Fluctuating asymmetry (FA), defined as the random non-directional deviation from perfect bilateral symmetry, is considered to reflect developmental instability in bilaterally symmetrical organisms. It has been commonly used as a reliable biomarker of environmental stresses in natural populations. In agroecosystems, FA may arise through transgenerational influence of insecticides. We measured fluctuating asymmetry on unexposed progeny of Forficula auricularia mothers sampled in orchards that used no to frequent insecticides’ applications. Seven morphometric traits were measured on both right and left side and fluctuating asymmetry was calculated. Among these seven traits, four were more asymmetric when the progeny was born from a mother who lived in conventional orchards that used insecticides frequently compared to a mother who lived in insecticide free orchards. Males were more asymmetric than females for all traits but pest management strategy influenced FA in a similar way for both sexes. Measuring FA on unexposed daughters and sons of individuals originating from natural ecosystems may thus be a reliable easy-to-use tool to monitor mid-term effects of insecticides in the environment.

International audience; Apple orchards are heavily treated crops and some sprayed insecticides are recognized to have toxic effects on non-target arthropods. Earwigs are important natural enemies in pip-fruit orchards and contribute to the biological control of aphids. In addition, due to their ease of capture and identification, they are an interesting potential bioindicator of the possible detrimental effects of different orchard management strategies. In this study, we measured the energy reserves and some morphological traits of Forficula auricularia L. sampled in apple orchards under management strategies (organic versus integrated pest management (IPM)). We observed a significant decrease in mass (22 to 27%), inter-eye width (3%), and prothorax width (2 to 5%) in earwigs from IPM compared to organic orchards. Energy body reserves also confirmed these results with a significant decrease of 48% in glycogen and 25 to 42% in lipid content in earwigs from IPM compared to organic orchards. However, the protein content was approximately 70% higher in earwigs from IPM than in organic orchards. Earwigs sampled in IPM orchards may adapt to minimize the adverse toxic effects of pesticide treatments using a large number of strategies, which are reflected in changes to their energy reserves. These strategies could, in turn, influence the population dynamics of natural enemies and impair their role in the biological control of pests in apple orchards.

Since climate change is expected to bring more severe and frequent extreme weather events such as heat waves, assessing the physiological and behavioural sensitivity of organisms to temperature becomes a priority. We therefore investigated the responses of honeybees, an important insect pollinator, to simulated heat waves (SHW). Honeybees are known to maintain strict brood thermoregulation, but the consequences at the colony and individual levels remain poorly understood. For the first time, we quantified and modelled colony real-time activity and found a 70% increase in foraging activity with SHW, which was likely due to the recruitment of previously inactive bees. Pollen and nectar foraging was not impacted, but an increase in water foragers was observed at the expense of empty bees. Contrary to individual energetic resources, vitellogenin levels increased with SHW, probably to protect bees against oxidative stress. Finally, though immune functions were not altered, we observed a significant decrease in deformed wing virus loads with SHW. In conclusion, we demonstrated that honeybees could remarkably adapt to heat waves without a cost at the individual level and on resource flow. However, the recruitment of backup foraging forces might be costly by lowering the colony buffering capacity against additional environmental pressures.

In a rapidly changing environment, honeybee colonies are increasingly exposed to diverse sources of stress (e.g., new parasites, pesticides, climate warming), which represent a challenge to individual and social homeostasis. However, bee physiological responses to stress remain poorly understood. We therefore exposed bees specialised in different tasks (nurses, guards and foragers) to ancient (immune and heat stress) or historically more recent sources of stress (pesticides), and we determined changes in the expression of genes linked to behavioural maturation (vitellogenin – vg and juvenile hormone esterase – jhe) as well as in energetic metabolism (glycogen level, expression level of the receptor to the adipokinetic hormone – akhr, and endothermic performance). While acute exposure to sublethal doses of two pesticides did not affect vg and jhe expression, immune and heat challenges caused a decrease and increase in both genes, respectively, suggesting that bees had responded to ecologically relevant stressors. Since vg and jhe are expressed to a higher level in nurses than in foragers, it is reasonable to assume that an immune challenge stimulated behavioural maturation to decrease potential contamination risk and that a heat challenge promoted a nurse profile for brood thermoregulation. All behavioural castes responded in the same way. Though endothermic performances did not change upon stress exposure, the akhr level dropped in immune and heat-challenged individuals. Similarly, the abdomen glycogen level tended to decline in immune-challenged bees. Altogether, these results suggest that bee responses are stress specific and adaptive but that they tend to entail a reduction of energetic metabolism that needs to be studied on a longer timescale.

Summary 1. Declines in availability of plant resources to pollinators are a major cause of pollinator loss. The management of plant communities to enhance floral resources is often proposed as a way to sustain pollinator populations. Nectar, the main energetic resource for pollinators, plays a central role in behaviour and composition of pollinator communities. Abiotic and biotic factors are known to influence nectar traits at both the species and community levels, but the impact of plant community composition itself has never been investigated. 2. Below-ground interactions in plant communities can induce changes in plant development through (i) plant-derived litter amendment of the soil and (ii) competition for soil resources between plants. We tested how plant below-ground interactions affect above-ground nectar traits involved in plant attractiveness to pollinators. 3. A short-term pot experiment was carried out with three temperate grassland species Mimulus guttatus, Lamium amplexicaule, and Medicago sativa, showing distinct litter stoichiometry and competitive abilities for soil resources. Litter amendment (none, mono and tri-specific litter) and plant interaction treatments (monocultures, two- and three-species mixtures) were crossed in a factorial design. 4. Litter amendment to the soil led to an increase in total nectar sugar content in L. amplexicaule plants but not in the two other species. We also found that the presence of M. guttatus, a competitive species, reduced the total nectar sugar content in L. amplexicaule through a concomitant decrease in nectar volume per flower and in floral display size, but not in other species. Species-specific responses of nectar traits to variation in soil nitrogen availability were thus observed, suggesting consequences for plant species and community attractiveness to pollinators. However, we did not find evidence that the legume M. sativa affected nectar traits of any neighbouring plants. 5. Synthesis. Our results demonstrate that litter inputs and competition between plants for soil resources can alter nectar traits linked to plant attractiveness to pollinators. This supports the idea that below-ground plant–plant interactions for soil resources can influence above-ground plant–plant interactions for pollination services. This offers promising perspectives in studying the role of below-ground–above-ground interactions on higher trophic levels.

International audience; In vivo distribution of the neonicotinoid insecticide, imidacloprid, was followed during 72 h in six biological compartments of Apis mellifera L: head, thorax, abdomen, haemolymph, midgut and rectum. Honeybees were treated orally with 100 µg of 14 C-imidacloprid per kg of bee, a dose close to the median lethal dose. Elimination half-life of total radioactivity in honeybee was 25 h. Haemolymph was the compartment with the lowest and rectum that with the highest level of total radioactivity during the whole study, with a maximum 24 h after treatment. Elimination half-life of imidacloprid in whole honeybee was 5 h. Imidacloprid was readily distributed and metabolised only by Phase I enzymes into five metabolites: 4/5-hydroxy-imidacloprid, 4,5-dihydroxy-imidacloprid, 6-chloronicotinic acid, and olefin and urea derivatives. The guanidine derivative was not detected. The urea derivative and 6-chloronicotinic acid were the main metabolites and appeared particularly in midgut and rectum. The olefin derivative and 4/5-hydroxy-imidacloprid preferentially occurred in head, thorax and abdomen, which are nicotinic acetylcholine receptor-rich tissues. Moreover, they presented a peak value around 4 h after imidacloprid ingestion. These results explain the prolongation of imidacloprid action in bees, and particularly the differences between rapid intoxication symptoms and late mortality.

We examined the effects of sublethal doses (0.1, 1, and 10 ng per animal) of a new neonicotinoid insecticide, Imidacloprid, on habituation of the proboscis extension reflex (PER) in honeybees (Apis mellifera) reared under laboratory conditions. In untreated honeybees, the habituation of the proboscis extension reflex is agedependent and there is a significant increase in the number of trials required for habituation in older bees (8‐10 days old) as compared to very young bees (4‐7 days old). Imidacloprid alters the number of trials needed to habituate the honeybee response to multiple sucrose stimulation. In 7-day-old bees, treatment with Imidacloprid leads to an increase in the number of trials necessary to abolish the response, whereas in 8-day-old bees, it leads to a reduction in the number of trials for habituation (15 min and 1 h after treatment), and to an increase 4 h after treatment. The temporal effects of Imidacloprid in both 7- and 8-day-old bees suggest that 4 h after treatment the observed effects are due to a metabolite of Imidacloprid, rather than to Imidacloprid itself. Our results suggest the existence of two distinct subtypes of nicotinic receptors in the honeybee that have different affinities to Imidacloprid and are differentially expressed in 7- and 8-day-old individuals. q 2001 Academic Press

International audience; Honey bee colonies are highly dependent upon the availability of floral resources from which they get the nutrients (notably pollen) necessary to their development and survival. However, foraging areas are currently affected by the intensification of agriculture and landscape alteration. Bees are therefore confronted to disparities in time and space of floral resource abundance, type and diversity, which might provide inadequate nutrition and endanger colonies. The beneficial influence of pollen availability on bee health is well-established but whether quality and diversity of pollen diets can modify bee health remains largely unknown. We therefore tested the influence of pollen diet quality (different monofloral pollens) and diversity (polyfloral pollen diet) on the physiology of young nurse bees, which have a distinct nutritional physiology (e.g. hypopharyngeal gland development and vitellogenin level), and on the tolerance to the microsporidian parasite Nosema ceranae by measuring bee survival and the activity of different enzymes potentially involved in bee health and defense response (glutathione-S-transferase (detoxification), phenoloxidase (immunity) and alkaline phosphatase (metabolism)). We found that both nurse bee physiology and the tolerance to the parasite were affected by pollen quality. Pollen diet diversity had no effect on the nurse bee physiology and the survival of healthy bees. However, when parasitized, bees fed with the polyfloral blend lived longer than bees fed with monofloral pollens, excepted for the protein-richest monofloral pollen. Furthermore, the survival was positively correlated to alkaline phosphatase activity in healthy bees and to phenoloxydase activities in infected bees. Our results support the idea that both the quality and diversity (in a specific context) of pollen can shape bee physiology and might help to better understand the influence of agriculture and land-use intensification on bee nutrition and health.

International audience; Many laboratory studies have demonstrated that parasitoids of various species depend on sugar sources such as nectar or honeydew. However, studies about nectar acquisition by parasitoids foraging in the field are scarce. Tersilochus heterocerus Thomson is one of the more abundant and widespread parasitoids of the pollen beetle (Meligethes aeneus F.) but nothing is known about the nutritional ecology of this species. In this study we examined the nutritional state of T. heterocerus at the time of emergence and at various time periods throughout the season while foraging in the field using high-performance anion-exchange chromatography. We found that: (i) T. heterocerus emerge with relatively small amounts of sugar, composed mainly of trehalose, glucose and fructose; (ii) the first parasitoids caught just after they appeared in the field at the beginning of oilseed rape flowering had already consumed significant amounts of sugar reserves; and (iii) the total amount of sugar at the end of flowering was always significantly higher than the total amount of sugar at the beginning of flowering. This study provides valuable insights into the acquisition of sugar in the field by the parasitoid T. heterocerus and suggests that nectar acquisition takes place in the oilseed rape field or in the surrounding landscape.

Biotransformation of imidacloprid and the appearance of olefin and 5-hydroxyimidacloprid metabolites in the honeybee were studied by HPLC-MS/MS analysis. Honeybees were treated orally with imidacloprid at 20 and 50 µgkg −1 bee. Imidacloprid was metabolised relatively quickly and thoroughly. Twenty minutes after the beginning of imidacloprid ingestion, the sum of the residues from the three compounds amounted to only 70% of the actual given dose. Imidacloprid, 5-hydroxyimidacloprid and olefin represented, respectively, 50%, 9% and 8% of the actual ingested dose. Six and 24h, respectively, after ingestion of imidacloprid at 20 and 50 µgkg −1 bee, imidacloprid could no longer be detected in the honeybee. Imidacloprid had a half-life ranging between 4.5 and 5h and was rapidly metabolised into 5-hydroxyimidacloprid and olefin. Except 5-hydroxyimidacloprid in the 20 µgkg −1 treatment, these two metabolites presented a peak value 4h after ingestion of the 20 and 50 µgkg −1 doses. This time fully coincided with the appearance of mortality induced by imidacloprid after acute oral intoxication. These results suggested that the immediate neurotoxicity symptoms are due to the action of imidacloprid, whereas 5-hydroxyimidacloprid and/or olefin are involved in honeybee mortality. In addition, it was likely that the 30% of residues undetected 20min after intoxication were imidacloprid metabolites, although not 5-hydroxyimidacloprid or olefin. Imidacloprid is a systemic nitroguanidine insecticide that belongs to the neonicotinoid family. As an agonist of the acetylcholine receptor, it attacks the insect nervous system and is extremely effective against various sucking and mining pests. Oral acute and chronic toxicity of imidacloprid and its main metabolites (5-hydroxyimidacloprid, 4,5-dihydroxyimidacloprid, desnitroimidacloprid, 6-chloronicotinic acid, olefin, and urea derivative) were investigated in Apis mellifera. Acute intoxication by imidacloprid or its metabolites resulted in the rapid appearance of neurotoxicity symptoms, such as hyperresponsiveness, hyperactivity, and trembling and led to hyporesponsiveness and hypoactivity. For acute toxicity tests, bees were treated with doses of toxic compounds ranging from 1 to 1,000 ng/bee (10-10,000 microg/kg). Acute toxicity (LD50) values of imidacloprid were about 60 ng/bee (600 microg/kg) at 48 h and about 40 ng/bee (400 microg/kg) at 72 and 96 h. Out of the six imidacloprid metabolites tested, only two (5-hydroxyimidacloprid and olefin) exhibited a toxicity close to that of imidacloprid. Olefin LD50 values were lower than those of imidacloprid. The 5-hydroxyimidacloprid showed a lower toxicity than imidacloprid with a LD50 four to six times higher than that of imidacloprid. Urea also appeared as a compound of nonnegligible toxicity by eliciting close to 40% mortality at 1,000 ng/bee (10,000 microg/kg). However, no significant toxicity was observed with 4,5-dihydroxyimidacloprid, 6-chloronicotinic acid, and desnitroimidacloprid in the range of doses tested. To test chronic toxicity, worker bees were fed sucrose solutions containing 0.1, 1, and 10 microg/L of imidacloprid and its metabolites for 10 d. Fifty percent mortality was reached at approximately 8 d. Hence, considering that sucrose syrup was consumed at the mean rate of 12 microl/d and per bee, after an 8-d period the cumulated doses were approximately 0.01, 0.1, and 1 ng/bee (0.1, 1, and 10 microg/kg). Thus, all tested compounds were toxic at doses 30 to 3,000 (olefin), 60 to 6,000 (imidacloprid), 200 to 20,000 (5-OH-imidacloprid), and1,000 to 100,000 (remaining metabolites) times lower than those required to produce the same effect in acute intoxication studies. For all products tested, bee mortality was induced only 72 h after the onset of intoxication.

International audience; Imidacloprid (1-(6-chloro-3-pyridylmethyl)-N-nitroimidazolidin-2-ylideneamine) belongs to a new chemical family of chloronicotinyl compounds whose mode of action on the insect nervous system differs from that of traditional neurotoxic products. Imidacloprid, a strong systemic compound, is effective against several sucking and mining pests. The acute toxicity of contact and oral applications on two Apis mellifera subspecies, Apis mellifera mellifera and Apis mellifera caucasica, was investigated. In all toxicological studies, each dose included three cages of 20 individuals and each study was replicated three times. The dose-mortality relation revealed some unusual characteristics. At low imidacloprid concentrations, a biphasic mortality appeared, particularly with the contact exposure route. At moderate doses, mortality profiles at 24 and 48 h were different only after oral application. Response kinetics showed that mortality was delayed at the higher doses of imidacloprid. After oral intoxication, the LD50 values of imidacloprid at 24 and 48 h were about 5 ng/bee for both A. m. mellifera and A. m. caucasica. After contact application, the LD50 values at 24 and 48 h were approximately 24 ng/bee for A. m. mellifera and 14 ng/bee for A. m. caucasica. Imidacloprid ranks among the more potent contact insecticides in this important pollinator species.

For many flowering plant species cross pollination is necessary to ensure reproductive success. Most of these plants have evolved to encourage insect pollination which is mainly carried out by bees. Such is the case of plants in the Cucurbitaceae family, including the cantaloupe (Cucumis melo L.), an important crop of the South of France, and squirting cucumber (Ecballium elaterium (L.) A. Rich.), a wild plant restricted to the Mediterranean Basin, the two vegetal models used in this study. To better understand plant-pollinator relations, we proposed to characterise the interaction between Cucurbitaceae pollen and the honey bee (Apis mellifera L.) through pollen production, its presentation and its transport on the honey bee body. Our results showed that developmental anomalies took place during pollen ontogenesis in these species leading to 3% of the pollen grains produced to be empty. Moreover, we demonstrated that meiotic abnormalities in E. elaterium leading to the production of genetically abnormal pollen grains could be a source of autopolyploidy. At anthesis in C. melo, our results showed that pollen viability decreased to 30% when pollen was exposed to environmental conditions, then remained stable during the period of anthesis.The ability for pollen to germinate was completed just before anthesis then decreased to 12% at the end of anthesis. We determined that viability and germinability of cantaloupe pollen carried on the honey bee body can be enhanced or decreased according to which specific honey bee biochemical compounds were detected., Chez de nombreuses Angiospermes, la pollinisation croisée est nécessaire pour le succès de la reproduction. La plupart de ces plantes ont évolué afin de favoriser la pollinisation entomophile, principalement assurée par les abeilles. C’est le cas des Cucurbitacées, dont le melon (Cucumis melo L.), importante culture du sud de la France et le concombre d’âne (Ecballium elaterium (L.) A. Rich.) plante spontanée endémique du bassin méditerranéen, les deux modèles de cette étude. Afin de mieux comprendre les relations plante-pollinisateur,nous avons choisi de caractériser le couple pollen de Cucurbitacées/abeille domestique (Apis mellifera L.) à travers la production de pollen, sa présentation sur la fleur et son transport sur l’abeille. Nous avons montré que, chez ces plantes, l’ontogenèse du pollen s’accompagne d’erreurs développementales conduisant à la formation de près de 3% de grains de pollen vides. De plus, nous avons mis en évidence que chez E. elaterium, des erreurs méiotiques entraînaient la formation de grains de pollen génétiquement anormaux qui pourraient être source d’autopolyploïdie. Chez C. melo, nos résultats ont révélé que la mise en contact du pollen avec le milieu environnant entraîne une baisse de 30% de sa viabilité, mais que celle-ci reste stable jusqu’à la fin de l’anthèse. L’aptitude à germer, quant à elle, finit d’être acquise juste avant anthèse et décroît de 12% en fin de période d’anthèse. Nous avons montré que le pollen de C. melo transporté sur le corps de l’abeille pouvait soit avoir une viabilité et une aptitude à germer préservée, probablement par protection contre la déshydratation, soit perdre quasi-totalement sa viabilité et son aptitude à germer. Enfin, nous avons détecté des composés biochimiques spécifiques des abeilles protégeant ou diminuant la qualité reproductrice du pollen.
