the KEKB collider. No significant excess of such signal events is observed, and thus 90% credibility level upper limits are set on the τ → ℓV 0 branching fractions in the range of (1.7–4.3) × 10 −8. These limits are improved by 30% on average from the previous results.

45. Evaluating predictions of the patterning cascade model of crown morphogenesis in the human lower mixed and permanent dentition.

Abstract: ObjectiveThe patterning cascade model of crown morphogenesis has been studied extensively in a variety of organisms to elucidate the evolutionary history surrounding postcanine tooth form. The current research is the first to use a large modern human sample to examine whether the crown configuration of lower deciduous and permanent molars aligns with expectations derived from the model. This study has two main goals: 1) to determine if metameric and antimeric pairs significantly differ in size, accessory trait expression, and relative intercusp spacing, and 2) assess whether the relative distance among early-forming cusps accounts for observed variation in accessory cusp expression.MethodsTooth size, intercusp distance, and morphological trait expression data were collected from 3D scans of mandibular dental casts representing participants of the Harvard Solomon Islands Project. Paired tests were utilized to compare tooth size, accessory trait expression, and relative intercusp distance between diphyodont metameres and permanent antimeres. Proportional odds logistic regression was implemented to investigate how the odds of greater accessory cusp expression vary as a function of the distance between early-developing cusps.Results/significanceComparing paired molars, significant differences were identified for tooth size and cusp 5 expression. Several relative intercusp distances emerged as important predictors of cusp 6 expression, however, results for cusp 5 and cusp 7 did not match expected patterns. These findings support previous quantitative genetic results and suggest the development of neighboring crown structures represents a zero-sum partitioning of cellular territory and resources. As such, this study contributes to a better understanding of the foundations of deciduous and permanent molar crown variation in humans.

The main result in this paper is that $Q_{n,k}(E,\tau)$ has the same Hilbert series as the polynomial ring on $n$ variables when $\tau$ is not a torsion point. We also show that $Q_{n,k}(E,\tau)$ is a Koszul algebra, hence of global dimension $n$ when $\tau$ is not a torsion point, and, for all but countably many $\tau$, it is Artin-Schelter regular. The proofs use the fact that the space of quadratic relations defining $Q_{n,k}(E,\tau)$ is the image of an operator $R_{\tau}(\tau)$ that belongs to a family of operators $R_{\tau}(z):\mathbb{C}^n\otimes\mathbb{C}^n\to\mathbb{C}^n\otimes\mathbb{C}^n$, $z\in\mathbb{C}$, that (we will show) satisfy the quantum Yang-Baxter equation with spectral parameter., Comment: 51 pages + index + references

Abstract: Abstract We present the analysis of two-particle angular correlations using coordinate systems defined with the conventional beam axis and the event thrust axis. We propose the latter as a good representation for the correlation structure interpretation in the e + e − collision system. The e + e − collisions to hadronic final states at center-of-mass energies of s $$ \sqrt{s} $$ = 10.52 GeV and 10.58 GeV are recorded by the Belle detector at KEKB. In this paper, results on the first dataset are supplementary to the previous Belle publication [1]. At the same time, the latter is the first two-particle correlation measurement at collision energy on the Υ(4S) resonance and is sensitive to its decay products. Measurements are reported as a function of the charged-particle multiplicity. Finally, a qualitative understanding of the correlation structure is discussed using a combination of Monte Carlo simulations and experimental data.

Abstract: A set of measurements of azimuthal asymmetries in the production of pairs of identified hadrons in deep-inelastic scattering of muons on transversely polarised 6LiD (deuteron) and NH3 (proton) targets is presented. All available data collected in the years 2003–2004 and 2007/2010 with the COMPASS spectrometer using a muon beam of 160GeV/c at the CERN SPS were analysed. The asymmetries provide access to the transversity distribution functions via a fragmentation function that in principle may be independently obtained from e+e− annihilation data. Results are presented, discussed and compared to existing measurements as well as to model predictions. Asymmetries of π+π− pairs measured with the proton target as a function of the Bjorken scaling variable are sizeable in the range x>0.032, indicating non-vanishing transversity distribution and di-hadron interference fragmentation functions. As already pointed out by several authors, the small asymmetries of π+π− measured on the 6LiD target can be interpreted as indication for a cancellation of u and d-quark transversity distributions.
