Cutibacterium acnes (C. acnes) is a Gram-positive anaerobic facultative bacterium that is part of the human skin commensal microbiome. It colonizes various regions of the body, including the face, back, and chest. While typically a harmless commensal, under certain conditions, C. acnes can become pathogenic, leading to or promoting conditions such as acne vulgaris (AV), post-surgical infections, prostate cancer, and sarcoidosis. Current treatments for C. acnes infections often involve antibiotics, but the rise of antibiotic resistance has raised concerns. This review presents the virulence factors, clinical relevance, and current treatments of C. acnes, highlighting its association with AV, post-surgical infections, and other diseases. It also explores alternative innovative therapies such as phage therapy in development/research that are gaining prominence, with a growing focus on personalized medical approaches. To enhance C. acnes treatment while minimizing side effects and antibiotic prescription concerns, numerous clinical studies have been undertaken. These investigations span various pathological profiles and employ diverse strategies, such as utilizing bacterial extracts and compounds to restore healthy skin flora. The limitations and challenges of current and innovative treatments are also addressed, emphasizing the need for multidisciplinary strategies to combat C. acnes infections effectively. The objective of this work is to evaluate the influence of pomegranate peel extract (PPE) in the behavior of breast cell lines (epithelial and tumor type) and related oxidative metabolism. Fruit-based functional foods have been the target of increasing scientific research for their physiological and pathophysiological properties. Pomegranate (Punica granatum) is a suitable example with both prophylactic and medicinal effects. MCF-7 cell line from tumor breast carcinoma, and MCF-10A cell line from normal epithelial mammary gland were used and subjected to different concentrations of PPE, ranging from 1 to 5 mM of gallic acid equivalents (GAE). Viability, proliferation, mobility, and cytotoxicity assays were performed along with the quantification of antioxidant enzymes, namely, catalase, superoxide dismutase (SOD) and reduced (GSH) and oxidized (GSSG) glutathione. We observed a decrease in viability and proliferation of MCF-7 cells, at higher concentrations of PPE, with no influence in epithelial cells. Interestingly, in a concentration-dependent manner, PPE triggered a significant decrease in migration on both cell lines, with a more pronounced effect in breast cancer cell line. Regarding antioxidant enzyme activity, on tumor cells higher concentrations of PPE decreased catalase activity and significantly increased SOD activity. Regarding GSH and GSSG, we observed different expression levels between MCF-7 and MCF-10A, with MCF-7 presenting lower levels compared to MCF-10A. GSH/GSSG ratio was notably higher in MCF-7 at 5 mM GAE. PPE exhibits anti-tumor effects without significantly affecting normal epithelial cells. Our work strengthens the potential antitumoral effect of PPE by reducing MCF-7 cell viability and proliferation through the imbalance of antioxidant enzymes. [ABSTRACT FROM AUTHOR]

The material and technical study of the anthropomorphic coffin (No. Inv. MAC/MUM.1) belonging to the Archaeological Museum of Carmo, in Lisbon, Portugal, carried out by an interdisciplinary team, sought to complete the existing textual and iconographic study on the same coffin, as well as rectify and complete the data from another partial technical study. According to previous studies, the coffin was produced between the Low Period and the beginning of the Ptolemaic period (6th to 2nd centuries BCE), possibly in Akhmim. This work presents results regarding the coffin’s wooden support and polychromy, as well as its current state of conservation. It involved the macro and microscopic examination of the object and the analytical characterization of micro-samples by XRF, Raman spectroscopy, FTIR-ATR, and SEM. The materials identified both in polychromy and in the production of the support indicate the use of materials traditional from the Pharaonic Egypt., O estudo material e técnico do ataúde antropomórfico (Nº Inv.

- Abstract
EuroScitizen COST Action is a research network which aims to identify targeted strategies to raise levels of scientific literacy in Europe using evolution as a model. One of the activities developed by the EuroScitizen Media Working Group (WG4) was a set of Training Schools in Science and Media directed to young scientists and journalists. By uniting scientists and journalists, we aimed to promote better communication between the two groups while giving advanced training in science communication. The training activities were originally planned as one in-person Training School but converted to online events due to the COVID-19 pandemic. This required an adjustment of schedule and contents, and instead of having a training school for three full days, as in the planned in-person school, we developed several shorter sessions. Herein, we present the set of Science and Media training events we organised from June 2021 through February 2022, as well as the main challenges we encountered and what we found to be advantages of the online environment for training. We also share a checklist we developed for the organisation of training schools that may provide a useful tool for people planning other training events. CTX‐M‐151 new group ,polycyclic compounds ,CTX‐M β‐lactamases ,Extended‐spectrum β‐lactamases (ESBL) ,biochemical phenomena, metabolism, and nutrition ,bacterial infections and mycoses - Abstract
The production of extended‐spectrum β‐lactamases (ESBLs) is the main defense mechanism found in Gram negative bacteria. Among all the ESBLs, the CTX‐M enzymes appear as the most efficient in terms of dissemination in different epidemiological contexts. CTX‐M enzymes exhibit a striking plas-ticity, with a large number of allelic variants distributed in several sublineages, which can be associated with functional heterogeneity of clinical relevance. This observational analytical study provides an update of this family, currently with more than 200 variants described, from a phylogenetic, molecular, and structural point of view through homology in amino acid sequences. Our data, combined with described literature, provide phylogenetic and structural evidence of a new group. Thus, herein, we propose six groups among CTX‐M enzymes: the already stablished CTX‐M‐1, CTX‐M‐2, CTX‐M‐8, CTX‐M‐9, and CTX‐M‐25 clusters, as well as CTX‐M‐151 as the new cluster.

This work aimed to contribute to the deeper knowledge of Portuguese medieval polychrome sculpture. Polychrome sculpture had a significant role in the society of the Middle Ages, and today many objects survive throughout the country. However, the study of these objects’ materiality, and in particular of their polychromy, has been disregarded. This research consists of the first systematic study of the materials and techniques of the polychromy of Portuguese sculpture of the 14th and 15th centuries, a comparative study which envisioned understanding how polychromy was applied to objects on wood and on stone. In order to understand the production of these sculptures and to frame it in the universe of European practices, the defined methodology used various sources of information. The investigation of guild regulations, notarial and royal documentation, and other published written sources provided evidence regarding the professionals who were responsible for the different tasks of the sculptures’ production. Technical and artistic medieval treatises and recipe books were also investigated, and the recipes dealing with the painting of wood and stone supports were identified. A literature review of the materials and techniques of the polychromy of medieval European sculpture was carried out, looking to characterize these practices, compare the materials and techniques which have been identified in the polychromy of such distinct supports, and identify trends, continuing practices and peculiarities. This review confirmed the huge lack of knowledge of Portuguese objects regarding these subjects. The material and technical characterization study of a group of Portuguese polychrome sculptures with a multi-analytical approach provided novel information regarding the original polychromy of these sculptures and of the Portuguese polychrome production practices. The selected corpus of sculptures includes stone objects attributed to the workshops of masters Pero, João Afonso and Diogo Pires-oVelho, and a group of wooden sculptures of unknown authorship. Due to the importance of the results attained and the inherent need of confirming the production dates of the polychromies under study, it was tested the application of radiocarbon dating within the multi-analytical approach used for the study of these polychromies. Radiocarbon dating of the lead white pigment and organic binder of the paint layers proved to be a valuable tool to guide the study and interpretation of the complex paint stratigraphies of the polychrome sculptures. This thesis brings to light an ensemble of information unknown until now, contributing to the deeper knowledge of Portuguese polychrome sculpture, for the understanding of the organization of the workshop labour to produce these objects in the Middle Ages, and to contextualize Portuguese artistic practices within the universe of European practices.

Subcontracting ,Lean Methodology ,Automotive Industry - Abstract
The automotive industry represents one of the largest economic sectors in the world and most companies in this industry involve subcontracting in their strategy. However, subcontracting is one of the possible processes that can lead to inefficiencies for companies. Since lean arises as a response to the most varied problems of companies, the main goal of this work is to understand the importance of subcontracting in the automotive sector and to understand how lean and subcontracting are related. For this, an empirical study of papers and books related to these topics was performed, concluding that subcontracting is frequently used in the automotive sector, to obtain strategic advantages, and that it supports the foundations of the lean concept, as well as the lean can help manage subcontracting. The trade-off of the transfer of activities enables organizations to be flexible, lean and agile; they can then focus on their core activities, which generate more value, while the option to subcontract generates a significant reduction in costs by transferring non-core activities. In short, these two concepts are related to each other in the context of the automotive industry.

These achievements introduced novel opportunities in dating cultural heritage objects. Within this pioneering research, the possibility of a direct 14C dating of lead white pigment and organic binder in paint samples was investigated on polychrome sculptures, a foremost artistic expression in human history. The polychromy, an indivisible part of polychrome sculpture, holds a key role in the interpretation and understanding of these artworks. Unlike in other painted artworks, the study of polychromies is repeatedly hampered by repaints and degradation. The omnipresence of lead white within the original polychromy was thus pursued as dating proxy. Thermal decomposition allowed bypassing geologic carbonate interferences caused by the object's support material, while an added solvent extraction successfully removed conservation products. This radiocarbon dating survey of the polychromy from 16 Portuguese medieval limestone sculptures confirmed that some were produced within the proposed chronologies while others were revised. Within this multidisciplinary study, the potential of radiocarbon dating as a complementary source of information about these complex paint systems guiding their interpretation is demonstrated. The challenges of this innovative approach are highlighted and improvements on sampling and sample preparation are discussed. publishersversion published

Numa investigação entre memórias de manifestos e projectos experimentais, procura-se reflectir de que modo poderiam coincidir ou convergir, intersectando duas realidades, num paralelismo de possibilidades utópicas., Metabolism: Perpetuity of Impermanence proposes to evoke a journey to the Orient in a moment of retrospective of the sixties. We immerse ourselves in an experimental analysis of the modern memories of a singular architectural movement: Metabolism. Aiming at the study of its manifesto, a cultural spectrum extremely peculiar to the western reality is explored; involving international moments, notions are approached, sometimes parallel, almost tangent, which illusory touch on irreverent premises. It seeks to highlight certain historical moments that advocated Japanese modernism; understand the heuristic atmosphere that gave rise to the movement under study, exploring its ideologies and works chronologically. It proposes an analysis between traditionalism and modernism, involved in the Japanese identity, exploring its eclectic narrative. It is intended a dialectic, in rehearsal mode, which aims to observe the latest avant-garde movement of modern architecture. In an investigation between memories of manifestos and experimental projects, the aim is to reflect on how they could coincide or converge, intersecting two realities, in a parallelism of utopian possibilities.

The Burnout Syndrome is a psychological disturbance caused by excessive stress derived from an overburden or excess of work. This is a physical and mental breakdown as a consequence of an exhausting and overloaded professional life, which incapacitates the individual’s ability to perform daily duties, such as work. This report intends to contribute to the advance of the knowledge concerning the concept of “burnout”. The scope of this work is to demonstrate the historical and conceptual evolution of the conception of “burnout”, the factors that cause this syndrome, consequences, main theoretical model and measures for its prevention and intervention. There are also presented the approaches of the most notable authors working on this topic. The Burnout Syndrome is a multidimensional concept characterized by three main features: exhaustion, depersonalization and absenteeism, which differentiate it from the symptoms of depression. Thus, this report exposes the former concept.“Burnout” or Burnout Syndrome is spread worldwide, affecting millions of workers, particularly education and health workers. Portugal is no exception. Considering the increasing presence of this reality, it has been carried out a questionnaire directed to the nurses of the Instituto Português de Oncologia de Coimbra Francisco Gentil, E.P.E., in Coimbra. The intent of this is to determine how much these workers are affected by the symptoms of the Burnout Syndrome. Besides this, this questionnaire tries to identify the impacts of the symptoms in these workers and its impact on the decision to withdraw or not their profession and the workplace and how the organization’s administration can proceed. The results obtained with this study are impacting and significative. From 100 people polled from a total of 165 professionals, 63 in the age group 35-50 years old show symptoms of “burnout” and 74 disclose that they would withdraw their profession and current workplace.

The polychromy is an inseparable part of medieval polychrome sculptures, without which it cannot be fully understood. The degradation and loss of materials, as well as the frequent renovations of the polychromy over time result in a difficult reading and understanding of these surfaces. Material and technical studies provide a fundamental insight into the original appearance and intention of the artworks. However, the knowledge on Portuguese polychromy is scant, while in Europe it has been more systematically studied. This paper is a literature review on the materials and techniques of the polychromy of medieval sculptures, aiming to contextualize Portuguese polychromies within the European practices. Despite the difficulties in undertaking such comparisons among dissimilar studies, this review enabled to observe and identify geographical and chronological trends, patterns, and singularities. Above all, this review underlined that little is known regarding the polychromy of Portuguese medieval sculpture and reinforced the urgency of its systematic study.

Ecomuseu Municipal do Seixal is aiming to exhibit three oil portraits on canvas from the 19th century in the Tide Mill of Corroios. Due to its location, intrinsic to its function of Tide Mill, the building exhibits environmental conditions characterized by severe fluctuations in temperature and relative humidity. This work aims to propose a model of microclimate frame that offers a solution for the continued preservation of the three paintings. The frame’s design was chosen based on the study of the environmental conditions (temperature and relative humidity) of the exhibition room at the Tide Mill as well as in the results of an in situ experiment involving model frames. The proposal here described, a microclimate frame with silica gel as buffering material, considered the designs presented in the literature, the individual needs of the paintings and, finally, the available budget. publishersversion published

The main objective of this dissertation project in the 5th year of the Integrated Master in Industrial Engineering and Management of the University of Minho was to improve the analysis of the quantities to be produced, as well as the number of lines optimizing these same quantities in the section Product and Process Engineer - industrial engineering strand - from Bosch Car Multimedia Portugal, SA For this improvement a tool has been created from Excel that normalizes the process and eliminates waste. The research methodology used in this dissertation was the case study that began with a diagnosis of the current situation of the area under study. For this, it participated in several workshops, namely lean line design, scalling and study of the line number of so as to be able to map the flow of decision-making, identify the main wastes associated and identify possible proposals for improvement. The study of Excel tool implementation was carried out on a Housing assembly line characterized by a large diversity of product families with different production quantities and different process flows. With the implementation of this project it is expected to normalize and reduce the time of the weeks analysis process to only one hour. The cost of investment is also reduced from € 7 391 700.00 to € 7 175 500 €, which is a € 216 200.00 saving. It is also estimated the reduction of rework related to the creation of a new Excel is always necessary to study the number of lines, whether relative to the reduction of the processing times of the equipment, or to the increase of the quantities.

The collaborative robots, designed to share the workspace with the humans, have been used in several areas of industry. Despite the advantages associated with their use, currently the movements of these robotic platforms are generated through trajectory replication systems. Based in Bicho et al. (2011), Costa e Silva (2011) and Gulletta et al. (2015) works, in this dissertation was developed a method for motion planning of the collaborative robot Sawyer, making it capable to autonomously reach, grasp and manipulate objects, similar to that performed by humans. Considering the model based on the characteristics of the upper limbs movements by Rosenbaum et al. (2001), in this dissertation the joints angles of the robotic platform are the solution of nonlinear constrained optimization problems. This methodology allows the process of motion planning to be independent of the derivation of Sawyer’s inverse kinematics. This process is divided into two subproblems: (i) selection of final posture and (ii) selection of trajectory. The validation of the proposed method was performed in the context of two interaction tasks: (i) assembling a Toy Vehicle and (ii) assisting a human with reduced mobility. The obtained results demonstrate that the proposed planning method allows: (i) to generate trajectories with characteristics similar to those observed in the experimental work performed with human upper limbs and (ii) prevent collisions with existing obstacles in the workspace, with the operator and the robot’s own body.

The present work is divided in two parts: Part 1 is focused on the analysis and treatment of a 19th century portrait of Domingos Affonso, which belongs to the Ecomuseu Municipal do Seixal; and Part 2, which is entitled “The Microclimate Frame Project” is focused on the study of Artsorb® and on the planning of a microclimate frame for the painting. In Part 1, a study of the painting’s materials was performed using complementary analytical techniques and the painting’s condition was carefully evaluated. The painting exhibited signs of mould growth, and a more detailed investigation was made of this topic to understand if the fungal community was active and if it represented a real danger to the painting. A treatment was proposed, appropriate to the painting’s condition. A description of the treatment carried out, comprising the treatment options, is also present in this section. Within the study of the microclimate frame, in Part 2, the study of the potential corrosiveness of Artsorb® was a central subject. Artsorb® sheets are one of the most widely used materials for buffering relative humidity fluctuations in microclimate frames and its reported excellent performance is enhanced by its availability in lightweight sheets that can be easily placed inside microclimate frames. However, concerns have arisen regarding the presence of the corrosive salt lithium chloride in the composition of this buffer. Consequently, the present work also aimed to understand the potential risks of using Artsorb® and the possibility of avoiding exposure of lithium chloride to the artworks through the use of Tyvek®. Results from the preliminary tests seem to indicate that Artsorb® releases lithium chloride into air. This study also showed that a Tyvek® cover over Artsorb® reduces but does not eliminate evidence of chlorine contamination, and it significantly reduces the effectiveness of the buffering material. Considering that Artsorb® appears to be unsuitable due to the release of the corrosive salt, that Tyvek® was not efficient as a barrier for lithium chloride or as a permeable material to enable the proper functioning of Artsorb®, the buffering material proposed for the use in the microclimate frames is silica gel without indicator. Based on the choice of buffering material, as a result of this study, a microclimate frame is proposed.

Mestrado em Biologia Aplicada - Biologia Marinha Floating marine debris are present throughout the oceans and may have negative impacts on marine fauna, such as entanglement, ingestion and transport of exotic species. The present work aims at obtaining estimates of abundance and density of floating marine debris and vessels, as well as their spatial distributions along the entire study area. It also aims at identifying areas of spatial overlap between marine debris, cetaceans and vessels. The sighting surveys were performed during the summer of 2011, aboard the sailing vessel Santa Maria Manuela. This campaign was part of the project LIFE+ MARPRO - Conservation of marine protected species in Mainland Portugal, and the study area ranged between the 50 and the 220 nautical miles west of Portugal mainland, covering an area of about 252833 Km2. The survey was conducted along line transects, following a methodology that allowed to using conventional distance sampling methods to estimate the abundance and density of floating marine debris and vessels. Mean density (objects per km2) of debris in the study area overall was 2.96 objects per km2 and the total abundance was 747720 floating pieces. Floating plastic was the most common type of debris. In respect of vessels, mean density was 0.0013434 vessels per km2 and the estimated total abundance was 340 vessels.
