Abstract: Objectives Informing research participants of the results of studies in which they took part is viewed as an ethical imperative. However, there is little guidance in the literature about how to do this. The Fluoxetine Or Control Under Supervision trial randomised 3127 patients with a recent acute stroke to 6 months of fluoxetine or placebo and was published in the Lancet on 5 December 2018. The trial team decided to inform the participants of the results at exactly the same time as the Lancet publication, and also whether they had been allocated fluoxetine or placebo. In this report, we describe how we informed participants of the results.Design In the 6-month and 12-month follow-up questionnaires, we invited participants to provide an email address if they wished to be informed of the results of the trial. We re-opened our trial telephone helpline between 5 December 2018 and 31 March 2019.Setting UK stroke services.Participants 3127 participants were randomised. 2847 returned 6-month follow-up forms and 2703 returned 12-month follow-up forms; the remaining participants had died (380), withdrawn consent or did not respond.Results Of those returning follow-up questionnaires, a total of 1845 email addresses were provided and a further 50 people requested results to be sent by post. Results were sent to all email and postal addresses provided; 309 emails were returned unrecognised. Seventeen people replied, of whom three called the helpline and the rest responded by email.Conclusion It is feasible to disseminate results of large trials to research participants, though only around 60% of those randomised wanted to receive the results. The system we developed was efficient and required very little resource, and could be replicated by trialists in the future.Trial registration number ISRCTN83290762; Post-results.

Abstract: Current evidence supports the use of intravenous thrombolysis with alteplase in patients with wake-up stroke selected with MRI or perfusion imaging and is recommended in clinical guidelines. However, access to advanced imaging techniques is often scarce. We aimed to determine whether thrombolytic treatment with intravenous tenecteplase given within 4·5 h of awakening improves functional outcome in patients with ischaemic wake-up stroke selected using non-contrast CT.TWIST was an investigator-initiated, multicentre, open-label, randomised controlled trial with blinded endpoint assessment, conducted at 77 hospitals in ten countries. We included patients aged 18 years or older with acute ischaemic stroke symptoms upon awakening, limb weakness, a National Institutes of Health Stroke Scale (NIHSS) score of 3 or higher or aphasia, a non-contrast CT examination of the head, and the ability to receive tenecteplase within 4·5 h of awakening. Patients were randomly assigned (1:1) to either a single intravenous bolus of tenecteplase 0·25 mg per kg of bodyweight (maximum 25 mg) or control (no thrombolysis) using a central, web-based, computer-generated randomisation schedule. Trained research personnel, who conducted telephone interviews at 90 days (follow-up), were masked to treatment allocation. Clinical assessments were performed on day 1 (at baseline) and day 7 of hospital admission (or at discharge, whichever occurred first). The primary outcome was functional outcome assessed by the modified Rankin Scale (mRS) at 90 days and analysed using ordinal logistic regression in the intention-to-treat population. This trial is registered with EudraCT (2014-000096-80), ClinicalTrials.gov (NCT03181360), and ISRCTN (10601890).From June 12, 2017, to Sept 30, 2021, 578 of the required 600 patients were enrolled (288 randomly assigned to the tenecteplase group and 290 to the control group [intention-to-treat population]). The median age of participants was 73·7 years (IQR 65·9-81·1). 332 (57%) of 578 participants were male and 246 (43%) were female. Treatment with tenecteplase was not associated with better functional outcome, according to mRS score at 90 days (adjusted OR 1·18, 95% CI 0·88-1·58; p=0·27). Mortality at 90 days did not significantly differ between treatment groups (28 [10%] patients in the tenecteplase group and 23 [8%] in the control group; adjusted HR 1·29, 95% CI 0·74-2·26; p=0·37). Symptomatic intracranial haemorrhage occurred in six (2%) patients in the tenecteplase group versus three (1%) in the control group (adjusted OR 2·17, 95% CI 0·53-8·87; p=0·28), whereas any intracranial haemorrhage occurred in 33 (11%) versus 30 (10%) patients (adjusted OR 1·14, 0·67-1·94; p=0·64).In patients with wake-up stroke selected with non-contrast CT, treatment with tenecteplase was not associated with better functional outcome at 90 days. The number of symptomatic haemorrhages and any intracranial haemorrhages in both treatment groups was similar to findings from previous trials of wake-up stroke patients selected using advanced imaging. Abstract: BACKGROUND: The prevalence of atrial fibrillation(AF) and diabetes mellitus is rising to epidemic proportions. We aimed to assess the impact of diabetes on the management and outcomes of patients with AF.METHODS: The EORP-AF General Long-Term Registry is a prospective, observational registry from 250 centres across 27 European countries. Outcomes of interest were as follows: i)rhythm control interventions; ii)quality of life; iii)healthcare resource utilisation; and iv)major adverse events.RESULTS: Of 11,028 patients with AF, the median age was 71 (63-77) years and 2537 (23.0%) had diabetes. Median follow-up was 24 months. Diabetes was related to increased use of anticoagulation but less rhythm control interventions. Using multivariable analysis, at 2-year follow-up, patients with diabetes were associated with greater levels of anxiety (p = 0.038) compared to those without diabetes. Overall, diabetes was associated with worse health during follow-up, as indicated by Health Utility Score and Visual Analogue Scale. Healthcare resource utilisation was greater with diabetes in terms of length of hospital stay (8.1 (±8.2) vs. 6.1 (±6.7) days); cardiology and internal medicine/general practitioner visits; and emergency room admissions. Diabetes was an independent risk factor of major adverse cardiovascular event (MACE; HR 1.26 [95% CI, 1.04-1.52]), all-cause mortality (HR 1.28 [95% CI, 1.08-1.52]), and cardiovascular mortality (HR 1.41 [95% CI, 1.09-1.83]).CONCLUSION: In this contemporary AF cohort, diabetes was present in 1 in 4 patients and it served as an independent risk factor for reduced quality of life, greater healthcare resource utilisation and excess MACE, all-cause mortality and cardiovascular mortality. There was increased use of anticoagulation therapy in diabetes but with less rhythm control interventions.

Abstract: Background Increased understanding of whether individuals who have recovered from COVID-19 are protected from future SARS-CoV-2 infection is an urgent requirement. We aimed to investigate whether antibodies against SARS-CoV-2 were associated with a decreased risk of symptomatic and asymptomatic reinfection. Methods A large, multicentre, prospective cohort study was done, with participants recruited from publicly funded hospitals in all regions of England. All health-care workers, support staff, and administrative staff working at hospitals who could remain engaged in follow-up for 12 months were eligible to join The SARS-CoV-2 Immunity and Reinfection Evaluation study. Participants were excluded if they had no PCR tests after enrolment, enrolled after Dec 31, 2020, or had insufficient PCR and antibody data for cohort assignment. Participants attended regular SARS-CoV-2 PCR and antibody testing (every 2–4 weeks) and completed questionnaires every 2 weeks on symptoms and exposures. At enrolment, participants were assigned to either the positive cohort (antibody positive, or previous positive PCR or antibody test) or negative cohort (antibody negative, no previous positive PCR or antibody test). The primary outcome was a reinfection in the positive cohort or a primary infection in the negative cohort, determined by PCR tests. Potential reinfections were clinically reviewed and classified according to case definitions (confirmed, probable, or possible) and symptom-status, depending on the hierarchy of evidence. Primary infections in the negative cohort were defined as a first positive PCR test and seroconversions were excluded when not associated with a positive PCR test. A proportional hazards frailty model using a Poisson distribution was used to estimate incidence rate ratios (IRR) to compare infection rates in the two cohorts. Findings From June 18, 2020, to Dec 31, 2020, 30 625 participants were enrolled into the study. 51 participants withdrew from the study, 4913 were excluded, and 25 661 participants (with linked data on antibody and PCR testing) were included in the analysis. Data were extracted from all sources on Feb 5, 2021, and include data up to and including Jan 11, 2021. 155 infections were detected in the baseline positive cohort of 8278 participants, collectively contributing 2 047 113 person-days of follow-up. This compares with 1704 new PCR positive infections in the negative cohort of 17 383 participants, contributing 2 971 436 person-days of follow-up. The incidence density was 7·6 reinfections per 100 000 person-days in the positive cohort, compared with 57·3 primary infections per 100 000 person-days in the negative cohort, between June, 2020, and January, 2021. The adjusted IRR was 0·159 for all reinfections (95% CI 0·13–0·19) compared with PCR-confirmed primary infections. The median interval between primary infection and reinfection was more than 200 days. Interpretation A previous history of SARS-CoV-2 infection was associated with an 84% lower risk of infection, with median protective effect observed 7 months following primary infection. This time period is the minimum probable effect because seroconversions were not included. This study shows that previous infection with SARS-CoV-2 induces effective immunity to future infections in most individuals. Abstract: BACKGROUND: Cardiac troponins (cTn) have been reported to be predictors for adverse outcomes in atrial fibrillation (AF), patients, but their actual use is still unclear.AIM: To assess the factors associated with cTn testing in routine practice and evaluate the association with outcomes.METHODS: Patients enrolled in the ESC-EHRA EORP-AF General Long-Term Registry were stratified into 3 groups according to cTn levels as (i) cTn not tested, (ii) cTn in range (≤99th percentile), (iii) cTn elevated (>99th percentile). The composite outcome of any thromboembolism /any acute coronary syndrome/cardiovascular (CV) death, defined as Major Adverse Cardiovascular Events (MACE) and all-cause death were the main endpoints.RESULTS: Among 10 445 AF patients (median age 71 years, 40.3% females) cTn were tested in 2834 (27.1%). cTn was elevated in 904/2834 (31.9%) and in-range in 1930/2834 (68.1%) patients. Female sex, in-hospital enrollment, first-detected AF, CV risk factors, history of coronary artery disease, and atypical AF symptoms were independently associated with cTn testing. Elevated cTn were independently associated with a higher risk for MACE (Model 1, hazard ratio [HR] 1.74, 95% confidence interval [CI] 1.40-2.16, Model 2, HR 1.62, 95% CI 1.28-2.05; Model 3 HR 1.76, 95% CI 1.37-2.26) and all-cause death (Model 1, HR 1.45, 95% CI 1.21-1.74; Model 2, HR 1.36, 95% CI 1.12-1.66; Model 3, HR 1.38, 95% CI 1.12-1.71).CONCLUSIONS: Elevated cTn levels were associated with an increased risk of all-cause mortality and adverse CV events. Clinical factors that might enhance the need to rule out CAD were associated with cTn testing.

Abstract: Summary Background BNT162b2 mRNA and ChAdOx1 nCOV-19 adenoviral vector vaccines have been rapidly rolled out in the UK from December, 2020. We aimed to determine the factors associated with vaccine coverage for both vaccines and documented the vaccine effectiveness of the BNT162b2 mRNA vaccine in a cohort of health-care workers undergoing regular asymptomatic testing. Methods The SIREN study is a prospective cohort study among staff (aged ≥18 years) working in publicly-funded hospitals in the UK. Participants were assigned into either the positive cohort (antibody positive or history of infection [indicated by previous positivity of antibody or PCR tests]) or the negative cohort (antibody negative with no previous positive test) at the beginning of the follow-up period. Baseline risk factors were collected at enrolment, symptom status was collected every 2 weeks, and vaccination status was collected through linkage to the National Immunisations Management System and questionnaires. Participants had fortnightly asymptomatic SARS-CoV-2 PCR testing and monthly antibody testing, and all tests (including symptomatic testing) outside SIREN were captured. Data cutoff for this analysis was Feb 5, 2021. The follow-up period was Dec 7, 2020, to Feb 5, 2021. The primary outcomes were vaccinated participants (binary ever vacinated variable; indicated by at least one vaccine dose recorded by at least one of the two vaccination data sources) for the vaccine coverage analysis and SARS-CoV-2 infection confirmed by a PCR test for the vaccine effectiveness analysis. We did a mixed-effect logistic regression analysis to identify factors associated with vaccine coverage. We used a piecewise exponential hazard mixed-effects model (shared frailty-type model) using a Poisson distribution to calculate hazard ratios to compare time-to-infection in unvaccinated and vaccinated participants and estimate the impact of the BNT162b2 vaccine on all PCR-positive infections (asymptomatic and symptomatic). This study is registered with ISRCTN, number ISRCTN11041050, and is ongoing. Findings 23 324 participants from 104 sites (all in England) met the inclusion criteria for this analysis and were enrolled. Included participants had a median age of 46·1 years (IQR 36·0–54·1) and 19 692 (84%) were female; 8203 (35%) were assigned to the positive cohort at the start of the analysis period, and 15 121 (65%) assigned to the negative cohort. Total follow-up time was 2 calendar months and 1 106 905 person-days (396 318 vaccinated and 710 587 unvaccinated). Vaccine coverage was 89% on Feb 5, 2021, 94% of whom had BNT162b2 vaccine. Significantly lower coverage was associated with previous infection, gender, age, ethnicity, job role, and Index of Multiple Deprivation score. During follow-up, there were 977 new infections in the unvaccinated cohort, an incidence density of 14 infections per 10 000 person-days; the vaccinated cohort had 71 new infections 21 days or more after their first dose (incidence density of eight infections per 10 000 person-days) and nine infections 7 days after the second dose (incidence density four infections per 10 000 person-days). In the unvaccinated cohort, 543 (56%) participants had typical COVID-19 symptoms and 140 (14%) were asymptomatic on or 14 days before their PCR positive test date, compared with 29 (36%) with typical COVID-19 symptoms and 15 (19%) asymptomatic in the vaccinated cohort. A single dose of BNT162b2 vaccine showed vaccine effectiveness of 70% (95% CI 55–85) 21 days after first dose and 85% (74–96) 7 days after two doses in the study population. Interpretation Our findings show that the BNT162b2 vaccine can prevent both symptomatic and asymptomatic infection in working-age adults. This cohort was vaccinated when the dominant variant in circulation was B1.1.7 and shows effectiveness against this variant. Abstract: Significance Hydrothermal veins supply much of the Earth’s gold. Their propensity to contain “bonanza” occurrences of gold that have concentrations millions of times greater than the concentration of gold in Earth’s crust makes them important targets for exploration and resource development. The mechanisms by which such hyperenrichment occurs are enigmatic. The accepted wisdom is that this enrichment reflects the saturation and precipitation of gold from hydrothermal fluids. Laboratory experiments and measurements of active hydrothermal systems, however, have shown that the solubility of this noble metal is exceptionally low. Here, we demonstrate that this issue is resolved by the physical transport of gold in the solid state as nanoparticles and their flocculated aggregates, thereby explaining the paradox of bonanza gold ore formation., Aqueous complexation has long been considered the only viable means of transporting gold to depositional sites in hydrothermal ore-forming systems. A major weakness of this hypothesis is that it cannot readily explain the formation of ultrahigh-grade gold veins. This is a consequence of the relatively low gold concentrations typical of ore fluids (tens of parts per billion [ppb]) and the fact that these “bonanza” veins can contain weight-percent levels of gold in some epithermal and orogenic deposits. Here, we present direct evidence for a hypothesis that could explain these veins, namely, the transport of the gold as colloidal particles and their flocculation in nanoscale calcite veinlets. These gold-bearing nanoveinlets bear a remarkable resemblance to centimeter-scale ore veins in many hydrothermal gold deposits and give unique insight into the scale invariability of colloidal flocculation in forming hyperenriched gold deposits. Using this evidence, we propose a model for the development of bonanza gold veins in high-grade deposits. We argue that gold transport in these systems is largely mechanical and is the result of exceptionally high degrees of supersaturation that preclude precipitation of gold crystals and instead lead to the formation of colloidal particles, which flocculate and form much larger masses. These flocculated masses aggregate locally, where they are seismically pumped into fractures to locally form veins composed largely of gold. This model explains how bonanza veins may form from fluids containing ppb concentrations of gold and does not require prior encapsulation of colloidal gold particles in silica gel, as proposed by previous studies.

Abstract: The Brucejack intermediate-sulfidation epithermal Au-Ag deposit, located 65 km north of Stewart, BC, forms part of a well-mineralized, structurally controlled, north-south gossanous trend associated with Early Jurassic intrusions straddling the Late Triassic-Early Jurassic Stuhini-Hazelton Group unconformity in the Sulphurets mineral district. Mining of the deposit commenced in mid-2017 after a long history of exploration dating back to the 1880s. Mineralization is hosted in deformed Lower Jurassic island-arc volcanic rocks of the Hazelton Group exposed on the eastern limb of the Cretaceous McTagg anticlinorium. High-grade Au-Ag mineralization was formed from ~184 to 183 Ma in association with a telescoped, multipulsed magmatic-hydrothermal system beneath an active local volcanic center. Precious metal mineralization occurs as coarse aggregates of electrum and silver sulfosalts in steeply dipping, E- to SE-trending quartz-carbonate vein stockwork zones cutting low-grade intrusion-related phyllic alteration. Epithermal vein development is interpreted to have occurred during the waning stages of Early Jurassic sinistral transpression in a compressive arc environment, followed by a limited Cretaceous deformation overprint.

Abstract: The high-grade (8.1 million ounces of gold; 15.6 Mt grading 16.1 g/t Au; Pretium, 2016) Brucejack epithermal Au-Ag deposits are located in the Canadian Cordillera of northwestern British Columbia, and formed in association with extensive early Mesozoic island arc magmatism. Porphyry-type Cu-Au-Mo mineralization occurs nearby at the Kerr-Sulphurets-Mitchell (196–190 Ma; Febbo et al., 2015), Bridge Zone (191.7 ± 0.8 Ma, 191.5 ± 0.8 Ma), and West Zone (188.9 ± 0.9 Ma) prospects. Gold-silver vein-type mineralization at Brucejack is hosted by variably altered and deformed Early Jurassic porphyritic latite lava flows, volcaniclastic rocks, and volcanic-derived sandstones, siltstones, and conglomerates. This study focuses on the Valley of the Kings Zone at Brucejack, where host rocks have been dated at 188–184 Ma (U-Pb, zircon). Post-mineralization (but not post-alteration) basaltic and trachybasaltic dikes that cut the veins are geochemically similar to the host volcanic sequence and follow the same structural trends of the mineralized vein corridors. We suggest these dikes are broadly coeval with mineralization, and that ore formation at the Valley of the Kings formed during the overall Jurassic tectonomagmatic event. Gold-silver mineralization is hosted by quartz-carbonate veins that cut sericitized and pyritized volcaniclastic rocks. Six stages of veining are defined, with Au-Ag mineralization (electrum) focused in stages III–V: stage I–III veins consist of quartz with minor carbonate, chlorite, sericite, and sulfide minerals; stage IV quartz veins contain more abundant base metal sulfides (pyrite, sphalerite, and galena with minor sulfosalts); stage V veins are dominantly calcitic; late post-mineralization stage VI veins are quartz-calcite and contain sparse pyrite and chlorite, but no electrum. The veins are interpreted to be syn- to late-tectonic, with deformation decreasing from locally penetrative in the host-rocks prior to veining (resulting in local foliation of sericite and pressure shadows around pyrite), to dismemberment and shearing of early stage I–II quartz veins, brittle disruption of stage III–IV quartz and stage V carbonate veins, and minimal deformation of post-mineralization stage VI quartz-carbonate veins. A much younger set of extensional muscovite veins locally cuts the deposit with Late Cretaceous apparent ages, and appears to be associated with a weak thermal overprint that has reset K-Ar and 40Ar/39Ar ages in sericite throughout the district. The deformation experienced by the vein system, particularly in the later stages (III–V), was not uniform, and original undeformed vein textures and fluid inclusion assemblages are locally preserved. We report fluid inclusion microthermometric data from carefully selected primary fluid inclusion assemblages from quartz, calcite, and sphalerite from vein stages III–IV, many of which show evidence for boiling (coexisting liquid- and vapor-rich primary fluid inclusions). Liquid-rich fluid inclusions from stage III and V veins have moderate homogenization temperatures (∼170 °C and ∼160 °C respectively) and salinities of 2–8 wt% NaCl equiv., whereas inclusions from base-metal-sulfide-bearing stage IV veins show evidence of mixing with a cooler, more saline brine (∼140 °C, ∼10–15 wt% NaCl equiv.). Carbon dioxide was observed as clathrate during cooling in some fluid inclusions, suggesting that minor amounts of CO2 were present in the fluids. Calculated oxygen isotopic compositions of fluids in equilibrium with quartz and calcite from vein stages III–V range from δ18Ofluid = −10.7 to +1.8‰, whereas δ13CCO2 ranges from −9.5 to −4.5‰, and δ34Spyrite ranges from −1.7 to +0.6‰. Taken together, these data suggest a magmatic source for S and some C, carried by a fluid of evolved or diluted magmatic origin, which variably mixed with meteoric-derived groundwater or seawater containing carbon of sedimentary (organic) origin. Mixing is supported by large salinity ranges of vein stage IV fluid inclusions, where individual assemblages range from ∼1 to 15 wt% NaCl equiv. These fluid temperatures, salinities, and isotopic compositions are typical of epithermal deposits where distal magmatic fluids mix with local heated groundwaters. The low fluid temperatures but evidence of boiling suggest formation at shallow crustal depths. This is despite the evidence for penetrative deformation in the sericitized volcanosedimetary host rocks, which we attribute to rapid uplift immediately prior to or during the earliest stages of mineralization. The vein system is therefore interpreted to have formed during the later stages of a deformational event related to arc accretion.

Abstract: Background: High blood pressure is common in acute stroke and is a predictor of poor outcome; however, large trials of lowering blood pressure have given variable results, and the management of high blood pressure in ultra-acute stroke remains unclear. We investigated whether transdermal glyceryl trinitrate (GTN; also known as nitroglycerin), a nitric oxide donor, might improve outcome when administered very early after stroke onset.Methods: We did a multicentre, paramedic-delivered, ambulance-based, prospective, randomised, sham-controlled, blinded-endpoint, phase 3 trial in adults with presumed stroke within 4 h of onset, face-arm-speech-time score of 2 or 3, and systolic blood pressure 120 mm Hg or higher. Participants were randomly assigned (1:1) to receive transdermal GTN (5 mg once daily for 4 days; the GTN group) or a similar sham dressing (the sham group) in UK-based ambulances by paramedics, with treatment continued in hospital. Paramedics were unmasked to treatment, whereas participants were masked. The primary outcome was the 7-level modified Rankin Scale (mRS; a measure of functional outcome) at 90 days, assessed by central telephone follow-up with masking to treatment. Analysis was hierarchical, first in participants with a confirmed stroke or transient ischaemic attack (cohort 1), and then in all participants who were randomly assigned (intention to treat, cohort 2) according to the statistical analysis plan. This trial is registered with ISRCTN, number ISRCTN26986053.Findings: Between Oct 22, 2015, and May 23, 2018, 516 paramedics from eight UK ambulance services recruited 1149 participants (n=568 in the GTN group, n=581 in the sham group). The median time to randomisation was 71 min (IQR 45–116). 597 (52%) patients had ischaemic stroke, 145 (13%) had intracerebral haemorrhage, 109 (9%) had transient ischaemic attack, and 297 (26%) had a non-stroke mimic at the final diagnosis of the index event. In the GTN group, participants' systolic blood pressure was lowered by 5·8 mm Hg compared with the sham group (pvs 3 [2–5]; n=558, in the sham group; 1·04 [0·84–1·29]; p=0·69). We found no difference in secondary outcomes, death (treatment-related deaths: 36 in the GTN group vs 23 in the sham group [p=0·091]), or serious adverse events (188 in the GTN group vs 170 in the sham group [p=0·16]) between treatment groups.Interpretation: Prehospital treatment with transdermal GTN does not seem to improve functional outcome in patients with presumed stroke. It is feasible for UK paramedics to obtain consent and treat patients with stroke in the ultra-acute prehospital setting.Funding: British Heart Foundation.

16. Geochemistry and tectonic setting of mafic rocks in western Dronning Maud Land, East Antarctica: implications for the geodynamic evolution of the Proterozoic Maud Belt

Abstract: On the basis of new bulk major and trace element (including REE) as well as Sm–Nd and Rb–Sr isotope data, used in conjunction with available geochronological data, a post-tectonic mafic igneous province and four groups of pre- to syntectonic amphibolite are distinguished in the polymetamorphic Maud Belt of western Dronning Maud Land, East Antarctica. Protoliths of the Group 1 amphibolites are interpreted as volcanic arc mafic intrusions with Archaean to Palaeoproterozoic Nd model ages and depletion in Nb and Ta. Isotopic and lithogeochemical characteristics of this earliest group of amphibolite indicate that the Maud Belt was once an active continental volcanic arc. The most likely position of this arc, for which a late Mesoproterozoic age ( c . 1140 Ma) is indicated by available U–Pb single-zircon age data, was on the southeastern margin of the Kaapvaal–Grunehogna Craton. The protoliths of Group 2 amphibolites are attributed to the 1110 Ma Borgmassivet–Umkondo thermal event on the basis of comparable Nd model ages and trace element distributions. Group 3 amphibolite protoliths are characterized by mid-ocean ridge basalt-type REE patterns and low Th/Yb ratios, and they are related to Neoproterozoic extension. Group 4 amphibolite protoliths are distinguished by high Dy/Yb ratios and are attributed to a phase of syntectonic Pan-African magmatism as indicated by Rb–Sr isotope data.

17. Tectono-thermal evolution of the Maud Belt: New SHRIMP U–Pb zircon data from Gjelsvikfjella, Dronning Maud Land, East Antarctica

Abstract: The Maud Belt in Dronning Maud Land (western East Antarctic Craton) preserves a high-grade polyphase tectono-thermal history with two orogenic episodes of Mesoproterozoic (1.2–1.0 Ga) and Neoproterozoic (0.6–0.5 Ga) age. New SHRIMP U–Pb zircon data from southern Gjelsvikfjella in the northeastern part of the belt make it possible to differentiate between a series of magmatic and metamorphic events. The oldest event recorded is the formation of an extensive 1140–1130 Ma volcanic arc. This was followed by 1104 ± 8 Ma granitoids that might represent, together with so far undated mafic dykes, part of a decompression melting-related bimodal suite that reflects the sub-continental Umkondo igneous event. The first high-grade metamorphism is constrained at 1070 Ma. The metamorphic age data are similar to those obtained from other parts of the Maud Belt, but also from the Namaqua-Natal Belt in South Africa, but the preceding arc formation was diachronous in the two belts. This indicates that the two belts did not form a continuous volcanic arc unit as suggested in previous models, but became connected only at the end of the Mesoproterozoic. Intense reworking during the Neoproterozoic, probably as a result of continent–continent collision between components of Gondwana, is indicated by ductile refliation, further high-grade metamorphic recrystallisation and metamorphic zircon overgrowths at approximately 530 Ma. This was followed by late- to post-tectonic magmatism, reflected by 500 Ma granite bodies and 490 Ma aplite dykes as well as a 480 Ma gabbro body.

18. Pan-African Tectonism in the Western Maud Belt: P–T–t Path for High-grade Gneisses in the H.U. Sverdrupfjella, East Antarctica

Abstract: Extensive high-grade polydeformed metamorphic provinces surrounding Archaean cratonic nuclei in the East Antarctic Shield record two tectono-thermal episodes in late Mesoproterozoic and late Neoproterozoic–Cambrian times. In Western Dronning Maud Land, the high-grade Mesoproterozoic Maud Belt is juxtaposed against the Archaean Grunehogna Province and has traditionally been interpreted as a Grenvillian mobile belt that was thermally overprinted during the Early Palaeozoic. Integration of new U–Pb sensitive high-resolution ion microprobe and conventional single zircon and monazite age data, and Ar–Ar data on hornblende and biotite, with thermobarometric calculations on rocks from the H.U. Sverdrupfjella, northern Maud Belt, resulted in a more complex P–T–t evolution than previously assumed. A c. 540Ma monazite, hosted by an upper ampibolite-facies mineral assemblage defining a regionally dominant top-to-NW shear fabric, provides strong evidence for the penetrative deformation in the area being of PanAfrican age and not of Grenvillian age as previously reported. Relics of an eclogite-facies garnet–omphacite assemblage within strainprotected mafic boudins indicate that the peak metamorphic conditions recorded by most rocks in the area (T ¼ 687–758 � C, P ¼ 9� 4–11� 3kbar) were attained subsequent to decompression from P >12� 9kbar. By analogy with limited U–Pb single zircon age data and on circumstantial textural grounds, this earlier eclogitefacies metamorphism is ascribed to subduction and accretion around 565Ma. Post-peak metamorphic K-metasomatism under amphibolite-facies conditions is ascribed to the intrusion of postorogenic granite at c. 480Ma. The recognition of extensive PanAfrican tectonism in the Maud Belt casts doubts on previous Rodinia reconstructions, in which this belt takes a pivotal position between East Antarctica, the Kalahari Craton and Laurentia. Evidence of late Mesoproterozoic high-grade metamorphism during the formation of the Maud Belt exists in the form of c. 1035Ma zircon overgrowths that are probably related to relics of granulite-facies metamorphism recorded from other parts of the Maud Belt. The polymetamorphic rocks are largely derived from a c. 1140Ma volcanic arc and 1072 � 10Ma granite.

Abstract: A regional fluid inclusion and stable isotope (H, O, C) study of the external part of the Pan-African Gariep Belt, with emphasis on the Rosh Pinah Zn-Pb sulphide deposit, demonstrated that the bulk of those fluids present at the time of syn-tectonic peak metamorphism and subsequent uplift and exhumation are of aqueous composition with low salinity. Exceptions are few moderately saline inclusions that are limited to the tectonic contact with the pre-Gariep basement near the volcano-sedimentary Rosh Pinah Formation. These are explained by derivation from a nearby, locally developed lagoonal, possibly evaporitic, facies. The infiltrated host rocks largely controlled the composition of the syn-orogenic fluids and no evidence was found for a regional pervasive flux of orogenic brines through the external parts of the belt. Very rare pre-orogenic fluids discovered in Rosh Pinah mine samples display markedly different composition and density that are in line with a derivation from a volcanic source. These fluid inclusions are interpreted as remnants of the original ore fluid that infiltrated the host sediments and pyroclastic rocks at a temperature of around 400°C and a hydrostatic pressure of a few hundred bars. This infiltration led not only to intense brecciation and silicification of arenitic footwall and sulphide precipitation but also to the dolomitisation of original limestone host rock. The good preservation of the syn-rift Rosh Pinah ore bodies during the Gariep orogeny is explained by the lack of regional orogenic brine flow.

Those sites at which the air had a common geographical origin and transport history are grouped together, and statistical measures of chemical characteristics are computed. Temporal changes in potential temperature are used to determine whether trajectories experience a significant convective influence during the 10-day period. Those trajectories that do not experience a significant convective influence are divided into four geographical categories depending on their origins and paths. Air parcels originating over Africa and South America are characterized by enhanced mixing ratios of O3, CO, HNO3, and PAN. The backward trajectories travel at high altitudes (∼10-11 km), covering long distances due to strong upper-tropospheric westerly winds. The observed enhancement of combustion-related species is attributed to biomass burning from distant sources to the west, extending even to South America. The relatively large value of Be-7 probably is due either to less efficient removal of aerosols from upper tropospheric air or to small stratospheric contributions. Aged marine parcels are found to have relatively small concentrations of burning-related species. Although these trajectories arrive at a wide range of aircraft altitudes, they do not pass over a land mass during the preceding 10-day period. Air passing over Australia but no other land mass exhibits a combustion signature; however, photochemical product species such as O3 and PAN are less enhanced than in the long-range transport category. These trajectories travel shorter distances and are at lower altitudes (∼5-8 km) than those reaching Africa and/or South America. The combustion influence on these parcels is attributed to biomass burning emissions injected over Australia. That burning is less widespread than in Africa and South America. Finally, trajectories originating over Southeast Asia appear to receive a weak combustion influence. However, compared to Africa and South America, Southeast Asia has a relatively small incidence of biomass burning. There is little combustion input from Australia due to the high transport altitudes compared to the lower heights of the convection. The Southeast Asian parcels exhibit the greatest NOx to ΣNOi ratio of any category, perhaps due to lightning. Parcels experiencing a significant convective influence also are examined. Most of these parcels pass through widespread, persistent convection along either the South Pacific Convergence Zone or Intertropical Convergence Zone approximately 5 days prior to arriving at the aircraft locations. Thus the category mostly represents marine convection. Mixing ratios of peroxides and acids in the convective category are found to be smaller than in parcels not experiencing convection. Abstract: The purpose of the present study was to investigate the relation between Type A behavior and adipose tissue linoleic acid composition.The sample consisted of 73 male and female Cretan adults, 23-64 years of age. Anthropometric and arterial blood pressure measurements were taken. Adipose tissue samples were collected by aspiration and data concerning general health habits were obtained. Food intake was estimated by a 24-hour dietary recall, while Type A profile was assessed with a Greek translation of the Bortner scale.Factors associated with Type A profile are adipose tissue linoleic acid (p < 0.0004), annual income (p < 0.001), total serum cholesterol (p < 0.009), and adipose fat myristoleic acid (p < 0.02). Negative correlations were observed between adipose linoleic and myristoleic acids and the dependent variable.It appears that adipose tissue linoleic acid may be one of the etiological factors for Type A behavior in adults.

Abstract: Extensive high-grade polydeformed metamorphic provinces surrounding Archaean cratonic nuclei in the East Antarctic Shield record two tectono-thermal episodes in late Mesoproterozoic and late NeoproterozoicCambrian times. In Western Dronning Maud Land, the high-grade Mesoproterozoic Maud Belt is juxtaposed against the Archaean Grunehogna Province and has traditionally been interpreted as a Grenvillian mobile belt that was thermally overprinted during the Early Palaeozoic. Integration of new UPb sensitive high-resolution ion microprobe and conventional single zircon and monazite age data, and ArAr data on hornblende and biotite, with thermobarometric calculations on rocks from the H.U. Sverdrupfjella, northern Maud Belt, resulted in a more complex PTt evolution than previously assumed. Abstract: Antibody-dependent cell-mediated cytotoxicity, mediated by natural killer (NK) cells and antibodies, emerged as a secondary correlate of protection in the RV144 HIV vaccine clinical trial, the only vaccine thus far demonstrating some efficacy in human. Therefore, leveraging NK cells with enhanced cytotoxic effector responses may bolster vaccine induced protection against HIV. Here, we investigated the effect of orally administering indole-3-carbinol (I3C), an aryl hydrocarbon receptor (AHR) agonist, as an adjuvant to an RV144-like vaccine platform in a mouse model. We demonstrate the expansion of KLRG1-expressing NK cells induced by the vaccine together with I3C. This NK cell subset exhibited enhanced vaccine antigen-specific cytotoxic memory-like features. Our study underscores the potential of incorporating I3C as an oral adjuvant to HIV vaccine platforms to enhance antigen-specific (memory-like) cytotoxicity of NK cells against HIV-infected cells. This approach may contribute to enhancing the protective efficacy of HIV preventive vaccines against HIV acquisition.

Abstract: Protein-protein interactions (PPIs) can be detected through selective complementation of split fluorescent reporters made of two complementary fragments that reassemble into a functional fluorescent reporter when in close proximity. We previously introduced splitFAST, a chemogenetic PPI reporter with rapid and reversible complementation. Here, we present the engineering of splitFAST2, an improved reporter displaying higher brightness, lower self-complementation, and higher dynamic range for optimal monitoring of PPI using an original protein engineering strategy that exploits proteins with orthology relationships. Our study allowed the identification of a system with improved properties and enabled a better understanding of the molecular features controlling the complementation properties. Because of the rapidity and reversibility of its complementation, its low self-complementation, high dynamic range, and improved brightness, splitFAST2 is well suited to study PPI with high spatial and temporal resolution, opening great prospects to decipher the role of PPI in various biological contexts.

29. Structural anisotropy results in mechano-directional transport of proteins across nuclear pores.

Abstract: The nuclear pore complex regulates nucleocytoplasmic transport by means of a tightly synchronized suite of biochemical reactions. The physicochemical properties of the translocating cargos are emerging as master regulators of their shuttling dynamics. As well as being affected by molecular weight and surface-exposed amino acids, the kinetics of the nuclear translocation of protein cargos also depend on their nanomechanical properties, yet the mechanisms underpinning the mechanoselectivity of the nuclear pore complex are unclear. Here we show that proteins with locally soft regions in the vicinity of the nuclear-localization sequence exhibit higher nuclear-import rates, and that such mechanoselectivity is specifically impaired upon knocking down nucleoporin 153, a key protein in the nuclear pore complex. This allows us to design a short, easy-to-express and chemically inert unstructured peptide tag that accelerates the nuclear-import rate of stiff protein cargos. We also show that U2OS osteosarcoma cells expressing the peptide-tagged myocardin-related transcription factor import this mechanosensitive protein to the nucleus at higher rates and display faster motility. Abstract: Means to analyse cellular proteins and their millions of variants at the single-molecule level would uncover substantial information previously unknown to biology. Nanopore technology, which underpins long-read DNA and RNA sequencing, holds potential for full-length proteoform identification. We use electro-osmosis in an engineered charge-selective nanopore for the non-enzymatic capture, unfolding and translocation of individual polypeptides of more than 1,200 residues. Unlabelled thioredoxin polyproteins undergo transport through the nanopore, with directional co-translocational unfolding occurring unit by unit from either the C or N terminus. Chaotropic reagents at non-denaturing concentrations accelerate the analysis. 31. Enhanced statistical sampling reveals microscopic complexity in the talin mechanosensor folding energy landscape.

Abstract: Statistical mechanics can describe the major conformational ensembles determining the equilibrium free-energy landscape of a folding protein. The challenge is to capture the full repertoire of low-occurrence conformations separated by high kinetic barriers that define complex landscapes. Computationally, enhanced sampling methods accelerate the exploration of molecular rare events. However, accessing the entire protein's conformational space in equilibrium experiments requires technological developments to enable extended observation times. We developed single-molecule magnetic tweezers to capture over a million individual transitions as a single talin protein unfolds and refolds under force in equilibrium. When observed at classically-probed timescales, talin folds in an apparently uncomplicated two-state manner. As the sampling time extends from minutes to days, the underlying energy landscape exhibits gradually larger signatures of complexity, involving a finite number of well-defined rare conformations. A fluctuation analysis allows us to propose plausible structures of each low-probability conformational state. The physiological relevance of each distinct conformation can be connected to the binding of the cytoskeletal protein vinculin, suggesting an extra layer of complexity in talin-mediated mechanotransduction. Abstract: Non-native disulfide bonds are dynamic covalent bridges that form post-translationally between two cysteines within the same protein (intramolecular) or with a neighboring protein (intermolecular), frequently due to changes in the cellular redox potential. The reversible formation of non-native disulfides is intimately linked to alterations in protein function; while they can provide a mechanism to protect against cysteine overoxidation, they are also involved in the early stages of protein multimerization, a hallmark of several protein aggregation diseases. Yet their identification using current protein chemistry technology remains challenging, mainly because of their fleeting reactivity. Here, we use single-molecule spectroscopy AFM and molecular dynamics simulations to capture both intra- and intermolecular disulfide bonds in γD-crystallin, a cysteine-rich, structural human lens protein involved in age-related eye cataracts. Our approach showcases the power of mechanical force as a conformational probe in dynamically evolving proteins and presents a platform to detect non-native disulfide bridges with single-molecule resolution.

Abstract: Molecular fluctuations directly reflect the underlying energy landscape. Variance analysis examines protein dynamics in several biochemistry-driven approaches, yet measurement of probe-independent fluctuations in proteins exposed to mechanical forces remains only accessible through steered molecular dynamics simulations. Using single molecule magnetic tweezers, here we conduct variance analysis to show that individual unfolding and refolding transitions occurring in dynamic equilibrium in a single protein under force are hallmarked by a change in the protein's end-to-end fluctuations, revealing a change in protein stiffness. By unfolding and refolding three structurally distinct proteins under a wide range of constant forces, we demonstrate that the associated change in protein compliance to reach force-induced thermodynamically stable states scales with the protein's contour length increment, in agreement with the sequence-independent freely jointed chain model of polymer physics. Our findings will help elucidate the conformational dynamics of proteins exposed to mechanical force at high resolution which are of central importance in mechanosensing and mechanotransduction.
